Iodine is a trace element required to produce the thyroid hormones, which are critical for development, growth and metabolism. To ensure appropriate population iodine nutrition, convenient and accurate methods of monitoring are necessary. Current methods for determining iodine status either involve a significant participant burden or are subject to considerable intra-individual variation. The continuous secretion of iodide in saliva potentially permits its use as a convenient, non-invasive assessment of status in populations. To assess its likely effectiveness, we reviewed studies analysing the association between salivary iodide concentration (SIC) and dietary iodine intake, urinary iodide concentration (UIC) and/or 24-h urinary iodide excretion (UIE). Eight studies conducted in different countries met the inclusion criteria, including data for 921 subjects: 702 healthy participants and 219 with health conditions. SIC correlated positively with UIC and/or UIE in four studies, with the strength of relationship ranging from r = 0·19 to r = 0·90 depending on sampling protocol, age, and if salivary values were corrected for protein concentration. Additionally, SIC positively correlated with dietary intake, being strongest when saliva was collected after dinner. SIC varied with external factors, including thyroid function, use of some medications, smoking and overall health status. Evidence provided here supports the use of SIC as a viable, low-burden method for determining iodine status in populations. However, small sample sizes and high variability indicates the need for more extensive analyses across age groups, ethnicities, disease states and dietary groups to clarify the relative accuracy and reliability in each case and standardise procedure.

A complete protocol is presented for the speciation analysis of 129 I for both inorganic and organic iodine in seawater using coprecipitation and solid-phase extraction (SPE) combined with accelerator mass spectrometry (AMS). By modifying the iodide separation process and adding a crossover removal step, the improved coprecipitation method significantly reduces the cross-contamination of iodide and iodate to less than 0.05% in the speciation analysis of inorganic 129 I, with the separation efficiencies of about 95% and 93% for iodide and iodate, respectively. The SPE-DOI method for the dissolved organic 129 I (DO 129 I) analysis was developed, whereby we report the first direct observation of DO 129 I/DO 127 I atom ratios in seawater in this paper. 129 I species in seawater from Tokyo Bay were analysed. The 129 I results demonstrated that our protocol for speciation analysis of 129 I is reliable and provided new insights into understanding the iodine cycle.

Despite its paramount role in the functioning of coastal ecosystems, relatively little is known about halogen metabolism in giant kelp (Macrocystis pyrifera). This is an important shortcoming given the potential implications for marine and atmospheric chemical processes in the wide distribution range of Macrocystis. The work presented here constitutes the first in depth investigation of the uptake, efflux, and of the physiological function of iodide in this important kelp species. Iodide uptake and efflux rates were measured in adult sporophytes of Macrocystis under normal and stressed (exogenous hydrogen peroxide and an elicitor-triggered oxidative burst) conditions. Kelp tissue took up iodide according to Michaelis-Menten type kinetics when incubated in seawater enriched with various concentrations of iodide. Upon the addition of exogenous hydrogen peroxide, simulating oxidative stress, a marked efflux of iodide occurred. In situ generation of hydrogen peroxide was elicited in Macrocystis upon the addition of oligomeric degradation products of alginate as well as arachidonic acid and methyl jasmonate constituting a defensive oxidative burst that could be linked to iodine accumulation. H 2 O 2 was detected at the single cell level using dichlorohydrofluorescein diacetate, a fluorogenic probe capable of detecting intracellular H 2 O 2 . When assayed for vanadium haloperoxidase activity, several bromoperoxidase isoforms were detected as well as a single iodoperoxidase. Altogether, the results of this study show that Macrocystis has an elaborate iodine metabolism, which is likely significant for impacting iodine speciation in seawater around kelp beds and for volatile halogen emissions into the coastal atmosphere., (Copyright © 2017 Elsevier Inc. Majority of iodine found in dairy milk comes from the diet and teat disinfection products used during milking process. The objective of this study was to evaluate the effects of 4 iodine-based teat dips on milk iodide concentrations varying in iodine level (0.25% vs. 0.5%, w/w), normal low viscosity dip versus barrier dip, and application method (dip vs. spray) to ensure safe iodine levels in dairy milk when these products are used. The iodine exposure study was performed during a 2-wk period. The trial farm was purged of all iodine-based disinfection products for 21 d during a prestudy "washout period," which resulted in baseline milk iodide range of 145 to 182 ppb. During the experiment, iodine-based teat dips were used as post-milking teat disinfectants and compared to a non-iodine control disinfectant. Milk iodide residue levels for each treatment was evaluated from composited group samples. Introduction of different iodine-based teat disinfectants increased iodide residue content in milk relative to the control by between 8 and 29 μg/L when averaged across the full trial period. However, residues levels for any treatment remained well below the consumable limit of 500 μg/L. The 0.5% iodine disinfectant increased milk iodide levels by 20 μg/L more compared to the 0.25% iodine. Compared to dip-cup application, spray application significantly increased milk iodide residue by 21 μg/L and utilized approximately 23% more teat dip. Objective: To investigate the iodine status and its change among school-aged children in their morning urine and eating salt from 2012 to 2014 in Yuhuan County, Zhejiang Province, China., Methods: Three repeated cross-sectional studies were carried out at a same primary school in 2012, 2013 and 2014, respectively. Three classes were randomly selected from each of 3 to 5 grade by the cluster-stratified sampling every year. Totally, 1343 out of 1350 eligible children aged 8 to 10 years old were involved into this study. Their morning urine and salt eating at home were collected and tested., Results: The overall median of urine iodine was 116.0 μg/L, and no significant change was found over year. The overall proportions of subjects with urine iodine < 50 μg/L, < 100 μg/L, and ≥ 300 μg/L were 4.8%, 38.6% and 3.5%, respectively, and there were little differences over year, gender and grade of children. An overall median of iodine from 1343 salt samples was 0.0 mg/kg and no year difference was statistically observed. The proportions of subjects consumed iodized salt significantly decreased from 25.1% in 2012 to 21.8% in 2013 and to 14.2% in 2014. There was a significant difference in urine iodine between subjects taken iodized salt or not and also a weak positive correlation between salt iodine and urine iodine., Conclusion: The nutritional status of iodine is overall stable, proper and safety in recent 3 years among school children in Yuhuan County. The coverage rate of iodized salt is very low.

The present work describes the development of a sequential injection standard addition method for iodine speciation in bathing waters and seaweeds extracts without prior sample treatment. Iodine speciation was obtained by assessing the iodide and iodate content, the two inorganic forms of iodine in waters. For the determination of iodide, an iodide ion selective electrode (ISE) was used. The indirect determination of iodate was based on the spectrophotometric determination of nitrite (Griess reaction). For the iodate measurement, a mixing chamber was employed (flow batch approach) to explore the inherent efficient mixing, essential for the indirect determination of iodate. The application of the standard addition method enabled detection limits of 0.14 µM for iodide and 0.02 µM for iodate, together with the direct introduction of the target water samples, coastal and inland bathing waters. The results obtained were in agreement with those obtained by ICP-MS and a colorimetric reference procedure. The dissolved iodine species that dominate aquatic systems are iodide, iodate and organo-iodine. These species may undergo transformation to one another and thus affect the formation of iodinated disinfection byproducts during disinfection of drinking waters or wastewater effluents. In this study, a fast, sensitive and accurate method for determining these iodine species in waters was developed by derivatizing iodide and iodate to organic iodine and measuring organic iodine with a total organic iodine (TOI) measurement approach. Within this method, organo-iodine was determined directly by TOI measurement; iodide was oxidized by monochloramine to hypoiodous acid and then hypoiodous acid reacted with phenol to form organic iodine, which was determined by TOI measurement; iodate was reduced by ascorbic acid to iodide and then determined as iodide. The quantitation limit of organo-iodine or sum of organo-iodine and iodide or sum of organo-iodine, iodide and iodate was 5 μg/L as I for a 40 mL water sample (or 2.5 μg/L as I for an 80 mL water sample, or 1.25 μg/L as I for a 160 mL water sample). This method was successfully applied to the determination of iodide, iodate and organo-iodine in a variety of water samples, including tap water, seawater, urine and wastewater. The recoveries of iodide, iodate and organo-iodine were 91-109%, 90-108% and 91-108%, respectively. Individual and simultaneous electrochemical detection of iodide and iodine has been performed via cyclic voltammetry on boron doped diamond (BDD) electrodes in a 1M NaClO(4) (pH 8) solution, representative of typical environmental water conditions. It is feasible to compute accurate calibration curve for both compounds using cyclic voltammetry measurements by determining the peak current intensities as a function of the concentration. A lower detection limit of about 20 μM was obtained for iodide and 10 μM for iodine. Based on the comparison between the peak current intensities reported during the oxidation of KI, it is probable that iodide (I(-)) is first oxidized in a single step to yield iodine (I(2)). The latter is further oxidized to obtain IO(3)(-). This technique, however, did not allow for a reasonably accurate detection of iodate (IO(3)(-)) on a BDD electrode., (Copyright © 2012 Elsevier B.V. Selective turn-on fluorescence detection of I(-) was accomplished using fluorescein isothiocyanate-decorated gold nanoparticles (FITC-AuNPs). FITC molecules, which fluoresce strongly in an alkaline solution, were severely quenched when they were attached to the surface of AuNPs through their isothiocyanate group. Upon the addition of I(-), FITC molecules were detached because of I(-) adsorption on the surface of AuNPs. As a result, released FITC molecules were restored to their original fluorescence intensity. Because I(-) has a higher binding affinity to the surface of Au than do Br(-), Cl(-), or F(-), the FITC-AuNPs obviously have a higher selectivity toward I(-) than toward these other anions. Meanwhile, after IO(3)(-) was reduced to I(-) with ascorbic acid, the detection of IO(3)(-) was successfully achieved using the FITC-AuNPs. Under an optimum pH and AuNP concentration, the lowest detectable concentrations of I(-) and IO(3)(-) using this probe were 10.0 and 50.0 nM, respectively. The FITC-AuNPs provide a number of advantages, including easy preparation, selectivity, sensitivity, and low cost. This unique probe was applied to an analysis of the total iodine in edible salt and seawater.

Reactive iodine species have been suggested to play an important role in the atmosphere (e.g. tropospheric ozone depletion, coastal new particle formation). However, there still exist major uncertainties about their atmospheric chemistry, mostly due to the lack of analytical approaches for the accurate speciation of certain key compounds. In this study, 1,3,5-trimethoxybenzene (1,3,5-TMB)-coated denuder proved to be suitable for the differentiation between gaseous interhalogens (iodine monochloride (ICl), iodine monobromide (IBr)) and molecular iodine (I2) based on a selective collection/derivatization method. The results of the denuder sampling were compared with the results of impinger sampling in water, methanol and carbon tetrachloride solutions of 1,3,5-TMB. ICl and IBr are converted into 1-iodo-2,4,6-trimethoxybenzene (1-iodo-2,4,6-TMB) and 1-bromo-2,4,6-trimethoxybenzene (1-bromo-2,4,6-TMB), respectively, in the denuder systems. The respective collection efficiency is 99.2% for ICl and 92.6% for IBr, at 500mLmin(-1) gas flow rate. The collection efficiency for I2 is lower than 1% in the same denuder system, but significantly increases to about 90% in the aqueous 1,3,5-TMB loaded impinger. The denuder-impinger coupled system was then used to differentiate and to collect the ICl, IBr and I2 gas mixtures, followed by gas chromatography-ion trap mass spectrometry (GC-MS) determination. The precision of the method is in general better than 9.1%. The parameters affecting denuder operation including sampling flow rate, sampling duration, and relative humidity have been evaluated. The presented method provides an attractive protocol for iodine species analysis for atmospheric chemistry research.

A two-channel flow-injection (FI) method is reported for the determination of iodide and iodine by its enhancement effect on the Ru(bpy)(3)(3+)-NADH chemiluminescence (CL) system. The limit of detection (3 s of blank) was 1.0 x 10(-9) mol/L iodide/iodine, with a sample throughput of 60/h. The calibration graphs over the range 1.0-50 x 10(-8) mol/L gave correlation coefficients of 0.9994 and 0.999 (n = 5) with relative standard deviations (RSD; n = 4) of 1.0-2.5%, respectively. The effects of interfering cations, anions and some organic compounds were also studied. The method was applied to iodized salts and pharmaceutical samples and the results obtained were in good agreement with the value quoted. The CL method developed was compared with spectrophotometric method.

Equilibrium dialysis on povidone-iodine-solutions (Braunol, standardized Betadine and non-standardized iso-Betadine reveal that the amount of available iodine, free iodine, iodide and triiodide varies significantly both in the undiluted and diluted forms. These differences are reflected in the different bactericidal activity against Staphyloccus aureus as determined by the standard quantitative in vitro suspension test. The amount of available iodine is not an appropriate measure for an assessment of the microbicidal activity. For this, the free iodine has to be determined by means of equilibrium dialysis. The free iodine concentration in the Braunol concentrate was found to be 22 mg/L, in the standardized Betadine 9.7 mg/L and in the non-standardized Betadine concentrate only 2.1mg/L. Because of the atypical behaviour of iodophores and the increase of free iodine at dilution and because of a bactericidal level of free iodine of 5mg/L, Braunol and standardized Betadine can be employed as disinfectant as such, iso-Betadine has to be diluted before use. Summarizing all results, it can be stated that Braunol is superior to standardized Betadine and unstandardized iso-Betadine both as to the release of free iodine in the undiluted and in the diluted forms as in the killing rate of S. aureus.

The electrochemical oxidation of iodide ion in the presence of 4-hydroxycoumarin (1) was studied using cyclic voltammetry and controlled-potential coulometry. The result indicates that the resulting iodine takes part in a halogenation reaction and reacts with 4-hydroxycoumarin (1). According to the obtained results, a new and simple coulometric titration method with potentiometric end-point detection for the determination of 4-hydroxycoumarin (1) is presented. In the presented method, 2-200 micromol of 4-hydroxycoumarin (1) was successfully determined.

A simple and sensitive gas chromatography (GC) method is described for the trace analysis of iodide anion (iodide) in processed seaweed as an organic derivative. The method is based on the derivatization of aqueous iodide extracted from seaweed with 2-(pentafluorophenoxy)ethyl 2-(piperidino)ethanesulfonate in toluene using tetra-n-hexylammonium bromide as a phase-transfer catalyst. The resulting pentafluorophenoxyethyl iodide is highly responsive to an electron-capture detector (ECD) and was analyzed by GC-ECD, giving a low detection limit of approximately 2.7 nM (2.7 fmol/microL injected). Interferences of some common anions in the analysis of iodide were studied and proved to be minimal. Application of the method to the analysis of iodide in processed seaweed was performed.

The iodine value (iodine number) is an important analytical characteristic of fats and oils. Leading pharmacopeias determine it using iodine monobromide (Hanus method). We used methyl oleate as a simple analog of unsaturated triacylglycerols to identify the products. After performing the reaction in deuterated solvents under pharmacopeial conditions, NMR spectroscopy revealed the presence of the 9, 10-diiodo, 9, 10-dibromo, and 9, 10-bromoiodo adducts, leaving no educt olefin. The prescribed subsequent addition of potassium iodide led to the formation of methyl 9, 10-diiodo and bromoiodo stearate in equal amounts.

A new method for determining iodine had been developed in ICP-AES. An oxidant, NaNO2 and a sample containing iodine ion were mixed in the PHD hydroid generation device, the producing iodine molecules were nebulized in nebulizer and then the iodine gas came into troch directly. The problem that the intensity of iodine decreases with the increase of time was solved. It was found that NaNO2 decreased the effect of Cl- in the low acidity. A detection limit of 14 ng.mL-1 was obtained and the RSD of the measurement for 2 micrograms.mL-1 of iodine was 1.2% (n = 10). The average recoveries of iodine were 99.44%. The method was applied to the determination of salt and seawater.

Background and Objective: Administration of levothyroxine and/or iodide can effectively reduce the volume of endemic goitre. However, TSH suppression during levothyroxine treatment may increase the number of recurrences through the persistence of intrathyroidal iodine deficiency. With special attention paid to the level of levothyroxine, a comparison was made of two dosages of combined levothyroxine and iodide., Patients and Methods: 44 patients with diffuse euthyroid goitre were randomized to two treatment groups. Group A received 100 micrograms levothyroxine + 100 micrograms iodide, group B 75 micrograms levothyroxine + 150 micrograms iodide, all of them for three months. This was followed by three months without the medication. Intrathyroidal iodine concentration was measured at the onset of the study, then three months and six months later. At these same times thyroid volume was measured by ultrasound, as well as urinary iodine and various parameters of thyroid function., Results: Thyroid gland volume was reduced in both groups (group A: -17.3%; group B: -14.8%; p < 0.001). There was no significant difference of intrathyroidal iodine concentration and thyroid volume between both groups. After the treatment period, TSH suppression was more marked in group A, while TSH rise was greater in group B., Conclusions: Both drug combinations resulted in comparable reduction of thyroid volume, while the intrathyroid concentration of iodine remained unchanged. The smaller rise of TSH after the treatment suggests that the dosage of 75 micrograms levothyroxine + 150 micrograms iodine is to be preferred.

Background: The National Aeronautics and Space Administration (NASA) has used iodination as a method of microbial disinfection of potable water systems in U.S. spacecraft and long-duration habitability modules. A review of thyroid function tests of NASA astronauts who had consumed iodinated water during spaceflight was conducted., Methods: Thyroid function tests of all past and present astronauts were reviewed. Medical records of astronauts with a diagnosis of thyroid disease were reviewed. Iodine consumption by space crews from water and food was determined. Serum thyroid-stimulating hormone (TSH) and urinary iodine excretion from space crews were measured following modification of the Space Shuttle potable water system to remove most of the iodine., Results: Mean TSH significantly increased in 134 astronauts who had consumed iodinated water during spaceflight. Serum TSH, and urine iodine levels of Space Shuttle crewmembers who flew following modification of the potable water supply system to remove iodine did not show a statistically significant change. There was no evidence supporting association between clinical thyroid disease and the number of spaceflights, amount of iodine consumed, or duration of iodine exposure., Conclusions: It is suggested that pharmacological doses of iodine consumed by astronauts transiently decrease thyroid function, as reflected by elevated serum TSH values. Although adverse effects of excess iodine consumption in susceptible individuals are well documented, exposure to high doses of iodine during spaceflight did not result in a statistically significant increase in long-term thyroid disease in the astronaut population.

A real-time determination of iodide is proposed which involves the oxidation of iodide with 2-iodosobenzoate in the presence of N,N-dimethylaniline. The reaction is completed within 1 min to yield 4-iodo-N,N-dimethylaniline, which is extracted in cyclohexane and determined by GC-MS. It was also possible to determine iodine by derivatization in the absence of 2-iodosobenzoate, and iodate by its reduction with ascorbic acid to iodide and subsequent derivatization. A rectilinear calibration graph was obtained for 0.02-50 micrograms l-1 iodide with a correlation coefficient of 0.9998. The limit of detection was 8 ng l-1 iodide. The method was applied to the determination of iodate in iodized table salt and free iodide and total iodine in sea-water, and to spiked samples when the recovery was in the range 96.8-104.3% (RSD 1.9-3.6%). A sample clean-up by solid-phase extraction with a LiChrolut EN cartridge is proposed.

Dissolved iodine, iodide, and iodate are determined in environmental matrixes by in situ chemical manipulation and inductively coupled plasma atomic emission spectrometry (ICPAES). The method uses equipment commonly available to most laboratories involved in environmental inorganic analysis. Total dissolved iodine, iodide, and iodate are determined by ICPAES using iodine vapor generation. Total iodine is determined directly by ICPAES after filtration. Total dissolved iodide (I-) is oxidized in situ to iodine by the addition of sodium nitrite in sulfuric acid in a simplified continuous flow manifold. Iodate is determined by prereduction at the instrument before analysis by the in situ oxidation ICPAES procedure. A standard nebulizer produces the gas-liquid separation of the total iodine, which is then quantified by ICPAES at 206.16 nm. The instrument detection limit for the iodine analysis was 0.04 microgram/mL. Recoveries from seawater, saltwater, and freshwater standard reference materials ranged from 85 to 118% and averaged 98%. For samples containing both iodine and iodide, the total is determined with in situ oxidation, iodine is determined without the oxidizing reagents, and iodine is calculated from the difference. For samples containing all 3 species, pre-reduction is used and the iodine and iodide concentrations are subtracted for quantitation of iodate. The analysis is selective for these 3 species (I-, I2, and IO3). A group of 20-30 samples may be analyzed and quantitated for all 3 individual, commonly occurring iodide species in less than 1 h. The procedure is considerably faster than any other reported techniques. This method is especially well-suited to the analysis of small environmental samples.

The difficulty in ion-chromatographic determination of nitrite in aqueous solutions containing a high concentration of chloride arises mainly from incomplete resolution of the peaks for these anions on the separation column whose efficiency is not high. A photometric measurement of iodine formed by a reaction of nitrite with iodide has been found to make it possible to determine, chromatographically, trace amounts of nitrite without any interference from chloride; chloride does not oxidize iodide to produce iodine. The proposed method was based on the separation of nitrite from matrix anions on a silica-based anion-exchange column with a 1.5.10(-3) M phthalate eluent (pH 5.0), followed by photometric measurement of the iodine (as triiodide) formed via a post-column reaction of the separated nitrite with iodide. The optimal conditions for the post-column reaction were established by varying the concentrations of iodide, copper(II) and nitric acid in a post-column-reaction solution and the length of a reaction tube. A calibration graph for nitrite, plotted as peak heights versus concentrations, was linear up to 1.50.10(-5) M (690 ppb). The detection limit, defined at S/N = 3, was 1.00.10(-7) M (4.60 ppb) nitrite. The presence of chloride ions up to 0.01 M did not give any interference to the determination of nitrite. This method was successfully applied to the determination of nitrite in lake water, river water, sewage works water and snow samples without any pretreatment.

In the Xinjiang province of western China, conventional methods of iodine (I) supplementation (i.e, goiter pills and iodinated salt) used to mitigate I deficiencies were ineffectual. However, the recent addition of KIO3 to irrigation waters has proven effective. This study was conducted to determine the effects of I form and concentration on rice (Oryza sativa L.) growth, I partitioning within the plant, and ultimately to assist in establishing guidelines for incorporating I into the human food chain. We compared IO3- vs. I- in order to determine how these chemical species differ in their biological effects. Rice was grown in 48 L aerated tubs containing nutrient solution and IO3- or I- at 0, 1, 10, or 100 micromoles concentrations (approximately 0, 0.1, 1, and 10 mg kg-1 I). The IO3- at 1 and 10 micromoles had no effect on biomass yields, and the 100 micromole treatment had a small negative effect. The I- at 10 and 100 micromoles was detrimental to biomass yields. The IO3- treatments had more I partitioning to the roots (56%) on average than did the I- treatments (36%), suggesting differences in uptake or translocation between I forms. The data support the theory that IO3- is electrochemically or biologically reduced to I- prior to plant uptake. None of the treatments provided sufficient I in the seed to meet human dietary requirements. The I concentration found in straw at 100 micromoles IO3- was several times greater than seed, and could provide an indirect source of dietary I via livestock feeding on the straw.

In this study we present the molecular basis of a total iodide organification defect causing severe congenital hypothyroidism. In the thyroid gland of the patient, thyroid peroxidase (TPO) activity and the iodination degree of thyroglobulin were below detection limits, and no TPO messenger ribonucleic acid was detectable by Northern blot analysis. Denaturing gradient gel electrophoretic analysis of the TPO gene of the patient revealed a homozygous mutation in exon 2. Sequence analysis showed the presence of a 20-basepair duplication, 47 basepairs down-stream of the ATG start codon. This duplication generates a frame shift, resulting in a termination signal in exon 3, compatible with the complete absence of TPO. Both parents of the patient are heterozygous for the same duplication, confirming the recessive mode of inheritance of the mutation.

Clinical, scintigraphic and laboratory findings in a group of 23 hyperthyroid patients with inappropriate total intrathyroidal iodine pool (ITI), measured by X-ray fluorescence, were compared to those of 25 hyperthyroid patients with low ITI. Differences are discussed which may be useful in the diagnosis of this clinical entity.

Relationships between I intake by lactating Holstein cows and iodine concentrations in milk and meat were investigated. Six treatment groups with seven cows assigned to each treatment were fed a basal diet containing .8 mg I/kg alone or supplemented with I at 1, 2, or 4 mg/kg in four 5-wk periods. Basal alone was fed in the first and third periods and the I supplement was potassium iodide in the second period and ethylenediamine dihydroiodide in the fourth period. Iodine concentrations in milk increased with each increase in dietary I from 205 ng/ml for basal periods to 404, 477, and 757 ng/ml when 1, 2, or 4 mg/kg I was fed as potassium iodide; and 467, 535, and 869 ng/ml when 1, 2, or 4 mg/kg I was fed as ethylenediamine dihydroiodide. Concentrations of I in skeletal muscle after the fourth period were not affected by 2 mg/kg I and only increased from 166 to 199 ng/g when supplemental I was 4 mg/kg. Moderate changes in dietary I are quickly reflected in milk I, but I in meat is relatively stable.

Twelve yearling Holstein steers were blocked by weight and assigned to receive one of three dietary I levels (0, 50 or 400 mg ethylenediamine dihydriodide [EDDI]/head/day) for 4 weeks. Average daily gain, body temperature and respiration rate were measured before and during the I feeding period, and no significant differences among treatment groups were found. Serum total I showed a significant increase with each increment of I. When animals were slaughtered, five muscle samples and liver and thyroid samples were dissected for I analysis. Mean total muscle I levels for the 0-, 50- and 400-mg treatment groups were .092, .127 and .406 micrograms/g wet tissue, respectively, each being different (P less than .05) from the other two. I concentrations did not differ statistically between individual muscles within the 0- and 50-mg treatment groups, but in the 400-mg EDDI group, the trapezius (.469 micrograms/g) and biceps brachii (.569 micrograms/g) muscles had higher (P less than .05) I concentrations than the semimembranous (.316 micrograms/g), psoas major (.365 micrograms/g) and longissimus (.307 micrograms/g). Dietary I increased (P less than .05) liver and thyroid I concentrations in the 400-mg treatment group over those observed for the 0- and 50-mg treatment groups.

The adsorption behavior of radioiodine on soils has been studied. Soils were collected in Tokai, Ibaraki Pref . and classified with soil texture and contents of organic matter. Carrier-free radioiodine in iodide form is adsorbed on soil independent of soil properties, while radioiodine in iodate form is more readily adsorbed on soil which is abundant in organic, matter.

Mature human milk samples from young healthy women on an equilibrated diet contained a mean of 81 ng/ml total iodine. Iodide represented a mean of 77% of the total iodine. Of the 22 ng/ml organic iodine, there was about 1 ng thyroxine and triiodothyronine and, after pepsin hydrolysis, up to 40% of organic iodine in monoiodotyrosine form. By electrophoresis, after incubation with radioactive thyroid hormone, we found an absence of binding on thyroxine-binding globulin, but thyroxine and triiodothyronine were bound to albumin with a maximal capacity 50 times higher than in human serum. All these differences did not favor transport of iodinated compounds from maternal serum to milk. In conclusion, milk iodide was taken up by newborn thyroid to make thyroid hormones.

In a total of 195 children and adolescents of both sex (mean age 12.9, range 5-17 years) with endemic non-toxic goitre the thyroidal iodine concentration (IC) was determined using X-ray fluorescent scanning on admission and during iodine (100 micrograms daily) and L-thyroxine (3 micrograms/kg body weight daily) treatment respectively. Additionally the thyroid volume was measured sonographically in a longitudinal study including 46 patients before and after 4-8 months of iodine supplementation (100 micrograms daily). The IC was 305 +/- 144 micrograms/g. It compared well with that of adult goitre patients (288 +/- 109 micrograms/g) and was significantly inferior to the value of normal controls (389 +/- 170 micrograms/g). Under L-thyroxine therapy the IC further decreased (243 +/- 144 micrograms/g), whereas patients receiving iodide showed an increase of the IC (570 +/- 197 micrograms/g). The mean TSH level fell from 2.3 +/- 0.9 microU/ml to 1.4 +/- 0.6 microU/ml. The average T4/TBG (thyroxine binding globulin) ratio showed a slight increase which, however, was not significant. The mean goitre volume decreased by 40%. It was evidenced that iodide is useful not only in the prophylaxis of non-toxic goitre but also as a more physiologic treatment than thyroid hormones, at least for young subjects with simple diffuse goitres.

1. Baboons and cats were given radioactive iodine and, at varying times after the injection, samples of thyroid and non-thyroid lymph and of thyroid venous and peripheral venous blood were collected. The radioactive material in these samples was separated by paper chromatography or by differential precipitation and solvent extraction into three main fractions: inorganic iodide, iodoamino acid iodine and high molecular-weight material (retained at the origin of chromatograms and not extractable by butanol or ethanol). The latter was considered to be iodoprotein.2. Thyroxine was the main component of the radioactive iodoamino acid iodine in thyroid lymph, in thyroid venous and in peripheral blood. The concentration of thyroxine radioactivity in the thyroid lymph was several times greater than that in either thyroid venous or peripheral blood plasma, both before and after giving thyroid stimulating hormone.3. An unidentified thyronine compound that was frequently found in thyroid lymph and also in blood samples contained an appreciable proportion of the radioactive iodine. This substance was probably a metabolite of one of the iodothyronines. In addition, iodotyrosines probably accounted for up to 4% of the radioactivity in the lymph and blood samples but radioactive triiodothyronine could not be detected with certainty.4. In some experiments lymph was obtained from the cervical lymph trunk at a considerable distance from the thyroid gland, in order to avoid operative trauma to the gland. This cervical lymph contained diluted thyroid lymph but even so it contained more radioactivity in the form of iodoamino acid and iodoprotein than did peripheral blood or non-thyroid lymph. Cervical lymph and thyroid lymph both contained less radioactive inorganic iodide than did non-thyroid lymph.5. The administration of thyroid stimulating hormone caused a prolonged rise in the output of radioactive iodoamino acid in the thyroid venous blood and, for a limited period, an increase in the output of radioactive inorganic iodide. At the same time there was an increase in the output of radioactive material in the thyroid lymph. The main part of the latter was due to a rise in the output of iodoprotein.6. The lymphatics and the venous system provide alternative pathways for the outflow of iodine compounds from the thyroid gland. These pathways differ in relative importance for different iodine compounds. For iodoprotein the lymphatic pathway is the important one since there is no appreciable release of this substance into the thyroid venous blood. In the case of inorganic iodide, where there is no appreciable difference in concentration between thyroid lymph and blood, the greater flow of blood makes the venous pathway relatively more important than the lymphatic one. Thyroxine is released into the thyroid lymph in higher concentration than into the blood but the faster flow of blood makes the venous pathway relatively more important.
