Anti-flt1 peptide (GNQWFI, AF) specifically binds to Vascular Endothelial Growth Factor Receptor 1 (VEGFR1), thereby inhibiting the interaction of VEGFR1 with a series of ligands. ES2 (IVRRADRAAVP) can effectively inhibit the proliferation and invasion of endothelial cells and play a key role in anti-angiogenesis. AF and ES2 peptides differ in their activity. To better exploit the advantages of both, we designed a new peptide called ES2-AF (IVRRADRAAVPGGGGGGNQWFI). Hyaluronic acid (HA) is widely used in the pharmaceutical industry because of its biodegradable and high load performance. The HA-specific cell surface receptor CD44 was highly expressed in the tumour site during the anti-tumour study. Therefore, we used HA as a modifier to chemically modify ES2-AF; it was expected that the modified compound would have preferable solubility, stronger targeting, longer half-life, and better anti-angiogenesis effects in vivo. In this study, the anti-proliferative, anti-migration and targeting activities of HA-ES2-AF in vitro were studied by MTT, ELISA, transwell and SPR assays. Meanwhile, the anti-neovascularization activity of HA-ES2-AF in vivo was studied by CAM assay, and the targeting of HA-ES2-AF to tumour tissue was studied by bioimaging techniques. Finally, we also studied the half-life of HA-ES2-AF in vivo. In short, the bioactivity of the new peptide ES2-AF was enhanced to a certain extent, and ES2-AF modified by HA had higher anti-neovascularization activity in vitro and in vivo, had stronger targeting to tumour tissue, and had a significantly prolonged half-life in vivo. This study reports the use of diethylenetriamine as background electrolyte for the simultaneous separation of hyaluronan acid, chondroitin sulfate, dermatan sulfate and heparin. The analytes were baseline separated by using an uncoated fused silica capillary at 37°C with a run time of 23min. The migration order, with hyaluronan acid at first and heparin at last, was related to the sulfation degree. The effect of salt concentration on resolution and migration order was also investigated. The developed method was applied to the simultaneous determination of hyaluronan acid and chondroitin sulfate in mouse plasma. Interferences in plasma were removed by protein precipitation and glycosaminoglycans were further purified by ethanol precipitation. The method was validated over the concentration range from 50 to 600μg/mL for hyaluronan acid and 500 to 6000μg/mL for chondroitin sulfate in mouse plasma. Objective: To compare the cytotoxicities and efficacy of hyaluronan (HA), carbomer, and sodium alginate on repairing thermal-injured cells and promoting cell migration., Design: The 3-(4,5)-dimethylthiahiazo(-z-y1)-3,5-di-phenytetra-zoliumromide method was used to evaluate the cytotoxicities of HA, carbomer, and sodium alginate on L929 mouse fibroblasts and their repairing ability to thermal-injured HaCaT keratinocytes. A scratch test was used to observe the effects of the 3 materials on cell migration., Results: Hyaluronan with different molecular weights were nontoxic, even at the concentration of 0.5%, whereas carbomer and sodium alginate showed mild or moderate cytotoxicities when their concentrations were higher than 0.1%. Cell viability and cell density of the thermal-injured keratinocytes treated with HA (600, 1070, and 1500 kDa) were increased significantly compared with that of model control (P < .05), whereas carbomer aggravated cell injury, and sodium alginate had no obvious repairing ability. Hyaluronan promoted cell migration significantly with higher cell density in the scratch area, compared with the control after culture for 48 hours; both carbomer and sodium alginate inhibited the cell migration, and carbomer altered the cell morphology completely., Conclusions: Hyaluronan can repair cell injury and promote cell migration and proliferation. It also has good biocompatibility. As a new type of hydrogel matrix, HA is superior to carbomer and sodium alginate if it is used in wound caring preparations.

Hyaluronic acid (HA), a natural high molecular weight polysaccharide, is produced by Streptococcus zooepidemicus. However, Streptococcus has several drawbacks including its potential to produce exotoxins, so there is demand for an alternative HA source. Here, a recombinant HA biosynthesis operon, as well as the HA biosynthesis operon of S. zooepidemicus were introduced into L. lactis using the nisin-controlled expression system, respectively. HA was successfully synthesized by recombinant L. lactis. Furthermore, overexpression of the endogenous enzymes directing the synthesis of precursor sugars was effective at increasing HA production, and increasing the supply of UDP-activated monosaccharide donors aided synthesis of monodisperse HA polysaccharides. Besides GRAS host strain (L. lactis) and NICE system, the selecting marker (lacF gene) of the recombinant strain is also food grade. Therefore, HA produced by recombinant L. lactis overcomes the problems associated with Streptococcus and provides a source of food-grading HA appropriate for widespread biotechnological applications.

A simple and accurate capillary electrophoresis (CE) method was developed to simultaneously separate and quantify heparin, chondroitin sulfate and hyaluronic acid. The relative standard deviations (intra-day) of migration time, peak height and peak area for heparin, chondroitin sulfate and hyaluronic acid were lower than 1.11, 5.45 and 2.82%, respectively. The limits of detection of heparin, chondroitin sulfate and hyaluronic acid were 0.91, 0.12 and 9.04 × 10(-3) mg/mL, respectively. The developed electrophoretic method was successfully applied to the analysis of commercial drug products and biological samples containing chondroitin sulfate and/or hyaluronic acid. The recoveries for chondroitin sulfate and hyaluronic acid were in the range of 95.9 ~107.0%. This was the first time the content of hyaluronic acid in the synovial fluids from osteoarthritic rabbits was investigated by CE. The results suggested that hyaluronic acid in the synovial fluids from osteoarthritic rabbits may be further metabolized and the administration of chondroitin sulfate or hyaluronic acid could affect the content and metabolism of hyaluronic acid in the synovial fluids. Aims: Oligosaccharides of hyaluronan (o-HAs) were proved to have pro-angiogenic activities. The aim of this study was to obtain four hyaluronan oligosaccharides (o-HAs) of defined molecular weight including 4 saccharide residues (o-HA4), 6 saccharide residues (o-HA6), 8 saccharide residues (o-HA8), and 10 saccharide residues (o-HA10) under optimum conditions, and compare their angiogenic activities., Main Methods: The four o-HAs were prepared by digesting the native high molecular weight HA with hyaluronidase under optimum conditions. The effects of the four o-HAs on human umbilical vein endothelial cell (HUVEC) proliferation were measured using 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenil tetrazolium bromide (MTT) assay. Angiogenic effects of the four o-HAs were evaluated using chicken chorioallantoic membrane (CAM) assay. The effects of the four o-HAs on the vascular endothelial growth factor (VEGF) and basic fibroblast growth factor (bFGF) messenger ribonucleic acid (mRNA) expressions were detected by semi-quantitative polymerase chain reaction (PCR)., Key Findings: O-HA6, o-HA8 and o-HA10 could effectively promote the proliferation of HUVEC, induce angiogenesis in the CAM assay and increase VEGF mRNA levels in HUVEC. No significant effects were found with o-HA4., Significance: These results suggested that o-HA6, o-HA8, and o-HA10, but not o-HA4 were effective angiogenic factors.

As the interaction of hyaluronan (HA) with its receptor CD44 contributes to multidrug resistance (MDR) of tumor cells, HA oligosaccharides (o-HAs), as HA antagonists, may be useful to reverse the MDR. The objective of this study was to investigate the reversal effects of four o-HAs, including 4 saccharide residue (o-HA4), 6 saccharide residue (o-HA6), 8 saccharide residue (o-HA8), and 10 saccharide residue (o-HA10) fragments, on adriamycin (ADR)-resistant K562/A02 cells. The four o-HAs were prepared by digesting the native high molecular weight HA with hyaluronidase and gel filtration chromatography. 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyl tetrazolium bromide (MTT) assay was used to assess the cytotoxicity of the four o-HAs and/or ADR on K562/A02 and K562 cells. The intracellular accumulation of ADR in K562/A02 cells was measured by flow cytometry. By comparing the IC50 (concentration resulting in 50% inhibition of cell growth) of ADR with K562/A02 cells in the presence and absence of a series of different concentrations of o-HAs, the reversal folds of the four o-HAs were calculated. The reversal folds of o-HA4, o-HA6, o-HA8, and o-HA10 were 2.04, 2.05, 1.91, and 1.84, respectively. After o-HA4, o-HA6, o-HA8, and o-HA10 treatment, the intracellular amounts of ADR were increased to 3.90, 3.92, 3.76, and 3.39 times, respectively. Shorter o-HAs (o-HA4 and o-HA6) showed stronger reversal effects than longer o-HAs (o-HA8 and o-HA10). In conclusion, the results showed that the four o-HAs could effectively reverse the ADR resistance of K562/A02 cells by increasing the intracellular accumulation of ADR. O-HAs may be used as MDR reversal drugs to increase the effectiveness of chemotherapy.

Hyaluronidases are a family of enzymes that degrade hyaluronic acid (hyaluronan, HA) and widely used in many fields. A hyaluronidase producing bacteria strain was screened from the air. 16S ribosomal DNA (16S rDNA) analysis indicated that the strain belonged to the genus Bacillus, and the strain was named as Bacillus sp. A50. This is the first report of a hyaluronidase from Bacillus, which yields unsaturated oligosaccharides as product like other microbial hyaluronate lyases. Under optimized conditions, the yield of hyaluronidase from Bacillus sp. A50 could reach up to 1.5×104 U/mL, suggesting that strain A50 is a good producer of hyaluronidase. The hyaluronidase (HAase-B) was isolated and purified from the bacterial culture, with a specific activity of 1.02×106 U/mg protein and a yield of 25.38%. The optimal temperature and pH of HAase-B were 44°C and pH 6.5, respectively. It was stable at pH 5–6 and at a temperature lower than 45°C. The enzymatic activity could be enhanced by Ca2+, Mg2+, or Ni2+, and inhibited by Zn2+, Cu2+, EDTA, ethylene glycol tetraacetic acid (EGTA), deferoxamine mesylate salt (DFO), triton X-100, Tween 80, or SDS at different levels. Kinetic measurements of HAase-B towards HA gave a Michaelis constant (Km) of 0.02 mg/mL, and a maximum velocity (Vmax) of 0.27 A232/min. HAase-B also showed activity towards chondroitin sulfate A (CSA) with the kinetic parameters, Km and Vmax, 12.30 mg/mL and 0.20 A232/min respectively. Meanwhile, according to the sequences of genomic DNA and HAase-B’s part peptides, a 3,324-bp gene encoding HAase-B was obtained. [ABSTRACT FROM AUTHOR]
