Key message: Knowledge of genetic variation, genetics, physiology/molecular basis and breeding (including biotechnological approaches) for biofortification and bioavailability for Zn, Fe and Se will help in developing nutritionally improved wheat. Biofortification of wheat cultivars for micronutrients is a priority research area for wheat geneticists and breeders. It is known that during breeding of wheat cultivars for productivity and quality, a loss of grain micronutrient contents occurred, leading to decline in nutritional quality of wheat grain. Keeping this in view, major efforts have been made during the last two decades for achieving biofortification and bioavailability of wheat grain for micronutrients including Zn, Fe and Se. The studies conducted so far included evaluation of gene pools for contents of not only grain micronutrients as above, but also for phytic acid (PA) or phytate and phytase, so that, while breeding for the micronutrients, bioavailability is also improved. For this purpose, QTL interval mapping and GWAS were carried out to identify QTLs/genes and associated markers that were subsequently used for marker-assisted selection (MAS) during breeding for biofortification. Studies have also been conducted to understand the physiology and molecular basis of biofortification, which also allowed identification of genes for uptake, transport and storage of micronutrients. Transgenics using transgenes have also been produced. The breeding efforts led to the development of at least a dozen cultivars with improved contents of grain micronutrients, although land area occupied by these biofortified cultivars is still marginal. In this review, the available information on different aspects of biofortification and bioavailability of micronutrients including Zn, Fe and Se in wheat has been reviewed for the benefit of those, who plan to start work or already conducting research in this area. Key message: A review of the available literature on genetics of yield and its component traits, tolerance to abiotic stresses and biofortification should prove useful for future research in wheat in the genomics era. The work reviewed in this article mainly covers the available information on genetics of some important quantitative traits including yield and its components, tolerance to abiotic stresses (heat, drought, salinity and pre-harvest sprouting = PHS) and biofortification (Fe/Zn and phytate contents with HarvestPlus Program) in wheat. Major emphasis is laid on the recent literature on QTL interval mapping and genome-wide association studies, giving lists of known QTL and marker-trait associations. Candidate genes for different traits and the cloned and characterized genes for yield traits along with the molecular mechanism are also described. For each trait, an account of the present status of marker-assisted selection has also been included. A family of putative PECTIN ESTERASE INHIBITOR (PEI) genes, which were detected in the genomic region co-segregating with the resistance gene Rrs2 against scald caused by Rhynchosporium commune in barley, were characterized and tested for their possible involvement in mediating resistance to the pathogen by complementation and overexpression analysis. The sequences of the respective genes were derived from two BAC contigs originating from the susceptible cultivar ‘Morex’. For the genes HvPEI2, HvPEI3, HvPEI4 and HvPEI6, specific haplotypes for 18 resistant and 23 susceptible cultivars were detected after PCR-amplification and haplotype-specific CAPS-markers were developed. None of the tested candidate genes HvPEI2, HvPEI3 and HvPEI4 alone conferred a high resistance level in transgenic over-expression plants, though an improvement of the resistance level was observed especially with OE-lines for gene HvPEI4. These results do not confirm but also do not exclude an involvement of the PEI gene family in the response to the pathogen. A candidate for the resistance gene Rrs2 could not be identified yet. It is possible that Rrs2 is a PEI gene or another type of gene which has not been detected in the susceptible cultivar ‘Morex’ or the full resistance reaction requires the presence of several PEI genes. Advances in genome sequencing technologies have enabled researchers and breeders to rapidly associate phenotypic variation to genome sequence differences. We recently took advantage of next-generation sequencing technology to develop MutMap, a method that allows rapid identification of causal nucleotide changes of rice mutants by whole genome resequencing of pooled DNA of mutant F2 progeny derived from crosses made between candidate mutants and the parental line. Here we describe MutMap+, a versatile extension of MutMap, that identifies causal mutations by comparing SNP frequencies of bulked DNA of mutant and wild-type progeny of M3 generation derived from selfing of an M2 heterozygous individual. Notably, MutMap+ does not necessitate artificial crossing between mutants and the wild-type parental line. This method is therefore suitable for identifying mutations that cause early development lethality, sterility, or generally hamper crossing. Furthermore, MutMap+ is potentially useful for gene isolation in crops that are recalcitrant to artificial crosses. [ABSTRACT FROM AUTHOR]

Morpho-agronomic traits and microsatellite markers were used to survey genetic diversity in 115 common bean genotypes that included 70 Indian landraces, 24 released varieties and 21 exotic accessions. Twelve morpho-agronomic traits, namely, days to 50% flowering, leaflet length, leaflet width, pod length, pod width, number of pods per plant, days to maturity, seed length, seed width, number of seeds per pod, 100 seed weight and seed yield per plant were studied. Field data of two consecutive years were subjected to multivariate analysis as proposed by Mahalanobis’s D2-statistics, Tochers method of clustering and combined analysis of variance. Seventeen microsatellite markers were also used to examine genetic diversity at molecular level that showed polymorphic information content (PIC) in the range of 0.00–0.684. Dendrograms based on Euclidean distances and UPGMA analysis showed the presence of majority of released varieties into single cluster, which pointed toward their low genetic base in comparison to indigenous landraces and exotic germplasm. Significant correlation existed between morphological genetic distance and microsatellite genetic distance tested by Mantel test ( r = 0.876). [ABSTRACT FROM AUTHOR]

In eukaryotic genomes, ribosomal DNA is organized in tandem repeat units at the nucleolar organizing regions (NORs) on each satellited chromosome. Each repeat unit consists of a coding region and an intergenic spacer (IGS), the latter generally varying in length at different loci of the same genotype and also at the same locus in different genotypes. The spacer length variants (slvs) are sometimes correlated with climatic and ecological variables. In a study of 42 accessions of wild barley (Hordeum spontaneum) collected from four different microniches (sun-deep soil, sun-shallow soil, shade-deep soil and shade-shallow soil) at the Newe Ya’ar microsite (3182 m2), in Israel, we observed eight slvs constituting 10 different rDNA phenotypes, correlated with edaphic conditions prevalent at the four microniches. We conclude that rDNA diversity in wild barley is distributed nonrandomly and adaptively in the four microniches of Newe Ya’ar. A novel feature in this study is also the homogenization of IGS length at the two loci located at two nonhomologous chromosomes.
