Abstract Background DNA methylation profiling may provide a more accurate measure of the smoking status than self-report and may be useful in guiding clinical interventions and forensic investigations. In the current study, blood DNA methylation profiles of nearly 800 Polish individuals were assayed using Illuminia EPIC and the inference of smoking from epigenetic data was explored. In addition, we focused on the role of the AHRR gene as a top marker for smoking and investigated its responsiveness to other lifestyle behaviors. Results We found > 450 significant CpGs associated with cigarette consumption, and overrepresented in various biological functions including cell communication, response to stress, blood vessel development, cell death, and atherosclerosis. The model consisting of cg05575921 in AHRR (p = 4.5 × 10–32) and three additional CpGs (cg09594361, cg21322436 in CNTNAP2 and cg09842685) was able to predict smoking status with a high accuracy of AUC = 0.8 in the test set. Importantly, a gradual increase in the probability of smoking was observed, starting from occasional smokers to regular heavy smokers. Furthermore, former smokers displayed the intermediate DNA methylation profiles compared to current and never smokers, and thus our results indicate the potential reversibility of DNA methylation after smoking cessation. The AHRR played a key role in a predictive analysis, explaining 21.5% of the variation in smoking. In addition, the AHRR methylation was analyzed for association with other modifiable lifestyle factors, and showed significance for sleep and physical activity. We also showed that the epigenetic score for smoking was significantly correlated with most of the epigenetic clocks tested, except for two first-generation clocks. Conclusions Our study suggests that a more rapid return to never-smoker methylation levels after smoking cessation may be achievable in people who change their lifestyle in terms of physical activity and sleep duration. As cigarette smoking has been implicated in the literature as a leading cause of epigenetic aging and AHRR appears to be modifiable by multiple exogenous factors, it emerges as a promising target for intervention and investment.

Abstract This study investigated the impact of active rest intervals within a lower body complex training session on post-activation performance enhancement (PAPE) response in amateur soccer players. Twelve soccer players took part in four different experimental conditions. These sessions included 2 sets of lower body complex-paired exercises, each involving 3 repetitions of either back squats or hip thrusts at 90% one-repetition maximum (1RM) as a conditioning activity paired with a broad jump and countermovement jump. Between those exercises in active intra-complex rest interval conditions, participants were performing 8 repetitions of bench press at 75%1RM or, in passive intra-complex rest interval conditions, rested while seated. A significant main effect of a set to increase broad jump length (p = 0.002), countermovement jump height (p = 0.002), and modified reactive strength index (p = 0.005) was revealed, without any significant differences between conditions. Post-hoc comparisons showed a significant increase in broad jump length from baseline to Set-2 (231 ± 13 vs. 234 ± 13 cm; p = 0.003; ES = 0.22). On the other hand, countermovement jump height and modified reactive strength index significantly increased from baseline to Set-1 (34.4 ± 3.6 vs. 35.6 ± 3.9 cm; p = 0.027; ES = 0.31 and 0.4 ± 0.05 vs. 0.45 ± 0.09; p = 0.005; ES = 0.66). Results of this study showed that to sustain a high training density, sports practitioners may incorporate upper body exercises within the intra-complex rest interval during lower limb complex training sessions and still elicit a significant PAPE effect.

Abstract The aim of this study was to examine whether acute whole-body vibration, a single bout of drop jumps, or a combination of both may enhance countermovement jump (CMJ) and would affect volitional pace 3 km running performance. Twelve healthy and recreationally active males completed 4 conditions in randomized order: (i) 5 sets of 30 s calf raises on the platform but without vibration; (ii) 5 sets of 30 s calf raises on the vibration platform with 30 s rest intervals between sets; (iii) 5 sets of 6 drop jump with a 30 s rest interval between sets; (iv) 5 sets of 30 s calf raises on the vibration platform followed by 6 drop jumps with a 30 s rest interval between sets. Before, 3-min after, and immediately after a 3 km run each participant performed CMJ. No significant difference between conditions (p = 0.327) for the 3 km time trial was found. PurposeEvidence directly comparing the effects of bilateral and unilateral conditioning activities is limited. Therefore, the aim of this study was to assess the acute effect of unilateral and bilateral conditioning activity on vastus lateralis stiffness, countermovement jump parameters, and 10 m sprint.MethodsTwelve semi-professional basketball players participated in this study (age: 23 ± 4 yrs; body mass: 84.7 ± 10.6 kg; body height: 192 ± 6 cm; basketball training experience: 11 ± 4 yrs) performed four experimental sessions to compare the acute effects of bilateral, stronger-only, weaker-only lower limb or no conditioning activity on vastus lateralis stiffness, countermovement jumps variables (height; peak velocity; peak force, contraction time, countermovement depth, and modified reactive strength index and 10 m sprint time. Measurements were performed 5 minutes before and in the 5th and 10th minutes after CA.ResultsBilateral conditioning activity significantly increase the countermovement jump height (p = 0.002; ES = 0.71) and the reactive strength index modified (p = 0.010; ES = 0.59). Moreover, a significantly higher peak force in the stronger than in the weaker limb was found (p0.05). However, there were no significant (p>0.05) interactions and effects of conditions or time-point in the case of the other countermovement jump variables, vastus lateralis stiffness, and 10m sprint time.ConclusionUnilateral and bilateral drop jumps (3 sets of 5 repetitions) did not affect the vastus lateralis stiffness and time of the 10m sprint. However, only bilateral drop jumps effectively enhanced the countermovement jump height and modified reactive strength index. Bilateral drop jumps might be a useful part of a warm-up to improve jumping performance in basketball players.

Therefore, this study aimed to examine differences in training volume assessed by a number of performed repetitions, time under tension, and load–displacement as well as peak barbell velocity between the cambered and standard barbell bench press training session. The participants performed 3 sets to muscular failure of bench press exercise with the cambered or standard barbell at 50% of one-repetition maximum (1RM). Abstract The study aimed to evaluate the impact of ischemia, used only before particular sets of a lower limb resistance exercise on power output. Ten healthy resistance-trained males (age = 26 ± 6 years; body mass = 90 ± 9 kg; training experience = 9 ± 7 years) performed two experimental sessions (with ischemia; control without ischemia) following a randomized crossover design. During the ischemic condition, the cuffs were inflated to 60% of arterial occlusion pressure. The cuffs were applied before each set for 4.5 min and released 30 s before the start of the set as the reperfusion (4.5 min ischemia + 0.5 min reperfusion). In the control condition, ischemia was not applied. During the experimental sessions, the subjects performed the Keiser machine squat exercise protocol which consisted of 5 sets of two repetitions, at a load of 60% of one-repetition maximum (1RM), with 5 min rest intervals between sets. Background The study aimed to evaluate the effects of ischemia used during the rest periods between successive sets on maximal number of performed repetitions, time under tension and bar velocity during the bench press exercise. Methods and materials Thirteen healthy resistance trained men volunteered for the study (age = 28.5 ± 7.1 years; body mass = 87.2 ± 8.6 kg; bench press 1RM = 143.1 ± 20.7 kg; training experience = 11.0 ± 6.9 years). In experimental protocol the subjects performed 5 sets of bench press exercise at 70%1RM with maximal number of repetitions in each and with 5 minutes rest periods between each set. During the ischemia condition occlusion with 80% arterial occlusion pressure (AOP) was applied using a 10 cm wide cuff, before the first set of the bench press exercise and during all rest periods between sets (for 4.5 minute). During the control condition no ischemia was applied. Results The two-way repeated measures ANOVA showed a statistically significant interaction effect for time under tension (p = 0.022; η2 = 0.20). However, the results did not show a statistically significant interaction effect for peak bar velocity (p = 0.28; η2 = 0.10) mean bar velocity (p = 0.38; η2 = 0.08), and for number of performed repetitions (p = 0.28; η2 = 0.09). The post hoc analysis for interaction showed significantly shorter time under tension for ischemia condition compared to control in set 1 (p < 0.01). The post hoc analysis for main effect of condition revealed that time under tension was significantly shorter for ischemia compared to control condition (p = 0.04). Conclusion The results of this study indicate that ischemia intra-conditioning does not increase strength-endurance performance as well as bar velocity during bench press exercise performed to muscle failure.

BackgroundThe importance of leptin in controlling body mass has recently gained more attention. Its levels are directly associated with the amount of fat mass, but not necessarily dependent on it. Exercise has great potential in reducing leptin levels, however the response of exercise to this cytokine is still not well understood.ObjectiveThe objective of the review was to analyze the effects of physical exercise on plasma leptin concentration, either acutely (post-exercise/training session) and/or after a training period (short- or long-term), as well as to investigate the existence of possible moderating variables.MethodsThe studies included in this systematic review were published between 2005 and May 2023. Only peer-reviewed studies, available in English, performed with humans that evaluated the effects of any form of exercise on leptin levels were included. The search was conducted on May 03, 2023, in Embase (Elsevier), MEDLINE via PubMed®, and Web of Science (Core collection). The risk of bias in the included trials was assessed by the Physiotherapy Evidence Database tool, considering 11 questions regarding the methodology of each study with 10 questions being scored. The data (n, mean, and standard deviation) were extracted from included studies to perform random effects meta-analyses using standardized mean difference between the pre- and post-intervention effects.ResultsTwenty-five studies (acute effect: 262 subjects; short- and long-term effect: 377 subjects) were included in this systematic review and meta-analysis. Short- and long-term physical exercise and caloric restriction plus exercise reduce plasma leptin levels, presenting statistically significant differences (pConclusionBoth acute and chronic exercise reduce leptin levels, yet the acute effect is dependent on the pre-exercise meal. In addition to having a long-term reduction in leptin levels, the minimum amount of weekly exercise to have a significant reduction in plasma leptin is 180 minutes of moderate-intensity exercise and 120 minutes of high-intensity exercise.
