Poultry production plays an important role in the economy and livelihoods of rural households in Kenya. As part of a surveillance program, avian influenza virus (AIV)-specific real-time RT-PCR (RRT-PCR) was used to screen 282 oropharyngeal swabs collected from chickens at six live bird markets (LBMs) and 33 backyard poultry farms in Kenya and 8 positive samples were detected. Virus was isolated in eggs from five samples, sequenced, and identified as H9N2 low pathogenic AIV (LPAIV) G1 lineage, with highest nucleotide sequence identity (98.6-99.9%) to a 2017 Ugandan H9N2 isolate. The H9N2 contained molecular markers for mammalian receptor specificity, implying their zoonotic potential. Virus pathogenesis and transmissibility was assessed by inoculating low and medium virus doses of a representative Kenyan H9N2 LPAIV isolate into experimental chickens and exposing them to naïve uninfected chickens at 2 -days post inoculation (dpi). Virus shedding was determined at 2/4/7 dpi and 2/5 days post placement (dpp), and seroconversion determined at 14 dpi/12 dpp. None of the directly-inoculated or contact birds exhibited any mortality or clinical disease signs. All directly-inoculated birds in the low dose group shed virus during the experiment, while only one contact bird shed virus at 2 dpp. Only two directly-inoculated birds that shed high virus titers seroconverted in that group. All birds in the medium dose group shed virus at 4/7 dpi and at 5 dpp, and they all seroconverted at 12/14 dpp. This is the first reported detection of H9N2 LPAIV from Kenya and it was shown to be infectious and transmissible in chickens by direct contact and represents a new disease threat to poultry and potentially to people., Competing Interests: Declaration of Competing Interest The authors declare that they have no conflict of interests. Background & Objectives: The susceptibility of influenza viruses to neuraminidase inhibitors (NAIs) is studied using enzyme-based assays, sequence analysis and in vitro and in vivo studies. Oseltamivir carboxylate (OC) is the active prodrug of the NAI oseltamivir. There is lack of information on the use of embryonated chicken eggs for studying susceptibility of highly pathogenic avian influenza (HPAI) H5N1 viruses to antiviral drugs. The aim of the present study was to assess the use of 10 day old embryonated chicken eggs for studying antiviral susceptibility of HPAI H5N1 viruses., Methods: Two HPAI H5N1 viruses isolated from India were used in the study. Fluorescence-based NAI assay was performed to determine antiviral susceptibility of these viruses. In ovo antiviral assays were carried out using 10 day old embryonated chicken eggs. The virus dilutions were incubated with 14 μg/ml of OC and inoculated in the allantoic cavity. In the eggs, 50 per cent egg infectious dose (EID 50 ) titres as well as mortality were quantitated., Results: The two viruses used were susceptible to OC in the NAI assay. It was found that there was a significant drop in EID 50 titres; however, no significant protection from mortality after OC treatment was observed., Interpretation & Conclusions: By measuring viral titres, the egg model was suitable to study the susceptibility of HPAI viruses to antiviral drugs along with NAI assay. Influenza is a viral infectious disease considered as a source of many health problems and enormous socioeconomic disruptions. Conventional methods are inadequate for in-field detection of the virus and generally suffer from being laborious and time-consuming. Thus, studies aiming to develop effective alternatives to conventional methods are urgently needed. In this work, we developed an approach for the isolation and detection of influenza A virus subtype H9N2. For this aim, two specific influenza receptors were used. The first, anti-matrix protein 2 (M2) antibody, was attached to iron magnetic nanoparticles (MNPs) and used for the isolation of the virus from allantoic fluid. The second biomolecule, Fetuin A, was attached to an electrochemical detectable label, gold nanoparticles (AuNPs), and used to detect the virus tacking advantage from fetuin-hemagglutinin interaction. The MNP-Influenza virus-AuNP formed complex was isolated and treated by an acid solution then the collected gold nanoparticles were deposited onto a screen printed carbon electrode. AuNPs catalyzes the hydrogen ions reduction in acidic medium while applying an appropriate potential, and the generated current signal was proportional to the virus titer. High pathogenicity avian influenza virus (HPAIV) infections in chickens negatively impact egg production and cause egg contamination. Previously, vaccination maintained egg production and reduced egg contamination when challenged with a North American H5N2 HPAIV. However, Asian H5N1 HPAIV infection has some characteristics of increased pathogenicity compared to other H5 HPAIV such as more rapid drop and complete cessation in egg production. Sham (vaccinated at 25 and 28 weeks of age), inactivated H5N1 Once (1X-H5-Vax; vaccinated at 28 weeks of age only) and inactivated H5N1 Twice (2X-H5-Vax; vaccinated at 25 and 28 weeks of age) vaccinated adult White Leghorn hens were challenged intranasally at 31 weeks of age with 6.1 log10 mean embryo infectious doses (EID50) of clade 2.3.2.1a H5N1 HPAIV (A/chicken/Vietnam/NCVD-675/2011) which was homologous to the inactivated vaccine. Sham-vaccinated layers experienced 100% mortality within 3 days post-challenge; laid soft and thin-shelled eggs; had recovery of virus from oral swabs and in 53% of the eggs from eggshell surface (35%), yolk (24%), and albumin (41%); and had very high titers of virus (average 7.91 log10 EID50/g) in all segments of the oviduct and ovary. By comparison, 1X- and 2X-H5-Vax challenged hens survived infection, laid similar number of eggs pre- and post-challenge, all eggs had normal egg shell quality, and had significantly fewer contaminated eggs with reduced virus quantity. The 2X-H5-Vax hens had significantly higher HI titers by the day of challenge (304 GMT) and at termination (512 GMT) than 1X-H5-Vax hens (45 GMT and 128 GMT). The current study demonstrated that AIV infections caused by clade 2.3.2.1a H5N1 variants can be effectively controlled by either double or single homologous vaccination., (Published by Elsevier Ltd.)

Chicks possess maternally derived antibody (MDA) against pathogens and vaccines previously encountered by the dams. This passive immunity is important in early life, when the immune system is immature and unable to fight off infection. On the other hand, MDA can also affect the development of the immune system and interfere with vaccination against avian diseases such as Newcastle disease (ND) and avian influenza (AI). The effect of MDA is generally investigated by studying the progeny of vaccinated dams, which is time-consuming, poorly flexible and expensive. Moreover, the antibody titres obtained are not homogeneous. In this study, a model was developed to offer a faster, more reproducible and cheaper way to study passive immunity in specific pathogen free chickens by injection of a polyclonal serum into the egg yolk at embryonic day 14, combined with an intraperitoneal injection at day 1. A satisfactory model, with consistent, homogeneous antibody titres, as well as persistence close to natural passive immunity, could be obtained for ND virus. On the other hand, the application of this optimized protocol in an H5 AI context induced only a low artificial passive immunity compared with that described in the literature for the progeny of AI vaccinated dams. This artificial model should facilitate future studies regarding the effect of passive immunity on vaccine efficacy at a young age and its effect on immune system development.

Infection with high-pathogenicity avian influenza virus (HPAIV) has been associated with a wide range of clinical manifestations in poultry, including severe depression in egg production and isolation of HPAIV from eggs laid by infected hens. To evaluate the pathobiology in the reproductive tract of chickens, adult hens were inoculated intranasally with 3 HPAIV strains. All 3 strains induced lesions in the reproductive tract 36 to 72 hours after inoculation. Positive immunostaining was observed in all segments of the reproductive tract, occurring predominantly in stromal cells and superficial germinal epithelium of the ovary, in mucosal epithelial cells and less often glandular epithelium throughout the oviduct, and in vascular endothelium. This study generates important data and explains previously reported virus isolation from yolk, due to ovarian virus replication, and virus recovery from albumin, due to virus replication in epithelial cells in several segments of the oviduct.

The 2009 swine-origin influenza A virus (H1N1) and its initial reassortant vaccine strains did not grow well in embryonated eggs. The glutamine to arginine mutation at the amino acid position 223 (Q223R) of the hemagglutinin (HA) gene is the major mutation previously found in egg-adapted 2009 H1N1 strains and shown to enhance viral growth in embryonated eggs. However, the effect of this mutation on the receptor-binding preference had not been directly demonstrated. In this study, the Q223R mutation was shown to change the viral HA binding preference from the human-type receptor, α2,6-linked sialic acid, to the avian-type receptor, α2,3-linked sialic acid; and to enhance the viral growth in embryonated eggs but not in cell culture.

Even though low pathogenic avian influenza viruses (LPAIv) affect the poultry industry of several countries in the world, information about their transmission characteristics in poultry is sparse. Outbreak reports of LPAIv in layer chickens have described drops in egg production that appear to be correlated with the virus transmission dynamics. The objective of this study was to use egg production data from LPAIv infected layer flocks to quantify the within-flock transmission parameters of the virus. Egg production data from two commercial layer chicken flocks which were infected with an H7N3 LPAIv were used for this study. In addition, an isolate of the H7N3 LPAIv causing these outbreaks was used in a transmission experiment. The field and experimental estimates showed that this is a virus with high transmission characteristics. Furthermore, with the field method, the day of introduction of the virus into the flock was estimated. The method here presented uses compartmental models that assume homogeneous mixing. This method is, therefore, best suited to study transmission in commercial flocks with a litter (floor-reared) housing system. It would also perform better, when used to study transmission retrospectively, after the outbreak has finished and there is egg production data from recovered chickens. This method cannot be used when a flock was affected with a LPAIv with low transmission characteristics (R(0)<2), since the drop in egg production would be low and likely to be confounded with the expected decrease in production due to aging of the flock. Because only two flocks were used for this analysis, this study is a preliminary basis for a proof of principle that transmission parameters of LPAIv infections in layer chicken flocks could be quantified using the egg production data from affected flocks., (Copyright © 2012 Elsevier B.V. The negative impact of high pathogenicity avian influenza virus (HPAIV) infection on egg production and deposition of virus in eggs, as well as any protective effect of vaccination, is unknown. Individually housed non-vaccinated, sham-vaccinated and inactivated H5N9 vaccinated Once or Twice adult White leghorn hens were challenged intranasally/intratracheally 3-weeks post-vaccination with H5N2 HPAIV. The non-/sham-vaccinated layers experienced 100% mortality (0% survivability) within 3-4 days post-challenge (DPC), and major changes to reproductive parameters including precipitous drops in egg production (79-0% in <5 days), production of soft and thin-shelled eggs, and deposition of virus in albumin and yolk, and on the egg shell surface of 53% of eggs. By comparison, the three H5-vaccinated groups had 83%, 100% and 100% survivability after challenge; the two H5-vaccinated Once hens that died had low pre-challenge HI titers (GMT=16). H5-vaccinated Once or Twice groups maintained egg production after challenge (63%), but there was a mild and significant reduction in egg production as compared to pre-challenge egg production (79%). H5-vaccinated groups had reduced number of virus contaminated eggs (28%), and in most groups, reduced quantity of virus in contaminated eggs compared to non-/sham-vaccinated groups. No HPAIV-positive eggs were laid on or after 5 DPC. In conclusion, HPAIV infection had major negative impact on egg production and other reproductive parameters. H5-vaccination Once or Twice prevented declines in egg production after HPAIV challenge, reduced number of virus-infected eggs, and typically reduced the titer of virus in internal contents and on eggshell surface., (Published by Elsevier Ltd.)

Circulation of an antigenically variant lineage of highly pathogenic avian influenza (HPAI) H5N1 virus in chicken breeder flocks in Egypt is a continuing problem. The protective efficacy of multiple repeated vaccinations using the currently available H5N2 vaccines is unclear. Here, broiler breeder chickens were vaccinated at weeks 6, 12 and 18 with an inactivated H5N2 commercial vaccine. HI-titer against an Egyptian H5N1 field isolate of classic clade 2.2.1 (EGYcls/H5N1) were significantly lower after the first immunization but increased after booster vaccinations. In contrast, no HI titers were induced against an antigenically distinct field virus of the variant lineage of clade 2.2.1 (EGYvar/H5N1). Upon challenge at week 50 mild, if any, clinical signs were observed in the group infected with EGYcls/H5N1 although one of eight (12.5%) birds died. Mortality reached 6/8 (75%) in the EGYvar/H5N1 challenge group. Virus excretion in all vaccinated groups was reduced in amplitude, but in vaccinated surviving birds, time of virus excretion was extended to up to ten days. Strikingly, challenged vaccinated birds kept laying eggs almost throughout the observation period. Virus was detected on the outer egg-shell of 17 of 40 eggs. The majority of the infected eggs were derived from the EGYcls/H5N1 challenged animals; here the virus was detected also in the yolk and albumin. Repeated vaccination using a commercial H5N2-based vaccine broadened the antigen profile of induced antibodies but did not provide adequate protection against heterologous virus variant. In addition, the observation of AIV contaminated eggs from infected flocks highlights the risk of silent virus spread by vaccinated animals and point to eggs as a possible vector., (Copyright © 2011. The extensive circulation of the H5N1 highly pathogenic avian influenza (HPAI) virus in animals and the human health implications which it poses have led to extensive research in unexplored fields and thus a re-assessment of our understanding of this infection. Moreover, widespread infection of poultry has raised concerns about the food safety and trade implications of this infection, necessitating revised international trade regulations. The role of wild birds has been much debated and resources have been invested to clarify the role that they may play in the spread of infection. It is now clear that wild birds may be responsible for primary introduction in a previously free area. To date it is still unclear whether HPAI infection may be maintained in wild bird populations for extended periods of time. This paper reviews existing knowledge on the transboundary spread of HPAI through poultry and poultry commodities and summarises evidence of spread through wild birds.

In May 2009, during routine monitoring of a commercial layer flock of about 87,000 birds kept in cages in 4 different houses that had been vaccinated 3 times with an inactivated H5N1 vaccine at weeks 1, 7, and 16, highly pathogenic avian influenza (HPAI) virus of subtype H5N1 was isolated and detected by real-time reverse transcriptase-polymerase chain reaction (RT-PCR) in tracheal and cloacal swabs collected from houses 3 and 4; 7 days after onset of clinical signs, there was an increase in mortality accompanied by a decrease in egg production and egg quality. In addition, using RT-PCR, the viral RNA could be detected from albumin and eggshell as well. Seven days after the onset of the clinical signs, the hemagglutination inhibition (HI) titers in the affected houses were 3.2 and 1.9 log2. In the other two houses, there were no clinical signs, and all tested samples were negative using virus isolation and real-time RT-PCR. The HI titers were 6.6 and 7.0 log2 in nonaffected houses. The isolated virus from egg albumin showed high nucleotides and amino-acid identities and clustered with viruses from recently H5N1-confirmed human infections and poultry from different places in Egypt. Moreover, several amino-acid substitutions of viral H5 protein were observed. The vaccinal break seems to be associated with immune escape mutants and/or improper vaccination. The role of contaminated eggs as a source of infection and as a vehicle for spread of the virus should be considered in area with avian influenza outbreaks.

The aim of this study was to develop and evaluate an early warning system (EWS) for commercial egg laying flocks to detect the subtle mortality and egg production changes that characterize low pathogenic avian influenza virus (LPAIV) infections. An EWS will create an alert when the recommended 'trigger point' is reached or exceeded. Previously used EWSs are based on fixed alert levels, while the proposed EWS customizes the alert level to each flock. While a fixed approach may be valid for highly pathogenic diseases, it results in a lower detection probability for low pathogenic diseases. The EWS was based on daily data collected from flocks affected by the 2000-2004 H6N2 LPAI epidemic in California. Three EWSs were evaluated: (1) EWS1, which is triggered when the observed mortality increase or production decrease exceeds more than "x" times the expected daily value (2.75-3.50 times the expected mortality), (2) EWS2, which is triggered when the observed mortality increase or production decrease exceeds more than "y" times during each of 2 consecutive days the expected daily values (1.75-2.15 times the expected mortality), and (3) a combination of the two. The EWSs were evaluated according to three parameters: detection delay (days) of a LPAI outbreak, false alerts (%) and outbreaks missed (%). Results showed that an egg production-based EWS added no benefit to a mortality-based system, mainly because H6N2 LPAI-related egg production decrease always occurred after increase in mortality. Combining the two EWSs resulted in a reduced detection delay and no missed outbreaks, but at the expense of a slight increase in the number of false alerts triggered. The system presented in this study also outperformed fixed EWSs in all three evaluated parameters. The proposed EWS, if used as part of a poultry cooperative program and combined with a rapid laboratory diagnosis, could be a useful tool in the detection and control of LPAI outbreaks and other poultry diseases. Built in a spreadsheet, the system could be inexpensively, easily and quickly incorporated into a commercial egg production farm decision support system. In addition, the proposed system could be quickly adjusted to changing epidemic situations, and easily customized to individual flocks.

Intercontinental movements of northern pintail (Anas acuta) ducks wintering in Japan create a high-risk of both incursion and dispersion of avian influenza viruses (AIVs) that circulate in the ducks' breeding grounds in Siberia and Alaska. This predisposition is likely amplified by bi-directional conveyance of AIVs between Japan and China. In this study, H5N2 viruses were characterized by means of HA cleavage site sequencing and found to be low pathogenic. Through entire genome analysis, as well as in ovo and in vitro pathogenicity tests, one isolate - A/northern pintail/Akita/714/06 H5N2 (Akita/714/06 H5N2) - was characterized. Comparative molecular analysis revealed that genes of this virus have 97.5-99.6% and 96.7-100% likeness at nucleotide and at amino acid level, respectively, with genes of different subtypes of viruses isolated from China, Korea, Russia, and Italy. Phylogenetically, Akita/714/06 (H5N2) clustered with viruses isolated from Eurasian countries. Partial affinity to a recent Korean porcine strain is noticeable. Related evolutionary, ecological, and phenotypic aspects were discussed, thereby signifying the importance of the described pathogen-host-ecosystem interfaces.

Real-time reverse transcription polymerase chain reaction (real-time RT-PCR) is routinely used for the rapid detection of Avian influenza virus (AIV) in clinical samples, but inhibitory substances present in some clinical specimens can reduce or block PCR amplification. Most commercial RNA extraction kits have limited capacity to remove inhibitors from clinical samples, but using a modified commercial protocol (Ambion MagMAX, Applied Biosystems, Foster City, CA) with an added high-salt wash of 2 M NaCl and 2 mM ethylenediamine tetra-acetic acid was shown to improve the ability of the kit to remove inhibitors from cloacal swabs and some tissues. Real-time RT-PCR was carried out in the presence of an internal positive control to detect inhibitors present in the purified RNA. Cloacal swabs from wild birds were analyzed by real-time RT-PCR comparing RNA extracted with the standard (MagMAX-S) and modified (MagMAX-M) protocols. Using the standard protocol on 2,668 samples, 18.4% of the samples had evidence of inhibitor(s) in the samples, but the modified protocol removed inhibitors from all but 21 (4.8%) of the problem samples. The modified protocol was also tested for RNA extraction from tissues using a TRIzol-MagMAX-M hybrid protocol. Tissues from chickens and ducks experimentally infected with high-pathogenicity Asian H5N1 AIV were analyzed by real-time RT-PCR, and the limit of detection of the virus was improved by 0.5-3.0 threshold cycle units with the RNA extracted by the MagMAX-M protocol. The MagMAX-M protocol reported in the present study can be useful in extracting high-quality RNA for accurate detection of AIV from cloacal swabs and tissues by real-time RT-PCR.

The extensive circulation of the H5N1 highly pathogenic avian influenza virus, and the human health threat that it poses, has raised concerns over the food safety implications of this virus infecting poultry. In addition, among the most important risk factors for the possible emergence of avian influenza in the European Union and the United States, the European and Food Safety Agency and the US Department of Agriculture's Animal and Plant Health Inspection Service, respectively, have identified legal and illegal importations of infected poultry commodities. The present paper reviews existing knowledge on the presence of viable avian influenza viruses in poultry commodities.

The panzootic of the H5N1 strain of highly pathogenic avian influenza has become an international crisis. All parts of the world are now considered at risk due to trade globalisation, with the worldwide movement of animals, products and humans, and because of the possible spread of the virus through the migration of wild birds. The risk of introducing notifiable avian influenza (NAI) through trade depends on several factors, including the disease status of the exporting country and the type of products. The highest risk occurs in the trade of live birds. It is important to assess and manage these risks to ensure that global trade does not result in the dissemination of NAI. However, it is also important that the risk of infection is not used as an unjustified trade barrier. The role of the regulatory authorities is thus to facilitate the safe trade of animal products according to international guidelines. Nevertheless, the balance between acceptable risk and safe trade is difficult to achieve. Since the movements of poultry and birds are sometimes difficult to trace, the signature or 'identity card' of each isolated virus can be very informative. Indeed, sequencing the genes of H5N1 and other avian influenza viruses has assisted greatly in establishing links and highlighting differences between isolates from different countries and tracing the possible source of introduction. Recent examples from Asia, Europe and Africa, supported by H5N1 molecular fingerprinting, have demonstrated that the sources of introduction can be many and no route should be underestimated.

H9N2 influenza viruses are endemic in many Asian countries including China and Korea, and cause a considerable economic loss to chicken industry by reduction in egg production and about 30% mortality. Here we developed live cold-adapted attenuated H9N2 influenza vaccine by adaptation of viruses in hen's eggs at 25 degrees C. Genetic analysis shows that the cold-adapted H9N2 (A/Chicken/Korea/S1/03) viruses contain a total of 44 amino acid substitutions, of which 7 amino acids are identical to the loci identified in the cold-adapted H2N2 (A/Ann Arbor/6/60) vaccine strain compared to genes in wild-type H9N2 (A/Chicken/Korea/S1/03) influenza viruses. When cold-adapted H9N2 (A/Chicken/Korea/S1/03) influenza viruses were inoculated in layers viruses were detectable in the tracheas, not in the lungs, no reduction of egg production and mortality was observed in contrast to the infection of wild-type H9N2 influenza viruses, and CD8+ T lymphocytes expressing IFN-gamma were induced. When layers vaccinated with cold-adapted attenuated H9N2 (A/Chicken/Korea/S1/03) influenza viruses were challenged with wild-type H9N2 (A/Chicken/Korea/521/04) influenza viruses, they were protected from the loss of egg production and mortality. Our results suggest that cold-adapted attenuated H9N2 vaccine can be used for controlling the infection of H9N2 influenza viruses in chickens.

Outbreaks of highly pathogenic avian influenza (HPAI) (2000-2003) resulted in 50 million EU birds culled or dead. The circulation of H5N1 in Asia could represent the origin of a human pandemic. Questions have been raised to combat the ongoing AI crisis. HPAI H5N1 has spilled over to resident and migratory wild bird populations which could represent a means of the virus reaching the EU, but lack of data make any forecast imprudent. Poultry holdings located close to migratory bird breeding and resting sites are considered at greater risk of exposure and methods to prevent exposure should be implemented. Legal safeguards for importation of poultry commodities currently only apply to HPAI and rely on detection of clinical signs that may not be observable during incubation period. Illegal imports represent an additional risk. Insufficient data on the effectiveness of commodity processing are available and few indications can be deducted. Biosecurity is the primary tool to prevent AI introduction and secondary spread. Massive spread was observed in densely populated poultry areas resulting in vaccination programs. Vaccination should be used to support eradication together with enhanced biosecurity and restriction measures, which shall also be implemented in case of prophylactic vaccination. Animal welfare aspects of AI include use of appropriate culling methods, correct vaccine application, and availability of trained staff. EFSA has recently set up a new scientific work group to further assess the risk of HPAI introduction and spread posed in particular by wild, migratory birds, as well as further follow-up of recent AI developments.

Until recently, most vaccines against avian influenza were based on oil-emulsified inactivated low- or high-pathogenicity viruses. Now, recombinant fowl pox and avian paramyxovirus type 1 vaccines with avian influenza H5 gene inserts (+ or - N1 gene insert) are available and licensed. New technologies might overcome existing limitations to make available vaccines that can be grown in tissue culture systems for more rapid production; provide optimized protection, as a result of closer genetic relations to field viruses; allow mass administration by aerosol, in drinking-water or in ovo; and allow easier strategies for identifying infected birds within vaccinated populations (DIVA). The technologies include avian influenza viruses with partial gene deletions, avian influenza-Newcastle disease virus chimeras, vectored vaccines such as adenoviruses and Marek's disease virus, and subunit vaccines. These new methods should be licensed only after their purity, safety, efficacy and potency against avian influenza viruses have been demonstrated, and, for live vectored vaccines, restriction of viral transmission to unvaccinated birds. Use of vaccines in countries affected by highly pathogenic avian influenza will not only protect poultry but will provide additional safety for consumers. Experimental studies have shown that birds vaccinated against avian influenza have no virus in meat and minimal amounts in eggs after HPAI virus challenge, and that replication and shedding from their respiratory and alimentary tracts is greatly reduced.

Avian influenza virus (AIV) was recovered from the internal contents of eggs, including mixture of albumen and allantoic fluid, and from the oviduct of naturally infected Japanese quail (Coturnix coturnix japonica) flocks in the southern part of Thailand. The virus titers of 10(4.6)-10(6.2) ELD(50)/mL were directly measured from the internal content of infected eggs. The virus was isolated by chorioallantoic sac inoculation of embryonating chicken eggs. Infected allantoic fluid was identified as hemagglutinating virus and then was indicated the presence of H5 hemagglutinin. The virus was confirmed to be H5N1 subtype influenza A virus by reverse transcriptase-polymerase chain reaction. Additionally, real-time reverse transcriptase-polymerase chain reaction assay could specifically detect influenza virus subtype H5. Furthermore, indirect fluorescent antibody (IFA) test by using specific anti-influenza A monoclonal antibody indicated that virus antigens were detected in the parenchyma of multiple tissues. Systemic localization of viral antigen detected was certainly considered to be viremic stage. In addition, influenza virus antigen was also detected by IFA in allantoic fluid sediments isolated from internal content of egg or oviduct. The conclusion of isolated AIV type A subtype H5N1 from these two infected materials was correlated to the viremic stage of infection because the virus antigens could be observed in almost all tissues. Conclusively, the need for adequate safeguards to prevent contamination and spread of the virus to the environment during movement of eggs--including hatching eggs, cracked eggs, and other relevant infected materials-- or egg consumption from area of outbreak is emphasized and must not be ignored for the reasons of animal, public, and environmental health.

Three H5N1 influenza viruses were isolated from shell washes of duck and goose eggs confiscated from travelers coming from Vietnam. All eight gene segments of these viruses share high sequence identity with the H5N1 avian influenza viruses that caused outbreaks in poultry and humans in Hong Kong in 1997. Animal studies indicate that these isolated viruses are able to replicate in mouse lung and could be found in the organs of ducks without causing any clinical signs or death. However, the viruses are highly pathogenic for chickens. Although the source of these recently isolated Hong Kong 97-like H5N1 viruses is undetermined, their detection in the egg shell of duck and goose suggests that this particular genotype of H5N1 virus may have re-emerged in nature or may have been circulating continuously.

Using clinical materials from experimentally infected poultry, we established an effective method for the preparation of viral RNA directly from tissue samples and eggs. Furthermore, our type A-specific matrix reverse transcription-polymerase chain reaction (RT-PCR) test was improved, and an H7 subtype-specific nested RT-PCR, which includes the hemagglutinin cleavage site, was designed. Both RT-PCR systems proved to be as sensitive as virus isolation. In addition, the labeled H7 HA-nested PCR primers were suitable for sequencing of the PCR products. The RT-PCR amplification of viral RNA and sequencing of the PCR product allows for the sensitive and rapid differentiation between low-pathogenic and highly pathogenic avian influenza viruses.

The 1985 outbreak of high-pathogenicity avian influenza (HPAI) in Victoria, Australia, took 5 days to confirm by standard laboratory tests, during which time infected chickens continued excreting virus, thus creating the opportunity for transmission to other farms. An immunofluorescence test for the detection of viral antigen in tissue impression smears was evaluated as a rapid diagnostic test for HPAI virus infections of poultry. Several test configurations were compared for background reactions and strength of fluorescence, with the optimum combination found to be an influenza A group-specific monoclonal antibody, detected by an anti-mouse fluorescein isothiocyanate conjugate. Immunohistochemical examination of tissues from chickens experimentally infected with low-pathogenicity and HPAI viruses identified the pancreas as the organ most consistently containing high concentrations of HPAI viral antigen. This test has since been used in Australia in the rapid laboratory confirmation of three avian influenza outbreaks and in showing that numerous other suspect cases were not caused by avian influenza.

Between January 1997 and March 1998, 11 cases of H7N2 avian influenza (nonpathogenic) were diagnosed at the Laboratory of Avian Medicine and Pathology, Kenneth Square, PA. These cases involved either commercial leghorn laying hens or leghorn pullets raised in Pennsylvania. Grossly and histologically, the most striking lesion associated with disease was salpingitis, usually with edema and occasionally with oviduct necrosis. Fluid, fibrinous, and egg yolk material in the peritoneum (egg yolk peritonitis) as well as pulmonary congestion and pulmonary edema were also frequently seen. Oviduct lesions have rarely been described in association with avian influenza infections in previous outbreaks. Mortality in affected houses was mild to moderate (less than 4% total mortality during the outbreak), with concurrent mild to moderate egg production declines (2%-4% at the time of disease onset).

Serologic screening of avian sera for group-specific antibodies to type A influenza is currently accomplished by using the avian influenza (AI) agar gel immunodiffusion (AGID) test. A competitive enzyme-linked immunosorbent assay (CELISA) was developed using a baculovirus vector, Autographa californica nuclear polyhedrosis virus, expressing the nucleoprotein (NP) gene of A/Ann Arbor/6/60 influenza virus. The recombinant NP was obtained by inoculation of Spodoptera frugiperda (Sf9) insect cells or Trichoplusia ni insect larvae with the recombinant baculovirus. A hybridoma cell line producing monoclonal antibody against influenza virus A nucleoprotein was used to generate mouse ascitic fluid for the CELISA. The nucleoprotein and the monoclonal antibody were used without further purification in a CELISA for detection of avian-origin serum antibodies to type A influenza. The AI AGID and CELISA tests were compared for sensitivity and specificity using 1651 experimental and reference antisera. Samples discrepant in AGID and CELISA test results were further evaluated by the AI indirect fluorescent antibody (IFA), hemagglutination-inhibition (HI), and neuraminidase-inhibition (NI) tests. The results demonstrated a high degree of correlation between the AGID and CELISA test results, with the IFA, HI, and NI tests offering additional support of CELISA test specificity. The CELISA is a rapid, economical, sensitive, and specific serodiagnostic method for screening large numbers of avian sera for antibodies to avian influenza virus.
