Insecticides efficient against the target species while conserving natural enemies in the agroecosystem are required for IPM. With the imminent discontinuation of fipronil, a broad-spectrum insecticide, ethiprole, which belongs to the same group as phenylpyrazole (2B), and isocycloseram, a novel isoxazoline insecticide with distinct mode of action (30), provide options for controlling boll weevil. The susceptibility of the boll weevil, Anthonomus grandis grandis (Boh.), and two natural enemies [Eriopis connexa (Germar) and Bracon vulgaris Ashmead] to these insecticides were studied. Furthermore, the survival and biological traits of the lady beetle, E. connexa, exposed to fipronil, isocycloseram, and ethiprole were assessed. The LC50s values for fipronil, ethiprole, and isocycloseram for A. grandis grandis were 2.71, 0.32, and 0.025 mg a.i./L, respectively; 0.86, > 200, and 3.21 mg a.i./L for E. connexa; and 2.31, 592.94, and 0.18 mg a.i./L for B. vulgaris, respectively. The recommended rates of ethiprole did not cause mortality in adult lady beetles, although fipronil and isocycloseram were highly toxic. Lady beetle larvae and adults survived more than 80% when exposed to dried residues of ethiprole, but less than 10% when exposed to fipronil and isocycloseram. Lady beetle larvae development, reproduction, and predation rates of adults were similar between ethiprole and the control group. Although fipronil and ethiprole belong to the same insecticide group, the difference in toxicity to boll weevils and natural enemies is presented and discussed. Fluralaner, a systemic pesticide, was originally registered with the US Food and Drug Administration in 2014 under the trade name Bravecto for flea treatment for pets. As a GABA antagonist, the footprint of fluralaner has expended beyond medical and veterinary pests in recent years. In this study, we examined the acute toxicity of fluralaner against three pests of Henosepilachna vigintioctopunctata, Megalurothrips usitatus, and Phyllotreta striolata in the Solanaceae, Fabaceae, and Cruciferae families, respectively, and the sublethal impact of fluralaner on Propylaea japonica, a widely distributed predatory ladybeetle. Based on LC 50 , fluralaner was effective against H. vigintioctopunctata (0.098 mg a.i. L -1 for the second instar larvae), M. usitatus (0.134 mg a.i. L -1 for adult females), and P. striolata (0.595 mg a.i. L -1 for adults). For P. japonica, however, fluralaner was substantially less effective (1.177 mg a.i. L -1 for the third instar larvae). Furthermore, the LC 10 and LC 30 of P. japonica were also consistently higher than the LC 50 of the three pests. In addition, we did not observe any significant impacts of fluralaner at LC 10 and LC 30 on the life history traits, including body weight, developmental time, pre-oviposition period, and fecundity of P. japonica. Filth flies cause billions of dollars of losses annually to the animal production industry. Fluralaner is a relatively new pesticide currently sold for control of fleas, ticks, and mites on companion animals and poultry. We examined the efficacy of fluralaner against three species of filth flies. Insecticide-susceptible horn flies and stable flies were tested topically. Fluralaner outperformed permethrin by > 2-fold for the horn flies but underperformed permethrin by > 45-fold for stable flies at 24 h. House flies were tested topically with fluralaner in comparison to permethrin at 48 h and orally with fluralaner in comparison to imidacloprid at 24 h. Topical fluralaner was 6- to 28-fold as toxic as permethrin in four pyrethroid-resistant strains and not significantly less toxic than permethrin in a susceptible strain and a mildly pyrethroid-resistant strain. There was slight cross-resistance between topically applied fluralaner and permethrin in all five insecticide-resistant strains tested. Oral fluralaner was more toxic than imidacloprid in all four house fly strains tested, 9- to 118-fold as toxic. Oral cross-resistance between imidacloprid and fluralaner was not detected, but imidacloprid resistance was not high in any of the tested strains. Fluralaner shows promise for control of horn flies and house flies.

Background: Fluralaner, a novel pesticide that targets the γ-aminobutyric acid (GABA) receptor (GABAR) subunit of resistant to dieldrin (RDL), exhibits strong potential to be an insecticide to control agricultural insect pests. However, the risk and action of fluralaner to economic insects, e.g., honeybee Apis mellifera Linnaeus, remains unclear., Results: In this study, both oral and contact toxicity of fluralaner to honeybee were found to be 0.13 μg adult -1 . Abamectin, dieldrin, ethiprole, α-endosulfan, fipronil and fluralaner strongly inhibited the GABA-induced current in A. mellifera RDL (AmRDL), expressed in Xenopus laevis oocytes, with median inhibitory concentration (IC 50 ) values of 7.99, 868.1, 27.10, 412.0, 11.21 and 13.59 nM, respectively. The binding free energy and electrophysiological response of AmRDL and insecticides were opposite. The correlation values between toxicity (to A. mellifera) and binding free energy/electrophysiological inhibition (to AmRDL) were at a moderate level., Conclusion: In conclusion, we report for the first time the notable risk of fluralaner to honeybee in vivo and compared the actions of GABAR-targeted insecticides on the AmRDL receptor. © 2019 Society of Chemical Industry., (© 2019 Society of Chemical Industry.)

Background: Leishmania infantum is the parasite responsible for the disease in humans known as zoonotic visceral leishmaniasis (ZVL). Dogs are considered the main domestic reservoir of ZVL and sand flies are the proven vectors. The use of systemic insecticides in dogs has been studied as an alternative strategy to control ZVL in endemic areas. One systemic insecticide in dogs, fluralaner, has a proven anti-sand fly effect in membrane-fed studies. However, the efficacy and duration on sand flies directly feeding from dogs treated with fluralaner remains unknown., Methods: Direct feeding bioassays were performed on 10 beagle dogs that had been randomly assigned to two groups: one with five dogs orally treated with Bravecto® (fluralaner) and other five as a control. About 30 females of Phlebotomus papatasi were allowed to directly feed from dogs at seven days before the administration of the treatment and Days 3, 17, 31, 45 and 73 post-treatment. Sand fly mortality after feeding was observed every 24 h for 5 days. The Kaplan-Meyer method, Henderson-Tilton formula and a negative binomial mixed model were used to respectively calculate: (i) mortality and its 95% confidence interval (CI); (ii) efficacy of the insecticide at killing sand flies in 24 h; and (iii) differences in the risk of sand fly death at 24 h after feeding., Results: Control sand fly mortality 24 h after feeding was always ≤ 20% and mortality in the fluralaner group ranged from 2% (95% CI: 0-4%) 7 days before treatment to 100% at 3 days post-treatment. Fluralaner efficacy was 100, 93, 94 and 75% at Days 3, 17, 31 and 45, respectively (P < 0.0001). The increase in the risk of sand fly death was 32.9 (95% CI: 4-263), 76 (95% CI: 8-705), 95.8 (95% CI: 9-1029) and 10.6 times (95% CI: 1.43-79) on Days 3, 17, 31 and 45, respectively CONCLUSIONS: The efficacy of fluralaner, orally administered to dogs, against sand-flies was above 90% for 31 days. Fluralaner administered to dogs should be further evaluated as a control strategy in ZVL endemic areas.

The increasing occurrence of resistance to chemical insecticides in insect pest populations is a serious threat to the integrity of current pest management strategies, and exploring new alternative chemistries is one important way to overcome this obstacle. Fluralaner, as a novel isoxazoline insecticide, has broad spectrum activity against a variety of insect pests, but little data is available about its effect on Lepidopterans. The effects of fluralaner on Spodoptera litura Fabricius, a widespread and polyphagous pest, were evaluated in the present study. Our results showed younger larvae were more susceptible to fluralaner treatment, but feeding and topical applications were similarly effective in 3rd instar larvae. Synergism assays indicated that piperonyl butoxide (PBO) could increase the toxicity of fluralaner to S. litura to a certain degree and P450 may be involved in the detoxification of fluralaner in vivo. Sublethal developmental effects included reduced larval body weight, decreased pupation and emergence, and notched wings in adults, accompanied by changes in the transcript levels of chitinase 5 (CHT5) and juvenile hormone acid methyltransferase (Jhamt), genes vital for insect development. Above results manifested that fluralaner is highly toxic to S. litura larvae via either topical or oral application and provide an indication of how this insecticide is metabolized in vivo. The ability to recover to original states after disturbances makes macroinvertebrates useful tools for assessing the impacts of pesticides. Many studies showed that direct exposure to pesticides decreases macroinvertebrate richness and alters their composition. The main objective of this study was to assess recovery patterns in macroinvertebrate communities after pesticide application in irrigated rice fields. We analyzed short-term temporal dynamics of macroinvertebrate communities after application of the herbicides bispyribac-sodium and clomazone and the insecticide chlorantraniliprole, over the rice-growing season in southern Brazil. We selected three conventional rice fields and the recovery of macroinvertebrate communities was also compared with three adjacent natural ponds. The study was developed from November 2011 to February 2012 (rice-growing season). Five macroinvertebrate collections were carried out 3, 7, 14, 38, and 60 days after pesticide application (November 25). Rice fields showed lower richness and abundance than ponds in the period immediately after pesticide application, and recovery rates in the richness of macroinvertebrate communities were more conspicuous as pesticide residuals dissipated from the fields. Macroinvertebrate community structure in rice fields also became more similar to natural ponds as pesticide traces were scarcer. However, macroinvertebrate abundance patterns were not related to pesticide concentrations in the fields. Our results supported the general hypothesis on the negative effects of pesticide application on macroinvertebrate community in irrigated rice fields, although other environmental features (e.g., length of the flooded period) also contributed to explain temporal dynamics in the macroinvertebrate communities from irrigated rice fields.

Fluralaner is a novel isoxazoline insecticide which shows high insecticidal activity against parasitic, sanitary and agricultural pests, but there is little information about the effect of fluralaner on non-target organisms. This study reports the acute toxicity, bioconcentration, elimination and antioxidant response of fluralaner in zebrafish. All LC 50 values of fluralaner to zebrafish were higher than 10 mg L -1 at 24, 48, 72 and 96 h. To study the bioconcentration and elimination, the zebrafish were exposed to sub-lethal concentrations of fluralaner (2.00 and 0.20 mg L -1 ) for 15 d and then held 6 d in clean water. The results showed medium BCF of fluralaner with values of 12.06 (48 h) and 21.34 (144 h) after exposure to 2.00 and 0.20 mg L -1 fluralaner, respectively. In the elimination process, a concentration of only 0.113 mg kg -1 was found in zebrafish on the 6th day after removal to clean water. After exposure in 2.00 mg L -1 fluralaner, the enzyme activities of SOD, CAT, and GST, GSH-PX, CarE and content of MDA were measured. Only CAT and CarE activities were significantly regulated and the others stayed at a stable level compared to the control group. Meanwhile, transcriptional expression of CYP1C2, CYP1D1, CYP11A were significantly down-regulated at 12 h exposed to 2.00 mg L -1 of fluralaner. Except CYP1D1, others CYPs were up-regulated at different time during exposure periods. Fluralaner and its formulated product (BRAVECTO ® ) are of low toxicity to zebrafish and are rapidly concentrated in zebrafish and eliminated after exposure in clean water. Antioxidant defense and metabolic systems were involved in the fluralaner-induced toxicity. Among them, the activities of CAT and CarE, and most mRNA expression level of CYPs showed fast response to the sub-lethal concentration of fluralaner, which could be used as a biomarker relevant to the toxicity., (Copyright © 2017 Elsevier Ltd. The serendipitous observation of the insecticidal activity of a candidate herbicide was the first in a series of surprises that changed the course of insecticide research and opened the "Golden Age of Diamide and Isoxazoline Insecticides" which have a common genesis. Two novel modes of action were discovered, one involving the γ-aminobutyric acid (GABA) receptor of the chloride channel and the other the ryanodine receptor (RyR) of the calcium-activated calcium channel. These are old insecticide targets, but physiological assays and radioligand binding studies reveal that the new diamides and isoxazolines act at previously unrecognized sites without cross-resistance to other chemotypes and more important differing between insects and mammals resulting in selective toxicity and mechanistically based safety. The phthalic diamide flubendiamide and anthranilic diamides chlorantraniliprole and cyantraniliprole act at an allosteric site of the RyR to activate calcium release in insects but not mammals. They are the most important insecticide introductions of the past decade. Isoxazoline and meta-diamide insecticides and their previously unrecognized GABA-R target are more recent discoveries. Isoxazolines are currently important in flea and tick control in dogs and cats, and meta-diamides show promise for pest management and crop protection. These 21st century RyR and GABA-R diamides and isoxazolines were serendipitous discoveries and developments showing the importance of mechanism studies in maintaining the arsenal of safe and effective insecticides.

Background: The Asian citrus psyllid, Diaphorina citri (Hemiptera: Psyllidae), is the most destructive pest of citrus in Florida. The development of insecticide resistance in several populations of D. citri has been documented. There is an urgent need to develop and integrate novel tools for the successful management of D. citri and also to prevent the development of insecticide resistance., Results: The effects of a relatively newer chemistry, cyantraniliprole, against D. citri were investigated. The contact toxicity of cyantraniliprole was 297-fold higher against D. citri than its primary parasitoid, Tamarixia radiata (Hymenoptera: Eulophidae). D. citri settled and fed less on cyantraniliprole-treated plants than controls at concentrations as low as 0.025 and 0.125 µg AI mL⁻¹ respectively. D. citri egg production, first-instar emergence and adult emergence were significantly reduced on plants treated with 0.25, 0.02 and 0.25 µg AI mL⁻¹ of cyantraniliprole, respectively, when compared with control plants. Under field conditions, foliar and drench treatments with cyantraniliprole (1436.08 g ha⁻¹) reduced numbers of D. citri adults and nymphs, as well as of a secondary pest, citrus leafminer, Phyllocnistis citrella (Lepidoptera: Gracillariidae), more than a standard insecticide., Conclusions: These results suggest that cyantraniliprole should be a valuable new tool for rotation into D. citri management programs. For insecticide resistance management, cyantraniliprole may be particularly useful for rotation with neonicotinoids. In addition, cyantraniliprole was much less toxic to T. radiata than to D. citri and thus may have less impact on biological control than other currently used broad-spectrum insecticides, such as organophosphates and pyrethroids., (© 2012 Society of Chemical Industry.)

Twenty 3,5-disubstituted isoxazoles have been synthesized and tested against fourth instar Aedes aegypti larvae. In the synthesis of title compounds, modifications have been made in the C-5 side-chain with a view to test their larvicidal activity. These isoxazoles have been obtained by 1,3-dipolar cycloaddition of arylnitrile oxides to terminal alkynes which furnished the desired products in 20% to 79% yields. A comparative study of the larvicidal activity between 3-(3-aryl-isoxazol-5-yl)-propan-1-ols and 3-(3-aryl-isoxazol-5-yl)-propionic acids clearly demonstrated that the latter compounds possess much better larvicidal activity than the former. We also tested two esters, viz., methyl 3-[3-(phenyl)-isoxazole-5-yl] propionate and methyl 3-[3-(4-chlorophenyl)-isoxazole-5-yl] propionate, where the latter presented an excellent larvicidal profile., (Copyright © 2012 Elsevier Ltd.
