Bacterial infected wounds bring an economic burden to the worldwide medical care field. A variety of bioactives-integrated hydrogel patches are developed in response to this challenge. Here, the melanin hydrogel inverse opal microneedle patches (MNs) with antioxidant and visual color sensing abilities for the management of bacterial infected wounds are proposed. The MNs are fabricated by applying melanin-loaded polyethylene glycol diacrylate (PEGDA) as the inverse opal hydrogel and using bacitracin-carried gelatin to fill those nanopores of hydrogel scaffold. Benefitting from the antioxidant capacity of melanin nanoparticles and the local antimicrobial ability of bacitracin, the resulting MNs possess the integrated functions of reactive oxygen species scavenging and antibacterial. Besides, the inverse opal structure endows the MNs with vivid structure color and detectable reflected wavelength, which can gradually shift with the release of the drug, thus allowing MNs to assess the drug delivery. Based on these characteristics, MNs perform excellent in in vitro drug delivery and monitoring, as well as the promotion of bacterial infected wound recovery in vivo, indicating the potential of MNs in the future wound management field., (© 2024 Wiley‐VCH GmbH.)

Hydrogel-derived implants have proven value in bone tissue regeneration, and current efforts have concentrated on devising strategies for producing functional implants with desired structures and functions to improve therapeutic outcomes. Herein, a novel black phosphorus (BP) tagged responsive strontium (Sr) hydrogel particles are presented for bone defect repair. By applying microfluidic technology, Sr and carboxymethyl chitosan, and BP are integrated into poly(N-isopropyl acrylamide) (pNIPAM) hydrogel matrix to generate such microparticles called pNBCSMs. Upon exposure to near-infrared irradiation, the pNBCSMs experience volume shrinkage and provoke the extrusion of the incorporated Sr, ascribed to the photothermal conversion ability of BP and the thermosensitivity of pNIPAM. In vitro and in vivo experimental results reveal that pNBCSMs subjected to near-infrared light display superior anti-inflammatory, anti-apoptotic, bacterial inhibitory, as well as osteogenesis-promoting effects, thereby effectively improving defective cranial bone repair. Constructing hydrogels with spatially heterogeneous structures are crucial for unlocking novel applications. To this end, selectively removing a specific portion of hydrogels by facile and intricate destructive strategies is worth exploring. Herein, a "contact-destructive" hydrogel actuator is presented, composed of a dynamic hydrogel network doped with hydrophilic polyethylene glycol (PEG). The destructive behavior of the hydrogel actuator is attributed to the surface tension-induced spreading effect and the enhanced water absorption due to the additive PEG. Parameters that act on these mechanisms are used to control the destruction of the hydrogel. During the destructive process, the hydrogel actuator exhibits locomotion routes predetermined by the graphic pattern with the aid of 3D printing. Additionally, such self-destructive behavior can be terminated by UV light irradiation when PEG is replaced with poly(ethylene glycol) diacrylate (PEGDA). Significantly, diverse applications including controllable 3D structures collapse, self-erasing, and on-demand cell release, are realized with such self-destructive hydrogel. Several natural Chinese herbal medicines have demonstrated considerable potential in facilitating wound healing, while the primary concern remains centered around optimizing formulation and structure to maximize their efficacy. To address this, a natural microneedles drug delivery system is proposed that harnesses gelatinized starch and key Chinese herbal ingredients-aloe vera and berberine. After gelatinized and aged in a well-designed mold, the starch-based microneedles are fabricated with suitable mechanical strength to load components. The resulting Chinese herbal hydrogel microneedles, enriched with integrated berberine and aloe, exhibit antibacterial, anti-inflammatory, and fibroblast growth-promoting properties, thereby facilitating wound healing in the whole process. In vivo experimental results underscore the notable achievements of the microneedles in early-stage antibacterial effects and subsequent tissue reconstruction, contributing significantly to the overall wound healing process. With the accelerated aging tendency, osteoarthritis (OA) has become an intractable global public health challenge. Stem cells and their derivative exosome (Exo) have shown great potential in OA treatment. Research in this area tends to develop functional microcarriers for stem cell and Exo delivery to improve the therapeutic effect. Herein, we develop a novel system of Exo-encapsulated stem cell-recruitment hydrogel microcarriers from liquid nitrogen-assisted microfluidic electrospray for OA treatment. Benefited from the advanced droplet generation capability of microfluidics and mild cryogelation procedure, the resultant particles show uniform size dispersion and excellent biocompatibility. Moreover, acryloylated stem cell recruitment peptides SKPPGTSS are directly crosslinked within the particles by ultraviolet irradiation, thus simplifying the peptide coupling process and preventing its premature release. The SKPPGTSS-modified particles can recruit endogenous stem cells to promote cartilage repair and the released Exo from the particles further enhances the cartilage repair performance through synergistic effects. Nerve guidance conduits (NGCs) are considered as promising treatment strategy and frontier trend for peripheral nerve regeneration, while their therapeutic outcomes are limited by the lack of controllable drug delivery and available physicochemical cues. Herein, novel aligned piezoelectric nanofibers derived hydrogel NGCs with ultrasound (US)-triggered electrical stimulation (ES) and controllable drug release for repairing peripheral nerve injury are proposed. The inner layer of the NGCs is the barium titanate piezoelectric nanoparticles (BTNPs)-doped polyvinylidene fluoride-trifluoroethylene [BTNPs/P(VDF-TrFE)] electrospinning nanofibers with improved piezoelectricity and aligned orientation. The outer side of the NGCs is the thermoresponsive poly(N-isopropylacrylamide) hybrid hydrogel with bioactive drug encapsulation. Such NGCs can not only induce neuronal-oriented extension and promote neurite outgrowth with US-triggered wireless ES, but also realize the controllable nerve growth factor release with the hydrogel shrinkage under US-triggered heating. Thus, the NGC can positively accelerate the functional recovery and nerve axonal regeneration of rat models with long sciatic nerve defects. Organ-on-chips can highly simulate the complex physiological functions of organs, exhibiting broad application prospects in developmental research, disease simulation, as well as new drug research and development. However, there is still less concern about effectively constructing cochlea-on-chips. Here, a novel cochlear organoids-integrated conductive hydrogel biohybrid system with cochlear implant electroacoustic stimulation (EAS) for cochlea-on-a-chip construction and high-throughput drug screening, is presented. Benefiting from the superior biocompatibility and electrical property of conductive hydrogel, together with cochlear implant EAS, the inner ear progenitor cells can proliferate and spontaneously shape into spheres, finally forming cochlear organoids with good cell viability and structurally mature hair cells. By incorporating these progenitor cells-encapsulated hydrogels into a microfluidic-based cochlea-on-a-chip with culture chambers and a concentration gradient generator, a dynamic and high-throughput evaluation of inner ear disease-related drugs is demonstrated. Wound healing remains a critical challenge in regenerative engineering. Great efforts are devoted to develop functional patches for wound healing. Herein, a novel sprayable black phosphorus (BP)-based multifunctional hydrogel with on-demand removability is presented as a joints' skin wound dressing. The hydrogel is facilely prepared by mixing dopamine-modified oxidized hyaluronic acid, cyanoacetategroup-functionalized dextran containing black phosphorus, and the catalyst histidine. The catechol-containing dopamine can not only enhance tissue adhesiveness, but also endow the hydrogel with antioxidant capacity. In addition, benefiting from the photothermal conversion ability of the BP and thermally reversible performance of the formed C═C double bonds between aldehyde groups and cyanoacetate groups, the resulting hydrogel displays excellent antibacterial performance and on-demand dissolving ability under NIR irradiation. Moreover, by loading vascular endothelial growth factor into the hydrogel, the promoted migration and angiogenesis effects of endothelial cells can also be achieved. Based on these features, it is demonstrated that such sprayable BP hydrogels can effectively facilitate joint wounds healing by accelerating angiogenesis, alleviating inflammation, and improving wound microenvironment. Self-healing hydrogels have emerged as the most promising alternatives to conventional brittle hydrogels used in the biomedical field due to the features of long-term stability and durability. However, the incompatibility between the fast self-healing property and enough mechanical strength of hydrogels remains a challenge. Therefore, hydrogels that possess not only mechanical toughness but also autonomous self-healing capacity are sought after. This review presents a comprehensive summary of the latest self-healing mechanisms. Specifically, we review various systems based on dynamic bonds, ranging from dynamic covalent bonds to non-covalent bonds. Additionally, this review presents different characterization methods for self-healing hydrogels, and also highlights their potential applications in the biomedical field, such as tissue engineering, drug delivery, cell therapy, and wound dressing. Furthermore, this review aims to provide valuable guidance for constructing diverse self-healing hydrogels with tailored functions.

Many hydrogel patches are developed to solve the pervasive and severe challenge of complex wound healing, while most of them still lack satisfactory controllability and comprehensive functionality. Herein, inspired by multiple creatures, including octopuses and snails, a novel muti-functional hydrogel patch is presented with controlled adhesion, antibacterial, drug release features, and multiple monitoring functions for intelligent wound healing management. The patch with micro suction-cup actuator array and a tensile backing layer is composed of tannin grafted gelatin, Ag-tannin nanoparticles, polyacrylamide (PAAm) and poly(N-isopropylacrylamide) (PNIPAm). In virtue of the photothermal gel-sol transition of tannin grafted gelatin and Ag-tannin nanoparticles, the patches exert a dual anti-microbial effect and temperature-sensitive snail mucus-like features. In addition, as the "suction-cups" consisting of thermal responsive PNIPAm can undergo a contract-relax transformation, the medical patches can adhere to the objects reversibly and responsively, and release their loaded vascular endothelial growth factor (VEGF) controllably for wound healing. More attractively, benefiting from their fatigue resistance, self-healing ability of the tensile double network hydrogel, and electrical conductivity of Ag-tannin nanoparticles, the proposed patches can report multiple wound physiology parameters sensitively and continuously. Hydrogel patches have outstanding values in wound treatment; challenges in this field are concentrated on developing functional and intelligent hydrogel patches with new antibacterial strategies for improving healing process. Herein, a novel melanin-integrated structural color hybrid hydrogel patches for wound healing is presented. Such hybrid hydrogel patches are fabricated by infusing asiatic acid (AA)-loaded low melting-point agarose (AG) pregel into the melanin nanoparticles (MNPs)-integrated fish gelatin inverse opal film. In this system, MNPs not only impart the hybrid hydrogels with properties of photothermal antibacterial and antioxidant, but also improve the visibility of structural colors by providing an inherent dark background. Besides, the photothermal effect of MNPs under near-infrared irradiation can also trigger liquid transformation of AG component in hybrid patch, resulting in the controllable release of its loaded proangiogenic AA. Attracting, this drug release induced refractive index variations in the patch can be detected as visible structural color shifting, which can be used to monitor their delivery processes. Benefiting from these features, the hybrid hydrogel patches are demonstrated to achieve excellent therapeutic effects for in vivo wound treatment. Heart-on-chips have emerged as a powerful tool to promote the paradigm innovation in cardiac pathological research and drug development. Attempts are focused on improving microphysiological visuals, enhancing bionic characteristics, as well as expanding their biomedical applications. Herein, inspired by the bright feathers of peacock, we present a novel optical and electrical dual-responsive heart-on-a-chip based on cardiomyocytes hybrid bright MXene structural color hydrogels for hormone toxicity evaluation. Such hydrogels with inverse opal nanostructure are generated by using pregel to replicate MXene-decorated colloidal photonic crystal (PhC) array templates. The attendant MXene in the hydrogels could not only enhance the saturation of structural color, but also ensure the composite hydrogel with excellent electroconductivity to facilitate the synergetic beating of their surface cultured cardiomyocytes. In this case, the hydrogels would undergo a synchronous deformation and generate shift in corresponding photonic band gap and structural color, which could be employed as visual signal for self-reporting of the cardiomyocyte mechanics. Based on these features, we demonstrated the practical value of the optical and electrical dual-responsive structural color MXene hydrogels constructed heart-on-a-chip in hormone toxicity testing. Hard-healing diabetic wound brings burgeoning physical and mental burdens to patients. Current treatment strategies tend to achieve multistage promotion and real-time reporting to facilitate wound management. Herein, a biomimetic enzyme cascade inverse opal microparticles system for wound healing, which is intergated with glucose oxidase (GOD) and copper peroxide (CP). Such microparticles are composed of biofriendly hyaluronic acid methacryloyl (HAMA) and pH-responsive acrylic acid (AA), which provided abundant binding sites and spaces for chemical immobilizing and physical doping of enzymes and metal bioinorganics. When the cascade catalytic system is applied on wound sites, hyperglycemia environment would serve as a hydrogen peroxide (H 2 O 2 ) generator through GOD catalysis, while acidic environment triggers the decomposition of CP, further catalyzing H 2 O 2 to generate reactive oxygen species (ROS). Additionally, the distinctive structural color of the microparticles can visually reflect the wound pH and intelligently estimate the healing state. It is demonstrated that such microparticle systems exhibit excellent broad-spectrum antibacterial and angiogenesis-promoting properties, as well as significant real-time reporting ability for wound healing. Due to their intrinsic anti-inflammatory and immunomodulatory properties, adipose-derived stem cells (ADSCs) are explored as a promising alternative in treating rheumatoid arthritis (RA). To address the poor survival and function loss of directly injected stem cells, efforts in this area are focus on the generation of efficient cell delivery vehicles. Herein, a novel extracellular matrix (ECM)-inspired injectable hydrogel for ADSCs encapsulation and RA treatment is proposed. The hydrogel with dendritic polylysine and polysaccharide components is formed through the reversible Schiff base crosslinking. It possesses self-healing capability, superior mechanical properties, minimal toxicity, and immunomodulatory ability. When encapsulated with ADSCs, the hydrogel could recover chronic inflammation by directly reversing the dominant macrophage phenotype from M1 to M2 and inhibiting the migration of fibroblast-like synoviocytes. Through a collagen-induced arthritis rat model, the tremendous therapeutic outcomes of this ADSCs-laden hydrogel, including inflammation attenuation, cartilage protection, and bone mineral density promotion are demonstrated. Graphene oxide (GO) hydrogels have provided tremendous opportunities in designing and fabricating complex constructs for diverse applications, while their 3D printing without photocuring is still a challenging task due to their low viscosity, uncontrollable gelation, and low interfacial tension. Here, we report a histidine-assisted printing strategy to prepare GO hybrid hydrogels through the microfluidic 3D printing technique. We found that the GO additive could significantly hamper the Knoevenagel condensation (KC) reaction between benzaldehyde and cyanoacetate group-functionalized polymers to form a hydrogel, while these GO mixed solutions were rapidly solidified into a hydrogel when histidine was added. This fascinating phenomenon enabled us to prepare low-viscosity GO mixed polymer solutions as printable inks and generate hydrogel microfibers in histidine solutions. The hydrogel fibers could support cell survival and be further constructed into complex 3D structures through microfluidic 3D printing techniques. Moreover, due to the addition of GO, the microfibers exhibited excellent electrical conductivity and could sense the motion changes and convert these stimuli as electrical resistance signals. This strategy adds an option for the design and application of 3D printable aqueous GO inks in many fields.

Developing a self-reporting bioadhesive patch that has strong adhesion to the wet tissues and meanwhile can avoid adhering to the adjacent tissues is a current research difficulty and challenge. In this paper, inspired by the wet adhesion of spider web, slippery surface of Nepenthes, and structural color phenomena of chameleons, a novel structural color medical patch with surface dual-properties of wet bioadhesion and slipperiness for internal tissue repair based on inverse opal scaffold is presented. The adhesive surface made by poly(acrylic acid)-polyethylene glycol-N-hydroxysuccinimide ester and gelatin hydrogel can attain tough adhesion to internal wet tissues by absorbing tissue interfacial water and the covalent cross-linking between the hydrogel and tissue. Besides, the slippery surface made by liquid paraffin infused inverse opal scaffold can avoid adhesion to the adjacent tissues. It is demonstrated that the designed patch can adhere tightly to the defect tissue and improve the tissue repair without adjacent adhesion when applied in a rat model with full-thickness perforation of the stomach wall. In addition, the responsive structural color can supply a color-sensing monitoring to evaluate the adhesive and repair process. Background: Wound healing has become a worldwide healthcare issue. Attempts in the area focus on developing patches with the capabilities of avoiding wound infection, promoting tissue remolding, and reporting treatment status that are of great value for wound treatment., Results: In this paper, we present a novel inverse opal film (IOF) patch based on a photo-crosslinking fish gelatin hydrogel with the desired features for wound healing and dynamic monitoring. The film with vibrant structure colors was constructed by using the mixture of fish gelatin methacryloyl, chitosan, and polyacrylic acid (PAA) to replicate colloidal crystal templates. As the structures of these natural biomolecules are well-retained during the fabrication, the resultant IOF was with brilliant biocompatibility, low immunogenicity, antibacterial property, as well as with the functions of promoting tissue growth and wound healing. In addition, the IOF presented interconnected nanopores and high specific surface areas for vascular endothelial growth factor loading, which could further improve its angiogenesis capability. More attractively, as the pH-responsive PAA was incorporated, the IOF patch could report the wound healing status through its real-time structural colors or reflectance spectra., Conclusions: These features implied the practical value of the multifunctional fish gelatin hydrogel IOFs in clinical wound management., (© 2022. Heart-on-a-chip plays an important role in revealing the biological mechanism and developing new drugs for cardiomyopathy. Tremendous efforts have been devoted to developing heart-on-a-chip systems featuring simplified fabrication, accurate imitation and microphysiological visuality. In this paper, the authors present a novel electroconductive and anisotropic structural color hydrogel by simply polymerizing non-close-packed colloidal arrays on super aligned carbon nanotube sheets (SACNTs) for visualized and accurate heart-on-a-chip construction. The generated anisotropic hydrogel consists of a colloidal array-locked hydrogel layer with brilliant structural color on one surface and a conductive methacrylated gelatin (GelMA)/SACNTs film on the other surface. It is demonstrated that the anisotropic morphology of the SACNTs could effectively induce the alignment of cardiomyocytes, and the conductivity of SACNTs could contribute to the synchronous beating of cardiomyocytes. Such consistent beating rhythm caused the deformation of the hydrogel substrates and dynamic shifts in structural color and reflection spectra of the whole hybrid hydrogels. More attractively, with the integration of such cardiomyocyte-driven living structural color hydrogels and microfluidics, a visualized heart-on-a-chip system with more consistent beating frequency has been established for dynamic cardiomyocyte sensing and drug screening. Structural colors, derived from existing natural creatures, have aroused widespread attention in the materials regulation for different applications. Here, inspired by the color adjusting mechanism of hummingbird, we present a novel shape-memory structural color hydrogel film by introducing shape memory polymers (SMPs) into synthetic inverse opal scaffold structure. The excellent flexibility as well as the inverse opal structure of the hydrogel films imparts them with stable stretchability and brilliant structural colors. Benefiting from the transient structural anisotropy of copolymers, the hybrid films are possessed with shape-morphing behaviors capability. Based on the shape transformations and color responsiveness performance, we have demonstrated diverse structural color actuators with complex shapes for different tasks. Notably, as the photothermal responsive graphene quantum dots were integrated into the hydrogel, the hybrid films could also be endowed with the feature of light-controlled reversible deformation with synchronous structural color variation. Wound healing is an important issue for regenerative medicine. Attempts in this area tend to develop functional wound patches to promote the healing. Here, we present self-bonded hydrogel inverse opal particles as sprayed flexible patch for wound healing. Such particles were fabricated by infusing drugs-loaded gelatin (GT) and carrageenan (CG) pregel into inverse opal scaffolds, which were composed of biocompatible hyaluronic acid methacryloyl (HAMA) and gelatin methacryloyl (GelMA) with graphene oxide quantum dots (GO QDs) doping. Due to the photothermal conversion capability of GO QDs and temperature reversible phase-changing performance of GT/CG, the hybrid particles could undergo GT/CG liquid transformation under the near-infrared (NIR) irradiation, which made them adhere to each other and finally form a flexible patch. Following by the phase-change of GT/CG hydrogel, the encapsulated drugs were also controllably released from the inverse opal scaffold. As the inverse opal scaffolds of the hybrid particles were maintained, their drug release induced refractive index changes could be detected as visual structural color shifting, which could be utilized to monitor their delivery processes. Based on these features, we have demonstrated that the self-bonded particles, administered in the form of spray, could be applied for wound tissue healing and drug delivery monitoring. These results indicate that the self-bonded hydrogel particles have potential value as a multifunctional patch for clinical applications.

Micromotors have opened novel avenues for drug delivery due to their capacity for self-propelling. Attempts in this field trend towards ameliorating their functions to promote their clinical applications. In this paper, an ingenious suction-cup-inspired micromotor is presented with adhesive properties for drug delivery in the stomach. The micromotors are fabricated by using hydrogel replicating the structure of suction-cup-like microparticles, which derive from self-assembly of colloidal crystals under rapid solvent extraction, followed by loading magnesium (Mg) in the bottom spherical surface. The Mg-loaded micromotors can realize spontaneous movement due to the continual generation of hydrogen bubbles in gastric juice. The combination of unique suction-cup-like structure with excellent motion performance makes the micromotor an ideal carrier for drug delivery as they can efficiently adhere to the tissue. Moreover, benefiting from the porous structure, the hydrogel micromotors exhibit a high volume-surface ratio, which enables efficient drug loading. It is demonstrated that the suction-cup-inspired micromotors can adhere efficiently to the ulcer-region in the stomach and release drugs due to their distinctive architecture and spontaneous motion, exhibiting desirable curative effect of gastric ulcer. Effective and timely joint monitoring has been a significantly vital research direction in human healthcare. As an emerging technology, flexible electronics provides more possibilities and applicabilities for practical sensing and signal transmission. Here, we provide novel elastic MXene microfibers of controllable morphologies at a microscale through microfluidic technology for actual joint motion monitoring. Double-network hydrogels including covalently cross-linking polyacrylamide and ionically cross-linking alginate were chosen for superelasticity. For the improvement of the electrical conductivity of superelastic hydrogel microfibers, MXene was selected to mix with them. By introducing the cross-linker to the outer channel, microfibers with controllable diameters along with high electrical conductivities and tensile properties could be fabricated successfully. The practical value of the synthesized microfibers in joint movement sensing has been demonstrated by acting as the element of new motion sensors. Based on these features, it is believed that these elastic MXene hydrogel microfibers have high potential for rapid sensing and diagnosis of joint diseases.

Stratagems of drug delivery are important for disease treatment and other biomedical areas. In this study, a novel stratagem is presented for versatile and controllable drug delivery by integrating photoresponsive drug delivery microspheres (PDDMs) into pyramid microneedle (MN) arrays. The PDDMs, containing black-phosphorus (BP) and poly (N-isopropylacrylamide) (pNIPAM), are generated by a flexible capillary microfluidic method. Benefiting from the high water content of the pNIPAM hydrogel, various bioactive substances can be loaded and maintain biological activity. Furthermore, due to the near-infrared (NIR) absorption and conversion capabilities of the contained BP, the PDDMs can increase temperature, shrink volume, and release their encapsulated bioactives under the trigger of biocompatible NIR. In addition, as the PDDMs are stuffed into the solid MN arrays of porous ethoxylated trimethylolpropane triacrylate (ETPTA), the composited PDDMs-MNs system has enough mechanical strength to penetrate into the skin and can deliver drugs underneath the skin uniformly. Based on the resultant PDDMs-MNs, it is demonstrated that insulin can be controllably released to adjust blood glucose levels of streptozotocin (STZ)-induced diabetic mice. Bionic electronic skin (E-skin) that could convert external physical or mechanical stimuli into output signals has a wide range of applications including wearable devices, artificial prostheses, software robots, etc. Here, we present a chameleon-inspired multifunctional E-skin based on hydroxypropyl cellulose (HPC), Poly(Acrylamide-co-Acrylic acid) (PACA), and carbon nanotubes (CNTs) composited liquid-crystal hydrogel. We found that the HPC could still form cholesteric liquid-crystal photonic structures with the CNTs additive for enhancing their color saturation and PACA polymerization for locating their assembled periodic structures. As the composite hydrogel containing HPC elements and the PACA scaffold responds to different stimuli, such as temperature variations, mechanical pressure, and tension, it could correspondingly change its volume or internal nanostructure and report these as visible color switches. In addition, due to the additive of CNTs, the composite hydrogel could also output these stimuli as electrical resistance signals. Thus, the hydrogel E-skins had the ability of quantitatively feeding back external stimuli through electrical resistance as well as visually mapping the stimulating sites by color variation. Patch plays an important role in clinical medicine for its broad applications in tissue repair and regeneration. Here, inspired by the diverse adhesion, anti-adhesion, and responsive structural color phenomena in biological interfaces, we present a hybrid hydrogel film with an adhesive polydopamine (PDA) layer and an anti-adhesive poly(ethylene glycol) diacrylate (PEGDA) layer in an inverse opal scaffold. It was demonstrated that the resultant hydrogel film could serve as a functional tissue patch with an excellent adhesion property on one surface for repairing injured tissues and an anti-adhesion property on the other surface for preventing adverse adhesion. Besides, because of the responsive structural color, the patch was imparted with self-reporting mechanical capability, which could provide a real-time color-sensing feedback to monitor the heartbeat activity. Moreover, the catechol groups on PDA imparted the patch with high tissue adhesiveness and self-healing capability in vivo. Islet transplantation has been one promising strategy in diabetes treatment, which can maintain patient's insulin level long-term and avoid periodical insulin injections. However, donor shortage and temporal mismatch between donors and recipients has limited its widespread use. Therefore, searching for islet substitutes and developing efficient cryopreservation technology (providing potential islet bank for transplantation on demand) is in great need. Herein, a novel cryopreservation method is developed for islet β cells by combining microfluidic encapsulation and cold-responsive nanocapsules (CR-NCs). The cryopreserved cell-laden hydrogels (calcium alginate hydrogel, CAH) can be transplanted for diabetes treatment. During the freezing process, trehalose is released inside β cells through the CR-NCs and serves as the sole cryoprotectant (CPA). Additionally, CAH helps cells to survive the freeze-thaw process and provide cells with a natural immune barrier in vivo. Different from traditional cryopreservation methods, this method combining the CR-NCs and hydrogel encapsulation replaces the toxic CPAs with natural trehalose. Great preservation results are obtained and transplantation experiments of diabetic rats further prove the excellent glucose regulation ability of such β cell-laden hydrogels post cryopreservation. Structural color hydrogels with healable capability are of great significance in many fields, however the controllability of these materials still needs optimizing. Thus, this work presents a healable structural color hydrogel with photocontrolling properties. The component parts of the hydrogel are a graphene oxide (GO) integrated inverse opal hydrogel scaffold and a hydrogel filler with reversible phase transition. The inverse opal scaffold provides stable photonic crystal structure and the hydrogel filler is the foundation of healing. Taking advantage of the prominent photothermal conversion efficiency of GO, the healable structural color material is imparted with photocontrolled properties. It is found that the structural color hydrogel shaped in complex patterns can heal under near-infrared (NIR) irradiation. Structural color hydrogels are promising candidates as scaffold materials for tissue engineering and for matrix cell culture and manipulation, while their super-elastic features are still lacking due to the irreconcilable interfere of the precursor and the self-assembly unit. This hinders many of their practical biomedical applications where elasticity is required. Herein, hydrophilic and size-controllable Fe 3 O 4 @poly(4-styrenesulfonic acid-co-maleic acid) (PSSMA)@SiO 2 magnetic response photonic crystals are fabricated as the assembly units of the structural color hydrogels by orderly packing of core-shell colloidal nanocrystal clusters via a two-step facile synthesis approach. These units are capable of responding instantaneously to an external magnetic field with resistance to interference of ions, thus, by integrating super-elastic hydrogels, super-elastic magnetic structural color hydrogels can be achieved. The structural color arises from the dynamic ordering of the magnetic nanoparticles through the contactless control of external magnetic field, allowing regional polymerization of hydrogels via changing orientation and strength of external magnetic field. Multidrug resistance is one of the leading causes of chemotherapy failure in cancer patients. Early detection and capture of drug-resistant tumor cells can facilitate the monitoring of the therapy process and improve the prognosis of patients. In this study, novel P-glycoprotein (P-gp) antibody modified porous hydrogel particles are proposed for drug-resistant tumor cells capture. The hydrogel particles employ a highly biocompatible hydrogel, methacrylate gelatin (GelMA), as the carrier and replicate from the silica colloidal crystal beads. By the modification of P-gp antibody probes on their surfaces, the hydrogel particles are endowed with the ability to capture drug-resistant tumor cells, which overexpress specific components of P-gp on their membranes. Additionally, the acquired ordered porous nanostructure of the particles can provide not only more surface area for antibody immobilization but also a nanopatterned platform for highly efficient target cell capture. Early detection of cardiac troponin I (cTnI), B-type natriuretic peptide (BNP), and myoglobin (Myo) is essential for the diagnosis of acute myocardial infarction (AMI) and heart failure (HF). We designed a porous hydrogel-encapsulated photonic crystal (PhC) barcode-based suspension array for multiple cardiovascular marker detection. The hybrid hydrogel was composed of polyethylene glycol diacrylate (PEGDA) and gelatin, resulting in a porous and hydrophilic scaffold which ensured stability of the PhC in aqueous solutions. The encapsulated PhC barcodes had stable diffraction peaks for the corresponding markers. Using a sandwich format, the proposed suspension array was used for simultaneous multiplex detection of cardiovascular biomarkers in a single tube. The immunoassay results we tested on cTnI, BNP, and Myo could be assayed in the ranges of 0.01 to 1000 ng/mL, 0.1 to 10 000 pg/mL, and 1 to 10 000 ng/mL with limits of detection of 0.009 ng/mL, 0.084 pg/mL, and 0.68 ng/mL at 3σ, respectively. This method also showed acceptable accuracy and repeated detection, and the results were consistent with the results of conventional clinical methods for detecting actual clinical samples. Therefore, suspension arrays based on hydrogel-encapsulated PhC barcodes are highly promising for AMI diagnosis.

The ultrasensitive detection of multiple nucleic acids has great significance in a broad range of medical fields since nucleic acid samples are usually of small volumes and highly diluted. Herein, we present a novel magnetic photonic crystal (PhC) barcode-integrated condensing-enriched superhydrophobic platform for the ultrasensitive multiple miRNA detection. Droplets containing targets could suck the hydrogel PhC barcodes without losing their targets due to the difference in wettability between the hydrophilic PhC barcodes and hydrophobic substrate. During the evaporation of water from the droplet microreactors, these targets could be effectively enriched in the barcodes for achieving higher sensitivities. As the encoding signals of the PhC barcodes are the characteristic reflection peaks generated by their ordered physical nanostructures, the barcodes are stable and free from any fluorescent background and photobleaching; thus, they are accurate for distinguishing different targets. In addition, with the integration of magnetic nanoparticles into the hydrogel PhC barcodes, they could be imparted with the features of immobilization during change of medium and flexible movement for controlling the reaction, both of which could facilitate the detection processes. Based on this platform, we have demonstrated that the detection limit of multiple miRNAs is improved by about three orders. Thus, our platform is an ideal alternative for the ultrasensitive simultaneous multiplex analysis in medical fields.

Although some advanced treatments of heart failure (HF) have experienced great development in recent decades, morbidity and mortality associated with HF have not improved significantly. Hydrogels have some unique advantages for the delivery and controlled release of bioactive molecules, drugs, and cells, which can directly affect the heart locally. Many studies have shown that hydrogels represent a new approach for the treatment of HF. In this review, we present a current perspective of hydrogel-based approaches for cardiac tissue engineering and myocardial regeneration in ischemia-induced HF, with an emphasis on functional studies, translation, and clinical advancements.

Liver cancer is one of the malignant cancers that seriously threatens human health. Although some common treatments including chemotherapy have been applied in oncotherapy, there are often serious shortcomings such as frequent and uncontrollable drug infusion. To overcome these limitations, here, we introduced responsive porous hydrogel microparticles loaded with 5-fluorouracil and metformin for oncotherapy. Because of the interconnected porous structures, various forms of active molecules could be loaded into the particles. In addition, the relatively higher temperature of the tumor site and the temperature-responsive shape transition of pNIPAM hydrogel enabled controllable drug release. The porous pNIPAM particles not only exhibited large loading efficiency and sustained release for the 5-fluorouracil and metformin co-delivery, but also protected drugs from being resolved. Thus, it can be anticipated that the porous microparticles will have great potential in oncotherapy.

Biohybrid actuators composed of living tissues and artificial materials have attracted increasing interest in recent years because of their extraordinary function of dynamically sensing and interacting with complex bioelectrical signals. Here, a compound biohybrid actuator with self-driven actuation and self-reported feedback is designed based on an anisotropic inverse opal substrate with periodical elliptical macropores and a hydrogel filling. The benefit of the anisotropic surface topography and high biocompatibility of the hydrogel is that the planted cardiomyocytes could be induced into a highly ordered alignment with recovering autonomic beating ability on the elastic substrate. Because of the cell elongation and contraction during cardiomyocyte beating, the anisotropic inverse opal substrates undergo a synchronous cycle of deformation actuations, which can be reported as corresponding shifts of their photonic band gaps and structural colors. These self-driven biohybrid actuators could be used as elements for the construction of a soft-bodied structural color robot, such as a biomimetic guppy with a swinging tail. Besides, with the integration of a self-driven biohybrid actuator and microfluidics, the advanced heart-on-a-chip system with the feature of microphysiological visuality has been developed for integrated cell monitoring and drug testing. This anisotropic inverse opal-derived biohybrid actuator could be widely applied in biomedical engineering.

Rapid wound healing is a fundamental health concern and poses a critical challenge for healthcare systems worldwide. Here, we developed an injectable, dynamic, self-healing hydrogel serving as a fibroblast growth factor 2 (FGF2) delivery system for tissue repair applications. Benzaldehyde-terminated polyethylene glycol (BAPEG) was synthesized and cross-linked with N-Succinyl-chitosan (SCS) through reversible Schiff-base reaction to form a dynamic self-healing hydrogel. Because it can be injected after gelation and self-heal into an integral part, the hydrogel could not only auto-adapt to the irregular wound surface to provide better protection but also encapsulate drugs homogeneously and implant into tissue with a less invasive strategy. As a result, the FGF2-loaded SCS/BAPEG hydrogel is able to significantly promote the process of angiogenesis, collagen deposition, and granulation tissue formation, as well as reduce inflammation. This study opens the door to self-healing hydrogels serving as a drug delivery system for tissue repair applications.

Structural color hydrogels with lasting survivability are important for many applications, but they still lack anti-biodegradation capability. Thus, we herein present novel antibacterial structural color hydrogels by simply integrating silver nanoparticles (AgNPs) in situ into the hydrogel materials. Because the integrated AgNPs possessed wide and excellent antibacterial abilities, the structural color hydrogels could prevent bacterial adhesion, avoid hydrogel damage, and maintain their vivid structural colors during their application and storage. It was demonstrated that the AgNP-tagged poly(N-isopropylacrylamide) structural color hydrogels could retain their original thermal-responsive color transition even when the AgNP-free hydrogels were degraded by bacteria and that the AgNP-integrated self-healing structural color protein hydrogels could save their self-repairing property instead of being degraded by bacteria. These features indicated that the antibacterial structural color hydrogels could be amenable to a variety of practical biomedical applications.

Diagnosing hematological disorders based on the separation and detection of cells in the patient's blood is a significant challenge. We have developed a novel barcode particle-based suspension array that can simultaneously capture and detect multiple types of blood cells. The barcode particles are polyacrylamide (PAAm) hydrogel inverse opal microcarriers with characteristic reflection peak codes that remain stable during cell capture on their surfaces. The hydrophilic PAAm hydrogel scaffolds of the barcode particles can entrap various plasma proteins to capture different cells in the blood, with little damage to captured cells.

Cell adhesion and alignment are two important considerations in tissue engineering applications as they can regulate the subsequent cell proliferation activity and differentiation program. Although many effects have been applied to regulate the adhesion or alignment of cells by using physical and chemical methods, it is still a challenge to regulate these cell behaviors simultaneously. Here, we present novel substrates with tunable nanoscale patterned structures for regulating the adhesion and alignment of cells. The substrates with different degrees of pattern orientation were achieved by customizing the amount of stretching applied to polymer inverse opal films. Cells cultured on these substrates showed an adjustable morphology and alignment. Moreover, soft hydrogels, which have poor plasticity and are difficult to cast into patterned structures, were applied to infiltrate the inverse opal structure. We demonstrated that the adhesion ratio of cells could be regulated by these hybrid substrates, as well as adjusting the cell morphology and alignment. These features of functional inverse opal substrates make them suitable for important applications in tissue engineering.

An angle-independent photonic crystal (PhC) colorimetric sensor was developed by using a stimuli-response hydrogel to replicate the template arrays of isotropic photonic crystal beads (PCBs) for the detection of Hg(2+).

We present a new method to prepare inverse opaline photonic beads with good spherical shape and superior optical performance by simply introducing an interfacial tension system into a template replication method. When the scaffolds of these beads were composed of poly(ethylene glycol) diacrylate hydrogel, they could provide a homogeneous water surrounding, which remedied many shortcomings of biomolecular microcarriers introduced by the presence of the solid surface of them. The suspension array, which used these macroporous hydrogel photonic beads as coding elements, showed obvious advantages in multiplexed capability, rapid biomolecular screening (within 12 min), and highly sensitive detection (with limit of detection of approximately 10(-12) M).

Highlights: Derived from silica photonic crystals, inverse opal microspheres have a regularly connected porous structure and inherit structural color properties. Combined with the stable scaffold and the photothermal phase-transition of the secondary filling material, the inverse opal composite microspheres are endowed with self-healing properties and the ability for controllable drug release. Inverse opal microspheres were significantly treated for diabetic wound, via promoting tissue regeneration, collagen deposition and angiogenesis. Meanwhile, the release of drugs could be monitored by the structural color characteristic. Chronic diabetic wounds confront a significant medical challenge because of increasing prevalence and difficult-healing circumstances. It is vital to develop multifunctional hydrogel dressings, with well-designed morphology and structure to enhance flexibility and effectiveness in wound management. To achieve these, we propose a self-healing hydrogel dressing based on structural color microspheres for wound management. The microsphere comprised a photothermal-responsive inverse opal framework, which was constructed by hyaluronic acid methacryloyl, silk fibroin methacryloyl and black phosphorus quantum dots (BPQDs), and was further re-filled with a dynamic hydrogel. The dynamic hydrogel filler was formed by Knoevenagel condensation reaction between cyanoacetate and benzaldehyde-functionalized dextran (DEX-CA and DEX-BA). Notably, the composite microspheres can be applied arbitrarily, and they can adhere together upon near-infrared irradiation by leveraging the BPQDs-mediated photothermal effect and the thermoreversible stiffness change of dynamic hydrogel. Additionally, eumenitin and vascular endothelial growth factor were co-loaded in the microspheres and their release behavior can be regulated by the same mechanism. Moreover, effective monitoring of the drug release process can be achieved through visual color variations. The microsphere system has demonstrated desired capabilities of controllable drug release and efficient wound management. Biomedical patches offer significant potential for keratitis treatment. Building on advances in multifunctionality and biomimicry, an innovative, multifunctional hydrogel patch with high therapeutic efficacy, inspired by the native architecture and functions of the cornea, is introduced. By engineering a composite patch comprising recombinant human collagen (RHC) hydrogel, near‐field electrospinning (NFES) microfibers, and gold‐nanoparticle‐decorated tetragonal barium titanates (BTO@Au), structural mimicry, mechanical reinforcement, tissue‐specific adhesion, and bacterial inhibition are achieved. The RHC hydrogel recreates a three‐dimensional (3D) microenvironment that emulates the natural structure of the corneal tissue, demonstrating excellent tissue adhesion. Integrated within this hydrogel, the NFES microfibers, designed to emulate the orthogonal arrangement of native corneal stroma, not only reinforce the mechanical strength of the RHC hydrogel but also act as scaffolds to guide the aligned growth of human keratocytes. A unique aspect of this advanced patch is the incorporation of BTO@Au nanoparticles, which generate reactive oxygen species for effective bacterial eradication when subjected to ultrasound stimulation. Through in vivo studies on rat models with infected corneal wounds, this hydrogel patch exhibits superior therapeutic efficacy compared to the current treatment. It is posited that these cornea‐inspired ultrasound‐responsive adhesive hydrogel patches represent a significant scientific advancement with high potential for clinical applications. Rheumatoid arthritis (RA) is one of the leading causes of disability due to the autoimmune destruction of synovial joints. Great efforts have been put into developing multifunctional drug delivery systems for delaying the progression of RA. Herein, a novel locally injectable, hierarchically structured delivery system with a hyaluronic acid (HA) microparticle and an MTX‐conjugated nanoparticle for promoted RA treatment is proposed. In this system, MTX is chemically grafted onto a peptide dendrimer‐based nanoparticle (G‐MTX). These nanoparticles are encapsulated inside HA microparticles (H‐G‐MTX) using microfluidics to realize the system. The G‐MTX can be released from the degradable HA microparticles in the acidic environment of inflamed RA joints. As the G‐MTX shows an enhanced intracellular delivery of the conjugated MTX, the H‐G‐MTX can reverse the dominant macrophage phenotype from M1 to M2. Through a collagen‐induced arthritis (CIA) rat model, the great therapeutic outcomes of this hierarchically structured delivery system are demonstrated, including inflammation reduction, cartilage protection, and bone mineral density promotion. Hydrogel microrobots have blazed new avenues for local drug administration, while their values in inner ear drug delivery remain to be explored. Here, novel magnetic alpha‐lipoic acid (ALA)‐conjugated gelatin methacryloyl hydrogel microrobots are presented for inner ear drug delivery and deafness prevention. The magnetic hydrogel microrobots with features of slow drug release, targeted movement, and biosafety are generated from microfluidic devices. After intratympanic injection, the microrobots can be oriented in the middle ear cavity under a specific magnetic field. In addition, the ALA drug released from the magnetic hydrogel microrobots can remain in the inner ear for a long time. Based on these advantages, the practical value of the magnetic hydrogel microrobots in deafness prevention is demonstrated by pre‐injecting them in the middle ear of cisplatin‐deafened mice. These results indicate that the proposed smart magnetic hydrogel microrobots can effectively deliver drugs to the inner ear through the middle ear drug delivery route, thus opening a new chapter for middle ear deafness treatment. Chemotherapy is an essential postoperative treatment for pancreatic cancer, while due to the lack of effective drug evaluation platforms, the therapeutic outcomes are hampered by tumor heterogeneity among individuals. Here, a novel microfluidic encapsulated and integrated primary pancreatic cancer cells platform is proposed for biomimetic tumor 3D cultivation and clinical drug evaluation. These primary cells are encapsulated into hydrogel microcapsules of carboxymethyl cellulose cores and alginate shells based on a microfluidic electrospray technique. Benefiting from the good monodispersity, stability, and precise dimensional controllability of the technology, the encapsulated cells can proliferate rapidly and spontaneously form 3D tumor spheroids with highly uniform size and good cell viability. By integrating these encapsulated tumor spheroids into a microfluidic chip with concentration gradient channels and culture chambers, dynamic and high‐throughput drug evaluation of different chemotherapy regimens could be realized. It is demonstrated that different patient‐derived tumor spheroids show different drug sensitivity on‐chip, which is significantly consistent with the clinical follow‐up study after the operation. Marine organisms provide novel and broad sources for the preparations and applications of biomaterials. Since the urgent requirement of bio‐hydrogels to mimic tissue extracellular matrix (ECM), the natural biomacromolecule hydrogels derived from marine sources have received increasing attention. Benefiting from their outstanding bioactivity and biocompatibility, many attempts have been made to reconstruct ECM components by applying marine‐derived natural hydrogels. Moreover, marine hydrogels have been successfully applied in biomedicine by means of microfluidics, electrospray, and bioprinting. In this review, the classification and characteristics of marine‐derived hydrogels are summarized. In particular, their role in the development of biomaterials is also introduced. Then, the recent advances in bio‐fabrication strategies for various hydrogel materials are focused upon. Besides, the influences of hydrogel types on their functions in biomedical applications are discussed in depth. Developing a biocompatible and multifunctional adhesive hydrogel with injectability and self-healing ability for promoting wound healing is highly anticipated in various clinical applications. In this paper, we present a novel natural biopolymer-derived hydrogel based on the aldehyde-modified oxidized guar gum (OGG) and the carboxymethyl chitosan (CMCS) for efficiently improving wound healing with the encapsulation of vascular endothelial growth factor (VEGF). As the hydrogels are synthesized via the dynamically reversible Schiff base linkages, it is imparted with excellent self-healing ability and good shear thinning behavior, which make the hydrogel be easily and conveniently injected through a needle. Besides, the physiochemical properties, including porous structure, mechanical strength and swelling ratio of the hydrogel can be well controlled by regulating the concentrations of the OGG. Moreover, the hydrogel can attain strong adhesion to the tissues at physiological temperature based on the Schiff base between the aldehyde group on the hydrogel and the amino group on the tissue. Based on these features, we have demonstrated that the VEGF encapsulated hydrogel can adhere tightly to the defect tissue and improve wound repair in the rat model of defected skin by promoting cell proliferation, angiogenesis, and collagen secretion. Stem cells have demonstrated values in diabetic ulcer (DU) treatments. Challenges in this area are focused on enhancing the localized curative effects of stem cells and improving diabetic wound healing efficiently. Herein, a novel living microneedle (MN) patch is presented as a localized delivery system of bioactive platelet derived growth factor D (PDGF‐D) and human adipose‐derived stem cells (ADSCs) for DU wound treatment. Compared with traditional complicated stem cell carriers, the MN patch can keep stem cell viability for ADSCs encapsulation and delivery, and possesses good mechanical strengths to penetrate the local skin wounds noninvasively. It is demonstrated that the delivery ADSCs are with the abilities of angiogenesis promotion during the DU wound healing; while the additive PDGF‐D can contribute significantly to the proliferation of ADSCs, strengthening the cell function of ADSCs and further facilitating the healing processes. Thus, living MN patches accelerate vascularization, tissue regeneration, and collagen deposition in a wounded diabetic mouse model, suggesting their potential application to DU wound healing and other therapeutic applications. Structural colors have a demonstrated value in constructing various functional materials. Efforts in this area are devoted to developing stratagem for generating heterogeneous structurally colored materials with new architectures and functions. Here, inspired by icing process in nature and ice-templating technologies, we present freeze-derived heterogeneous structural color hydrogels with multiscale structural and functional features. We find that the space-occupying effect of ice crystals is helpful for tuning the distance of non-close-packed colloidal crystal nanoparticles, resulting in corresponding reflection wavelength shifts in the icing area. Thus, by effectively controlling the growth of ice crystals and photo-polymerizing them, structural color hydrogels with the desired structures and morphologies can be customized. Other than traditional monochromatic structure color hydrogels, the resultant hydrogels can be imparted with heterogeneous structured multi-compartment body and multi-color with designed patterns through varying the freezing area design. Based on these features, we have also explored the potential value of these heterotypic structural color hydrogels for information encryptions and decryptions by creating spatiotemporally controlled icing areas. We believe that these inverse ice-template structural color hydrogels will offer new routes for the construction and modulation of next generation smart materials with desired complex architectures. While most of the structural colored materials only present isotropic colors, limiting their functions and many practical applications, while realizing anisotropic structural color materials remains challenging. Microneedle (MN) patches are promising stratagems for transdermal drug delivery owing to its minimal invasiveness and portability. Particularly, separable MN patches attract more attention for durable and long‐term drug delivery. Here, a new strategy for designing controllably separable MN patches by integrating photothermal responsive phase‐changing microspheres (MPs) into conical hydrogel MNs, is presented. Such MPs consist of near‐infrared (NIR)‐responsive black‐phosphorus (BP) and phase‐changing gelatin (GT), and are fabricated using a simple capillary microfluidic device. When the composite MN patch is exposed to NIR, the contained BP/GT MPs can transform light into heat, increase local temperature, turn into a liquid state, and form cavities in MNs, resulting in the convenient detachment of the backing plate and the needles by shear force. Subsequently, the embedded needles can sustainably release the drug. In addition to intelligently controlling the detachment of MNs, the BP/GT MPs can load macromolecular drugs and release them in response to NIR stimulation. Results further demonstrated that such controllably separable MN patch can effectively load and release two drugs to ameliorate disease progression for systemic lupus erythematosus treatment.
