The immune response represents a suite of evolved traits that can involve energetic and evolutionary trade‐offs with other energy‐demanding and fitness‐related processes. Here, we tested the hypothesis that aquatic beetles living in inland hypersaline habitats have lower immune capacity than freshwater congeners.Phenoloxidase activity, encapsulation response and antimicrobial peptide activity were compared in freshwater/hypersaline species pairs with differing osmoregulatory capacity and cuticular waterproofing properties in the genera Nebrioporus (Dytiscidae) and Enochrus (Hydrophilidae), independent evolutionary lineages that have colonised saline media separately.Hypersaline species (N. ceresyi and E. jesusarribasi) showed significantly lower phenoloxidase activity and antimicrobial peptide responses than their freshwater relatives (N. clarkii and E. salomonis). Encapsulation responses in freshwater species also appeared to be higher than in hypersaline relatives.Our results reinforce the complex nature of immune responses and suggest that adaptation to saline environments may have involved a trade‐off between osmoregulation and investment in immune defences, but also are consistent with relaxed selection pressures on basal immune responses resulting from lower microbial infection load in saline habitats. In addition, the more resistant cuticle of species occupying such habitats may protect against the entry of parasites, reducing selection pressure on immunity.Because the evolution of salinity tolerance is associated with reduced immune capacity, saline specialists may be particularly vulnerable to the dilution of saline waters and consequent changes in pathogen communities and load following colonisation by more generalist microorganisms. The climatic variability hypothesis predicts the evolution of species with wide thermal tolerance ranges in environments with variable temperatures, and the evolution of thermal specialists in thermally stable environments. In caves, the extent of spatial and temporal thermal variability experienced by taxa decreases with their degree of specialization to deep subterranean habitats. We use phylogenetic generalized least squares to model the relationship among thermal tolerance (upper lethal limits), subterranean specialization (estimated using ecomorphological traits), and habitat temperature in 16 beetle species of the tribe Leptodirini (Leiodidae). We found a significant, negative relationship between thermal tolerance and the degree of subterranean specialization. Conversely, habitat temperature had only a marginal effect on lethal limits. In agreement with the climatic variability hypothesis and under a climate change context, we show that the specialization process to live in deep subterranean habitats involves a reduction of upper lethal limits, but not an adjustment to habitat temperature. Thermal variability seems to exert a higher evolutionary pressure than mean habitat temperature to configure the thermal niche of subterranean species. Our results provide novel insights on thermal physiology of species with poor dispersal capabilities and on the evolutionary process of adaptation to subterranean environments. We further emphasize that the pathways determining vulnerability of subterranean species to climate change greatly depend on the degree of specialization to deep subterranean environments. One of the main challenges in ecology, biogeography and evolution is to understand and predict how species may respond to environmental changes. Here we focus on the deep subterranean environment, a system that minimizes most of the typical uncertainties of studies on epigean (surface) environments. Caves are relatively homogeneous habitats with nearly constant environmental conditions and simplified biological communities, allowing to control for biotic interactions. Thus, this particular system could be considered a natural habitat whose environmental conditions are similar to what can be reproduced in a laboratory, being an ideal model system for ecological, biogeographical and evolutionary studies. Subterranean species may potentially be used to assess the capability to persist in situ in a global change scenario, as they cannot accommodate to drastic changing conditions by behavioural plasticity, microhabitat use or by migrating to distant, more suitable areas, something frequent in epigean environments. In order to provide accurate predictions of the response of the subterranean biodiversity to climate change, we encourage evolutionary biologist, biogeographers and conservation biologist to work in this interesting ecosystem. Transitions from fresh to saline habitats are restricted to a handful of insect lineages, as the colonization of saline waters requires specialized mechanisms to deal with osmotic stress. Previous studies have suggested that tolerance to salinity and desiccation could be mechanistically and evolutionarily linked, but the temporal sequence of these adaptations is not well established for individual lineages. We combined molecular, physiological and ecological data to explore the evolution of desiccation resistance, hyporegulation ability (i.e., the ability to osmoregulate in hyperosmotic media) and habitat transitions in the water beetle genus Enochrus subgenus Lumetus (Hydrophilidae). We tested whether enhanced desiccation resistance evolved before increases in hyporegulation ability or vice versa, or whether the two mechanisms evolved in parallel. The most recent ancestor of Lumetus was inferred to have high desiccation resistance and moderate hyporegulation ability. There were repeated shifts between habitats with differing levels of salinity in the radiation of the group, those to the most saline habitats generally occurring more rapidly than those to less saline ones. Significant and accelerated changes in hyporegulation ability evolved in parallel with smaller and more progressive increases in desiccation resistance across the phylogeny, associated with the colonization of meso- and hypersaline waters during global aridification events. All species with high hyporegulation ability were also desiccation-resistant, but not vice versa. Overall, results are consistent with the hypothesis that desiccation resistance mechanisms evolved first and provided the physiological basis for the development of hyporegulation ability, allowing these insects to colonize and diversify across meso- and hypersaline habitats. As species' physiological breadth determines their potential to deal with environmental changes, and influences individuals' survival and the persistence of populations, information about lethal and sublethal responses could be fundamental for conservation purposes. 2. We used a standard experimental approach to explore mortality and behavioural avoidance responses (i.e. flight and emersion from the water) to a combination of acute heat and osmotic stress on six species of saline water beetles (belonging to Enochrus, Nebrioporus, and Ochthebius genera). 3. Heat stress affected survival and behavioural responses in all of the species, whereas osmotic stress and the interaction between both stressors only showed significant effects for the Ochthebius genus. Behavioural and survival patterns were highly interrelated across the stress gradients. The Enochrus and Nebrioporus studied species showed maximum avoidance activity at 35-40 °C, and a short (< 30 min) exposure to 45 °C was lethal. Ochthebius species were the most heat tolerant and displayed increasing behavioural responses with increasing temperature. In the Nebrioporus and Ochthebius genera, the species occupying lotic, more environmentally stable habitats, showed greater mortality, and avoidance responses were higher or initiated at lower stress thresholds than lentic species. In contrast, both Enochrus species displayed a similar mortality, and the lentic species E. bicolor emerged and flew more than the lotic E. falcarius, in concordance with its higher dispersal capacity. 4. Avoidance responses could provide interesting information about species' physiological amplitudes as a complement to lethal responses. The lotic species here studied showed narrower physiological amplitude (i.e. N. baeticus and O. glaber) or lower dispersal ability (i.e. E. falcarius) than their lentic relatives; both traits could result in a higher vulnerability of lotic species to thermal habitat changes.
