The present study involves the development of citric acid-cross-linked carboxymethyl cellulose (C3CA) scaffolds by a freeze-drying process. Scaffolds were fabricated at different freezing temperatures of -20, -40, or -80 °C to investigate the influence of scaffold pore size on bone regeneration. All three scaffolds were porous in structure, and the pore size was measured to be 74 ± 4, 55 ± 6, and 46 ± 5 μm for -20, -40, and -80 °C scaffolds. The pores were larger in scaffolds processed at -20 °C compared to -40 and -80 °C, indicating the reduction in pore size of the scaffolds with a decrease in freezing temperature. The cytocompatibility, cell proliferation, and differentiation in C3CA scaffolds were assessed with the Saos-2 osteoblast cell line. These scaffolds supported the proliferation and differentiation of Saos-2 cells with significant matrix mineralization in scaffolds processed at -40 °C. Subcutaneous implantation of C3CA scaffolds in the rat model was investigated for its ability of vascularization and new matrix tissue formation. The matrix formation was observed at the earliest of 14 days in the scaffolds when processed at -40 °C while it was observed only after 28 days of implantation with the scaffolds processed at -20 and -80 °C. These results suggest that the citric acid-cross-linked CMC scaffolds processed at -40 °C can be promising for bone tissue engineering application.

Inhibition of collagen fibrillation by small molecules is of growing interest to develop therapeutics for the illnesses related to excess deposition of collagen. In this context, we have studied the inhibitory effect of N-Vanillylnonanamide (NVA), a natural product from capsicum oleoresin and an analog of capsaicin (a known inhibitor of collagen fibrillation), on collagen self-assembly that leads to fibrillation in vitro. Commercially, capsaicin was found to be expensive than NVA. Therefore, it would be an advantage economically if NVA could display a similar/better inhibitory activity compared to capsaicin. The conventional turbidity measurements indicate that NVA completely inhibits collagen fibrillation at body temperature (37 °C) and its inhibition were concentration-dependent. The inhibition efficiency was observed to reduce at room temperature (25 °C). NVA protects the triple helical structure of collagen while it increases the thermal stability of collagen compared to collagen alone. Fluorescence results suggest that NVA binds in both telopeptide and triple helical regions of collagen and thereby prevents collagen self-assembly. The present results thus indicate that NVA is a potential inhibitor and, economically, it could be a better choice as a therapeutic agent compared to capsaicin in evolving treatment for disorders associated with excessive collagen deposition.

Salt-based preservation of hides/skins contributes to about 50% total dissolved solids (TDS) in tannery wastewaters. In this study, raw skins have been preserved by exposing them to a continuous flow of supercritical carbon dioxide (SCCO2) in a pressurized reactor. The process was carried out in reactors of two different capacities to ensure scalability. The skins thus dried could be stored at room temperature for a period of 30 days. The SCCO2–dried skins were less conducive for microbial growth than wet-salted skins. The soak liquor of SCCO2–dehydrated skin showed a 90% reduction in chloride content and significantly lower BOD and COD levels than soak liquor from wet-salted skins. The leathers produced from SCCO2-preserved skins and wet-salted skin had no significant quality differences. As SCCO2 systems have been reported as alternatives to all other unit operations, establishing SCCO2–based preservation will complete the circle of total leather manufacture with SCCO2.

Zinc Oxide (ZnO) nanoparticles are used in the field of nanomedicine. ZnO is non-toxic and compatible with biological system. The major problem of wound is infections. The infections delays wound healing. Usage of Nanomaterials like ZnO nanoparticles for wound healing is promising. ZnO nanoparticles are synthesised by wet chemical method. It is annealed at 300, 500, 700, and 900 °C for 3 h. ZnO nanoparticle morphology and size distribution are analysed by XRD, FTIR and TEM. The influences of particle size on fibroblast cell growth (NIH3T3) and on antimicrobial activity are analysed. The growth of fibroblast cell is higher with ZnO nanoparticles of larger particle size. Further the MTT analysis, fluorescence calcein AM cell staining and F-actin analysis confirms the growth of fibroblast cells with improved biocompatibility behaviour. The antimicrobial activity is higher for ZnO nanoparticles of lower particle size. Confocal microscope analysis and Zone of inhibition analysis confirms the antimicrobial activity of ZnO nanoparticles.

Biomimetic nanofibrous scaffolds targeting multiple dysfunctional processes provide a multi-pronged strategy to restore functions and regenerate the damaged tissue. This study investigates a strategy of combining a regenerative component, Type I collagen Peptide (CP), along with a nitric oxide donor, S-Nitrosoglutathione (GSNO), in the form of nanofibrous scaffold to address the non-healing diabetic ulcer. Silk Fibroin-Polyvinyl alcohol (SF-PVA) nanofibrous scaffold is used as a carrier for delivering functional moieties. The developed nanofibrous electrospun mats (SF-PVA, CP-SF-PVA, and CP-GSNO-SF-PVA) showed continuous, bead-less and randomly oriented fibers with highly porous morphology. The in vitro biocompatibility was assessed by MTT assay, DAPI-Rhodamine 123 and FITC-Phalloidin imaging studies. CP-GSNO-SF-PVA nanofibrous scaffold showed a high degree of cell attachment, spreading of F-actin with viable cell morphology and appreciable inter-cellular connection. Thus the study showed that the proliferation of fibroblast cells are mainly facilitated by the presence of collagen peptide in the nanofibrous matrix. Griess assay demonstrated immediate release of NO for a day from the developed multifunctional scaffold. These results demonstrate the in vitro efficacy of CP-GSNO and indicate the opportunity of CP-GSNO-SF-PVA nanofibrous scaffold for the treatment of ischemic non-healing ulcers.

Collagen is a unique protein abundantly present in the connective tissues of mammals and widely used for biomaterial preparation. In this study, we synthesized and characterized collagen-fucoidan blend films for tissue regenerative properties. Thermogravimetric analysis (TGA) and Differential Scanning Calorimetry (DSC) were used for thermal analysis of the blend films, and the films exhibited higher thermal stability and denaturation temperature (Td) than those of native collagen due to intramolecular hydrogen bonding interaction between collagen and fucoidan, which was analyzed by FTIR spectroscopy. Morphological evaluation of these films using Scanning Electron Microscopy (SEM) showed smaller pore size than the control. Moreover, fucoidan protects collagen against enzymatic degradation and thereby increases the structural stability of collagen. Further, the in vitro studies of the synthesized films showed that they effectively facilitated the proliferation and migration of fibroblast cells without exhibiting toxicity. These study results suggested that the collagen-fucoidan blend films are a favorable substrate for growth of fibroblast cells, and may have great potential for tissue engineering applications.

Bioactive compounds are extracted from mekmeko and characterized for antioxidant, phytochemical, and antibacterial activities. The phytochemical studies of the extracts revealed the presence of condensed tannins, flavonoid, and phenolic compounds. Radical scavenging activity of the extract demonstrates that the ethanol extract has more significant antioxidant activity than the aqueous extract. Ethanolic extract of mekmeko is more effective against gram-positive bacteria compared to gram-negative. A Box–Behnken-design was used to study the response pattern and to determine the optimum combination of variables for maximization of extraction yield. Experiments were carried out to evaluate the four independent variable effects (particle size, temperature, time, and solid–solvent-ratio) on the maximum extraction yield of bioactive compounds. Second order polynomial mathematical model was developed with a high coefficient of determination ( R 2 = 0.99) from the experimental data. A maximum of 18.85% total extraction yield was achieved at the optimal conditions of particle size of 350 mesh, extraction temperature at 41 °C, solid–solvent-ratio of 0.07, and extraction time of 24 h. The extracted constituents are predominantly phenolics (39%) and condensed tannins (21%). Under theses optimal conditions, the experimental values agreed well with the predicted values and indicated the suitability of the developed models.

Ulcerative colitis, particularly the chronic persistent form is characterized by the presence of active inflammation and extensive areas of ulceration in the colonic mucosa. The existing treatment protocol aims at only reducing intestinal inflammation, rather than targeting mucosal ulceration. In this study, type I collagen and its daughter peptides called collagen hydrolysate, highly popular reconstructive materials for tissue engineering applications, are hypothesized as healing matrices to target the recuperation of internal mucosal ulceration. The clinical assessments on day 10 of dextran sodium sulfate induced colitis in mice model revealed that both the collagen (1.56±0.29) and collagen hydrolysate treatments (1.33±0.33) showed a significant reduction in the rectal bleeding compared to the reference mesalamine treatment (2.50±0.33) and untreated negative control (2.40±0.40). VEGF, a potent angiogenic growth factor, over expressed during UC was down-regulated by collagen hydrolysate (1.06±0.25) and collagen (1.76±0.45) to a greater extent than by mesalamine (2.59±0.51) and untreated control (4.17±0.15). The down-regulation of proinflammatory cytokines such as TNF-α, IL-1β, and IL-6 also follows the same pattern. Histological observations were in accordance with the clinical indicators. Both collagen and collagen hydrolysate treatments showed significant reduction in mucosal damage score and facilitated faster regeneration of damaged mucosa.

Human life expectancy has been steadily increasing at a rapid rate, but this increasing life span also brings about increases in diseases, dementia, and disability. A global burden of disease 2010 study revealed that hip and knee osteoarthritis ranked the 11th highest in terms of years lived with disability. Wear and tear can greatly influence the quality of life during ageing. In particular, wear and tear of the articular cartilage have adverse effects on joints and result in osteoarthritis. The articular cartilage uses longevity of type II collagen as the foundation around which turnover of proteoglycans and the homeostatic activity of chondrocytes play central roles thereby maintaining the function of articular cartilage in the ageing. The longevity of type II collagen involves a complex interaction of the scaffolding needs of the cartilage and its biochemical, structural and mechanical characteristics. The covalent cross-linking of heterotypic polymers of collagens type II, type IX and type XI hold together cartilage, allowing it to withstand ageing stresses. Discerning the biological clues in the armamentarium for preserving cartilage appears to be collagen cross-linking. Therapeutic methods to crosslink in in-vivo are non-existent. However intra-articular injections of polyphenols in vivo stabilize the cartilage and make it resistant to degradation, opening a new therapeutic possibility for prevention and intervention of cartilage degradation in osteoarthritis of aging.

Background: Deformities and neuropathic chronic ulcers are the common features associated with leprosy-cured individuals that impact their quality of life and impair rehabilitation efforts. The challenging aspects for treatment of chronic wounds are the factors that inhibit healing. We reasoned that limited success of various therapeutic interventions could be due to the fact that leprosy-cured individual's physiology gets acclimatized to having a chronic wound that any therapeutic intervention is counterbalanced to maintain status quo at the wound site. Therefore, an alternative strategy would be to use biomaterials that gradually alter the wound site allowing the individual's physiology to participate in the healing process. Aims: Developing the human amnion (Amn)-derived biomaterial scaffolds and evaluating its use to heal chronic wounds in leprosy-cured but deformed persons (LCDPs). Materials and Methods: Using an enzymatic protocol, we have developed a rapid method to generate biomaterial scaffolds from discarded human Amn. A clinical trial on 26 LCDPs was performed with the biomaterial, and its wound-healing potential was then compared with LCDPs undergoing standard treatment procedure. Results: Biomaterial-based treatment of chronic wounds on LCDP displayed a higher efficiency in healing when compared to standard treatment. Conclusions: This study exemplifies that biomaterial-based treatment of leprosy-wounds offers an excellent affordable alternative for wound management. This study underlines the importance of involving both local wound environment and systemic effects for healing. In addition, we highlight wound healing as a necessity for successful rehabilitation and reintegration of leprosy-cured person into the society.

Matrix metalloproteinases (MMPs) have been investigated as a potential target for treating invasive breast cancers. The chemotherapy for breast cancer is often prescribed as a combination of drugs. The present study investigates a novel strategy of combining a MMP inhibitor, Epigallocatechin gallate (EGCG), along with an anticancer drug, Paclitaxel (PTX), in the form of a liposomal co-delivery system. The developed PTX/EGCG co-loaded liposomes showed an entrapment of 77.11 � 2.30% and 59.11 � 3.51% for PTX and EGCG, respectively. The in vitro efficacy of the liposomes was assessed by their ability to promote apoptosis and curtail cell invasion. On all parameters, namely cytotoxicity and caspase-3 activity that are indicators of apoptosis, and MMP-2 and - 9 inhibition and invasion assays that are indicators of cell invasion, the PTX/EGCG co-loaded liposomes showed better results than each of the individual drug loaded liposomes. These findings demonstrate the synergistic outcome of PTX/EGCG combination and indicate the suitability of PTX/EGCG co-loaded liposomes for the treatment of invasive breast cancer. � 2014 Elsevier B.V.

Excess accumulation of collagen (fibrosis) is the hallmark of many fibrotic diseases such as keloids, hypertrophic scars, etc. The inhibition of collagen fibrillation during its accumulation is a therapeutic way to limit the fibrosis. Herein, the effect of Ferulic acid (FA), a natural phenolic acid compound, on collagen fibrillation is studied using biophysical methods. Optical density (OD) and microscopic analysis indicate that FA inhibits collagen self-association, and the inhibitory efficiency depends on the concentration and temperature. The absence of an increase in OD for matured collagen fibrillar solution upon addition of FA followed by collagen solution indicates that FA could also terminates the progression of preformed collagen fibrils. Spectroscopic measurements indicate that collagen retains its unique triple helical structure in the presence of FA. Saturation Transfer Difference NMR suggests that FAs are in proximity to collagen while fluorescence quenching upon addition of FA proposes that FA most likely binds to the telopeptide regions of collagen. Enzymatic studies suggest that FA protects collagen from enzymatic degradation. The current study demonstrates that FA is a potential inhibitor of collagen fibrillation and its propagation and thus it can be considered for therapeutic studies to make FA as a remedy for the plaques related to collagen deposition.

Excess accumulation of collagen (fibrosis) undergoes self-aggregation, which leads to fibrillar collagen, on the extracellular matrix is the hallmark of a number of diseases such as keloids, hypertrophic scars, and systemic scleroderma. Direct inhibition or disintegration of collagen fibrils by small molecules offer a therapeutic approach to prevent or treat the diseases related to fibrosis. Herein, the anti-fibrotic property of Glucono-δ-lactone (GdL), known as acidifier, on the fibrillation and its disintegration of collagen was investigated. As collagen fibrillation is pH dependent, the pH modulation property of GdL is attractive to inhibit self-association of collagen. Optical density and microscopic data indicate that GdL elicits concentration-dependent fibril inhibition and also disintegrates pre-formed collagen fibrils. The simultaneous pH analysis showed that the modulation(lowering) of pH by GdL is the primary cause for its anti-fibrotic activity. The intact triple helical structure of collagen upon treatment of GdL suggests that collagen fibril disintegration can be achieved without affecting the native structure of collagen which is essential for any anti-fibrotic agents. Saturation transfer difference (STD) NMR result reveals that GdL is in proximity to collagen. The present results thus suggest that GdL provides a lead to design novel anti-fibrotic agents for the pathologies related to collagen deposition.

The potential of multifunctional wound heal biomaterial relies on the optimal content of therapeutic constituents as well as the desirable physical, chemical, and biological properties to accelerate the healing process. Formulating biomaterials such as amnion or collagen based scaffolds with natural products offer an affordable strategy to develop dressing material with high efficiency in healing wounds. Using image based phenotyping and quantification, we screened natural product derived bioactive compounds for modulators of types I and III collagen production from human foreskin derived fibroblast cells. The identified hit was then formulated with amnion to develop a biomaterial, and its biophysical properties, in vitro and in vivo effects were characterized. In addition, we performed functional profiling analyses by PCR array to understand the effect of individual components of these materials on various genes such as inflammatory mediators including chemokines and cytokines, growth factors, fibroblast stimulating markers for collagen secretion, matrix metalloproteinases, etc., associated with wound healing. FACS based cell cycle analyses were carried out to evaluate the potential of biomaterials for induction of proliferation of fibroblasts. Western blot analyses was done to examine the effect of biomaterial on collagen synthesis by cells and compared to cells grown in the presence of growth factors. This work demonstrated an uncomplicated way of identifying components that synergistically promote healing. Besides, we demonstrated that modulating local wound environment using biomaterials with bioactive compounds could enhance healing. This study finds that the developed biomaterials offer immense scope for healing wounds by means of their skin regenerative features such as anti-inflammatory, fibroblast stimulation for collagen secretion as well as inhibition of enzymes and markers impeding the healing, hydrodynamic properties complemented with other features including non-toxicity, biocompatibility, and safety.

Missense mutations, which replace one Gly with a larger residue in the repeating sequence of the type I collagen triple helix, lead to the hereditary bone disorder osteogenesis imperfecta (OI). Previous studies suggest that these mutations may interfere with triple-helix folding. NMR was used to investigate triple-helix formation in a series of model peptides where the residue replacing Gly, as well as the local sequence environment, was varied. NMR measurement of translational diffusion coefficients allowed the identification of partially folded species. When Gly was replaced by Ala, the Ala residue was incorporated into a fully folded triple helix, whereas replacement of Gly by Ser or Arg resulted in the presence of some partially folded species, suggesting a folding barrier. Increasing the triple-helix stability of the sequence N-terminal to a Gly-to-Ser replacement allowed complete triple-helix folding, whereas with the substitution of Arg, with its large side chain, the peptide achieved full folding only after flexible residues were introduced N-terminal to the mutation site. These studies shed light on the factors important for accommodation of Gly mutations within the triple helix and may relate to the varying severity of OI.

Collagenase enzymes are nontoxic and eco-friendly biocatalysts. Dyeing is an important process in the leather industry, which employs many synthetic colorants. Many good dyes suffer from incomplete exhaustion and this causes concern, as the biotreatability of the unexhausted dyes in effluent is normally difficult. Hence in the present study, an attempt has been made to improve the exhaustion of dyes by using bacterial collagenase enzymes as biocatalysts. The effect of process parameters of enzymatic treatment such as pH, temperature and duration on the exhaustion of the dye, levelness of dyeing, shade brightness, dye penetration and color intensity have been studied and the conditions are optimized. Uptake of dye as high as 99% has been observed by the treatment of collagenase. The change in shades due to enzymatic treatment has been quantified by reflectance measurements and compared with the visual assessment data. Scanning electron microscopy analysis showed a well opened-up fibre matrix for the collagenase treated leather. The strength properties are not significantly altered and the bulk properties like softness have been found to be improved by the use of collagenase.

The central role of collagen as the major structural fibrous protein in the mammalian extracellular matrix has motivated a significant effort toward the determi- nation of its mechanical properties at all levels, ranging from single monomers and long-chain polymers to a structural element within a biological tissue. However, the stabiliza- tion of collagen against collagenolytic degradation finds sig- nificance in biomedical and industrial applications. Tannins are plant-derived polyphenols that have the ability to inhibit the collagenase activity at minimum concentration. The in- hibitory effect of wattle (Acacia mollissima) and myrobalan (Terminalia chebula) on the action of collagenase against colla- gen was probed in this study. The kinetics of the inhibition of collagenase by wattle and myrobalan was deduced from the extent of hydrolysis of 2-furanacryloyl-L-leucyl-glycyl- L-prolyl-L-alanine. Both wattle and myrobalan tannin exhib- ited competitive modes of inhibition against collagenase. Circular dichroism studies of collagenase on treatment with wattle and myrobalan revealed changes in the secondary structure of collagenase. These results suggest that the tan- nins of A. mollissima and T. chebula extracts facilitated colla- gen stabilization through collagenase inhibition. 2007

In the recent past, keratin has been fabricated into different forms of biomaterials like scaffold, gel, sponge, film etc. In lieu of the myriad advantages of the hydrogels for biomedical applications, a keratin-silica hydrogel was fabricated using tetraethyl orthosilicate (TEOS). Textural analysis shed light on the physical properties of the fabricated hydrogel, inturn enabling the optimization of the hydrogel. The optimized keratin-silica hydrogel was found to exhibit instant springiness, optimum hardness, with ease of spreadability. Moreover, the hydrogel showed excellent swelling with highly porous microarchitecture. MTT assay and DAPI staining revealed that keratin-silica hydrogel was biocompatible with fibroblast cells. Collectively, these properties make the fabricated keratin-silica hydrogel, a suitable dressing material for biomedical applications.

Inhibitory effect of green tea polyphenols viz., catechin and epigallocatechin gallate (EGCG) on the action of collagenase against collagen has been probed in this study. Catechin and EGCG treated collagen exhibited 56 and 95% resistance, respectively, against collagenolytic hydrolysis by collagenase. Whereas direct interaction of catechin and EGCG with collagenase exhibited 70 and 88% inhibition, respectively, to collagenolytic activity of collagenase against collagen and the inhibition was found to be concentration dependent. The kinetics of inhibition of collagenase by catechin and EGCG has been deduced from the extent of hydrolysis of (2-furanacryloyl-L-leucyl-glycyl-L-prolyl-L-alanine), FALGPA. Both catechin and EGCG exhibited competitive mode of inhibition against collagenase. The change in the secondary structure of collagenase on treatment with catechin and EGCG has been monitored using circular dichroism spectropolarimeter. CD spectral studies showed significant changes in the secondary structure of collagenase on treatment with higher concentration of catechin and EGCG. Higher inhibition of EGCG compared to catechin has been attributed to the ability of EGCG to exhibit better hydrogen bonding and hydrophobic interaction with collagenase.

Collagen, a unique connective tissue protein finds extensive application as biocompatible biomaterial in wound healing, as drug carriers, cosmetics, etc. A work has been undertaken to study the stabilization of type I collagen using the plant polyphenol catechin. Catechin treated collagen fibres showed a shrinkage temperature around 70 degrees C implying that catechin is able to impart thermal stability to collagen. Catechin treated collagen fibres has been found to be stable even after treatment with high concentration of the secondary structural destabilizer, urea. Circular dichroism studies revealed that there is no major alteration in the structure of collagen on treatment with catechin. The study has demonstrated the involvement of hydrogen bonding and hydrophobic interactions as the major forces involved in the stabilization of collagen by the plant polyphenol, catechin.

Arthritic diseases, such as osteoarthritis and rheumatoid arthritis, inflict an enormous health care burden on society. Osteoarthritis, a degenerative joint disease with high prevalence among older people, and rheumatoid arthritis, an autoimmune inflammatory disease, both lead to irreversible structural and functional damage to articular cartilage. The aim of this study was to investigate the effect of polyphenols such as catechin, quercetin, epigallocatechin gallate, and tannic acid, on crosslinking type II collagen and the roles of these agents in managing in vivo articular cartilage degradation. The thermal, enzymatic, and physical stability of bovine articular cartilage explants following polyphenolic treatment were assessed for efficiency. Epigallocatechin gallate and tannic acid-treated explants showed >12 °C increase over native cartilage in thermal stability, thereby confirming cartilage crosslinking. Polyphenol-treated cartilage also showed a significant reduction in the percentage of collagen degradation and the release of glycosaminoglycans against collagenase digestion, indicating the increase physical integrity and resistance of polyphenol crosslinked cartilage to enzymatic digestion. To examine the in vivo cartilage protective effects, polyphenols were injected intra-articularly before (prophylactic) and after (therapeutic) the induction of collagen-induced arthritis in rats. The hind paw volume and histomorphological scoring was done for cartilage damage. The intra-articular injection of epigallocatechin gallate and tannic acid did not significantly influence the time of onset or the intensity of joint inflammation. However, histomorphological scoring of the articular cartilage showed a significant reduction in cartilage degradation in prophylactic- and therapeutic-groups, indicating that intra-articular injections of polyphenols bind to articular cartilage and making it resistant to degradation despite ongoing inflammation. These studies establish the value of intra-articular injections of polyphenol in stabilization of cartilage collagen against degradation and indicate the unique beneficial role of injectable polyphenols in protecting the cartilage in arthritic conditions.

Keratin from the hoof is a less explored source for making valuable products. In this paper we present the extraction of pure keratin from bovine hooves and characterized them to better address the possible exploitation of this bio-resource as an alternative material for tissue engineering applications. The keratin protein from the pulverized hooves was extracted by reduction, which was observed to be pure, and two polypeptide chains of molecular weight in the range of 45–50 and 55–60 KDa were determined using SDS-PAGE assay. FTIR analysis complementing circular dichroism (CD) data, established that hoof keratin predominantly adopted α-helical conformation with admixture of β-sheet. The keratin was shown to have appreciably high denaturation temperature (215°C) as indicated by differential scanning calorimetric (DSC) analysis. Thermogravimetric analysis (TGA) also showed the retention of 50% of the original weight of the sample even at a temperature of 346°C. The keratin from the hoof had been observed to be biocompatible when analyzed with MTT assay using fibroblast cells, showing more than 90% cell viability. Hence, hoof keratin would be useful for high value biomedical applications.

Capsaicin is a versatile plant product which has been ascribed several health benefits and anti-inflammatory and analgesic properties. We have investigated the effect of capsaicin on the molecular stability, self-assembly, and fibril stability of type-I collagen. It was found that capsaicin suppresses collagen fibril formation, increases the stability of collagen fibers in tendons, and has no effect on the molecular stability of collagen. Turbidity assay data show that capsaicin does not promote disassembly of collagen fibrils. However, capsaicin moderately protects collagen fibrils from enzymatic degradation. Computational studies revealed the functions of the aromatic group and amide region of capsaicin in the collagen–capsaicin interaction. The results may have significant implications for capsaicin-based therapeutics that target excess collagen accumulation-linked pathology, for example thrombosis, fibrosis, and sclerosis.

Collagen is one of the most widely used biomaterial for various biomedical applications. In this Research Article, we present a novel approach of using collagen hydrolysate, smaller fragments of collagen, as an alternative to traditionally used collagen scaffold. Collagen hydrolysate composite scaffold (CHCS) was fabricated with sol-gel transition procedure using tetraethoxysilane as the silica precursor. CHCS exhibits porous morphology with pore sizes varying between 380 and 780 μm. Incorporation of silica conferred CHCS with controlled biodegradation and better water uptake capacity. Notably, 3T3 fibroblast proliferation was seen to be significantly better under CHCS treatment when compared to treatment with collagen scaffold. Additionally, CHCS showed excellent antimicrobial activity against the wound pathogens Staphylococcus aureus, Bacillus subtilis, and Escherichia coli due to the inherited antimicrobial activity of collagen hydrolysate. In vivo wound healing experiments with full thickness excision wounds in rat model demonstrated that wounds treated with CHCS showed accelerated healing when compared to wounds treated with collagen scaffold. These findings indicate that the CHCS scaffold from collagen fragments would be an effective and affordable alternative to the traditionally used collagen structural biomaterials.

Development of a bio-composite using synergistic combination is a promising strategy to address various pathological manifestations of acute and chronic wounds. In the present work, we have combined three materials viz., mupirocin as an antimicrobial drug, sol-gel processed silica microsphere as drug carrier for sustained delivery of drug and collagen, an established wound healer as scaffold. The mupirocin-loaded silica microspheres (Mu-SM) and Mu-SM loaded collagen scaffold were characterized for surface morphology, entrapment efficiency and distribution homogeneity, in vitro drug release, water uptake capacity, cell proliferation and antibacterial activity. In vivo wound healing efficacy of the bio-composite was experimented using full thickness excision wound model in Wistar albino rats. The Mu-SM incorporated collagen scaffold showed good in vitro characteristics in terms of better water uptake, sustained drug availability and antimicrobial activity. The wound closure analysis revealed that the complete epithelialisation was observed at 14.2 ± 0.44 days for Mu-SM loaded collagen, whereas this was 17.4 ± 0.44 days and 20.6 ± 0.54 days for collagen and control groups, respectively. Consequently, the synergistic strategy of combining mupirocin-loaded silica microspheres and collagen as a Mu-SM loaded collagen dressing material would be an ideal biomaterial for the treatment of surface wounds, burns and foot ulcers.

Fibrils of Type I collagen in the skin are exposed to ultraviolet (UV) light and there have been claims that collagen photo-degradation leads to wrinkles and may contribute to skin cancers. To understand the effects of UV radiation on collagen, Type I collagen solutions were exposed to the UV-C wavelength of 254 nm for defined lengths of time at 4°C. Circular dichroism (CD) experiments show that irradiation of collagen leads to high loss of triple helical content with a new lower thermal stability peak and SDS-gel electrophoresis indicates breakdown of collagen chains. To better define the effects of UV radiation on the collagen triple-helix, the studies were extended to peptides which model the collagen sequence and conformation. CD studies showed irradiation for days led to lower magnitudes of the triple-helix maximum at 225 nm and lower thermal stabilities for two peptides containing multiple Gly-Pro-Hyp triplets. In contrast, the highest radiation exposure led to little change in the Tm values of (Gly-Pro-Pro)10 and (Ala-Hyp-Gly)10, although (Gly-Pro-Pro)10 did show a significant decrease in triple helix intensity. Mass spectroscopy indicated preferential cleavage sites within the peptides, and identification of some of the most susceptible sites of cleavage. The effect of radiation on these well defined peptides gives insight into the sequence and conformational specificity of photo-degradation of collagen. © 2011 Wiley Periodicals, Inc. Osteogenesis imperfecta (OI), or brittle bone disease, often results from missense mutation of one of the conserved glycine residues present in the repeating Gly-X-Y sequence characterizing the triple-helical region of type I collagen. A composite model was developed for predicting the clinical lethality resulting from glycine mutations in the alpha1 chain of type I collagen. The lethality of mutations in which bulky amino acids are substituted for glycine is predicted by their position relative to the N-terminal end of the triple helix. The effect of a Gly --> Ser mutation is modeled by the relative thermostability of the Gly-X-Y triplet on the carboxy side of the triplet containing the substitution. This model also predicts the lethality of Gly --> Ser and Gly --> Cys mutations in the alpha2 chain of type I collagen. The model was validated with an independent test set of six novel Gly --> Ser mutations. The hypothesis derived from the model of an asymmetric interaction between a Gly --> Ser mutation and its neighboring residues was tested experimentally using collagen-like peptides. Consistent with the prediction, a significant decrease in stability, calorimetric enthalpy, and folding time was observed for a peptide with a low-stability triplet C-terminal to the mutation compared to a similar peptide with the low-stability triplet on the N-terminal side. The computational and experimental results together relate the position-specific effects of Gly --> Ser mutations to the local structural stability of collagen and lend insight into the etiology of OI.

Stabilization of type I rat tail tendon (RTT) collagen by various aldehydes, viz. formaldehyde, gluteraldehyde, glyoxal and crotanaldehyde was studied to understand the effect of each on the thermal, enzymatic and conformational stability of collagen. The aldehydes have been found to increase the heat stability of rat tail tendon collagen fibres from 62 to 77-86 degrees C. The increase in thermal stability was found to be in a species dependent manner. The variation in the thermal stability of collagen brought about by aldehydes was in the order of formaldehyde > gluteraldehyde > glyoxal > crotanaldehdye. The aldehydes also impart a high degree of stability to collagen against the activity of the degrading enzyme, collagenase. The order of enzymatic stability brought about by aldehydes follows the same trend as the thermal stability brought about by them. This shows that the number of cross-links formed influence both the thermal and enzymatic stability in the similar manner. The effect of various aldehydes on the secondary structure of collagen was studied using circular dichroism and it was found that the aldehydes lead to changes in the amplitude of the circular dichroic (CD) spectrum but did not alter the triple helical conformation of collagen. The secondary structure of collagen is not significantly altered on interaction with different aldehydes.

Collagen, a unique connective tissue protein finds extensive application as biocompatible biomaterial in wound healing, as drug carriers, cosmetics, etc. A study has been undertaken to stabilise Type-I collagen of rat-tail tendon using plant polyphenol (Acacia Mollissima) in the presence of an acrylic polymer. It has been found that collagen fibres pre-treated with acrylic polymer followed by the treatment with Acacia Mollissima exhibited an increase in hydrothermal stability by 25°C. Infrared spectroscopic studies display the changes in the spectral characteristics of native and treated collagen films. Transmission electron microscopic and circular dichroic studies provide an insight into the understanding of the improved stabilisation of collagen, due to treatment with acrylic polymer and plant polyphenols. The study is expected to enhance the biomaterial applications of collagen tissues.
