Long non-coding RNAs (lncRNAs) play vital roles in the pathogenesis and development of multiple cancers, including colorectal cancer (CRC). Nevertheless, the regulatory mechanisms of LINC01278 in CRC remain unknown. Our research aims to identify the regulatory mechanisms of LINC01278 in CRC.The expression of LINC01278 was examined by quantitative real-time polymerase chain reaction (RT-qPCR). StarBase and TargetScan websites were used to predict the interaction between miR-134 and LINC01278 or KDM2A, which was further confirmed by Dual-Luciferase reporter assay and RNA immunoprecipitation (RIP) assay. Cell viability, migration, and invasion were detected by Cell Counting Kit-8 (CCK-8) and transwell assays.LINC01278 was upregulated in CRC tissues and cell lines, and knockdown of LINC01278 suppressed CRC cell progression. In addition, LINC01278 inhibited miR-134 expression by direct interaction, and the inhibition of miR-134 abolished the suppressive effects of LINC01278 knockdown on viability, migration, and invasion of CRC cells. Furthermore, KDM2A was confirmed to be a target gene of miR-134. Overexpression of KDM2A facilitated the tumorigenesis of CRC, while this effect was reversed by the upregulation of miR-143. Finally, it was demonstrated that miR-134 inhibitor reversed the shLINC01278‑mediated inhibitory effect on KDM2A expression.Our study demonstrated that LINC01278 upregulated KDM2A to promote CRC progression by interacting with miR-143, suggesting that LINC01278 might be a new therapeutic target of CRC.

To explore the clinical significance of circRNF20 in non-small-cell lung carcinoma (NSCLC), and its regulatory effects on NSCLC cell functions by activating MAPK9.Relative levels of circRNF20 and MAPK9 in NSCLC tissues were detected by quantitative Real Time-Polymerase Chain Reaction (qRT-PCR). The relationship between circRNF20, MAPK9 and pathological factors in NSCLC patients was analyzed. Prognostic potentials of circRNF20 and MAPK9 in NSCLC were assessed by Kaplan-Meier method. The interaction between circRNF20 and MAPK9 was tested by Dual-Luciferase reporter assay. Regulatory effects of circRNF20 and MAPK9 on proliferative abilities in H358 and SPC-A1 cells were examined by Cell Counting Kit-8 (CCK-8) and colony formation assay.CircRNF20 and MAPK9 were upregulated in NSCLC tissues than normal ones. They were correlated to T stage and poor prognosis in NSCLC patients, while their levels were unrelated to gender, age, and incidences of lymphatic and distant metastasis. Knockdown of circRNF20 attenuated proliferative abilities in H358 and SPC-A1 cells. On the contrary, the overexpression of MAPK9 yielded the opposite results. MAPK9 was the target gene binding circRNF20, which was able to reverse the regulatory effect of circRNF20 on NSCLC proliferation.CircRNF20 and MAPK9 are upregulated in NSCLC cases, which are closely linked to T stage in NSCLC patients. They are independent prognostic factors for NSCLC. By activating MAPK9, circRNF20 stimulates NSCLC proliferation.

This study aims to explore the role of parecoxib in improving postoperative cognitive dysfunction (POCD) in rats, and to investigate the possible underlying mechanism.60 Sprague Dawley (SD) rats were randomly divided into the control group (n=20), the model group (n=20), and the para group (n=20). The Morris water maze test was conducted to detect the postoperative cognitive ability of rats. The hematoxylin-eosin (HE) staining was applied to observe the neuronal density in the hippocampus of rats. The TdT-mediated dUTP Nick-End Labeling (TUNEL) was used to detect the hippocampal apoptosis in rats. Meanwhile, relative levels of inflammatory factors in rat hippocampus were detected by enzyme-linked immunosorbent assay (ELISA), including interleukin-1β (IL-1β), IL-6, tumor necrosis factor-α (TNF-α), and prostaglandin E2 (PGE2). Furthermore, Western blot was conducted to detect the protein levels of cyclooxygenase-2 (COX-2), PGE2, EP1, and p-Akt in rat hippocampus.Behavioral test results showed that the escape latency of rats in the para group was significantly shorter than that of the model group (p0.05). Compared with the model group, rats in para group showed significantly longer time in target quadrant and more times across the platform, as well as higher Tn/Tn+Tf (p0.05). In the control group, pathological changes of hippocampal neurons were slighter with rare apoptotic cells. Rats in model group showed great pathological lesions with abundant apoptotic neurons, which were markedly alleviated in the para group (p0.05). Meanwhile, ELISA showed that the levels of IL-1β, IL-6, TNF-α, and PGE2 in the para group were remarkably lower than those of the model group (p0.05). Expression levels of COX-2, PGE2, and EP1 in the para group were significantly lower than those of the model group (p0.05). However, the expression level of p-Akt was significantly higher than the model group (p0.05).Parecoxib improves the cognitive function of POCD rats via inhibiting COX-2 overexpression in rat brain.

The correlation of the embryo matrix metalloproteinase-9 (MMP-9) secretion, embryonic development and clinical pregnancy was investigated.The embryo culture from in vitro fertilization-embryo transfer (IVF-ET) patients were collected in the Xuzhou Central Hospital from January 2013 to September 2014. At the same time, the embryo grade was recorded. The secretion of the MMP-9 in the embryo culture was detected through hybridization (Dot-blot) and enzyme-linked immunosorbent assay (ELISA). The clinical pregnancy outcome was followed after one month of the embryo transfer.With the embryonic development from 2-cell to 8-cell, embryonic MMP-9 secretion increased gradually (p0.05). Though 8/I embryo, the secretion of MMP-9 are not identical. The quantitative detection of the MMP-9 secretion of the 8-cell embryo by ELISA is higher as is the embryo score, but the low secretion of embryo score is lower (p0.001). As 8/I embryos, the clinical pregnancy rate (77.3%) of the MMP-9 high secretion embryos is higher than the low secretion embryos (16.7%) (p0.001).The secretion of the embryonic MMP-9 is closely related to the quality of the embryo and embryo implantation. It is speculated that MMP-9 may become one of the criteria to evaluate the quality of the embryos.

Meningiomas represent one-third of all primary brain tumors and cause 35,000 new cases each year. Because of this high incidence, we sought to determine if there are proteomic differences between meningiomas and neighboring tissues. Two-dimensional gel electrophoresis and mass spectrometry were used to detect differentially expressed proteins in tumor samples, using arachnoid tissue as a control. Western blot analysis was used to validate the identified candidate proteins. We obtained quantitative data on 112 proteins, 17 of which were down-regulated and 26 of which were up-regulated in meningiomas relative to normal arachnoid tissue. Our analysis showed that the expression of galectin-3, vimentin, and endoplasmin was decreased significantly in meningiomas. The expression of 40S ribosomal protein S12, glutathione S-transferase P, and hypoxia up-regulated protein 1 was increased significantly (P < 0.05). The six above-mentioned differentially expressed proteins might be closely involved with the development of meningiomas. The results of this study provide basic insights into the proteome of meningiomas and provide a preliminary database for further research to enhance understanding of meningioma development.

Background: Henoch Schonlein Purpura (HSP) is a systemic vasculitic disease which is common in children. It is very important to understand the clinical features of this disease for doctors and nurses. Objectives: To study the clinical characteristics of HSP in children. Methods: Collect the clinical data of the HSP children, and analyze the clinical characteristics of these HSP patients. Results: The ratio of M:F was 1.9:1. The mean age was 6.6 ± 1.6 years. The typical onset seasons were spring, winter and autumn. Infection and food allergy were the main etiological factors. The first symptom was skin purpura and these purpura mainly concentrated the lower extremities and buttocks. The dominant digestive clinical features were abdominal pains and vomiting. The knee joint and ankle joint were most frequently affected. The typical kidney symptoms were microscopic hematuria and albuminuria. An increased ESR was reported in 68 patients (56.7%). Serum C3 decreased in 13 cases (10.8%). ASO titer was higher in 57 children (47.5%). Conclusion: There were gender, season and area differences for the HSP patients. The etiological factors were diverse. HSP patients could have various clinical symptoms and rare complications. Key words: Henoch Schonlein Purpura, Children

To identify the potential role of macrophage inflammatory protein-1α (MIP-1α) with its C-C chemokine receptor 5 (CCR5) in epileptogenic brain injury, we examined their expression in juvenile rat hippocampus and explored the potential link between MIP-1α, CCR5 and neuropathological alterations after status epilepticus (SE) induced by intracerebroventricular (i.c.v.) kainic acid (KA) injection.Based on the determination of the development of spontaneous seizures initiated by SE in developing rat brain, we firstly examined hippocampal neurone damage through Nissl and Fluoro-Jade B staining, and evaluated microglial reaction during the early phase following KA-induced SE in 21-day-old rats. MIP-1α and CCR5 protein were quantified by ELISA and Western blot respectively following mRNA by real-time PCR. We also mapped MIP-1α and CCR5 expression in the hippocampus by immunohistochemistry and identified their cellular sources using double-labelling immunofluorescence.In juvenile rats, KA caused characteristic neurone damage in the hippocampal subfields, with accompanying microglial accumulation. In parallel with mRNA expression, MIP-1α protein in hippocampus was transiently increased after KA treatment, and peaked from 16 to 72 h. Double-labelling immunofluorescence revealed that MIP-1α was localized to microglia. Up-regulated CCR5 remained prominent at 24 and 72 h and was mainly localized to activated microglia. Further immunohistochemistry revealed that MIP-1α and CCR5 expression were closely consistent with microglial accumulation in corresponding hippocampal subfields undergoing degenerative changes.Our data indicated that MIP-1α as a regulator, linking with the CCR5 receptor, may be involved within the early stages of the epileptogenic process following SE by i.c.v. KA injection.

Hepatitis B virus (HBV) is the major cause of chronic hepatitis,cirrhosis, and hepatocellular carcinoma. The global epidemiological scenario of HBV infection has been changing rapidly over the last two decades due to an effective immunization programme initiated by the World Health Organization. The objective of this study is to estimate the prevalence of HBV in apparently adult people who were taken health examination in our Hospital.A cross-sectional seroprevalence analysis of hepatitis B virus infection was performed in 12037 adult residents (agedor =18 years) in Chengdu, who visited Health Examination Center of West China Hospital of Sichuan University for routine medical check-up during the period from March to December 2008. A structured medical form was used to collect data on demographic characteristics and risk factors. ELISA was used to test sera for HBV markers. Descriptive and logistic regression models were used for analysis.A total of 12,037 urban residents were involved. Prevalence of positive HBsAg was 6.1%, lower than the level of national seroepidemiological survey in (7.18%). Among HBsAg negative people, anti-HBs and anti-HBc was 60.2% and 13.6% respectively. There was a maximum between 18 to 29 years of age (61.8%) in anti-HBs positive people. Multivariate conditional logistic regressive analysis showed that, except for blood and vertical transmission, factors of male gender (OR, 1.876; 95% CI, 1.519-2.316; P0.001) and alcohol intake (OR, 0.689; 95% CI, 0.571-0.832; P0.001) were associated with a higher risk of positive HBsAg.Among the medical examination people in Chengdu, HBsAg positive rate was lower than the national general population, the epidemiological characteristics of hepatitis B has been changed, because of vacination policies to the newborn; therefore, the necessity to continue to carry on the vaccination program.

The aim of this study was to explore the effects and mechanisms of Icarisid II (ICA-II) on enhancing the cellular cGMP in rat corpus cavernosum tissue (RCCT). Diabetes mellitus Wistar rats were induced by streptozotocin, and diabetic ED rats were selected for the RCCT culture by apomorphine. ICA-II was extracted and purified from Icariin (ICA) by enzymatic method. The RCCT was treated with ICA-II, ICA and Sildenafil at different concentrations. cGMP and nitric oxide synthase (NOS) activities were checked respectively by enzyme immunoassay. Meanwhile, nNOS, iNOS and eNOS in RCCT were checked by western blot. ICA-II evaluated the intracellular cGMP to 8.01 ± 1.02 pmol mg(-1) min(-1), which is much weaker than that from Sildenafil (12.4 ± 1.16 pmol mg(-1) min(-1)) (P0.05). There is no significant difference between ICA-II and ICA. With the treatment of 10 μm ICA-II for 24 and 48 h, nNOS expression was significantly increased in RCCT (P0.05), while the eNOS expression level was very low without any change. Notably, ICA-II increased the intracellular NOS activity significantly in vitro in RCCT. Except the PDE5 inhibitory effect, ICA-II increases the intracellular cGMP through the enhancement of nNOS expression and NOS activity in RCCT in vitro. ICA-II implies a potential compound for neurogenic erectile dysfunction by NO-cGMP pathway.

28 adult Min sows (3-7 parities), represented 9 sire families were selected to conduct the continual blood collection from the tail for two hours at the interval of ten minutes between 9:00-11:00 in the morning on the sixth day after the last mating. The content of progesterone was assayed by radio-immunological assay. The amplitudes of the secretion of progesterone were generated by the procedure of HORM fft. exe complied by the author. It is difficult to get a reasonable conclusion from the content analysis, because the content varied widely between different points of blood collection in single sow. After Fourier conversion, the heritability of amplitude for the fifth partial wave was high (h2 = 0.932); middle for the basal, 2nd and 3rd partial waves; lower for others. This supported the idea that prolific sows had more active secretion of progesterone in luteal cells. The coexist of low and high heritability components in amplitude of progesterone secretion showed that the activities of progesterone secretion were influenced by both the genetics and environments. The genetic correlation of basal, 1st, 5th partial waves were negative, the others were positive, with the litter size and litter size alive. It showed that the reproductive performance of Min pigs were not expressed perfectly, as the balance of secretion of progesterone did not reach the best. The genetic correlation of all partial waves with duration of estrus was negative which supported that the progesterone suppresses the estrus. The genetic correlation of 3rd, 5th partial waves were stronger than others, but selection for 5th partial wave would make high responses than the 3rd wave, because the genetic correlation of 3rd partial wave with litter size and litter size alive was positive, therefore, the more appropriats method of selection for progesterone was adopted, the better results would be achieved in the improvement of indirect selection response for litter size.

Postoperative reflux esophagitis is a common complication after partial removal of the esophagus and the whole gastric cardia for reconstruction through esophagogastrostomy. In 10 cases, the lower esophageal sphincter (LES) was preserved for a length of 2.05 +/- 0.33cm (mean +/- S) during partial esophagectomy for benign or malignant lesion at the middle and lower portion of the thoracic esophagus. The mean value of the LES pressures measured two weeks after operations was 2.40 +/- 0.64 kPa (18 +/- 4.8 mmHg) and the postoperative X-ray barium meal examination revealed no evidence of gastroesophageal reflux. Our study suggested that the preserved LES should effectively act as a functional barrier against the development of reflux esophagitis.

167 cases of amyotrophic lateral sclerosis treated in Peking Union Medical College Hospital were analyzed. There was a significant difference between some of the clinical features of amyotrophic lateral sclerosis in China and western countries. The age of onset in China was 10 to 20 years earlier than and the male to female ratio (4.6:1) was 2 times higher than those in the west. Average age of death was 47 years, being earlier than that in the west. Some specific clinical features for early clinical diagnosis were also discussed.

Upper esophageal sphincter dysfunction is a diseases of a altered esophageal motility characterized by muscular functional obstruction with cervical dysphagia as the main clinical manifestation. It is complicated sometimes by incidental aspiration. Pulmonary infection and even choking. The cricopharyngeal anatomically acts as the upper esophageal sphincter Uncoordinated contraction between cricopharyngeal and pharyngeal muscle could be one of the mechanisms for the development of cervical dysphagia. Cricopharyngeal myotomy has been used to treat functional disorder of the upper esophagus. The rationale for such procedure is to relieve functional obstruction by producing a wide "Opening door" for the upper esophagus. This paper presents 5 cases suffering from severe dysphagia treated with above mentioned procedure. The result is satisfactory that all patients became symptom. Free another female patient who did not undergo the operation has died of sudden choking. Authors concluded that cricopharyngeal myotomy is a simple but effective procedure for treating upper esophageal sphincter dysfunction.

Nasopharyngeal carcinoma (NPC) is etiologically closely connected to the Epstein Barr virus. NPC is the only carcinoma in man that can be diagnosed serologically. In the serum of patients with NPC characteristic antibody titres of certain viral antigens can be demonstrated during the disease. Conclusions regarding the success of treatment and on possible relapse can be deduced from these antibody titres. Criteria of histological differention for NPC prognosis are described, and the possible mode of infection is discussed. Since the disease also occurs in adolescents the treatment of a typical case of NPC is described.
