Summary: Although most xyloglucans (XyGs) biosynthesis enzymes have been identified, the molecular mechanism that defines XyG branching patterns is unclear. Four out of five XyG xylosyltransferases (XXT1, XXT2, XXT4, and XXT5) are known to add the xylosyl residue from UDP‐xylose onto a glucan backbone chain; however, the function of XXT3 has yet to be demonstrated.Single xxt3 and triple xxt3xxt4xxt5 mutant Arabidopsis (Arabidopsis thaliana) plants were generated using CRISPR‐Cas9 technology to determine the specific function of XXT3.Combined biochemical, bioinformatic, and morphological data conclusively established for the first time that XXT3, together with XXT4 and XXT5, adds xylosyl residue specifically at the third glucose in the glucan chain to synthesize XXXG‐type XyGs. We propose that the specificity of XXT3, XXT4, and XXT5 is directed toward the prior synthesis of the acceptor substrate by the other two enzymes, XXT1 and XXT2. We also conclude that XXT5 plays a dominant role in the synthesis of XXXG‐type XyGs, while XXT3 and XXT4 complementarily contribute their activities in a tissue‐specific manner.The newly generated xxt3xxt4xxt5 mutant produces only XXGG‐type XyGs, which further helps to understand the impact of structurally deficient polysaccharides on plant cell wall organization, growth, and development. The plant's recalcitrant cell wall is composed of numerous polysaccharides, including cellulose, hemicellulose, and pectin. The most abundant hemicellulose in dicot cell walls is xyloglucan, which consists of a β-(1- > 4) glucan backbone with α-(1- > 6) xylosylation producing an XXGG or XXXG pattern. Xylose residues of xyloglucan are branched further with different patterns of arabinose, fucose, galactose, and acetylation that varies between species. Although xyloglucan research in other species lag behind Arabidopsis thaliana , significant advances have been made into the agriculturally relevant species Oryza sativa and Solanum lycopersicum , which can be considered model organisms for XXGG type xyloglucan. In this review, we will present what is currently known about xyloglucan biosynthesis in A. thaliana , O. sativa , and S. lycopersicum and discuss the recent advances in the characterization of the glycosyltransferases involved in this complex process and their organization in the Golgi. The plant cell wall has many significant structural and physiological roles, but the contributions of the various components to these roles remain unclear. Modification of cell wall properties can affect key agronomic traits such as disease resistance and plant growth. The plant cell wall is composed of diverse polysaccharides often decorated with methyl, acetyl, and feruloyl groups linked to the sugar subunits. In this study, we examined the effect of perturbing cell wall acetylation by making transgenic Arabidopsis (Arabidopsis thaliana) and Brachypodium (Brachypodium distachyon) plants expressing hemicellulose- and pectin-specific fungal acetylesterases. All transgenic plants carried highly expressed active Aspergillus nidulans acetylesterases localized to the apoplast and had significant reduction of cell wall acetylation compared with wild-type plants. Partial deacetylation of polysaccharides caused compensatory up-regulation of three known acetyltransferases and increased polysaccharide accessibility to glycosyl hydrolases. Transgenic plants showed increased resistance to the fungal pathogens Botrytis cinerea and Bipolaris sorokiniana but not to the bacterial pathogens Pseudomonas syringae and Xanthomonas oryzae. These results demonstrate a role, in both monocot and dicot plants, of hemicellulose and pectin acetylation in plant defense against fungal pathogens. [ABSTRACT FROM AUTHOR]

The hemicellulosic polysaccharide xyloglucan (XyG), found in the primary cell walls of most plant tissues, is important for structural organization of the cell wall and regulation of growth and development. Significant recent progress in structural characterization of XyGs from different plant species has shed light on the diversification of XyG during plant evolution. Also, identification of XyG biosynthetic enzymes and examination of their interactions suggests the involvement of a multiprotein complex in XyG biosynthesis. Xyloglucans are the main hemicellulosic polysaccharides found in the primary cell walls of dicots and nongraminaceous monocots, where they are thought to interact with cellulose to form a three-dimensional network that functions as the principal load-bearing structure of the primary cell wall. To determine whether two Arabidopsis thaliana genes that encode xylosyltransferases, XXT1 and XXT2, are involved in xyloglucan biosynthesis in vivo and to determine how the plant cell wall is affected by the lack of expression of XXT1, XXT2, or both, we isolated and characterized xxt1 and xxt2 single and xxt1 xxt2 double T-DNA insertion mutants. Although the xxt1 and xxt2 mutants did not have a gross morphological phenotype, they did have a slight decrease in xyloglucan content and showed slightly altered distribution patterns for xyloglucan epitopes. More interestingly, the xxt1 xxt2 double mutant had aberrant root hairs and lacked detectable xyloglucan. The reduction of xyloglucan in the xxt2 mutant and the lack of detectable xyloglucan in the xxt1 xxt2 double mutant resulted in significant changes in the mechanical properties of these plants. We conclude that XXT1 and XXT2 encode xylosyltransferases that are required for xyloglucan biosynthesis. The acquisition of freeze tolerance in winter plants involves, among other cellular responses, activated catabolism of cell wall polysaccharides, thereby liberating oligosaccharides. One of these was identified as an oligosaccharin (physiologically active fragment) that most likely originates from hemicelluloses. Treatment of winter wheat seedlings with the oligosaccharin at 2°C increased their freeze tolerance by ~30%. Results obtained to date suggest that the oligosaccharin acts as an endogenous and systemic signaling molecule during cold adaptation. [ABSTRACT FROM AUTHOR]

The polysaccharide structure and dynamics in the primary cell wall of the model grass Brachypodium distachyon are investigated for the first time using solid-state nuclear magnetic resonance (NMR). While both grass and non-grass cell walls contain cellulose as the main structural scaffold, the former contains xylan with arabinose and glucuronic acid substitutions as the main hemicellulose, with a small amount of xyloglucan (XyG) and pectins, while the latter contains XyG as the main hemicellulose and significant amounts of pectins. We labeled the Brachypodium cell wall with 13C to allow two-dimensional (2D) 13C correlation NMR experiments under magic-angle spinning. Well-resolved 2D spectra are obtained in which the 13C signals of cellulose, glucuronoarabinoxylan (GAX), and other matrix polysaccharides can be assigned. The assigned C chemical shifts indicate that there are a large number of arabinose and xylose linkages in the wall, and GAX is significantly branched at the developmental stage of 2 weeks. 2D 13C-13C correlation spectra measured with long spin diifusion mixing times indicate that the branched GAX approaches cellulose microfibrils on the nanometer scale, contrary to the conventional model in which only unbranched GAX can bind cellulose. The GAX chains are highly dynamic, with average order parameters of ∼0.4. Biexponential 13C T1 and 1H T1ρ, relaxation indicates that there are two dynamically distinct domains in GAX: the more rigid domain may be responsible for cross-linking cellulose microfibrils, while the more mobile domain may fill the interfibrillar space. This dynamic heterogeneity is more pronounced than that of the non-grass hemicellulose, XyG, suggesting that GAX adopts the mixed characteristics of XyG and pectins. Moderate differences in cellulose rigidity are observed between the Brachypodium and Arabidopsis cell walls, suggesting different effects of the matrix polysaccharides on cellulose. Abstract: Cell wall components, such as lignin, cellulose, and hemicelluloses, play an important role in the conversion efficiency of corn stover into ethanol. Understanding the molecular basis of cell wall formation is fundamental for marker assisted selection to develop lines more suitable for ethanol production. In this study, we evaluated a set of 40 European forage maize lines for cellulose, lignin, total hemicellulose, glucuronoarabinoxylan (GAX), and monosaccharides, such as arabinose (ara), xylose (xyl), and glucuronic acid (GlcA). The most significant correlations were observed between hemicelluloses and GAX (0.9), and hemicelluloses and cellulose (−0.81). Cell wall digestibility (CWD, estimated by digestible neutral detergent fiber, DNDF) was negatively correlated with Xyl (−0.34). The association analysis between the evaluated traits and polymorphisms within ten “lignin” genes revealed significant associations between polymorphisms within CCoAOMT1, CCoAOMT2, 4CL2 and C4H, and cellulose/xyl, cellulose, cellulose, and GclA, respectively. None of the QTPs identified in this study corresponded to previously reported CWD QTPs. [Copyright &y& Elsevier]
