Caesalpinia digyna (Rottl.) (Family: Fabaceae) is well known for its numerous medicinal values against several human disorders including fever, senile pruritis, diarrhea, tuberculosis, tonic disorder, diabetes, etc. The current study is intended to investigate the in vivo antipyretic activity of the methanol extract of C. digyna leaves (MECD) and its carbon-tetrachloride (CTCD) and butanol fraction (BTCD). As there is a resurgence of interest in plants as promising sources of new active pharmaceutical constituents, the present study has been designed to establish the preliminary biological activities of Blumea clarkei Hook.f. leaves. Here, the crude methanol extract of the leaves along with its organic and aqueous soluble fractions were subjected to different in vitro and in vivo assays. The ethyl acetate soluble fraction exhibited the highest total phenolic content (71.59 mg of GAE/gm of extract) and DPPH free radical scavenging (IC50 value 19.25 μg/ml) whereas another polar fraction; the aqueous soluble extractive showed the maximum cytotoxicity (LC50 value of 18.60 μg/ml) in the brine shrimp lethality assay. Piper nigrum L. also called black pepper is popular for its numerous uses. The present research is designed to investigate the pharmacological potential of methanol extract of Piper nigrum (MEPN). The antidepressant investigation was performed by using both in vivo forced swimming test (FST) and tail suspension test (TST) methods while the anxiolytic research by hole-board test (HBT) method. Again, the antipyretic analysis was conducted through yeast-induced pyrexia method, whereas clot lysis activity was employed by the thrombolytic method. Furthermore, in silico studies followed by molecular docking analysis of several secondary metabolites, pass prediction, and ADME/T were evaluated with AutoDock Vina, Discovery Studio 2020, UCSF Chimera software PASS online, and ADME/T online tools. The plant extract demonstrated dose-dependent potentiality in antidepressant, anxiolytic, antipyretic, and thrombolytic activities. Induction of MEPN produced a significant (p < .5, p < .001) increase of mobility in FST and TST, and increased the head dipping and decreased the latency of time (p < .01, p < .001) in HBT. MEPN 400 (mg/kg; b.w.; p.o.) lowered the rectal temperature of yeast-induced pyrexia substantially (p < .001). Besides, MEPN produced promising (p < .001) clot lysis activity. In the computational approach, among all the proteins, a docking score was found ranging from −1.0 to −7.90 kcal/mol. Besides, all the compounds were found safe in ADME/T study. Achyranthes aspera L. (Prickly Chaff flower) is an imperative Asian herb that has been known for numerous healing powers. The aim of this research is to investigate the pharmacological i.e., antidepressant and anxiolytic properties of the methanol extract of aerial part of A. aspera (MEAA) by in vivo and in silico approaches. Additionally, thrombolytic potential of the crude extract has also been explored. The antidepressant and anxiolytic investigations were performed by force swimming test (FST), tail suspension test (TST), and hole board test (HBT) in mice, whereas clot lysis activity test was conducted using thrombolytic experimental method. Furthermore, in silico studies were appraised with Discovery studio, PyRx, UCSF Chimera whereas, PASS prediction, and ADME/T was implemented using online tools. Treatment with MEAA revealed significant (p < 0.05) reduction of immobility time in FST and TST, increased head dipping and decreased the latency of time in HBT. Besides, MEAA produced significant (p < 0.001) clot lysis activity at a minimum dose. In the computer-aided approach, among all the proteins, the docking score has been revealed ranging from − 2.31 kcal/mol to − 7.4 kcal/mol. Spathulenol, diazepam and hydroquinone showed the best binding affinity and all compounds were found safe in ADME/T study. The outcomes of this research demonstrate that MEAA may provide potential for mitigating depression, anxiety, and coagulation.

Methods The antidiarrheal investigation was performed by using in vivo castor oil-induced diarrheal method whereas in vitro antimicrobial and antioxidant investigation have been implemented by disc diffusion and DPPH scavenging method respectively. Moreover, in silico studies were followed by molecular docking analysis of several secondary metabolites that were appraised with Schrödinger-Maestro v11.1 and Biovia Discovery Studio. Results The induction of plant extract (200 and 400 mg/kg, b.w, p.o) has minimized the castor oil mediated diarrhea by 16.96% (p p C. gigantea showed mild sensitivity against almost all the tested strains but it shows high consistency of phenolic content and yielded 67.68 μg/mL of IC50 value in the DPPH test. In the PASS prediction, selected isolated compounds have demonstrated significant antidiarrheal and antimicrobial activity following the Lipinski drug rules which have ascertained efficacy with the compounds in molecular docking study. Conclusion The results of this scientific research reflects that the methanol soluble extract of C. gigantea is safe and may provide possibilities of alleviation of diarrhea along with being a potential wellspring of antioxidant and antimicrobial agents which can be considered as an alternate source for exploration of new medicinal products in near future.

Tetrastigma leucostaphylum (Family: Vitaceae) is popular for its medicinal value in Bangladeshi tribal communities. This study aims to investigate several pharmacological values of methanol extract of T. leucostaphylum (METL). In vivo analgesic and anti-inflammatory researches have been implemented by using acetic acid-induced writhing and formalin-induced paw licking test protocols in mice. Furthermore, in vitro thrombolytic and anthelmintic studies have been performed by following the blood clot lysis method and nematode mortality measurement method. In the in vivo study, METL did not minimize the acetic acid-induced writhes prominently but significantly attenuate both the peripheral and inflammatory pain in mice in a dose-dependent manner. In early and late phase, METL 400 (mg/kg, b.w; p.o) showed 39.63 % and 48.73 % paw licking inhibition. Again, METL (100 µL) reflected 56.62 % clot lysis in thrombolytic research. Besides, METL causes the death of nematodes in a dose-dependent manner. The bioassay of the methanol extract of T. leucostaphylum justified the analgesic, anti-inflammatory, thrombolytic and anthelmintic activities of the crude extract and finally suggests the test extract as a wellspring of anti-inflammatory, thrombolytic and anthelmintic agents as a crude drug source.
