Gastrointestinal nematodes (GIN) use flexible life history strategies to maintain their fitness under environmental challenges. Costs incurred by a challenge to one life trait can be recouped by increasing the expression of subsequent life traits throughout their life cycle. Anticipating how parasites respond to the challenge of control interventions is critical for the long-term sustainability of the practice and to further ensure that the parasites withstand favourable adaptive responses. There is currently limited information on whether distinct populations of a GIN species respond to the same environmental challenge in a consistent manner, with similar alterations to their life history strategies or comparable fitness outcomes. This study compared the life history traits and experimental fitness of three distinct Haemonchus contortus isolates exposed to environmental challenges at both the parasitic (i.e., passage through resistant or susceptible sheep) and free-living (i.e., exposure to diverse climatic conditions) life stages. The key findings show that H. contortus maintain their fitness under challenge with isolate-specific alterations to their life history strategies. Further, partial exploration of the H. contortus isolates transcriptomes using cDNA-AFLP methods confirmed disparate expression profiles between them. These results bring fresh insights into our understanding of the non-genetic adaptive processes of GIN that may hinder the efficacy of parasite control strategies.

Background and Aim: Promotions of goat farming by both public and private sectors encouraged considerable goat raising in central Thailand. Gastrointestinal nematodes (GIN) infection is a major health and economic problem; however, evidence of resistance to broad-spectrum anthelmintics is frequently reported. Investigation of anthelmintic resistance (AR) status and identification of factors related to the development of AR is important components for sustainable GIN control. However, no information is available on this topic in the study area. The present study aimed to gather information on GIN control practices and to evaluate the effectiveness of albendazole, ivermectin, and levamisole for treating GIN infestation in goat herds in Sing Buri Province. Materials and Methods: Twenty-nine herds were randomly selected. Information on management practices was collected by face-to-face interview using a structured questionnaire. Three field experiments for routinely used anthelmintics, including albendazole, ivermectin, and levamisole were conducted from June 2019 to November 2019. Fecal samples were collected pre- and post-treatment and examined for fecal egg count reduction to determine the status of anthelmintic resistance of goat GIN. Results: Several improper practices were identified that lead to AR, especially chronic use of albendazole and ivermectin. All herds were considered resistant to albendazole and ivermectin, and levamisole resistant nematodes were detected in two herds. AR was strongly linked with the continuous use of anthelmintics. Conclusion: Levamisole, which was still effective in the province, should be used with caution to minimize the selection of resistant strains. Farmers should be provided with updated information for sustainable parasite control. Further, the efficacy of anthelmintics should be routinely monitored.

This study examined the gastrointestinal parasitological status of three endangered Sub-Saharan antelope species (Addax nasomaculatus, Oryx dammah, Gazella dorcas) hosted at Souss-Massa National Park in Morocco. A total of 254 faecal samples (80 samples from the addax population, 81 from the oryx population and 93 from the dorcas population) were analysed to determine the prevalence and the intensity of the parasites in host faeces (expressed as the mean EPG: egg per gram), using microscopic methods (Flotation and McMaster) and the molecular identification of parasites using PCR and sequencing of the second internal transcribed spacer region of the rDNA (ITS-2).The prevalence results in the addax, oryx and dorcas gazelle were 43.7%, 2.4%, and 61.3%, respectively, for Nematodirus spp.; 21.2%, 12.3%, and 16.13%, respectively, for Trichuris spp.; and 36.2%, 39.5%, and 53.7%, respectively, for other, undistinguished strongylids.The means of EPG values for parasites in addax, oryx and dorcas gazelle were 8.9, 2.4, and 61.3, respectively, for Nematodirus spp.; 4.3, 2.4, and 4.8, respectively, for Trichuris spp.; and 18.1, 16.6, and 50.1, respectively, for other undistinguished strongylids. Sequencing of the ITS-2 rDNA region of the isolated parasites allowed the identification of Camelostrongylus mentulatus and Nematodirus spathiger in these three antelope species. Da bi se ispitala prevalencija i intenzitet infestacije parazitima kod životinja, ukupno je ispitano 254 uzoraka fecesa (80 uzoraka iz addax populacije, 81 uzoraka iz orix populacije i 93 uzoraka iz dorcas populacije) pri čemu su rezultati prikazivani kao srednja vrednost EPG (jajašca parazita po gramu fecesa). Korišćene su mikroskopske metode (Flotacija i McMaster) kao i molekularna identifi kacija parazita upotrebom PCR testa i sekvencioniranjem rDNK (ITS-2). Rezultati prevalence Nematodirus spp kod addax, oryx i dorcas gazela bili su: 43,7%, 2,4% odnosno 61,3%; za Trichuris spp vrste 21,2%, 12,3% i 16,3%, a za strongilidne vrste koje nisu mogle da se determinišu: 36,2%, 39,5% i 53,7%. Srednje EPG vrednosti za parazite kod adax, oryx i dorcas gazele bili su za Nematodirus spp vrste 8,9, 2,4 i 61,3; za Trichuris 4,3, 2,4 i 4,8 a za na strongilidne vrste koje nisu mogle da se determinišu, 18,1, 16,6 odnosno 50,1. Rezultati sekvencioniranja ITS-2 regiona rDNK izolovanih parazita omogućio je identifi kaciju Camelostrongylus mentulatus i Nematodirus spathinger u navedene tri vrste antilopa. Može da se zaključi da su infestacije parazitima kod ove tri vrste antilopa na podnošljivom nivou pri čemu je po prvi put dijagnostikovana infestacija antilopa u Maroku sa Camelostrongylus mentulatus i Nematodirus spathinger

The objective of this study was to describe the serum protein pattern in sheep with naturally acquired gastrointestinal parasitosis and to compare the distribution of protein fractions with the results obtained in animals with negative fecal examination results. Fecal and blood samples were taken from twenty-nine sheep positive for nematode eggs and twenty-four animals with negative results of fecal examination. Between the evaluated groups of sheep significant differences were noted in the relative mean values for all protein fractions and for albumin/globulin ratio (p2-globulins and A/G ratio were significantly lower, the mean concentrations of α1-, β-, γ1- and γ2-globulins significantly higher in the nematode positive group of sheep (p2-zone in three sheep and the γ-globulin zones were characterized by a diffuse higher broad and wide peaks. The presented results indicate that the gastrointestinal parasitic infections in sheep alter the distribution of serum proteins, and suggest their usefulness in animals with unchanged serum protein concentrations. The study brings new findings and extends the knowledge about the metabolic responses and consequences of gastrointestinal parasitic infections in sheep, particularly with regard to alterations in protein metabolism.

Gastrointestinal nematode (GIN) infestations are a major constraint to sheep production in the West Indies (WI). Intensive and semi-intensive management systems are most commonly employed. These islands display tropical weather patterns with wet and dry seasons. Semi-intensive farming combined with increased rainfall during the wet season has been reported to be most favourable for development and survival of GIN. This study was conducted to determine whether there was a relationship between GIN burdens in sheep with seasonality and management practices of farmers in Trinidad and Tobago (T&T). Farms were visited on a monthly basis from January to December 2017. A maximum of ten sheep, three to nine months of age, were selected from each farm. A total of 3,053 faecal samples were collected and analysed using the Modified McMaster technique. Environmental data on daily precipitation and temperature were collected from the Trinidad and Tobago Meteorological Office during the period of sampling. A mixed effects negative binomial regression model was constructed to analyse the relationship between gastrointestinal nematode faecal egg counts (GINFEC) with season, management system and use of dewormers as fixed effects and farm as a random effect. Average diurnal temperatures of T&T fluctuated between 23.2°C to 32.6°C and 23.9°C to 32.3°C in the dry and wet seasons, respectively. Sheep gastrointestinal nematode (GIN) infestation represents a limiting factor for sheep farming and milk production in Italy. The development of anthelmintic resistance to conventionally used drugs suggests the path towards the use of natural remedies as a possible alternative. The purpose of this study is to evaluate the in vitro anthelmintic efficacy of the hydroalcoholic extracts of basal leaves (It-BL), cauline leaves (It-CL) and flowers (It-F) of Isatis tinctoria (Brassicaceae), a spontaneous Sicilian species renowned as an important source of bioactive compounds. The dry extracts of the different parts of the plant were tested using the egg hatch test (EHT) in vitro to verify the efficacy against ovine GIN at different concentrations (1.00, 0.5, 0.25, 0.125 mg/mL). Thiabendazole and deionized water were used as positive and negative controls, respectively. The results obtained from EHT indicated that all the I. tinctoria extracts were highly effective (p < 0.0001) in inhibiting egg hatching within 48 h of exposure. The in vitro inhibitory effect was never less than 84% in all doses tested, and it was only slightly lower than the standard drug thiabendazole (95.6%). The current study documents the anthelmintic activity of I. tinctoria against sheep’s GIN, suggesting its application as alternative natural method to limit the use of antiparasitic drugs.

Herbal products such as essential oils may play a promising role in the treatment of infections caused by gastrointestinal nematodes (GINs). The aim of this study was to evaluate the in vitro potential of 11 essential oils (EOs) and one binary combination of isolated EO compounds, as well as the in vivo anthelmintic efficacy of two EO formulations. Four GIN genera were identified in the coproculture examination: Haemonchus, Trichostrongylus, Teladorsagia and Chabertia. The in vitro egg hatch test (EHT) was performed at six different concentrations (50, 12.5, 3.125, 0.781, 0.195 and 0.049 mg/mL) for each EO, whereas in the in vivo faecal egg count reduction test (FECRT), each EO sample was diluted in sunflower oil and orally administrated at a dose of 100 mg/kg to the different group of animals. In the EHT, the EOs of Origanum vulgare, Foeniculum vulgare, Satureja montana, Satureja hortensis and two types of Thymus vulgaris were the most effective. The dominant compounds of these EOs were carvacrol, thymol, anethol, p-cymene and γ-terpinene, indicating their importance for the anthelmintic activity. In the FECRT, both T. vulgaris EO type 1 and linalool:estragole combination show an anthelmintic potential with a mean effect on FECR of approximately 25%. The results suggest the possible role of tested EOs as anthelmintic agents in sheep farms, although further in vivo tests are needed. © 2022 by the authors. Licensee MDPI, Basel, Switzerland.

The development of anthelmintic resistance in sheep gastrointestinal nematodes (GINs) requires novel strategies for the sustainable control of these parasites. This study aimed to evaluate the anthelmintic efficacy of the Origanum vulgare (L.) essential oil (EO) against sheep GINs and to evaluate the possibility of its use in control practice. The in vitro egg hatch test was conducted at eight different concentrations (50, 12.5, 3.125, 0.781, 0.195, 0.049, 0.025, and 0.0125 mg/mL) of the tested EO. For the in vivo fecal egg count reduction test, the EO of O. vulgare was administrated orally at a mean single dose of 150 mg/kg to sheep from two farms in Southern Italy, whereby potential toxic effects to the hosts were also evaluated. In the egg hatch test, the inhibition of egg hatchability varied from 71.3% to 93.7%, depending on the concentration used. The high anthelmintic potential was confirmed in the fecal egg count reduction test with an average reduction of nematode eggs in feces of 43.2% and 60.1% on days 7 and 14 after treatment, respectively. In addition, no toxic effects were noticed during the clinical examination of sheep or by observing blood count and liver or kidney function test results. The obtained results suggest the strong activity of the O. vulgare EO against sheep GINs, probably due to a high percentage of carvacrol (76.21%), whereby it can be considered safe for sheep at the dose tested in vivo. Therefore, it is suitable for use in veterinary practice as a part of an integrated strategy for the control of sheep GINs.

Haemonchus contortus is an infectious gastrointestinal nematode parasite of small ruminants. This study addresses the in vitro/in vivo anti-haemonchiasis potential, toxicological effects, and mechanism of action of nanoparticles. Online databases were used to search and retrieve the published literature (2000 to 2021). A total of 18 articles were selected and reviewed, out of which, 13 (72.2%) studies reported in vitro, 9 (50.0%) in vivo, and 4 (22.2%) both in vitro/in vivo efficacy of different nanoparticles. Mostly, organic nanoparticles (77.7%) were used including polymeric (85.7%) and lipid nanoparticles (14.3%). The highest efficacy, in vitro, of 100% resulted from using encapsulated bromelain against eggs, larvae, and adult worm mortality at 4, 2, and 1 mg/ml, respectively. While in vivo, encapsulated Eucalyptus staigeriana oil reduced worm burden by 83.75% and encapsulated Cymbopogon citratus nano-emulsion by 83.1%. Encapsulated bromelain, encapsulated Eucalyptus staigeriana oil, and encapsulated Cymbopogon citratus nano-emulsion were safe and non-toxic in vivo. Encapsulated bromelain damaged the cuticle, caused paralysis, and death. Nanoparticles could be a potential source for developing novel anthelmintic drugs to overcome the emerging issue of anthelmintic resistance in H. contortus. Studies on molecular effects, toxicological consequences, and different pharmacological targets of nanoparticles are required in future research.

Resistance to anthelmintic drugs in gastrointestinal nematodes (GIN) of sheep is of high concern for livestock production worldwide. In Calabria (southern Italy), many plants have been used in ethnoveterinary medicine for parasite control in small ruminants. Here, we present an in vivo evaluation of anthelmintic efficacy of three plant extracts. The first was based on bark and leaves of Salix caprea, the second and the third were based on the whole plant Artemisia campestris and whole fruit (seeds and peel) of Punica granatum, respectively. Anthelmintic efficacy was evaluated according to the fecal egg count reduction test (FECRT) performed with the FLOTAC technique. The results showed a significant anthelmintic effect of Punica granatum macerate (50%), a low effectiveness of the Artemisia campestris macerate (20%), and a complete ineffectiveness of Salix caprea macerate (0.1%). With these outcomes, we report a P. granatum-based remedy reducing 50% GIN egg output. This result was obtained without using any synthetic drug, paving the way for the employment of green veterinary pharmacology (GVP) as a complementary and sustainable method to reduce the use of chemicals and to counteract anthelmintic resistance.

Small ruminants are very affected by gastrointestinal nematode (GIN) parasitism worldwide. The improvement of the host protective response and the reduction of the probability of contact between hosts and parasites appear as very promising strategies. The objective of this study was to evaluate the effect of a combination of two components of these two strategies on natural GIN infection and production performances of Creole goat kids: dietary supplementation and a rotational mixed grazing system. One hundred and twenty weaned Creole kids and six Creole heifers were divided into four experimental batches: Splus (supplemented) vs. Szero (non-supplemented) × Mixed grazing (kids associated with heifers) vs. Control (kids alone), and grazing plots of forage grasses were balanced for live weight (LW) in both species. The feed intake, blood, and parasitological parameters and production performances between 3 and 11 months of age were monitored. The fecal egg count (FEC) was significantly lower and the packed cell volume (PCV) significantly higher in the Mixed grazing groups. No effect of supplementation was observed for FEC. In contrast, PCV, body condition score, and live weight were significantly higher in supplemented animals whatever the groups. Mixed grazing system and supplementation had significant effects on the slaughter parameters (P < 0.05), but there was no significant interaction. Our results suggested that the advantage driven either by mixed grazing or dietary supplementation on kids' performances would be equivalent, and the combination of both would improve the animal performances.

Gastrointestinal nematodes (GIN) negatively impact productivity and welfare in sheep globally and are estimated to cost the European sheep industry €157-477 million annually. GIN are mainly controlled by anthelmintic treatment, however, as anthelmintic resistance becomes prominent, the routine treatment of ewes against GIN has been questioned. A questionnaire survey of 383 sheep farmers in Great Britain was conducted to identify strategies currently used to control GIN infections in ewes. Ordinal and binary regression analysis were used to identify factors associated with use of practices known to influence anthelmintic resistance development, including number and timing of ewe GIN anthelmintic treatment, targeted selective treatment (TST) of ewes, drench and move of ewes and long-acting moxidectin treatment of periparturient ewes. Participating farmers treated their ewes against GIN 1.68 times per year on average, with 42.3% and 32.1% of participating farmers worming their ewes once or twice a year on average, respectively. 17.2% of participating farmers wormed their ewes more than twice a year, and 8.4% never worm their ewes. Participating farmers who devised GIN control strategies based on SCOPS guidelines treated their ewes significantly less per year (P 0.001), whilst those determining treatment timing based on ewe DAG scores or the time of year treated their ewes significantly more frequently (P 0.001). Farmers who devised GIN control strategies in conjunction with their vet had greater odds of using TST (P 0.001), as well as farmers who determined flock treatment timing based on ewe condition (P = 0.027). The use of narrow spectrum flukicides was significantly associated with reduced number of annual ewe GIN anthelmintic treatments (P 0.001), TST of ewes against GIN (P 0.001) and the avoidance of moving ewes to clean pastures following GIN treatment (P 0.001). The presence of sheep scab on a farm or in the area was significantly associated with increased annual GIN treatments for ewes (P = 0.002), not using TST strategies to control GIN in ewes (P 0.001) and moving ewes to clean pasture after anthelmintic treatment, whilst using macrocyclic lactones treatments to prevent sheep scab was significantly associated with the treatment of periparturient ewes with long-acting moxidectin (P = 0.001). This research suggests that by encouraging the application of evidence based targeted or targeted selective treatment strategies, further interaction between farmers and veterinarians/SCOPS guidance, and the uptake of best practices for controlling liver fluke and sheep scab on farms, sustainable GIN control strategies can become the common practice in ewes.

Our current understanding of differences in the epidemiology of gastrointestinalnematode (GIN) species in co-grazed sheep and goats is inadequate with reference to the development of sustainable control strategies. The next-generation metabarcoded sequencing method referred to as the ‘nemabiome’ allows some of these differences to be explored to describe the intensity of co-infecting GIN species. We applied this platform to study sheep and goats that were co-grazed on Guinea grass pasture in northeastern Brazil. Co-grazed goats and sheep were treated with a monepantel anthelmintic, then exposed to the same gastrointestinal nematode species. Overall, there were differences in the prevalence of GIN species identified in the sheep and goats;Trichostrongylus colubriformis and Teladorsagia circumcincta predominated in goat kids, while Haemonchus contortus predominated in adult does, ewes and lambs once burdens became re-established after anthelmintic treatment. Description of the pattern of re-infection following anthelmintic treatment was prevented by the unpredicted poor efficacy of 2.5 mg/kg and 5 mg/kg, respectively, of monepantel against O. columbianum and T. circumcincta in lambs, and T. circumcincta adult does. Differences in drug efficacy between host age and species groups may be important when considering sustainable GIN control strategies for co-grazed animals. The aggregated FECs of the adult does and goat kids representing re-established GIN burdens, were higher than those of the co-grazedadult ewes and lambs. This implies that there are inherent differences in GIN species adaptation to the two naïve small ruminant host species, and shows the need for better understanding of the factors giving rise to this situation associated with exposure to infective larvae and host responses. At the start of the study, the adult does were coinfected with several GIN species, with the highest intensity of T. circumcincta, contrasting with the situation in the adult ewes, in which H. contortus predominated.However, once burdens became re-established after treatment, H. contortus predominated in both adult does and ewes. This demonstrates the potential for host burdens of H. contortus to establish and predominate after anthelmintic treatment when burdens of coinfecting GIN species are low.

This study aimed to determine control measures for gastrointestinal nematodes in goats in the northeastern semiarid after analyzing the dynamics of gastrointestinal helminths during the drought, the evolution of the parasitic load after the first rains and the differences in susceptibility between goats of different categories and ages. Five farms were studied from March 2013 to January 2015. Feces were collected from all goats every month, for fecal egg counts (FECs). No treatment was required on any farm during the dry period. In 2013, with annual rainfall of 265-533 mm, treatments were not necessary during the rainy season. However, in 2014, with rainfall of 604-778 mm, treatments were necessary 60-90 days, after the first rains. On three farms, gastrointestinal nematodes showed multiple anthelmintic resistance. The FECs from lactating goats were significantly higher than from dry and young goats. In conclusion, in the Brazilian semi-arid region (Caatinga biome), it is generally unnecessary to treat grazing goats during the dry season. In the rainy season, the parasite load increases 2-3 months after the first rains. In both, the dry and the rainy season, farmers should monitor their herds by means of FEC or another criterion (anemia or submandibular edema), to determine the need to treat. RESUMO: Este trabalho objetivou determinar medidas de controle de nematódeos gastrintestinais de caprinos no semiárido nordestino após análise da dinâmica das infecções helmínticas durante a seca, a evolução da carga parasitária após as primeiras chuvas e as diferenças de susceptibilidade entre caprinos de distintas categorias e idades. Em cinco propriedades, de março de 2013 a janeiro de 2015, foram coletadas, mensalmente, fezes de todos os caprinos para contagem de ovos. Em nenhuma propriedade foi necessário vermifugar durante os períodos de seca. Em 2013, com precipitações de 265-533 mm anuais, não foi necessário vermifugar durante o período de chuva. No entanto, em 2014, com precipitações de 604-778 mm, foi necessário vermifugar 60-90 dias após as primeiras chuvas em três propriedades. Nessas três propriedades foi encontrada multirresistência aos anti-helmínticos. Foi constatado que o OPG das cabras lactantes foi significativamente maior do que o OPG das cabras secas e dos cabritos. Em conclusão, na região semiárida, geralmente, não é necessário o tratamento das cabras pastejando na caatinga durante a estação seca. Na estação chuvosa, a carga parasitária aumenta 2-3 meses após as primeiras chuvas. Tanto na seca quanto nas chuvas, o produtor deve monitorar o rebanho mediante OPG ou por outros critérios (anemia, edema submandibular) para determinar a necessidade de vermifugação.

Infections with gastrointestinal nematodes (GIN) adversely affect meat color in lambs. Although white-rot fungi (WRF) pretreatment increases nutritional value and fiber digestion of corn straw for lambs, whether it can improve meat quality of lambs infected with GINs is unknown. The objective of this experiment was to study effects of feeding WRF-pretreated corn straw on the health and meat quality of lambs infected with GINs. Sixteen healthy Ujumqin lambs were orally drenched with 3rd-stage GINs larvae and randomly divided into two dietary treatments of control (CON) and WRF diets for 70 days of feeding. Results showed that feeding WRF-pretreated corn straw decreased L* and b* values (p &lt, 0.05) and increased a* value (p &lt, 0.01) of both longissimus thoracis et lumborum (LTL) and semimembranosus (SM) muscles of lambs infected with GINs. Feeding WRF-pretreatment corn straw decreased fecal egg count (p = 0.014) and increased packed cell volume (p = 0.013) of lambs from 28 d of feeding and increased plasma iron content (p = 0.008) of lambs from 56 d of the feeding. Feeding WRF-pretreatment corn straw decreased myosin heavy-chain (MyHC)-I (p = 0.032) and MyHC-II&alpha, (p = 0.025) content in LTL muscle and MyHC-I (p = 0.022) and MyHC-II&beta, (p = 0.048) in SM muscle of lambs. In conclusion, although there were no significant changes in the content of most amino acids or increased intensity of better flavor compounds, meat quality and health of lambs infected with GINs was significantly improved by feeding WRF-pretreated corn straw due to increased PCV and meat color and tenderness.

The objective of the present study was to study, under field conditions, potential associations between gastrointestinal nematode parasitism and subclinical mastitis in ewes during the lactation period. Faecal and milk samples were collected from 240 ewes in 16 farms, for parasitological and bacteriological plus cytological examination, respectively. In the population sampled, prevalence of gastrointestinal nematode infection was 63.0%; mean faecal count was 357.7 ± 32.4 eggs per gram (epg); Teladorsagia spp. larvae were identified more frequently in coprocultures (median proportion among farms: 53.0%). The prevalence of subclinical mastitis was 22.6%; coagulase-negative Staphylococcus spp. were identified more frequently as causal agents (64.7% of isolates recovered from cases of the disease). There was clear evidence that the prevalence of subclinical mastitis was higher among ewes with gastrointestinal nematode infection than among ewes without: 26.4% and 16.1% (P = 0.047), respectively. Further, the prevalence of subclinical mastitis was higher in ewes with high faecal epg: 31.1% versus 18.6% in ewes with low faecal epg (P = 0.027). Mean epg counts in ewes with mastitis were significantly higher: 500 ± 84 (mean ± standard error of the mean), than epg counts in healthy ewes: 316 ± 36 (P = 0.024). The findings further underline the importance of concurrent parasitic and bacterial infections. Moreover, results also suggest further factors that can play a role in development of mastitis in ewes.

Gastrointestinal nematode infection is one of the major diseases affecting small ruminants. Although some breeds of goats are quite resistant, many breeds of goats are relatively susceptible. This study used a combined parasitological, immunological, bioinformatic and statistical approach to examine the role of goat IgA and eosinophils in protection against Teladorsagia circumcincta. Molecular modelling suggested that the transmembrane domain of the high affinity IgA receptor was dysfunctional in goats. Statistical analyses failed to find any association in naturally infected goats between high IgA or eosinophil responses and low faecal egg counts. Together these results indicate that IgA and eosinophil responses against T. circumcincta are less effective in goats than sheep.

Gastrointestinal nematode infection in sheep is a significant challenge for production with implications for animal health and welfare, and lamb growth. Flocks rely on the chemical control of worm burdens, but with the increasing challenge of anthelmintic resistance there is a necessity for farmers to uptake SCOPs (Sustainable Control of Parasites) guidance. Vets need to communicate practical strategies to farmers and suggest novel approaches that address these challenges so that holistic and sustainable parasite management can be achieved. This article examines the challenge of nematode infections and anthelmintic resistance and suggests how hazard analysis and critical control point (HACCP) application may facilitate a whole-flock strategy.

Cross-sectional surveys of sheep parasite control practices in Australia were conducted in 2004, 2012 and 2019 to document parasite problems, control practices and measure change over time. This article reports the results pertaining to gastrointestinal nematode infection; comparisons between years are mostly descriptive and not based on statistical inference. There was a general increase in the use of grazing management to prepare clean pastures for sheep to control gastrointestinal nematode infections with the largest increases in the use of: cropping, long acting anthelmintics to prepare clean pastures, feeding strategies, selecting rams for resistance to nematodes, and leaving some sheep untreated. The proportion of respondents using faecal worm egg count monitoring (WEC) and the number of WEC monitors per year were similar in 2003 (weaners: 3.0 WEC/year, adult ewes: 2.6 WEC/year) and 2018 (lambs and weaners and adult ewes both 3.1 WEC/year) but lower in 2011(lambs and weaners: 2.0 WEC/year, adult ewes: 2.9 WEC/year), whilst there was a higher number of anthelmintic treatments given in 2011 than 2003 and 2018 which were similar. There was an increase in the proportion of respondents carrying out WEC monitors themselves from 2011 (21%) to 2018 (30%) and a corresponding reduction in the use of private laboratories, government laboratories and veterinarians with regional differences in who carried out WECs. The top three anthelmintic groups used did not differ between the three survey years with macrocyclic lactones the most used followed by benzimidazoles (BZ) and levamisole (LEV) although the percentage of MLs used in 2011 and 2018 was lower than in 2003 with higher proportions of respondents using BZ and LEV groups in the latter two survey years. Moxidectin was in the most commonly used active in all survey years. There was an increase in the use of combination of anthelmintics from different groups, especially for a combination of three anthelmintics (2003: 4.4%, 2011: 19.1%, 2018: 21.5%), with lower use of single anthelmintics in 2011 (57.0%) and 2018 (55.4%) compared with 2003 (74.5%). The use of testing for anthelmintic resistance was generally low across the survey years with a lower proportion of respondents using tests in 2011 and 2018 than in 2003 (2003:48%, 2011: 29%, 2018: 37%). Time of year, results from WEC and seasonal weather condition were the most important factors when deciding when to apply anthelmintic treatment.

Excreta egg counting techniques are used for indirectly estimating the magnitude of gastrointestinal nematode infection in live animals. The aim of this study was to optimise laboratory and field sampling methods for routine monitoring of nematode infections in chickens by evaluating the sensitivity, accuracy, and precision of the Modified McMaster (MM) and Mini-FLOTAC (MF) methods using laying chicken excreta samples spiked with estimated true numbers of eggs (Experiment 1 = 5-1500 EPG (eggs/g); Experiment 2 = 5-500 EPG) without and with operator effects, respectively or using individual fresh excreta (n = 230) and fresh floor excreta (n = 42) from naturally infected free-range layer farms. The Coefficient of Variation (CV) was assessed within and between operators and the time spent on sample preparation and counting was also evaluated. MF was more sensitive than MM at ≤ 50 EPG level but not above this while MM had a significantly higher egg recovery rate than MF for ≥ 50 EPG levels (MM = 89.7 %, MF = 68.2 %; P 0.0001). Operator factors did not have a significant effect (P = 0.358-0.998) on egg counts across methods and EPG levels. The CV between replicates of the MM and MF methods for ≥ 50 EPG was 43.4 and 36.5 %, respectively. The inter-observer CV of the MM and MF methods for ≥ 50 EPG levels was 63.8 and 44.3 % respectively. When the naturally infected free-range layers which were individual caged for excreta sampling, the proportion of samples positive for MM and MF were 91.7 and 96.5 %, respectively (P = 0.023). MM resulted in significantly (P = 0.029) higher excreta egg counts (604) than MF (460) with the difference between methods greatest at higher EPG levels. Fresh floor excreta (pooled or individual) and individual caged chicken excreta did not have significant effect on egg counts (P = 0.274). The total time taken for sample preparation and egg counting was significantly lower using the MM method (4.3-5.7 min) than the MF method (16.9-23.8 min) (P 0.0001). In conclusion, MM was more accurate than MF, particularly at higher EPG levels, but slightly less precise and sensitive, particularly at low EPG levels, while taking less than 25 % of the laboratory time per sample. Our observations indicate that the MM method is more appropriate for rapid diagnosis of chicken nematodes in the field. Pooled fresh floor excreta samples would be sufficient to indicate infection level in free range farms.

Background According to targeted treatment (TT), the whole flock is dewormed based on knowledge of the risk, or parameters that quantify the mean level of infection, whereas according to targeted selective treatment (TST), only individual animals within the grazing group are treated, based on parasitological, production and/or morbidity parameters. The aim of this study was to compare two different treatment protocols on sheep farms in Lithuania. The study was conducted from 15 April to 31 October 2014 on three sheep farms. On the TT (the whole flock) and T(S)T (with FECs ≥ 300, respectively) farms all adult animals were treated orally with fenbendazole irrespective of EPG counts before the grazing season. The second treatment was applied with injectable ivermectin on both farms. However, on the TT farm all sheep were also treated on 2nd August regardless of their EPG counts, while on the T(S)T farm only those animals with an EPG ≥ 300 were treated on 1 July using a threshold of ≥ 300 EPG. No treatments were administered on the control farm (n = 1) during the study. Results Spring treatment of ewes significantly reduced nematode faecal egg counts (FEC) both on the TT and T(S)T farms, with the benefit of lowering pasture contamination with infective L3 stage larvae at the start of grazing season, while it remained significantly higher on the control farm. The positive effect of the spring treatment of ewes was reflected by increased body weight gains (BWG) in lambs in the first half of the grazing season. Following the second treatment, the weight gains in lambs on the T(S)T farm were higher compared to lambs on the TT farm, while BWG in the control lambs started to decrease. The difference was also substantiated by the body condition scores (BCS) and dag scores (DS) of lambs, which were highest on the T(S)T farm compared with those on the control and TT farms. Conclusions The results of this study show that both treatment strategies were useful in reducing clinical effects (BCS and DS) of gastrointestinal nematode parasitism and increasing the performance in lambs. Furthermore, on the T(S)T farm some of animals were left in refugia, helping to slow down the development of anthelmintic resistance (AR) in future.

Background The anthelmintic efficacy of the 0.5% w/v topical formulation of eprinomectin (EPN), EPRINEX® Pour-on (Merial) when administered at 1 mg/kg body weight was evaluated in sheep in two dose confirmation laboratory studies and one multicenter field study. In addition, the pharmacokinetics of EPN when administered at that dosage to adult sheep was determined. Results In the two dose confirmation studies, which included 10 sheep each, sheep treated with topical EPN had significantly (p 99%: adult Dictyocaulus filaria, Haemonchus contortus, Teladorsagia circumcincta(pinnata/trifurcata), Trichostrongylus axei, T. colubriformis, T. vitrinus, Cooperia curticei, Nematodirus battus, Strongyloides papillosus, Chabertia ovina and Oesophagostomum venulosum, and inhibited fourth-stage Teladorsagia larvae. A total of 196 sheep harboring naturally acquired gastrointestinal nematode infections were included in the field efficacy study at two sites each in Germany (48 Merino x Ile de France lambs, 52 adult Merino females) and in Italy (adult male and female Bagnolese, Lacaune, Lacaune x Bagnolese, Bagnolese x Sarda sheep; 48 animals per site). Animals were blocked on pre-treatment body weight and within each block, one animal was randomly assigned to the control (untreated) group and three animals were randomly assigned to be treated with topical EPN. This study compared the phenotypic susceptibility of Pelibuey and Katahdin female lambs against gastrointestinal nematodes (GINs) under grazing conditions in the hot humid tropics of Mexico. The study was performed during the rainy season (August to October, 2014). It included 27 Pelibuey and 12 Katahdin female lambs from 6 months of age and live weight of 21.0 ± 3.7 and 23.3 ± 3.6 kg, respectively. Lambs were reared free of GIN infection before the study. The study lasted 91 days. Animals were weighed and sampled (blood and feces) on days 0 and 28 and every 7 days onwards. Fecal samples were obtained to determine fecal eggs of GIN per gram (EPG), and blood samples were used to determine the packed cell volume (PCV), the peripheral eosinophil counts (PECs), and optical densities (ODs) for IgA. The EPG counts were significantly lower for Pelibuey lambs compared to Katahdin throughout the study (P 0.001). Similarly, Pelibuey lambs had higher mean PCV (P 0.01) and PEC (P 0.05) than Katahdin lambs during the study. The total weight gain and OD for IgA were similar between breeds (P 0.05). Negative associations (P 0.05) between EPG and PCV or PEC were moderate to strong for the lambs of both breeds. No association was found between EPG and IgA. In conclusion, Pelibuey lambs showed phenotypic evidence of higher resistance to natural GIN infections compared to Katahdin lambs sharing the same grazing conditions in the hot humid tropics. The most accurate phenotypic markers to identify a difference in susceptibility were EPG and PEC.

Semi-intensive equine breeding system favors gastrointestinal nematode infections. The treatment of these infections is based on the use of anthelmintics. However, the inappropriate use of these drugs has led to parasitic resistance to the available active principles. The objective of this study was to evaluate the efficacy of the main classes of antiparasitic (ATP) used in control in adult and young animals, including: benzimidazoles (fenbendazole), pyrimidines (pyrantel pamoate), macrocyclic lactones (ivermectin and moxidectin), as well as the combination of active ingredients (ivermectin + pyrantel pamoate). The study was carried out in two military establishments, located in Rio Grande do Sul (RS), from January to December, 2018. The intervals between the treatments of the animals were performed from 30 to 90 days. Coproparasitological evaluations were determined by the egg count reduction in the faeces. Cyatostomine larvae were identified in pre and post-treatment cultures. The results demonstrated the multiple parasitic resistance of cyathostomins to fenbendazole, moxidectin in young animals, and to fenbendazole, pyrantel pamoate in adult animals. Thus, it is necessary to define or diagnose parasitic resistance to assist in the creation of prophylactic parasitic control, using suppressive treatment with ATP associated with integrated alternatives. O objetivo deste estudo foi avaliar a eficácia das principais classes de antiparasitários (ATP), utilizados no controle em animais adultos e jovens, incluindo: benzimidazois (fenbendazol), pirimidinas (pamoato de pirantel), lactonas macrocíclicas (ivermectina e moxidectina), bem como a combinação de ingredientes ativos (ivermectina + pamoato de pirantel). O estudo foi realizado em dois estabelecimentos militares, localizados no Rio Grande do Sul (RS), de janeiro a dezembro de 2018. Os intervalos entre os tratamentos foram realizados de 30 a 90 dias. As avaliações coproparasitológicas foram determinadas pela redução da contagem de ovos nas fezes. Foram identificadas larvas de ciatostomíneos nas coproculturas pré e pós tratamentos. Os resultados demonstraram a resistência parasitária múltipla dos ciatostomíneos ao fenbendazol, moxidectina em animais jovens, febendazole e pamoato de pirantel em animais adultos. Estabelecer o diagnóstico da resistência parasitária auxiliará na elaboração de um controle parasitário profilático, reduzindo o tratamento supressivo com ATP juntamente com alternativas de controle integrado. Dessa forma, o avanço da resistência parasitária poderá ser retardado.

The treatments of gastrointestinal nematodes (GIN) infection in sheep is almost exclusively based on the use of synthetic drugs. In some European regions the intensive use of antiparasitic drugs is leading to widespread development of anthelmintic resistance (AR). Currently in southern Italy AR is rare, but a constant monitoring of anthelmintic efficacy and the use of effective alternative therapies is strongly recommended. The aim of our study was to evaluate the effectiveness of a complementary natural feed (natural vegetable mixture), based on natural extracts, registered for GIN treatment in sheep, and its comparison with the drug ivermectin. The study was conducted in two sheep breeding farms in southern Italy and 75 sheep were divided in groups of 15 animals each (treated and untreated groups), homogeneous by GIN eggs per gram (EPG) of faeces, using the natural anthelmintic administered at full dose (10 g/sheep/orally) in the first breeding and at double dose (20 g/sheep/orally) in the second. In the latter we compared the effectiveness of mixture with ivermectin administered at full dose (200 &mu, g/kg/BW). To determine the effectiveness, individual faecal samples were collected to evaluate the faecal eggs count (FEC) using FLOTAC technique and FEC reduction (FECR) on different days. The formula used FECR = 100 &times, (1 &minus, (T2/C2)), based on the comparison of post-treatment EPG mean of the treated and untreated group (T2 and C2, respectively), is the one recommended by World Association for the Advancement of Veterinary Parasitology (W.A.A.V.P.) guidelines to monitor drug efficacy against GIN in livestock. The results reported that complementary natural feed, at two different dosages, was ineffective against GIN, while the drug, at conventional dosage, showed good anthelmintic efficacy, also confirming the importance of in vivo effectiveness studies.

Animals suffering from parasitism typically display altered grazing behaviour and a voluntary reduction in feed intake. These changes are potentially important as indicators of disease. Recent advances in sensor technologies provide the opportunity to objectively measure animal activity while on pasture. Tri-axial accelerometers measure body movement in terms of acceleration, which can then be used to estimate physical activity over time. This study investigated if tri-axial measures of overall activity can be used to assess the impact of gastrointestinal nematode (GIN) infection in young sheep. To address this, the overall activity, faecal nematode egg count (FEC) and body weight of two treatment groups of Romney X Suffolk ram lambs were compared. Animals were monitored for four days using tri-axial accelerometer sensors mounted on a ram mating harness after 42-days grazing on contaminated pasture. On Day 0, all lambs were given anthelmintics. Subsequently, a Suppressive Treatment Group (n = 12) was treated with anthelmintics every two weeks. An Untreated Group (n = 12) did not receive further anthelmintics. Overall activity levels were monitored from Day 42 – 46. Activity level was calculated as vectorial dynamic body acceleration (VeDBA). Anthelmintic treatment had a significant effect on FEC but there was no evidence found for a treatment effect on body weight growth over the 42-day period. An effect of treatment and lamb starting weight on overall activity was found (beta = –0.74, 95 % CI –1.17 to –0.30, p = 0.002), identifying a negative impact of parasitism on activity in heavier animals. These results highlight the usefulness of this approach in assessing the effect of GIN parasitism on sheep monitored remotely. If a threshold value of activity could be determined, it could provide a useful tool for farmers and managers that serves as an early indicator of parasitism in sheep.

Endoparasite infections can lead to considerable economic losses in dairy cattle due to decreases in milk yield and quality. Environmental and host-related factors contribute to endoparasite infection intensity and probability. Moreover, advancing urbanization influences parasite infection dynamics in livestock due to close human-animal cohabitation and changes in animal housing conditions. The aim of the present study was to investigate social-ecological effects on gastrointestinal nematode (GIN) and Eimeria spp. infections in dairy cattle along a rural-urban gradient in the emerging Indian megacity Bangalore. In this regard, 726 faecal samples from 441 dairy cattle of different ages and physiological stages were collected from 101 farms and examined at three visits between June 2017 and April 2018. Based on a survey stratification index (SSI) comprising built-up density and distance to the city center, we assigned the farms to urban, mixed and rural areas. GIN eggs were identified in the faeces of 243 cattle (33.5%; 95% confidence interval [CI]: 30.1-36.9%), and Eimeria spp. oocysts in the faeces of 151 cattle (20.8%; 95% CI: 17.9-23.7%). Co-infection rates of GIN and Eimeria spp. were 8.5 to 12.2% higher in rural compared to urban and mixed areas. The SSI effect significantly influenced Eimeria spp. infection probability and oocyst per gram of faeces (OpG; P 0.001) with an infection probability and OpG higher than 26% and 40% for cattle kept in rural areas compared to cattle from urban areas. However, the SSI effect was not significant for the infection probability of GIN and for GIN eggs per gram of faeces (EpG). Infection probabilities and EpG/OpG were significantly higher in calves and heifers compared to lactating and dry cows. Moreover, we estimated significantly lower OpG values in summer compared to the other seasons. No differences were estimated for GIN and Eimeria spp. infection probabilities and EpG/OpG with regard to pasture access and breed. The variations in endoparasite infection intensity and probability observed along the rural-urban gradient of Bangalore reflect the variability in dairy husbandry systems governed by the social-ecological context.

Gastrointestinal nematode infection is an important cause of high economic losses in livestock production. Nematode control based on a synthetic chemical approach is considered unsustainable due to the increasing incidence of anthelmintic resistance. Control alternatives such as the use of natural products are therefore becoming relevant from an environmental and economic point of view. Proteins are macromolecules with various properties that can be obtained from a wide range of organisms, including plants and fungi. Proteins belonging to different classes have shown great potential for the control of nematodes. The action of proteins can occur at specific stages of the nematode life cycle, depending on the composition of the external layers of the nematode body and the active site of the protein. Advances in biotechnology have resulted in the emergence of numerous protein and peptide therapeutics; however, few have been discussed with a focus on the control of animal nematodes. Here, we discuss the use of exogenous proteins and peptides in the control of gastrointestinal. O controle de nematoides com compostos químicos sintéticos é considerado insustentável devido ao aumento da resistência anti-helmíntica. Alternativas de controle, como o uso de produtos naturais, estão se tornando relevantes do ponto de vista ambiental e econômico. As proteínas são macromoléculas com várias propriedades que podem ser obtidas de uma ampla gama de organismos, incluindo plantas e fungos. Proteínas pertencentes a diferentes classes têm mostrado grande potencial para o controle de nematoides. A ação das proteínas pode ocorrer em estágios específicos do ciclo de vida do nematoide, dependendo da composição das camadas externas do parasito e do sítio ativo da proteína. Avanços na biotecnologia resultaram no surgimento de numerosas terapias de proteínas e peptídeos; no entanto, pouco foi discutido com foco no controle de nematoides parasitos de animais. Na presente revisão foi discutido o uso de proteínas exógenas e peptídeos no controle de nematoides gastrintestinais, os mecanismos sugeridos de ação, e os desafios e perspectivas para o uso dessas biomoléculas como uma classe de anti-helmínticos.

International audience; Foraging behavior by parasitized herbivores can be interpreted as a decision-making process where individuals are faced with trading-off the ingestion of nutrients with the ingestion of potentially medicinal -and toxic- plant secondary compounds. We determined how parasitized sheep prioritize selection of crude protein, energy and a medicinal plant secondary compound (quebracho tannins-QT). Foraging preferences were tested in 40 lambs before experiencing a parasitic infection (Phase 1), during an infection (Phase 2; 10,000 L3 Haemonchus contortus per lamb) and after chemotherapy (Phase 3). Lambs were assigned to four groups (10 lambs/group) such that animals in Group 1 (Control) could choose between foods of high (HEP) or low (LEP) energy to protein ratios. The other groups received the same choice, but QT were added (4%) to HEP (Group 2), to LEP (Group 3) or to both foods (Group 4). All groups under a parasitic infection (Phase 2) increased their preference for HEP (from 0.44 to 0.66 ± 0.042; P 0.05). The importance of protein nutrition on parasitized animals has been highlighted before, but these results suggest that lambs prioritized the ingestion of energy-dense over protein-dense foods or medicinal condensed tannins when challenged by gastrointestinal parasitism. Consumption of medicinal tannins represented a side-effect of the preference manifested for energy-dense foods during testing.

Gastrointestinal nematode (GIN) infections are a common constraint in pasture-based herds and cause a decrease in animal health, productivity and farm profitability. Current control practices to prevent production losses of GIN infections in livestock depend largely on the use of anthelmintic drugs. However, due to the continued use of these drugs over more than three decades, the industry is now increasingly confronted with nematode populations resistant to the available anthelmintics. This emerging anthelmintic resistance (AR) in cattle nematodes emphasizes the need for a change toward more sustainable control approaches that limit, prevent or reverse the development of AR. The uptake of diagnostic methods for sustainable control could enable more informed treatment decisions and reduce excessive anthelmintic use. Different diagnostic and targeted or targeted selective anthelmintic control approaches that slow down the selection pressure for anthelmintic resistance have been developed and evaluated recently. Now it is time to transform these insights into guidelines for sustainable control and communicate them across the farmer community. This article reviews the current uptake of such sustainable practices with a focus on farmer's socio-psychological factors affecting this uptake. We investigate communication as a possible tool to change current behavior and successfully implement more sustainable anthelmintic treatment strategies.

The anthelminthic efficacy of the crude extracts of Hypoestes forskaolii (Vahl) R.Br (Acanthaceae) against gastrointestinal nematodes (GIN) in sheep was investigated using the in vitro egg hatch inhibition assay. Faecal samples were collected from sheep with naturally occurring infection of GIN (Trichostrongylus spp., Chabertia ovina, Cooperia spp., Haemonchus contortus and Teladorsagia spp.). Crude leaf extracts of H. forskaolii was obtained using increasing polarity solvents: n-hexane, chloroform, chloroform:methanol 9:1, methanol. Thiabendazole (0.2 &micro, g/mL and 0.5 &micro, g/mL) was used as a positive control and untreated GIN eggs in deionised water served as the negative control. All the extracts exhibited a weak ovicidal activity against GIN (less than 50% of egg hatch). Noteworthy, the n-hexane extract showed a percentage of inhibition of egg hatching greater than other extracts inhibiting the 30.8% at the concentration of 1 mg/mL showing a dose-dependent effect on nematode eggs hatching. Further studies are needed to investigate the effects of extracts used and to evaluate the ovicidal effects of other extracts of H. forskaolii.

Gastrointestinal nematode parasites are important pathogens of all domesticated livestock species. Fecal egg counts (FEC) are routinely used for evaluating anthelmintic efficacy and for making targeted anthelmintic treatment decisions. Numerous FEC techniques exist and vary in precision and accuracy. These performance characteristics are especially important when performing fecal egg count reduction tests (FECRT). The objective of this study was to compare the accuracy and precision of three commonly used FEC methods and determine if differences existed among livestock species. In this study, we evaluated the modified-Wisconsin, 3-chamber (high-sensitivity) McMaster, and Mini-FLOTAC methods in cattle, sheep, horses, and llamas in three phases. In the first phase, we performed an egg-spiking study to assess the egg recovery rate and accuracy of the different FEC methods. In the second phase, we examined clinical samples from four different livestock species and completed multiple replicate FEC using each method. In the last phase, we assessed the cheesecloth straining step as a potential source of egg loss. In the egg-spiking study, the Mini-FLOTAC recovered 70.9% of the eggs, which was significantly higher than either the McMaster (P = 0.002) or Wisconsin (P = 0.002). In the clinical samples from ruminants, Mini-FLOTAC consistently yielded the highest EPG, revealing a significantly higher level of egg recovery (P 0.0001). For horses and llamas, both McMaster and Mini-FLOTAC yielded significantly higher EPG than Wisconsin (P 0.0001, P 0.0001, P 0.001, and P = 0.024). Mini-FLOTAC was the most accurate method and was the most precise test for both species of ruminants. The Wisconsin method was the most precise for horses and McMaster was more precise for llama samples. We compared the Wisconsin and Mini-FLOTAC methods using a modified technique where both methods were performed using either the Mini-FLOTAC sieve or cheesecloth. The differences in the estimated mean EPG on log scale between the Wisconsin and mini-FLOTAC methods when cheesecloth was used (P 0.0001) and when cheesecloth was excluded (P 0.0001) were significant, providing strong evidence that the straining step is an important source of error. The high accuracy and precision demonstrated in this study for the Mini-FLOTAC, suggest that this method can be recommended for routine use in all host species. The benefits of Mini-FLOTAC will be especially relevant when high accuracy is important, such as when performing FECRT.

International audience; Gastrointestinal (GI) nematode control has an important role to play in increasing livestock production from a limited natural resource base and to improve animal health and welfare. In this synthetic review, we identify key research priorities for GI nematode control in farmed ruminants and pigs, to support the development of roadmaps and strategic research agendas by governments, industry and policymakers. These priorities were derived from the DISCONTOOLS gap analysis for nematodes and follow‐up discussions within the recently formed Livestock Helminth Research Alliance (LiHRA). In the face of ongoing spread of anthelmintic resistance (AR), we are increasingly faced with a failure of existing control methods against GI nematodes. Effective vaccines against GI nematodes are generally not available, and anthelmintic treatment will therefore remain a cornerstone for their effective control. At the same time, consumers and producers are increasingly concerned with environmental issues associated with chemical parasite control. To address current challenges in GI nematode control, it is crucial to deepen our insights into diverse aspects of epidemiology, AR, host immune mechanisms and the socio‐psychological aspects of nematode control. This will enhance the development, and subsequent uptake, of the new diagnostics, vaccines, pharma‐/nutraceuticals, control methods and decision support tools required to respond to the spread of AR and the shifting epidemiology of GI nematodes in response to climatic, land‐use and farm husbandry changes. More emphasis needs to be placed on the upfront evaluation of the economic value of these innovations as well as the socio‐psychological aspects to prioritize research and facilitate uptake of innovations in practice. Finally, targeted regulatory guidance is needed to create an innovation‐supportive environment for industries and to accelerate the access to market of new control tools

Background The gastrointestinal parasitic nematode Haemonchus contortus is a pathogenic organism resistant to several anthelmintics. This study assessed the efficacy of a medicinal herbal mixture (Herbmix) and organic zinc, as an essential trace element for the proper functioning of both unspecific and specific immune defensive mechanisms, against experimental infections with H. contortus in lambs. All lambs were infected orally with approximately 5000 third-stage larvae of a strain of H. contortus susceptible to anthelmintics (MHco1). Twenty-four female lambs 3–4 months of age were divided into four groups: unsupplemented animals (control), animals supplemented with Herbmix (Hmix), animals supplemented with organic zinc (Zn) and animals supplemented with Herbmix and organic zinc (Hmix+Zn). Eggs per gram (EPG) of faeces were quantified 20, 28, 35, 42, 49, 56, 62 and 70 d post-infection and mean abomasal worm counts were assessed 70 d post-infection. Samples of blood were collected from each animal 7, 35, 49 and 70 d post-infection. Results Quantitative analyses of the bioactive compounds in Herbmix identified three main groups: flavonoids (9964.7 μg/g), diterpenes (4886.1 μg/g) and phenolic acids (3549.2 μg/g). Haemonchus (H.) contortus is a blood feeding gastrointestinal nematode, and is considered one of the major threats for goat health and production globally. Crude somatic antigens (CSA) have been reported as sources of immunogens and provide better level of immunity against gastrointestinal nematodes. The present study aimed to quantify CSA of H. contortus worms (n=25) of both sexes followed by isolation of peptides through Sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE) and their characterization. The triturate contained proteins having concentration of 1.3mg/ml as measured through Bradford assay. Seven different bands were appeared on gel ranging from 35KDa to 170KDa. The future prospects may include identification of the immunogenic peptides which can be used as vaccine candidates against H. contortus.

Background Gastrointestinal nematode (GIN) infections are the main impediments that restrict the welfare and productivity of small ruminant in the world. Effective management of GINs in grazing sheep relies heavily on the use of highly efficacious anthelmintic drugs. However, anthelmintic resistance is becoming a significant concern in the world, and this phenomenon severely threatens the potential utilisation of this control strategy. Therefore, this study was conducted 1) to evaluate the efficacy of commonly used anthelmintic on GINs in naturally infected sheep and 2) to assess the farmers’ perception on anthelmintics utilisation practices in Dabat district, Northwest Ethiopia. Methods One hundred twenty nematode infected sheep were used in this study. Sheep were selected based on the egg count (≥150 eggs per gram of faeces). The animals were allocated randomly into four groups (30 animals per group). Group-I, II and III were treated with Albendazole, Tetramisole, and Ivermectin, respectively. The 4th group was left untreated (as control). Faecal samples were collected on day 0 (before treatment), on day 3, 7, 10 and 14 (post-treatment). The modified McMaster technique was used for quantifying the eggs. Faecal egg count reduction test (FECRT) was applied to determine the efficacy of anthelmintic at day 14 (post-treatment). In addition, a questionnaire survey was conducted on 100 randomly selected sheep owners. Results All anthelmintics tested revealed significant (P 0.05) among the treatment groups. The nematode genera identified before treatment were Haemonchus, Trichostrongylus, Cooperia, Trichuris, Teladorsagia, Bunostomum, and Strongyloides. Haemonchus and Trichostrongylus were detected after treatment with Albendazole and Ivermectin. The questionnaire survey revealed that Albendazole was the most commonly (90%) used anthelmintic to treat nematodes in sheep, followed by Tetramisole (36%) and Tetraclozan (Tetramisole-Oxyclozanide combination) (20%). Respondents expressed that anthelmintic selection was made based on veterinarian prescription (84%), colour (27%), efficacy (4%), price affordability (1%) and availability (1%). Conclusion This study demonstrated that the tested anthelmintics had an acceptable level of efficacy against GINs of sheep.

Their close morphological similarity, common occurrence as co-infections and ability to hybridize makes definitive diagnosis and epidemiological studies in field populations challenging. In this paper, we describe the development of a panel of microsatellite markers that can be used to discriminate and study the genetics of these two parasite species in co-infections and mixed field populations. We have identified two additional microsatellites (Hp52 and Hp53), in addition to three previously reported microsatellites (Hcms3561, Hcms53265 and Hcms36) that have a discrete set of alleles between the two species. Multilocus genotyping of worms with this 5 marker panel from 3 geographically diverse H. placei isolates and 4 geographically diverse H. contortus populations allows unambiguous species assignment of individual worms. This panel of markers should provide a valuable resource in studying the biology and epidemiology of these important ruminant parasite species in the field. The aim of this study was to compare two fecal egg count (FEC) techniques; McMaster (McM) and Mini-FLOTAC (mF), for the detection of cattle and horse gastrointestinal nematode eggs, in different locations. Experiment 1: feces were collected from 16 cattle and FEC was performed individually, using mF with the sensitivity of 5 eggs per gram of feces (EPG) and McM with a sensitivity of 50 EPG at Empresa de Pesquisa Agropecuaria de Minas Gerais - EPAMIG and the Laboratory of Parasitic Diseases of the University of Parana - LDP/UFPR. Experiment 2: Fecal samples from 30 horses were analyzed with mF (sensitivity of 5 EPG) and McM (sensitivity of 25 EPG) at the University of Mato Grosso do Sul - UFMS and LPD/UFPR. Experiment 3: feces were collected from 14 foals and FEC was performed using mF (sensitivity of 5 EPG); and McM (sensitivity of 25 EPG) only at the LPD/UFPR. For cattle, the average FEC of mF was 962 at LPD; and 1248 at EPAMIG; for McM it was 1393 at LPD and 1563 at EPAMIG. For horses, the FEC average using the mF was 650 at LPD and 469 at UFMS; and for McM it was 677 at LPD and 554 at UFMS. For foals, the average FEC for mF was 404 and 436 for McM. In all experiments, the standard deviation and the coefficient of variation values were significantly lower for mF. Therefore, it is recommended the use of the Mini-FLOTAC technique, which is a method with less variability and higher accuracy, particularly for animals with low FEC.

Control of sheep gastrointestinal nematode (GIN) infections mostly relies on the use of anthelmintics. Refugia-based control strategies as targeted selective treatments (TST) can delay anthelmintic resistance development, but the optimal decision criteria for selecting individuals to be treated in subclinical infection scenarios remain unclear. The present study aimed to evaluate the suitability of body condition score (BCS) and faecal egg counts (FEC) as treatment indicators and to determine their optimized threshold values for treatment in TST by determining the relationships of BCS, FEC and anthelmintic treatment with several productive parameters in pre-mating and pre-partum periods at an individual level. Deworming in pre-mating period increased BCS gain, but its magnitude was directly associated with strongyle FEC before treatment. Deworming also increased fertility in ewes with BCS < 2.75 and the proportion of lambing ewes that got pregnant during the first ovulation cycle. Nevertheless, treatment did not yield productive benefits in ewes with higher BCS values. On the other hand, deworming in the pre-partum lowered lamb peri-partum mortality and increased lamb weight at birth and the growth of lambs during lactation, especially in lambs born from ewes with strongyle FEC > 400 epg before treatment. These results showed that FEC and especially BCS can be potential decision criteria for the implementation of TST in these types of scenarios. The TST scheme derived from the present results on the control of GIN infections should include anthelmintic treatment of ewes with BCS lower than ˜ 3 approximately five weeks before mating and lambing. In the pre-lambing period, treatment may be unnecessary if flock mean strongyle FEC is lower than ˜ 200 epg, as the proportion of ewes with individual FEC > 400 epg would be very low. The results suggest that implementation of this TST scheme would provide benefits, such as the improvement of productivity, a rational management of parasites in refugia, and preservation of future efficacy of anthelmintics, in comparison to traditional deworming schemes.

The difference between castrated male lambs and entire female lambs in susceptibility to gastrointestinal nematode infection was investigated in a flock of Scottish Blackface sheep. Castrated male lambs had higher faecal egg counts, higher body weights, lower parasite-specific plasma IgA activity and more adult nematodes. There were no detectable differences in the length of adult Teladorsagia circumcincta. Heavier egg shedding by males was associated with greater worm burdens. The difference in plasma IgA activity may have been a consequence of larger numbers of adult nematodes inhibiting the transfer of mucosal IgA to the plasma. At least part of the sex difference in egg production could be a consequence of males ingesting larger amounts of grass and hence numbers of infective larvae because of their larger size. The differences between castrated males and females contribute to the observed variation in nematode egg production in naturally infected lambs.

Monthly canine parasite prophylactic products prevent not only adult heartworm infection, but also patent infections with specific gastrointestinal parasites. While most monthly products control and treat certain hookworm and roundworm infections, fewer are labeled for whipworm (Trichuris vulpis). Therefore, we hypothesized that fecal samples collected from municipal dog parks will have a greater prevalence of whipworm eggs compared to hookworm and roundworm eggs. In this study, canine fecal samples were collected from municipal dog parks in three southeastern states, with up to 20 fecal samples were collected from each park. A total of 200 fecal samples were obtained from dog parks in Georgia, North Carolina, and South Carolina. All fecal samples were examined for the presence of gastrointestinal helminths by a simple centrifugal flotation using sheather's sugar flotation solution. Of the 200 samples collected, 27% were positive for gastrointestinal helminths by fecal flotation. Of these infected fecal samples, 8.5%, 17%, and 1.5% contained whipworm, hookworm, and roundworm, respectively. However, the majority of hookworm-positive samples were collected from one park, whereas whipworm and roundworm samples were collected from multiple parks. These results could indicate that dogs are at risk of infection by all three parasites at dog parks, and that preventive strategies may need to be tailored not only to the specific region, but to specific infected dog parks.

Gastrointestinal nematodes (GINs) have been identified in Australia as a major problem in goat production, with few anthelmintics registered for use in goats. Therefore, anecdotally many producers use anthelmintics that have not been registered for goats. Using unregistered products could increase selection pressure for anthelmintic resistance as well as safety and/or meat or milk chemical residues of products from treated goats. This producer survey was conducted in 2014 to establish Australian goat producer knowledge, perception and practises of GIN treatment and control. Eighty-eight producers responded to the survey. Of these respondents, 90% thought that GINs were a problem for the Australian goat industry, and 73% considered GINs had caused production losses or health impacts for their goats during the 5 years prior to the survey. With regard to anthelmintic resistance, 7% believed that anthelmintic resistance was not a problem at all, 93% acknowledged anthelmintic resistance was a problem in Australian goats herds, with 25% of these reporting their properties as being affected. The majority (81%) of respondents believed the number of anthelmintics registered for goats was inadequate for effective GIN control. Of the 85% of producers who used an anthelmintic during the survey period, 69% had used a treatment not registered for use in goats. Fifty respondents listed the anthelmintic dosage used, and 50% of those had used a dose rate greater than the recommended label dose. The average frequency of administration of anthelmintic was 2.5 times per annum. Of the 51% of respondents who listed the frequency of their treatments given during the survey period, 16% administered four or more treatments annually to the majority of their goats and 8% administered treatments on an “as needed” basis. Faecal egg count (FEC) had been performed on 72% of properties in at least one of the six years covered by the survey. These results indicated that the majority of surveyed producers use anthelmintics that are not registered for use in goats and at different dose rates to label. These practises have the potential for increasing the spread of anthelmintic resistance in the GIN populations of goats and sheep. Further, giving dose rates in excess of label recommendations could impact goat safety and/or product residues. Further research is needed to investigate these risks and evaluate more sustainable GIN control options for goat herds. In addition more effective dissemination of information is necessary for the improvement of the Australian goat industry.

Fecal egg count reduction tests (FECRT) and larval migration inhibition tests (LMIT) were conducted to assess the efficacy of ivermectin (IVM) against gastrointestinal nematodes on 2 cattle farms in northern Japan in 2009 and 2010. Twelve to 20 calves on each farm were treated topically with 0.5 mg IVM/kg 2 (Farm 2) or 4 times (Farm 1) during the grazing season (May–October). On Farm 1, fecal egg count (FEC) reduction at 14 days post-treatment ranged from 16 to 87% in 2009 and from 24 to 96% in 2010, with relatively low reductions in August and October (16–53%). Conversely, IVM treatment on Farm 2 reduced FEC by 97% in September 2009. Larvae obtained from fecal cultures and identified by PCR-RFLP analysis revealed that the dominant species on both farms prior to IVM administration was Cooperia oncophora. In 2009, the FEC reduction of C. oncophora on Farm 1 decreased from 85% in May to 56% in August. In 2010, the reduction in C. oncophora in August was 28%. In the LMIT using larvae collected from the fecal cultures on Farm 1 in May and August 2009, the EC50 value of IVM in C. oncophora in August (0.892 µg/ml) was 3 times higher than that in May (0.296 µg/ml). The results of the LMIT corroborated the FECRT data, indicating the presence of IVM-resistant C. oncophora on Farm 1, at least in August. This is the first report of IVM-resistant nematodes in Japanese cattle.

Gastrointestinal nematode (GIN) parasite control recommendations are in a state of flux because of the increase in anthelmintic resistant cattle parasites, such as Cooperia spp. In addition, Cooperia spp. infection is typically high in warm-season grass pastures and can affect growth performance of grazing stocker calves in the Gulf Coast Region. This study evaluated the effects of moxidectin pour-on, oxfendazole oral suspension, or a combination of the two given at separate times on infection and performance of weaned beef calves grazing summer forages. Steers (n=42) and heifers (n=31) were stratified by sex, d-11 fecal egg count (FEC), and d-1 shrunk body weight (BW) to one of 10 pastures with four anthelmintic treatments and one control. Treatments included: (1) oxfendazole given on d 0 and moxidectin on d 73 (O+M), (2) moxidectin given on d 0 and oxfendazole on d 73 (M+O), (3) moxidectin given on d 0 (M), (4) oxfendazole given on d 0 (O) and (5) no anthelmintic given (CON). International audience; Background: Gastrointestinal nematode (GIN) remains the most important pathogenic constraint of small ruminant production worldwide. The improvement of the host immune response against GIN though breeding for improved animal resistance, vaccination and nutritional supplementation appear as very promising methods. The objective of this study was to investigate the effect of four nutritional status differing in protein and energy levels (Hay: 5.1 MJ/Kg of dry matter (DM) and 7.6% of crude protein (CP), Ban: 8.3 MJ/Kg of DM and 7.5% of CP, Soy: 7.6 MJ/Kg of DM and 17.3% of CP, BS: 12.7 MJ/Kg of DM and 7.4% of CP) on the haematological disturbances due to Haemonchus contortus infection in Creole kid goats. Results: No significant effect of the nutritional status was observed for faecal egg count (FEC) but the experimental infection induced haematological disturbances whose intensity and lengthening were dependent on the nutritional status. A transient marked regenerative macrocytic hypochromic anaemia as revealed by a decrease of packed cell volume (PCV), red blood cells (RBC) and hemoglobin and an increase of reticulocytes was observed in all infected groups except Hay. In this latter, the anaemia settled until the end of the experiment. Furthermore, H. contortus induced a thrombocytopenia significantly more pronounced in the group under the lowest nutritional status in term of protein (Hay and Ban). A principal component analysis revealed that the variables that discriminated the nutritional status were the average daily gain (ADG) and the PCV, considered as measures of the level of resilience to H. contortus infection. Moreover, the variables that discriminated infected and non-infected animals were mostly related to the biology of RBC (i.e. size and hemoglobin content) and they were correlated with FEC. Conclusions: The severity and the lengthening of the regenerative anaemia and the thrombocytopenia induced by H. contortus have been affected by the nutritional status. The protein enriched diets induced resilience to the infection rather than resistance. This suggests that resilience is associated with an improved regenerative capacity of the bone marrow. However, this needs to be further investigated to understand the relationships between resistance, resilience and dietary supplementation.

El objetivo del estudio fue determinar la prevalencia de nematodos gastrointestinales en sistemas de producción de ganado ovino y caprino en el trópico de Antioquia, Colombia, mediante un estudio descriptivo transversal. Se incluyeron 17 apriscos de 9 municipios y 302 animales (ovejas y cabras). Se evaluó la frecuencia de parasitismo gastrointestinal y carga parasitaria de especies y géneros determinados a través de la caracterización taxonómica por identificación de las larvas (L3), considerando variables clínico-epidemiológicas. El 76% de los animales se encontraba infectado, donde el 69.5% presentó cargas parasitarias bajas (menos de 200 hpg de heces). Se concluyó que los apriscos de Antioquia presentan alta prevalencia de infección por Tricostrongilidos, siendo Haemonchus contortus (61.3%), Teladorsagia (Ostertagia) circumcincta (25.5%) y Trichostrongylus sp (21.5%) los parásitos más frecuentes., The aim of this study was to determine the prevalence of gastrointestinal nematodes in sheep and goat production systems in the tropics of Antioquia, Colombia through a cross-sectional study. Faecal samples were collected from 302 sheep and goats from 17 farms in 9 municipalities. The prevalence of nematode infection and the parasite burden based on species and genus identified through taxonomic characterization of infective third-stage larvae (L3) and considering clinical and epidemiological variables were evaluated. The prevalence of infected ruminants was 76%, where 69.5% showed low parasite burden (less than 200 epg of faeces). It is concluded that ovine and caprine farms in Antioquia had high prevalence of Trichostrongylids infection, where Haemonchus contortus (61.3%), Teladorsagia (Ostertagia) circumcincta (25.5%) and Trichostrongylus sp (21.5%) were the most common parasites.

A quantificação de ovos de nematoides em amostras de fezes e a identificação morfológica das larvas de terceiro estágio a partir de culturas de fezes têm sido amplamente utilizadas, com resultados satisfatórios para o diagnóstico das infecções por nematoides gastrintestinais em ruminantes. Com o intuito de melhorar a sensibilidade na detecção dos ovos, outras técnicas, tais como Flotac e Miniflotac, têm sido empregadas. Resultados obtidos em diferentes estudos indicam que a contagem de ovos nas fezes é uma medida confiável e representativa da magnitude da carga parasitária. No entanto, o estado imunológico dos animais deve ser considerado na interpretação dos resultados coproparasitológicos. As técnicas moleculares também têm sido úteis no diagnóstico das enfermidades parasitárias. Os maiores avanços no diagnóstico têm sido propiciados pelo desenvolvimento de plataformas robóticas, que separam os ovos das fezes com pouca manipulação, propiciando a obtenção de DNA de ovos de boa qualidade, o que resulta em uma amplificação que possibilita indicar o grau da infecção pelas diferentes espécies de parasitas em infecções mistas. O método ideal deve ser preciso, rápido e de fácil execução por não-especialistas. Com o avanço da robótica, bioinformática e biologia molecular, espera-se que equipamentos com tais características se tornem disponíveis e acessíveis para serem utilizados na rotina laboratorial para o diagnóstico de nematoides gastrintestinais de ruminantes.

Maternally derived antibodies can provide partial protection against certain bacterial and viral infections. We investigated whether chicks descending from nematode-infected hens are more resistant against Ascaridia galli, a prevalent gastrointestinal nematode, than chicks from nematode-free mothers. One-day-old chicks (N=153) from infected (mab+; maternal antibody+) or uninfected control dams (mab-) were experimentally infected with A. galli at two different levels (100 or 1000 eggs/chick). The worm burdens of the chicks were determined at 6 weeks post infection. There was a high correlation (r=0.89) between A. galli-specific antibody concentrations in dam plasma and egg yolk. There was no difference between worm burdens of chicks descending from infected or uninfected dams (P=0.892), indicating no maternally derived protection against A. galli. Chicks receiving the higher infection dose had higher worm burdens (P0.05). Although there was no difference (P0.05) between worm counts of female and male chicks infected with 100 eggs, females chicks infected with 100 eggs harboured longer and heavier female worms. We conclude that there is no protective maternal immunity against A. galli infection.

Diarrhoea is a major impediment to profitable sheep production in many countries as it predisposes animals to blowfly strike and contaminates wool and meat carcasses. While it is accepted that nematode parasites are a major cause of diarrhoea in grazing animals, less is known about what facets of the host-parasite relationship lead to diarrhoea and what the most appropriate control strategies are. In this review, the relationship between gastrointestinal nematode infection and diarrhoea is discussed and it is concluded that in many cases, particularly in immunologically mature sheep, diarrhoea is not due to parasite infection per se but rather due to immunopathological processes. Mechanisms that lead to faecal softening in immune sheep are considered, and the question addressed as to whether anthelmintic treatment and selective breeding of naturally parasite-resistant sheep will effectively reduce the occurrence of diarrhoea.

The increasing cost of production loss caused by gastrointestinal nematodes (GIN) highlights the importance of good control programs. However, the endemic nature of anthelmintic resistance also reminds of the need for nonchemical options. Both chemical and nonchemical control options need to be integrated into regional parasite management programs (IPM) with the emphasis on component options determined by factors such as major GIN species, regional climate, property size and enterprise structure. The Integrated Parasite Management of Sheep project was established to develop and demonstrate regional parasite control programs, that integrated chemical and nonchemical options, for the main sheep-producing regions of Australia. The project included research about the ecology of the main endo and ecto-parasites of sheep and a national survey of parasite control practices by sheep producers. IPM approaches developed for two contrasting regions of Australia are discussed. Barriers for the adoption of IPM programs include perceived complexity associated with a multi-component approach, time requirements and difficulty. Facilitating the industry adoption of IPM programs is discussed with relevance to the use of small group extension and involvement of the commercial sector. Perceptions of complexity of IPM may be managed by facilitating adoption of components in a step-wise process such that learning outcomes accumulate over time. Extension efforts must address the needs of industry sectors other than sheep producers and explore user pay approaches. The success of these approaches will depend on the relation of the extra profit to producers, through adoption of IPM programs, with remuneration sufficient to attract a commercial service.
