Organic contaminants, and herbicides in particular, represent a risk for aquatic ecosystems. The primary target of herbicides are producers, the base of food webs, but frequently they end up far from the application point affecting non-target species. Its presence can work as sub-lethal stimulus, which sort the genetic and phenotypic differences within a species. Intraspecific variation allows adaptation to changes in the environment but also to new niches due to variations in species' sensitivity and biochemical response to a certain chemical. A better understanding of these variations can lead to the development of improved strategies for ecosystem protection. This research aimed to compare a sensitive and a tolerant strain of the freshwater diatom Nitzschia palea to atrazine. Strains were exposed to three concentrations within their tolerance range, during 96 h. The activity of the antioxidant enzymes superoxide dismutase, catalase, glutathione-S-transferases and glutathione peroxidases was determined. In addition, chlorophylls a and c, carotenoids, reduced glutathione, proteins and lipid peroxidation were quantified. Both strains displayed different strategies to deal with atrazine toxicity: while the sensitive strain decreased the oxidative stress, increasing the activity of antioxidant enzymes such as superoxide dismutase, the tolerant strain invested in conjugation pathways and carotenoids' maintenance.

Diatoms are used as indicators of freshwater ecosystems integrity. Developing diatom-based tools to assess impact of herbicide pollution is expected by water managers. But, defining sensitivities of all species to multiple herbicides would be unattainable. The existence of a phylogenetic signal of herbicide sensitivity was shown among diatoms and should enable prediction of new species sensitivity. However, diatoms present a cryptic diversity that may lead to variation in their sensitivity to herbicides that would need to be taken into account. Using bioassays, the sensitivity to four herbicides (Atrazine, Terbutryn, Diuron, Isoproturon) was evaluated for 11 freshwater diatom taxa and intraspecific variability was assessed for two of them (Nitzschia palea and Achnanthidium spp.). Intraspecific variability of herbicide sensitivity was always smaller than interspecific variability, but intraspecific variability was more important in N. palea than in Achnanthidium spp. Indeed, one species showed no intraspecific phylogenetic signal (N. palea) whereas the other did (Achnanthidium spp.). On one hand, species boundaries are not set properly for Achnanthidium spp. which encompass several taxa. On the other hand, there is a higher phenotypic plasticity for N. palea. Finally, a phylogenetic signal of herbicide sensitivity was measured at the interspecific level, opening up prospects for setting up reliable biomonitoring tools based on sensitivity prediction, insofar as species boundaries are correctly defined.

Chemical monitoring revealed a regular decrease in herbicide concentration in Lake Geneva since last decades that may be linked to an ecotoxic restoration of nontarget phytoplanktonic communities. The Pollution-induced community tolerance (PICT) approach was tested as a tool to monitor the ecotoxic restoration of Lake Geneva for herbicides from 1999 to 2011. We conducted monthly assessments in 1999 and in 2011 for the tolerance of the phytoplankton communities to two herbicides (atrazine and copper), using PICT bioassays. The taxonomical composition of the communities was determined on the same collecting dates. The herbicide concentration decrease during the 12 years significantly influenced the composition of communities. The PICT monitoring indicated that a significant tolerance decrease in the community to both herbicides accompanied the herbicide concentration decrease. PICT measurements for atrazine and copper also changed at the intra-annual level. These variations were mainly due to community composition shifts linked to seasonal phosphorus and temperature changes. PICT monitoring on a seasonal basis is required to monitor the mean tolerance of communities. PICT appeared to be a powerful tool that reflected the toxic effects on environmental communities and to monitor ecotoxic ecosystem restoration.

Seasonal changes in the structure and composition of a benthic microalgal community may lead to different responses to herbicide contamination during different seasons. Consequently, the thresholds derived from risk assessment tools such as species sensitivity distributions (SSDs) must allow for these changes. We built a single-substance SSD for each of four herbicides (atrazine, terbutryn, diuron and isoproturon), which was specific to the sensitivity of the benthic diatoms found in Lake Geneva, in order to derive protective thresholds for a mixture of these four herbicides using the concentration addition model. We then investigated (1) the structural parameters of a Lake Geneva benthic microalgal community during two contrasting seasons (summer 2012 and winter 2013), (2) the response of these communities to a herbicide mixture, and (3) the protective levels of the thresholds derived. The winter community was characterized by having greater biomass, diatom species richness, and diversity metrics, and lower non-diatom species richness than the summer community. The differences in the diatom communities composition in these seasons appeared to be primarily driven by the environmental nitrate concentrations and the temperature. Moreover, the species in the winter community were more resistant to herbicides than those found in the summer community. Consequently, the protective threshold for this herbicide mixture obtained in this study was in fact protective for the winter community, but not for the summer community based on their structural parameters. Thus, the protective level against herbicides of the threshold for the benthic microalgal community should take into account changes in the environmental physico-chemical conditions that strongly influence the structure and composition of the community. The fact that the succession of species over time (i.e., over the seasons) is difficult to predict introduces uncertainties into the estimation of protective thresholds and questions their applicability year round.

Phylogeny has not yet been fully accepted in the field of ecotoxicology, despite studies demonstrating its potential for developing environmental biomonitoring tools, as it can provide an a priori assessment of the sensitivity of several indicator organisms. We therefore investigated the relationship between phylogeny and sensitivity to herbicides in freshwater diatom species. This study was performed on four photosystem II inhibitor herbicides (atrazine, terbutryn, diuron, and isoproturon) and 14 diatom species representative of Lake Geneva biofilm diversity. Using recent statistical tools provided by phylogenetics, we observed a strong phylogenetic signal for diatom sensitivity to herbicides. There was a major division in sensitivity to herbicides within the phylogenetic tree. The most sensitive species were mainly centrics and araphid diatoms (in this study, Thalassiosirales and Fragilariales), whereas the most resistant species were mainly pennates (in this study, Cymbellales, Naviculales, and Bacillariales). However, there was considerable variability in diatom sensitivity within the raphid clade, which could be explained by differences in trophic preferences (autotrophy or heterotrophy). These traits appeared to be complementary in explaining the differences in sensitivity observed at a refined phylogenetic level. Using phylogeny together with complementary traits, as trophic preferences, may help to predict the sensitivity of communities with a view to protecting their ecosystem.

Temperature is a strong driver of biofilm formation and of the dynamics of microalgae in freshwater. Moreover, exposure to herbicides is a well-known stressor of periphytic communities in anthropized aquatic environments. We tested these two environmental factors on periphytic communities that had been sampled from the littoral zone of Lake Geneva and acclimatized in the lab for 3 weeks at 18, 21, 24 and 28 °C. After this acclimation period, differences in the composition of the diatom community and decreases in cell density were observed corresponding to the temperature gradient. These acclimated communities were then exposed to 23 and 140 nM of a mixture composed of equitoxic quantities of atrazine, terbutryn, diuron and isoproturon. The periphytic community was more sensitive to the herbicide mixture at 18 °C than at higher temperatures, suggesting that higher temperature reduced its toxicity. Small and pioneer diatom species known to be promoted by contamination also appeared to benefit from higher temperatures. Mixture risk assessment predictions have rarely been confronted with biological changes observed in the environment. In this study, long-term monitoring of a European great lake, Lake Geneva, provides the opportunity to assess to what extent the predicted toxicity of herbicide mixtures explains the changes in the composition of the phytoplankton community next to other classical limnology parameters such as nutrients. To reach this goal, the gradient of the mixture toxicity of 14 herbicides regularly detected in the lake was calculated using concentration addition and response addition models. A temporal gradient of toxicity was observed which decreased from 2004 to 2009. Redundancy analysis and partial redundancy analysis showed that this gradient explains a significant portion of the variation in phytoplankton community composition with and without having removed the effect of all other co-variables. Moreover, species that are significantly influenced, positively or negatively, by the decrease of toxicity in the lake over time are highlighted. It can be concluded that the herbicide mixture toxicity is one of the key parameters to explain phytoplankton changes in Lake Geneva.

Although benthic diatoms are widely used in ecological studies of aquatic systems, there is still a dearth of data concerning species sensitivities towards several contaminants. Within the same community, different species may respond differently depending on their physiological and ecological characteristics. This lack of knowledge makes specific appropriate risk assessment impossible. To find out whether species sensitivity distribution (SSD) could be used to estimate the risk of herbicide toxicity for diatoms, we need to know whether their sensitivity depends on their physiological and ecological characteristics. We carried out single-species bioassays on 11 diatom species exposed to 8 herbicides. Dose-responses relationships were used to extrapolate the Effective Concentration 5 (EC(5)) and the Effective Concentration 50 (EC(50)) for each exposure. These data were used to fit a SSD curve for each herbicide, and to determine the Hazardous concentration 5 (HC(5)) and 50 (HC(50)). Our results revealed a high level of variability of the sensitivity in the set of species tested. For photosystem-II inhibitor (PSII) herbicides, diatoms species displayed a typical grouping of sensitivity levels consistent with their trophic mode and their ecological guild. N-heterotroph and "motile" guild species were more tolerant of PSII inhibitors, while N-autotroph and "low profile" guild species were more sensitive. Comprehensive SSD curves were obtained for 5 herbicides, but not for sulfonylurea herbicides or for dimetachlor, which had toxicity levels that were below the range of concentration tested. The SSD curves provided the following ranking of toxicity: diuron> terbutryn> isoproturon> atrazine> metolachlor. The HC that affected 5% of the species revealed that, even at the usual environmental concentrations of herbicides, diatom assemblages could be affected, especially by isoproturon, terbutryn, and diuron.

The potential of microbenthic algal assemblages to recover after diuron exposure was investigated. Microbenthic algal assemblages (periphyton) were grown on glass slides in correspondence to a diuron-polluted and adiuron-free sampling site of a river. After 5 weeks of colonization, the impacted periphyton was transferred by translocating the colonized glass slides to the unpolluted site. To monitor the changes in functional and structural parameters and to assess recovery, both the transferred and the local reference periphyton were sampled at the day of transfer (t0) and 1, 3, and 5 weeks after the transfer (t1, t3,t5). Structural transitions of eukaryotic communities were characterized by denaturing gradient gel electrophoresis (DGGE) and functional ones were characterized by short-term incubation toxicity tests with diuron. As shown by Bray–Curtis similarity values based on DGGE band patterns, almost total structural recovery of the transferred periphyton took place 5 weeks after transfer. For the transferred periphyton, previous diuron exposure at the contaminated site induced the development of diuron tolerance,which decreased after 1 week and became very similar to the tolerance of the nonimpacted community after 5 weeks. It is concluded that 5-week-old eukaryotic periphyton communities are capable of restoring their structural and functional attributes after 5 weeks within a noncontaminated environment.

Recovery of bacterial and eukaryotic communities in biofilms naturally grown on stones was studied for 9 weeks after transferring them from a pesticide polluted downstream site of the river Morcille (Beaujolais, France) to a non-contaminated upstream site. Site-specific periphyton present on stones at both the down- and the upstream sampling site were collected to analyze the site-specific colonization. Throughout the experiment, structural and functional parameters were analyzed for the periphyton transferred and for the site-specific up- and downstream periphyton. Comparison between these three communities allowed quantifying recovery of the transferred one. Denaturing Gradient Gel Electrophoresis of PCR-amplified 16S and 18S rRNA gene fragments were used to assess prokaryotic and eukaryotic microbial community composition, respectively. Microscopy counts allowed characterizing the diatom taxa abundances. The sensitivity of the microalgal communities towards diuron and copper was investigated at the laboratory by short-term photosynthesis inhibition assays. The functional reaction of the bacterial communities towards copper was assessed by short-term respiration inhibition assays. The structure of transferred eukaryotic, bacterial and diatom communities was more similar to the structure of the downstream communities than to upstream ones even after 9 weeks acclimatization in particular for the bacterial community. In the same way, the community tolerance towards diuron and copper, as estimated by the EC50 values, was intermediate for the transferred biofilms compared to the local up- or downstream biofilm, even after 9 weeks of acclimatization. These results strongly suggest slow recovery, likely to be linked to long lasting exposure of pesticides and in particular copper adsorbed to the biofilm matrices and to the toughness for pioneer microorganisms to invade mature biofilms., (Copyright (c) 2010 Elsevier B.V. We have tested the sensitivity of phytoplankton to the herbicides atrazine and nicosulfuron in experiments conduced in increasingly complex systems, from single strain phytoplankton cultures (microplates) to mesocosms mimicking whole ecosystems. The endpoints used to assess sensitivity to atrazine and nicosulfuron were total biomass increase, photosynthetic efficiency, and community diversity, depending on the system considered. Nicosulfuron appeared to be very much less toxic to phytoplankton than atrazine, in accord with the planned changes in agricultural practices to reduce the effects of surface water contamination on aquatic biota. Nevertheless, nicosulfuron had significant effects in some systems (principally microcosms), whereas the single monocultures were almost insensitive to it. This points out the inaccuracy of standardized toxicity test on phytoplanktonic algae alone for predicting the effects of xenobiotics on natural communities and the need for tests in microcosms and mesocosms to obtain reliable evidence about the toxicity of a given chemical on freshwater aquatic ecosystems.

Growth modifications caused by various concentrations of atrazine and nicosulfuron were monitored in closed and continuous culture of Chlorella vulgaris (chlorophyta), Navicula accommoda (diatomophyta), and Oscillatoria limnetica (cyanophyta). The concentration at which algal growth rate was reduced twofold (EC50) was determined in the three species for both herbicides. Comparatively, the two toxicants were applied at 10 microg/l level in microcosms inoculated with natural phytoplankton from Lake Geneva. The relative abundances of major phytoplanktonic species were measured by algal cell count at the beginning and at the end of each experiment. Atrazine and nicosulfuron have different targets in plant metabolism, respectively, photosystem II (PSII) and acetolactate synthase (ALS), and the expected effects were different. Generally, the cultured phytoplankton exhibited various sensitivities, depending on species or herbicide. In the microcosms, the major taxa of natural phytoplanktonic samples exhibited various patterns, from acute toxicity to growth enhancement. For example, the diatoms inside the community were not affected by atrazine and nicosulfuron, except for Stephanodiscus minutulus that was sensitive to both, and Asterionella/formosa that was sensitive only to nicosulfuron. The specific physiology and the relationships among the phytoplanktonic communities have to be carefully considered when one would try to predict the extent of herbicide action on natural phytoplankton using in vitro tests. There is a need to test the toxic effect on various cultured strains, representative of most of the taxonomic composition of natural communities, to take into account the wide range of sensitivities and reaction to herbicide contamination. But this is not enough to give a solid frame when transposing the results to the field, and the use of more ecologically relevant systems is recommended.
