A PEG-Tyr block copolymer forms a kinetically stable colloidal solution in water at room temperature which undergoes an irreversible conversion to a gel phase upon heating. A micellar solution and a gel can therefore be studied under identical experimental conditions. This made it possible to compare physical properties and chemical reactivity of micelles and gels in identical chemical environments and under identical conditions. EPR spectra of the spin-labelled copolymer showed that tyrosine mobility in gels was slightly reduced compared to micelles. Chemical reactivity was studied using photochemical degradation of tyrosine and tyrosine dimerization, in the absence and in the presence of an Fe(iii) salt. The reactivity trends were explained by reduced tyrosine mobility in the gel environment. The largest reactivity difference in gels and micelles was observed for bimolecular dityrosine formation which was also attributed to the reduction in molecular mobility.

Host-guest interactions between nitroxide stable radicals and supramolecular coordination cages were investigated using electron paramagnetic resonance (EPR) spectroscopy in water and acetonitrile. TEMPO showed negligible association with the cages in water, while 4-oxo-TEMPO bound with a strength comparable to that previously reported for related ketones. Carboxylic acid-functionalized nitroxides bound strongly to the acetonitrile-soluble coordination cages. In all cases, host-guest complex formation resulted in significant decreases in the molecular tumbling rate of the guests, with tumbling becoming strongly anisotropic. The polarity of the cage environment in both solvents was found to be intermediate between water and acetonitrile.

EPR spectroscopy was used to monitor formation of free radicals in human hair upon UV irradiation. While the EPR spectra of brown hair were dominated by melanin signal, those of white hair were keratin-derived. The decay of UV induced keratin radicals was enhanced at increased ambient humidity. We argue that at higher humidity the swollen hair provides a more liquid-like environment, and higher molecular mobility in this environment leads to faster radical reactions. This interpretation is consistent with the increased UV-triggered protein damage in hair at high humidity as demonstrated by the protein loss, MALDI-TOF and FT-IR data.

Intramolecular spin exchange in four flexible PEG-based bis-nitroxides was studied by X-band electron paramagnetic resonance (EPR) spectroscopy as a function of temperature and solvent viscosity. A series of biradicals with different lengths of the ethylene glycol bridge connecting the two nitroxide groups was investigated in aqueous and mixed 1:1 i-propanol:water solutions. Conformational transitions in liquid solutions of the biradicals were analyzed, and thermodynamic parameters of the conformational reorganization were calculated from the EPR spectra. Intramolecular distances r between paramagnetic >N–O• fragments in biradicals in frozen solutions were calculated and compared with dynamic properties of the biradicals.

The formation of alginate gel from low molecular weight alginate and very low molecular weight alginate in the presence of divalent cations was investigated using Electron Paramagnetic Resonance (EPR) spectroscopy. The transition from sol to gel in the presence of divalent cations was monitored by the changes in the dynamics of spin labelled alginate. The immobilisation of the spin labelled alginate in the gel reflects the strength of interaction between the cation and alginate chain. Diffusion experiments showed that both the cation and alginate polyanion in the gel fibres can exchange with molecules in solution. In particular, we showed that dissolved alginate polyanions can replace alginates in the gel fibres, which can hence diffuse through the bulk of the gel. This illustrates the surprisingly highly dynamic nature of these gels and opens up the possibility of preparing multicomponent alginate gels via polyanion exchange process.

We describe the synthesis of spin-labeled bis-ureas which coassemble with bis-urea gelators and report on self-assembly as detected using electron paramagnetic resonance spectroscopy (EPR). Specifically, EPR detects the gel-sol transition and allows us to quantify how much spin-label is immobilized within the gel fibers and how much is present in mobile solvent pools-as controlled by temperature, gelator structure, and thermal history. EPR is also able to report on the initial self-assembly processes below the gelation threshold which are not macroscopically visible and appears to be more sensitive than NMR to intermediate-sized nongelating oligomeric species. By studying dilute solutions of gelator molecules and using either single or double spin-labels, EPR allows quantification of the initial steps of the hierarchical self-assembly process in terms of cooperativity and association constant. Finally, EPR enables us to estimate the degree of gel-fiber solvation by probing the distances between spin-labels. Comparison of experimental data against the predicted distances assuming the nanofibers are only composed of gelator molecules indicates a significant difference, which can be assigned to the presence of a quantifiable number of explicit solvent molecules. In summary, EPR provides unique data and yields powerful insight into how molecular-scale mobility and solvation impact on assembly of supramolecular gels.

Non-thermal atmospheric pressure ('cold') plasmas have received increased attention in recent years due to their significant biomedical potential. The reactions of cold plasma with the surrounding atmosphere yield a variety of reactive species, which can define its effectiveness. While efficient development of cold plasma therapy requires kinetic models, model benchmarking needs empirical data. Experimental studies of the source of reactive species detected in aqueous solutions exposed to plasma are still scarce. Biomedical plasma is often operated with He or Ar feed gas, and a specific interest lies in investigation of the reactive species generated by plasma with various gas admixtures (O2, N2, air, H2O vapor, etc.) Such investigations are very complex due to difficulties in controlling the ambient atmosphere in contact with the plasma effluent. In this work, we addressed common issues of 'high' voltage kHz frequency driven plasma jet experimental studies. A reactor was developed allowing the exclusion of ambient atmosphere from the plasma-liquid system. The system thus comprised the feed gas with admixtures and the components of the liquid sample. This controlled atmosphere allowed the investigation of the source of the reactive oxygen species induced in aqueous solutions by He-water vapor plasma. The use of isotopically labelled water allowed distinguishing between the species originating in the gas phase and those formed in the liquid. The plasma equipment was contained inside a Faraday cage to eliminate possible influence of any external field. The setup is versatile and can aid in further understanding the cold plasma-liquid interactions chemistry.

Electron paramagnetic resonance (EPR) spectroscopy and spin trapping were used to explore the mechanism of alcohol oxidation over gold catalysts. Reaction of secondary alcohols with supported and unsupported gold catalysts (e.g., Au/CeO(2), polymer-incarcerated Au nanoparticles, PPh(3)-protected Au nanoparticles) in the presence of spin traps led to the formation of a hydrogen spin adduct. Using isotope labeling, we confirmed that the hydrogen in the spin adduct originates from the cleavage of the C-H bond in the alcohol molecule. The formation of the hydrogen spin adduct most likely results from the abstraction of hydrogen from the Au surface by a spin trap. These results thus strongly suggest intermediate formation of Au-H species during alcohol oxidation. The role of oxygen in this mechanism is to restore the catalytic activity rather than oxidize alcohol. This was further confirmed by carrying out gold-catalyzed alcohol oxidation in the absence of oxygen, with nitroxides as hydrogen abstractors. The support (e.g., metal oxides) can activate oxygen and act as an H abstractor from the gold surface and hence lead to a faster recovery of the activity. Peroxyl radicals were also observed during alcohol oxidation, consistent with a free-radical autoxidation mechanism. However, this mechanism is likely to be a minor side reaction, which does not lead to the formation of an appreciable amount of oxidation products.

Electron spin-echo envelope modulation (ESEEM) spectroscopy was used to investigate intramolecular and intermolecular complexes of cyclodextrins (CDs) with a nitroxide group. The interaction with solvent molecules (D(2)O) was followed through the (2)H modulation depth. Competition experiments with adamantane-type guests were used to confirm complexation. The shielding of the nitroxide group from solvent upon inclusion into a CD cavity made this technique more sensitive to complexation than cw EPR spectroscopy. ESEEM analysis of a series of CDs mono and bis spin-labeled on the primary rim of the cavity showed that only one compound formed a self-inclusion complex. This suggests that significant linker length/flexibility is required for formation of inclusion complexes in functionalized CDs. DEER (double electron-electron resonance) experiments confirmed that the self-inclusion complex of the spin-labeled CD was intra- rather than intermolecular.

Cross-linking of ethylene propylene diene monomer (EPDM) rubbers containing different amounts of dicyclopentadiene (DCPD), 5-ethylidene-2-norbornene (ENB) and 5-vinyl-2-norbornene (VNB) dienes was examined by EPR spectroscopy. The cross-linking was initiated by thermal decomposition of dicumyl peroxide at 440 K. The concentration of free radicals increased towards the end of the cross-linking process before reaching a maximum and decaying to zero. This is explained by the spatial confinement of the radicals in the cross-linked rubber, which leads to increased life time and, hence, higher radical concentration at a time when most peroxide has decomposed. The EPR spectra showed the presence of two components: a well-resolved spectrum overlapping a broad line. Both components are assigned to allyl radicals possessing different mobility. The more mobile component is assigned to allyl radicals along the EPDM chains, whereas the immobilised allyl radicals are formed in the cross-links. The stability of the allyl radicals decreases in the order DCPD > ENB > VNB. EPDMs containing two dienes show more persistent radicals than their single-diene counterparts. The most persistent radicals are observed for highly cross-linked (e.g., 28% ENB) or mixed diene EPDMs (e.g., 2.2% DCPD–4.4% ENB); the EPR spectra of free radicals in these systems can be observed for several hours.

Variable temperature electron paramagnetic resonance (EPR) spectra of spin-labeled Au nanoparticles in toluene solution were recorded at X-, Q- and W-bands. The chain length of the nitroxide-based spin-labeled ligand and the surrounding alkanethiol ligands was systematically varied. The spectra were analyzed using the NLSL program (Budil, D. E.; Lee, S.; Saxena, S.; Freed, J. H. J. Magn. Reson., Ser. A 1996, 120, 155) which simulates slow motion spectra using the stochastic Liouville approach. The simulation results show that the rotational diffusion parameters for particles (∼2.6 nm Au core diameter) are dominated by the local motion of the ligand. The length of the spin label has the biggest effect on the nitroxide dynamics followed by the chain length of the surrounding ligands. The results are consistent with the loose packing of ligands on the nanoparticle surface. The packing density of ligands further decreases with the increasing distance from the Au surface. A series of Au nanoparticles modified with a nitroxide-functionalized ligand was prepared with a range of spin-label coverage. The X-band EPR spectra of frozen solutions of these nanoparticles showed coverage-dependent line-broadening due to dipole-dipole interactions between spin labels. We developed a methodology to analyze such spectra in terms of geometrical features of the nanoparticles (e.g., Au core size and the length of the spin-labeled ligand). Our method is based on the assumption that the spectral line shape is determined by the average distance between nearest-neighboring spin labels adsorbed on the Au particle. Geometrical and statistical analysis then relates this distance to the line shape parameter d1/d, which was calibrated using a model system. Application of this methodology to the experimental spectra provided information about the conformation of ligands on the Au surface. We found that, if the spin-labeled ligand is substantially longer than the surrounding protecting layer, it does not adopt a fully stretched conformation but wraps around the particle immediately above the layer of surrounding ligand. Our results also show that the ligands do not adsorb cooperatively on the Au surface.

The mechanism of a place-exchange reaction of ligand-protected Au nanoparticles was investigated using diradical disulfide spin labels. Analysis of reaction mixtures using a combination of GPC and EPR allowed us to determine concentration profile and propose a kinetic model for the reaction. In this model, only one branch of the disulfide ligand is adsorbed on the Au surface during exchange; the other branch forms mixed disulfide with the outgoing ligand. The two branches of the disulfide ligand therefore do not adsorb in adjacent positions on the surface of Au nanoparticles; this was ultimately proven by the powder EPR spectra of frozen exchange reaction mixtures. Our data also suggest the presence of different binding sites with different reactivity in the exchange reaction. The most-active sites are likely to be nanoparticle surface defects.

The properties of a gel consisting of a covalent network formed by the reaction of isocyanate end-capped polyethylene glycol (PEG) with β-cyclodextrin, were investigated by EPR spectroscopy. Spin-labelled cyclodextrin was incorporated into the cross-link points of the gel and at the chain ends. The dynamics of the gel fibres as reported by the spin label, was found to be sensitive to the H-bonding ability of the solvent, density of cross-links and temperature. Addition of spin probes (e.g., TEMPO and adamantane-TEMPO) to the unlabelled gel made it possible to characterise the solvent pools in the gel. While TEMPO was uniformly distributed throughout the solvent pools, the adamantane derivative was located at the gel fibre-solvent pool interface; these two probes thus reported on the different locations in the solvent pools. At low temperature, the gels were shown to prevent ice crystallisation in the solvent pools resulting in the formation of supercooled water. Both probes showed that the water froze at ca. 250 K, thus suggesting that the properties of the supercooled water are uniform across the solvent pools.

EPR spectroscopy and spin-trapping experiments showed that polymer-encapsulated Au nanoparticles promote aldehyde oxidation via a radical pathway by initiating formation of acyl radicals.

We have synthesised a series of amphiphilic spin probes containing two nitroxide-functionalised lipophilic branches connected by a disulfide group. The sensitivity of EPR spectroscopy to the interactions between the radical centres in these bisnitroxides makes it possible to monitor the thiol-disulfide exchange reaction in colloidal assemblies.

A series of gold nanoparticles functionalized with TEMPO-modified disulfide 2 have been prepared and studied by electron paramagnetic resonance (EPR) spectroscopy, UV-vis, transmission electron microscopy (TEM), energy dispersive X-ray analysis (EDX), and thermogravimetric analysis (TGA). In order to increase the packing of spin labels on the particle surface, heat-induced size evolution and ligand exchange reactions were used. The optimized synthesis included a one-pot reaction at room temperature that led to gold nanoparticles with a controlled large size (ca. 7 nm) and high coverage of radicals. These nanoparticles showed a |Δms| = 2 transition at half-field, which gives direct evidence of the presence of a high-spin state and permits an EPR study of the nature of the magnetic coupling between the spins. Low temperature (‘cold’) atmospheric pressure plasmas have gained much attention in recent years due to their biomedical effects achieved through the interactions of plasma induced species with the biological substrate. Monitoring of the radical species in an aqueous biological milieu is usually performed via electron paramagnetic resonance (EPR) spectroscopy using various nitrone spin traps, which form persistent radical adducts with the short-lived radicals. However, the stability of these nitroxide radical adducts in the plasma-specific environment is not well known. In this work, chemical transformations of nitroxide radicals in aqueous solutions using a model nitroxide 4-oxo-TEMPO were studied using EPR and LC-MS. The kinetics of the nitroxide decay when the solution was exposed to plasma were assessed, and the reactive pathways proposed. The use of different scavengers enabled identification of the types of reactive species which cause the decay, indicating the predominant nitroxide group reduction in oxygen-free plasmas. The 2H adduct of the PBN spin trap (PBN-D) was shown to decay similarly to the model molecule 4-oxo-TEMPO. The decay of the spin adducts in plasma-treated solutions must be considered to avoid rendering the spin trapping results unreliable. In particular, the selectivity of the decay indicated the limitations of the PTIO/PTI nitroxide system in the detection of nitric oxide.

Aging of thiolate protected gold nanoparticles (AuNPs) results in reduced reactivity in the disulfide exchange as monitored by electron paramagnetic resonance (EPR) spectroscopy with a bisnitroxide disulfide incoming ligand. Factors determining the reactivity of the aged particles were investigated. The presence of different binding sites on the surface of AuNPs and a surface reorganization process during aging can explain observed reactivity trends.

The factors limiting the relaxivity (r) of MRI contrast agents based on small (∼2.0 nm) gold nanoparticles functionalised with paramagnetic chelates were explored using EPR spectroscopy. The EPR analysis suggested that nanoparticle-attached chelates exhibit relatively high tumbling rates which restrict their relaxivity. Two different strategies were employed in order to test this hypothesis and hence improve the relaxivity of the nanoparticle-based contrast agents. In the first approach, the particle diameter was increased. This resulted in lower surface curvature and hence tighter ligand packing, which in turn led to increased relaxivity. In the second approach, the nanoparticles were overcoated with multilayers of oppositely charged polyelectrolytes. The restricted motion of Gd(3+) chelates coated by 2-4 polymer layers led to increased relaxivity which was dramatically reduced for thicker layers, presumably due to restricted diffusion of water molecules.

During asymmetric cyanohydrin synthesis catalysed by vanadium(V)oxo(salen) complexes, the catalysts are reduced to vanadium(IV)oxo(salen) as determined by EPR spectroscopy; the reducing agent is cyanide which is oxidised to cyanogen via a non-radical mechanism.

We present a molecular probe technique for accessing interfacial surface electrostatics of ligand-protected gold nanoparticles. A series of ligands with variable length of the hydrocarbon bridge between the anchoring sulfur and the reporting pH-sensitive nitroxide is described. The protonation state of this probe is directly observed by EPR spectroscopy. For tiopronin-protected Au nanoparticles, we observed an increase in pKa of up to ca. 1.1 pH units that was affected by the position of the reporter moiety with respect to the monolayer interface.

The lateral mobility of the thiolate ligands on the surface of Au nanoparticles was probed by EPR spectroscopy. This was achieved by using bisnitroxide ligands, which contained a disulfide group (to ensure attachment to the Au surface) and a cleavable ester bridge connecting the two spin-labeled branches of the molecule. Upon adsorption of these ligands on the surface of Au nanoparticles, the two spin-labeled branches were held next to each other by the ester bridge as evidenced by the spin-spin interactions. Cleavage of the bridge removed the link that kept the branches together. CW and pulsed EPR (DEER) experiments showed that the average distance between the adjacent thiolate branches on the Au nanoparticle surface only marginally increased after cleaving the bridge and thermal treatment. This implies that the lateral diffusion of thiolate ligands on the nanoparticle surface is very slow at room temperature and takes hours even at elevated temperatures (90 degrees C). The changes in the distance distribution observed at high temperature are likely due to ligands hopping between the nanoparticles rather than diffusing on the particle surface.

Complexation of beta-cyclodextrin with flexible nitroxide biradicals linked by a polyethylene glycol chain was monitored by EPR spectroscopy. The EPR spectra of the uncomplexed biradicals show an exchange interaction due to the flexibility of the polyethylene glycol chain. Complexation with cyclodextrin leads to the disappearance of the exchange interaction in the EPR spectra. The complexation can be reversed by the addition of competing guests (e.g., adamantane derivatives). At high concentration, the inclusion complexes precipitate, and differential scanning calorimetry (DSC) of the precipitates proved the formation of complexes. Elemental analysis data revealed that the complexes contain several cyclodextrin units per biradical but that the composition was not stoichiometric.

EPR spectra of cyclodextrins labelled with TEMPO derivatives (SL-CDs) are sensitive to complexation with large guest molecules. We used SL-CDs to explore the behaviour of concentrated PEG/PPG solutions. The relationship between rotational correlation times and solvent viscosity showed significant deviations from the Debye–Stokes–Einstein equation, probably due to self-aggregation of alkylene glycols in concentrated solutions. The data fit the fractional Debye–Stokes–Einstein equation well. We have also studied complexation of SL-CDs with adamantane-functionalised DAB dendrimers. The strength of binding increases with dendrimer generation; formation of supramolecular aggregates at high concentrations was observed with the generation 3 dendrimer.

EPR spectroscopy was used to study formation of inclusion complexes of monofunctionalised spin-labelled beta-cyclodextrins; this method is very sensitive to the interactions of cyclodextrins with large guest molecules.

Spin-trapping experiments show the formation of sulfur-centered radicals during a ligand-exchange reaction between Ph3P-protected Au nanoparticles and alkanethiols in air (see picture). Oxidation of the alkanethiols by molecular O2 adsorbed on the nanoparticles is proposed as the key step. The feasibility of such a process is demonstrated by the Au-nanoparticle-catalyzed oxidation of BH4- and tBuOOH with air.

Using EPR spectroscopy and spin-labeled ligands, we have studied product distribution in the ligand exchange reactions of Au nanoparticles. When the incoming ligand was used in excess, the exchanged nanoparticles had uniform composition of the organic shell. Preparative GPC was employed to fractionate these materials according to the diameter of the Au core, as confirmed by TEM, UV, and EPR measurements. When the exchange reaction was carried out with less than a stoichiometric amount of the incoming ligand, nanoparticles with polydisperse composition of the organic shell were formed. This was established by the EPR analysis of the exchange reaction mixtures following their GPC fractionation. The GPC separation of these mixtures is controlled by both Au core size and shell composition.

A series of Au nanoparticles functionalised with nitroxide spin labels has been prepared and studied by EPR spectroscopy. Samples with low coverage of the spin label were used to investigate the dynamics of the surface-attached labels at different distances from the Au surface. The rotational correlation times of spin labels vary from 10(-10) s to more than 3 x 10(-9) s, depending on the chain length of the label and the surrounding ligand. The samples with higher coverage of the spin label show an increasing contribution of the exchange interaction between nitroxides adsorbed in a close proximity to each other on the same nanoparticle. Quantitative analysis of the EPR spectra of these samples suggests the presence of non-equivalent binding sites on the surface of Au nanoparticles. Additionally, EPR signals of isolated radical pairs were observed at intermediate coverage.

An EPR study of a place-exchange reaction of a diradical disulfide with butanethiol-protected Au nanoparticles showed that the two branches of the disulfide molecule do not adsorb adjacent to each other on the Au surface. The most likely mechanism includes adsorption of only one branch of the disulfide molecule in the exchange process. The exchange reaction was found to be zeroth-order with respect to the diradical, indicative of a dissociative "SN1"-type mechanism.

Formation of organogels from covalently linked polyethylene glycols and cyclodextrins and their encapsulation/release properties were monitored with EPR spectroscopy using spin labels and spin probes; changes in molecular tumbling rates provided information on spin probe/label location inside the gel network, and molecular properties of the gel.

The formation of radicals in the gas phase oxidation of cyclohexane over metal oxides was studied using spin trapping and EPR spectroscopy; the results confirmed that during the catalytic initiation, the oxygen responsible for the radicals formation originates from the lattice of the metal oxide.

Convolution-based fitting of EPR spectra makes it possible to simultaneously determine concentrations of TEMPO and oxygen in TEMPO-inhibited polymerisations and autoxidations; this method is useful for understanding the chemistry of inhibitor mixtures.
