This work aimed to evaluate the effect of platelet-rich plasma (PRP) on advancement skin flaps in dogs regarding improvement of vascularization, with focus on increasing its viable area, since there are reports that it is a potential angiogenesis stimulator. The experimental group was composed of eight adult bitches, in which two advancement skin flaps were made in the ventral abdominal region. No product was applied in the control flap (CF), while PRP was used in the contralateral flap, called treated flap (TF). The areas were clinically evaluated every two days until the 7th postoperative day regarding skin color and presence of necrosis. At 10 days, both flaps were removed and submitted to histological examination and blood vessel morphometry. The vessels counted in each group were statistically analyzed by the F-test at 1% probability. Results showed no significant difference in macroscopic changes in the wound, or CF and TF vascularization, thus suggesting that PRP gel did not improve advancement skin flap angiogenesis in bitches under the experimental conditions in which this research was developed.

The goal of this study is to characterize the variations in glutamine (GLN) concentration and other blood biomarkers in dogs after blood donation. Blood samples were collected from 12 dogs that were used regularly for blood donation. The procedure was performed on the following occasions: before blood donation and +24 h, +7 days, +15 days, and +30 days after blood donation in order to obtain GLN, glutamate (GLU), white blood cells (WBC), red blood cells (RBC), urea, albumin (ALB), creatinine (CREAT), total protein (TP), and globulin (GLOB). The results were analyzed by ANOVA, for repeated measures with one factor, and by Tukey’s test with a P value of 5 %. There are significant differences in GLN after blood donation (P < 0.05), with an elevation in concentration in two phases, 24 h after donation and after 15 days and then return to the initial levels after 30 days. Decreases in UREA and ALB were also observed (P < 0.05). No variations were observed in GLU, CREAT, TP, and GLOB after blood donation (P < 0.05). The red blood cells, hemoglobin, and hematocrit were significantly reduced 24 h after blood donation (P < 0.05). The platelet count shows large elevation after 7 days of blood donation (P < 0.05). There is no variation in WBC, whereas lymphocytes and eosinophils varied during the experiment (P < 0.05). It is concluded that after blood donation, there is an increase in GLN, platelets, and eosinophils. More studies are needed to be done to understand how the interorgan transport of GLN contributes to the recovery of these dogs after blood donation.

Avaliaram-se dois protocolos para a produção de plasma rico em plaquetas (PRP) com o sangue de 20 cães adultos. Foram coletados três frascos de sangue em que um deles foi usado para produção do PRP por meio do protocolo A - centrifugação única a 1200rpm/10min -, o outro para fabricação do PRP pelo protocolo B - primeira centrifugação a 1200rpm/10min e a segunda centrifugação a 1600rpm/10min - e o terceiro para realização da contagem plaquetária no sangue total, que serviu de parâmetro para os valores alcançados no PRP. O protocolo no qual foi possível alcançar maior concentração plaquetária foi testado em outros 20 cães para avaliar sua reprodutibilidade. Constatou-se que o protocolo B resultou em maior plaquetometria em 100% das amostras e concluiu-se ser ele eficiente para a produção do PRP em cães. The objective of this paper was to analyze two protocols for the production of platelet rich plasma (PRP) in dogs. Peripheral blood of 20 adult dogs was collected into three tubes. The first was processed through protocol A - single centrifugation at 1200rpm for 10min - , the second was submitted to protocol B - a first centrifugation at 1200rpm for 10min and a second centrifugation at 1600rpm for 10min - and the third was used to perform platelet count in whole blood, which served as a parameter for values obtained in PRP. The protocol in which it was possible to achieve a higher platelet count was tested in other 20 dogs to evaluate its reproducibility. Protocol B resulted in a superior platelet count in 100% of the samples, concluding that the referred protocol is effective for PRP production in dogs.

O meio de contraste ioversol 240mg de iodo(I) por ml foi administrado na dose de 0,5ml/kg de peso por via intracisternal em 30 cães sadios, com o objetivo de verificar as possíveis alterações liquóricas e histopatológicas geradas pelo mesmo. Foram realizadas no líquor, análises físicas, contagens total e diferencial de leucócitos e determinação da concentração total de proteínas. A análise do líquor pós-mielográfica revelou aumento no número de leucócitos totais em três animais e da concentração de proteína em quatro. Alterações histopatológicas foram observadas em dois deles: infiltrado de células inflamatórias mononucleares na fissura ventral e no canal central da medula espinhal e infiltrado discreto de células inflamatórias mononucleares no canal central da medula espinhal, respectivamente. Concluiu-se que a mielografia com o meio de contraste ioversol 240mg I/m pode causar alterações citológicas e na concentração de proteínas no líquor, e histopatológicas, porém discretas, durante o período estudado. Ioversol contrast agent, at a concentration of 240mg of iodine/ml, was administered via intrathecal routes to 30 normal dogs at a dose of 0.5ml/kg weight to determine possible cerebrospinal fluid and histopathological changes. Physical analysis, total and differential white cell counts and quantitative protein determination of the cerebrospinal fluid were done. Cerebrospinal fluid analysis revealed increase in the total white cell count in three animals, and protein in four animals of different groups post-myelography. Histopathological changes were seen in spinal cords of two dogs: mononuclear infiltrate in the ventral fissure and in the central canal, and mononuclear infiltrate in the central canal, respectively. Myelography with ioversol 240mg I/m can cause mild cytological and protein alterations, and histopathological changes under the experimental conditions.
