Objective: A meta-analysis was conducted to assess the effects of dietary chromium picolinate (CrPic) supplementation on broiler growth performance and to determine whether such effects are regulated by broiler strains, sex, environmental stress, or contextual factors including study area and years., Methods: Eligible studies were identified by searching the Web of Science, Springer, Elsevier, ScienceDirect, Taylor & Francis Online databases. Weighted average differences with corresponding 95% confidence intervals were computed with a random-effects model. We performed subgroup analysis stratified by study area, published years, broiler strains and sex, and environmental stress. Publication bias was assessed with Egger's test method. A total of 15 studies eligible for inclusion., Results: The results indicated that CrPic supplementation significantly improved broiler growth performance and subgroup analysis confirmed this conclusion. We also found that Ross 308 or male broilers might be more sensitive to CrPic supplementation and showed better growth performance. A model was used to obtain the amount of chromium addition under the optimal growth performance, which suggested that the maximum value of average daily gain (ADG) was reached when chromium addition was 1810 μg/kg. The results of the sensitivity analysis showed low sensitivity and high stability of the meta-analysis., Conclusions: CrPic supplementation had a positive effect on the growth performance of broilers, and this meta-analysis provides a more accurate value of chromium addition, which may be beneficial for the practice of the broiler industry., Competing Interests: The authors have declared that no competing interests exist.

Picolinic acid (PiA) is an endogenous metabolite of tryptophan that has been reported to possess a wide range of physiological actions. We investigated the effects of dietary PiA on the metabolism of tryptophan to nicotinamide in growing rats. Feeding an ordinary diet containing 1% PiA to growing rats (6 wk) caused death within a few days. Toxicity of PiA was higher than that of analogs such as nicotinic acid and quinolinic acid. Feeding an ordinary diet containing 0.05% and 0.1% PiA did not elicit decreased intake of food or loss in body weight. PiA did not affect the in vitro liver activities of quinolinic acid phosphoribosyltransferase or α-amino-β-carboxymuconate-ε-semialdehyde decarboxylase (ACMSDase, a Zn-dependent enzyme). Concentrations of NAD and NADP in the liver and blood were not affected by PiA. PiA administration did not affect tryptophan metabolites such as anthranilic acid, kynurenic acid, and xanthurenic acid. However, quinolinic acid and subsequent metabolites such as nicotinamide and its catabolites were increased by administration of a diet containing 0.05% PiA but not by a 0.1% PiA diet. These results suggest that the in vivo activity of ACMSDase is controlled by the Zn level. Therefore, a small amount of PiA has a beneficial effect for conversion of tryptophan to nicotinamide, but an excessive amount of PiA can be very toxic.

Objective: Chromium treatment has been shown to improve mood, appetite, and glucose regulation in various psychiatric and medical patient populations. The authors propose that chromium may be useful in the treatment of binge eating disorder (BED)., Method: Twenty-four overweight adults with BED were enrolled in a 6-month double-blind placebo-controlled trial and randomly assigned to receive either 1000mcg chromium/day ("high dose"; n=8) or 600mcg chromium/day ("moderate dose"; n=9) as chromium picolinate or placebo (n=7). Mixed linear regression models were used to estimate mean change in binge frequency and related psychopathology, weight, symptoms of depression, and fasting glucose., Results: Fasting glucose was significantly reduced in both chromium groups compared to the placebo group; similarly, numerically, but not significantly, greater reductions in binge frequency, weight, and symptoms of depression were observed in those treated with chromium versus placebo, although statistical power was limited in this pilot trial. For fasting glucose, the findings suggest a dose response with larger effects in the high dose compared to moderate dose group., Conclusion: These initial findings support further larger trials to determine chromium's efficacy in maintaining normal glucose regulation, reducing binge eating and related psychopathology, promoting modest weight loss, and reducing symptoms of depression in individuals with BED. Studies designed to link the clinical effects of chromium with changes in underlying insulin, serotonin, and dopamine pathways may be especially informative. If efficacious, chromium supplementation may provide a useful, low-cost alternative to or augmentation strategy for selective serotonin reuptake inhibitors, which have partial efficacy in BED. The relationships between chromium and metabolism are sophisticated. Organic nucleic acids and serum biochemistry parameters are affected by dietary chromium levels. The objective of this work was to study the effect of chromium picolinate (CrPic) supplementation on total DNA and RNA contents, the ratio of RNA/DNA in muscle and in pancreatic tissue, the level of insulin receptor (IR) mRNA and some serum biochemistry parameters in a porcine model. Young animals (48) were assigned randomly into three groups of 16 piglets, fed with three different dietary levels of Cr (common basal feedstuff alone or supplemented with CrPic at a dose of 1.61 μg/g or 3.22 μg/g, which corresponds to 0.2 μg/g and 0.4 μg/g Cr). After 80 days, the animals were sacrificed and skeletal muscle and pancreatic tissues were analyzed to detect differences caused by different levels of dietary Cr. The total content of RNA in muscle was increased significantly (P<0.05) in the CrPic supplemented groups. There was no significant difference between groups in the concentrations of total RNA in the pancreas or DNA in the muscle and pancreatic tissues. The RNA/DNA ratio in pancreas showed no significant change but the ratio was increased significantly (P<0.05) in muscle. There was a slight increase of the mRNA level of IR but there was no significant difference between groups. The content of serum cholesterol and insulin were reduced significantly (P<0.05) in the CrPic-supplemented groups and the content of high-density lipoprotein cholesterol (HDLC) was increased significantly (P<0.05) as the CrPic dose increased. There was a slight (non-significant) reduction of the concentrations of serum triglyceride and low-density lipoprotein cholesterol (LDLC) in the CrPic supplementation groups. Supplementary CrPic caused no significant change of muscular mRNA level of IR in healthy animals. An increased content of RNA in muscle, improved cholesterol metabolism and improved insulin sensitivity were found in these CrPic-treated groups in the porcine model., (Copyright © 2011 Elsevier GmbH. Aim: Multidrug regimens in HIV disease are associated with an increased incidence of insulin resistance, by as much as 50%. Not only does insulin resistance predisposes subjects to diabetes but also it is associated with the metabolic syndrome and increased risk of cardiovascular disease. Previous studies suggest that chromium picolinate can improve insulin resistance in patients with type 2 diabetes. The objective was to study the efficacy and safety of chromium picolinate as a treatment of insulin resistance in subjects infected with HIV., Methods: The ability of chromium picolinate (1000 mug/day) to improve insulin sensitivity, determined with a hyperinsulinaemic-euglycaemic insulin clamp, was determined in eight HIV-positive subjects on highly active antiretroviral therapy., Results: The mean rate of glucose disposal during the clamp was 4.41 mg glucose/kg lean body mass (LBM)/min (range 2.67-5.50), which increased to 6.51 mg/kg LBM/min (range 3.19-12.78, p = .03), an increase of 25% after 8 weeks of treatment with chromium picolinate. There were no significant changes in blood parameters, HIV viral burden or CD4+ lymphocytes with chromium picolinate treatment. Two subjects experienced abnormalities of liver function during the study. Another subject experienced an elevation in blood urea nitrogen., Conclusions: The study shows that chromium picolinate therapy improves insulin resistance in some HIV-positive subjects, but with some concerns about safety in this population.

In women with polycystic ovary syndrome, chromium picolinate (200 microg/d) improves glucose tolerance compared with placebo but does not improve ovulatory frequency or hormonal parameters. This pilot study indicates that future studies in the polycystic ovary syndrome population should examine higher dosages or longer durations of treatment.

This paper addresses the potential economic benefits of chromium picolinate plus biotin (Diachrome) use in people with Type 2 diabetes (T2DM). The economic model was developed to estimate the impact on health care systems' costs by improved HbA1C levels with chromium picolinate plus biotin (Diachrome). Lifetimes cost savings were estimated by adjusting a benchmark from the literature, using a price index to adjust for inflation. The cost of diabetes is highly dependent on the HbA1C level with higher initial levels and higher annual increments increasing the cost. Improvement in glycemic control has proven to be cost-effective in delaying the onset and progression of T2DM, reducing the risk for diabetes-associated complications and lowering utilization and cost of care. Chromium picolinate plus biotin (Diachrome) showed greater improvement of glycemic control in poorly controlled T2DM patients (HbA(1C) > or = 10%) compared to their better controlled counterparts (HbA(1C) < 10%). This improvement was additive to that achieved by oral hypoglycemic medications and correlates to calculated levels of cost savings. Average 3-year cost savings for chromium picolinate plus biotin (Diachrome) use could range from 1,636 dollars for a poorly controlled patient with diabetes without heart diseases or hypertension, to 5,435 dollars for a poorly controlled patient with diabetes, heart disease, and hypertension. Average 3-year cost savings was estimated to be between 3.9 billion dollars and 52.9 billion dollars for the 16.3 million existing patients with diabetes. Chromium picolinate plus biotin (Diachrome) use among the 1.17 million newly diagnosed patients with T2DM each year could deliver lifetime cost savings of 42 billion dollars, or 36,000 dollars per T2DM patient. Affordable, safe, and convenient, chromium picolinate plus biotin (Diachrome) could prove to be a cost-effective complement to existing pharmacological therapies for controlling T2DM.

Chromium picolinate (CrP) supplementation has been studied as a potential therapy of insulin resistance and lipid abnormalities. There have been some reports involving chromium supplementation in patients with diabetes, but the results are varied. The present study was conducted to assess the effects of CrP on insulin sensitivity and body weight in Goto-Kakizaki (GK) diabetic rats. We supplemented normal Sprague-Dawley (SD) rats and GK diabetic rats with supplemental CrP, 100 mg/kg/day once a day for 4 weeks. In the normal SD rats, the mean body weight of the control group increased by 50.5%, whereas that of the CrP-treated group increased by 65.9% (P < 0.05 vs control). Similarly, in the diabetic GK rats, CrP supplementation showed increased weight gain compared to the control group (133.4% vs 119.6% of the baseline weight, P < 0.01). Glucose tolerance tests (GTT) [ip injection of glucose; 2 g/kg] and insulin sensitivity tests [SQ injection of insulin (5 U/kg) plus ip injection of glucose (30 min after insulin injection)] were conducted. During insulin sensitivity tests at the end of treatment, the glucose levels were significantly lower in CrP-treated rats compared with the control rats (AUC0-->120; 113.1 +/- 32.0 vs 170.5 +/- 49.0 mg-min/mL, P < 0.05). During GTTs, the glucose levels and insulin concentrations in the CrP-treated rats were not different from those in the control rats. The results of these studies suggest that CrP supplementation in GK diabetic rats leads to increase of weight gain and improvement of insulin sensitivity. This raises the possibility that CrP supplementation can be considered to improve carbohydrate metabolism in patients with type 2 diabetes mellitus.

Earlier studies from our laboratory have indicated insulin sensitizing action of chromium picolinate as the mechanism of its anti-diabetic activity in experimental models of type I and type II diabetes. In the present investigation, we have evaluated the effects of chronic administration of chromium picolinate on the functional and histological alterations of streptozotocin (STZ)-induced diabetes in rats. Type I diabetes was induced by intravenous injection of STZ (40 mg/kg) in adult rats, whereas, type II diabetes was induced by intraperitoneal injection of STZ (90 mg/kg) in 2-day old rat pups which in adulthood develop abnormalities resembling type II diabetes. Chromium picolinate was administered at 8 microg/ml in drinking water for 6 weeks and was found to improve glucose tolerance and increase insulin sensitivity of STZ-diabetic rats. This treatment decrease elevated serum creatinine and urea levels as well as elevated serum levels of hepatic enzymes of both groups of diabetic rats. Histopathological studies of kidney and liver show decrease in the intensity and incidence of vacuolations, cellular infiltration and hypertrophy of STZ and nSTZ (neonatal STZ) diabetic rats. Chronic treatment with chromium picolinate however, did not alter the normal function or morphology of control rats. Chronic chromium picolinate at the therapeutic doses that improved glucose tolerance, was observed to have no hepatotoxic or nephrotoxic potential. It was rather found to improve renal and hepatic function and to reduce abnormalities associated with STZ-diabetes. Chromium picolinate could play an important role in the long term management of diabetes mellitus.

Human studies suggest that chromium picolinate (CrPic) decreases insulin levels and improves glucose disposal in obese and type 2 diabetic populations. To evaluate whether CrPic may aid in treatment of the insulin resistance syndrome, we assessed its effects in JCR:LA-corpulent rats, a model of this syndrome. Male lean and obese hyperinsulinemic rats were randomly assigned to receive oral CrPic [80 microg/(kg. d); n = 5 or 6, respectively) in water or to control conditions (water, n = 5). After 3 mo, a 120-min intraperitoneal glucose tolerance test (IPGTT) and a 30-min insulin tolerance test were performed. Obese rats administered CrPic had significantly lower fasting insulin levels (1848 +/- 102 vs. 2688 +/- 234 pmol/L; P < 0.001; mean +/- SEM) and significantly improved glucose disappearance (P < 0.001) compared with obese controls. Glucose and insulin areas under the curve for IPGTT were significantly less for obese CrPic-treated rats than in obese controls (P < 0.001). Obese CrPic-treated rats had lower plasma total cholesterol (3.57 +/- 0.28 vs. 4.11 +/- 0.47 mmol/L, P < 0.05) and higher HDL cholesterol levels (1.92 +/- 0.09 vs. 1.37 +/- 0.36 mmol/L, P < 0.01) than obese controls. CrPic did not alter plasma glucose or cholesterol levels in lean rats. Total skeletal muscle glucose transporter (Glut)-4 did not differ among groups; however, CrPic significantly enhanced membrane-associated Glut-4 in obese rats after insulin stimulation. Thus, CrPic supplementation enhances insulin sensitivity and glucose disappearance, and improves lipids in male obese hyperinsulinemic JCR:LA-corpulent rats.

This study assessed the effect of resistive training (RT), with or without highdose chromium picolinate (Cr-pic) supplementation, on body composition and skeletal muscle size of older women. Seventeen sedentary women, age range 54-71 years, BMI 28.8 +/- 2.4 kg/m2, were randomly assigned (double-blind) to groups (Cr-pic, n = 9; Placebo, n = 8) that consumed either 924 micrograms Cr/d as Cr-pic or a low-Cr placebo (< 0.2 microgram Cr/d) during a 12-week RT program (2 day/week, 3 sets.exercise-1.d-1, 80% of 1 repetition maximum). Urinary chromium excretion was 60-fold higher in the Cr-pic group, compared to the Placebo group (p < .001), during the intervention. Resistive training increased maximal strength of the muscle groups trained by 8 to 34% (p < .001), and these responses were not influenced by Cr-pic supplementation. Percent body fat and fat-free mass were unchanged with RT in these weight-stable women, independent of Cr-pic supplementation. Type I and type II muscle fiber areas of the m. vastus lateralis were not changed over time and were not influenced by Cr-pic supplementation. These data demonstrate that high-dose Cr-pic supplementation did not increase maximal strength above that of resistive training alone in older women. Further, these data show that, under these experimental conditions, whole body composition and skeletal muscle size were not significantly changed due to resistive training and were not influenced by supplemental chromium picolinate.

This study was conducted to determine the effects of Cr (chromium picolinate, CrPic) supplementation at various levels (0, 200, 400, 800 or 1200 microg/kg of diet) on egg production, egg quality and serum concentrations of insulin, corticosterone and glucose in laying Japanese quail (Coturnix coturnix japanica) reared under conditions of heat stress (32.5 degrees C). Laying Japanese quail (n = 150; 45 d old) were divided into five groups of 30 birds. The quail were fed either a control diet containing 965 microg Cr/kg diet or the control diet supplemented with 200, 400, 800 or 1200 microg of Cr/kg diet. Increased supplemental chromium increased body weight (P = 0.05, linear), feed intake (P = 0.05, linear), egg production (P = 0.01, linear) and also improved feed efficiency (P = 0.01, linear). Increased supplemental chromium linearly increased egg weight (P = 0.01), eggshell thickness, egg specific gravity (P = 0.05) and Haugh unit (P = 0.01). Serum insulin concentration increased linearly (P = 0.01), whereas corticosterone and glucose concentration decreased linearly (P = 0.05) as dietary chromium increased. The best results were obtained with 1200 microg Cr/kg diet, and chromium supplementation at such a level can be considered to be protective management practice in a quail diet, reducing the negative effects of heat stress.

Chromium picolinate, [Cr(pic)(3)], is the second most popular nutritional supplement after calcium supplements. However, the supplement, unlike simple inorganic Cr(III) salts, has been shown in the presence of biological reducing agents in vitro to catalytically generate appreciable quantities of hydroxyl radicals, resulting in DNA damage. The complex has also been shown to be remarkably stable in vitro at neutral, basic, or weakly acidic pHs. Thus, the significance of this ability to generate hydroxyl radicals depends on whether the complex is absorbed by cells intact along with the stability and concentration of the complex in cells. Consequently, male Sprague Dawley rats have been injected with (51)Cr- and (3)H-labeled [Cr(pic)(3)]. The tissue distribution, urinary and fecal loss, and subcellular hepatocyte distribution and concentration of the labels suggest that [Cr(pic)(3)] has a lifetime of less than 1 day in vivo, minimizing the potential threat from the supplement itself.

Chromium is an essential dietary trace mineral involved in carbohydrate and lipid metabolism. Chromium is required for cellular uptake of glucose, and chromium deficiency causes insulin resistance. Chromium supplementation may improve insulin sensitivity and has been used as adjunct treatment of diabetes mellitus in humans. In this study, 13 dogs with naturally acquired diabetes mellitus were treated with insulin for 3 months, then with insulin and chromium picolinate for 3 months. Dogs weighing <15 kg (33 lb: n = 9) were administered 200 microg of chromium picolinate PO once daily for I month, then 200 microg of chromium picolinate twice daily for 2 months. Dogs weighing >15 kg (n = 4) received 200 microg of chromium picolinate once daily for 2 weeks, then 200 microg twice daily for 2 weeks, then 400 microg twice daily for 2 months. Type of insulin, frequency of insulin administration, and diet were kept constant, and insulin dosage was adjusted, as needed, to maintain optimal control of glycemia. Mean body weight, daily insulin dosage, daily caloric intake, 10-hour mean blood glucose concentration, blood glycated hemoglobin concentration, and serum fructosamine concentration were not markedly different when dogs were treated with insulin and chromium picolinate, compared with insulin alone. Adverse effects were not identified with chromium picolinate administration. Results of this study suggest that, at a dosage range of 20-60 microg/kg/d, chromium picolinate caused no beneficial or harmful effects in insulin-treated diabetic dogs.

This experiment was conducted to evaluate the effects of chromium (chromium picolinate) on performance and plasma concentrations of insulin and corticosterone of laying hens (Ross Brown) under a low ambient temperature (6.9 degrees C). One hundred and twenty laying hens (46 weeks old) were divided into four groups, 30 hens per group. The laying hens were fed either a control diet containing 710.3 p.p.b. chromium or the control diet supplemented with 100, 200 or 400 microg chromium/kg diet. Increasing supplemental chromium increased live weight change (p < or = 0.001, linear) and egg production (p < or = 0.001, linear) and also improved feed efficiency linearly (p < or = 0.001). Live weight change and egg production also had quadratic responses (p < or = 0.001) to increasing chromium supplementation. Plasma insulin concentration increased linearly (p < or = 0.001), whereas corticosterone concentration decreased linearly (p < or = 0.001) as dietary chromium supplementation increased. The results of this study indicate that supplemental dietary chromium, 200 p.p.b. in particular, had a positive effect on performance and increased the plasma insulin concentration of laying hens under cold stress.

Background: Chromium is an essential nutrient required for carbohydrate and lipid metabolism. Chromium supplementation in humans has been reported to improve glucose metabolism and improve serum lipid parameters and to reduce body fat; parameters that worsen with aging. As a result, chromium picolinate has been widely promoted as a health aid for the general population. The purpose of the study was to examine the effects of chromium supplementation on insulin sensitivity, serum lipids, and body composition in nonobese, healthy men and women of advanced age., Methods: A randomized, double-blind, placebo-controlled study with 19 subjects (9 men and 10 women), aged 63-77, were given either chromium picolinate, 1,000 microg/d, or a placebo for 8 weeks. Serum lipids were measured at baseline and 8 weeks. Insulin sensitivity and body composition were measured with the minimal-model intravenous glucose tolerance test and dual-energy x-ray absorptiometry scan, respectively, at baseline and after 8 weeks of chromium or placebo supplementation., Results: No significant change in serum lipids, insulin sensitivity, or body composition was observed in the chromium group compared with the placebo group., Conclusions: Chromium picolinate supplementation alone does not appear to improve insulin sensitivity, serum lipids, or change body composition in nonobese, healthy men and women of advanced age.

Recent in vitro studies have shown that chromium (III) compounds such as chromium picolinate, a popular dietary supplement among people trying to lose weight, produce chromosome damage. We monitored levels of DNA damage in a chromium picolinate supplement trial by measuring antibodies titers to an oxidized DNA base, 5-hydroxymethyl-2'-deoxyuridine (HMdU), by enzyme-linked immunosorbent assays. Ten obese volunteer women completed a 8-week course of 400 micrograms chromium picolinate per day. In either absolute titers or percent of the baseline value, there were no changes in antibody titers at 4 or 8 weeks. The titers were very stable within individuals and those of one individual rarely crossed over others, which was reflected in an intraclass correlation coefficient of 0.99 (95% confidence interval: 0.96-1.00). There were no effects on glucose and lipid metabolism in this period. The results of this trial suggest that chromium (III) picolinate in a dose typically used for nutrient supplementation dose not increase oxidative DNA damage, as measured by anti-HMdU antibody levels.

Rhabdomyolysis results from skeletal muscle injury and release of muscle cell contents into plasma. A number of etiologic factors have been reported for the condition, including strenuous exercise, weight lifting, trauma, seizure, sepsis, and alcohol and drug abuse. Hundreds of drugs also reportedly cause rhabdomyolysis. A 24-year-old body builder developed the disease after ingesting 1200 microg of chromium picolinate (6-24 times the daily recommended allowance of 50-200 microg) over 48 hours. We believe this to be the first reported case of chromium-induced rhabdomyolysis.

Chromium competes with iron for binding to transferrin, and high-dose chromium supplementation has been hypothesized to adversely affect iron status. This study examined the effects of chromium picolinate supplementation on hematologic indexes and selected indexes of iron status in 18 men aged 56-69 y who participated in an introductory resistive training program. The men were randomly assigned (double-blind design) to groups (n = 9) that consumed either 17.8 mumol Cr/d (924 micrograms Cr/d) as chromium picolinate or a low-chromium placebo for 12 wk while engaging in resistive training twice weekly (3 sets of 8-12 repetitions at 80% of one repetition maximum for 5 exercises). Hematocrit, hemoglobin, red blood cell (erythrocyte) count, mean corpuscular volume, mean corpuscular hemoglobin, mean corpuscular hemoglobin concentration, red blood cell distribution width, platelet count, and mean platelet volume were within normal clinical ranges and were unchanged by either chromium picolinate supplementation or resistive training. Resistive training decreased total-iron-binding capacity from 38.4 +/- 9.3 to 27.3 +/- 5.6 mumol/L (P < 0.0001) and increased transferrin saturation from 35.7 +/- 16.3% to 45.4 +/- 16.9% (P = 0.050). Chromium picolinate supplementation did not influence these responses. Serum iron concentrations and serum ferritin concentrations were unchanged by either resistive training or chromium picolinate supplementation. These data suggest that high-dose chromium picolinate supplementation for 12 wk did not influence hematologic indexes or indexes of iron metabolism or status in older men. The decrease in total-iron-binding capacity and increase in transferrin saturation (%) with resistive training are largely opposite to changes associated with iron depletion and suggest a novel effect of resistive training on iron transport.
