As pure antipodes may differ in biological interactions, pharmacology, and toxicity, discrimination of enantiomers is important in the pharmaceutical and agrochemical industries. Two major challenges in enantiomer determination are transducing and amplifying the distinct chiral-recognition signals. In this study, a light-sensitive organic photoelectrochemical transistor (OPECT) with homochiral character is developed for enantiomer discrimination. Demonstrated with the discrimination of glucose enantiomers, the photoelectrochemically active gate electrode is prepared by integrating Au nanoparticles (AuNPs) and a chiral Cu(II)-metal-organic framework (c-CuMOF) onto TiO 2 nanotube arrays (TNT). The captured glucose enantiomers are oxidized to hydrogen peroxide (H 2 O 2 ) by the oxidase-mimicking AuNPs-loaded c-CuMOF. Based on the confinement effect of the mesopocket structure of the c-CuMOF and the remarkable charge transfer ability of the 1D nanotubular architecture, variations in H 2 O 2 yield are translated into significant changes in OPECT drain currents (I D ) by inducing a catalytic precipitation reaction. Variations in I D confer a sensitive discrimination of glucose enantiomers with a limit of detection (LOD) of 0.07 μM for L-Glu and 0.05 μM for D-Glu. A co-metabolization of xylose and glucose by Schizophyllum commune 227E.32 wild mushroom for exopolysaccharide (EPS) production is presented. Cultivations performed with S. commune 227E.32 at different xylose concentrations demonstrated that the concentration of 50 g·L -1 of xylose achieved the highest EPS production, around 4.46 g·L -1 . Scale-up in a stirred tank reactor (STR) was performed. 10 % inoculum showed the highest cost/benefit ratio regarding sugar conversion and EPS production (Y P/S  = 0.90 g·g -1 ), achieving 1.82 g·L -1 of EPS. Isolation, purification, and characterization were conducted with EPS produced in flasks and STR. GC-MS analysis showed glucose as main monosaccharide constituents for both isolates. 13 C NMR and HSQC-edited showed that both EPS isolated consisted of a β-D-Glcp (1 → 3) main chain, partially substituted at O-6 with nonreducing β-D-Glcp ends on every third residue, similar to β-D-glucan isolated from S. commune basidiomes known as schizophyllan (SPG). The M w was determined by GPC to 1.5 × 10 6  Da (flasks) and 1.1 × 10 6  Da (STR). Sulfur containing glycosides offer an exciting prospect for inclusion within noncanonical glycan sequences, particularly as enabling probes for chemical glycobiology and for carbohydrate-based therapeutic development. In this context, we required access to 4-thio-d-glucopyranose and sought its chemical synthesis. Unable to isolate this material in homogenous form, we observed instead a thermodynamic preference for interconversion of the pyranose to 4-thio-d-glucofuranose. Accordingly, we present an improved method to access both bis(4-thio-d-glucopyranoside)-4,4'-disulfide and 4-thio-d-glucofuranose from a single precursor, demonstrating that the latter compound can be accessed from the former using a dithiothreitol controlled reduction of the disulfide. The dithiothreitol-mediated interconversion between pyranose (monomer and disulfide) and furanose forms for this thiosugar is monitored by 1 H NMR spectroscopy over a 24-h period. Here we report synthetic monosaccharide channels built with shape-persistent organic cages, porphyrin boxes (PBs), that allow facile transmembrane transport of glucose and fructose through their windows. PBs show a much higher transport rate for glucose and fructose over disaccharides such as sucrose, as evidenced by intravesicular enzyme assays and molecular dynamics simulations. The transport rate can be modulated by changing the length of the alkyl chains decorating the cage windows. Insertion of a linear pillar ligand into the cavity of PBs blocks the monosaccharide transport. In vitro cell experiment shows that PBs transport glucose across the living-cell membrane and enhance cell viability when the natural glucose transporter GLUT1 is blocked. Time-dependent live-cell imaging and MTT assays confirm the cyto-compatibility of PBs. Like glucose, fructose is a monosaccharide, but the mechanisms of its absorption and metabolism in the body are very different between the 2 molecules. In this study, we investigated the effects of oral administration of glucose and fructose on food intake, diencephalic gene expression, and plasma metabolite concentrations in broiler chicks. The animals used in this study were 4-day-old male broiler chicks (Ross 308). They were given glucose, fructose (200 mg/ 0.5 mL/ bird), or a similar volume of distilled water orally after 6 h fasting. After treatment, measurements of food intake (at 0, 30, and 60 min), and blood glucose as well as insulin concentrations were measured over time; however, diencephalic (hypothalamus) gene expression and plasma metabolites were measured at 30 min. The results showed that glucose administration suppressed food intake, but fructose administration did not suppress food intake and it was at the same level as distilled water administration. In addition, fructose administration did not increase plasma glucose and insulin levels as did glucose administration. In the diencephalon, expression levels of genes related to the melanocortin system were unaffected by the treatment, while gene expression levels related to intracellular energy regulation, such as AMP-activated protein kinase were affected by the glucose treatment in the fasted chicks. These results suggest that fructose administration does not suppress feeding behavior as a result of possible reduction in the energy levels in the diencephalon and associated energy metabolism., (Copyright © 2022 The Authors. Isomaltose (6-O-α-d-glucopyranosyl-d-glucose) and isomaltulose (palatinose; 6-O-β-d-glucopyranosyl-d-fructose) were heated to 90 °C in 100 mM sodium phosphate buffer (pH 7.5). Aldose-ketose isomerization between isomaltose, isomaltulose, and epi-isomaltulose was observed in the early stage of the reaction, alongside the release of a small amount of glucose. The total concentration of these disaccharides gradually decreased as the heating time increased. However, this decrease did not correlate with the amount of glucose or fructose released, suggesting that the releases of these monosaccharides were not caused by the hydrolysis of glycosidic linkages. A slight decrease in the pH of the reaction solution was attributed to the formation of two organic acids, 6-O-β-d-glucopyranosyl-3-deoxy-d-arabino-hexonic acid (1) and 6-O-β-d-glucopyranosyl-3-deoxy-d-ribo-hexonic acid (2). These compounds were formed from the β-elimination of the hydroxyl group at the C-3 of fructose, leaving a substituted glucose residue at the C-6 position, followed by keto-enol tautomerization and benzilic acid rearrangement. Although approximately 30% of 1 and 2 were degraded after 360 min of heating at 90 °C in 100 mM sodium phosphate, a little release of glucose was observed, indicating no hydrolysis of the glucoside bond at C-6. Besides 1 and 2, time-dependent changes in the NMR spectra of the reaction mixture in water indicated the formation of formic acid and the presence of species possibly resulting from the β-elimination of the hydroxyl group from 3- and 4-ulose. Motivated by underestimation of the diffusion constant of glucose in aqueous solution at high glucose concentrations we performed additional optimization of the Drude polarizable hexopyranose monosaccharide force field. This indicated aggregation of the glucose at higher concentrations, which is a concern for studies of complex glycan systems such as the HIV Envelope where high effective concentrations of sugars are present. High-level quantum mechanical calculations were undertaken on water monohydrate-glucose interactions, on water cluster-glucose interactions and on glucose-glucose dimers in stacked (parallel) and perpendicular orientations. Optimization of the nonbond and dihedral parameters targeting these data yielded a revised model that showed improved agreement with experimental aqueous diffusion data. However, limitations in the diffusion constants were still present. These were due to the SWM4-NDP inherently overestimating the diffusion constant of water, a problem that was validated by calculation of the aqueous diffusion constants using the SWM6-NDP water model. In addition, results show the water diffusion constant to be significantly overestimated at high glucose concentrations though the glucose diffusion is in satisfactory agreement with experiment. These results indicate the subtle balance of water-sugar, water-water and sugar-sugar interactions that needs to be properly modeled to account for the full range of aqueous behavior of sugars in aqueous solution., (Copyright © 2018 Elsevier Ltd. Pollen tubes are fast growing, photosynthetically inactive cells. Their energy demand is covered by specific transport proteins in the plasma membrane that mediate the uptake of sugars. Here we report on the functional characterization of AtSTP10, a previously uncharacterized member of the SUGAR TRANSPORT PROTEIN family. Heterologous expression of STP10 cDNA in yeast revealed that the encoded protein catalyses the high-affinity uptake of glucose, galactose and mannose. The transporter is sensitive to uncouplers of transmembrane proton gradients, indicating that the protein acts as a hexose-H(+)symporter. Analyses of STP10 mRNA and STP10 promoter-reporter gene studies revealed a sink-specific expression pattern of STP10 in primordia of lateral roots and in pollen tubes. This restriction to sink organs is mediated by intragenic regions of STP10 qPCR analyses with cDNA of in vitro grown pollen tubes showed that STP10 expression was down-regulated in the presence of 50mM glucose. However, in pollen tubes of glucose-insensitive plants, which lack the glucose sensor hexokinase1 (HXK1), no glucose-induced down-regulation of STP10 expression was detected. A stp10T-DNA insertion line developed normally, which may point towards functional redundancy. The data presented in this paper indicate that a high-affinity glucose uptake system is induced in growing pollen tubes under low glucose conditions and that this regulation may occur through the hexokinase pathway., (© The Author 2016. Analyses of free glucose and glucose 6-phosphate in milk have until now been dependent upon several time consuming and troublesome procedures. This has limited investigations in the area. The present article presents a new, reliable, analytical procedure, based on enzymatic degradation and fluorometric detection. Standards and control materials were based on milk that was stripped of intrinsic glucose and glucose 6-phosphate in order to obtain standards and samples based on the same matrix. 1. The utilization of specifically labelled d-glucose in the biosynthesis of plum-leaf polysaccharides has been studied. After these precursors had been metabolized in plum leaves, the polysaccharides were isolated from the leaves, and their monosaccharide constituents isolated and purified. 2. Both the specific activities and the distribution of (14)C along the carbon chains of the monosaccharides were determined. Significant (14)C activity was found in units of d-galactose, d-glucose, d-xylose and l-arabinose, but their specific activities varied widely. The labelling patterns suggest that in the leaves the other monosaccharides all arise directly from d-glucose without any skeletal change in the carbon chain, other than the loss of a terminal carbon atom in the synthesis of pentoses. 3. The results indicated that within the leaf there are various precursor pools for polysaccharide synthesis and that these pools are not in equilibrium with one another.

Group D Types 1 and 26 cell walls and the corresponding type-specific carbohydrates, extracted from the walls by various means, contain rhamnose, glucose, galactose, N-acetylglucosamine, and N-acetylgalactosamine. Mucopeptide elements are also present in the walls and in enzymatically-extracted carbohydrates. Types 1 and 26 carbohydrates extracted by formamide contain no mucopeptide, but the serologic reactivity of Type 1 antigen is destroyed by this procedure. The Type 1 antigen was successfully extracted, however, by a new technique involving autolysis of cell walls at pH 6.2. The type carbohydrate prepared by this procedure has chemical and serological similarities to the antigens prepared by the S. albus enzyme and the lysozyme methods. Quantitative precipitin inhibition studies with Type 1 antigen and antibody indicate that D-glucose and N-acetylglucosamine may be components of the antigenic determinant. The terminal residue is probably bound by an beta-glycosidic bond to the subterminal sugar. Similar studies with the Type 26 carbohydrate have not revealed any of the chemical features of the antigenic determinant.

A study was carried out of the mechanisms of biological clogging of sand during prolonged percolation of water containing high levels of organic matter. It was found that polysaccharide-producing microorganisms predominated in clogged layers of sand. A positive correlation was observed between accumulation in the profile of polysaccharides and clogging of columns of sand in permeameters. The level of oxygen in the system appears to determine the equilibrium between production of clogging materials and their decomposition.

Phospholipids extracted from "ghosts" of human erythrocytes or from other sources carry substantial quantities of glucose or other monosaccharides from the dry state into highly nonpolar solvents. Various characteristics of this weak association phenomenon show suggestive parallels with known properties of the mediated sugar-transfer system in the membrane of the intact red cell.

Development of elegant, selective, and efficient strategies for the production of value‐added platform chemicals from renewable feedstocks are in high demand to achieve the future needs and sustainable goals. In this context, an efficient acid‐promoted synthesis of highly valuable hydroxymethylfurfural (HMF) has been demonstrated from glucose, a major constituent of lignocellulosic biomass. The major challenge in the conversion of glucose to HMF is the selective isomerization of glucose to ketose, which in the present work has been successfully addressed through the amine‐mediated rearrangement of glucose to aminofructose under Amadori rearrangement. Importantly, subsequent dehydration step affords HMF and regenerates the amine employed in the first step, which could be readily recovered. In addition, scale‐up and successful integration into one‐pot synthesis of HMF proves the efficiency and applicability of the present transformation in large scale application. The epimerization of glucose into mannose is of great significance for high-value utilization of monosaccharides. Extensive studies on the epimerization of glucose into mannose have been conducted using molybdenum-based catalysts, owing to their good activities for the epimerization reaction. Nonetheless, a great challenge for this reaction is that molybdenum species are prone to leaching into solution, which is a primary reason for catalyst deactivation. Additionally, the reaction usually gives a very low mannose yield and poor selectivity because it is a thermodynamic equilibrium reaction with sugar prone to side reactions at high temperatures. Herein, a metal–organic framework (MOF) material incorporating Keggin-type phosphomolybdate was explored as the catalyst for the epimerization of glucose. Under optimal conditions, an excellent mannose yield of up to 32.5% with an impressive selectivity of 94% was achieved. When other aldoses such as arabinose were used as starting materials, the catalyst also afforded high yield and product selectivity in the epimerization reaction. This novel hybrid material, a polyoxometalate-based metal–organic framework (POMOF), has been introduced to catalyze the conversion of sugars for the first time. Notably, in contrast to most molybdenum-based catalysts, the target hybrid showed good stability after five successive cycles. Furthermore, the reaction mechanism of the glucose-to-mannose epimerization was proposed, confirming the intramolecular carbon-skeleton rearrangement pathway (involving C-1 and C-2 positions) by isotope-labeling trace experiments. Chilling stress caused by extreme weather is threatening global rice (Oryza sativa L.) production. Identifying components of the signal transduction pathways underlying chilling tolerance in rice would advance molecular breeding. Here, we report that OsMST6 , which encodes a monosaccharide transporter, positively regulates the chilling tolerance of rice seedlings. mst6 mutants showed hypersensitivity to chilling, while OsMST6 overexpression lines were tolerant. During chilling stress, OsMST6 transported more glucose into cells to modulate sugar and abscisic acid signaling pathways. We showed that the transcription factor OsERF120 could bind to the DRE/CRT element of the OsMST6 promoter and activate the expression of OsMST6 to positively regulate chilling tolerance. Genetically, OsERF120 was functionally dependent on OsMST6 when promoting chilling tolerance. In summary, OsERF120 and OsMST6 form a new downstream chilling regulatory pathway in rice in response to chilling stress, providing valuable findings for molecular breeding aimed at achieving global food security. The Ruminiclostridium thermocellum cellulosome system is one of the most efficient cellulase systems in nature. The main hydrolysis product of cellulosic saccharification by R. thermocellum was cellobiose, which served as a strong feedback inhibitor to the cellulosome. In this study, R. thermocellum M3, which directly hydrolyzes cellulosic biomass into monosaccharides, was used to degrade cellobiose to investigate the kinetic characteristics of cellobiose degradation. Moreover, the transcriptomic characteristics of strain M3 under cellobiose and Avicel culture conditions were compared. The results indicated that strain M3 was able to grow and hydrolyze cellobiose under cellobiose concentrations higher than 20 g/L. The best growth performance and glucose production were obtained under 12 g/L cellobiose. Compared with that of Avicel, cellobiose had greater β-glucosidase activity under cellobiose culture, with a maximum glucose yield of 767.97±19.13 mg/g cellobiose. The transcription results indicated that R. thermocellum M3 resisted the feedback inhibition of cellobiose by regulating genes encoding β-glucosidase, cellobiose phosphorylase and ABC transporters. The high glucose yield of strain M3 was closely related to the expression of the bglX gene. In addition, the gene related to the chemotactic pathway of strain M3 also increased its perception and adaptation to cellobiose. This study demonstrates that Lactobacillus can produce exopolysaccharides (EPSs) using alternative carbon sources, such as sugarcane molasses and glycerol. After screening 22 strains of Lactobacillus to determine which achieved the highest production of EPS based on dry weight at 37 °C, the strain Ke8 (L. casei) was selected for new experiments. The EPS obtained using glycerol and glucose as carbon sources was classified as a heteropolysaccharide composed of glucose and mannose, containing 1730 g.mol−1, consisting of 39.4% carbohydrates and 18% proteins. The EPS obtained using molasses as the carbon source was characterized as a heteropolysaccharide composed of glucose, galactose, and arabinose, containing 1182 g.mol−1, consisting of 52.9% carbohydrates and 11.69% proteins. This molecule was characterized using Size Exclusion Chromatography (HPLC), Gas chromatography–mass spectrometry (GC-MS), Fourier-transform infrared spectroscopy (FTIR), and proton nuclear magnetic resonance spectroscopy (1H-NMR). The existence of polysaccharides was confirmed via FT-IR and NMR analyses. The results obtained suggest that Lacticaseibacillus casei can grow in media that use alternative carbon sources such as glycerol and molasses. These agro-industry residues are inexpensive, and their use contributes to sustainability. The lack of studies regarding the use of Lacticaseibacillus casei for the production of EPS using renewable carbon sources from agroindustry should be noted. [ABSTRACT FROM AUTHOR]

The detection of biomolecules utilizing surface-enhanced Raman spectroscopy (SERS) is highly trending in research, but the unlabeled and sensitive detection of saccharides in body fluids is challenging. Here, a novel SERS substrate (AgNS600) is fabricated by physical scratching of smooth silver sheets, which successfully achieves enhanced SERS fingerprinting by capturing glucose molecules through nanoscale grooves on the surface of the substrate to form multi-dimensional "hot spot" regions. The platform proves to be rapid, reliable, and reproducible, with a lower detection limit of 0.5 amol L-1 in deionized water. Further detection of multiple monosaccharides, polysaccharides, and mix samples, which are distinguished with machine learning and heat map, demonstrates the versatility and specificity of the platform. Importantly, real samples are explored and the results show that the platform is able to detect and recognize SERS signals of glucose in the blood, urine, tears, and perspiration, and non-destructive predict diabetes by analyzing the signal intensity of different characteristic peaks. In this work, it is the first non-chemical, unlabeled determination of glucose with high sensitivity via SERS, which provides a potential strategy for advancing the clinical development of SERS technology and the early diagnosis of glucose metabolic diseases. The therapeutic success of nanomedicines requires nanomaterials to either adhere to the surface or internalize within the cytoplasm. The endocytosis phenomenon is controlled by the nanomaterial's shape, size, composition, charge, and capping molecules. The membrane potential‐based non‐specific internalization of therapeutic nanomedicines offers limited benefits than receptor‐based specific delivery. Glut receptor‐based internalization of glucose molecules is a well‐known process in cancerous cells, which is one of the most exploited strategies to target cancer cells using nanoparticles. However, the internalization process of other structurally similar monosaccharides (D‐Galactose, Mannose, and D‐Fructose) conjugated nanoparticles remains to be unexplored. Herein, D‐Glucose, D‐Galactose, Mannose, and D‐Fructose‐coated AuNPs and AgNPs have been synthesized and studied the role of Glut receptors in their internalization in liver cancer cells, and compared them with non‐cancerous cells. Results revealed that almost all monosaccharide‐coated NPs exhibited high uptake in liver cancer cells than non‐cancerous cells. Glut‐1 receptor is observed to play a key role in the uptake and inhibition of Glut‐1 receptors by genistein lead to a significant decrease in nanoparticle uptake. In conclusion, monosaccharide‐conjugated nanoparticles can be used to direct the selective internalization of AuNPs and AgNPs in hepatic cancer cells to realize therapeutic and imaging applications. Sulfite and kraft pulping are two principal methods of industrial delignification of wood. In recent decades, those have been considered as possibilities to pretreat recalcitrant wood lignocellulosics for the enzymatic hydrolysis of polysaccharides and the subsequent fermentation of obtained sugars to valuable bioproducts. Current work compares chemistry and technological features of two different cooking processes in the preparation of polysaccharide substrates for deep saccharification with P. verruculosum glycosyl hydrolases. Bleached kraft and sulfite pulps were subjected to hydrolysis with enzyme mixture of high xylanase, cellobiohydrolase, and β-glucosidase activities at a dosage of 10 FPU/g of dry pulp and fiber concentration of 2.5, 5, and 10%. HPLC was used to analyze soluble sugars after hydrolysis and additional acid inversion of oligomers to monosaccharides. Kraft pulp demonstrated higher pulp conversion after 48 h (74–99%), which mostly resulted from deep xylan hydrolysis. Sulfite-pulp hydrolysates, obtained in similar conditions due to higher hexose concentration (more than 50 g/L), had higher fermentability for industrial strains producing alcohols, microbial protein, or organic acids. Along with saccharification, enzymatic modification of non-hydrolyzed residues occurred, which led to decreased degree of polymerization and composition changes in two industrial pulps. As a result, crystallinity of kraft pulp increased by 1.3%, which opens possibilities for obtaining new types of cellulosic products in the pulp and paper industry. The ionization potentials and total ionization cross sections of glucose and ribose monosaccharide molecules were measured by electron impact at energies up to 70 eV. Using two methods, Hartree–Fock (HF) and density functional theory (DFT), the structure of d- and l-forms of glucose and ribose molecules were calculated using Gaussian. The energy characteristics of the molecular orbitals (MOs) were used to calculate the summarized single ionization cross sections (CS) based on the MOs by the Binary–Encounter–Bethe (BEB) and Gryzinsky (Gryz) models. By normalizing to Gryz-DFT cross sections at thresholds, at 11 eV (Glucose) and 12.65 eV (Ribose), the absolute values of the measured total ionization cross sections of these molecules were obtained. The ionization potentials of glucose and ribose molecules were evaluated from binding HOMO MOs energies and compared with the measured values: 12.25 ± 0.25 eV (Glucose) and 10.46 ± 0.25 eV (Ribose).
