Background Neuropathic pain in neuroimmunological disorders refers to pain caused by a lesion or disease of the somatosensory system such as multiple sclerosis (MS) and neuromyelitis optica spectrum disorder (NMOSD). MS and NMOSD are autoimmune disorders of the central nervous system, and ≥ 50% of patients with these disorders experience chronic neuropathic pain. The currently available medications for the management of neuropathic pain have limited effectiveness in patients with MS and NMOSD, and there is an unmet medical need to identify novel therapies for the management of chronic neuropathic pain in these patients. In this review article, we summarize the role of interleukin-6 (IL-6) in the pathogenesis of MS and NMOSD and the ameliorative effects of anti–IL-6 therapies in mouse models of experimental autoimmune encephalomyelitis (EAE). Main body Intraperitoneal injection of MR16-1, an anti–IL-6 receptor (IL-6R) antibody, reduced mechanical allodynia and spontaneous pain in EAE mice, which was attributed to a reduction in microglial activation and inhibition of the descending pain inhibitory system. The effect of anti–IL-6 therapies in ameliorating neuropathic pain in the clinical setting is controversial; a reduction in pain intensity has been reported with an anti–IL-6 antibody in four studies, namely a case report, a pilot study, a retrospective observational study, and a case series. Pain intensity was evaluated using a numerical rating scale (NRS), with a lower score indicating lesser pain. A reduction in the NRS score was reported in all four studies. However, in two randomized controlled trials of another anti–IL-6R antibody, the change in the visual analog scale pain score was not statistically significantly different when compared with placebo. This was attributed to the low mean pain score at baseline in both the trials and the concomitant use of medications for pain in one of the trials, which may have masked the effects of the anti–IL-6R antibody on neuropathic pain. Conclusion Thus, anti–IL-6 therapies might have a potential to reduce neuropathic pain, but further investigations are warranted to clarify the effect of inhibition of IL-6 signaling on neuropathic pain associated with MS and NMOSD.

Background Using a controlled trial, this study aimed to evaluate the effectiveness of adhesive strapping to improve the natural healing rate of umbilical hernia. Methods This prospective, observational study included 128 patients from Kumamoto, Japan (97 in the adhesive strapping group, and 31 in observation group), from 2012–2015. The duration from first hospital visit to the hernia orifice closure was compared between the two groups. Result Kaplan‐Meier curves showed that the probability of umbilical hernia in the adhesive strapping group was lower until approximately 200 days, but it was not statistically significant in the log rank test. According to multivariate Cox proportional hazard models, the hazard risk of umbilical hernia in the adhesive strapping group was significantly higher within 0–60 days after adjusting for confounding factors such as hernial cavity and hernia orifice area (P < 0.0001). Conclusion Adhesive strapping of umbilical hernia was significantly associated with earlier closure of the hernia orifice from baseline until at least 60 days.

At the Institute of Laser Engineering, Osaka, the first steps are taken towards the development of a table-top laser-driven neutron source, capable of producing a wide range of energies with high brightness and temporal resolution. By employing a pure hydrogen moderator, maintained at cryogenic temperature, a cold neutron ($$\le 25\hbox { meV}$$ ≤ 25 meV ) flux of $$\sim 2\times 10^3\hbox { n/cm}^2$$ ∼ 2 × 10 3 n/cm 2 /pulse was measured at the proximity of the moderator exit surface. The beam duration of hundreds of ns to tens of $$\upmu \hbox {s}$$ μ s is evaluated for neutron energies ranging from 100s keV down to meV via Monte-Carlo techniques. Presently, with the upcoming J-EPoCH high repetition rate laser at Osaka University, a cold neutron flux in orders of $$\sim 1\times 10^{9}\hbox { n/cm}^2/\hbox {s}$$ ∼ 1 × 10 9 n/cm 2 / s is expected to be delivered at the moderator in a compact beamline.

We demonstrate that $(n,2n)$ reactions are induced by a high-flux pulse of fast neutrons ($\ensuremath{\sim}{10}^{10}$ neutrons in $\ensuremath{\sim}1\phantom{\rule{0.16em}{0ex}}\mathrm{ns}$) provided from a laser-driven neutron source (LDNS). The maximum energy of the broadband neutrons reaches a few tens MeV. Several kinds of metal targets are exposed to the fast neutrons. As a result, unstable isotopes, such as $^{54}\mathrm{Mn}, ^{58}\mathrm{Co}, ^{175}\mathrm{Hf}$, and $^{196}\mathrm{Au}$ are produced by $(n,2n)$ reactions and $^{180}\mathrm{Hf}^{m}, ^{181}\mathrm{Hf}, ^{56}\mathrm{Mn}, ^{198}\mathrm{Au}$, and $^{60}\mathrm{Co}$ are produced by $(n,\ensuremath{\gamma})$ reactions. We evaluate the neutron fluence and energy spectrum using the activation method in conjunction with a time-of-flight measurement. The neutron fluence is determined to be $(4.3\ifmmode\pm\else\textpm\fi{}0.5)\ifmmode\times\else\texttimes\fi{}{10}^{8}\phantom{\rule{0.16em}{0ex}}\mathrm{neutrons}/\mathrm{c}{\mathrm{m}}^{2}$ in the energy range from approximately 8 to 20 MeV at 8-mm downstream of the neutron source. The present scheme provides a method to evaluate high-density neutrons seen in stellar environments, which are expected to be generated from future LDNSs.

A dosimetric calibration of three types of radiochromic films (GafChromicTM HD-V2, MD-V3, and EBT3) was carried out for absorbed doses (D) ranging up to 100 kGy using a 130 TBq Co60 γ-ray source. The optical densities (ODs) of the irradiated films were acquired with the transmission-mode flatbed film scanner EPSON GT-X980. The calibration data were cross-checked using the 20-MeV proton beam from the azimuthally varying field cyclotron at the Research Center for Nuclear Physics in Osaka University. These experimental results not only present the measurable dose ranges of the films depending on the readout wavelength, but also show consistency with our hypothesis that the OD response curve [log(OD)–log(D) curve] is determined by the volumetric average of the absorption dose and does not strongly depend on the type of radiation for the excitation.

CHA zeolite (chabazite) crystals and membranes were prepared from organic structural directing agents-free (OSDA-free) gels. Chabazite membranes were grown on the tubular alumina supports. The effect of cesium and fluoride salts on the crystallization of chabazite crystals and membranes were investigated. Highly crystalline chabazite and uniform chabazite membranes were only obtained from the gel containing the cesium and fluoride salts. Single CO2, N2 and CH4 permeances and mixed gas separations in CO2/N2 and CO2/CH4 binary mixtures through chabazite membranes were measured. Temperature and pressure affected the permeances and separation selectivities in two mixtures. The best membrane showed CO2/N2 and CO2/CH4 separation selectivities as high as 16 and 46 together with CO2 permeances of ~ 1.0 × 10-7 mol/(m2 s Pa) at a high temperature of 473 K in equimolar dry CO2/N2 and CO2/CH4 binary mixtures, respectively. The CO2/N2 and CO2/CH4 selectivities lost only 11% and 15%, and CO2 permeances lost only 18% and 20% in equimolar wet CO2/N2 and CO2/CH4 mixtures (8% in mole of water vapor) at 473 K compared to these values in dry mixtures, respectively. The OSDA-free chabazite membranes were hydrothermally stable in the presence of vapor water for 4 d at 378 K.

Resorcinol/formaldehyde (RF) foam resin is an attractive material as a low-density target in high-power laser–plasma experiments because of its fine network structure, transparency in the visible region, and low-Z element (hydrogen, carbon, and oxygen) composition. In this study, we developed disk-shaped RF foam and deuterated RF foam targets with 40–200 μm thickness and approximately 100 mg/cm3 density having a network structure from 100 nm to a few micrometers cell size. By deuteration, the polymerization rate was drastically slowed down owing to kinetic isotope effects. These targets were used in high-power laser experiments where a megaelectronvolt proton beam was successfully generated.

Baculovirus-infected silkworms are promising bioreactors for producing recombinant glycoproteins, including antibodies. Previously, we developed a method for isotope labeling of glycoproteins for nuclear magnetic resonance (NMR) studies using silkworm larvae reared on an artificial diet containing 15N-labeled yeast crude protein extract. Here, we further develop this method by introducing a technique for the expression of isotope-labeled glycoproteins by silkworm pupae, which has several potential advantages relative to larvae-based techniques in terms of production yield, ease of handling, and storage. Here, we fed fifth instar larvae an artificial diet with an optimized composition containing [methyl-13C]methionine, leading to pupation. Nine-day-old pupae were then injected with recombinant Bombyx mori nucleopolyhedrovirus (BmNPV) bacmid for expression of recombinant human immunoglobulin G (IgG). From the whole-body homogenates of pupae, 0.35 mg/pupa of IgG was harvested, which is a yield that is five times higher than can be obtained from larvae. Recombinant IgG, thus prepared, exhibited mainly three kinds of pauci-mannose-type oligosaccharides and had a 13C-enrichment ratio of approximately 80%. This enabled selective observation of NMR signals originating from the methionyl methyl group of IgG, confirming its conformational integrity. These data demonstrate the utility of silkworm pupae as factories for producing recombinant glycoproteins with amino-acid-selective isotope labeling.

Fast isochoric heating of a pre-compressed plasma core with a high-intensity short-pulse laser is an attractive and alternative approach to create ultra-high-energy-density states like those found in inertial confinement fusion (ICF) ignition sparks. Laser-produced relativistic electron beam (REB) deposits a part of kinetic energy in the core, and then the heated region becomes the hot spark to trigger the ignition. However, due to the inherent large angular spread of the produced REB, only a small portion of the REB collides with the core. Here, we demonstrate a factor-of-two enhancement of laser-to-core energy coupling with the magnetized fast isochoric heating. The method employs a magnetic field of hundreds of Tesla that is applied to the transport region from the REB generation zone to the core which results in guiding the REB along the magnetic field lines to the core. This scheme may provide more efficient energy coupling compared to the conventional ICF scheme., It is desirable to deposit more energy in the dense plasma core to trigger the fusion ignition. Here the authors demonstrate enhanced energy coupling from laser to plasma core by using solid targets and guiding the transport of relativistic electron beam with external magnetic field.

We present the results of theoretical studies of formation and evolution of the current sheet in a colliosionless plasma during magnetic reconnection in relativistic limit. Relativistic magnetic reconnection is driven by parallel laser pulses interacting with underdense plasma target. Annihilation of laser created magnetic field of opposite polarity generates strong non-stationary electric field formed in between the region with opposite polarity magnetic field accelerating charged particles within the current sheet. This laser-plasma target configuration is discussed in regard with the laboratory modeling of charged particle acceleration and gamma flash generation in astrophysics. We present the results of 3-dimensional kinetic simulations and theoretical studies on the formation and evolution of the current sheet in a collisionless plasma during magnetic field annihilation in the ultra-relativistic limit. Annihilation of oppositively directed magnetic fields driven by two laser pulses interacting with underdense plasma target is accompanied by an electromagnetic burst generation. The induced strong non-stationary longitudinal electric field accelerates charged particles within the current sheet. Properties of the laser-plasma target configuration are discussed in the context of the laboratory modeling for charged particle acceleration and gamma flash generation in astrophysics.

OBJECTIVE:Transcranial magnetic stimulation (TMS) produces characteristic deflections in the EEG signal named TMS-evoked EEG potentials (TEPs), which can be used to assess drug effects on cortical excitability. TMS can also be used to determine the resting motor threshold (RMT) for eliciting a minimal muscle response, as a biomarker of corticospinal excitability. XEN1101 is a novel potassium channel opener undergoing clinical development for treatment of epilepsy. We used TEPs and RMT to measure the effects of XEN1101 in the human brain, to provide evidence that XEN1101 alters cortical excitability at doses that might be used in future clinical trials.METHODS:TMS measurements were incorporated in this Phase I clinical trial to evaluate the extent to which XEN1101 modulates TMS parameters of cortical and corticospinal excitability. TEPs and RMT were collected before and at 2-, 4-, and 6-hours post drug intake in a double-blind, placebo-controlled, randomized, two-period crossover study of 20 healthy male volunteers.RESULTS:Consistent with previous TMS investigations of antiepileptic drugs (AEDs) targeting ion channels, the amplitude of TEPs occurring at early (15-55 msec after TMS) and at late (150-250 msec after TMS) latencies were significantly suppressed from baseline by 20 mg of XEN1101. Furthermore, the RMT showed a significant time-dependent increase that correlated with the XEN1101 plasma concentration.INTERPRETATION:Changes from baseline in TMS measures provided evidence that 20 mg of XEN1101 suppressed cortical and corticospinal excitability, consistent with the effects of other AEDs. These results support the implementation of TMS as a tool to inform early-stage clinical trials.

When an outer panel of a B-pillar is manufactured with the hot stamping process, reinforcements are spot welded on its inner side. Before reinforcements are added, the microstructure of the outer panel is martensite. However, reheating during spot welding changes the martensite to ferrite, which has a lower hardness in the heat-affected zone than in other areas. If spot welding is conducted before hot stamping for making a spot welded tailored blank, the microstructure in the spot welded tailored blank after hot stamping is martensite. This sequence of processes avoids hardness reduction due to spot welding. In this study, the hardness and microstructure around spot welded parts of the tailored blank were investigated. The results clearly showed that areas close to the spot welded parts are severely stretched during hot stamping. In addition, stretching suppresses the martensitic phase transformation and reduces the hardness. To characterize this phenomenon, a simulation was conducted that considered the effects of pre-strain on the phase transformation. A continuous cooling transformation (CCT) diagram and a deformation continuous cooling transformation (DCCT) diagram were made in order to quantify the effect of the cooling rate and pre-strain on the phase transformation and hardness. The hardness was then calculated using the experimentally measured CCT and DCCT diagrams and the finite element analysis results. The calculated hardness was compared with the experimental hardness. Good agreement was found between the calculated and experimental results.

BackgroundTriage has an important role in providing suitable care to the largest number of casualties in a disaster setting, but there are no secondary triage methods suitable for children. This study developed a new secondary triage method named the Pediatric Physiological and Anatomical Triage Score (PPATS) and compared its accuracy with current triage methods.MethodsA retrospective chart review of pediatric patients under 16 years old transferred to an emergency center from 2014 to 2016 was performed. The PPATS categorized the patients, defined the intensive care unit (ICU)-indicated patients if the category was highest, and compared the accuracy of prediction of ICU-indicated patients among PPATS, Physiological and Anatomical Triage (PAT), and Triage Revised Trauma Score (TRTS).ResultsAmong 137 patients, 24 (17.5%) were admitted to ICU. The median PPATS score of these patients was significantly higher than that of patients not admitted to ICU (11 [IQR: 9-13] versus three [IQR: 2-4]; P2=0.139; P2=0.320; P2=0.362; P2=0.308; PConclusionsThe accuracy of PPATS was superior to other methods for secondary triage of children.ToidaC, MugurumaT, AbeT, ShinoharaM, GakumazawaM, YogoN, ShirasawaA, MorimuraN. Neutron beams have been providing indispensable tools for a wide range of fields in modern science and engineering. Recently, a new type of pulsed neutron source has been developed, known as laser-driven neutron source (LDNS). The LDNSs utilize laser-accelerated ions, including protons and deuterons as a primary beam and generate neutrons from a secondary target (lithium, beryllium, etc) via a nuclear reaction. Applying an additional moderator part, LDNSs can provide a broad energy range of neutrons (meV ∼ MeV). This paper aims to introduce the current status of LDNS and the results of application-oriented experiments implemented at the Institute of Laser Engineering (ILE) of Japan.

Thermal neutron (∼25 meV) beam is a powerful tool for investigating the structure and properties of materials used in science and technology. A laser-driven neutron source generating 1010 neutrons within 1 ns duration is utilized to a single shot radiography with a dual beam of thermal neutrons and X-rays. As a proof of principle, we show the non-destructive inspection of hazardous substances (Cadmium) contained in a typical battery, when the cadmium anode thickness is evaluated from the transmittance of thermal neutrons. The fact that the neutron inspection above is performed with a single laser shot, i.e. with a single bunch of neutrons leads to a safer neutron source that is optically controlled on/off, and provides a novel tool for science and engineering.

Tetraethylenepentamine (TEPA) components are separated by fractional distillation, and the structure of each fraction is established using spectroscopic methods. Then, solid sorbents incorporating these components are prepared, and the capacities and rates of CO2 absorption of the sorbents are evaluated to elucidate how the molecular structure affects the absorption performance.

To understand a biological system, it is important to observe structures of biomolecules in the solution where the system is functionalized. Small-Angle X-ray Scattering coupled with Size Exclusion Chromatography (SEC-SAXS) is one of techniques to selectively observe the target molecules in the multi-component system. However, this technique is believed to be available only with a synchrotron-based SAXS instrument due to requirement of high beam intensity and, therefore, the limitation of the beam time was obstacle to satisfy demands from many bio-researchers. We newly developed Laboratory-based Size exclusion chromatography SAXS System (La-SSS) by utilizing a latest laboratory-based SAXS instrument and finely optimization of the balance between flow rate, cell volume, irradiation time and so on. La-SSS succeeded not only decoupling of target protein(s) from non-specific aggregates but also measurement of each concerned component in a multi-component system. In addition, an option: “stopping mode”, which is designed for improving statistics of SAXS profile, realized a high S/N data acquisition for the most interesting protein in a multi-component system. Furthermore, by utilizing a column having small bed volume, the small-scale SEC-SAXS study makes available. Through optimization of instrumental parameters and environments, La-SSS is highly applicable for experimental requirements from various biological samples. It is strongly expected that a La-SSS concept must be a normal option for laboratory-based SAXS in the near future.

Among various CO2-capture technologies, membrane separation is considered as one of the promising solutions, because of its energy efficiency and operation simplicity. Many research and development are conducted for the (1) CO2/N2 (post-combustion: CO2 separation from flue gas), (2) CO2/CH4 (CO2 separation from natural gas), and (3) CO2/H2 (pre-combustion: CO2 separation from integrated gasification combined cycle (IGCC) processes). In this chapter, research and development of various types of membranes (polymeric membranes, inorganic membranes, ionic liquid membranes, and facilitated transport membranes) for these applications are briefly reviewed. In the latter part of the chapter, the development of “molecular gate” membrane modules by the authors for the pre-combustion to separate CO2 from H2 at the integrated coal gasification combined cycle (IGCC) power plants are reviewed.

Fast isochoric laser heating is a scheme to heat a matter with relativistic-intensity ($>$ 10$^{18}$ W/cm$^2$) laser pulse or X-ray free electron laser pulse. The fast isochoric laser heating has been studied for creating efficiently ultra-high-energy-density (UHED) state. We demonstrate an fast isochoric heating of an imploded dense plasma using a multi-picosecond kJ-class petawatt laser with an assistance of externally applied kilo-tesla magnetic fields for guiding fast electrons to the dense plasma.The UHED state with 2.2 Peta-Pascal is achieved experimentally with 4.6 kJ of total laser energy that is one order of magnitude lower than the energy used in the conventional implosion scheme. A two-dimensional particle-in-cell simulation reveals that diffusive heating from a laser-plasma interaction zone to the dense plasma plays an essential role to the efficient creation of the UHED state., 8 pages, 4 figures, 1 table

Ceramic powders are synthesized using chemical solution methods, such as organic acid salt, precipitant generation, alkaline hydrolysis, and alkoxide hydrolysis. The powder synthesis by precipitation and hydrolysis and the synthesis of monodispersed particles are described. The formation of complex alkoxide of lithium niobate (LN) precursor is confirmed by 93Nb nuclear magnetic resonance spectroscopy. Stoichiometric LN films are synthesized on various substrates via chemical solution route. The synthesis of the designed precursor in solution is a key for the low-temperature crystallization of high-quality LN films with preferred orientation. Several functional ceramic thin films of multicomponent oxides are also synthesized using chemical solution process.

A large-aperture high-sensitivity image intensifier panel that consists of an avalanche photodiode array and a light-emitting diode array is presented. The device has 40% quantum efficiency, over 104 optical gain, and 80-ns time resolution. The aperture size of the device is 20 cm, and with the current manufacturing process, it can be scaled to arbitrarily larger sizes. The device can intensify the light from a single particle scintillation emission to an eye-visible bright flash. The image resolution of the device is currently limited by the size of the avalanche photodiode that is 2 mm, although it can be scaled to smaller sizes in the near future. The image intensifier is operated at a small voltage, typically +57 V. The device can be applied to various applications, such as scintillation imaging, night vision cameras, and an image converter from non-visible light (such as infrared or ultraviolet) to visible light.

Study of warm dense matter remains a very important task for understanding of many unique phenomena observing as in astrophysical research as in inertial fusion and fast ignition. In this work, we studied the parameters of plasma created by 1.7 ps laser pulses of relativistic intensity of 7 × 1018W/cm2in a specially designed Al–Cu wire-shape target, in comparison with a flat Cu and Al foil targets. We observed the strong emission of neutral or virtually neutral CuKαline from both Cu foil and Cu wire part of targets, which indicates the creation of a dense state exposed to the intense flow of hot electrons. Parameters of the plasma were evaluated by comparison of experimental spectra with the results of modeling by collisional-radiative kinetic code PrismSpec under the plasma zone approach. The using of Al foil in front of Cu wire part of target allowed avoiding the direct heating of Cu-wire and acquiring spectra of Cu K-shell emission evidently belonging to emission of warm dense matter (WDM) state. The upper estimate for the electron temperature in WDM region was found to be below 80 eV.

In recent years, the deterioration of infrastructure facilities such as bridges has become a problem. Precautionary measures such as visual inspection and repair by humans are in place as countermeasures for aging; however, there are issues with cost and safety in such inspections. If inspection by robots becomes possible, both these aspects will be improved, which will significantly contribute to the maintenance of infrastructure facilities. In this paper, we propose a complex image processing technique to specify the location of feature points as coordinates through smartphone cameras to obtain the location information of feature points needed for positioning BIREM-IV-P developed to support bridge inspection. The corners located in the bridge inspection environment are used as feature points, and the corners are specified using Harris corner detection, which is a conventional corner detection method, to obtain the position of the feature points. In addition, to compensate for the shortcomings of Harris corner detection, a line segment in the image is detected using the Hough transform, and the intersection points of the line segments are recognized as corners. By combining the results of the two detection methods in this manner, the target feature points can be accurately specified. Then, the position of the feature points of the specified image coordinate system can be changed to the world coordinate system. As a result, it was possible to detect the location of the target feature points in a three-dimensional coordinate system.

Laser-driven neutrons have been expected as an optimum source for neutron radiography applications. Since the neutron fluence is lower than that of accelerator-based neutron source a new device for amplifying scintillation light has been required. The avalanche image intensifier panel discussed here has a large aperture and fast response which has been developed specifically for laser-driven neutron radiography applications. The gain uniformity for pulsed light sources was improved by optimizing electrical circuit. The performances of the improved image intensifier panel were experimentally evaluated. With the improved circuit, the gain non-uniformity was σ = 5 % and impulse response was 50 ns in full width half maximum (FWHM), and dynamic range was 100 which is invariable for a light pulse duration from 15 ns to 500 ns. Laser-driven neutron radiography has been successfully demonstrated in spite of the low neutron flux.

In order to enhance the conversion efficiency of ultra-intense laser energy into plasma leading to ion acceleration, we have performed an experiment with over-picosecond laser pulses having a relativistic intensity interacting with foam targets. We adjust the prepulse of the laser to control the plasma density near the critical density. The combination of hydrodynamic and particle-in-cell simulations suggest that the region of high energy density is induced in the near-critical plasma, leading to enhance the proton energy up to 23 MeV with the laser intensity of 6 × 1018 W/cm2.

In a previous study, we developed a novel in situ analysis and observation system that allows for simultaneous synchrotron X-ray diffraction (XRD) and optical observations of a frictional surface. This in situ system was used to investigate the scuffing phenomena of SUJ2 bearing steel (AISI 52100); characteristic surface deteriorations occurred during the scuffing process, including plastic flow, heat-spot formation, austenite transformation, and a decrease in the width of the XRD peaks (indicating a decrease of dislocations and strain). These surface deteriorations are not observed during normal wear, hence it is possible that they cause catastrophic wear during the scuffing of steel. In this study, to elucidate the scuffing mechanism of steel, we focused on the following two points: (1) whether the above surface deteriorations are unique to SUJ2 steel or whether they occur in general steels as well, and (2) the extent to which these surface deteriorations contribute to the wear amount. To achieve these objectives, we performed scuffing tests on four types of steel using the previously developed in situ system. In particular, we focused on the first stage of the scuffing process. The present test results suggest that these surface deteriorations also occur in general steels, and that plastic flow and heat-spot formation, which originate from the same phenomenon, are the dominant contributors to the wear amount during the scuffing of steel. Furthermore, the wear amount per unit plastic flow appears to be independent of steel composition.

Increasing the laser energy absorption into energetic particle beams represents a longstanding quest in intense laser-plasma physics. During the interaction with matter, part of the laser energy is converted into relativistic electron beams, which are the origin of secondary sources of energetic ions, γ-rays and neutrons. Here we experimentally demonstrate that using multiple coherent laser beamlets spatially and temporally overlapped, thus producing an interference pattern in the laser focus, significantly improves the laser energy conversion efficiency into hot electrons, compared to one beam with the same energy and nominal intensity as the four beamlets combined. Two-dimensional particle-in-cell simulations support the experimental results, suggesting that beamlet interference pattern induces a periodical shaping of the critical density, ultimately playing a key-role in enhancing the laser-to-electron energy conversion efficiency. This method is rather insensitive to laser pulse contrast and duration, making this approach robust and suitable to many existing facilities., Enhanced coupling of laser energy to the target particles is a fundamental issue in laser-plasma interactions. Here the authors demonstrate increased photon absorption leading into higher laser to electron and proton energy transfer through the interference of multiple coherent beamlets.

BACKGROUND: Staphylococcus aureus bacteraemia is a common cause of severe community-acquired and hospital-acquired infection worldwide. We tested the hypothesis that adjunctive rifampicin would reduce bacteriologically confirmed treatment failure or disease recurrence, or death, by enhancing early S aureus killing, sterilising infected foci and blood faster, and reducing risks of dissemination and metastatic infection.METHODS: In this multicentre, randomised, double-blind, placebo-controlled trial, adults (≥18 years) with S aureus bacteraemia who had received ≤96 h of active antibiotic therapy were recruited from 29 UK hospitals. Patients were randomly assigned (1:1) via a computer-generated sequential randomisation list to receive 2 weeks of adjunctive rifampicin (600 mg or 900 mg per day according to weight, oral or intravenous) versus identical placebo, together with standard antibiotic therapy. Randomisation was stratified by centre. Patients, investigators, and those caring for the patients were masked to group allocation. The primary outcome was time to bacteriologically confirmed treatment failure or disease recurrence, or death (all-cause), from randomisation to 12 weeks, adjudicated by an independent review committee masked to the treatment. Analysis was intention to treat. This trial was registered, number ISRCTN37666216, and is closed to new participants.FINDINGS: Between Dec 10, 2012, and Oct 25, 2016, 758 eligible participants were randomly assigned: 370 to rifampicin and 388 to placebo. 485 (64%) participants had community-acquired S aureus infections, and 132 (17%) had nosocomial S aureus infections. 47 (6%) had meticillin-resistant infections. 301 (40%) participants had an initial deep infection focus. Standard antibiotics were given for 29 (IQR 18-45) days; 619 (82%) participants received flucloxacillin. By week 12, 62 (17%) of participants who received rifampicin versus 71 (18%) who received placebo experienced treatment failure or disease recurrence, or died (absolute risk difference -1·4%, 95% CI -7·0 to 4·3; hazard ratio 0·96, 0·68-1·35, p=0·81). From randomisation to 12 weeks, no evidence of differences in serious (p=0·17) or grade 3-4 (p=0·36) adverse events were observed; however, 63 (17%) participants in the rifampicin group versus 39 (10%) in the placebo group had antibiotic or trial drug-modifying adverse events (p=0·004), and 24 (6%) versus six (2%) had drug interactions (p=0·0005).INTERPRETATION: Adjunctive rifampicin provided no overall benefit over standard antibiotic therapy in adults with S aureus bacteraemia.FUNDING: UK National Institute for Health Research Health Technology Assessment.

A relativistic laser pulse, confined in a cylindrical-like target, under specific conditions may perform multiple scattering along the internal target surface. This results in the confinement of the laser light, leading to a very efficient interaction. The demonstrated propagation of the laser pulse along the curved surface is just yet another example of the “whispering gallery” effect, although nonideal due to laser–plasma coupling. In the relativistic domain its important feature is a gradual intensity decrease, leading to changes in the interaction conditions. The process may pronounce itself in plenty of physical phenomena, including very efficient electron acceleration and generation of relativistic magnetized plasma structures.

Glycoproteins are characterized by the heterogeneous and dynamic nature of their glycan moieties, which hamper crystallographic analysis. NMR spectroscopy provides potential advantages in dealing with such complicated systems, given that the target molecules can be isotopically labeled. Methods of metabolic isotope labeling in recombinant glycoproteins have been developed recently using a variety of eukaryotic production vehicles, including mammalian, yeast, insect, and plant cells, each of which has a distinct N-glycan diversification pathway. Yeast genetic engineering has enabled the overexpression of homogeneous high-mannose-type oligosaccharides with 13C labeling for NMR characterization of their conformational dynamics. The utility of stable isotope-assisted NMR spectroscopy has also been demonstrated using the Fc fragment of immunoglobulin G (IgG) as a model glycoprotein, providing useful information regarding intramolecular carbohydrate–protein interactions. Transverse relaxation optimization of intact IgG with a molecular mass of 150 kDa has been achieved by tailored deuteration of selected amino acid residues using a mammalian expression system. This offers a useful probe for the characterization of molecular interaction networks in multimolecular crowded systems typified by serum. Perspectives regarding the development of techniques for tailoring glycoform designs and isotope labeling of recombinant glycoproteins are also discussed.

Lead-free ferroelectric (Bi0.5Na0.5)TiO3 (BNT) thin films were prepared on Pt/TiOx/SiO2/Si substrates via chemical solution deposition. Mn doping of these BNT films was very effective for improving the ferroelectric properties, and the doped Mn ion affected the leakage current properties in both low- and high-applied-field region. Specifically, the leakage current density was effectively reduced by doping with an appropriate amount of Mn. For example, a 1 mol% Mn-doped BNT thin film exhibited a well-shaped ferroelectric polarization–electric field hysteresis loop with a remanent polarization of 22 μC/cm2 and a coercive field of 165 kV/cm at room temperature. In addition, electron spin resonance and X-ray photoelectron spectroscopic analyses revealed that the Mn ions are doped into the Ti sites of BNT as Mn2+ or Mn3+.

Using one of the world most powerful laser facility, we demonstrate for the first time that high-contrast multi-picosecond pulses are advantageous for proton acceleration. By extending the pulse duration from 1.5 to 6 ps with fixed laser intensity of 1018 W cm−2, the maximum proton energy is improved more than twice (from 13 to 33 MeV). At the same time, laser-energy conversion efficiency into the MeV protons is enhanced with an order of magnitude, achieving 5% for protons above 6 MeV with the 6 ps pulse duration. The proton energies observed are discussed using a plasma expansion model newly developed that takes the electron temperature evolution beyond the ponderomotive energy in the over picoseconds interaction into account. The present results are quite encouraging for realizing ion-driven fast ignition and novel ion beamlines.

The identification of the key nanostructural features in supramolecules and the understanding of the relationship between structure and function are important factors in the development of novel molecular-based materials. The ionic conduction path is one of the most attractive key structures because of its ability to generate solid-state ionic diffusive properties for application as a solid electrolyte. We herein report the generation of ionic conduction paths using crystallization of supramolecules with a channel structure. We succeeded in precise structural control of the conduction paths by varying the component diamines or lithium salts. We also show the structure–conductivity relationships of the obtained compounds based on a single crystal X-ray diffraction study and electrochemical measurements.

The measurements of the three-dimensional (3D) dose distribution of proton beams in water are critical for proton therapy for quality assessment (QA). Although ionization chambers are commonly used for this purpose, such measurements take a long time to calculate precise 3D dose distribution. To solve this problem, we measured 3D dose distributions using a glass plate. We placed a 1-mm thick float glass plate on the upper inside of a black box with a water phantom set above the float glass plate outside the black box and irradiated the proton beam to the water phantom from the upper side. The attenuated proton beam by water in the phantom was detected by the float glass plate and a scintillation image was formed in the plate. The image was reflected by a surface mirror set below the float glass plate and detected by a cooled charge-coupled device (CCD) camera from the side. We changed the water depths in the phantom and measured the scintillation images at each depth. Then we calculated the 3D scintillation images from the measured images by stacking them in the depth direction. Measurements were made for 71.2- and 100-MeV proton pencil beams and a spread-out Bragg peak (SOBP) using the imaging system. From the images, we successfully formed 3D scintillation images without quenching. The depth profiles measured from the scintillation images showed almost identical distribution with those measured by the ionization chamber within a difference less than 5%. The lateral profiles were also almost identical within width differences less than 2 mm. We conclude that our proposed method is promising for the 3D dose distribution measurements of proton beams.

We developed a small field-of-view (FOV) high energy gamma camera and measured the gamma photons from an Ir-192 source for imaging of the source. The developed gamma camera consists of a 10 mm thick GAGG pixel scintillator block optically coupled to a position sensitive photomultiplier (PSPMT). It was encased in a 2 cm thick tungsten shield and a 1 mm diameter pinhole collimator was mounted in its head. The gamma camera was positioned 1 m from the Ir-192 source delivered from the brachytherapy system and imaged the Ir-192 source while the source changed dwell positions in the FOV of the gamma camera. We could image the source with 1 s interval and observed the source changed its position in the images. The spatial resolution of the gamma camera system at 1 m from the Ir-192 source was 28 mm FWHM and position accuracy was 5 mm. With the 10 cm thick water phantom positioned between the source and gamma camera, the counts were decreased to half, but the Ir-192 source images could still be observed clearly. We conclude that the developed gamma camera system is promising for the imaging of the Ir-192 source during high-dose-rate brachytherapy pending future improvements in spatial resolution.

In selecting drug target candidates for pharmaceutical research, the linkage to disease and the tractability of the target are two important factors that can ultimately determine the drug efficacy. Several existing resources can provide gene-disease associations, but determining whether such a list of genes are attractive drug targets often requires further information gathering and analysis. In addition, few resources provide the information required to evaluate the tractability of a target. To address these issues, we have updated TargetMine, a data warehouse for assisting target prioritization, by integrating new data sources for gene-disease associations and enhancing functionalities for target assessment. As a data mining platform that integrates a variety of data sources, including protein structures and chemical compounds, TargetMine now offers a powerful and flexible interface for constructing queries to check genetic evidence, tractability and other relevant features for the candidate genes. We demonstrate these features by using several specific examples.

Carbon dioxide (CO2) capture and storage (CCS) is a promising countermeasure against global warming, but installing CCS into a power supply system causes a significant decrease in power output. Much research has already focused on the issue of how to facilitate implementation of CCS technology. This paper reviews recent studies on the efficiency penalty of coal-fired power plants with CCS. Efficiency penalty, which represents a net decrease in the power efficiency caused by the CO2 capture and compression process, can be estimated using process simulation that considers factors such as the power generation steam cycle, coal type, and CO2 capture and compression process. According to previous research, the efficiency penalty for current applications was about 10%. The ratio of efficiency penalty caused by CO2 capture to the total efficiency penalty was about two thirds. It appears that while the types of power plant and coal had little influence on efficiency penalty, the CO2 capture technology was critically important. By reducing the regeneration energy of the CO2 scrubbing solvent by 1 GJ/t-CO2, an approximate 2% efficiency improvement can be expected.

A design of multichannel gated photomultiplier tube (PMT) is presented for the 960-channel neutron time-of-flight detector at the Institute of Laser Engineering of Osaka University. This is important for the fusion science and the nuclear photonics where intense hard X-rays are generated from the interaction of ultra-short laser pulse of petawatt power density with matter. The hard X-rays often overload PMTs and cause signal-induced background noises called afterpulses, making the detection of subsequent neutrons impossible. For this reason, the PMTs are coupled with an electrical time-gating (ETG) system to avoid overloading. The ETG system disables the PMT by modulating the dynode potential during the primary X-ray flash. An after-pulsing suppression technique is demonstrated by applying a reverse bias voltage between the photocathode and the first dynode. The presented multichannel scheme provides a gate response time of 80 ns, a signal cutoff ratio of 2.5 × 102, and requires reasonably low power consumption.

Mesoporous silica materials with different pore volumes were functionalized by wet impregnation with a variety of amines for the purpose of CO2 adsorption. The effects of the concentration and composition of amine blends, silica supports, and adsorption temperature on CO2 adsorption were investigated. The mechanism of an observed synergistic effect between blended amines is also discussed. The experimental results showed that in addition to the pore volume of the supports and the adsorption temperature the molecular functional groups of the blended amines play an important role and greatly affect the CO2 adsorption capacity. The silica material with the largest pores (MSU-F) had the highest CO2 adsorption capacity after impregnation with a mixture of 40 wt % tetraethylenepentamine and 30 wt % diethanolamine. For 100 kPa CO2, the adsorption capacity at 50 °C was 5.91 mmol/g.

A new diagnosis method for high energy ions utilizing a single CR-39 detector mounted on plastic plates is demonstrated to identify the presence of the high energy component beyond the CR-39's detection threshold limit. On irradiation of the CR-39 detector unit with a 25 MeV per nucleon He ion beam from conventional rf-accelerators, a large number of etch pits having elliptical openings are observed on the rear surface of the CR-39. Detailed investigations reveal that these etch pits are created by heavy ions inelastically backscattered from the plastic plates. This ion detection method is applied to laser-driven ion acceleration experiments using cluster-gas targets, and ion signals with energies up to 50 MeV per nucleon are identified.
