This article reports a facile synthesis of radiolabeled PdCu@Au core-shell tripods for use in positron emission tomography (PET) and image-guided photothermal cancer treatment by directly incorporating radioactive (64)Cu atoms into the crystal lattice. The tripod had a unique morphology determined by the PdCu tripod that served as a template for the coating of Au shell, in addition to well-controlled specific activity and physical dimensions. The Au shell provided the nanostructure with strong absorption in the near-infrared region and effectively prevented the Cu and (64)Cu atoms in the core from oxidization and dissolution. When conjugated with D-Ala1-peptide T-amide (DAPTA), the core-shell tripods showed great enhancement in targeting the C-C chemokine receptor 5 (CCR5), a newly identified theranostic target up-regulated in triple negative breast cancer (TNBC). Specifically, the CCR5-targeted tripods with an arm length of about 45 nm showed 2- and 6-fold increase in tumor-to-blood and tumor-to-muscle uptake ratios, respectively, relative to their nontargeted counterpart in an orthotopic mouse 4T1 TNBC model at 24 h postinjection. The targeting specificity was further validated via a competitive receptor blocking study. We also demonstrated the use of these targeted, radioactive tripods for effective photothermal treatment in the 4T1 tumor model as guided by PET imaging. The efficacy of treatment was confirmed by the significant reduction in tumor metabolic activity revealed through the use of (18)F-fluorodeoxyglucose PET/CT imaging. Taken together, we believe that the (64)Cu-doped PdCu@Au tripods could serve as a multifunctional platform for both PET imaging and image-guided photothermal cancer therapy.

In metal-enhanced fluorescence (MEF), the localized surface plasmon resonances of metallic nanostructures amplify the absorption of excitation light and assist in radiating the consequent fluorescence of nearby molecules to the far-field. This effect is at the base of various technologies that have strong impact on fields such as optics, medical diagnostics, and biotechnology. Among possible emission bands, those in the near-infrared (NIR) are particularly intriguing and widely used in proteomics and genomics due to its noninvasive character for biomolecules, living cells, and tissues, which greatly motivates the development of effective and, eventually, multifunctional NIR-MEF platforms. Here, we demonstrate NIR-MEF substrates based on Au nanocages micropatterned with a tight spatial control. The dependence of the fluorescence enhancement on the distance between the nanocage and the radiating dipoles is investigated experimentally and modeled by taking into account the local electric field enhancement and the modified radiation and absorption rates of the emitting molecules. At a distance around 80 nm, a maximum enhancement up to 2-7 times with respect to the emission from pristine dyes (in the region 660-740 nm) is estimated for films and electrospun nanofibers. Due to their chemical stability, finely tunable plasmon resonances, and large light absorption cross sections, Au nanocages are ideal NIR-MEF agents. When these properties are integrated with the hollow interior and controllable surface porosity, it is feasible to develop a nanoscale system for targeted drug delivery with the diagnostic information encoded in the fluorophore.

We report a facile synthesis of Au tetrahedra in high purity and with tunable, well-controlled sizes via seed-mediated growth. The success of this synthesis relies on the use of single-crystal, spherical Au nanocrystals as the seeds and manipulation of the reaction kinetics to induce an unsymmetrical growth pattern for the seeds. In particular, the dropwise addition of a precursor solution with a syringe pump, assisted by cetyltrimethylammonium chloride and bromide at appropriate concentrations, was found to be critical to the formation of Au tetrahedra in high purity. Their sizes could be readily tuned in the range of 30-60 nm by simply varying the amount of precursor added to the reaction solution. By manipulating the kinetics of seeded growth through judicious control of reaction conditions, edge-selective metal deposition can be achieved to synthesize new Au nanostructures with face-centered concavities, referred to herein as Au overgrown trisoctahedra. These nanostructures display higher sensitivity to changes in refractive index compared to both Au traditional trisoctahedra and the Au nanocube seeds from which they are grown. Often, concave nanostructures are achieved by selective etching processes or corner-selective overgrowth and adopt a stellated profile rather than a profile with face-centered concavities. Single-crystal gold nanospheres with controlled diameters in the range 5-30 nm were synthesized by using a facile approach that was based on successive seed-mediated growth. The key to the success of this synthesis was the use of hexadecyltrimethylammonium chloride (CTAC) as a capping agent and a large excess of ascorbic acid as a reductant to ensure fast reduction and, thus, single crystallinity and a spherical shape of the resultant nanoparticles. The diameters of the gold nanospheres could be readily controlled by varying the amount of seeds that were introduced into the reaction system. The gold nanospheres could be produced with uniform diameters of up to 30 nm; thus, their localized surface plasmon resonance properties could be directly compared with the results that were obtained from theoretical calculations. Interestingly, we also found that these gold nanospheres self-assembled into dimers, larger aggregates, and wavy nanowires when they were collected by centrifugation, dispersed in deionized water, and then diluted to different volumes with deionized water., (Copyright © 2013 WILEY-VCH Verlag GmbH & Co. Molecular imaging techniques based on surface-enhanced Raman scattering (SERS) face a lack of reproducibility and reliability, thus hampering its practical application. Flower-like gold nanoparticles have strong SERS enhancement performance due to having plenty of hot-spots on their surfaces, and this enhancement is not dependent on the aggregation of the particles. These features make this kind of particle an ideal SERS substrate to improve the reproducibility in SERS imaging. Here, the SERS properties of individual flower-like gold nanoparticles are systematically investigated. The measurements reveal that the enhancement of a single gold nanoparticle is independent of the polarization of the excitation laser with an enhancement factor as high as 10(8) . After capping with Raman signal molecules and folic acid, the gold nanoflowers show strong Raman signal in the living cells, excellent targeting properties, and a high signal-to-noise ratio for SERS imaging., (Copyright © 2013 WILEY-VCH Verlag GmbH & Co. Second-harmonic generation (SHG) from periodic arrays of subwavelength rectangular air hole with various aspect ratios perforated in gold thin films can get resonantly enhanced for some specific geometric shapes. Here we clarify the physical origin of this shape resonance effect. A nonlinear coupled-mode theory is set up to solve energy conversion from fundamental wave (FW) mode to second-harmonic wave (SHW) mode within the nanoscale air hole. It reveals that several physical mechanisms, including the FW mode excitation amplitude, FW-SHW modal spatial overlap, FW-SHW mode phase mismatch, and SHW mode attenuation, are all geometric shape sensitive and altogether act to induce the SHG shape resonance effect. The theory agrees well with experimental observations and provides an accurate and complete explanation for the long-emphasized but elusive shape effect. The study may stimulate deeper insights to visualize general nonlinear nanophotonic processes and pave the way to engineering high-efficiency nonlinear nanophotonic devices.

Gold nanorods are too tiny to be manipulated using conventional mechanical methods. In this paper, we demonstrate the trapping, transferring, positioning and patterning of gold nanorods with dual-optical tweezers. The convenient manipulations are achieved by taking advantage of the longitudinal surface plasmon resonance of gold nanorods and the anisotropic optical trapping forces formed by two linearly polarized Gaussian beams. The trapped gold nanoparticles are positioned extremely firmly and quickly on a substrate compared with randomly dispersed ones. It is observed that gold nanorods show advantages over gold nanospheres with regard to positioning speed and stability. More importantly, versatile plasmon coupling effects have been achieved in some patterned nanorods.

We report a facile method for the synthesis of uniform Au octahedra with well-controlled sizes and optical properties by seed-mediated growth. Starting from single-crystal seeds of Au spheres with a uniform size, we could reproducibly obtain Au octahedra with a narrow size distribution (<7% in standard deviation) and in high purity (>90%). Moreover, the edge lengths of these Au octahedra could be readily tuned in a controllable fashion from 16 to 77 nm by varying the amount of seeds, the concentration of HAuCl(4) , or both. A simple Ar(+)-ion irradiation route has been developed to prepare gold nanoneedle arrays on glass substrates for surface-enhanced Raman scattering (SERS)-active substrates. The nanoneedles exhibited very sharp tips with an apex diameter of 20 nm. These arrays were evaluated as potential SERS substrates using malachite green molecules and exhibited a SERS enhancement factor of greater than 10(8), which is attributed to the localized electron field enhancement around the apex of the needle and the surface plasmon coupling originating from the periodic structure. This work demonstrates a new technique for producing controllable and reproducible SERS substrates potentially applicable for chemical and biological assays.

This paper describes the synthesis of Pd-Au bimetallic nanocrystals with controlled morphologies via a one-step seeded-growth method. Two different reducing agents, namely, L-ascorbic acid and citric acid, were utilized for the reduction of HAuCl(4) in an aqueous solution to control the overgrowth of Au on cubic Pd seeds. When L-ascorbic acid was used as the reducing agent, conformal overgrowth of Au on the Pd nanocubes led to the formation of Pd-Au nanocrystals with a core-shell structure. On the contrary, localized overgrowth of Au was observed when citric acid was used as the reducing agent, producing Pd-Au bimetallic dimers. Through this morphological control, we were able to tune the localized surface plasmon resonance peaks of Pd-Au bimetallic nanostructures in the visible region.

Gold nanocages with an average edge length of 65 +/- 7 nm and a strong absorption peak at 800 nm were conjugated with monoclonal antibodies (anti-HER2) to target breast cancer cells (SK-BR-3) through the epidermal growth factor receptor (in this case, HER2), which is overexpressed on the surfaces of the cells. Both the number of immuno Au nanocages immobilized per cell and the photothermal therapeutic effect were quantified using flow cytometry. The targeted cells were irradiated with a pulsed near-infrared laser, and by varying the power density, the duration of laser exposure, and the time of response after irradiation, we were able to optimize the treatment conditions to achieve effective destruction of the cancer cells. We found that cells targeted with the immuno Au nanocages responded immediately to laser irradiation and that the cellular damage was irreversible at power densities greater than 1.6 W/cm(2). The percentage of dead cells increased with increasing exposure time up to 5 min and then became steady. By quantifying the photothermal effect of immuno Au nanocages, critical information with regards to both the optimal dosage of nanocages and parameters of the laser irradiation has been garnered and will be applied to future in vivo studies.

Poly(ethylene glycol)-coated Au nanocages have been evaluated as a potential near-infrared (NIR) contrast agent for photoacoustic tomography (PAT). Previously, Au nanoshells were found to be an effective NIR contrast agent for PAT; however, Au nanocages with their more compact sizes (<50 nm compared to >100 nm for Au nanoshells) and larger optical absorption cross sections should be better suited for in vivo applications. We sequentially injected Au nanocages into the circulatory system of a rat in three administrations and in vivo PAT was conducted immediately prior to the first injection and continued until 5 h after the final injection. A gradual enhancement of the optical absorption in the cerebral cortex, by up to 81%, was observed over the course of the experiment.

This paper describes a two-step procedure for generating cubic nanocages and nanoframes. In the first step, Au/Ag alloy nanoboxes were synthesized through the galvanic replacement reaction between Ag nanocubes and an aqueous HAuCl4 solution. The second step involved the selective removal (or dealloying) of Ag from the alloy nanoboxes with an aqueous etchant based on Fe(NO3)3 or NH4OH. The use of a wet etchant other than HAuCl4 for the dealloying process allows one to better control the wall thickness and porosity of resultant nanocages because there is no concurrent deposition of Au. By increasing the amount of Fe(NO3)3 or NH4OH added to the dealloying process, nanoboxes derived from 50-nm Ag nanocubes could be converted into nanocages and then cubic nanoframes with surface plasmon resonance (SPR) peaks continuously shifted from the visible region to 1200 nm. It is also possible to obtain nanocages with relatively narrow SPR peaks (with a full width at half-maximum as small as 180 nm) by controlling the amount of HAuCl4 used for the galvanic replacement reaction and thus the optimization of the percentage of Au in the alloy nanoboxes.

Gold nanocages with a relatively small size (e.g., approximately 45 nm in edge length) have been developed, and the structure of these nanocages was tailored to achieve strong absorption in the near-infrared (NIR) region for photothermal cancer treatment. Numerical calculations show that the nanocage has a large absorption cross section of 3.48 x 10(-14) m(2), facilitating conversion of NIR irradiation into heat. The gold nanocages were conjugated with monoclonal antibodies (anti-HER2) to target epidermal growth factor receptors (EGFR) that are overexpressed on the surface of breast cancer cells (SK-BR-3). Our preliminary photothermal results show that the nanocages strongly absorb light in the NIR region with an intensity threshold of 1.5 W/cm(2) to induce thermal destruction to the cancer cells. In the intensity range of 1.5-4.7 W/cm(2), the circular area of damaged cells increased linearly with the irradiation power density. These results suggest that this new class of bioconjugated gold nanostructures, immuno gold nanocages, can potentially serve as an effective photothermal therapeutic agent for cancer treatment.

This article presents a mechanistic study on the galvanic replacement reaction between 11- and 14-nm multiply twinned particles (MTPs) of Ag and HAuCl4 in chloroform. We monitored both morphological and spectral changes as the molar ratio of HAuCl4 to Ag was increased. The details of reaction were different from previous observations on single-crystal Ag nanocubes and cuboctahedrons. Because Au and Ag form alloys rapidly within small MTPs rich in vacancy and grain boundary defects, a complete Au shell did not form on the surface of each individual Ag template. Instead, the replacement reaction resulted in the formation of alloy nanorings and nanocages from Ag MTPs of decahedral or icosahedral shape. For the nanorings and nanocages derived from 11-nm Ag MTPs, the surface plasmon resonance (SPR) peak can be continuously shifted from 400 to 616 nm. When the size of Ag MTPs was increased to 14 nm, the SPR peak can be further shifted to 740 nm, a wavelength sought by biomedical applications. We have also investigated the effects of capping ligands and AgCl precipitate on the replacement reaction. While hollow structures were routinely generated from oleylamine-capped Ag MTPs, we obtained very few hollow structures by using a stronger capping ligand such as oleic acid or tri-n-octylphosphine oxide (TOPO). Addition of extra oleylamine was found to be critical to the formation of well-controlled, uniform hollow structures free of AgCl contamination thanks to the formation of a soluble complex between AgCl and oleylamine.

Gold-silver alloy nanocages with controllable pores on the surface have been synthesized via galvanic replacement reaction between truncated Ag nanocubes and aqueous HAuCl4. Unlike in the previous studies, the initiation of replacement reaction started in a controllable way, simultaneously from eight corners of the truncated Ag nanocubes where {111} facets were exposed. The formation of cubic nanocages with pores at all the corners was determined by the capping agent, poly(vinyl pyrrolidone) (PVP), which preferentially covered the {100} facets of a truncated Ag nanocube.

The surface plasmon resonance peaks of gold nanostructures can be tuned from the visible to the near infrared region by controlling the shape and structure (solid vs. hollow). In this tutorial review we highlight this concept by comparing four typical examples: nanospheres, nanorods, nanoshells, and nanocages. A combination of this optical tunability with the inertness of gold makes gold nanostructures well suited for various biomedical applications.

The optical properties of two Au-Ag nanobox samples with average edge lengths of 44 and 58 nm and wall thicknesses of 6 and 8 nm, respectively, have been studied by single particle spectroscopy. The measurements gave an average line width of Gamma = 306 +/- 7 meV with a standard deviation of sigma = 30 meV for the 44-nm boxes, and Gamma = 350 +/- 9 meV with sigma = 35 meV for the 58-nm boxes. These line widths are much broader than those of gold nanorods with comparable resonance energies. The increased broadening is attributed to a combination of surface scattering of electrons, as well as increased radiation damping for the nanoboxes. Discrete dipole approximation calculations have been performed with and without surface scattering of electrons to compare with the experimental spectra. The calculations confirm that both electron-surface scattering and radiation damping are important effects in this system.

Gold nanocages of <40 nm in dimension have been synthesized using the galvanic replacement reaction between Ag nanocubes and HAuCl4 in an aqueous solution. By controlling the molar ratio between Ag and HAuCl4, the gold nanocages could be tuned to display surface plasmon resonance peaks around 800 nm, a wavelength commonly used in optical coherence tomography (OCT) imaging. OCT measurements on phantom samples indicate that these gold nanocages have a moderate scattering cross-section of approximately 8.10 x 10(-16) m2 but a very large absorption cross-section of approximately 7.26 x 10(-15) m2, suggesting their potential use as a new class of contrast agents for optical imaging. When bioconjugated with antibodies, the gold nanocages have also been demonstrated for specific targeting of breast cancer cells.

The coupling effect between two gold nanospheres (GNSs) is investigated for second-harmonic generation (SHG) in this paper. When the two GNSs approach each other, the coupling effect becomes stronger, and it consequently leads to stronger enhancement of SHG. A strong coupling area between two GNSs is also found. We propose a physical quantity, the coupling strength to describe the coupling effect on SHG enhancement and show that it can fully illustrate the relationship between the SHG enhancement and the gap of two GNSs. Moreover, further calculations reveal the existence of hot spots of both fundamental wave (FW) and excited second harmonic wave in the intermediate region of two GNSs. Our study can not only help to understand the coupling effect on SHG precisely but also offer some possibility to design a nonlinear plasmonic nano-ruler. [ABSTRACT FROM AUTHOR]
