Depletion of ascorbic acid from adrenals, brain and epididymis along with loss in weight were noticed in the state of thyroidectomy. This decrease appears to be due to an effect of thyroidectomy on the membrane integrity since the membrane bound sialic acid was found to be significantly lowered in these tissues as a consequence of the elevated activity of sialidase. Thyroidectomy was also found to cause an adverse effect on the activities of hepatic UDP-glucuronyl transferase and beta-glucuronidase with no alteration in UDP-glucose dehydrogenase.

Time-courses of changes in the activities of liver and kidney glucose-6-phosphatase [EC 3.1.3.9] and hepatic tryptophan pyrrolase [EC 1.13.1.12; TPO] in rats pre-fed high-protein diets for 5 days and then shifted to zero-protein diets were studied. Liver glucose-6-phosphatase activity decreased 1 day after the dietary shift but then increased and remained significantly higher than the 0 day value for the next 2 days. Changes in liver glycogen were found to be intimately and inversely related to liver glucose-6-phosphatase activity. Changes in kidney glucose-6-phosphatase activity paralleled the pattern of changes observed in liver activity. An initial decrease in TPO activity was followed by increased enzyme activity up to the 3rd day of the dietary shift. Later there was a rapid fall in tryptophan pyrrolase activity. Changes observed in these specific enzyme proteins differed from those observed in total tissue proteins. Alterations in the activities of these enzymes and changes in other parameters are compared with those observed earlier with the reverse type of dietary shift.

Hepatic drug metabolism in patients with acute viral hepatitis was investigated under different conditions: pregnancy, postpartum, non-pregnancy, and among males. Liver function tests were altered in all of these conditions. The relationship between in vivo and in vitro drug metabolism was studied in twenty-two patients using diagnostic liver needle biopsies by comparing the drug metabolising enzymes (aminopyrine-N-demethylase and bilirubin-UDP-glucuronyl transferase) in these biopsies with the elimination kinetics of antipyrine. Drug clearance tests were repeated in the patients who had recovered. All patients gave altered liver function tests results. Antipyrine half-life was significantly higher in females as compared to males. Among females, it was maximum in the pregnant group. Activities of drug metabolising enzymes were found to be significantly lower in liver biopsy material. The half-life of antipyrine showed a significant correlation with drug metabolising enzymes. After recovery patients showed a normal antipyrine half-life.
