Ethylene glycol (EG)-based lubricant was prepared with dissolved organosolv lignin from birch wood (BL) and softwood (SL) biomass. The effects of different lignin types on the rheological, thermal, and tribological properties of the lignin/EG lubricants were comprehensively investigated by various characterization techniques. Dissolving organosolv lignin in EG results in outstanding lubricating properties. Specifically, the wear volume of the disc by EG-44BL is only 8.9% of that lubricated by pure EG. The enhanced anti-wear property of the EG/lignin system could be attributed to the formation of a robust lubrication film and the strong adhesion of the lubricant on the contacting metal surface due to the presence of a dense hydrogen bonding (H-bonding) network. The lubricating performance of EG-BL outperforms EG-SL, which could be attributed to the denser H-bonding sites in BL and its broader molecular weight distribution. The disc wear loss of EG-44BL is only 45.7% of that lubricated by EG-44SL. Overall, H-bonding is the major contributor to the different tribological properties of BL and SL in EG-based lubricants. Specific degradation of the bonds between lignin and carbohydrates is an important step towards separating individual lignocellulosic biopolymers for sustainable production of materials and chemicals. One of the most established covalent lignin-carbohydrate (LC) interactions is the ester bond between the α- or γ-hydroxyl group of a lignin phenylpropane unit and a glucuronic acid side chain of xylan. In this work, a model of the LC benzyl ester bond was synthesized in a one-pot reaction from a β-O-4 lignin unit and D-glucuronic acid, both from commercial sources. The resulting lignin-carbohydrate complex (LCC) model was unstable in aqueous solution. However, at pH 4, the rate of spontaneous hydrolysis was sufficiently low to allow for enzymatic splitting experiments. The enzymatic hydrolysis of the LC benzyl ester bond of the LCC model was demonstrated by means of the glucuronoyl esterase StGE2 from Sporotrichum thermophile, which showed a preference for erythro forms of the LCC model. A comparative study among different pretreatment methods used for the fractionation of the birch outer bark components, including steam explosion, hydrothermal and organosolv treatments based on the use of ethanol/water media, is reported. The residual solid fractions have been characterized by ATR-FTIR, 13C-solid-state NMR and morphological alterations after pretreatment were detected by scanning electron microscopy. The general chemical composition of the untreated and treated bark including determination of extractives, suberin, lignin and monosaccharides was also studied. Composition of the residual solid fraction and relative proportions of different components, as a function of the processing conditions, could be established. Organosolv treatment produces a suberin-rich solid fraction, while during hydrothermal and steam explosion treatment cleavage of polysaccharide bonds occurs. This work will provide a deeper fundamental knowledge of the bark chemical composition, thus increasing the utilization efficiency of birch outer bark and may create possibilities to up-scale the fractionation processes. Glucuronoyl esterases (GEs) are recently discovered enzymes that are suggested to cleave the ester bond between lignin alcohols and xylan-bound 4- O-methyl- d-glucuronic acid. Although their potential use for enhanced enzymatic biomass degradation and synthesis of valuable chemicals renders them attractive research targets for biotechnological applications, the difficulty to purify natural fractions of lignin-carbohydrate complexes hampers the characterization of fungal GEs. In this work, we report the synthesis of three aryl alkyl or alkenyl d-glucuronate esters using lipase B from Candida antarctica (CALB) and their use to determine the kinetic parameters of two GEs, StGE2 from the thermophilic fungus Myceliophthora thermophila (syn. Sporotrichum thermophile) and PaGE1 from the coprophilous fungus Podospora anserina. PaGE1 was functionally expressed in the methylotrophic yeast Pichia pastoris under the transcriptional control of the alcohol oxidase ( AOX1) promoter and purified to its homogeneity (63 kDa). The three d-glucuronate esters contain an aromatic UV-absorbing phenol group that facilitates the quantification of their enzymatic hydrolysis by HPLC. Both enzymes were able to hydrolyze the synthetic esters with a pronounced preference towards the cinnamyl- d-glucuronate ester. The experimental results were corroborated by computational docking of the synthesized substrate analogues. In this study, formaldehyde-free bioresin adhesives were synthesised from lignin and tannin, which were obtained from softwood bark. The extraction was done via organosolv treatment and hot water extraction, respectively. A non-volatile, non-toxic aldehyde, glyoxal, was used as a substitute for formaldehyde in order to modify the chemical structure of both the lignin and tannin. The glyoxal modification reaction was confirmed by ATR–FTIR spectroscopy. Three different resin formulations were prepared using modified lignin along with the modified tannin. The thermal properties of the modified lignin, tannin, and the bioresins were assessed by DSC and TGA. When the bioresins were cured at a high temperature (200 °C) by compression moulding, they exhibited higher thermal stability as well as an enhanced degree of cross-linking compared to the low temperature-cured bioresins. A shift towards an economically viable biomass biorefinery concept requires the use of all biomass fractions (cellulose, hemicellulose, and lignin) for the production of high added-value products. As lignin is often underutilized, the establishment of lignin valorization routes is highly important. In-house produced organosolv as well as commercial Kraft lignin were used in this study. The aim of the current work was to make a comparative study of thermoplastic biomaterials from two different types of lignins. Native lignins were alkylate with two different alkyl iodides to produce ether-functionalized lignins. Successful etherification was verified by FT-IR spectroscopy, changes in the molecular weight of lignin, as well as 13C and 1H Nuclear Magnetic Resonance (NMR). The thermal stability of etherified lignin samples was considerably improved with the T2% of organosolv to increase from 143 °C to up to 213 °C and of Kraft lignin from 133 °C to up to 168 °C, and glass transition temperature was observed. The present study shows that etherification of both organosolv and Kraft lignin with alkyl halides can produce lignin thermoplastic biomaterials with low glass transition temperature. This study demonstrates the effects of structural variations of lignins isolated via an organosolv process from different woody and herbaceous feedstocks on their thermal stability profiles. The organosolv lignins were first analysed for impurities, and structural features were determined using the default set of gel permeation chromatography, FT-IR spectroscopy, quantitative 31 P NMR spectroscopy and semi-quantitative 1 H- 13 C HSQC analysis. Pyrolysis-, O 2 - and CO 2 -reactivity of the organosolv lignins were investigated by thermogravimetric analysis (TGA), and volatile formation in various heating cycles was mapped by head-space GC-MS analysis. Revealed reactivities were correlated to the presence of identified impurities and structural features typical for the organosolv lignins. Lignin, one of the three main structural biopolymers of lignocellulosic biomass, is the most abundant natural source of aromatics with a great valorization potential towards the production of fuels, chemicals, and polymers. Although kraft lignin and lignosulphonates, as byproducts of the pulp/paper industry, are available in vast amounts, other types of lignins, such as the organosolv or the hydrolysis lignin, are becoming increasingly important, as they are side-streams of new biorefinery processes aiming at the (bio)catalytic valorization of biomass sugars. Within this context, in this work, we studied the thermal (non-catalytic) and catalytic fast pyrolysis of softwood (spruce) and hardwood (birch) lignins, isolated by a hybrid organosolv–steam explosion biomass pretreatment method in order to investigate the effect of lignin origin/composition on product yields and lignin bio-oil composition. The catalysts studied were conventional microporous ZSM-5 (Zeolite Socony Mobil–5) zeolites and hierarchical ZSM-5 zeolites with intracrystal mesopores (i.e., 9 and 45 nm) or nano-sized ZSM-5 with a high external surface. All ZSM-5 zeolites were active in converting the initially produced via thermal pyrolysis alkoxy-phenols (i.e., of guaiacyl and syringyl/guaiacyl type for spruce and birch lignin, respectively) towards BTX (benzene, toluene, xylene) aromatics, alkyl-phenols and polycyclic aromatic hydrocarbons (PAHs, mainly naphthalenes), with the mesoporous ZSM-5 exhibiting higher dealkoxylation reactivity and being significantly more selective towards mono-aromatics compared to the conventional ZSM-5, for both spruce and birch lignin. Biomass fractionation, as an alternative to biomass pretreatment, has gained increasing research attention over the past few years as it provides separate streams of cellulose, hemicellulose, and lignin. These streams can be used separately and can provide a solution for improving the economics of emerging biorefinery technologies. The sugar streams are commonly used in microbial conversions, whereas during recent years lignin has been recognized as a valuable compound as it is the only renewable and abundant source of aromatic chemicals. Successfully converting lignin into valuable chemicals and products is key in achieving both environmental and economic sustainability of future biorefineries. In this work, lignin retrieved from beechwood sawdust delignification pretreatment via an organosolv process was depolymerized with thermal and catalytic pyrolysis. ZSM-5 commercial catalyst was used in situ to upgrade the lignin bio-oil vapors. Lignins retrieved from different modes of organosolv pretreatment were tested in order to evaluate the effect that upstream pretreatment has on the lignin fraction. Both thermal and catalytic pyrolysis yielded oils rich in phenols and aromatic hydrocarbons. Use of ZSM-5 catalyst assisted in overall deoxygenation of the bio-oils and enhanced aromatic hydrocarbons production. The oxygen content of the bio-oils was reduced at the expense of their yield. Organosolv lignins were successfully depolymerized towards phenols and aromatic hydrocarbons via thermal and catalytic pyrolysis. Background: Lytic polysaccharide monooxygenases (LPMOs) are copper-dependent enzymes that oxidatively cleave recalcitrant lignocellulose in the presence of oxygen or hydrogen peroxide as co-substrate and a reducing agent as electron donor. One of the possible systems that provide electrons to the LPMOs active site and promote the polysaccharide degradation involves the mediation of phenolic agents, such as lignin, low-molecular-weight lignin-derived compounds and other plant phenols. In the present work, the interaction of the bulk insoluble lignin fraction extracted from pretreated biomass with LPMOs and the ability to provide electrons to the active site of the enzymes is studied. Results: The catalytic efficiency of three LPMOs, namely MtLPMO9 with C1/C4 regioselectivity, PcLPMO9D which is a C1 active LPMO and NcLPMO9C which is a C4 LPMO, was evaluated in the presence of different lignins. It was correlated with the physicochemical and structural properties of lignins, such as the molecular weight and the composition of aromatic and aliphatic hydroxyl groups. Moreover, the redox potential of lignins was determined with the use of large amplitude Fourier Transform alternating current cyclic voltammetry method and compared to the formal potential of the Cu (II) center in the active site of the LPMOs, providing more information about the lignin-LPMO interaction. The results demonstrated the existence of low-molecular weight lignin-derived compounds that are diffused in the reaction medium, which are able to reduce the enzyme active site and subsequently utilize additional electrons from the insoluble lignin fraction to promote the LPMO oxidative activity. Regarding the bulk lignin fractions, those isolated from the organosolv pretreated materials served as the best candidates in supplying electrons to the soluble compounds and, finally, to the enzymes. This difference, based on biomass pretreatment, was also demonstrated by the activity of LPMOs on natural substrates in the presence and absence of ascorbic acid as additional reducing agent. Conclusions: Lignins can support the action of LPMOs and serve indirectly as electron donors through low-molecular-weight soluble compounds. Abstract Lignin, one of the most abundant natural polymers, has been successfully used as an effective lubricant additive with high value. The chemical structure of lignin is very diverse and strongly affected by both the source of lignin (i.e. plant species) and the lignin extraction process. In this work, a series of lignin from different biomass sources (hard or soft wood) and extraction process (organosolv with or without acid catalyst) has been successfully incorporated into poly(ethylene glycol) (PEG) and fortified lubricating properties were achieved. The effects of different lignin on the rheological, thermal and tribological properties of the lignin/EG lubricants were systematically investigated by different characterization techniques. Lignin in PEG significantly improves the lubricating property, where a wear reduction of 93.8% was observed. The thermal and lubrication properties of the PEG lubricants filled with different kinds of lignin are tightly related to the synergistic state of hydrogen bonding and molecular weight distribution. Lignin with broader molecular weight distribution and higher hydroxyl content shows better adhesion on metal surfaces and strengthened lubricating film, which could be used as the efficient lubricating additives. This work provides a criterion for selecting appropriate lignin as the efficient lubricant additive and accelerates the application of lignin. Abstract For a transition to a sustainable society, fuels, chemicals, and materials should be produced from renewable resources. Lignocellulosic biomass constitutes an abundant and renewable feedstock; however, its successful application in a biorefinery requires efficient fractionation into its components; cellulose, hemicellulose and lignin. Here, we demonstrate that a newly established hybrid organosolv – steam explosion pretreatment can effectively fractionate spruce biomass to yield pretreated solids with high cellulose (72% w/w) and low lignin (delignification up to 79.4% w/w) content. The cellulose-rich pretreated solids present high saccharification yields (up to 61% w/w) making them ideal for subsequent bioconversion processes. Moreover, under high-gravity conditions (22% w/w) we obtained an ethanol titer of 61.7 g/L, the highest so far reported for spruce biomass. Finally, the obtained high-purity lignin is suitable for various advanced applications. Organosolv lignins (OSLs) are important byproducts of the cellulose-centred biorefinery that need to be converted in high value-added products for economic viability. Yet, OSLs occasionally display characteristics that are unexpected looking at the lignin motifs present. Applying advanced NMR, GPC, and thermal analyses, isolated spruce lignins were analysed to correlate organosolv process severity to the structural details for delineating potential valorisations. Very mild conditions were found to not fractionate the biomass, causing a mix of sugars, lignin-carbohydrate complexes (LCCs), and corresponding dehydration/degradation products and including pseudo-lignins. Employing only slightly harsher conditions promote fractionation, but also formation of sugar degradation structures that covalently incorporate into the oligomeric and polymeric lignin structures, causing the isolated organosolv lignins to contain lignin-humin hybrid (HLH) structures not yet evidenced as such in organosolv lignins. These structures effortlessly explain observed unexpected solubility issues and unusual thermal responses, and their presence might have to be acknowledged in downstream lignin valorisation. Careful structural analyses of spruce organosolv lignin isolates by state-of-the-art techniques revealed a new structural component: humin-lignin hybrids. These novel structures can help to understand the complex interplay between the structural polymers during common biorefinery approaches, and can explain puzzling physico-chemical behaviours of organosolv lignins. Herein, we have investigated how pure lignin extracted from birch and spruce via a hybrid organosolv/steam explosion method reacts under hydrothermal carbonization (HTC) to produce hydrochar, a product that has found applications in environmental remediation, energy storage and catalysis. We subjected thirteen lignin samples obtained from birch and spruce under different extraction conditions to HTC at 260 ℃ for four hours. The yield of hydrochar varied between the different extraction conditions and source, although no clear correlation between extraction conditions and yield could be observed. For instance, lignin from birch pretreated in 60%v/v ethanol for 15 min resulted in a hydrochar yield of 39 wt%. Increasing the time to 30 and 60 resulted in a hydrochar yield of 27 wt% and 23 wt%, respectively. This suggested that small changes in the organosolv reaction conditions might produce highly structurally different lignin, resulting in the difference in HTC yield. Thus, we chose a subset of four lignin samples to investigate in-depth, subjecting these samples to a range of hydrothermal reaction temperatures and residence times. Solid State NMR and FTIR analysis indicated that the most significant structural changes occurred below 230 ℃ resulting in the breaking of C-O- linkages. Increasing the temperature or time had minimal impact, with no further C-O- linkages broken and no changes to the ring structure of C-C groups. Size exclusion chromatography indicated that the degree of micro and macromolecules in the liquid product varied significantly with lignin source and HTC reaction conditions. Overall, this study demonstrated that lignin has a large reaction range where it produces a very chemically similar solid product, with the only major difference being the yield of material. An enzyme belonging to the glycoside hydrolase family 61 from the thermophilic fungus Sporotrichum thermophile , was functionally expressed in the methylotrophic yeast Pichia pastoris under the transcriptional control of the alcohol oxidase ( AOX1 ) promoter. The enzyme hydrolyzed barley β-glucan, carboxymethyl cellulose, lichenan, wheat arabinoxylan and birchwood xylan showing optimal activity at pH 8 and 65 °C. A 2:1 mixture of Celluclast 1.5 L and St Cel61a was capable of increasing the degree of spruce conversion by 42%. The use of substrates with varying lignin content permitted the detection of a dependence of the enhancing capacity of St Cel61a on the radical scavenging capacity of the different lignocellulosics. In the presence of a reductant, St Cel61a boosted the efficiency of a mixture of purified cellulases (EGII, CBHI, β-GLUC) by 20%. The synergistic activity exhibited by St Cel61a and its dependence on reducing substances provide guidelines for process design towards the production of economically viable bioethanol. This study investigates whether the variability in lignin, created from different plant types, growing conditions and extraction methods, influences the properties of the final hydrothermal carbon material. Seven different lignin samples, three from Kraft extraction and four from organosolv extraction, were subjected to hydrothermal treatment at 260℃ for four hours to assess the impact of lignin type on the physicochemical properties of the hydrothermal material. The 13C Solid state NMR, XPS, FTIR and SEM analysis revealed that the different sources of lignin and the extraction conditions created variations in the degree of syringyl and guaiacyl subunits, inter-unit bonding arrangements, morphology and surface composition. Hydrothermal carbonization appeared to "normalize" the differences between each of the lignin samples, via breaking β-O-4 or α-O-4 linkages, removal of methoxy and syringyl subunits, and creation of C C and 4-O-5 linkages to polymerization into large 100−200 μm amorphous carbon particles. The oil scarcity and the rise in earth temperature have elevated the interest in lignocellulosic biorefineries. Lignin has high potential to be used in various applications including the production of biomaterials and transportation fuels. Among the different sources of lignin, organosolv lignin has the advantage of being sulphur-free and of low ash content compared to other types of industrial lignin. The present study focuses on cradle-to-gate life cycle and cost assessment of a novel organosolv lignin production process from spruce bark. The system boundary included production of tannin, lignin from spruce bark and handling of waste including all the inputs (material and energy) and outputs (emissions) in the process. Baseline scenario and scenarios S1 and S2 were compared to identify the most environmentally and economically suitable scenario. The baseline scenario is lignin production with co-production of tannin and tannin free bark (TFB) from spruce bark; scenario S1 is lignin production from TFB; and scenario S2 is lignin production from TFB with mass allocation. The functional unit was 1 kg lignin produced and ReCiPe 2016 Midpoint (H) method was used for the environmental impact assessment. The results showed that the baseline scenario had higher global warming potential (GWP) (2.14 kg CO 2 eq.) and total cost (1.959 €/kg) than S1 (1.39 kg CO 2 eq. and 1.377 €/kg respectively) and S2 (0.23 kg CO 2 eq. and 0.998 €/kg respectively) scenarios. Flotation is a key step during mineral separation. Xanthates are the most commonly used collectors for recovering Cu, Ni, and Zn from sulphide ores. However, xanthates are fossil-based and toxic for the environment. The aim of this study was to evaluate the use of lignin nanoparticles and microparticles as sustainable and environmentally friendly collectors. Lignin particles demonstrated good selectivity toward Cu (chalcopyrite), with total recoveries exceeding 80% and grades of up to 8.6% w/w from a Cu-Ni ore in rougher flotation tests. When floating Zn-Pb-Cu ore, lignin nanoparticles could reduce the use of xanthates by 50%. Moreover, they outperformed xanthates alone, achieving total recoveries of up to 91%, 85%, and 98% for Cu, Pb, and Zn, respectively. Lignin still remains an underutilized plentiful resource whose conversion to high-added value products is a cornerstone towards establishing a viable biomass biorefinery. Bio-materials in the form of nanoparticles represent promising high-value products with numerous downstream applications. The aim of the current work was to develop a method that would allow controlling the size of (birch and spruce) lignin nano- and micro-particles for their subsequent recovery into a solid product. We tested different two-step and one-step isolation processes and demonstrated that particle size could be easily controlled to meet different ranges (<100 nm, <500 nm, and>1 μm). In general, two-step isolation methods, i.e. a step of decrease of solvent concentration followed by isolation of lignin particles, were better for the isolation of well-defined spherical particles. In particular, the rate at which ethanol concentration was decreased played a significant role in determining the size of lignin particles. Moreover, when lignin concentration was increased from 1 % to 5 % and 10 % (w/v), particle size and homogeneity decreased slightly, but productivity augmented.
