Endostatin (ES) has attracted considerable attention for the treatment of anti-angiogenesis-related disorders. An 11-amino-acid peptide (ES2, IVRRADRAAVP) from the amino terminal of ES is of interest because it is the main active fragment of ES. However, both ES and ES2 have a poor stability and a short half-life, and other disadvantages need to be further resolved. Thus, we conjugated ES2 to glycol-split heparin derivatives (GSHPs) to yield the polymer-peptide conjugate, GSHP-ES2. This study showed that GSHP-ES2 exhibited increased stability, a wider pH activity range, better inhibition of endothelial cell proliferation, migration and tube formation in vitro, better anti-angiogenic activity and a longer half-life in vivo compared with ES2. These results also indicate that GSHP-ES2 has good potential for the treatment of angiogenesis-related diseases, either alone or in combination with other chemicals., (Copyright © 2018 Elsevier Ltd. This study reports the use of diethylenetriamine as background electrolyte for the simultaneous separation of hyaluronan acid, chondroitin sulfate, dermatan sulfate and heparin. The analytes were baseline separated by using an uncoated fused silica capillary at 37°C with a run time of 23min. The migration order, with hyaluronan acid at first and heparin at last, was related to the sulfation degree. The effect of salt concentration on resolution and migration order was also investigated. The developed method was applied to the simultaneous determination of hyaluronan acid and chondroitin sulfate in mouse plasma. Interferences in plasma were removed by protein precipitation and glycosaminoglycans were further purified by ethanol precipitation. The method was validated over the concentration range from 50 to 600μg/mL for hyaluronan acid and 500 to 6000μg/mL for chondroitin sulfate in mouse plasma. Ion-exchange chromatography is a widely used purification technology in the heparin manufacturing process. To improve the efficiency and understand the process directly, a rapid and equally precise method needs to be developed to measure heparin concentration in chromatography process. Here, two robust partial least squares regression (PLS-R) models were established for quantification of heparin based on the near-infrared (NIR) spectroscopy with 80 samples of adsorption process and 76 samples of elution process. Several variables selection algorithms, including correlation coefficient method, successive projection algorithm (SPA) and interval partial least squares (iPLSs), were performed to remove non-informative variables. The results showed that the correlation coefficient of validation (Rp) and the residual predictive deviation (RPD) corresponded to 0.957 and 3.4472 for adsorption process, 0.968 and 3.9849 for elution process, respectively. Heparin and heparosan have been confirmed to be effective blockers in inhibiting adhesion of pathogens in vitro. However, their effects on gut microbiota in vivo remain unknown. Here we have studied the effects of oral administration of heparin or heparosan on gut microbiota in rats by polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE). Results showed that the predominant bacterial communities in the feces of heparin- or heparosan-treated animals were different from those of the saline-treated animals, with increased Lactobacillus spp. and decreased Enterococcus sp. Different DGGE banding patterns were also observed for the subpopulations of Lactobacillus and Bacteroides groups. A simple and accurate capillary electrophoresis (CE) method was developed to simultaneously separate and quantify heparin, chondroitin sulfate and hyaluronic acid. The relative standard deviations (intra-day) of migration time, peak height and peak area for heparin, chondroitin sulfate and hyaluronic acid were lower than 1.11, 5.45 and 2.82%, respectively. The limits of detection of heparin, chondroitin sulfate and hyaluronic acid were 0.91, 0.12 and 9.04 × 10(-3) mg/mL, respectively. The developed electrophoretic method was successfully applied to the analysis of commercial drug products and biological samples containing chondroitin sulfate and/or hyaluronic acid. The recoveries for chondroitin sulfate and hyaluronic acid were in the range of 95.9 ~107.0%. This was the first time the content of hyaluronic acid in the synovial fluids from osteoarthritic rabbits was investigated by CE. The results suggested that hyaluronic acid in the synovial fluids from osteoarthritic rabbits may be further metabolized and the administration of chondroitin sulfate or hyaluronic acid could affect the content and metabolism of hyaluronic acid in the synovial fluids. Potency is an important parameter for evaluation of quality of heparin active pharmaceutical ingredient (API). In this paper the feasibility to determine potency of heparin API with near infrared reflectance spectroscopy (NIRS) coupled with partial least squares (PLS) algorithm is attempted. PLS factors, correlation coefficient of calibration set (R(c)), the root mean square of cross-validation (RMSECV), correlation coefficient of prediction set (R(p)) and the root mean square of prediction (RMSEP) were used to evaluate the performance of the models. The optimal calibration model was obtained with R(p)=0.9721 and RMSEP=0.55 in the 1700-1898nm spectral region when using SG-1st derivative spectral transform method and division of calibration/prediction samples was 1/1. Three other additional samples demonstrated good prediction capability of the final model and three validation samples gave good repeatability result. In this study, the effects of heparin-superoxide dismutase conjugate (heparin-SOD) on bleomycin-induced pulmonary fibrosis in vivo and on inflammatory cytokine expression in vitro were evaluated. To investigate the effects of heparin-SOD on pulmonary fibrosis, heparin-SOD was administered to bleomycin (BLM)-treated mice by intraperitoneal injection once a day and the hydroxyproline content in lung was determined per 7days. The degree of fibrosis was assessed quantitatively using histopathologic features. The results showed that heparin-SOD inhibited BLM-induced lung fibrotic lesions as reflected by the decrease of lung hydroxyproline content and lung fibrosis grade 28days after BLM instillation. The in vitro effects on the cytokine level expressed by irradiated 3T3 fibroblasts showed that heparin-SOD significantly lowered the levels of transforming growth factor-beta1 and interleukin-1beta. These results strongly demonstrated that heparin-SOD might be useful in the prevention and treatment of pulmonary fibrosis.

In this study, the effects of heparin-superoxide dismutase conjugate (heparin-SOD) on carbon tetrachloride (CCl4)-induced acute liver failure and hepatic fibrosis were evaluated. To investigate the effects of heparin-SOD on acute liver failure, heparin-SOD was administered to CCl4-treated mice by intravenous injection. Biochemical indicators, such as glutamic oxaloacetic transaminase/glutamic pyruvic transaminase (GOT/GPT), GSH (glutathione), lactate dehydrogenase (LDH), and malondialdehyde (MDA) were determined 24 h after CCl4 treatment. The development of CCl4-induced acute liver failure altered the redox state with a decreased hepatic GSH and increased formation of lipid peroxidative products, which were partially normalized by treatment with heparin-SOD or heparin + SOD. Compared with other groups, the acute liver injury of heparin-SOD group was significantly lessened (reduced activities of GOT/GPT, MDA, and increased activities of GSH). To investigate the effects of heparin-SOD on hepatic fibrosis, heparin-SOD and CCl4 were co-administered by intraperitoneal injection twice a week for 12 weeks. Histological and hepatic hydroxyproline examination revealed that heparin-SOD could significantly prevent the progression of hepatic fibrosis. Moreover, real-time PCR was used to determine transforming growth factor-beta1 (TGF-beta1), metalloproteinase-2 (MMP-2), fibronectin, and collagen-I expression. Significantly, greater fibrosis and TGF-beta1, MMP-2, fibronectin, and collagen-I expression were found in the liver of CCl4-induced mice at the end of 12th week. Heparin-SOD could markedly attenuate the mRNA expression of TGF-beta1, MMP-2, and collagen-I. Western blots of tissue homogenates revealed that the protein expression of TGF-beta1 was substantially reduce also by heparin-SOD treatment. These results demonstrate that administration of heparin-SOD may be useful in the treatment and prevention of acute liver failure and hepatic fibrosis.

To improve the ability of superoxide dismutase (SOD) to suppress reactive oxygen species (ROS)-mediated injury, chemically modified derivatives of SOD with N,N,N-trimethyl chitosan chloride (TMC) and heparin, cationized SOD (TMC-SOD), and anionized SOD (heparin-SOD) were designed and prepared. In this study, the inhibitory effect of TMC-SOD and heparin-SOD on superoxide anion release from macrophages was studied in vitro. Both TMC-SOD and heparin-SOD exhibited excellent inhibitory effects on superoxide anion release from macrophages, and the effects of TMC-SOD surpassed those of native SOD and heparin-SOD. The effects of TMC-SOD and heparin-SOD on inflammatory cytokine expression in vitro were also evaluated. The results showed that both TMC-SOD and heparin-SOD could significantly lower the levels of transforming growth factor-beta1 (TGF-beta1) and interleukine-1beta (IL-1beta) expressed by irradiated 3T3 fibroblasts. These results demonstrated that cationic polysaccharide or anionic polysaccharide SOD derivatives might be useful in the prevention and treatment of ROS-mediated inflammatory diseases. This study also demonstrated that chemical modification of SOD, especially cationization, greatly enhanced SOD's intracellular delivery and, consequently, produced a significant protective effect against ROS-mediated injury.

Heparan sulfate ( HS) is a highly sulfated glycosaminoglycan and exists in all animal tissues. HS and heparin are very similar, except that heparin has higher level of sulfation and higher content of iduronic acid. Despite the fact that it is a century-old drug, heparin remains as a top choice for treating thrombotic disorders. Pharmaceutical heparin is derived from porcine intestine or bovine lung via a long supply chain. This supply chain is vulnerable to the contamination of animal pathogens. Therefore, new methods for manufacturing heparin or heparin-like substances devoid of animal tissues have been explored by many researchers, among which, modifications of heparosan, the capsular polysaccharide of Escherichia coli K5 strain, is one of the promising approaches. Heparosan has a structure similar to unmodified backbone of natural HS and heparin. It is feasible to obtain HS or heparin derivatives by modifying heparosan with chemical or enzymatic methods. These derivatives display different biological activities, such as anticoagulant, anti-inflammatory, anticancer, and antiviral activities. This review focuses on the recent studies of synthesis, activity, and structure-activity relationship of HS/heparin-like derivatives prepared from heparosan. [ABSTRACT FROM AUTHOR]

Abstract: Endostatin (ES), a potent endogenous angiogenesis inhibitor found in 1997 by O’Reilly, can effectively inhibit angiogenesis, inhibit the growth and metastasis of tumors. ES can also decrease drug resistance in long term and repeated treatment when it is used in combination with other chemotherapeutic agents. But there are still lots of obstacles on its clinical application, such as the need of a high dose to maintain its efficacy short half-life, poor stability, expensive, and some other shortcomings just like other protein drugs. Chemical modification on ES by polyethylene glycol (PEG) and low molecular weight heparin (LMWH) were successfully carried out in order to obtain a better ES derivative. Several classic experimental models were employed to study the bioactivity of ES and modified ES, including chicken chorioallantoic membrane (CAM) assay, corneal neovascularization (CNV) assay and Sarcoma 180 tumor bearing mice assay. The results showed that PEG-ES and LMWH-ES had better anti-angiogenesis and anti-tumor activity than ES, which indicates that LMWH was also a good protein modifier. [Copyright &y& Elsevier]

Abstract: Low-molecular-weight heparin (LMWH) and Cu,Zn-superoxide dismutase (SOD) were extensively investigated on preventing brain reperfusion injury after ischemia (BRII) in the past few years and both exhibited some advantages as well as limits in practice. To explore whether chemical modification for LMWH and SOD can lead to improved bioactivity, in our present study, we examined the efficacy of LMWH conjugated SOD (LMWH−SOD) in the model of BRII gerbils. Ischemia/reperfusion was performed for 5 min by clamping the bilateral common carotid arteries of gerbils. LMWH−SOD, SOD and the mixture of LMWH and SOD (LMWH+SOD) were administered intravenously to corresponding animals just before ischemia. 24 h after reperfusion, serum malondialdehyde (MDA) content and SOD activity were measured, the expression of intercellular adhesion molecule-1 (ICAM-1) was examined by immunohistochemistry, and the brain sections were processed for Nissl staining and terminal deoxynucleotidyl transferase (TdT)-mediated dUTP nick end-labeling. The results showed that LMWH−SOD significantly lowered MDA content (P <0.001, versus SOD and LMWH+SOD) and elevated SOD activity (P <0.05, versus SOD and LMWH+SOD) in the serum of BRII gerbils. Immunohistochemical results indicated ICAM-1 positive staining was lighter, pyramidal cells of hippocampal CA1 region were more regular and the changes in cell edema were minor, and apoptosis of hippocampal cells was milder in LMWH−SOD treated animals than in SOD or LMWH+SOD treated animals, untreated BRII animals and sham-operated animals. The results suggest that the novel LMWH−SOD conjugate can inhibit upregulation of ICAM-1 and prevent neuronal cell apoptosis in BRII gerbils, and the LMWH−SOD conjugate has better anti-inflammatory and neuroprotective effects in BRII than native SOD and the mixture of LMWH and SOD.
