Capecitabine is converted into 5'-deoxy-5-fluorocytidine (5'DFCR), 5'-deoxy-5-fluorouridine (5'DFUR) and 5-fluorouracil (5-FU) by CES1 and 2, CDD, and TP, in both liver and tumour. 5-FU is catabolised by DPD. Gene expression analysis of these enzymes was undertaken in fresh human hepatocytes, mouse liver, colorectal cancer cell lines and xenografts. Cell lines with low CDD expression (<1.5) had 5'DFCR/5'DFUR cytotoxicity ratios>2 and cell lines with TP/DPD<0.6 had 5'DFUR IC50>50 microM (SRB assay). A pharmacokinetic/pharmacodynamic study in nude mice bearing HCT 116 xenografts and treated with capecitabine by oral gavage assessed pharmacokinetic, gene expression and antitumour activity. Low liver CDD correlated with high 5'DFCR plasma concentrations in mice. CDD expression was approximately 100-fold higher in fresh human hepatocytes than mouse liver, explaining the higher plasma 5'DFUR concentrations reported previously in humans. Tumour 5-FU concentration correlated with TP/DPD and with tumour response. These studies identify the potential utility of gene expression analysis and drug monitoring in tumour in patients.

Plasma-membrane fragments recovered in the microsomal fraction of rat liver homogenates were shown to be heterogeneous in density. It was demonstrated that 5'-nucleotidase, the most commonly used plasma-membrane marker, is concentrated in the lightest subfraction. Two of the published procedures for the isolation of plasma-membrane fragments from the microsomal fraction (Touster et al., 1970; Hinton et al., 1971) are shown to give products which are not representative of all the plasma-membrane fragments of microsomal size, and it is argued that a third procedure (House & Weidemann, 1970) is likely to give a similar product.

The ribonuclease and phosphodiesterase activities of rat liver plasma membranes, purified from the crude nuclear fraction by centrifugation in an A-XII zonal rotor and flotation, were examined and compared. The plasma membrane is responsible for between 65 and 90% of the phosphodiesterase activity of the cell and between 25 and 30% of the particulate ribonuclease activity measured at pH8.7 in the presence of 7.5mm-MgCl(2). Both enzymes were most active between pH8.5 and 8.9. Close to the pH optimum, both enzymes were more active in Tris buffer than in Bicine or glycine buffer. Both plasma-membrane phosphodiesterase and ribonuclease were strongly activated by Mg(2+), there being at least a 12-fold difference between the activity in the presence of Mg(2+) and of EDTA. There is, however, a difference in the response of the enzymes to Mg(2+) and EDTA in that the phosphodiesterase is fully activated by 1.0mm-MgCl(2) and fully inhibited by 1.0mm-EDTA, whereas the ribonuclease requires 7.5mm-MgCl(2) for full activation and 5mm-EDTA for full inhibition. Density-gradient centrifugation has indicated that on solubilization in Triton X-100 most of the ribonuclease activity is released into a small fragment of the same size as that containing the phosphodiesterase activity. The relationship between the two activities is discussed in view of these results.

In view of the unsatisfactory appearance, under the electron microscope, of liver mitochondria isolated in isotonic sucrose medium, alternative media have been examined. It was found to be advantageous to replace sucrose by raffinose, and to add levan or, preferably, dextran, together with heparin in suitable concentration. With the optimal medium, the constituents of which are raffinose, versene (optional), dextran of high molecular weight, heparin, and AMP (optional), most of the mitochondria in the osmium-fixed pellet are apparently intact, and show the membranes characteristic of mitochondria as seen in cell sections. The optimal medium has no adverse effect on the activity of the several tissue enzymes which have been studied, except that Mg(++)-activated ATPase is partially inhibited if the medium is present in high concentration in the assay system. Mitochondrial fractions isolated in the new medium have, in common with sucrose fractions, appreciable "free" ATPase activity, this activity being evidently a poor criterion of mitochondrial integrity. Use of the new medium does not decrease the proportion of cytoplasmic ATPase which fails to sediment with the mitochondria, but does give a mitochondrial fraction low in RNA and in acid phosphatase activity and little contaminated with microsomal material. Particles tentatively identified as "lysosomes" have been seen in certain sections.

The behaviour of heavy subcellular particles from rat liver during centrifugation in an A-XII zonal rotor has been studied with the help of enzymic markers and light and electron microscopy, A simple and reproducible method, not entailing hypotonic conditions, was developed for the isolation of plasma membrane fragments from liver homogenates, after study of factors affecting the yield and purity of such fragments. The livers were perfused with warm 0.25 M sucrose—5 mM NaHCO3, pH 7.5. Homogenisation and resuspension of the nuclear fraction were carried out in the same medium. Zonal centrifugation gave a band of plasma membrane fragments which showed a 14-fold increase in 5′-nucleotidase specific activity over the homogenate. This purification was increased to 30-fold by vigorous rehomogenisation in sucrose of density 1.19 followed by flotation of the plasma membrane fragments. These were now almost free of succinatedehydrogenase activity although still possessing some glucose-β-phosphatase and acid β-glycerophosphatase activity. [ABSTRACT FROM AUTHOR]
