Most aptasensors are designed based on aptamers' conformational change or structural transformation upon interaction with their targets. A limitation of these strategies is the unpredictability of such structural alterations that may yield false-positive or nonspecific signals. In bioanalysis, a dominant strategy for protein determination is the sandwich assay because of its high specificity and sensitivity resulting from the dual recognition mechanism. However, the approach is poorly suited to detection of low molecular weight targets, which unlikely bind to two aptamers simultaneously due to steric hindrance. To circumvent this drawback, a split aptamer strategy was recently developed, in which nucleic acid aptamers were split into two fragments that could specifically form a ternary assembly in the presence of ligand. This elegant approach has been extensively adopted for detection of various targets, especially small molecular weight targets, with different transduction methods including colorimetric, fluorescence, and electrochemical techniques. This review first introduced split aptamers and how to obtain split aptamers, and then summarized recent advances in the development of sandwich-format biosensing assays based on split aptamer fragments, aiming at providing a general guide for the design of split-aptamer-based sandwich assays. Furthermore, we discussed possible challenges in the development of split-aptamer-based sandwich assay regarding stability, sensitivity and multiplicity, so as to offer future opportunities of this assay. A class-specific molecular imprinted polymer (MIP) is described for simultaneous recognition of prometryn and simetryn prior to their determination via a fingerprint signal (at 974 cm−1 and 1074 cm−1) in the surface enhanced Raman scattering (SERS) spectra that were acquired in the presence of gold nanoparticles. The imprinted nanoparticles were applied to the analysis of rice and wheat samples spiked with both herbicides. The method has fairly good recoveries (72.7-90.9%) with a relative standard deviation of 1.7-7.8%, and a 20 μg·kg−1 limit of detection. The imprint factors (compared to non-imprinted polymers) are 5.3 for prometryn and 4.2 for simetryn (both at 10 μg·mL−1 of the initial solution).A MIP-SERS method was developed for simultaneous detection of triazine herbicides (prometryn and simetryn) in food samples. Aptamer-based strip assay is an easy, highly efficient and low-cost detection method, which has been developed and easily applied to onsite detection. A new sensitive sandwich dipstick assay for adenosine triphosphate (ATP) detection was successfully developed based on specific recognition between split aptamer fragments and the target. In this method, the thiolated aptamer was first conjugated to the surface of gold nanoparticles (AuNPs), while the biotin-aptamer was immobilized on the surface of a nitrocellulose filter in the test line. In the presence of ATP, the thiol-aptamer/ATP/biotin-aptamer complexes were generated, which led to an obvious increase in optical signals at the test line. Under the optimal determination conditions, an excellent linear logarithmic response to the ATP concentration was obtained within the range of 0.5 μM to 5 mM. The limit of detection (LOD) of 0.5 μM was reached at a signal-to-noise ratio of 3. The dipstick assay showed a good average recovery of 96-108 % with the RSD of less than 20 % in urine samples. The proposed method exhibited high specificity against other nucleotides such as the uridine triphosphate (UTP), cytidine triphosphate (CTP), and guanosine triphosphate (GTP). In this study, we propose an interference-free SERS-based aptasensor for trace detection of chlorpyrifos (CPF) in real samples. In the aptasensor, gold nanoparticles coated with Prussian blue (Au@PB NPs) were employed as SERS tags to provide a sole and intense Raman emission at 2160 cm−1, which could avoid overlapping with the Raman spectrum of the real samples in 600–1800 cm−1 to improve the anti-matrix effect ability of the aptasensor. Under the optimum conditions, this aptasensor displayed a linear response for CPF detection in the range of 0.1–316 ng mL−1 with a low detection limit of 0.066 ng mL−1. In addition, the prepared aptasensor shows excellent application to determine CPF in cucumber, pear and river water samples. The recovery rates were highly correlated with high-performance liquid chromatography‒mass spectrometry (HPLC‒MS/MS). This aptasensor shows interference-free, specific and sensitive detection for CPF and offers an effective strategy for other pesticide residue detection. A competitive bio-barcode immunoassay is described for the trace detection of parathion in water, pear, cabbage, and rice samples. It is based on amplification by platinum nanoparticle acting as a nanozyme. Gold nanoparticles (AuNPs) were modified with (a) monoclonal antibodies (mAbs) against parathion, and (b) thiolated single-stranded DNA (ssDNA) oligonucleotides. Magnetic nanoparticles (MNPs) were functionalized with ovalbumin coupled with parathion hapten. Parathion and its hapten compete with mAbs on the surface of the AuNPs. Subsequently, the platinum nanoparticles (PtNPs) probe, which was functionalized with the complementary thiolated ssDNA (C-ssDNA), was added to the reaction mixture for the detection of parathion. The signal was catalytically amplified by coupling with platinum nanozyme using teramethylbenzidine and H2O2 as the chromogenic system. The immunoassay has a linear range that extends from 0.01–50 μg·L−1, and the limit of detection is 2.0 × 10−3 μg·L−1. The recoveries and relative standard deviations (RSDs) ranged from 91.1–114.4% and 3.6–15.8%, respectively. It is known that gold nanoparticles (AuNPs) possess peroxidase-like activity. They can catalyze the oxidation of 3,3,5,5-tetramethylbenzidine by HO which leads to a color change from red to blue. It is shown here that the peroxidase-like activity of AuNPs can be inhibited by passivating its surface passivation with a ssDNA aptamer against sulfadimethoxine. If, however, the target molecule (sulfadimethoxine) is present, the aptamer is desorbed from the AuNPs surface, and this results in the reactivation of the catalytic property of the AuNPs. The color change of the solution (from purple to blue) is related to the analyte concentration, and this can be judged visually or by UV-visible absorptiometry at 650 nm. The assay, under optimized conditions, has a detection limit of 10 ng·mL of sulfadimethoxine, and the calibration plot is linear over a rather wide concentration range (0.01-1000 μg·mL). The assay can be performed within <15 min, is sensitive, and therefore is well suited for fast screening in food analysis. Fast immunoassay-based screening methods are unavailable for most small-molecule pesticides because of a lack of immunogenicity and the difficulty in obtaining antibodies by animal immunization. Aptamers are single-stranded DNA molecules selected through an in vitro process, which can bind to any target including nonimmunogenic small molecules with high affinity and specificity. Although various aptamer-based sensing methods have been developed for antibiotics, microorganisms, heavy metal ions, and biotoxins, there are few reports on aptamer-based methods for quick detection of organophosphorous pesticides. The gold (Au) nanoparticle (AuNP) colorimetric assay is a widely utilized rapid detection method because of properties such as easy operation and visualized results. In the present study, organophosphorous pesticide aptamers were adsorbed on the surface of AuNPs to stabilize the AuNP solution against high concentrations of salt to prevent AuNP aggregation. After the addition of targets, the aptamers binding to the targets are detached from the AuNPs, resulting in aggregation of AuNPs and a color change from red to purple-blue. The proposed method can detect 6 organophosphorous pesticides with good recoveries from 72% to 135% in environmental river water samples.
