The immunity‐related GTPases (IRGs) belong to the interferon‐inducible GTPase protein family, which mediates cell autonomous and innate immunity response to intracellular pathogens. Yet, the cellular and physiological function of IRGC, a member of the IRG subfamily, has not been elucidated. Here, we show that testis‐specific IRGC is specifically and highly expressed in mature spermatozoa and is required for sperm motility. IRGC induction results in the clustering of lipid droplets and initiation of their physical contact with mitochondria. When examining clinical semen samples, IRGC expression is significantly lower in asthenozoospermia patients relative to healthy individuals. These unique effects of IRGC identify it as an important player in sperm motility, and show the potential of lipid metabolism‐targeting therapeutic intervention aimed at controlling asthenozoospermia. The subcellular location and cell biological function of small GTPase Rab40c in mammalian cells have not been investigated in detail. In this study, we demonstrated that the exogenously expressed GFP-Rab40c associates with lipid droplets marked by neutral lipid specific dye Oil red or Nile red, but not with the Golgi or endosomal markers. Further examination demonstrated that Rab40c is also associated with ERGIC-53 containing structures, especially under the serum starvation condition. Rab40c is increasingly recruited to the surface of lipid droplets during lipid droplets formation and maturation in HepG2 cells. Rab40c knockdown moderately decreases the size of lipid droplets, suggesting that Rab40c is involved in the biogenesis of lipid droplets. Stimulation for adipocyte differentiation increases the expression of Rab40c in 3T3-L1 cells. Rab40c interacts with TIP47, and is appositionally associated with TIP47-labeled lipid droplets. In addition, over-expression of Rab40c causes the clustering of lipid droplets independent of its GTPase activity, but completely dependent of the intact SOCS box domain of Rab40c. In addition, Rab40c displayed self-interaction as well as interaction with TIP47 and the SOCS box is essential for its ability to induce clustering of lipid droplets. Our results suggest that Rab40c is a novel Rab protein associated with lipid droplets, and is likely involved in modulating the biogenesis of lipid droplets. [ABSTRACT FROM AUTHOR]

Abstract: The conversion of lipid-extracted microalgal biomass residues (LMBRs) into hydrogen plays the dual role in renewable energy production and sustainable development of microalgal biodiesel industry. An anaerobic fermentation process to covert LMBRs into hydrogen was investigated in this work. Using batch experiments, the effects of pretreatment of inoculum (by acid, base, heat, and chloroform, respectively), initial pH (5.0–7.0), inoculum concentrations at 0.59–2.94gVSS/l (volatile suspended solids, VSS) and substrate concentrations at 4.5–45gVS/l (volatile solids, VS) were investigated, respectively. The results showed that the most effective hydrogen production was obtained from fermentation of LMBRs with a concentration of 36gVS/l at the initial pH 6.0–6.5 using the heat-treated anaerobic digested sludge as inoculum. Acetate, propionate and butyrate were the main fermentation byproducts in the conversion of LMBRs into hydrogen. [Copyright &y& Elsevier]

Abstract: A two-stage process to produce hydrogen and methane from lipid-extracted microalgal biomass residues (LMBRs) was developed. The biogas production and energy efficiency were compared between one- and two-stage processes. The two-stage process generated 46 ± 2.4 mL H2/g-volatile solid (VS), and 393.6 ± 19.5 mL CH4/g-VS. The methane yield was 22% higher than the one in the one-stage process. Energy efficiency increased from 51% in the one-stage process to 65% in the two-stage process. Additionally, it was found that repeated batch cultivation was a useful method to cultivate the cultures to improve the methane production rate and reduce the fermentation time. In the repeated batch cultivation, the methane yield slightly decreased if the ammonia levels rose, suggesting that the accumulation of ammonia could affect methane production.
