Survival, T-cell functions, and postmortem histopathology were studied in H-2 congenic strains of mice bearing H-2b, H-2k, and H-2d haplotypes. Males lived longer than females in all homozygous and heterozygous combinations except for H-2d homozygotes, which showed no differences between males and females. Association of heterozygosity with longer survival was observed only with H-2b/H-2b and H-2b/H-2d mice. Analysis using classification and regression trees (CART) showed that both males and females of H-2b homozygous and H-2k/H-2b mice had the shortest life-span of the strains studied. In histopathological analyses, lymphomas were noted to be more frequent in females, while hemangiosarcomas and hepatomas were more frequent in males. Lymphomas appeared earlier than hepatomas or hemangiosarcomas. The incidence of lymphomas was associated with the H-2 haplotype--e.g., H-2b homozygous mice had more lymphomas than did mice of the H-2d haplotype. More vigorous T-cell function was maintained with age (27 months) in H-2d, H-2b/H-2d, and H-2d/H-2k mice as compared with H-2b, H-2k, and H-2b/H-2k mice, which showed a decline of T-cell responses with age.

Tumor cell variants were derived from an AKR T-cell lymphoma cell line (BW5147, H-2k haplotype). These variants differed in their malignant potential and in their membrane expression of class I MHC antigens. High tumorigenic and spontaneous metastatic capacity was found to be predominantly associated with a decrease of H-2Kk class I major histocompatibility complex (MHC) antigen expression. In contrast, high experimental metastatic capacity correlated strongly with an increased H-2Dk antigen expression. The in vitro invasive potential and the LFA-1 expression of the BW variants showed no correlation with the differential MHC antigen expression and the differential metastatic and tumorigenic capacity of the BW variants. Furthermore, the susceptibility of the BW 5147 variants to TNF and NK-mediated cytotoxicity was not related to the differential metastatic potential and the expression of the class I MHC antigens.

The relationship between MHC class I (H-2) expression and tumorigenicity was investigated after intracerebral inoculation of the murine lymphoma YAC-1 and its H-2 negative variant, A.H-2-. YAC-1 was less tumorigenic than A.H-2- in normal as well as NK-depleted syngeneic A/Sn mice. However, in T-cell-depleted syngeneic mice YAC-1 was as tumorigenic as A.H-2-. Following intracerebral growth, the H-2 expression of YAC-1 was markedly enhanced in a similar fashion as after intraperitoneal passage. The A.H-2- variant remained H-2 negative after intracranial passage. The H-2 negative variant cells were not rejected from the brain even when intermixed with wild-type YAC-1 cells prior to intracerebral inoculation, excluding an "innocent bystander" effect. In vitro, the intracerebrally passaged YAC-1 line showed enhanced sensitivity to lysis by H-2 Kk Dd (H-2a) specific CTLs but decreased sensitivity to NK cells. The A.H-2- line was unchanged. Our data suggest that the lack of H-2 molecules may facilitate the growth of antigenic tumor cells in the brain due to escape from T-cell-mediated immunosurveillance. Our data also suggest, in line with other recent findings, that intracerebrally growing tumor cells are sheltered from NK cell-mediated rejection.

SJL/J lymphomas emerged as H-2Ds-negative variants upon transplantation of their respective H-2Ds-positive spontaneous tumors. Of nine spontaneous SJL/J lymphomas examined, only two contained detectable numbers of H-2Ds-negative tumor cells. Of seven successfully transplanted tumors, however, four emerged as predominantly H-2Ds-negative tumors by their second transplantation. Long term SJL/J lymphoma lines, RCS 5 and SJL/SJL, were also found to be H-2Ds-negative. The expression of H-2Ks and I-As were maintained on all tumor lines examined. H-2Ds-negative tumor cells were consistent stimulators of syngeneic lymphoproliferation, whereas tumors maintaining H-2Ds expression varied in their stimulatory capacity. Transplantation times required to achieve morbidity in syngeneic mice correlated with tumor cell expression of H-2Ds. Tumors maintaining H-2Ds expression were less aggressive and required 57 to 150 days to produce morbidity compared with highly aggressive H-2Ds-negative tumors that required only 7 to 10 days at one-quarter of the cell dose (10(7) cells). Growth curve analysis based on spleen weights (corrected for body weight) indicated that three types of SJL/J lymphomas could be identified based on their expression of H-2Ds and ability to stimulate syngeneic lymphoproliferation: 1) indolent, nonstimulatory H-2Ds-positive tumors; 2) indolent, stimulatory H-2Ds-positive tumors; and 3) highly aggressive, stimulatory H-2Ds-negative tumors.

Splenocytes of A(H-2a), BALB/c(H-2d) and C57BL/6(H-2b) mice that were sensitized against several allogeneic normal lymphocytes and lymphoma cells in mixed lymphocyte cultures (MLC) and mixed lymphocyte-tumor cell cultures (MLTC) exhibited the expected cell-mediated cytotoxic responses against normal and tumor cells carrying the relevant alloantigens. In contrast, strong cytotoxic reactivity cross-reactive with C57BL/6 target cells was observed when a variant of YAC lymphoma (YAC-b) of strain A mice were employed either as sensitizers or as targets. Furthermore, this 'unpredicted' cross-reactivity was also obtained with syngeneic responder splenocytes sensitized to YAC-b cells. The possibility that the nonspecific cytotoxicity was caused by activation of natural killer (NK) cells in the culture was excluded by the finding that the cytotoxic capability of sensitized effector cells was greatly diminished by treatment with anti-Thy 1.2 serum and complement. Serologic analysis with anti-H-2 alloantisera revealed that the YAC-b tumor line, but not the parental YAC lymphoma, carried H-2b alloantigens. It is concluded that the 'unpredicted' cytotoxic responses here observed is a result of conventional antigen recognition and of specific sensitization to alien H-2b components expressed on the YAC-b subline.

The role of protein glycosylation in the tumor lysis mediated by effector cells derived from Moloney-sarcoma-virus(MSV)-immune mice was studied. Treatment of the Moloney-virus-induced H-2b lymphoma target cells, MBL-2, with tunicamycin (TM), an inhibitor of the protein-N-linked glycosilation, was found to cause a loss of susceptibility to lysis by MSV-immune syngeneic effectors cells, while the same target cells remained fully sensitive to the lytic action of anti-H-2b-immune lymphocytes. Examination of MBL-2 cell surface by lactoperoxidase, 125I iodination, and immunoprecipitation by antiviral protein sera revealed that env but not gag viral gene-encoded products were expressed on the surface of this lymphoma. The TM-induced alteration of cell surface expression of H-2Db, H-2Kb, and gp70 antigens was examined by a combined approach of serological and biochemical techniques. The results were concordant in indicating that (1) after 16 h of TM treatment the cells showed a decreased expression of the three glycoproteins, (2) H-2Db (the restriction element in this system) resulted more affected by the treatment than its counterpart H-2Kb (75% vs 50% reduction as compared to untreated cells), (3) an additional lighter form of H-2Kb was found on the surface of TM-treated cells. In the context of an "associative recognition' of Db and gp70 by MSV-immune effector cells, our results may explain the loss of susceptibility to antitumor effectors of TM-treated MBL-2 cells by a quantitative reduction in the expression of both molecules which interact to create the target structure of syngeneic effectors.

Expression of several thymocyte surface antigens was monitored in a murine model system of thymic lymphoma induction in two different strains of mice. RF/J mice are sensitive to tumor induction by N-nitrosomethylurea (NMU) and by gamma-irradiation, while 129/J mice form tumors only upon NMU treatment. Latency periods for tumor formation were characteristically different depending upon the inducing agent and the mouse strain. We observed differences in thymic leukemia antigen and H-2K expression according to the mode of tumor induction and in relation to the mouse strain, implying multiple factors involved in target cell selection and tumor progression.

It has previously been reported that MHC class I-deficient RBL-5 and YAC-1 lymphoma sublines show an enhanced NK sensitivity in vitro. In the present study such lymphoma variants were found to have different defects in H-2 biosynthesis such as 1) reduced beta 2-microglobulin and H-2 H chain transcription, 2) block in beta 2-microglobulin translation, and 3) impaired association between beta 2-microglobulin and H-2 H chains. For lines with the latter two defects, the results suggested that a major part of the H chains were arrested before the middle Golgi complex as indicated by their failure to undergo carbohydrate side chain trimming. The data suggest that NK sensitivity can be directly influenced by the membrane expression of MHC class I gene products of tumor targets because three independent molecular defects, all interfering with the cell surface H-2 expression, gave rise to NK-sensitive phenotypes. These variant lines will also be useful tools for studies of H-2 glycoprotein assembly and transport and for Ag presentation to CTL.

Interferon-gamma (IFN-gamma) treatment or in vivo passage of the murine YAC-1 lymphoma resulted in reduced sensitivity to in vitro lysis by syngeneic murine spleen cells cultured in rIL-2 (LAK-cells). IFN-gamma treatment also rendered the murine B16 melanoma less sensitive to lysis by syngeneic LAK cells, whereas in vivo passage did not alter LAK sensitivity. The reduction in sensitivity to lysis correlated with enhanced expression of cell surface H-2 on the target cells. The possible role of H-2 was studied with a beta 2-microglobulin-deficient, and thus H-2-deficient, variant of the YAC-1 lymphoma. This variant line remained H-2 negative even after IFN-gamma treatment or in vivo passage, and was highly sensitive to LAK-cell-mediated lysis, even after IFN-gamma treatment or in vivo passage. The present results are discussed in relation to IFN-gamma and in vivo induced modulation of MHC class-1 molecules on target cells and the possible consequences for interaction with activated as well as "natural" effector cells.

BW 5147 lymphoma cells are non-invasive tumor cells which do not generate experimental metastases following i.v. inoculation. In contrast, s.c. and intra-splenic (i.s.) growth of BW cells resulted in widespread colonization of liver and spleen. Cells derived from either s.c. tumors or metastatic lesions did generate metastases after i.v. administration. The capacity of these tumor-derived BW cells to disseminate via blood-borne cells was irreversible and stable, indicating that one in vivo passage of BW cells results in the generation of new, metastatic BW variants. Concomitantly, these variants exhibited an inherent invasive potential as manifested by their capacity to infiltrate in vitro monolayers of hepatocytes and fibroblasts. The BW variants expressed new membrane markers such as H-2 antigens, the Lyt 1.2 T-cell differentiation antigen and the MTH antigen (a newly defined membrane antigen expressed predominantly on murine metastatic T-cell lymphomas and mature T lymphocytes). This phenomenon was observed with both cloned and uncloned BW populations, suggesting that an inductive rather than a selective mechanism accounts for the transition of BW cells towards a more malignant phenotype. These observations confirm the concept that local factors at the growth site of a tumor might influence the metastatic behavior of that tumor, possibly via induction of silent differentiation programs.

Five different sublines of the BALB/c murine S49.1 T cell lymphoma were found to exhibit distinct patterns of absence of detectable H-2d class I major histocompatibility antigen expression. The results were demonstrated and verified by a) the generation of H-2Kd-, H-2Dd,Ld-, and H-2Ld-specific cytotoxic T lymphocytes that were assayed on S49.1 target cell lines, b) antibody-mediated cytotoxicity with the use of anti-H-2d monoclonal reagents, and c) flow microfluorometry. The five lines investigated were S49.1, T-25, T-25ADH, Thy-1-, and 100/0. None of these lines expressed detectable levels of Ld. S49.1 expressed both Kd and Dd, T-25 and T-25ADH expressed Dd but not Kd or Ld, Thy-1- expressed Kd but not Dd or Ld, and 100.0 did not express any detectable amounts of Kd, Dd, or Ld. These results indicate that K and D (and L) antigens can be expressed independently of each other and suggest that expression of class I antigens is controlled in a locus-specific manner.

Vaccination of mice with tumor-derived idiotypic IgM from the B cell lymphoma, BCL1, induces an anti-idiotypic immune response which suppresses tumor development. One of the mechanisms by which tumor cells can escape attack is by failing to express significant levels of idiotypic immunoglobulin at the cell surface, and a stable variant of this phenotype has been isolated. The variant, termed SNAG 1, continues to synthesize idiotypic IgM, which can be detected in the cytoplasm, but it neither secretes nor expresses IgM on the cell surface (less than 10% of the levels of the original BCL tumor), even though the H and L chains show no gross structural changes. The SNAG 1 cells resemble the parent BCL cells in morphology, in expression of MHC class I and II Ag and in bearing FcR. A significant difference between the BCL lymphoma cells and the variant cells is that the latter fail to respond to LPS by either DNA synthesis or secretion of IgM, suggesting that surface Ig might be required for such a response. The variant has a slower rate of division than the parent tumor both in vitro and in vivo, and a rather different organ distribution. Study of such variants might allow analysis of the mechanisms involved in surface Ig expression and its possible role in tumor cell growth and migration.

The effects of dimethyl sulfoxide (DMSO) on H-2 expression and susceptibility to NK- and cytotoxic T-lymphocyte (CTL)-mediated lysis in the murine T-cell lymphoma YAC-1 and its beta-2-microglobulin (beta 2m)-deficient variant were studied. Fluorescence-activated cell sorter analysis revealed induction of H-2Kk and beta 2m 3 days after culture of YAC-1 with DMSO, whereas optimal H-2Dd induction required more than 1 week. H-2Kk and H-2Dd induction by DMSO was equal to pretreatment of YAC-1 cells with 50-100 and 10-20 U/ml interferon (IFN)-gamma, respectively, but the T-cell differentiation antigens Lyt-1, Lyt-2, Thy-1, and L3T4 remained unaffected. DMSO protected YAC-1 cells from NK lysis as efficiently as 10-20 U IFN/ml, whereas susceptibility to anti-H-2a-, H-2Kk-, and H-2Dd-specific CTLs was augmented as in IFN-treated YAC-1 cells. In contrast, the beta 2m-deficient variant, which remained H-2 negative at the cell surface after DMSO treatment, also remained NK sensitive. Thus DMSO can induce H-2 expression and alter the sensitivity of murine lymphoma cells to different effector cells.

Metastases of ESb lymphoma cells in syngeneic DBA/2 animals frequently are selectively immunoresistant to lysis by syngeneic cytotoxic T-lymphocytes (CTL). This immunoresistance of tumor cells to CTL lysis could be due to a defect in either of two structures: (a) tumor-associated transplantation antigens; or (b) mouse major histocompatibility complex (H-2) antigens serving as restricting elements. In this study, we have analyzed the possible involvement of major histocompatibility complex Class I antigens in the immunoresistance of ESb tumor variant cells. Syngeneic anti-ESb CTL appeared to be H-2Kd restricted since only antibodies to Kd but not D,Ld molecules could inhibit CTL lysis. Comparison of H-2 antigens expressed on immunosensitive and resistant ESb sublines by immunofluorescence and flow cytofluorography, alloreactive CTL, two-dimensional gel analysis did not reveal any differences either qualitatively or quantitatively. Southern blotting of tumor-derived DNA with H-2-specific probes did not reveal differences either. Serologically detectable cell surface differentiation antigens were expressed very similarly on immunosensitive and resistant tumor lines, and only minor differences were noted by biochemical analysis of plasma membrane proteins. C-Type viral Mr 70,000 glycoprotein antigens were also similar in both types of cells. We conclude that cell surface changes on immunoresistant ESb variant cells are very selective and involve only CTL-defined tumor-associated transplantation antigen determinants.

Multiple (up to 14) layers of lipid were deposited onto an alkylated glass surface by dialysis of egg phosphatidylcholine (PC) and deoxycholate mixed micelles in the presence of alkylated glass coverslips. The amount of lipid associated with the coverslips was measured by using radioactive PC. It was found that the number of PC molecules in the multilayer increased with increasing initial lipid concentration in the dialysis mixture. Inclusion of cholesterol resulted in a significant increase in the amount of total lipid deposited in the multilayer. However, the PC/cholesterol ratio was up to 2-fold higher in the multilayers than in the liposomes present in the same dialysis bag. In addition, mouse monoclonal anti-H2Kk antibody which had previously been derivatized with palmitic acid could be readily incorporated into the lipid multilayer during dialysis. Measurements of lateral mobility with the fluorescence recovery after photobleaching technique on fluorescently labeled lipid or antibody in the multilayer showed that the lipid molecules diffused rapidly while the antibodies were essentially immobile. Lymphoma cells such as RDM4 cells expressing surface H2Kk glycoproteins could rapidly bind to the antibody-containing multilayers. The binding was blocked by free antibody or by goat anti-mouse immunoglobulin G, indicating the immunospecificity of the binding. Cell binding to the multilayer also exhibited a threshold dependence on the antibody density of the multilayer. A lower threshold was found for cells expressing a higher surface density of H2Kk. This system may be useful for model studies of cellular recognition.

A stable subline of 38C13 B-cell lymphoma (SIR-1) resistant to the antiproliferative effects of alpha-interferon (IFN) was isolated. In addition to defects in antiproliferative effects of IFN, SIR-1 is defective in IFN-mediated antiviral activity against both encephalomyocarditis virus and vesicularstomatitis virus. It is also defective in the induction of 2'-5'-oligoadenylate synthetase mRNA and enzyme activity, enhancement of H-2 antigen expression, and transient induction and subsequent repression of c-myc by IFN. SIR-1, although completely resistant to IFN in vitro, is more sensitive to IFN than the parental cell line in vivo. IFN treatment at 10(4) units, three times weekly, resulted in a 28% increase in mean survival time and a 1.4% long term survival rate in the IFN-sensitive 38C13 cell line but resulted in a 275% increase in mean survival rate and a 27% long term survival rate in the interferon-resistant SIR-1 mutant. Statistical analysis of 38C13 and SIR-1 with and without IFN treatment demonstrate that: a) the SIR-1 mutant remains sensitive to the cytotoxic effects of IFN in vivo (P less than 0.0001); and b) the mean survival and long term survival of animals with the SIR-1 mutant is significantly greater than for animals with the IFN-sensitive 38C13 cell line (P less than 0.0001). Two additional independently isolated IFN-resistant cell lines (SIR-111 and SIR-E102) also demonstrate significantly enhanced in vivo response to IFN compared to the interferon-sensitive parental (38C13) cells. These results indicate that, for this cell line, the antitumor effects of IFN are mediated by activation of host defenses and that resistance to the in vitro cytotoxic effects of IFN results in a tumor phenotype that is more readily recognized by host defenses and eliminated.

Injection of a dual tropic virus (DTV) isolated from a T cell lymphoma AKR/J mice into the thymus of 14 day old AKR/J puppies accelerates lymphoma development; 90-100% of the injected mice develop the disease within 120 days. In contrast a cell free centrifuge CFC-666 prepared B cell lymphoma of AKR/J origin injected into the thymus of 14 day old AKR/J mice failed to accelerate T cell lymphomagenesis and actually prevented the spontaneous T cell lymphoma development. However, 50% of the treated mice developed B cell lymphoma with a latency of 417 + 18 days. DTV injection induces amplification of thymic expression of MuLV related antigens, besides changes in thymus subpopulation. Such changes emerge spontaneously in 5-6 month preleukemic AKR/J mice (at the time of spontaneous DTV formation in the thymus). These changes in the thymus were not observed following CFC-666 injection. We assume therefore that CFC-666 interferes with spontaneous DTV formation that contributes to T cell lymphomagenesis.

The authors have investigated intracerebral natural resistance mechanism after tumor transplantation into the brain, by using YAC-1 (Moloney leukemia virus-induced T cell lymphoma of A/Sn mouse origin) and its H-2 negative A. H-2-. It was found that highly immunogenic H-2+ YAC-1 was less tumorigenic than A. H-2- in untreated as well as NK-depleted syngeneic mice. The variant cells were not rejected even if inoculated together with YAC-1 cells into the brain. Furthermore, in T cell-depleted, thymectomized mice YAC-1 was as tumorigenic as A. H-2-. Thus, intracerebral natural resistance was expressed against YAC-1, suggesting that T cells but not NK cells might be involved in the tumor rejection with an MHC-restricted regulation. Contrary to this, A. H-2- cells escaped from the natural resistance of the brain. In vitro cytotoxicity assays showed that in relation to the enhancement of cell surface H-2 antigens, intracerebrally passaged YAC-1 cells decreased and increased the sensitivity to NK- and CTL-mediated lysis, respectively. In contrast, A. H-2- did not alter either susceptibility to cell-mediated lysis or cell surface H-2 expression. In vivo rapid elimination assays revealed that after intravenous or subcutaneous inoculation there was a more efficient abrogation of 125I-iododeoxyuridine (IUdR) labelled YAC-1 cells in normal untreated mice compared to NK-depleted mice. After intracerebral inoculation, however, no difference in remaining radioactivity was observed between untreated and NK-depleted mice. This indicates that selective NK-mediated elimination of tumor cells might occur after intravenous or subcutaneous but not after intracerebral inoculation.(ABSTRACT TRUNCATED AT 250 WORDS)
