Tissue culture methods are useful in assessing the tolerance of various stresses due to the ease of controlling stress under in vitro conditions. This study aimed to investigate the response of sugarcane genotyps to drought stress using calli as a model system. For inducing sugarcane callus, the medium of Murashige and Skoog (MS) was used with different mannitol concentrations (100, 200, and 300 mM) to measure their effects on callus frequency, the day of callus initiation, embryogenic potential, relative growth rate (RGR), water and proline contents, K+ and Na+ contents, as well as the formation of shoot and roots for three sugarcane genotypes (e.g., GT 54-9, G 84-47, and pH 8013). The RAPD-PCR analysis was carried out using five oligonucleotide primers to identify the genetic variation among sugarcane genotypes. The results indicated that the degree of callus proliferation varied from 70 − 86%. The highest value of callus proliferation, PGR, shoot formation was recorded for the genotype GT 54-9 compared to the other two genotypes (G 84-47 and pH 8013). Calli treated with 100 mM mannitol showed the highest RGR, proline and waer contents for the genotype GT 54-9, while, those treated with 300 mM recorded the lowest values of these parameters for the genotype pH 8013. The genotype G 84-47 collected highest Na+ content, while the genotype pH 8013 collected highest K+ content. The results of this study recommend preference for GT 54-9 genotype, which is considered the most promising genotype, showing more tolerance to drought stress based on all studied traits.

Summary Background The landscape of dermatology services, already rapidly evolving into an increasingly digital one, has been irretrievably altered by the COVID‐19 (SARS‐CoV‐2) pandemic. Data are needed to assess how best to deliver virtual dermatology services in specific patient subgroups in an era of ongoing social distancing and beyond. Initial studies of teledermatology in paediatric populations suggest that many of the problems experienced in adult telemedicine are more apparent when treating children and come with additional challenges. Aim To evaluate the efficacy of a virtual paediatric dermatology telephone clinic in comparison to traditional face‐to‐face (FTF) clinics, both from the clinician and patient/parental perspective. Methods We carried out a prospective service evaluation examining a single centre cohort of paediatric dermatology patients managed during the COVID‐19 pandemic via a telephone clinic supported by images. The study period covered June–September 2020. Data on outcomes were collected from clinicians and a qualitative patient/parental telephone survey was undertaken separately. A five‐point Likert scale was used to assess both satisfaction and levels of agreement regarding whether a telephone clinic was more convenient than an FTF clinic. Results Of 116 patients included, 24% were new and 76% were follow‐up patients, with a mixture of inflammatory dermatoses (75%) and lesions (25%). From the clinician's perspective, most consultations (91%) were successfully completed over the telephone. However, qualitative patient and parent feedback paradoxically illustrated that although nearly all (98%) respondents had no outstanding concerns, 52% felt highly unsatisfied and only 22% agreed that telephone clinics were more convenient. Most (65%) preferred FTF follow‐up in the future. Statistical analysis using χ² test showed that among those with established follow‐ups, the preference for future consultation type was independent of specific reasons for follow‐up. Conclusions Our study demonstrates a clear discrepancy between the practical successes of a virtual service from the clinician’s perspective compared with the patient/parental perspective. Parental anxiety appears to be less effectively allayed virtually than with FTF. This raises the question of whether there is a role for virtual paediatric telephone clinics in the postpandemic future, which may be better left to patients/parents to decide on an individual basis.

Thermostable DNA polymerases are being widely used in polymerase chain reaction for diagnosis, cloning and DNA sequencing purposes. We have cloned a gene from Pyrobaculum calidifontis encoding a thermostable DNA polymerase and expressed in Escherichia coli BL21(DE3). The gene product was produced in soluble and highly active form. However, the expression was low, which resulted in a poor yield of the recombinant enzyme. In order to get higher expression, we introduced a silent mutation at second codon by replacing the rare codon (AGG), for arginine, with an E. coli preferred codon (CGT). This resulted in nearly a 2-fold higher production of the recombinant protein. We further optimized the inducer concentration and time of cultivation after induction. Under optimized conditions nearly 3-fold higher production of the recombinant protein was observed. Furthermore, we changed the expression host to E. coli Rosetta DE3 cells. However, there was no increase in the expression level, rather it was slightly decreased. Such silent mutations and use of an alternate expression host can be used to enhance heterologous expression of other genes too.

Purpose The aim of this study was to assess the feasibility and safety of a novel IPOM procedure with peritoneal bridging (IPOM-pb) for laparoscopic ventral hernia repair, and to compare the outcomes of this procedure with IPOM with- (IPOM-plus) and IPOM without (sIPOM) defect closure. Method A single-centre retrospective study comparing a novel IPOM technique with peritoneal bridging (IPOM-pb) with the two commonly used IPOM techniques, IPOM with defect closure (IPOM-plus) and without defect closure (sIPOM). The intraoperative and postoperative data of patients who underwent laparoscopic IPOM ventral hernia repair were reviewed. Preoperative data, recurrence, and postoperative seroma, surgical site infection, and pain, were compared. Results From January 2017 to June 2020, a total of 213 patients underwent laparoscopic ventral and incisional hernia repair with IPOM technique. The mean length and width of the ventral hernia was 4.4 ± 1.8 cm and 3.6 ± 1.4 cm, respectively, and the mean BMI was 30.1 ± 5.2 kg/m2. The mean operating time was 67 ± 28 min and was longer for IPOM-pb (71 ± 27 min), less for IPOM-plus (63 ± 28 min), and least for sIPOM (61 ± 26 min). The incidence of early postoperative seroma was least in IPOM-pb (1/98, 1%), and similar in the IPOM-plus (4/94, 4%) and sIPOM (1/21, 5%) group. Late postoperative seroma was found only in IPOM-plus (2, 2%). The incidence of early and late postoperative pain was relatively higher in sIPOM (3, 14%; 1, 5%, respectively) compared to IPOM-pb and IPOM-plus in the early (5, 5% and 6, 6%) and late (2, 2% and 1, 1%) postoperative period, respectively. Surgical site infection was higher in sIPOM group (3, 14%), compared to IPOM-pb (1, 1%), and IPOM-plus (3, 3%). Recurrence rates were similar in IPOM-pb group (3/98, 3%) and IPOM-plus (3/94, 3%), and none in sIPOM (0/21). Conclusion IPOM with peritoneal bridging is as feasible and safe as conventional IPOM with defect closure and simple non-defect closure. However, a large randomised controlled trial is required to confirm this finding.

Seromas are a common and unavoidable complication following lymphadenectomy, and often become clinically significant with superseded infection requiring re-admission for prompt intervention. However, there is no consensus as to whether a formal surgical incision and drainage (ID), ultrasound (US)-guided aspiration or intravenous (IV) antibiotics alone is the most efficacious method of managing an infected seroma, the investigation of which formed the rationale for this study.This retrospective cohort study included a consecutive series of patients readmitted for infected seroma following a lymphadenectomy for melanoma at Leeds Teaching Hospitals Trust (LTHT) from 2006 to 2017. Details on management, length of hospital stay, length of follow-up and number of clinical appointments required were examined.Seventy-one cases of infected seroma were identified from the cohort of 1691 lymphadenectomies. Initially, 21 patients (29.5%) were managed by IV antibiotics alone (failure rate of 52.4%); 18 (25.4%) with US-guided aspiration (failure rate 27.8%) and 32 (45.1%) with surgical ID, which was 100% effective. Ultimately, 62.5% of the cohort required surgical management. Patients who underwent surgical ID were discharged significantly faster following the procedure (3 versus 5 days for US-guided aspiration, p = 0.002) and spent fewer days in hospital overall (p = 0.022). The overall average cost was comparable across the three treatment groups.Surgical management seemed preferential to conservative approaches in terms of efficacy and was not significantly more expensive overall; but carries anaesthetic risk. There may be a clinically significant difference in outcome depending on management; however, more evidence is required to investigate this.

The medical literature on health aspects of Ramadan fasting (RF) is widely spread in many journals of varying access, making it less readily available to those interested in the subject. We performed a nonsystematic review of the international literature from a major online database (PubMed in 1 year [2020]). The search term “Ramadan fasting” was used, and relevant literature was narrated in a concise thematic account excluding diabetes. The publications spanned fundamental, clinical, ethical, professional, cultural, and advocacy facets. The publications predictably crossed the conventional disciplinary lines and geographical locations and appeared in journals with different access systems. The contents are presented under the emerged themes depending on the retrieved literature. This year, the basic coverage included changes in physiology, nutrition during Ramadan. However, the clinical issues included a more comprehensive range such as the impact of RF on the liver and gut, and endocrine conditions such as hypothyroidism and adrenal insufficiency. Coverage also included chronic kidney disease, maternal health and fetal well-being, cardiovascular medicine, nature and function of eyes, and neurological conditions, especially epilepsy. Sports medicine and athletes' well-being received somewhat prominent coverage. Other researchers focused on documenting patients' and health-care professionals' perceptions, attitudes, and practices during Ramadan. Health aspects of RF received a sustained academic interest with a broad spectrum in 2020. This narrative provides a scoping overview to help researchers and clinicians catch up quickly with state-of-the-art science today.

Repeated traumatic brain injuries (TBIs) cause debilitating effects. Without understanding the acute effects of repeated TBIs, treatment options to halt further degeneration and damage cannot be developed. This study sought to examine the acute effects of blood–brain barrier (BBB) dysfunction, edema, inflammation and behavioral changes after either a single or double TBI using a C57BL/6 mouse model. We examined the effects of one or two TBIs, of either a mild or moderate severity. Double injuries were spaced 7 days apart, and all analysis was performed 24 h post-injury. To examine edema and inflammation, protein levels of glial fibrillary acidic protein (GFAP), S100 calcium-binding protein B, interleukin-6, and matrix metallopeptidase 9 (MMP9) were analyzed. Aquaporin-4 (AQP4) and zonula occludens-1 (ZO-1) were analyzed to observe BBB dysfunction. Ionized calcium-binding adapter molecule 1 (IBA1) was analyzed to observe microglial activation. Rotarod, beam walking, and grip strength tests were used to measure changes in physical behavior post-injury. A sample size of ≥5 was used for all analysis. Double injuries led to an increase in BBB breakdown, as indicated by altered MMP-9, AQP4, and ZO-1 protein expression. Single injuries showed an increase in microglial activation, astrocyte activation, and BBB breakdown. Behavioral tasks showed no significant differences between injured and control groups. Based on our findings, we suggest that behavioral studies should not be used as the sole clinical indicator on brain tissue recovery. Analysis of markers such as IBA1, GFAP, MMP-9, AQP4, and ZO-1 provide valuable insight on pathophysiological response to injury.

Dermoid cysts account for only a small fraction of intracranial masses, with extradural dermoid cysts being considered a much rarer entity than those located intradurally. Intracranial dermoid cysts vary in clinical presentations: some maybe asymptomatic whilst others harbor features of raised intracranial pressure, neurological deficits, or even aseptic meningitis. Dermoid cysts may also present with cutaneous lesions. Herein, we report a rare case of a 1-year-old female presenting with a midline, scalp abscess. Brain MRI revealed an intracranial, extradural tumor, with features suggestive of a dermoid cyst, unusually located in the crista galli, and complicated by the formation of two cutaneous sinus tracts. After identification and characterization by MRI, bitemporal craniotomy was performed with complete excision of the mass and sinus tracts. Histological analysis confirmed dermoid cyst as the final diagnosis. Postoperatively, the patient recovered fully and had no evidence of recurrence in subsequent visits. The case mentioned above highlights the rarity of such a presentation for an intracranial extradural dermoid cyst and the vitality of early imaging for midline cutaneous lesions for identification of intracranial extensions and avoidance of detrimental consequences.

The literature on health aspects of Ramadan fasting (RF) is widely spread in many journals, making it not readily accessible to those interested in the subject. We aimed to provide a narrative overview of the global literature production in 1 year (2020) on diabetes and RF. This was a narrative, nonsystematic review of the international literature from a single major medical online database (i.e., PubMed) during the year 2020. The search term “Ramadan fasting AND Diabetes” was used, and the relevant literature was narrated in a concise thematic account. The publications spanned a vast array of topics related to RF, including assessments of safety and efficacy profiles of older and newer diabetes therapies, modes of insulin delivery, evaluating the role of utilizing advanced technology for the treatment, and monitoring of blood glucose during RF. Increased interest was evident in capturing patients' perspectives and healthcare professionals' perceptions, attitudes, and practices during Ramadan. Fasting by high-risk groups was studied. Not surprisingly, some reports covered COVID-19 and Ramadan and the role of telemedicine in ramadan. The current literature review presents this year's research data on the safety of fasting practices, care models, and patients' experiences and perspectives. It emphasizes the need for more comprehensive interventions for high-risk patients, promoting newer antidiabetic medicines, and advanced technology for safer fasting practices.

Introduction Rheumatoid arthritis (RA) disease is a systemic progressive inflammatory autoimmune disorder. Elderly-onset RA can be assumed as a benign form of RA. Until recently, face-to-face therapeutic sessions between health professionals and patients are usually the method of its treatment. However, during pandemics, including coronavirus disease 2019 (COVID-19), teletherapeutic sessions can extensively increase the patient safety especially in elderly patients who are more vulnerable to these infections. Thus, the aim of this randomized clinical trial was to evaluate a novel teletherapy approach for management of elderly patients suffering from RA by utilizing laser acupuncture. Methods A teletherapy system was used for management of elderly patients suffering from RA. Sixty participants were allocated randomly into two groups and the ratio was 1:1. Patients in the first group were treated with laser acupuncture and telerehabilitation sessions, which consisted of aerobic exercise and virtual reality training. Patients in the second group received telerehabilitation sessions, which consisted of aerobic exercise and virtual reality training. Evaluation of patients was done by using the Health Assessment questionnaire (HAQ), the Rheumatoid Arthritis Quality of Life (RAQoL) questionnaire, and the analysis of interleukin-6 (IL-6), serum C-reactive protein (CRP), plasma adenosine triphosphate (ATP) concentration and plasma malondialdehyde (MDA). Results A statistically significant difference was found in CRP, RAQoL, IL-6 and MDA between the pre- and post-treatments in the first group ( p Discussion Laser acupuncture teletherapy could be suggested as a reliable treatment method for elderly patients suffering from RA, as it can provide a safe and effective therapeutic approach. Teletherapy provided safer access to health professionals and patients while giving a high patient satisfaction value with a relatively lower cost (ClinicalTrials.gov Identifier: NCT04684693).

Introduction Hormonal therapy with tamoxifen and aromatase inhibitors reduces breast cancer recurrence and mortality but represents a risk factor for thromboembolic events. Therefore, most surgeons discontinue hormonal agents before microvascular surgery and for a variable period thereafter. There are no guidelines regarding when therapy should be stopped (preoperatively) or when it should be resumed (post-operatively). We, therefore, audited our hospital practice with the objective of making recommendations for microvascular breast reconstruction patients. Patients and methods A review was performed of all free flap breast reconstructions between 2014 and 2019. Patients were classified according to hormone medication status at operation. Timings of drug cessation and recommencement were recorded. Thrombotic events, namely flap microvascular thrombosis, deep vein thrombosis, superficial vein thrombosis and pulmonary embolism, were compared. Results A total of 240 patients had 275 free flaps over five years with 36 receiving hormone therapy within one month prior to surgery, which was discontinued 8.5 days (range: 0–28 days) before surgery. Intraoperative microvascular thromboses (HT 2.0%, NHT 0%, and p = 0.869) and post-operative microvascular complications/flap re-explorations (HT 6.6%, NHT 0%, and p = 0.234) were comparable between the two groups. Systemic venous thromboembolic events were also similar (HT 8.3%, NHT 6.1%, and p = 0.893). Age, BMI, smoking status and preoperative chemotherapy did not influence the incidence of thrombotic complications. Conclusion Hormone therapy did not significantly increase the risk of thromboembolic events. Despite the widespread practice of withholding it for 2 weeks prior to reconstructive surgery, this study does not support such practice being beneficial in terms of thromboembolic events and flap viability. Large-scale trials are needed to establish definitive protocols.

The current work was undertaken to evaluate the nephrotoxic effect of Ketoprofen on adult male rats. Eighteen rats were divided into two groups. Ketoprofen- received group (I) included 10 rats were administered Ketoprofen at a therapeutic dose of 13.5 mg/kg by I/M injection daily for 4 successive weeks. Five rats were randomly selected from group I and sacrificed at 2 and 4 weeks of the experiment. The control group (II) that received olive oil included 8 rats, where 4 rats were sacrificed after2 weeks and the rest of rats were sacrificed after 4 weeks. Tissue specimensfrom kidneys of all groups were collected for histopathological examination as well as the serum was obtained for the determination of biochemical parameters. The histopathological examination of group I showed glomerular changes such as expanding of glomerular matrix, glomerular sclerosis and congestion of glomerular capillary in the cortex. Renal tubular degeneration and necrosis accompanied with infiltration of inflammatory cells in interstitial tissue in both cortex and medulla were also observed. The biochemical results revealed that animals in group I showed a significant increase in malondialdehyde, creatinine, and urea compared to the control group, while total antioxidant capacity was numerically decreased. In conclusion, the therapeutic dose of Ketoprofen caused damage in kidney tissue even if was taken for a short period as well as altered biochemical parameters.

For people diagnosed with a mental illness, psychiatric hospitalization is one step in a long journey, consisting of clinical recovery such as removal of symptoms, and social reintegration involving resuming social roles and responsibilities, overcoming stigma and self-maintenance of the condition. Both clinical recovery and social reintegration need to go hand-in-hand for the overall well-being of individuals. However, research exploring social media for mental health has considered narrower, disjoint conceptualizations of people with mental illness - either as a patient or as a support-seeker. In this paper, we combine medical records with social media data of 254 consented individuals who have experienced a psychiatric hospitalization to address this gap. Adopting a theory-driven, Gaussian Mixture modeling approach, we provide a taxonomy of six heterogeneous behavioral patterns characterizing peoples' mental health status transitions around hospitalizations. Then we present an empirically derived framework, based on feedback from clinical researchers, to understand peoples' trajectories around clinical recovery and social reintegration. Finally, to demonstrate the utility of this taxonomy and the empirical framework, we assess social media signals that are indicative of individuals' reintegration trajectories post-hospitalization. We discuss the implications of combining peoples' clinical and social experiences in mental health care and the opportunities this intersection presents to post-discharge support and technology-based interventions for mental health.

Despite the fact that damask rose (Rosa damascena Mill. var. trigintipetala Dieck) is an economically important aromatic plant, its growth and development has been severely suppressed by salt stress. Reports show that moringa leaf extract (MLE) or salicylic acid (SA) are involved in salt-stress tolerance. Nevertheless, scarce information is available about MLE or SA impacts on aromatic plants, especially damask rose under salinity. This investigation was, therefore, conducted to investigate the effects of MLE and (or) SA on salt-stress mitigation in damask rose. Rose plants were treated with 200 mmol·L−1 sodium chloride, and foliar applied with MLE at 1:30 (v:v) and (or) 0.2 mmol·L−1 SA. Foliar spraying with MLE and (or) SA enhanced the growth attributes, chlorophyll content, relative water content, proline content, and membrane stability index under salinity. Less accumulation of malondialdehyde and H2O2 was also observed when MLE and (or) SA were applied. Furthermore, MLE and (or) SA applications considerably increased the radical scavenging activity, total phenols, and antioxidant enzyme activity (catalase and superoxide dismutase) under stress. The mitigative effect of MLE or SA was more pronounced when they were applied in combination. Collectively, MLE and (or) SA had efficient antioxidant defense system that scavenged reactive oxygen species, and thus afforded considerable protection against salt-induced oxidative damage.

The trials of finding non-conventional and alternative aquafeed ingredients are increasing. In this sense, this study evaluated the influence of coconut oil on the growth, feed utilization, immune, and antioxidative responses of Nile tilapia. Five test diets were formulated by mixing coconut oil with the other ingredients at 0, 1, 2, 3, and 4% of the total ration and presented for tilapia for 60 successive days. The final weight, SGR, weight gain (WG), and feed intake were superior in fish delivered 2% of coconut oil (P < 0.05). Concurrently, fish that received 2% coconut oil had lower FCR and higher PER than fish of the control and 4% groups (P < 0.05). Higher lipase activity was observed in fish of 2% and 3% levels than the remaining groups (P < 0.05). Besides, the amylase and protease activities of fish in 1%, 2%, and 3% groups were higher than the 0% level (P < 0.05). The total blood cholesterol, RBCs, and PCV showed higher values in Nile tilapia fed 2% and 3% coconut oil (P < 0.05). The lysozyme and phagocytic activities were higher in fish fed 2% and 3% levels than the control (P < 0.05), while the phagocytic index in 2% and 3% levels was higher than 0% and 4% levels. Furthermore, SOD and CAT were higher in fish fed 1%, 2%, and 3% than fish fed 0% and 4% levels while GSH was higher in fish of 1%, 2%, and 3% than fish fed 0% level (P < 0.05). However, the MDA level was markedly lower in fish fed 25, 3%, and 4% coconut oil than the 0% level (P < 0.05). The intestine's histological structure in all groups appeared normal, forming of intestinal villi projecting from the intestinal wall. Also, the structure of the hepatopancreas had a normal architecture in all groups. To sum up, the inclusion of coconut oil at 2 to 3% is recommended as a replacer for fish oil in Nile tilapia diets.

This study aimed to use thyme volatile oil (TVO) to control potato bacterial wilt disease caused by Ralstonia solanacearum and to explore the biochemical responses in relation to defense enzymes that induce resistance against pathogens in plants. In vitro experiments showed that all tested concentrations of TVO (1 µl, 2, 4, 6, 8, and 10 µl ml−1) significantly (P ≤ 0.05) inhibited the growth of the pathogen, with the most significant effect recorded at a concentration of 10 µl ml−1. Under greenhouse conditions, the tested concentration of TVO (10 µl ml−1) significantly (P ≤ 0.05) reduced the incidence of potato bacterial wilt and the population of R. solanacearum pathogens in potato plants. The current study also revealed that TVO (10 µl ml−1) increased the activity of antioxidant enzymes, e.g.,. peroxidase (POX) and phenylalanine ammonia-lyase (PAL), and reduced the activity of catalase (CAT) in treated potato plants. In conclusion, our results showed that TVO (10 µl ml−1) afforded a strong reduction of R. solanacearum radial growth in vitro and a high reduction in the development of potato wilt in vivo. Moreover, we demonstrated that POX and PAL activities can be used as biochemical markers for biocontrol efficacy in the control of potato bacterial wilt.

Introduction: Peer review is vital to the scientific publishing process. However, the present system has been criticized and accused of bias, lack of transparency, and failure to detect significant breakthroughs. Peer reviewers usually work pro bono, and their efforts are not formally acknowledged. Some journals have difficulty finding appropriate reviewers who can complete timely reviews, resulting in significant publication delay. Materials and Methods: An online survey of a convenience sample of clinicians and biomedical scientists from the Middle East (107) and Africa (69) was conducted to explore why reviewers decline to review and to ascertain their opinions on reviewer incentives. Items were scored on 5-point Likert scales, with low scores indicating low importance or low agreement. Results: One hundred and seventy two respondents provided adequate responses for analysis. Factors rated most highly in importance for the decision to accept to review a paper included contribution of the paper to the subject area (69.8%), the relevance of the topic to own work (66.0%), and desire to keep up to date with research (63.8%). The most highly rated factor that was important in the decision to decline to review was conflict with other workloads (69.4%), followed by low quality of submissions and tight time scale (65.8% for both), and lack of interest (65.1%). Most respondents agreed that financial incentives would not be effective when time constraints are prohibitive. However, reviewers agreed that nonfinancial incentives might encourage reviewers to accept requests to review: annual acknowledgment on the journal's website (78.5%), more feedback about the outcome of the submission (74.3%) and quality of the review (73.0%), appointment of reviewers to the journal's editorial board (69.1%), and being offered free subscription to the journal content (68.7%). Conclusions: Reviewers are more likely to accept to review a manuscript when it is relevant to their area of interest. Lack of time is the principal factor in the decision to decline. Reviewing should be formally recognized by academic institutions, and journals should acknowledge reviewers' work.

Microbial pathogens drive tumorigenesis in 20% of cancer cases, so the present study is aimed to evaluate the carcinogenic activities, sperm abnormalities and other dangerous effects of the subcutaneous injection of extracts obtained from various clinical Gram-negative bacteria derived from cancer patients using albino rats. We isolated, identified and extracted of their secondary metabolites of carbapenem resistant Gram-negative bacteria derived from cancer patients. Various methods have been used to determine hepatotoxicity, nephrotoxicity, tumorigenesis, inflammatory and sperm abnormalities in the albino rats injected with extracts. In comparison with the normal animals group, all extracts induced hepatotoxicity which was evidenced by the significant elevation in the activity of the serum alanine aminotransferase, aspartate aminotransferase, gamma-glutamyltransferase and alkaline phosphatase; also, nephrotoxicity that was indicated through the marked increase in the serum urea and creatinine levels; tumorigenesis was achieved from the sharp elevation in serum levels of alpha fetoprotein, carcinoembryonic antigen and lactate dehydrogenase values as tumor markers; as well as severe inflammatory characteristics were monitored from the marked raise of tumor necrosis factor alpha and interleukin-1beta. Furthermore, the proportion of micronuclei in polychromatic erythrocytes and sperm abnormalities were statistically significant in all groups compared to control group. Various kinds of head abnormalities and coiled tail were noted. Histopathological examination of hepatic tissue came in line with the biochemical and cytological findings. It could conclude that the extracts of Serratia sp. Esraa 1, Stenotrophomonas sp. Esraa 2, Acinetobacter sp. Esraa 3, Escherichia sp. Esraa 4 and Pseudomonas sp. Esraa 5 were able to initiate cytotoxicity and tumorigenesis in rats.

BACKGROUND AND STUDY AIM Bile duct injuries are not infrequently seen during hepatobiliary surgery, particularly after liver transplantation and cholecystectomy. The current study aims to figure out the frequency of postcholecystectomy biliary injuries (PCBI) and the role of early versus late endoscopic retrograde cholangiopancreatography (ERCP) in their management. PATIENTS AND METHODS Totally 960 cases operated by both laparoscopic and open cholecystectomy were evaluated in the current study. In total, 942 cases were operated in our institutes, by both laparoscopic (n = 925) and open (n = 17) cholecystectomy, and the frequency of PCBI among patients operated in our institutes was (9/942) 0.95%. Additional 18 cases of PCBI referred to our centers were included in the study. One patient was treated by repair during the surgery, in the remaining 26 patients, ERCP management was attempted. The full details of the 26 patients regarding ERCP management were discussed. RESULTS The overall success rate of ERCP management was 88.46% (23/26), whereas 11.54% of cases were treated surgically by choledochal-jejunal anastomosis due to complete common bile duct ligation. There were no differences between patients treated by early (first week) versus late (after the first week) ERCP regarding the needed interventions, type of PCBI, type and diameter of the inserted stents, and the overall success. There were no adverse events associated with ERCP management. CONCLUSIONS ERCP was valuable in the treatment of 88.46% of injured cases. There were no differences between early and late ERCP in the treatment of PCBI. Furthermore, ERCP management was not associated with adverse events.

Background: Liver cancer ranks as the 7th and 5th leading cause of cancer morbidity worldwide in men and women, respectively. Hepatocellular Carcinoma (HCC) is the most common type of liver cancer and is associated with an increasing global burden of Nonalcoholic Fatty Liver Disease (NAFLD) and Nonalcoholic Steatohepatitis (NASH). Objective: The present study aimed to investigate the possible chemopreventive effect of etoricoxib on diethylnitrosamine (DENA) and 2-acetylaminofluorene (2AAF)-induced HCC in male Wistar rats. Methods: HCC was induced by DENA (150 mg/kg/week; i.p) for 2 weeks, then 2AAF (20 mg/kg; p.o) every other day for three successive weeks. Etoricoxib (0.6 mg/kg, p.o.) was given to DENA/ 2AAF-administered rats for 20 weeks. Results: Etoricoxib significantly suppressed alpha-fetoprotein (AFP) and carbohydrate antigen 19-9 (CA19.9) as liver tumor biomarkers. It also decreased serum aspartate aminotransferase (AST), alanine aminotransferase (ALT), and total bilirubin levels while increased serum albumin levels. Besides, it alleviated DENA/2AAF-induced histopathological abrasions and inflammatory cell infiltration. Furthermore, etoricoxib showed a potent antioxidant effect, supported by a significant lipid peroxide reduction and elevation in superoxide dismutase activity and GSH content. In addition, Etoricoxib significantly down-regulated the protein expression of interleukin 1 beta (IL-1β), tumor necrosis factor α (TNFα), nuclear Factor-kappa B (NF-κB), phosphorylated nuclear Factor-kappa B (p-NF-κB), cyclooxygenase-2 (COX-2), and prostaglandin E2 (PGE2). Conclusion: In conclusion, the current results proved that etoricoxib possesses an anticarcinogenic effect via its antioxidant, anti-inflammatory, and modulation of NF-κB/COX-2/PGE2 signaling.

The research, influences of different energy microwaves treated water on the antioxidant enzyme of Trigonella foenum-graecum (T. foenum ) were studied. The analysis was carried out in 2020 in the laboratories of Ecology Department, College of Science under stable variables such as temperature, humidity, sunlight. The antioxidant enzyme, The catalase (CAT), ascorbate peroxidase (APX), superoxide dismutase (SOD) and peroxidase (POD) activity in T. foenum (hypocotyl and radical) were measured after twenty days of experiments. Results showed that germination was increased with water energy, and all enzymes activity were increased in low energy treated water gradually but decreased rapidly in high energy treated water. hypocotyl showed a higher response than Radical. (CAT) and (SOD) enzymes activity rises slowly in 30w* until it reaches a peak in 50 w * 120 s then decreases rapidly in 70 w * 150 s and whenever the time of treatments increased the enzyme becomes less effective. Both (APX) and (POD) had the same response but showed an increase in activities at 30W. The results improve that microwaves treated water-induced antioxidant enzyme response in T. foenum. While the specified energy level and duration showed a better growth rate compared to regular watering water. The results of this research serve those interested in developing cultivation and water treatment methods and improving our understanding of T. foenum irrigation behaviour using microwave treated water.

Background and Aim: Camels are important livestock in Egypt on cultural and economic bases, but studies of etiological agents of camelid diseases are limited. The enteropathogen Escherichia coli is a cause of broad spectrum gastrointestinal infections among humans and animals, especially in developing countries. Severe infections can lead to death. The current study aimed to identify pathogenic E. coli strains that cause diarrhea in camel calves and characterize their virulence and drug resistance at a molecular level. Materials and Methods: Seventy fecal samples were collected from diarrheic neonatal camel calves in Giza Governorate during 2018-2019. Samples were cultured on a selective medium for E. coli, and positive colonies were confirmed biochemically, serotyped, and tested for antibiotic susceptibility. E. coli isolates were further confirmed through detection of the housekeeping gene, yaiO, and examined for the presence of virulence genes; traT and fimH and for genes responsible for antibiotic resistance, ampC, aadB, and mphA. The isolates in the important isolated serotype, E. coli O26, were examined for toxigenic genes and sequenced. Results: The bacteriological and biochemical examination identified 12 E. coli isolates from 70 fecal samples (17.1%). Serotyping of these isolates showed four types: O26, four isolates, 33.3%; O103, O111, three isolates each, 25%; and O45, two isolates, 16.7%. The isolates showed resistance to vancomycin (75%) and ampicillin (66.6%), but were highly susceptible to ciprofloxacin, norfloxacin, and tetracycline (100%). The structural gene, yaiO (115 bp), was amplified from all 12 E. coli isolates and traT and fimH genes were amplified from 10 and 8 isolates, respectively. Antibiotic resistance genes, ampC, mphA, and aadB, were harbored in 9 (75%), 8 (66.6%), and 5 (41.7%), respectively. Seven isolates (58.3%) were MDR. Real-time-polymerase chain reaction of the O26 isolates identified one isolate harboring vt1, two with vt2, and one isolate with neither gene. Sequencing of the isolates revealed similarities to E. coli O157 strains. Conclusion: Camels and other livestock suffer various diseases, including diarrhea often caused by microbial pathogens. Enteropathogenic E. coli serotypes were isolated from diarrheic neonatal camel calves. These isolates exhibited virulence and multiple drug resistance genes.

Omani (Dhofari) frankincense is world famous for its high quality and fragrance. The oleo-gum resin obtained from the tree of Omani species i.e., Boswellia sacra (local name: luban) is widely used by the natives. The aim of the study was to determine the presence of phytochemicals in chloroform and ethanol extracts of B. sacra resin and to evaluate the in vitro cytotoxic, antimicrobial- and antioxidant-activities. The ethanol and chloroform extracts of Hojari luban were obtained in good yield. The extracts were subjected to phytochemical screenings, quantification of total phenolics (TP) and evaluation of in vitro biological activities viz. antioxidant, antibacterial and cytotoxic potential. The bioactivities of four boswellic acids, the major constituents of the oleo-gum resin, were also predicted with the help of PASS and CLC-pred software. Both the extracts showed similar level of TP (14.67 mg/g and 14.79 mg/g in terms of gallic acid equivalent) but polar fraction was noted to be more active against E. coli. A moderate antioxidant activity was shown by the resin extracts however their cytotoxic activity was observed to be quite remarkable at 1000 μg/l which could be attributed to their high boswellic acid content. Results of in silico PASS studies showed boswellic acids to possess an array of useful biological activities including chemopreventive activity. Further, CLC-pred software revealed that only keto boswellic acid is expected to exert antineoplastic activity against pancreatic carcinoma cell lines, however all the four major boswellic acids are likely to be active against non-tumor embryonic lung fibroblast (MRC5). Introduction: Literature on the health aspects of Ramadan fasting (RF) is widespread in many journals, making it less readily available to those interested in the subject. Materials and Methods: This is a narrative, nonsystematic review of international literature from two major online databases (viz., Scopus and PubMed) in 1 year (2019). The search term “RF” was used, and relevant literature was narrated in a concise thematic account excluding diabetes. Results: The publications spanned the fundamental, clinical, ethical, professional, cultural, and advocacy facets of the subject. The publications crossed the conventional disciplinary lines and geographical locations and appeared in journals with different access systems. The content is presented under relevant themes depending on the available literature. Basic coverage included changes in physiology, nutrition, and metabolism during Ramadan. Clinical aspects such as the impact of RF on kidney function, pregnancy outcome, fetal life, structure and function of eyes, and athletes' well-being received comparatively prominent coverage by researchers in 2019. Gut, liver, skin, skeleton, and blood were also covered. Other workers focused on documenting the perception, attitudes, and practices of both patients and health-care professionals during Ramadan. Conclusions: The health aspects of RF received sustained academic interest with a wide spectrum in 2019. We provided a scoping overview to help researchers and clinicians catch up quickly with the state-of-the-art today.

Most anticancer medications undergo major first-pass metabolism in the intestinal wall, the liver, or both. 5-fluorouracil (5-FU) is known to have erratic oral bioavailability due to first-pass metabolism. The present study aimed to develop 5-FU-loaded microsponges (MS) compressed in enteric-coated tablets as a new colon targeting to colorectal cancer. MS was prepared as a controlled release system for 5-FU and characterized for drug encapsulation efficiency, and surface morphology. The modified Sgarbossa criteria have been established to aid in the diagnosis of ST-elevation myocardial infarction in patients with left bundle branch block. Thus far, the sensitivities and specificities of the Sgarbossa signs have only been evaluated retrospectively in cohorts of patients with and without occlusive myocardial infarctions. These statistical analyses were based on correlating ST abnormalities with serum markers of myocardial injury and/or results of emergent cardiac catheterization. We present a patient with acute cardiovascular emergency where electrocardiograms revealed intermittent left bundle branch block. In serial ECGs, highly dynamic ST abnormalities on the narrow QRS beats were associated with similarly dynamic ST changes in the left bundle branch block beats. Our findings provided direct and real-time confirmation of the usefulness of the Sgarbossa and the modified Sgarbossa criteria in the diagnosis of acute ST elevation in patients with left bundle branch block.

Endoscopic retrograde cholangiopancreatography (ERCP) followed by laparoscopic cholecystectomy (LC) is a commonly performed treatment modality for concomitant gallbladder stones and common bile duct stones (CBD stones). There has been a debate about the appropriate timing of laparoscopic cholecystectomy after ERCP. The present study aimed to comprehensively compare early and delayed laparoscopic cholecystectomy after ERCP for concomitant gallbladder stones and common bile duct stones using unique parameters like the Nassar scale with clarification of the benefits of the appropriate timing. In a randomized controlled trial, adult patients with concomitant gallstones and CBD stones were randomly assigned after ERCP to one of two equal groups; early and delayed laparoscopic cholecystectomy. The early cholecystectomy group underwent surgery within 72 h of ERCP while the delayed group underwent surgery after 6 weeks of ERCP. Operative parameters included the operative time, the grading of adhesions, the difficulty of dissection in Calot’s triangle, bleeding, and conversion to open cholecystectomy. We also used the Nassar scale (for grading of operative difficulty in laparoscopic cholecystectomy) in the comparison between the two groups and predicting the surgery outcome. The delayed cholecystectomy group was associated with significantly higher rates of acute cholecystitis while waiting for surgery, higher grades of the Nassar scale, more adhesions, more operative difficulties, difficult dissection of Calot’s triangle, and longer operative time. Early laparoscopic cholecystectomy was better as it significantly had less operative time, less intraoperative difficulties, and fewer interval complications between ERCP and surgery. Background Sacrococcygeal pilonidal sinus disease (PSD) is an infection of the skin and subcutaneous tissue at the upper part of the natal cleft of the buttocks. Excision and healing by granulation “lay-open” method is still more preferable than other methods of midline closure or using flaps but the healing time is lengthy. The present study was performed to assess the healing promotion effect of platelet-rich plasma (PRP) on the pilonidal sinus wounds treated by the lay-open method. Methods One hundred patients suffering from PSD were randomly divided into two groups, they were treated by the lay-open method, at General surgery department, Kafr El-Sheik University hospital, Egypt, during the period from December 2018 to December 2019. Group (A) was adopted the regular dressing postoperatively, while group (B) was treated with PRP injection into the wound at 4 and 12 postoperative days. Results Accelerated rate of wound healing was detected in group (B) in day 10, with a significant difference detected in days 15, 20, 25 and 30 postoperative, with a mean time of complete healing 45 ± 2.6 days in group B, while it was 57 ± 2.4 days in group A with a p-value of 0.001 which indicates considerable effect in the treated group. Conclusions PRP injection is an effective new technique in accelerating the healing of pilonidal wound after surgery, with a significant decrease in post-operative pain, complications and an early return to work. Psoriasis vulgaris (PsV) is an immune-mediated inflammatory disorder with devastating psychosocial consequences. Expression of immunoregulator molecules on leukocytes in PsV remains unclear. Leukocyte-associated Ig-like receptor-1 (LAIR-1) and complement receptor-1 (CR-1) are immunoregulator receptors reported to bind complement component 1q involved in phagocytosis. We aimed to explore if altered leukocyte expression of LAIR-1 and CR-1 is associated with PsV. This case–control study included 36 PsV patients and 36 healthy controls. Neutrophils, monocytes and B and T cells were examined by flow cytometry for LAIR-1 and CR-1 mean fluorescence intensity (MFI) and positive cell percentage. Although zinc deficiency increases the risk of oxidative DNA damage, data regarding the association between zinc and oxidative DNA damage in diabetes are controversial. In this article, we focus on serum zinc levels and its relation to an established biomarker of oxidative DNA damage (8-hydroxy-2-deoxyguanosine) in patients with type 2 diabetes, and to ascertain the beneficial effects of zinc supplementation on the level of oxidative DNA damage. The study consisted of 2 interrelated parts: The first part was a cross-sectional study conducted on patients with type 2 diabetes (n = 297) and healthy individuals (n = 188). The second part was an interventional study that enrolled 38 diabetic patients with low zinc status and high DNA damage. Introduction: Appropriate dose adjustments of glucocorticoids replacement therapy for adrenal insufficiency (AI) is vital. Objective: We sought to scope physicians’ perceptions, and practices regarding Ramadan fasting (RF) impact on the management of AI. Methods: A web-based survey of a convenience sample of endocrinologists. Results: Nearly two-thirds of 145 respondents (64.1%) were adult endocrinologists and almost half (49%) saw more than 10 hypoadrenal patients per year. Most respondents (78.6%) prescribed hydrocortisone, while the minority prescribed other preparations. The glucocorticoid doses were reportedly divided twice daily by 70.8% and thrice daily by 22.2% of respondents. Respondents recognized RF as having potential consequences in adrenal insufficiency patients included causing hypoglycaemia, undue tiredness, and fatigue, hypotension, feeling dizzy, and light-headedness. Symptoms of under-replacement were thought to happen in the late afternoon by 59.3% of respondents. Almost half (45.5%) of respondents thought that RF has some probable or definite impact on glucocorticoid therapy that certainly warrants specific concern and possible action. Three quarters (76.4%) of respondents confirmed providing specific management recommendations during RF. The most frequently reported recommendation was taking in the usual morning dose of hydrocortisone just before pre-dawn meal (Suhor) (57.8%). Half reported providing patients with specific recommendations regarding breaking their fast and/or seeking help if hypoadrenal symptoms occur. Prior research has identified perceived discrimination as being a contributing factor in health and mental health disparities. However, there is little research on the relationship between perceived discrimination and behaviors such as hazardous alcohol and illicit substance use and risky sexual behaviors that put people at risk for negative health consequences including HIV. The current research explores the role that cultural factors may play in a tendency for individuals to engage in unhealthy behaviors or an ability to avoid them. A total of 266 college students who self-identified as Black or African American were surveyed on measures of familial ethnic socialization, perceived discrimination, emotion regulation, substance use, and risky sexual behaviors. Findings indicate that perceived discrimination and emotion regulation-suppression were associated with higher levels of hazardous alcohol use, and that emotion regulation-cognitive reappraisal was associated with lower levels of illicit substance use. Implications for intervention and prevention in African American college students are discussed.

Postharvest senescence is a critical problem of carnation cut flowers, limiting their transportation and subsequent marketing chain. This study was designed to assess whether the application of 1-methylcyclopropene (1-MCP), silver nanoparticles (AgNPs), and nitric oxide (NO) released from donor sodium nitroprusside (SNP) could prolong the vase life of cut carnations through an influence on the physiological and biochemical mechanisms involved in aging process. 1-MCP was used in the concentrations of 0, 100, 200, and 300 mg·m−3; AgNPs in the concentrations of 0, 50, 100, and 150 mg·dm−3; and SNP in the concentrations of 0, 0.1, 0.2, 0.3, and 0.4 mM. All treatments significantly extended the cut flower life compared with untreated flowers, more so with 300 mg·m−3 of 1-MCP, 100 mg·dm−3 of AgNPs, or 0.3 mM of SNP. All these chemical compounds were able to considerably improve the relative water content (RWC), reduce the malondialdehyde (MDA) content and increase the membrane stability index (MSI) in petals and, therefore, maintain the membrane integrity. In addition, they decreased the activities of acetyl-CoA synthetase (ACS) and nitric oxide synthase (NOS) and, hence, depressed the production of ethylene in carnation cut flowers through downregulating the ethylene production, what prolonged the vase life. Altogether, the application of exogenous 1-MCP, AgNPs, or SNP may provide a promising avenue to improve the postharvest performance of carnation cut flowers.

The response of Damask rose to drought and the underlying mechanisms involved are not known. In this study, vegetative, propagated rose plants were grown under control and water-deficit conditions in a greenhouse at Taif University, south-west of Saudi Arabia. Control plants were irrigated to field capacity (FC), while water-stressed plants were irrigated to either 50% FC (mild stress) or 25% FC (severe stress). After 60 days, leaf, stem and root fresh and dry weights (g plant−1), photosynthetic activity, leaf water potential (Ψw), leaf water content (WC), apoplastic water fraction (AWF), osmotic potential at full turgor (Ψs100) and turgor loss point (Ψs0), cell wall elasticity, osmotic adjustment (OA), and some solutes (K+, Ca2+, Cl−, proline and soluble carbohydrates) were evaluated. Water stress significantly decreased fresh and dry weights of R. damascena and all photosynthetic parameters, apart from leaf temperature, which increased. Severe water stress (25% FC) resulted in more negative Ψs100 and Ψs0 values than the mild water stress and control. The AWF did not significantly change in response to water stress. The leaf bulk modulus of elasticity (e) increased from 2.5 MPa under well-watered conditions to 2.82 and 3.5 MPa under mild and severe water stress, respectively. R. damascena experienced OA in response to water stress, which was due to the active accumulation of soluble carbohydrates and, to a lesser degree, proline under mild stress, along with tissue dehydration (passive OA) under severe stress. Overall, we identified two important mechanisms of drought tolerance in R. damascena—osmotic and elastic adjustment—but they could not offer resistance to water stress beyond 25% FC.

Aim: To determine the association of left main stem disease with diabetes and hypertension in STEMI patients underwent coronary angiography Methodology: This study was conducted at Department of Cardiology, Sheikh Zayed Hospital, Lahore during January to June 2019 where 220 patients with age ranging from 40-70 years whether male or female, presenting with STEMI, In Exposed Group: Patients presenting with STEMI having diabetes and hypertension and Unexposed Group: Patients of STEMI without diabetes and hypertension. We excluded all cases with recurrent MI or undergoing recurrent PCI (on medical record), valvular heart disease, and cardiogenic shock. Then two groups were formed i.e. exposed group with diabetes and hypertension and unexposed group without diabetes and hypertension. All patients underwent angiography by a senior consultant cardiologist with assistance of researcher. On angiography, presence of LMS disease was ≥50% occlusion (stenosis) in LMS artery. Angiography reports were assessed and LMS disease was noted labeled. Patients with LMS disease was managed as per hospital protocol. Results: In this study risk of LMS was higher among exposed group as compared to un exposed group. i.e. exposed: 19.1% & Unexposed: 5.5%, RR=3.50 The risk of LMS among exposed group was higher for patients in the age group 51-60 years (RR=8.00), for male patients (RR=5.76) risk for LMS was higher as compared to female patients in exposed group, patients with normal BMI (RR:2.20) and patients with shorter duration of STEMI in exposed group had higher risk for LMS (1-4 weeks: RR=7.54). Conclusion: Results of this study showed that hypertensive and diabetic patients presenting with STEMI had higher risk for LMS disease. Aim: To determine the frequency of hydrocephalus in cases of Tuberculous meningitis Methods: This was a cross sectional study conducted during 01-07-16 to 31-12-2017 at Department of Medicine, Sheikh Zayed Hospital, Rahim Yar Khan. The cases of tuberculosis meningitis of age range of 15 to 50 years of either gender with tuberculous meningitis were included in this study. The diagnosis of TBM was made on the basis of combination of clinical symptoms and laboratory data. The symptoms were fever, weight loss and neck rigidity with or without fits. These cases then underwent CSF analysis under aseptic measures. The findings to label TBM on CSF were low glucose (< 40 mg/dl), increased protein content (> 100 mg/dl) and high WBC count with predominance of lymphocytes. The cases of TBM were divided according to standard BMRC scale into three stages. The cases with bacterial meningitis, having any SOL in brain and those with any previous history of neurosurgery were excluded from this study. Then these cases underwent CT brain in the same institute and diagnosis of Hydrocephalus was made when any of the ventricle is dilated more than 25% of the baseline. Results: In this study there were total 93 cases of TBM with mean age of 37.11±08.67years. There were 54(58.06%) males and 39(41.94%) females. Maximum cases were seen in stage II of TBM which affected 65 (69.89%) cases. Hydrocephalus was seen in 61(65.59%) of the cases as shown in figure 01. Hydrocephalus was significantly high in male gender as compared to females where it affected 39(72.22%) of cases with p value of 0.03. It was also more common in cases that had age group 15-29 years affecting 40(67.79%) of cases with p= 0.86. Hydrocephalus was also significantly high in cases with stage II and III of TB affecting 66.15% and 77.27% of cases respectively with p= 0.01. Respiratory disease or snuffling is a major problem associated with serious economic losses in the rabbit industry. The disease in rabbit is a polybacterial infection in nature, resulting from co-infection with more than one bacterial pathogen. To determine the involved bacterial pathogens in respiratory infections in terms of prevalence and pathogenicity, different specimens from rabbits with typically respiratory disorders are subjected to the bacteriological examination followed by molecular verification through RT-PCR or PCR followed by sequencing the PCR-product. The results identified Mycoplasma sp. as a major partner in mixed infection with virulent strains of P. multocida or S. aureus, suggesting that, initial infection with Mycoplasma sp creates vulnerability to subsequent infection with P. multocida or S. aureus. Gross and microscopic analysis were also applied to demonstrate lesions accompanied by the detected bacterial pathogens. In conclusion, this study described the prevalence and characteristic of respiratory illness in rabbits domesticated in Al-Sharkia Governorate, Egypt and highlighted for the first time the role of Mycoplasma sp in increasing the severity of rabbit-respiratory infections.

Increasing the antimicrobial resistance associated with S. aureus mastitis challenges the therapy of such infection and results in extensive economic losses. This study revealed an increase in the efficacy of eight different antibiotics upon their combination with polyvinylpyrrolidone-capped silver nanoparticles (PVP-AgNPs) against MDR S. aureus mastitis strain. Significant synergism was detected in PVP-AgNPs combinations with a group of antibiotics including oxytetracycline, vancomycin, ofloxacin, cefuroxime, and ampicillin/sulbactam, but the highest synergism was reported with kanamycin, chloramphenicol, and penicillin. Furthermore, the antibacterial activity of PVP-AgNPs was tested in milk compared to PBS, demonstrating that the particles remained effective against S. aureus in milk with a twofold increase in MIC compared to PBS. At a concentration of 50 μg/kg b.wt, the PVP-AgNPs showed no signs of toxicity in albino rats upon oral exposure, whereas the intraperitoneal route produced slight toxicity signs within the safety limit. Our results suggested that a strategy of combining PVP-AgNPs with antibiotics might be a magic bullet to beat S. aureus mastitis and save the dairy industry. However, further study is required to evaluate the effect in vivo using mastitis model and expand the toxicological study to include systemic absorption from mammary gland to body organs, distribution, and elimination.

The inflammatory response is an obstacle to success in both allogeneic and autologous islet transplantation. In autologous islet transplantation (AIT), however, the recipient is also the donor, permitting pretreatment of donor/recipient for a controlled duration prior to transplantation. We sought to exploit this feature of (AIT) by pretreating donor/recipients with chronic pancreatitis undergoing total pancreatectomy and autologous islet transplantation (TPAIT) to test the hypothesis that peri-transplant treatment with the FDA-approved anti-inflammatory hydroxychloroquine (HCQ) improves graft function. In this randomized placebo-controlled pilot clinical study, patients ( n = 6) were treated with oral HCQ for 30 days prior to and 90 days after TPAIT. In vivo islet function was assessed via Mixed Meal Tolerance Testing before HCQ treatment, 6- and 12-months after surgery. In vitro islet bioenergetics were assessed at the time of transplantation via extracellular flux analysis of islet preparation samples from the clinical trial cohort and six additional patients ( n = 12). Our study shows that HCQ did not alter clinical endpoints, but HCQ-treated patients showed greater spare respiratory capacity (SRC) compared to samples from control patients ( P=0.028). Glycolytic metabolism of islet preparations directly correlated with stimulated C-peptide secretion both before and after TPAIT ( P=0.01, R 2=0.489 and P=0.03, R 2=0.674, respectively), and predicted in vivo islet function better than mitochondrial metabolism of islet preps or islet equivalents infused. In this study, marine sediment (MS) was successfully used as a source of methanogenic bacteria for the anaerobic digestion (AD) of chicken manure (CM). Using MS showed high production in liquid and semi-solid conditions. Even in solid conditions, 169.3 mL/g volatile solids of chicken manure (VS-CM) was produced, despite the accumulation of ammonia (4.2 g NH3-N/kg CM). To the best of our knowledge, this is the highest methane production from CM alone, without pretreatment, in solid conditions (20%). Comparing MS to Ozouh sludge (excess activated sewage sludge) (OS), using OS under semi-solid conditions resulted in higher methane production, while using MS resulted in more ammonia tolerance (301 mL/gVS-CM at 8.58 g NH3-N/kg). Production optimization was carried out via a response surface methodology (RDM) model involving four independent variables (inoculum ratio, total solid content, NaCl concentration, and incubation time). Optimized methane production (324.36 mL/gVS-CM) was at a CM:MS ratio of 1:2.5 with no NaCl supplementation, 10% total solid content, and an incubation time of 45 days.

Exposure of banana plants, one of the most important tropical and subtropical plants, to low temperatures causes a severe drop in productivity, as they are sensitive to cold and do not have a strong defense system against chilling. Therefore, this study aimed to improve the growth and resistance to cold stress of banana plants using foliar treatments of chitosan nanoparticles (CH-NPs). CH-NPs produced by nanotechnology have been used to enhance tolerance and plant growth under different abiotic stresses, e.g., salinity and drought, however, there is little information available about their effects on banana plants under cold stress. In this study, banana plants were sprayed with four concentrations of CH-NPs—i.e., 0, 100, 200, and 400 mg L−1 of deionized water—and a group that had not been cold stressed or undergone CH-NP treatment was used as control. Banana plants (Musa acuminata var. Baxi) were grown in a growth chamber and exposed to cold stress (5 °C for 72 h). Foliar application of CH-NPs caused significant increases (p &lt, 0.05) in most of the growth parameters and in the nutrient content of the banana plants. Spraying banana plants with CH-NPs (400 mg L−1) increased the fresh and dry weights by 14 and 41%, respectively, compared to the control. A positive correlation was found between the foliar application of CH-NPs, on the one hand, and photosynthesis pigments and antioxidant enzyme activities on the other. Spraying banana plants with CH-NPs decreased malondialdehyde (MDA) and reactive oxygen species (ROS), i.e., hydrogen peroxide (H2O2), hydroxyl radicals (•OH), and superoxide anions (O2•−). CH-NPs (400 mg L−1) decreased MDA, H2O2, •OH, and O2•− by 33, 33, 40, and 48%, respectively, compared to the unsprayed plants. We hypothesize that CH-NPs increase the efficiency of banana plants in the face of cold stress by reducing the accumulation of reactive oxygen species and, in consequence, the degree of oxidative stress. The accumulation of osmoprotectants (soluble carbohydrates, proline, and amino acids) contributed to enhancing the cold stress tolerance in the banana plants. Foliar application of CH-NPs can be used as a sustainable and economically feasible approach to achieving cold stress tolerance.

Artificial intelligence (AI) is rapidly integrating into modern technology and clinical practice. Although in its nascency, AI has become a hot topic of investigation for applications in clinical practice. Multiple fields of medicine have embraced the possibility of a future with AI assisting in diagnosis and pathology applications. In the field of gastroenterology, AI has been studied as a tool to assist in risk stratification, diagnosis, and pathologic identification. Specifically, AI has become of great interest in endoscopy as a technology with substantial potential to revolutionize the practice of a modern gastroenterologist. From cancer screening to automated report generation, AI has touched upon all aspects of modern endoscopy. Here, we review landmark AI developments in endoscopy. Starting with broad definitions to develop understanding, we will summarize the current state of AI research and its potential applications. With innovation developing rapidly, this article touches upon the remarkable advances in AI-assisted endoscopy since its initial evaluation at the turn of the millennium, and the potential impact these AI models may have on the modern clinical practice. As with any discussion of new technology, its limitations must also be understood to apply clinical AI tools successfully.
