Historical salmon catch records suggest that climatic variability, and more recently human exploitation, control patterns of abundance in Atlantic salmon populations. We present the first long-term (2000-year) reconstruction of Atlantic Salmon population variations based on a Marine Derived Nutrient (MDN) lake sediment record. Our record is constructed from nitrogen isotopes (δ15N) measured from a lake sediment core, which we compare with an escapement index (EI) derived from historic net catch data on major Scottish salmon rivers. We used an isotope mixing model to demonstrate that the N isotope values are likely enriched with MDN and demonstrate that Loch Insh sediments are enriched compared with a control site (Loch Vaa) that has never had exposure to salmon. We demonstrate that current adult spawner returns are around half that of historic values prior to major human exploitation. Before the onset of widespread human exploitation and habitat degradation, large fluctuations in salmon abundance are attributed to variations in North Atlantic sea surface temperature. While our data support published reconstructions of declining Atlantic salmon stocks in Northwest European rivers over the last 1000 years, rather than point to a solely human cause, the human impact appears to be overprinted on larger-scale changes in marine habitat occurring at the transition from the warmer Medieval Climatic Anomaly (MCA) to the cooler Little Ice Age (LIA). The collapse of the steppe-tundra biome (mammoth steppe) at the end of the Pleistocene is used as an important example of topdown ecosystem cascades, where human hunting of keystone species led to profound changes in vegetation across high latitudes in the Northern Hemisphere. Alternatively, it is argued that this biome transformation occurred through a bottom-up process, where climate-driven expansion of shrub tundra (Betula, Salix spp.) replaced the steppe-tundra vegetation that grazing megafauna taxa relied on. In eastern Beringia, these differing hypotheses remain largely untested, in part because the precise timing and spatial pattern of Late Pleistocene shrub expansion remains poorly resolved. This uncertainty is caused by chronological ambiguity in many lake sediment records, which typically rely on radiocarbon (14C) dates from bulk sediment or aquatic macrofossils--materials that are known to overestimate the age of sediment layers. Here, we reexamine Late Pleistocene pollen records for which 14C dating of terrestrial macrofossils is available and augment these data with 14C dates from arctic ground-squirrel middens and plant macrofossils. Comparing these paleovegetation data with a database of published 14C dates from megafauna remains, we find the postglacial expansion of shrub tundra preceded the regional extinctions of horse (Equus spp.) and mammoth (Mammuthus primigenius) and began during a period when the frequency of 14C dates indicates large grazers were abundant. These results are not consistent with a model of top-down ecosystem cascades and support the hypothesis that climate-driven habitat loss preceded and contributed to turnover in mammal communities. While there are extensive macro‐ and microfossil records of a range of plants and animals from the Quaternary, earthworms and their close relatives amongst annelids are not preserved as fossils and therefore the knowledge of their past distributions is limited. This lack of fossils means that clitellate worms (Annelida) are currently underused in palaeoecological research, even though they can provide valuable information about terrestrial and aquatic environmental conditions. Their DNA might be preserved in sediments, which offers an alternative method for detection. Here we analyse lacustrine sediments from lakes in the Polar Urals, Arctic Russia, covering the period 24 000–1300 cal. a BP, and NE Norway, covering 10 700–3300 cal. a BP, using a universal mammal 16S rDNA marker. While mammals were recorded using the marker (reindeer was detected twice in the Polar Urals core at 23 000 and 14 000 cal. a BP, and four times in the Norwegian core at 11 000 cal. a BP and between 3600–3300 cal. a BP), worm extracellular DNA 'bycatch' was rather high. In this paper we present the first reported worm detection from ancient DNA. Our results demonstrate that both aquatic and terrestrial clitellates can be identified in late‐Quaternary lacustrine sediments, and the ecological information retrievable from this group warrants further research with a more targeted approach. Metabarcoding of lake sediments have been shown to reveal current and past biodiversity, but little is known about the degree to which taxa growing in the vegetation are represented in environmental DNA (eDNA) records. We analysed composition of lake and catchment vegetation and vascular plant eDNA at 11 lakes in northern Norway. Out of 489 records of taxa growing within 2 m from the lake shore, 17–49% (mean 31%) of the identifiable taxa recorded were detected with eDNA. Of the 217 eDNA records of 47 plant taxa in the 11 lakes, 73% and 12% matched taxa recorded in vegetation surveys within 2 m and up to about 50 m away from the lakeshore, respectively, whereas 16% were not recorded in the vegetation surveys of the same lake. The latter include taxa likely overlooked in the vegetation surveys or growing outside the survey area. The percentages detected were 61, 47, 25, and 15 for dominant, common, scattered, and rare taxa, respectively. Similar numbers for aquatic plants were 88, 88, 33 and 62%, respectively. Detection rate and taxonomic resolution varied among plant families and functional groups with good detection of e.g. Ericaceae, Roseaceae, deciduous trees, ferns, club mosses and aquatics. The representation of terrestrial taxa in eDNA depends on both their distance from the sampling site and their abundance and is sufficient for recording vegetation types. For aquatic vegetation, eDNA may be comparable with, or even superior to, in-lake vegetation surveys and may therefore be used as an tool for biomonitoring. For reconstruction of terrestrial vegetation, technical improvements and more intensive sampling is needed to detect a higher proportion of rare taxa although DNA of some taxa may never reach the lake sediments due to taphonomical constrains. Holocene tephrostratigraphy in Alaska provides independent chronology and stratigraphic correlation in a region where reworked old (Holocene) organic carbon can significantly distort radiocarbon chronologies. Here, we present new glass chemistry and chronology for Holocene tephras preserved in three Alaskan lakes: one in the eastern interior and two in the southern Brooks Range. Tephra beds in the eastern interior lake-sediment core are correlated with the White River Ash and the Hayes tephra set H (~4200–3700 cal yr BP), and an additional discrete tephra bed is likely from the Aleutian arc/Alaska Peninsula. Cryptotephras (nonvisible tephras) found in the Brooks Range include the informally named “Ruppert tephra” (~2700–2300 cal yr BP) and the Aniakchak caldera-forming event II (CFE II) tephra (~3600 cal yr BP). A third underlying Brooks Range cryptotephra is chemically indistinguishable from the Aniakchak CFE II tephra (4070–3760 cal yr BP) and is likely to be from an earlier eruption of the Aniakchak volcano. Sedimentary DNA (sedDNA) has recently emerged as a new proxy for reconstructing past vegetation, but its taphonomy, source area and representation biases need better assessment. We investigated how sedDNA in recent sediments of two small Scottish lakes reflects a major vegetation change, using well-documented 20th Century plantations of exotic conifers as an experimental system., We used next-generation sequencing to barcode sedDNA retrieved from subrecent lake sediments. For comparison, pollen was analysed from the same samples., The sedDNA record contains 73 taxa (mainly genus or species), all but one of which are present in the study area. Pollen and sedDNA shared 35% of taxa, which partly reflects a difference in source area. More aquatic taxa were recorded in sedDNA, whereas taxa assumed to be of regional rather than local origin were recorded only as pollen., The chronology of the sediments and planting records are well aligned, and sedDNA of exotic conifers appears in high quantities with the establishment of plantations around the lakes. Abstract: Burial Lake in northwest Alaska records changes in water level and regional vegetation since ∼39,000calyr BP based on terrestrial macrofossil AMS radiocarbon dates. A sedimentary unconformity is dated between 34,800 and 23,200calyr BP. During all or some of this period there was a hiatus in deposition indicating a major drop in lake level and deflation of lacustrine sediments. MIS 3 vegetation was herb-shrub tundra; more xeric graminoid-herb tundra developed after 23,200calyr BP. The tundra gradually became more mesic after 17,000calyr BP. Expansions of Salix then Betula, at 15,000 and 14,000calyr BP, respectively, are coincident with a major rise in lake level marked by increasing fine-grained sediment and higher organic matter content. Several sites in the region display disrupted sedimentation and probable hiatuses during the last glacial maximum (LGM); together regional data indicate an arid interval prior to and during the LGM and continued low moisture levels until ∼15,000calyr BP. AMS 14C dates from Burial Lake are approximately synchronous with AMS 14C dates reported for the Betula expansion at nearby sites and sites across northern Alaska, but 1000–2000yr younger than bulk-sediment dates. When tracing vegetation dynamics over long timescales, obtaining enough floristic information to gain a detailed understanding of past communities and their transitions can be challenging. The first high-resolution sedimentary DNA (sed aDNA) metabarcoding record from lake sediments in Alaska—reported here—covers nearly 15,000 years of change. It shows in unprecedented detail the composition of late-Pleistocene "steppe-tundra" vegetation of ice-free Alaska, part of an intriguing late-Quaternary "no-analogue" biome, and it covers the subsequent changes that led to the development of modern spruce-dominated boreal forest. The site (Chisholm Lake) lies close to key archaeological sites, and the record throws new light on the landscape and resources available to early humans. Initially, vegetation was dominated by forbs found in modern tundra and/or subarctic steppe vegetation (e.g., Potentilla , Draba , Eritrichium , Anemone patens), and graminoids (e.g., Bromus pumpellianus, Festuca, Calamagrostis, Puccinellia), with Salix the only prominent woody taxon. Predominantly xeric, warm-to-cold habitats are indicated, and we explain the mixed ecological preferences of the fossil assemblages as a topo-mosaic strongly affected by insolation load. At ca. 14,500 cal yr BP (calendar years before C.E. 1950), about the same time as well documented human arrivals and coincident with an increase in effective moisture, Betula expanded. Graminoids became less abundant, but many open-ground forb taxa persisted. This woody-herbaceous mosaic is compatible with the observed persistence of Pleistocene megafaunal species (animals weighing ≥44 kg)—important resources for early humans. The greatest taxonomic turnover, marking a transition to regional woodland and a further moisture increase, began ca. 11,000 cal yr BP when Populus expanded, along with new shrub taxa (e.g., Shepherdia , Eleagnus, Rubus, Viburnum). Picea then expanded ca. 9500 cal yr BP, along with shrub and forb taxa typical of evergreen boreal woodland (e.g., Spiraea, Cornus, Linnaea). We found no evidence for Picea in the late Pleistocene, however. Most taxa present today were established by ca. 5000 cal yr BP after almost complete taxonomic turnover since the start of the record (though Larix appeared only at ca. 1500 cal yr BP). Abstract: Physical and geochemical proxy analyses of sediment cores from Harding Lake in central Alaska are used to reconstruct paleoenvironmental change and millennial scale fluctuations in lake level for the last ∼31,000 years. We analyzed a composite 422 cm core from the lake depocenter (42.1 m water depth) and identified 4 distinct lithologic units based on variability in dry bulk density, organic matter, biogenic silica, carbon to nitrogen mass ratios (C/N), organic matter carbon isotopes (δ13C), pollen, and elemental abundances via scanning X-ray fluorescence, with age control provided by 16 Accelerator Mass Spectrometry radiocarbon dates and 210Pb dating. In addition, we analyzed a transect of cores from 7.1 m, 10.75 m, 15.91 m, and 38.05 m water depths to identify lake level fluctuations and to characterize sediment compositional changes as a function of water depth. Organic matter content and magnetic susceptibility values in surface sediments from all transect cores show a strong correlation with water depth. Interpretation of four lithologic units with well-dated contacts produced a record of water-depth variations that is consistent with independent climate records from eastern Beringia. Basal coarse-grained sediments (quartz pebble diamicton) were deposited prior to 30,700 calendar years before present (yr BP), possibly from fluvial reworking or deflation during a period of severe aridity. Unit 1 sediments were deposited between 30,700 and 15,700 yr BP and are characterized by a low organic matter content, a high magnetic susceptibility, and low biogenic silica concentrations resulting from very low lake levels, low terrestrial and in-lake productivity and a high flux of clastic sediment. An abrupt increase in organic matter and biogenic silica concentration marks the transition into Unit 2 sediments, which were deposited between 15,700 and 9,400 yr BP when lake levels were higher and variable (relative to Unit 1). The transition to full interglacial conditions at 9,400 yr BP marks the beginning of Unit 3. Here an abrupt increase in the sedimentation rate, organic matter and biogenic silica concentration occurs (along with a corresponding decrease to low magnetic susceptibility). These high values persist until 8,700 yr BP, signifying a rapid rise to higher lake levels (in comparison to Units 1 and 2). Unit 4 sediments were deposited between 8,700 yr BP to 2010 AD and generally contain high concentrations of organic matter and biogenic silica with low magnetic susceptibility, suggesting that lake levels were relatively high and stable during the middle to late Holocene. Andøya on the NW coast of Norway is a key site for understanding the Last Glacial Maximum (LGM) in northern Europe. Controversy has arisen concerning the local conditions, especially about the timing and extent of local glacial cover, maximum July temperatures and whether pine and/or spruce could have grown there. We reviewed all existing data and add newly analysed ancient sedimentary DNA (seda DNA), pollen, macrofossils, geochemistry and stable isotopes from three lake sediment cores from Øvre Æråsvatnet. A total of 23 new dates and age-depth modelling suggests the lake has been ice-free since GI2 (<23.4 cal ka BP) and possibly GS3 (<26.7 cal ka BP). Pinus and Picea seda DNA was found in all three cores but at such low frequencies that it could not be distinguished from background contamination. LGM samples have an exceptionally high organic matter content, with isotopic values indicating that carbon and nitrogen derive from a marine source. Along with finds of bones of the little auk (Alle alle), this indicates that the lake received guano from an adjacent bird colony. Seda DNA, pollen and macrofossil assemblages were dominated by Poaceae, Brassicaceae and Papaver , but scattered occurrence of species currently restricted to the Low Arctic Tundra Zone (July temperature of 8–9 °C) such as Apiaceae (seda DNA, 8–9 °C), and Alchemilla alpina (macrofossil, 8–9 °C) were also recorded. The review of >14.7 cal ka BP data recorded 94 vascular plant taxa, of which 38% have a northern limit in Shrub Tundra or more southern vegetation zones. This unusual assemblage likely stems from a combination of proximity to ice-free water in summer, geographical isolation linked with stochastic long-distance dispersal events, and the presence of bird-fertilized habitats. The environmental reconstruction based on all records from the area does not preclude local growth of tree species, as the local climate combined with high nutrient input may have led to periodically suitable environmental 'hotspot' conditions. • High organic content of Last Glacial Maximum sediments from a northern refugium. • Isotopes and bones indicate birds roosted near the ice edge. • Conifer DNA found in sediments, but frequency not significantly above background contamination. • Review of all Andøya LGM studies reveals presence of boreal and arctic taxa. • Environmental reconstruction does not preclude intermittent growth conditions for trees.
