Abstract

Occupational exposure to lead generally occurs by inhalation of lead dust or fumes. Symptoms of lead poisoning include weight loss, vomiting, anemia, and convulsions. The accepted standard level of lead exposure has been set at 400 micrograms per liter (ug/L) of blood by the World Health Organization. Therefore, the effects of chronic lead exposure were determined. Female rabbits were given subcutaneous injections of lead acetate three times a week to maintain a constant blood lead level of 400 ug/L (low dose) or 800 ug/L (high dose). Lead concentrations in blood and tissue samples were determined by atomic absorption spectroscopy. After seven and a half months of treatment, rabbits on the high dose regime gained less weight than those on the lower dose regime. The high dose group developed a slight anemia (decrease in the number of red blood cells), which resolved after lead administration was stopped. No significant effect on urinary output or enzyme levels was detected, and no change in liver metabolism (levels of cytochrome P-450) was observed. Lead was stored mainly in bones. Heart and liver weight increased during lead exposure, but returned to normal after treatment was discontinued. The results indicate that exposure to lead at the 400 ug/L level does not produce adverse physiological effects in rabbits. (Consumer Summary produced by Reliance Medical Information, Inc.)
