To advance laccase production from basidiomycetes, a newly designed 14-L low-shear aerated and agitated bioreactor provided enzyme titers up to 23.5 IU/mL from Trametes versicolor cultures. Produced enzymes underwent ultrafiltration and LC/MS-MS characterization, revealing the predominant production of only two out of the ten laccases predicted in the T. versicolor genome. Process simulation and economic analysis using SuperPro designer® suggested that T. versicolor laccase could be produced at US$ 3.60/kIU in a 200-L/batch enterprise with attractive economic parameters and a payback period of 1.7 years. The present study aimed to identify a pair of fungal strains that promote laccase production in the co-cultivation of white-rot basidiomycetes and to determine the optimum conditions to enhance enzyme synthesis under co-fermentation of mandarin peels. Co-cultivation of Cerrena unicolor with Trametes versicolor, Lenzites betulina, and Panus lecomtei led to up-regulation of laccase activity. Moreover, interspecific interaction of Cerrena unicolor and Trametes versicolor induced the production of two new laccase isoenzymes. By contrast, interactions of Cerrena unicolor with Trametes coccineus and Trametes hirsuta resulted in a multiple decreased ability of Cerrena unicolor to produce laccase. Co-cultivation of Cerrena unicolor with other fungi 3- to 12-fold down-regulated manganese peroxidase (MnP) activity. The outcomes of these fungal interactions are closely related to the initial concentration and availability of the nutrients, the partners' inoculum ratio, time, and sequence of their inoculation. Although, currently, more than 100 laccases have been purified from basidiomycete fungi, the majority of these laccases were obtained from fungi of the Polyporales order, and only scarce data are available about the laccases from other fungi. In this article, laccase production by the white-rot basidiomycete fungus Peniophora lycii, belonging to the Russulales order, was investigated. It was shown that, under copper induction, this fungus secreted three different laccase isozymes. Two laccase isozymes--Lac5 and LacA--were purified and their corresponding nucleotide sequences were determined. Both purified laccases were relatively thermostable with periods of half-life at 70 °C of 10 and 8 min for Lac5 and LacA, respectively. The laccases demonstrated the highest activity toward ABTS (97 U·mg-1 for Lac5 and 121 U·mg-1 for LacA at pH 4.5); Lac5 demonstrated the lowest activity toward 2,6-DMP (2.5 U·mg-1 at pH 4.5), while LacA demonstrated this towards gallic acid (1.4 U·mg-1 at pH 4.5). Both Lac5 and LacA were able to efficiently decolorize such dyes as RBBR and Bromcresol Green. Additionally, phylogenetic relationships among laccases of Peniophora spp. were reconstructed, and groups of orthologous genes were determined. An extensive screening of saprotrophic Basidiomycetes causing white rot (WR), brown rot (BR), or litter decomposition (LD) for the production of laccase and Mn-peroxidase (MnP) and decolorization of the synthetic dyes Orange G and Remazol Brilliant Blue R (RBBR) was performed. The study considered in total 150 strains belonging to 77 species. The aim of this work was to compare the decolorization and ligninolytic capacity among different ecophysiological and taxonomic groups of Basidiomycetes. WR strains decolorized both dyes most efficiently; high decolorization capacity was also found in some LD fungi. The enzyme production was recorded in all three ecophysiology groups, but to a different extent. All WR and LD fungi produced laccase, and the majority of them also produced MnP. The strains belonging to BR lacked decolorization capabilities. None of them produced MnP and the production of laccase was either very low or absent. The most efficient decolorization of both dyes and the highest laccase production was found among the members of the orders Polyporales and Agaricales. The strains with high MnP activity occurred across almost all fungal orders (Polyporales, Agaricales, Hymenochaetales, and Russulales). Synthetic dye decolorization by fungal strains was clearly related to their production of ligninolytic enzymes and both properties were determined by the interaction of their ecophysiology and taxonomy, with a more relevant role of ecophysiology. Recycling paper is worthy alternative to cut down the dependence on trees thereby preventing the hazards like deforestation. Deinking is an important step in the recycling process which involves the removal of ink particles from fiber surface and decolorization of the dislodged ink. Enzymes such as laccases may prove eco-friendly cost effective alternatives to the conventional chemicals methods used for the recycling process. However, the commercial application of this enzyme is limited by insufficient production and less stability. Hence, in this study, the laccase produced by basidiomycetes species, P. ostreatus was evaluated for its potential as a deinking agent. P. ostreatus was able to grow on many cheap substrates and when grown in the medium containing sawdust showed significant laccase production. Addition of 5g/L of Al2O3 microparticles (microparticle enhanced cultivation) improved the production of laccase. The maximum activity of P. ostreatus laccase was observed at pH 3 and at 28°C. Activity was further stabilized by incorporation of Copper ions and immobilization in Ca-alginate beads. The enzyme was capable of decolorizing ink from color printer cartridge and from low quality coloured papers such as bus tickets. In this study, five fungal strains (Aspergillus niger SCBM1 – Ni, Aspergillus fumigatus SCBM6 – Fu, Trametes versicolor 561 – Tr, Ganoderma lucidum 601 – Ga and Pleurotus ostreatus PL06 – Pl) were cultivated individually and in consortium for biosynthesis of lignocellulose-degrading enzymes by solid-state fermentation (SSF). The enzyme production was investigated using a 25−1 fractional factorial design, with a total of 16 experiments (F1–F16) using raw sugarcane bagasse and raw wheat bran as substrates. Among the enzymatic extracts produced, Ni (F1) exhibited the highest production of endoglucanase (82.70 U/gds) (units per gram of dry substrate), exoglucanase (80.48 U/gds), β-xylosidase (145.01 U/gds) and manganese peroxidase (3.38 U/gds). For filter paper cellulase, Tr cocktail (F5) was the one that stood out (9.45 U/gds). Among the extracts produced in consortium, Ni + Tr + Pl (F6) presented the highest production of β-glucosidase (171.09 U/gds), β-xylosidase (139.99 U/gds) and manganese peroxidase (3.29 U/gds). For FPase, Ni + Fu + Ga (F12) exhibited the best production (10.46 U/gds). The highest xylanase biosynthesis (2582.38 U/gds) was obtained in Ni + Fu + Pl extract (F4). For laccase, the maximum biosynthesis (25.27 U/gds) was obtained in Tr + Ga + Pl (F13). Laccases are multicopper oxidases that catalyze oxidation of a wide range of organic and inorganic substrates accompanied by the reduction of dioxygen to water. The physicochemical and catalytic properties of two new fungal laccases from basidiomycetes Antrodiella faginea (AfL) and Steccherinum murashkinskyi (SmL) with middle redox potential of the T1 copper site were studied. The X-ray structures of AfL and SmL were solved at 1.75 Å and 0.95 Å, respectively. The oxidized state of copper ions in the active site was observed in AfL structure, while the mixture of oxidized and reduced states was observed in SmL structure. These oxidized and reduced states relate to the position of copper ions, their coordination, and nature and position of oxygen ligands. Comparative analysis of the T1 site environment of laccases with known structure allowed us to highlight the six types of the secondary coordination sphere of the T1 copper. The solvent accessible surface area of the conservative region of the secondary coordination sphere of the T1 copper correlates with its the redox potential. It was shown that the laccase classification by the structure of the T1 copper secondary coordination sphere is in agreement to ecophysiological behavior of laccase producing fungi. Abstract The ability of seven distinct species of white-rot basidiomycetes (WRB) to produce lignin-modifying enzymes (LME) and reduce high phenolic content was assessed during submerged fermentation of oil mill by-products. Cerrena unicolor strains appeared to be the outstanding producers of laccase (56.3–78.5 U mL−1) and MnP (1.03–2.76 U mL−1) in the fermentation of olive pomace (OP), and laccase (26.9–93.8 U mL) in the fermentation of olive tree sawdust. Diluted 50% olive mill wastewater (OMW) can be used as a nutrient medium for laccase and MnP production but its supplementation with additional carbon and nitrogen sources and Cu2+ and Mn2= promoted the 3-, 28- and 18-fold increase of C. unicolor biomass and volumetric laccase and MnP activities, respectively, as well as 3- and 17-fold increase of Pleurotus ostreatus biomass and laccase activity, respectively. The tested fungi successfully reduced the phenolics content in the submerged fermentation of OP (72.0–81.3%) and OMW (62.4–83.5%). The laccase role in the OP phenolics reduction was proved through the treatment of 10% (wet weight) OP with commercial and lab-generated laccases preparations and reduction of total phenols by 66–70% during 24 h incubation. This work indicated that the olive mill by-products fermentation by individual substrate-specific WRB is an economically feasible and promising alternative for the simultaneous production of LME, cellulase, xylanase and detoxified materials suitable for further exploitation in biotechnological and agronomic applications. Wood-decaying fungi tend to have characteristic substrate ranges that partly define their ecological niche. Fomitopsis pinicola is a brown rot species of Polyporales that is reported on 82 species of softwoods and 42 species of hardwoods. We analyzed the gene expression levels and RNA editing profiles of F. pinicola from submerged cultures with ground wood powder (sampled at 5 days) or solid wood wafers (sampled at 10 and 30 days), using aspen, pine, and spruce substrates (aspen was used only in submerged cultures). Fomitopsis pinicola expressed similar sets of wood-degrading enzymes typical of brown rot fungi across all culture conditions and time points. Nevertheless, differential gene expression and RNA editing were observed across all pairwise comparisons of substrates and time points. Genes exhibiting differential expression and RNA editing encode diverse enzymes with known or potential function in brown rot decay, including laccase, benzoquinone reductase, aryl alcohol oxidase, cytochrome P450s, and various glycoside hydrolases. There was no overlap between differentially expressed and differentially edited genes, suggesting that these may provide F. pinicola with independent mechanisms for responding to different conditions. Comparing transcriptomes from submerged cultures and wood wafers, we found that culture conditions had a greater impact on global expression profiles than substrate wood species. In contrast, the suites of genes subject to RNA editing were much less affected by culture conditions. The laccase production by mycelial antagonistic interaction among white-rot fungi is a very important pathway for lignin degradation research. To gain a better understanding of competitive mechanisms under mycelial antagonistic interaction among three lignin-degrading white-rot basidiomycetes of Trametesversicolor (Tv), Pleurotusostreatus (Po) and Dichomitussqualens (Ds), mycelial morphology and proteins in three co-culture combinations TvPo (Tv cocultivated with Po), PoDs (Po cocultivated with Ds), TvDs (Tv cocultivated with Ds) were compared with corresponding each two mono-cultures. In this study, scanning electron microscopy detection of co-cultures indicated a highly close attachment of fungal hyphae with each other and conidiation could be inhibited under fungal interaction. In addition, a label-free proteomic analysis revealed changes on fungal proteomes existed in their counterpart competitors of co-culture. The maximum number of 1020 differentially expressed proteins (DEPs) were identified in PoDs relative to Po while the minimum number of 367 DEPs were identified in PoDs relative to Ds. Notably, we also found a large number of overexpressed proteins were oxidative stress-related proteins, followed by carbohydrate metabolism-related proteins and energy production-related proteins in all three co-culture combinations compared with control. Laccase production in saline conditions is still poorly studied. The aim of the present study was to investigate the production of laccase in two different types of bioreactors by the marine-derived basidiomycete Peniophora sp. CBMAI 1063. The highest laccase activity and productivity were obtained in the Stirred Tank (ST) bioreactor, while the highest biomass concentration in Air-lift (AL) bioreactor. The main laccase produced was purified by ion exchange and size exclusion chromatography and appeared to be monomeric with molecular weight of approximately 55 kDa. The optimum oxidation activity was obtained at pH 5.0. The thermal stability of the enzyme ranged from 30 to 50 °C (120 min). The Far-UV Circular Dichroism revealed the presence of high β-sheet and low α-helical conformation in the protein structure. Additional experiments carried out in flask scale showed that the marine-derived fungus was able to produce laccase only in the presence of artificial seawater and copper sulfate. Laccase (benzenediol:oxygen oxidoreductase, EC 1.10.3.2) is an enzyme that has been studied for over 100 y and is present in virtually all fungi. As increasing numbers of fungal genomes have been sequenced, it has become apparent that the laccase genes in white-rot fungi commonly form multigene families consisting of many nonallelic genes. Although a number of reports focussing on laccase gene expression in different fungal species were published over the decades, the fundamental questions of why fungi need such a redundant array of genes and how they manage this array to perform biological function(s) remain far from answered. In this article, we present a comprehensive study of the transcription of the whole Trametes hirsuta laccase multigene family under different conditions, including exposure to different nutritional factors such as nitrogen sources (organic and inorganic) and concentrations of nitrogen and carbon in the culture medium; in different growth phases (lag phase and stationary phase); and in the presence of different inducer agents (water-soluble lignin, bromocresol green dye, p -coumaric acid, ferulic acid, guaiacol, vanillin, veratryl alcohol, vanillic acid and syringic acid). A combined system of a unique dye-decolorizing peroxidase (Ftr-DyP) and a laccase obtained from the basidiomycete Funalia trogii converted the precursor (+)-valencene completely to the high-value grapefruit flavour constituent (+)-nootkatone, reaching a concentration maximum of 1100 mg/L. In the presence of 1 mM Mn2+ and 2.5 mM p-coumaric acid, (+)-nootkatone was the predominating volatile product, and only traces of substrate and the nootkatols were detectable after 24 h. Hence, the two-enzyme-system reproduced the oxidizing activity observed before for the crude culture supernatant. The newly discovered Ftr-DyP was purified, sequenced and further characterized as a thermostable, non-glycosylated protein with a pH-optimum in the acidic range and a calculated mass of 52.3 kDa. Besides the typical activity of DyPs towards anthraquinone dyes, Ftr-DyP also oxidized Mn2+ and showed activity in the absence of hydrogen peroxide. Neither the DyP from Mycetinis scorodonius nor the manganese peroxidase from Nematoloma frowardii were able to replace Ftr-DyP in this reaction. A hypothetical reaction mechanism is presented. Laccases are multicopper oxidases that are able to catalyze reactions involving a range of substrates, including phenols and amines, and this ability is related to the existence of different laccases. Basidiomycetes usually have more than one gene for laccase, but until now, this feature has not been demonstrated in a marine-derived fungus. Peniophora sp. CBMAI 1063 is a basidiomycete fungus isolated from a marine sponge that exhibits the ability to secrete significant amounts of laccase in saline conditions. In the present study, we identified laccase sequences from the transcriptome of Peniophora sp. CBMAI 1063 and used them to perform different molecular in silico analyses. The results revealed the presence of at least eight putative genes, which may encode ten different laccases with peptide lengths ranging from 482 to 588 aa and molecular weights ranging from 53.5 to 64.4 kDa. These laccases seem to perform extracellular activities, with the exception of one that may represent an intracellular laccase. The 10 predicted laccases expressed by Peniophora sp. CBMAI 1063 in laccase-induced media showed different patterns of N-glycosylation and isoelectric points and are divided into two classes based on the residue associated with the regulation of the redox potential of the enzyme. None of the predicted laccases showed more than 61% similarity to other fungal laccases. Based on the differences among the laccases expressed by Peniophora sp. A laccase of the basidiomycete Pleurotus pulmonarius (PpuLcc) possessed strong decolorizing abilities towards artificial and natural dyes. The PpuLcc was purified from the culture supernatant via FPLC, and the corresponding gene cloned and expressed in Pichia pastoris GS115. To examine the impact of the C-terminal tail region and the signal peptide on the recombinant expression of PpuLcc, a non-modified version or different truncations (−2, −5, −13 AA) of the target protein were combined with different secretion signals. Heterologous expression of codon optimized constructs resulted in extracellular activities of the PpuLcc variants of up to 7000 U L −1 (substrate ABTS) which was six times higher than non-codon optimized constructs. In contrast to previous works, altering the C-terminal end of the protein did not influence kinetic parameters or the rate of expression. The His -Tag purified enzymes showed high temperature optima (50–70 °C) and thermo stability. All of the recombinant variants degraded triarylmethane and azo dyes. Rapid bleaching of β -carotene (E 160a) and the polyene acid norbixin (E 160b) using a laccase was found for the first time. Thus, the enzyme may be useful in decolorizing unwanted polyene pigments, for example from the processing of cheese, bakery, desserts, ice cream or coloured casings. [ABSTRACT FROM AUTHOR]

Enzymatic grafting of kraft lignin (KL) on wood surfaces is presented as a non-leachable wood protection treatment. Scots pine and European beech mini-blocks were treated with KL solution in the presence of laccase isolated from Myceliophthora thermophila and the formation of a stable interaction between wood and lignin was observed. Furthermore, the same strategy was employed to graft KL with the simultaneous entrapment of copper in the polymeric net formed. Enzymatic treatment diminished the leachability of the compounds in accordance with the EN 84 standard. The durability of the leached wood blocks was evaluated by accelerated decay tests. Samples with grafted lignin on the surface lost their antifungal activity in long-term exposure at the concentration tested. This observation is in contradiction to the preliminary tests, where KL seemed to have some biocidal activity. Dye wastewater management has been a challenging task for the textile industry. In this study, basidiomycetes fungi was explored for its ability to degrade dye. Isolate EL2, collected from the decaying woods around Kampar, Perak, Malaysia was selected for this purpose. The collected isolate was identified asTrametes ljubarskyiby using a molecular characterization method and morphological observation. Then, it was screened with solid agar supplemented with 2,2-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid, guaiacol and Remazol Brillant Blue R dye for laccase production. Four different nitrogen sources were involved in further liquid screening for optimization of laccase production in EL2. Based on the results obtained, urea is the best candidate, as it was able to produce maximal laccase activity of 50.82 U/(L·d), as compared to three other nitrogen sources (1.11–5.08 U/(L·d)). Previously, we demonstrated that when Ustilago maydis (DC) Cda., a phytopathogenic basidiomycete and the causal agent of corn smut, is grown in the vicinity of maize embryogenic calli in a medium supplemented with the herbicide Dicamba, it developed gastroid-like basidiocarps. To elucidate the molecular mechanisms involved in the basidiocarp development by the fungus, we proceeded to analyze the transcriptome of the process, identifying a total of 2002 and 1064 differentially expressed genes at two developmental stages, young and mature basidiocarps, respectively. Function of these genes was analyzed with the use of different databases. MIPS analysis revealed that in the stage of young basidiocarp, among the ca. two thousand differentially expressed genes, there were some previously described for basidiocarp development in other fungal species. Additional elements that operated at this stage included, among others, genes encoding the transcription factors FOXO3, MIG3, PRO1, TEC1, copper and MFS transporters, and cytochromes P450. During mature basidiocarp development, important up-regulated genes included those encoding hydrophobins, laccases, and ferric reductase (FRE/NOX). Metabolite formation from radical-based oxidation of the environmental pollutant triclosan (TCS) was compared using an ascomycete ( Phoma sp. UHH 5-1-03) and a basidiomycete ( Trametes versicolor ) laccase, laccase-redox mediator systems, and electrochemical oxidation (EC). Laccase oxidation predominantly yielded TCS di- and trimers, but notably also caused TCS ether bond cleavage. The latter was more prominent during EC-catalysed TCS oxidation, which generally resulted in a broader and more divergent product spectrum. By contrast, only quantitative but not qualitative differences in TCS metabolite formation were observed for the two laccases. Application of the presumable natural laccase redox mediator syringaldehyde (SYD) shifted the TCS-transforming reactions of laccase systems from oligomerization more towards ether bond cleavage. However, the observed rapid removal of SYD from reaction systems caused by predominant adduct formation from SYD and TCS, and concomitant conversion of SYD into 2,6-dimethoxy-1,4-benzoquinone (DMBQ) clearly demonstrates that SYD does not function as a “true” laccase redox mediator in the sense of being recycled during TCS oxidation. Laccase treatment of TCS without SYD decreased the anti-bacterial TCS activity more than treatment employing SYD in addition, indicating that SYD and/or its transformation products contribute to bacterial toxicity. DMBQ was found to be about 80% more active in a bacterial growth inhibition test than its parent compound SYD in terms of IC 20 values. These observations establish DMBQ as a potential cause of toxicity effects of SYD-laccase systems. The dependence of the degree of fluorene and fluoranthene degradation by the fungus Pleurotus ostreatus D1 on the culture medium composition has been studied. Polycyclic aromatic hydrocarbons ( PAHs) have been transformed in Kirk's medium (under conditions of laccase production) with the formation of a quinone metabolite and 9-fluorenone upon the use of fluoranthene and fluorene as substrates, respectively. More complete degradation with the formation of an intermediate metabolite, phthalic acid that has undergone subsequent utilization, has occurred in basidiomycete-rich medium (under the production of both laccase and versatile peroxidase). The formation of phthalic acid as a metabolite of fluoranthene degradation by lignolytic fungi has been revealed for the first time. The data allow the supposition that both extracellular laccase and laccase on the mycelium surface can participate in the initial stages of PAH metabolism, while versatile peroxidase is necessary for the oxidation of the formed metabolites. A laccase from the basidiomycete Pycnoporus sanguineus strain RVAN5 was evaluated for its ability to decolorize synthetic dyes and denim bleaching. Dye color reduction and denim bleaching were monitored at different dye concentrations and incubation times. Dye decolorization by Pycnoporus sanguineus fungal crude extract (FCE) ranged from 80 to 96% within 2–4 h at 25–65 °C. Comparable results were obtained when violuric acid (VA) was added as mediator to the FCE, however, the number of decolorized dyes increased significantly. Dye decolorization rates with VA varied of initial and final optical density (595 nm) values of 2.5–3.0 and 0.2–0.02, respectively. P. sanguineus FCE had no substantial effect on denim bleaching when used alone, notwithstanding, the mixture of FCE with VA (10 mM) showed significant denim color reduction values and considerably higher than those obtained with a bleaching enzyme from a commercial formulation; CIElab values obtained with FCE/VA mixture were of ΔL = 6.4, versus a ΔL 1.4 value obtained with an enzyme from commercial formulation. A strain LN07 with high laccase yield was identified as basidiomycete fungus Lepista nuda from which a white laccase without type I copper was purified and characterized. The laccase was a monomeric protein with a molecular mass of 56 kDa. Its N-terminal amino acid sequence was AIGPAADLHIVNKDISPDGF. Besides, eight inner peptide sequences were determined and lac4, lac5 and lac6 sequences were in the Cu2+ combination and conservation zones of laccases. HIV-1 reverse transcriptase was inhibited by the laccase with a half-inhibitory concentration of 0.65 μM. Cu2+ ions (1.5 mM) enhanced the laccase production and the optimal pH and temperature of the laccase were pH 3.0 and 50 °C, respectively. The Km and Vmax of the laccase using ABTS as substrate were respectively 0.19 mM and 195 μM. Several dyes including laboratory dyes and textile dyes used in this study, such as Methyl red, Coomassie brilliant blue, Reactive brilliant blue and so on, were decolorized in different degrees by the purified laccase. By LC-MS analysis, Methyl red was structurally degraded by the laccase. Moreover, the laccase affected the absorbance at the maximum wavelength of many pesticides. The aim of this review is to determine the trends of state-of-art of laccase sources, properties, structure and recent application of fungal laccase in various fields. Laccases are biotechnologically important multi copper proteins that have broad substrate specificity towards aromatic and non-aromatic compounds. Fungi are the major laccase producers especially ascomycetes, deuteromycetes and basidiomycetes, and laccases have an average molecular weight between 50 and 130 kDa. Fungal laccases are used in biotechnological applications for preparation of anticancerous and anti-oxidant hormonal drugs, stabilization of food products, and laccase application is also extended to preparation of biosensors, DNA labeling, immunochemical assay, bioorganic compound synthesis etc. The environmental application of laccase is for biodegradation of dyes, phenols and pesticides, and the mechanism of degradation has been briefly explained. Analysis of the biodegraded dye sample by FT-IR and Mass (ESI)-spectrum has been discussed in a detailed manner. Epitheaflagallin 3- O -gallate (ETFGg) is a minor polyphenol found in black tea extract, which has good physiological functions. It is synthesized from epigallocatechin gallate (EGCg) with gallic acid via laccase oxidation. Various basidiomycetes and fungi were screened to find a suitable laccase for the production of ETFGg. A basidiomycete, Hericium coralloides NBRC 7716, produced an appropriate extracellular laccase. The purified laccase produced twice the level of ETFGg compared with commercially available laccase from Trametes sp. The enzyme, termed Lcc2, is a monomeric protein with an apparent molecular mass of 67.2 kDa. The N-terminal amino acid sequence of Lcc2 is quite different from laccase isolated from the fruiting bodies of Hericium . Lcc2 showed similar substrate specificity to known laccases and could oxidize various phenolic substrates, including pyrogallol, gallic acid, and 2,6-dimethoxyphenol. The full-length lcc2 gene was obtained by PCR using degenerate primers, which were designed based on the N-terminal amino acid sequence of Lcc2 and conserved copper-binding sites of laccases, and 5'-, and 3'-RACE PCR with mRNA. The Lcc2 gene showed homology with Lentinula edodes laccase (sharing 77% amino acid identity with Lcc6). We successfully produced extracellular Lcc2 using a heterologous expression system with Saccharomyces cerevisiae . Laccase is a key enzyme in the degradation of lignin by fungi. Reports indicate that the activity of this enzyme ranges from 3.5 to 484,000 U L − 1 . Our aim was to analyze how laccase activity is calculated in the literature, and to determine statistically whether variations in activity are due to biological properties or to inconsistencies in calculation. We found a general lack of consensus on the definition of enzyme activity, and enzymes are sometimes characterized in terms of reaction rate and specific activity. Moreover, enzyme activity is calculated using at least seven different equations. Therefore, it is critical to standardize the calculation of laccase activity in order to compare results directly. The strain Pleurotus ostreatus Florida f6, its 45 basidiospore-derived isolates (both monokaryons and dikaryons prepared in our laboratory), Trametes versicolor strain CCBAS 614 and 22 other T. versicolor isolates obtained from the sporocarps collected in distant localities were successfully preserved for 12 y using perlite and straw cryopreservation protocols. All tested isolates survived a 12-year storage in liquid nitrogen (LN) and their laccase production and Poly B411 decolorization capacity was preserved. Also mycelium extension rate and the types of colony appearance of individual isolates remained unchanged. Different cryopreservation techniques were also tested for the short time (24 h) and the long time (6 m) storage of the culture liquid with extracellular laccase produced by T. versicolor strain CCBAS 614. The results showed that 10 % glycerol was the most suitable cryopreservant. The absence of the cryopreservant did not cause high loss of laccase activity in the samples; the presence of DMSO (5 or 10 %) in LN-stored samples caused mostly a decrease of laccase activity. Collections of a pleurotoid fungus from dead aspen in eastern Russia were initially identified as Lentinus sp., then as Phyllotopsis nidulans. DNA sequencing of cultures derived from these specimens using the nuclear ribosomal 28S (nrLSU) and nuclear ribosomal ITS1-5.8S-ITS2 regions (nrITS) showed that they were neither Lentinus nor Phyllotopsis and were not related to other pleurotoid genera Hohenbuehelia and Pleurotus. Subsequent investigation showed that the Russian fungus was the same as Pleurotus vetlinianus described from Poland. Screening of thirteen mushroom fungal isolates collected from Alagar Hill and Thandikudi Hill of Tamil Nadu, India, for laccase production revealed the high laccase production potential of the isolate MI 51. The mushroom isolate MI 51 was identified as Polyporus sp., based on sporophore (fruiting body) morphology and spore characteristics. Molecular identification of the fungal isolate MI 51 using primer ITS1 and ITS4 showed that isolate MI 51 shared 98 % sequence similarity with Polyporus arcularius, a basidiomycete fungus. Initial screening of production parameters using Plackett-Burman design helped in identifying the system variables/parameters which directly influenced laccase production. Laccase production by P. arcularius was optimized using central composite design (CCD) of experiments and response surface methodology (RSM) by studying influence of the growth supplements and media additives on modulating laccase production by P. arcularius. High laccase production by P. arcularius growing in basal salt medium, (9.30 IU/ml), was observed in 21 days of incubation. Results from experiments designed by CCD indicated that nearly threefold increase in laccase production (28.30 IU/ml) over the control experiments in basal salt medium could be obtained earlier at 15 days of incubation by increasing the content of the nitrogen source, yeast extract to 0.5 g/l, and by adding 250 μM CuSO to the basal salt medium. A novel laccase gene Lac1 and its cDNA were cloned from a white-rot fungus Cerrena sp. and characterized. The 1554-bp cDNA of Lac1 encoded a mature protein with 497 amino acids, preceded by a signal peptide of 20 amino acids. An unconventional intron splice site and incomplete splicing variants of Lac1 were observed. Lac1 was heterologously expressed in the yeast host Pichia pastoris , and a maximal laccase activity of 6.3 U mL −1 in the fermentation broth was achieved after fermentation for 9 days. The recombinant protein rLac1 was purified, and its enzymatic properties and functional characteristics were investigated. When ABTS was used as the substrate, the enzyme was most active at pH 3.5 and 55 °C, and stable at pH 4–10 and 20–60 °C. The K m and k cat values of rLac1 toward ABTS were 28.9 μM and 332.4 s −1 , respectively. Furthermore, rLac1 was tolerant to common metal ions up to 100 mM concentration and capable of decolorizing structurally different dyes in the absence of a redox mediator. Laccases are members of a large family of multicopper oxidases that catalyze the oxidation of a wide range of organic and inorganic substrates accompanied by the reduction of dioxygen to water. A new laccase was isolated from the basidiomycete Coriolopsis caperata strain 0677 and its amino-acid sequence was determined. According to its physicochemical properties and spectroscopic features, the laccase from C. caperata is a high redox-potential blue laccase. Attempts to crystallize the native enzyme were unsuccessful. The copper type 2-depleted (T2D) laccase was prepared and crystallized. The structure of T2D laccase from C. caperata was solved at 1.6 Å resolution, and attempts to reconstruct the T2 copper centre were performed using Cu+ and Cu2+ ions. The structure of T2D+Cu+ laccase was solved at 1.89 Å resolution. It was shown that the T2D+Cu+ laccase structure contained four copper ions in the active site. Aspergillus flavus is a well-known ubiquitous fungus able to contaminate both in pre- and postharvest period different feed and food commodities. During their growth, these fungi can synthesise aflatoxins, secondary metabolites highly hazardous for animal and human health. The requirement of products with low impact on the environment and on human health, able to control aflatoxin production, has increased. In this work the effect of the basidiomycete Trametes versicolor on the aflatoxin production by A. flavus both in vitro and in maize, was investigated. The goal was to propose an environmental loyal tool for a significant control of aflatoxin production, in order to obtain feedstuffs and feed with a high standard of quality and safety to enhance the wellbeing of dairy cows. The presence of T. versicolor, grown on sugar beet pulp, inhibited the production of aflatoxin B1 in maize by A. flavus. Furthermore, treatment of contaminated maize with culture filtrates of T. versicolor containing ligninolytic enzymes, showed a significant reduction of the content of aflatoxin B1. For preservation of 31 basidiomycete strains on perlite in cryovials we used five different perlite protocols to compare their applicability in laboratories with different equipment, namely a viability of the controlled freezing device or the electric deep-freezer and liquid nitrogen supply. The viability of the strains, macromorphological characteristics and the production of laccase were tested after 48 h, six months and one year of storage in the respective device. Our results indicated that the different response to the freezing/thawing process is an intrinsic feature of the respective strain. Nevertheless, the highest viability and preservation of laccase production in our tested strains was found when we used pre-freezing to −80 °C at a freezing rate of 1 °C/min in a programmable IceCube 1800 freezer or in freezing container Mr. Frosty before storage in liquid nitrogen or at ultra-low temperature freezer at −80 °C, respectively. The two abovementioned protocols enable all tested strains to survive three successive freezing/thawing cycles without substantial reduction of growth rate. The majority of the strains also do not lose laccase production. Our results showed that direct immersion of the strains into liquid nitrogen or placing them into −80 °C without pre-freezing is not suitable for basidiomycete cryopreservation. Laccases (EC 1.10.3.2) are a class of multi-copper oxidases with important industrial values. A basidiomycete strain Cerrena sp. HYB07 with high laccase yield was identified. After cultivation in the shaking flask for 4 days, a maximal activity of 210.8 U mL−1 was attained. A 58.6-kDa laccase (LacA) with 7.2% carbohydrate and a specific activity of 1952.4 U mg−1 was purified. 2,2′-Azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) was the optimal substrate, with Km and kcat being 93.4 µM and 2468.0 s−1, respectively. LacA was stable at 60°C, pH 5.0 and above, and in organic solvents. Metal ions Na+, K+, Ca2+, Mg2+, Mn2+, Zn2+ enhanced LacA activity, while Fe2+ and Li+ inhibited LacA activity. LacA decolorized structurally different dyes and a real textile effluent. Its gene and cDNA sequences were obtained. Putative cis-acting transcriptional response elements were identified in the promoter region. The high production yield and activity, robustness and dye decolorizing capacity make LacA and Cerrena sp. Abstract: Laccases (p-diphenol: oxygen oxidoreductase; EC 1.10.3.2) are multi-copper oxidases encoded by gene family in white rot fungi. Auricularia auricula-judae is one kind of white rot fungi with a soft, jelly-like texture and an ear-like shape. In the present study, seven laccase genes containing the signature sequences L1–L4 were isolated from A. auricula-judae strain Au916 on the basis of the mycelium-derived transcriptome. In the basidiomycetes, the predicted substrate binding loops of the A. auricula-judae laccases were found to be uncommon. Phylogenetic analysis showed that the laccases of the Auricularia were nested into the ascomycete laccases, indicating that the laccase genes from Auricularia are distinctly different in function from other basidiomycetes. Among the seven laccases, the intron positions and cluster distributions in the NJ tree varied from each other and the expression patterns of seven genes estimated by qRT-PCR were also discrepant. The lcc3 gene was highly expressed not only in the free-living mycelium but also in substrate mycelium, furthermore, the lcc5 gene was mostly expressed during the fruiting body formation and maturation indicating that lcc5 might play a major role during the sexual reproduction stage. The detail understanding of physiological/biochemical characteristics of individual laccase isoenzymes in fungi is necessary for fundamental and application purposes, but our knowledge is still limited for most of fungi due to difficult to express laccases heterologously. In this study, two novel laccase genes, named lac3 and lac4, encoding proteins of 547 and 532-amino acids preceded by 28 and 16-residue signal peptides, respectively, were cloned from the edible basidiomycete Coprinus comatus. They showed 70% identity but much lower homology with other fungal laccases at protein level (less than 58%). Two novel laccase isoenzymes were successfully expressed in Pichia pastoris by fusing an additional 10 amino acids (Thr-Pro-Phe-Pro-Pro-Phe-Asn-Thr-Asn-Ser) tag at N-terminus, and the volumetric activities could be dramatically enhanced from undetectable level to 689 and 1465 IU/l for Lac3 and Lac4, respectively. Both laccases possessed the lowest Km and highest kcat/Km value towards syringaldazine, followed by ABTS, guaiacol and 2,6-dimethylphenol similar as the low redox potential laccases from other microorganisms. Lac3 and Lac4 showed resistant to SDS, and retained 31.86% and 43.08% activity in the presence of 100 mM SDS, respectively. Lac3 exhibited higher decolorization efficiency than Lac4 for eleven out of thirteen different dyes, which may attribute to the relatively higher catalytic efficiency of Lac3 than Lac4 (in terms of kcat/Km) towards syringaldazine and ABTS. In this study thirty wood rotting and litter degrading basidiomycetes were screened for the production of lignocellulolytic enzymes such as, laccase, peroxidase, and cellulase using rapid micro quantification assay. Out of the 30 indigenous isolates Trametes gibbosa was identified to be a potential lignocellulolytic enzyme producer, producing a maximum amount of cellulase (299.66 ± 1.59 IU/L) and laccase (257.94 ± 1.79 U/L). Moreover, it is the second leading producer of peroxidase enzyme (170.19 ± 1.98 U/L). Tricholomopsis sp. a wood rot basidiomycete was found to be the leading lignin decomposer with maximum peroxidase activity (287.84±2 U/L) and second maximum laccase activity (250.19 ±1.83 U/L). However, its cellulolytic potential was found to be moderate (100.04±1.13 U/L). A higher level of lignocellulolytic enzymes was recorded in wood rotting basidiomycetes, whereas very low levels of lignolytic enzymes were found in litter inhabiting basidiomycetes. A white-rot basidiomycete, isolated from decayed acacia wood (from Northwest of Tunisia) and identified as Trametes sp, was selected in a broad plate screening because of its ability to decolorize and dephenolize olive oil mill wastewater (OMW) efficiently. The major laccase was purified and characterized as a monomeric protein with apparent molecular mass of 61 kDa (SDS-PAGE). It exhibits high enzyme activity over broad pH and temperature ranges with optimum activity at pH 4.0 and a temperature of 60 °C. The purified laccase is stable at alkaline pH values. The enzyme retained 50 % of its activity after 90 min of incubation at 55 °C. Using ABTS, this laccase presented K and V values of 0.05 mM and 212.73 μmoL min mg, respectively. It has shown a degrading activity towards a variety of phenolic compounds. The purified laccase was partially inhibited by Fe, Zn, Cd and Mn, while Cu acted as inducer. Background: The basidiomycete Pleurotus ostreatus is an efficient producer of laccases, a group of enzymes appreciated for their use in multiple industrial processes. The aim of this study was to reveal the molecular basis of the superiority of laccase production by dikaryotic strains compared to their parental monokaryons. Methodology/Principal Findings: We bred and studied a set of dikaryotic strains starting from a meiotic population of monokaryons. We then completely characterised the laccase allelic composition, the laccase gene expression and activity profiles in the dikaryotic strain N001, in two of its meiotic full-sib monokaryons and in the dikaryon formed from their mating. Conclusions/Significance: Our results suggested that the dikaryotic superiority observed in laccase activity was due to non-additive transcriptional increases in lacc6 and lacc10 genes. Furthermore, the expression of these genes was divergent in glucose- vs. lignocellulose-supplemented media and was highly correlated to the detected extracellular laccase activity. Background: Basidiomycete high-redox potential laccases (HRPLs) working in human physiological fluids (pH 7.4, 150 mM NaCl) arise great interest in the engineering of 3D-nanobiodevices for biomedical uses. In two previous reports, we described the directed evolution of a HRPL from basidiomycete PM1 strain CECT 2971: i) to be expressed in an active, soluble and stable form in Saccharomyces cerevisiae, and ii) to be active in human blood. In spite of the fact that S. cerevisiae is suited for the directed evolution of HRPLs, the secretion levels obtained in this host are not high enough for further research and exploitation. Thus, the search for an alternative host to over-express the evolved laccases is mandatory. Results: A blood-active laccase (ChU-B mutant) fused to the native/evolved а-factor prepro-leader was cloned under the control of two different promoters (PAOX1 and PGAP) and expressed in Pichia pastoris. The most active construct, which contained the PAOX1 and the evolved prepro-leader, was fermented in a 42-L fed-batch bioreactor yielding production levels of 43 mg/L. The recombinant laccase was purified to homogeneity and thoroughly characterized. As happened in S. cerevisiae, the laccase produced by P. pastoris presented an extra N-terminal extension (ETEAEF) generated by an alternative processing of the а-factor pro-leader at the Golgi compartment. The laccase mutant secreted by P. pastoris showed the same improved properties acquired after several cycles of directed evolution in S. cerevisiae for blood-tolerance: a characteristic pH-activity profile shifted to the neutral-basic range and a greatly increased resistance against inhibition by halides. Slight biochemical differences between both expression systems were found in glycosylation, thermostability and turnover numbers. Conclusions: The tandem-yeast system based on S. cerevisiae to perform directed evolution and P. pastoris to over-express the evolved laccases constitutes a promising approach for the in vitro evolution and production of these enzymes towards different biocatalytic and bioelectrochemical applications. The presence of genes encoding lignin peroxidase, laccase, and manganese peroxidase was assessed in more than 20 types of polypore fungi collected in the woods of Kirov oblast; the fungi studied had not been previously characterized with regard to ligninolytic enzyme production. Fifteen isolates of eleven basidiomycete species were shown to contain genes coding for all three ligninolytic enzymes. Genes coding for these enzymes were detected in D. mollis, D. quercina, F. pinicola, G. trabeum, G. lucidum, H. fasciculare, L. betulina, P. betulinus, P. igniarus, P. pomaceus, P. pini, and P. cinnabarinus for the first time. Protein profiles of the basidiomycete Trametes hirsuta grown on standard medium without laccase as an inducer and on medium supplemented with CuSO were analyzed using a differential proteomics approach. Protocols developed for isolation and purification of extracellular and intracellular proteins of the mycelium allowed us to show extensive extraction of protein components. Simultaneously, components hampering two-dimensional electrophoresis (pigments, low molecular mass metabolites) were removed from the samples, and high-resolution protein maps were obtained. Analysis of the basidiomycete secretomes revealed qualitative changes in the protein profile: the addition of CuSO as an inducer resulted in increase in the produced laccase isoforms and/or isozymes from 7 to 11, whereas its p I range change exceeded 2 units. The number of separated intracellular protein components was 552 and 502 for the control medium and medium with the inducer, respectively. Comparative analysis of the protein maps revealed five regions with the most pronounced differences in the protein profiles. The proteins of interest were identified using MALDI-TOF/TOF mass spectrometry with subsequent peptide fingerprinting. Some intracellular proteins (β-subunits of ATP synthase, molecular chaperones, chaperone activator) upregulated during the growth on the inducer-containing medium were identified. Aims: This study was undertaken to investigate laccase-containing basidiomycete communities at the cDNA and DNA levels and to assess the influences of vegetation and soil types on the basidiomycete communities in forest soils. Materials and methods: Soil samples were collected from the upper soil layers of two typical subtropical forests (a broad-leaved forest developed in Karst limestone soil and an artificial coniferous forest with Pinus massoniana in red soil) in China. The basidiomycete communities were characterized by cloning and sequencing of the laccase genes at both the cDNA and DNA levels. Compositions of lignin monomers were determined by gas chromatography-mass spectrometry. Results: Most of the laccase genes obtained in this study were new, highlighting the research gap of this functional group. The trends of the basidiomycetous laccase gene diversity among the upper soil layers of the two forests were consistent between the cDNA and DNA levels. The Agaricales had high activity because they dominated all the tested soils. However, the total basidiomycete communities reflected at the cDNA and DNA levels were significantly different due to the presence of some quiescent basidiomycetous groups. Almost all of the lignin components were decomposed from the O to the A layers in the two forests, and laccases produced by Agaricales were likely responsible for the decomposition of guaiacyl monomers. Both vegetation and soil types had great influences on the active laccase-containing basidiomycete communities, primarily via the pH, C/N, and the contents of lignin monomers. Conclusions: The cDNA- and DNA-level approaches presented good consistency of diversities but different compositions of laccase-containing basidiomycete communities, thus emphasizing the importance of focusing on laccase genes at the cDNA level in future studies. The discharge of highly coloured synthetic dye effluents into rivers and lakes is harmful to the water bodies, and therefore, intensive researches have been focussed on the decolorization of wastewater by biological, physical or chemical treatments. In the present study, 12 basidiomycetes strains from the genus Pleurotus, Trametes, Lentinus, Peniophora, Pycnoporus, Rigidoporus, Hygrocybe and Psilocybe were evaluated for decolorization of the reactive dyes Cibacron Brilliant Blue H- GR and Cibacron Red FN-2 BL, both in solid and liquid media. Among the evaluated fungi, seven showed great ability to decolorize the synthetic textile effluent, both in vivo (74-77%) or in vitro (60-74%), and laccase was the main ligninolytic enzyme involved on dyes decolorization. Pleurotus ostreatus, Trametes villosa and Peniophora cinerea reduced near to 60% of the effluent colour after only 1 h of treatment. The decolorization results were still improved by establishing the nitrogen source and amount to be used during the fungal strains cultivation in synthetic medium previous their action on the textile effluent, with yeast extract being a better nitrogen source than ammonium tartarate. Abstract: Little is known on basidiomycete sources of feruloyl esterases (FAEs), although many wood-rotting representatives of these fungi typically grow on feruloyl-rich substrates. A major reason is that the almost ubiquitous presence of laccases interferes with the detection of FAE activity. Laccases polymerize the liberated ferulic acid (FA) in situ, thus detracting the product of enzymatic hydrolysis from its detection. A rapid HPLC-UV method was developed to detect the loss of FA, but also to quantify the hydrolysis of FA esters. The method allows at the same time to evaluate the substrate specificity of a FAE. Forty one basidiomycetes were tested for their FAE activities, and 25 out of the set were positive. The basidiomycetes hydrolyzing cinnamates with the highest conversion rates were Auricularia auricula-judae and Marasmius scorodonius. Moreover, a new FAE inducer, the nonionic detergent Tween 80, was found. This is the first comprehensive study on basidiomycete sources of FAEs. [Copyright &y& Elsevier]

The lignin-degrading, biopulping white-rot fungus Physisporinus rivulosus secretes several laccases of distinct features such as thermostability, extremely low pH optima and thermal activation for oxidation of phenolic substrates. Here we describe the cloning, heterologous expression and structural and enzymatic characterisation of two previously undescribed P. rivulosus laccases. The laccase cDNAs were expressed in the methylotrophic yeast Pichia pastoris either with the native or with Saccharomyces cerevisiae α-factor signal peptide. The specific activity of rLac1 and rLac2 was 5 and 0.3 μkat/μg, respectively. However, mutation of the last amino acid in the rLac2 increased the specific laccase activity by over 50-fold. The recombinant rLac1 and rLac2 enzymes demonstrated low pH optima with both 2,6-dimethoxyphenol (2,6-DMP) and 2,2′-azino-bis(3-ethylbenzathiazoline-6-sulfonate). Both recombinant laccases showed moderate thermotolerance and thermal activation at +60 °C was detected with rLac1. By homology modelling, it was deduced that Lac1 and Lac2 enzymes demonstrate structural similarity with the Trametes versicolor and Trametes trogii laccase crystal structures. Comparison of the protein architecture at the reducing substrate-binding pocket near the T1-Cu site indicated the presence of five amino acid substitutions in the structural models of Lac1 and Lac2. These data add up to our previous reports on laccase production by P. rivulosus during biopulping and growth on Norway spruce. Heterologous expression of the novel Lac1 and Lac2 isoenzymes in P. pastoris enables the detailed study of their properties and the evaluation of their potential as oxidative biocatalysts for conversion of wood lignin, lignin-like compounds and soil-polluting xenobiotics. During directed evolution to functionally express the high redox potential laccase from the PM1 basidiomycete in Saccharomyces cerevisiae, the characteristic maximum absorption at the T1 copper site (Abs610T1Cu) was quenched, switching the typical blue colour of the enzyme to yellow. To determine the molecular basis of this colour change, we characterized the original wild-type laccase and its evolved mutant. Peptide printing and MALDI-TOF analysis confirmed the absence of contaminating protein traces that could mask the Abs610T1Cu, while conservation of the redox potential at the T1 site was demonstrated by spectroelectrochemical redox titrations. Both wild-type and evolved laccases were capable of oxidizing a broad range of substrates (ABTS, guaiacol, DMP, synapic acid) and they displayed similar catalytic efficiencies. The laccase mutant could only oxidize high redox potential dyes (Poly R-478, Reactive Black 5, Azure B) in the presence of exogenous mediators, indicating that the yellow enzyme behaves like a blue laccase. The main consequence of over-expressing the mutant laccase was the generation of a six-residue N-terminal acidic extension, which was associated with the failure of the STE13 protease in the Golgi compartment giving rise to alternative processing. Removal of the N-terminal tail had a negative effect on laccase stability, secretion and its kinetics, although the truncated mutant remained yellow. The results of CD spectra analysis suggested that polyproline helixes were formed during the directed evolution altering spectral properties. Moreover, introducing the A461T and S426N mutations in the T1 environment during the first cycles of laboratory evolution appeared to mediate the alterations to Abs610T1Cu by affecting its coordinating sphere. Some of the industries that discharge highly colored effluents are paper and pulp mills, textiles and dye-making industries, alcohol distilleries and leather industries. Terrestrial white-rot basidiomycetous fungi and their lignindegrading enzymes laccase, manganese-peroxidase and lignin peroxidases are useful in the treatment of colored industrial effluents and other xenobiotics.In the present study different 13 species of fungi were isolated from the effluent. Among the isolated fungi Aspergillus was the dominant genus in the effluent.Except dissolved oxygen, all other parameters showed decreased level when compare to control.The BOD and COD level were reduced significantly. This study reports a new white-rot fungus Cerrena sp. WR1, identified based on an 18S rDNA sequence, which can secrete extracellular forms of laccase with a maximal activity reaching 202 000 U l−1 in a 5-l fermenter. A laccase protein, designated Lcc3, was purified and shown to be N-linked glycosylated by PNGase F and liquid chromatography tandem mass spectrometry analyses. The respective full-length cDNA gene (lcc3) of the Lcc3 protein was obtained using polymerase chain reaction-based methods. Kinetic studies showed that the Km and kcat of the native Lcc3 were 3.27 μM and 934.6 s−1 for 2,2′-Azino-bis-(3-ethylbenzthiazoline-6-sulfonic acid), 849.1 μM and 147.9 s−1 for guaiacol, 392.7 μM and 109.2 s−1 for 2,6-dimethoxyphenol, and 881 μM and 115.5 s−1 for catechol, respectively. The Tm of Lcc3 was determined at 73.9°C and it showed a long t1/2 (120 min) at 50°C. The laccase was highly ethanol resistant, with 80% of its original activity was detected when incubated in 25% ethanol for 14 days. Furthermore, crude enzyme broth or Lcc3 could degrade lignin in kraft paper (26.5%), and showed high decoloration efficiency (90%) on synthetic dye Remazol Brilliant Blue R. Abstract: This study revealed a dual effect of copper on D. squalens laccase activity, inducing and suppressing. The addition of copper sulfate led to enhancement of the enzyme activity until it reached a maximum value at peak copper sulfate concentration (PC) followed by suppression and even complete termination of the enzyme activity. Maximum laccase yield is a function of fungi tolerance and media composition. Nutrients had a significant impact on copper-independent and copper-induced laccase activity. Some nutrients also improved tolerance of cells and laccase-producing system to copper load. The effect of specific nutrient depended on the ratio of carbon and nitrogen sources. This research can be used to predict the effective copper concentration and nutrient composition to stimulate the laccase production without compromising the fungi growth. The methylated substrates (3-O-methylglucose, methylcellulose) in combination with casein and copper demonstrated high potential to support fungi tolerance and enhanced laccase activity.
