The prevalence of allergic disorders has been increasing globally, and it may be useful to identify the modifiable factors associated with it. Diet has been identified as one possible lifestyle factor implicated in the pathogenesis of allergic diseases. The goal of this study was to evaluate the associations between dietary patterns and the presence of allergies in a group of young women in eastern Iran. In all, 181 female students were enrolled. Screening for presence of allergic diseases, including allergic rhinitis (AR), asthma and eczema, was performed by an expert allergist. Information about dietary intake was collected using a validated 65-item food frequency questionnaire. Exploratory factor analysis was used to evaluate the relationships between food/nutrition variables. Serum immunoglobulin (Ig)E levels were significantly higher in the group in the third tertile (highest adherence) for a Western dietary pattern compared to the group in the first tertile (lowest adherence). The multivariate odds ratio for presence of allergic rhinitis (AR) was 2.5 (95% CI:1.1-5.1) for the highest compared to lowest tertile of the Western dietary pattern score. However, no significant relationship was found between a traditional dietary pattern and AR, asthma or eczema. Our findings indicate a potential role in the development of AR of the Western dietary pattern, which is characterized by high intake of dairy products, snacks, nuts and sugar. Since diet is a modifiable factor, the relationship between AR and a Western dietary pattern may have clinical implications, particularly in subjects with other risk factors for developing AR.

ObjectivePatients with malignancy suffer from a compromised immune system due to either the effects of malignancies or treatments. Cancer patients are at higher risk of different infections particularly SARS-CoV2 and usually produce weaker response to vaccines. The aim of this study was to evaluate the safety and immunogenicity of the inactivated SARS-CoV-2 vaccine (Sinopharm, BBIBP-CorV) in patients with malignancy.Material and MethodIn total 364 patients with cancer (median age: 54 years old, F/M ratio: 217/147) who received two doses of Sinopharm vaccine were enrolled in this study. Vaccine related side effects was assessed by a questionnaire and the presence of SARS-CoV-2 anti-Spike protein (S) IgG and neutralizing antibody two months following vaccination were measured by immunological methods.ResultsInjection site pain and fever were the most common local and systemic side effects in vaccine receivers. Two months after the first dose, anti-S IgG and neutralizing antibody were detectable in 77.1% and 80.7% of all participants, respectively with an overall response to either or both measured in 86.9% of patients The rate of seroconversion was lower in older age, those with hematological malignancies and chemotherapy receivers.ConclusionThe result of study confirmed the safety and short-term efficacy of Sinopharm inactivated vaccine (BBIBP-CorV) in patients with different type of malignancies.

BACKGROUND: The first case of coronavirus disease 2019 (COVID-19) was reported in December 2019 in Wuhan, China. Healthcare workers (HCWs) are at high risk of acquiring and spreading the COVID-19 infection; using personal protective equipment (PPE) reduces the risk of COVID-19 infection in HCWs. OBJECTIVE: Our study aimed to investigate the seroprevalence of COVID-19 IgG, IgM antibodies among HCWs as well as identifying the factors associated with this seroprevalence. METHODS: This cross-sectional study was performed from July to August 2020 on healthcare workers at two COVID-19 referral hospitals of Birjand University of Medical Sciences. The level of COVID-19 IgG and IgM antibodies in sera was measured by commercial qualitative ELISA kits. RESULTS: In total, 192 individuals participated in the study: physicians (31.25%), nurses (30.2%). 84.2% of participants had contact with confirmed COVID-19 cases and among them 42.9 % of had close contact with COVID-19 patients for more than 3 months, and 31% reported close contact with more than 50 confirmed COVID-19 cases. Mask and gloves were the most frequently used personal protective equipment (PPE) with 92.4% and 77.2% of usage. CONCLUSIONS: The results of the current study showed high level of adherence to the use of PPE among HCWs as well as very low prevalence of seropositivity for of COVID-19 antibodies, hence confirming the effectiveness of PPE in protecting HCWs among COVVID-19 and possibly any other similar infections.

Background Allergic disorders are common health problems worldwide with significant socio-economic impacts. The best diagnostic method using allergenic extract is the skin prick test. Regarding the effects of geo-climatic factors and allergenic extract source material quality, the aim of study was to determine the safety and efficacy of some in-house-developed allergenic extracts. Methods Forty-five different allergenic extracts, including common regional pollen, foods, and dog and cat hair, as well as positive and negative extracts, were prepared from domestic sources using optimum extraction methods. All extracts passed stability and sterility testing, and sterile final products containing 50% glycerin in 10 and 20 w/v concentrations were used. Skin prick testing was performed on volunteers and immediate or late side effects were recorded. Results In total, 56 students (mean age: 21.2±2.3y, M/F ratio: 1.07) participated in this study. For inhalant allergens, all extracts except dog hair extract caused positive responses. Salsola kali (Russian thistle) and Fraxinus velutina (ash tree) were the most common grass and tree pollen extracts, respectively. Of 18 different food extracts, five, including egg white, tomato, fig, melon, and green pepper caused skin reactivity in only one person. No participant reported any immediate or late side effects, including large local reaction or systemic response. Conclusion The result of the current study confirmed the safety of all our in-house-developed allergenic extracts. Regarding efficacy, almost all inhalant and five food allergens caused positive skin responses.

Background Allergic disorders may have a bidirectional causal relationship with mental disorders. In this cross-sectional study, we aimed to assess the associations between cognitive abilities and emotional function tests and quality of life with the presence of allergic disease in young women. Methods A diagnosis of allergic disorders, comprising allergic rhinitis (AR), asthma and atopic dermatitis (AD), was confirmed by a specialist in allergy. The presence and severity of depression, anxiety, stress, insomnia and sleepiness were evaluated using validated questionnaires. Cognitive abilities and quality of life were assessed using standard instruments. Results Among 181 female young participants, the prevalence of AR, asthma and AD were 26.5%, 2.8%, and 14.9% respectively. The AR group had higher scores than the non-AR group for depression, anxiety, insomnia, and lower scores for physical and mental health-related quality of life. Moreover, the AD cases had higher scores on the depression and stress scale compared to those without it (p Conclusion There was a high prevalence of psychological/psychiatric disorders that included: anxiety, and sleep problems among allergic women, and a reduced quality of life that may be associated with it.

Type 1 diabetes mellitus (T1DM) is an autoimmune disorder accompanied by activation of auto-reactive B cells, autoantibody production, and consequently insulin-producing beta cell destruction. Some evidences support the value of autoantibodies such as anti-islet cell antigen (IA-2) auto-antibody in T1DM diagnosis and prognosis. The present study investigated the presence of IA-2 antibody in T1DM patients and their first-degree relatives as well as T2DM patients and healthy people to evaluate the diagnostic and prognostic value of anti-IA-2 in T1DM. In total 264 cases including 38 T1DM patients, 36 first-degree relatives, 88 T2DM patients, and 102 age-/sex-matched healthy controls participated in this study. After venous blood collection, sera were isolated and the level of IA-2 antibody was measured by ELISA method. Routine hematologic and biochemical tests including complete blood count (CBC), fasting blood sugar (FBS), hemoglobin A1c (HbA1C), urea, creatinine, as well as lipid profile and liver function tests were analyzed. While activation of immune system may lead to a lower risk of some diseases, it has been shown that a history of atopic allergic disorders such as asthma, hay fever, eczema, and food allergies could be related to several types of cancer. However, the evidence is not entirely conclusive. Two proposals suggest a possible mechanism for the association between allergic disorders and cancers: immune surveillance and the antigenic stimulation. The association of allergy and cancer may vary by cancer site and the type of exposure. The aim of current review was to summarize the current knowledge of the association between allergic diseases and the risk of cancers with particular emphasis on case–controls and cohort studies to estimate the cancer risk associated with allergy.

Background Both home and school are important places where children are exposed to various indoor allergens. This study aimed to identify the profile of indoor allergens in schools and its impact on asthma development. Methods A total of 104 classrooms from 52 schools were selected for dust collection during the fall of 2017. The levels of indoor allergens including dust mite (Der f1, Der p1), cat (Fel d1), cockroach (Bla g1) and mouse (Mus m1) were measured by enzyme linked immunosorbent assay (ELISA). The diagnosis of asthma was made in all students of the selected classes by the allergist. The collected data were analyzed using SPSS version 21.0. Results Out of 2816 students in the selected classes, 180 students were involved with asthma. Students were mostly exposed to Bla g1 (83.1%), followed by Der f1 (51.5%), Mus m 1 (45.5%), Der p1 (8.9%) and Fel d1 (7.9%) in the dust collected from 101 classrooms. Although levels of all studied allergens in the settled dust of the classrooms were low, there was a relationship between Fel d1 in the classroom dust and development of asthma. Conclusion This study showed considerable levels of cockroach allergens in schools. Exposure to cat allergen in our schools played an important role in asthma development; further school-based investigations require evaluating the role of classroom allergen on asthma development.

Invariant natural killer T cells (iNKT) produce large amounts of different cytokines which can influence differentiation, polarization and activation of immune cells, particularly NK and T cells. iNKT have been shown to suppress GvHD and promote anti-tumor and anti-pathogen immunity. There are highly specific and safe synthetic ligands such as alpha-galactosylceramide (α-GalCer) and C20:2 which activate iNKT cells toward relatively Th1 and Th2 pathways, respectively. Bone marrow transplantation (BMT) or 'hematopoietic stem cell transplantation' (HSCT) is effective for leukemia and lymphoma through 'graft-versus-leukemia' (GVL) immunity. However, frequent serious complications include graft-versus-host-disease (GVHD), opportunistic infections and relapse. Both GVHD and GVL are mediated by T cells. Manipulating iNKT by different lipid analogues in early and late phases after transplantation may suppress GVHD and graft rejection and enhance GVL effect, as well as resistance to opportunistic infections and so, could be a novel and effective strategy for improving HSCT outcome.

Vitamin D exerts multiple immune-regulatory functions on the innate and adaptive immune systems. However, there is conflicting evidence concerning the contribution of vitamin D to the progression of allergies. The goal of this study was to investigate the connection between vitamin D and the prevalence of allergic disorders and serum IgE levels among young adults. The study samples included 272 students (120 males and 152 females) from Birjand University of Medical Sciences in Birjand, Iran. The allergic disorders studied, comprising allergic rhinitis (AR), asthma and atopic dermatitis (AD), were confirmed by an expert allergist. Commercial Elisa kits were used to measure serum levels of vitamin D and total IgE for all participants. The median (IQR) serum vitamin D value was 10.0 (6.0–17.0) ng/mL and the prevalence of vitamin D deficiency among the target population was 81.5%. Nasal polyps (NP) are associated with inflamed mucosa of unknown etiology. The role of T cells in nasal polyposis is unclear. Invariant natural killer T cells (iNKT) can promote Th2 responses and have been implicated in some types of asthma. As there are shared inflammatory pathways involved in asthma and NPs, we evaluated the frequency of iNKT in 17 patients with NPs, but without asthma. A median of 6% polyp cells were T lymphocytes, of which iNKT were 0 to 2.38% (mean 0.674%). In the matched group (n = 10), iNKT in NPs was significantly higher than PBMCs (1.057% vs 0.155%, P 0.05). Relative expression of Vα24 to TCR-beta genes in polyps (n = 14) was higher than blood in matched samples (n = 4). The presence of greater proportions of iNKT in NPs than in blood suggests that iNKT may play a role in the pathogenesis of nasal polyposis.

Allergic disorders are among the most common diseases around the world especially in children. Many factors contribute to the pathogenesis of atopic disorders, but early events during the pregnancy are very important. The aim of this study was to evaluate the level of cord blood immunoglobulin E (CB-IgE) and its association with maternal in a group of Iranian newborns. In a cross-sectional study, 163 pregnant women randomly selected and information about pregnancy and atopy were taken by questionnaire. Blood samples of mothers and matched cord blood were collected and total serum IgE levels were measured by enzyme-linked immunosorbent assay (ELISA) method. To rolling out the possibility of contamination with maternal blood, total IgA was checked for all the cord blood samples. Sixteen percent of mothers had the history of atopic diseases and the mean IgE level was significantly higher in an atopic than nonatopic mothers (241 vs 102, P 0.001). About 73.9% of cord blood samples, had high IgE level (0.9 IU/mL). The level of cord blood IgE (CB-IgE) was not significantly different in male and female newborns (2.14 vs 2.15 IU/mL). There was no significant correlation between maternal factors such as age, pregnancy variables, allergens exposure, smoking, and maternal IgE with cord blood IgE. The results of this study showed that CB-IgE is high in a remarkable number of samples; independent of maternal or fetal factors. Further studies need to evaluate the reasons for the high level of IgE in cord blood in our area.

Background: Hepatitis B Virus (HBV) is a universal health challenge all around the world. Several factors like viral load, genetic characteristics, age, sex, and immune status contribute to variable clinical outcomes of HBV infection. The sequels of HBV infection vary remarkably among persons ranging from the spontaneous deletion of infection to persistent infection. Objective: The present study aimed to evaluate the association of single nucleotide polymorphisms IL10-1082 with HBV clearance. Methods: Sixty subjects with Chronic Hepatitis B (CHB) infection and 60 subjects who spontaneously recovered HBV were enrolled in the study. The IL-10-1082 polymorphisms were determined by Polymerase Chain Reaction with Restriction Fragment Length Polymorphism (PCR–RFLP). Results: The clearance of HBV infection demonstrated a significant association with IL-10-1082 polymorphisms in the GG genotype (P = 0.03), while there was no association with other genotypes. Reduced risk of chronic hepatitis B infection was associated with IL-10-1082 GG (OR: 2.33, 95% CI: 1.07 - 5.09). Besides, IL-10-1082 A/G alleles did not differ clearly between the two study groups (P = 0.07) Conclusions: The IL-10-1082 polymorphisms may be associated with a reduced risk of CHB infection and recovery after HBV infection.

Background The prevalence of vaccine side effects plays an important role in public perception about vaccination programs. This study was designed to investigate the side effects of the first dose of COVID-19 vaccine; Sputnik-V, AZD-1222, and Covaxin. Methods A study was performed to evaluate the side effects of these vaccine among 503 health care workers in Birjand (Iran). Our study used the questionnaire consisted of 4 main categories including demographic data, previous COVID-19 infection, vaccine information, and local and systemic side effects of vaccines. Results 81.9%, 88.8%, and 92.9% of people who have been vaccinated with Sputnik-V, AZD1222, and Covaxin vaccines, respectively, have reported at least one side effect. The prevalence of systemic side effects in AZD-1222 vaccine was higher than Sputnik V and Covaxin vaccines. Injection site pain (62.1%), fatigue (43.9%), muscle pain (42.5%), and fever (40.6%) were the most common side effects in all three vaccines. Side effect frequency was higher in the female group (90.6%) than the male group (79.5%). The prevalence of side effects with Sputnik V and Covaxin vaccines was reduced in the elderly. Moreover, the prevalence of side effects was higher in the case of convalescent patients (92.4 %) than in the group with no history of infection. The prevalence of side effects was higher in person with a BMI above 25 in the AZD-1222 and Covaxin vaccines. Conclusions The most common side effects of the Sputnik-V, AZD-1222, and Covaxin vaccine among Birjand (Iran) healthcare workers were injection site pain, muscle pain, fatigue, fever, and headache. Age and gender were the most important variables in the prevalence of vaccine side effects.

Background The natural killer (NK) cells differentiated from umbilical cord blood (UCB) hematopoietic stem cells (HSCs) may be more suitable for cell-based immunotherapy compared to the NK cells from adult donors. This is due to the possibility to choose alloreactive donors and potentially more robust in vivo expansion. However, the cytotoxicity of UCB-HSC-derived NK cells against cancer cells might be suboptimal. To overcome this obstacle, we attempted to generate NK cells with potent antitumor activity by targeting RAS/MAPK, IGF-1R and TGF-β signaling pathways using IL-15, IGF-1 and SIS3 respectively. Methods The CD34 + cells were isolated from human UCB mononuclear cells through magnetic activation cell sorting (MACS) with purity of (≥ 90%) and were subjected to differentiate into NK cells. After 21 days of induction with SFTG36 (SCF, FLt-3L, TPO, GM-CSF, IL-3 and IL-6), IS721 (IGF-1, SIS3, IL-7 and IL-21) and IL-15/Hsp70 media, NK cells phenotypes were studied and their cytotoxicity against K562 human erythroleukemia cells and SKOV3 ovarian carcinoma cells was analyzed. Results The NK cells induced in SFTG36/IS721 medium were selected for activation due to their higher expression of CD56 + 16 + CD3 − (93.23% ± 0.75) and mean fluorescence intensity (MFI) of NKG2D + (168.66 ± 20.00) and also a higher fold expansion potential (11.893 ± 1.712) compared to the other groups. These cells once activated with IL-15, demonstrated a higher cytotoxicity against K562 (≥ 90%; P ≤ 0.001) and SKOV3 tumor cells (≥ 65%; P ≤ 0.001) compared to IL-15/Hsp70-activated NK cells. Conclusions The differentiation of ex vivo expanded CD34 + cells through manipulation of RAS/MAPK, IGF-1R and TGF-β signaling pathways is an efficient approach for generating functional NK cells that can be used for cancer immunotherapy.

Background: Viral infections are a public health problem. Objectives: We would like to evaluate the seroprevalence of hepatitis B, C, and human immunodeficiency virus infections (HIV) in a large sample of the Afghanistan population in Kabul. Methods: In total, 196516 Afghani citizens went to Fateme-al-Zahra clinic to perform obligatory checkup for traveling to Iran. The serum samples were primarily checked by rapid tests for HBV, HCV, and HIV, and in case of positive results, a commercial ELISA kit used as the confirmatory test. Results: Out of 196516 participants, 153763(78%) were men and 42753 (22%) were women. The seroprevalence of HBV, HCV, and HIV infections was 1.23% (2430), 0.13% (265) and 0.018% (16), respectively. The prevalence of HCV and HIV was significantly higher in males than females (m/f: 0.097%/0.037% vs. m/f: 0.008%/0%, respectively; P < 0.05). The simultaneous co-infection of HBV-HCV and HBV-HIV was 0.004% and 0.0005 %, respectively. Conclusions: This study showed a low prevalence of HBV, HCV, and HIV among the study group. Considering the selection bias, sensitivity, and specificity of rapid tests, the real prevalence expected to be quite higher. Proper strategies to improve the social awareness and implement preventive vaccination for HBV can decrease the incidence of these infections.

The coronavirus disease 2019 (COVID-19) outbreak is an ongoing global health emergence, but the pathogenesis remains unclear. Here, we applied weighted gene co-expression network analysis to comprehensively characterize transcriptional changes in bronchial epithelium cells (NHBE and A549 cells) during SARS-CoV-2 infection. Our analysis identified a network highly correlated to COVID-19 pathogenicity based on MX1, IFIT1, ISG15, IFI6, DDX60, IRF9, PARP9, PGLYRP4, IL36G, SAA2 and IL-8 hub genes. The results also indicated a unique transcriptional signatures of infected cells including IFI6 and IRF9 as novel gene candidates and suggested their prospective mechanism in COVID-19 pathogenesis. The result of hub genes enrichment showed that the most correlation topic in biological process and KEGG were type I interferon signaling pathway, IL-17 signaling pathway, cytokine mediated signaling pathway, and defense response to virus categories which all play significant roles in restricting viral infection. Also according to the drug-target network, we recognized 54 FDA-approved drug candidates for other indications could potentially use for the treatment of COVID-19 patients through regulation of six hub genes of the co-expression network. Our findings also showed that the 19 experimentally validated miRNAs regulated the co-expression network through 5 hub genes (SLC19A3, FAM13A, PLA2G16, and HRASLS5). In conclusion, these hub genes had potential roles in the translational medicine and might become promising therapeutic targets further in vitro and in vivo experimental studies are needed to evaluate the role of above mentioned genes in COVID-19.

Breast cancer (BC) is among the leading causes of mortality from cancer in women. Many of the available anticancer drugs have various side effects. Therefore, researchers are seeking novel anticancer agents particularly from natural compounds and in this regard, snake venom is still one of the main sources of drug discovery. Previous studies showed potential anticancer effects of Cytotoxin II (CTII) from Naja naja oxiana against the different types of cancers. In this study, a pET-SUMO-CTII vector was transformed into SHuffle® T7 Express, an Escherichia coli strain, for recombinant protein expression (rCTII) and the cytotoxic effects of this protein was assessed in MCF-7 cells. The flow cytometry assay was applied to measure the apoptosis and cell cycle. Also, mRNA levels of the Bax, Bcl2, P53, caspase-3, caspase-8, caspase-9, caspase-10, matrix metalloproteinases (MMP)-3, and MMP-9 were analyzed by quantitative real-time PCR to determine the underlying cellular pathways affected by rCTII. The results of this study showed that treatment with 4 µg mL-1 of rCTII enhanced apoptosis through the intrinsic and extrinsic pathways. Also, the increase of the cells' proportion in the sub-G1 phase as well as a reduction in S phase was observed. In addition, the expression of MMP-3 and MMP-9 was decreased in the treated group in comparison to the control group that may contribute to the reduced migratory ability of tumor cells. These experimental results indicate that rCTII has anti-proliferative potential, and so this protein could be a potential drug for BC therapy in combination with other drugs.

Warfarin is the cardinal anticoagulant drug prescribed around the world. Due to stochastic bleeding in patients, it is essential to adjust the dose for every individual. The aim of the present study was to evaluate the frequency of CYP2C9 and VKORC1 gene polymorphisms and their association with warfarin maintenance dose in a sample of cardiovascular patients in Birjand, South-Khorasan province of Iran. Patients with a history of cardiovascular disorders who take warfarin daily were selected. CYP2C9 and VKORC1 gene polymorphisms were detected by polymerase chain reaction-restriction fragment length polymorphism in all participants. A total of 114 patients (mean age: 52.7 ± 14.9 years, M/F ratio: 0.76) participated in this study. Regarding CYP2C9 gene polymorphisms, the most frequent genotype was 1*/1* (80.4% in females and 62.5% in males). The frequency of 1*/2* and 2*/2* variants was 13% and 6.5% in females and 25% and 12.5% in males, respectively. The frequency of VKORC1 gene (1639 G > A), was 31.5%, 39.5%, and 29% for GG, GA, and AA in males, respectively. Besides, the mentioned genotype frequencies for females were 50%, 40.5%, and 9.5%, respectively. Moreover, there was a statistically significant correlation between VKORC1 gene -1639 G > A variant and warfarin maintenance dose (P < 0.001) but not for CYP2C9 variants. The results of the current study confirmed that the mutant variants of CYP2C9 are not frequent and do not have any impact on warfarin dose. In the case of VKORC1, the mutant allele (A) showed a positive correlation with warfarin dose adjustment.

Vitiligo is the most common cause of skin, hair, and oral depigmentation which is known as an autoimmune disorder. Genetic and environmental factors have important roles in the progression of the disease. Dysregulation of gene expression, like microRNAs (miRNA), may serve as major relevant factors. Several biological processes are involved in vitiligo disease and developing a comprehensive approach helps us to better understand the molecular mechanisms of disease. In this research, we describe how a weighted gene co-expression network analysis as a systems biology approach assists to define the primary gene modules, hub genes, and messenger RNA (mRNA)-miRNA regulatory network in vitiligo disease as the novel biomarkers. The results demonstrated a module with a high correlation with vitiligo state. Moreover, gene enrichment analysis showed that this module's genes were mostly involved in some biological activities including G protein-coupled receptors signaling pathway, lymphocyte chemotaxis, chemokine activity, neutrophil migration, granulocyte chemotaxis, etc. The co-expression network was constructed using top hub genes of the correlated module which are named as CXCL10, ARL9, AKR1B10, COX7B, RPL26, SPA17, NDUFAF2, RPF2, DAPL1, RPL34, CWC15, NDUFB3, RPL26L1, ACOT13, HSPB11, and NSA2. MicroRNAs prediction tool (miRWalk) revealed top miRNAs correlated with the interested module. Finally, a drug-target network was constructed which indicated interactions of some food and drug administration (FDA) approved drugs with hub genes. Our findings specified one important module and main hub genes which can be considered as novel biomarkers for vitiligo therapeutic purposes.

Breast cancer (BC) is among the leading causes of mortality from cancer in women. Many of the available anticancer drugs have various side effects. Therefore, researchers are seeking novel anticancer agents particularly from natural compounds and in this regard, snake venom is still one of the main sources of drug discovery. Previous studies showed potential anticancer effects of Cytotoxin II (CTII) from Naja naja oxiana against the different types of cancers. In this study, a pET-SUMO-CTII vector was transformed into SHuffle® T7 Express, an Escherichia coli strain, for recombinant protein expression (rCTII) and the cytotoxic effects of this protein was assessed in MCF-7 cells. The flow cytometry assay was applied to measure the apoptosis and cell cycle. Also, mRNA levels of the Bax, Bcl2, P53, caspase-3, caspase-8, caspase-9, caspase-10, matrix metalloproteinases (MMP)-3, and MMP-9 were analyzed by quantitative real-time PCR to determine the underlying cellular pathways affected by rCTII. The results of this study showed that treatment with 4 μg mL−1 of rCTII enhanced apoptosis through the intrinsic and extrinsic pathways. Also, the increase of the cells' proportion in the sub-G1 phase as well as a reduction in S phase was observed. In addition, the expression of MMP-3 and MMP-9 was decreased in the treated group in comparison to the control group that may contribute to the reduced migratory ability of tumor cells. These experimental results indicate that rCTII has anti-proliferative potential, and so this protein could be a potential drug for BC therapy in combination with other drugs.

The inhalation of Chenopodium album ( C. album ) pollen, especially polcalcin (Che a 3), has been reported as a significant source of allergic respiratory symptoms. This study was conducted to characterize biophysical differences of recombinant polcalcin come from three different Escherichia coli strains using attenuated total reflectance Fourier transform infrared spectroscopy (ATR-FTIR), optimize recombinant polcalcin expression, and linear B-cell epitopes identification using in silico methods. ATR-FTIR results of purified proteins showed spectra intensity variance in the amide I region, while there were no changes in pick position and shape of the bands. SHuffle® T7 Express was selected for subsequent optimization due to ability in the correction of the mis-oxidized bonds and promotes proper folding which was validated by ATR-FTIR analysis results. Then, Response Surface Methodology was performed to optimize critical factors including induction temperature, duration of induction, and concentration of inducer. The best partitioning conditions were achieved in 1.5 mM IPTG for 10.97 h at 29.9 °C. Finally, prediction of polcalcin B-cell epitopes was carried out which indicated the presence of 4 different epitopes. Together, the results may help to the development of diagnostic approaches and also vaccine manufacture for desensitization and modulation of the allergic response in patients.

Tumor tolerance is known as a condition of systemic immune unresponsiveness or immunosuppression against cancers, in which malignant cells escape from the immune surveillance. Among the elements of immune tolerance orchestra, the CD4+CD25 + forkhead box protein3+ (Foxp3+) regulatory T cells (Tregs) have a great importance. Moreover, increased Treg populations and, particularly, reduced proportions of effector T cells to Tregs in tumor-infiltrating lymphocytes are closely linked to worse prognosis in different tumors. In the last years, several drugs for cancer immunotherapy have been developed with very promising results; however, different resistance mechanisms to immunotherapy may limit their effectiveness. Plant‐derived natural agents are emerging as an innovative therapeutic field. Particularly, a number of phytochemicals, by altering diverse immune signaling pathways, have shown to possess immunomodulating features; hence, they have been tested in multiple cancer experimental models in order to contrast the tumor-induced immunosuppressive behavior, particularly Tregs-mediated immune evasion in the tumor microenvironment. In this review, recent advances on the anti-cancer effect of natural agents, either alone or in combination with traditional drugs, due to the modulation of classic Treg cells, have been resumed with the aim to guide researchers for their potential future clinical use.

The coronavirus disease-2019 (COVID-19) pandemic has caused an enormous loss of lives. Various clinical trials of vaccines and drugs are being conducted worldwide; nevertheless, as of today, no effective drug exists for COVID-19. The identification of key genes and pathways in this disease may lead to finding potential drug targets and biomarkers. Here, we applied weighted gene co-expression network analysis and LIME as an explainable artificial intelligence algorithm to comprehensively characterize transcriptional changes in bronchial epithelium cells (primary human lung epithelium (NHBE) and transformed lung alveolar (A549) cells) during severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection. Our study detected a network that significantly correlated to the pathogenicity of COVID-19 infection based on identified hub genes in each cell line separately. The novel hub gene signature that was detected in our study, including PGLYRP4 and HEPHL1, may shed light on the pathogenesis of COVID-19, holding promise for future prognostic and therapeutic approaches. The enrichment analysis of hub genes showed that the most relevant biological process and KEGG pathways were the type I interferon signaling pathway, IL-17 signaling pathway, cytokine-mediated signaling pathway, and defense response to virus categories, all of which play significant roles in restricting viral infection. Moreover, according to the drug–target network, we identified 17 novel FDA-approved candidate drugs, which could potentially be used to treat COVID-19 patients through the regulation of four hub genes of the co-expression network. In conclusion, the aforementioned hub genes might play potential roles in translational medicine and might become promising therapeutic targets. Further in vitro and in vivo experimental studies are needed to evaluate the role of these hub genes in COVID-19.

There is increasing evidence that maternal diabetes mellitus during the pregnancy is associated with a higher risk of neurodevelopmental and neurofunctional anomalies including motor dysfunctions, learning deficits, and behavioral problems in offspring. The cerebellum is a part of the brain that has long been recognized as a center of movement balance and motor coordination. Moreover, recent studies in humans and animals have also implicated the cerebellum in cognitive processing, sensory discrimination, attention, and learning and memory. Synaptogenesis is one of the most crucial events during the development of the central nervous system. Synaptophysin (SYP) is an integral membrane protein of synaptic vesicles and is considered to be a marker for synaptic density and synaptogenesis. Here, we review the manuscripts focusing on the negative impacts of maternal diabetes in pregnancy on the expression or localization of SYP in the developing cerebellar cortex. We believe that the alteration in synaptogenesis or synapse density may be part of the cascade of events through which diabetes in pregnant women affects the newborn's cerebellum.

Introduction Cervical cancer is the fourth most common cancer in females demonstrating approximately more than 7% of all woman cancer mortality in 2018. HPV infection considered as the pivotal risk factor for cervical cancer induction. Permanent infection with carcinogenic genital HPVs increases the predisposition of cervical cancer. In this study, we investigated the frequency of HPV genotypes in a sample of Iranian population. Methods Therefore, 2135 individuals with genital warts between 2015 and 2018 recruited to the study. After DNA extraction, PCR amplification and genotyping were employed by HPV Direct Flow CHIP Kit and the e-BRID System®, supported by hybriSoft ™. HPV was found in 92% of the samples obtained from the patients. Results In total, 12.6% and 87.4% of warts were positive for any high-risk and low-risk genotypes, respectively. HPV 16 and 6 were the most predominant HR and LR genotypes in all categorized age groups (5.26% and 40.71%, respectively). Moreover, by getting older, the frequency of HPV 6 was decreased (P = 0 .018, r = −0.050) and the frequency of HPV 56 was increased (P =0 .026, r = 0.044). Conclusion Although we noticed lower prevalence of high risk HPV particularly 16 and 18 in genital warts in the East of Iran; however, genotyping of genital warts still recommended.

Activation of NOD-like receptor (NLR) family and pyrin domain containing 3 (NLRP3) inflammasome contributes to inflammation and may lead to atherosclerosis. The NLRP3 inflammasome as a molecular platform regulates the activation of ATP signaling, K+ efflux, cathepsin-B activity, lysosomal function and pro-inflammatory cytokines (i.e. IL-1β and IL-18). Statins has been widely prescribed for the treatment of hyperlipidemia and cardiovascular diseases. In addition to lipid-lowering effect, statins have immunomodulatory, anti-inflammatory, antioxidant and antiapoptotic functions. An increasing number of studies indicated NLRP3 inflammasome and their downstream mediators as important targets for statin drugs in inflammatory diseases. In this review, we discussed different aspect of the NLRP3 inflammasome signaling pathways and focused on the effect of statin drugs on NLRP3 inflammasomes in association to atherosclerosis in order to elucidate possible targets for future research and clinical settings.

Background and Objectives: Hepatitis C is a dangerous viral infection, which is the important cause of chronic liver diseases such as fibrosis, cirrhosis, and hepatocellular carcinoma. The hepatitis C virus (HCV) has six major genotype of HCV identified for which are different in prevalence, carcinogenicity, persistency, and response to the treatment. Identification of HCV genotype and viral load are main factors for planning of the treat and prediction of response. There has not been any study regarding the viral load and genotype distribution of HCV in our area and this study was done to evaluate risk factors, viral load, and genotype of HCV among prisoners in East Iran. Methods: In a cross-sectional study, 2,995 prisoners across the South Khorasan province were screened for the anti-HCV antibody by ELISA method. A total of 38 prisoners with positive results were selected for further evaluation. Demographic data as well as predisposing factors were obtained from the records. A total of 5 ml of venous blood was taken and after RNA extraction, viral load genotype and HCV were identified by using the commercial Real Time PCR and PCR method, respectively. Results: Among 2,995 prisoners, 34 cases (mean age 39 ± 8.8 years, M/F ratio: 13.1) were positive for HCV antibody. A total of 50% were married, 79.4% were urban citizens, and 97.1% of patients infected with HCV had drug addiction. In 28 cases (82.3%), the virus was detected by real time PCR method. Genotype 3a and 1a were the most prevalence genotypes (75% and 8.3%, respectively). The average of viral load was 123,798 IU/mL. There was no significant relation between age or sex with viral load and genotype. Discussion and Conclusions: This study showed that the predominant genotype in prisoners is the genotype 3a, which adds to the evidence of the predominant genotype in Iran. The genotype of injecting drugs prisoners is different to other groups infected with Hepatitis C in Iran.

This study was conducted to examine in vitro antibacterial potential from seed methanol extract of Peganum harmala (L) against some fish pathogenic bacteria including Lactococcus garvieae, Aeromonas hydrophila, Yersinia ruckeri and Pseuodomonas putida isolated from diseased rainbow trout (Oncorhynchus mykiss). The antibacterial activity of extracts was evaluated using disc diffusion assay, minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC). MICS were measured by serial dilution and the microplate assays. Results showed that the methanol extract of P. harmala was bactericidal for all test bacteria. The MICs of extract using serial dilution and microplate method were 0.6 mg mL -1 and 0.312 to 0.625 mg mL -1 against P. putida and 0.8 mg mL -1 and 0.625 to 1.25 mg mL -1 against L. garvieae, A. hydrophila, Y. ruckeri, respectively which was confirmed by MBC determination. Thus, the antibacterial activity of seed extract of P. harmala can be comparable as an alternative in the control of infectious by these microorganisms.

Background: Hepatitis B virus (HBV) is the main infectious agent that causes liver disease and may lead to an acute or chronic HBV infection. Evidence from many studies have determined that host genetic factors play a significant role in determining immunization, clinical course, and recovery from HBV infection. A 32-bp deletion in the region of CCR5 gene (CCR5 ∆32) is one of the mutations that is known to provide genetic protection against chronic HBV infections. Objectives: In this study, the researchers aimed at assessing the protective effect of CCR5 ∆32 in subjects, who had recovered from HBV infection, as well as subjects that developed chronic HBV infection, in Birjand, Iran. Methods: The study consisted of 60 patients with chronic HBV infection (patient group) and 120 patients, who had previously recovered from HBV infection (control group). Genomic DNA was extracted from blood samples by the salting out method, and then samples were analyzed for the CCR5 ∆32 genotype utilizing the gap-polymerase chain reaction (Gap-PCR) technique. Variables were analyzed using the Chi-square test and P values less than 0.05 were considered as statistically significant. Results: The researchers found only one heterozygous CCR5 ∆32 mutation in the control group and absolutely no homozygous CCR5 Δ32 mutation in either the patient or control group. Conclusions: Because of no significant visible correlation between carrying the mutation and the possibility of recovery from HBV infection in the city of Birjand, it seems the protective effect of this mutation is absent in Birjand’s population.

Background It is estimated that about 5% of hepatitis B surface antigen (HBsAg) carriers are infected with hepatitis D virus (HDV) worldwide. Moreover, dual infection with hepatitis B virus (HBV) / HDV causes more severe diseases and leads to a higher risk of mortality.Objectives This study was performed to determine the prevalence of HDV infection and identify HDV-associated factors among HBsAg-positive patients in Birjand, South Khorasan province, Iran.Methods This descriptive, cross sectional study was conducted on 5235 participants in line with a previous study on the seroprevalence of HBV in South of Khorasan. HDV was assessed through evaluating the blood serum of diagnosed HBsAg-positive patients (85 cases) by ELISA test. For statistical analysis, descriptive statistics were calculated, using SPSS version 22.Results In this study, the mean age of HBsAg-positive patients was 45.1 ± 12.9 years (minimum, 22 years; maximum, 70 years). In total, 1 out of 85 cases was positive for HDV virus. The prevalence of HDV infection among HBsAg-positive patients was 1.2% (95% CI, - 1.2 to 3.5).Conclusions The low prevalence of HDV infection in South of Khorasan was remarkable, considering the great area of this province, use of a comprehensive and inclusive sampling strategy, and mutual borders with Afghanistan.

Background: Hepatitis E infection is one of the most common causes of acute hepatitis. It is caused by the RNA virus of the hepatomegaly viridae family, which is transferred through stool in contaminated water. Cases of infection are most prevalent in developing countries. Objectives: This study was conducted to investigate the seroprevalence of hepatitis E among urban adults. The reason was the current lack of information on the hepatitis E prevalence among the general population in the city of Birjand in eastern Iran. This information is necessary because Southern Khorasan is in close proximity to Afghanistan. Methods: In this cross sectional study, 522 participants aged over 17 years were selected from the city of Birjand, eastern Iran, using the random cluster sampling. The participants completed a researcher-made questionnaire that included demographic characteristics, risk factors for infectious diseases such as records of blood transfusions, specific exposures, intravenous drug abuse, recent diagnosis or a history of liver disease, and other medical information. After selection, a blood sample was taken from each participant to a volume of 5 cc. Each blood sample was then measured for the title of IgG antivirus of hepatitis E by the enzyme-linked immunosorbent assay (ELISA) method using the third generation ELISA kit made by Delavara Company. Results: This study included 554 participants. The mean age of the participants was 41.4 years (age range, 17 - 72 years). From a total of 554 participants, 295 (53.2%) were females and the remainder were males. Results showed that the hepatitis E antibody title was positive in 139 (25.1%) participants and negative in the others. Comparison of the prevalence rates of positive HEV between men and women showed no statistically significant difference although it was significantly higher in married participants than in single ones (P < 0.001). Results also showed that the prevalence was less in participants with a tertiary education compared to those with a lower level education. Along with age, the prevalence of the positive antibody title increased significantly. The prevalence of HEV in participants who were smokers was significantly (P < 0.001) lower than in those who were nonsmokers. Conclusions: In this study, the seroprevalence of contact with HEV in urban adults in the city of Birjand was 25%. The results of this study showed that the prevalence of hepatitis E in the city of Birjand, eastern Iran, was higher than in other places compared with previous studies on communities in Iran. Also, the prevalence of HEV in single and educated people was significantly less than that in married and literate individuals.

In the current study, laboratory evaluations were made to assess the immunomodulatory effect of cypermethrin on fingerlings of common carp (Cyprinus carpio L.). Results showed that 96-h LC50 of cypermethrin in common carp was estimated at 0.85 μg/L. Fish were exposed for 21 days to cypermethrin at three sub-lethal concentrations of 0.042, 0.085, and 0.17 μg/L that represented 5, 10, and 20%, respectively, of the 96-h LC50 of the pesticide for this fish species. Blood samples were taken after 7, 14, and 21 days of exposure. Immunological indices and resistance against bacterial infection were determined. Allergic fungal sinusitis (AFS) is a noninvasive form of fungal rhinosinusitis with a prevalence of 6-9 % among all rhinosinusitis cases requiring surgery. The fungi causing AFRS have a great diversity and regional variation in the incidence of AFS has been reported worldwide. The aim of this study was to evaluate the prevalence of AFS among rhinosinusitis patients in the north east of Iran. In a prospective cross-sectional study 127 patients with sinonasal polyposis who were candidates for surgery were recruited. All cases were evaluated by sinonasal CT scan, Prick test for common regional allergens, total serum immunoglobulin E (IgE) level, and allergic mucin culture for fungi in suspected cases. Data analyses were done using SPSS version 13.0. Their mean age was 37.4 ± 11.6 years, the male-to-female ratio was 69/58. Eleven patients (9.45 %) met the AFS criteria. Nine cases had a positive culture for Aspergillus specimen and three for Alternaria. Prick test was positive for at least one allergen in 28 cases (22.05 %). The most common allergen was weed. The total IgE level was significantly higher in AFS patients. This study showed the prevalence of AFS among patients with nasal polyposis to be 9.45 %.

Background: Infection with the hepatitis B virus (HBV) is an important global health problem. Knowledge of the geographic dis- tribution pattern of HBV infection can help to control the spread of the disease. Objectives: To determine the prevalence of HBV infections and risk factors for the disease for the first time in Birjand, Southeastern Iran. Methods: This was an analytical cross-sectional study conducted in 2013 - 2014 of 5235 HBV cases in Birjand, South Khorasan Iran. Subjects aged 15 - 70 y were selected using the cluster sampling method. Blood samples were taken and tested at a reference labora- tory for the hepatitis B core antibody (anti-HBc). Seropositive specimens were tested for the hepatitis B surface antigen (HBsAg). Results: The mean age ( SD) was 39.07 ( 14.04) y, and 786 (15%) subjects were anti-HBc positive. The prevalence of HBsAg was 1.6% (n = 85). The prevalence of anti-HBc seropositivity was significantly higher in subjects with a lower level of education (P = 0.09), older subjects (P = 0.001), intravenous (IV) drug users (P

Allergic disorders are common ailments, which cause socioeconomic problems. The aim of the study was to determine the prevalence of allergic disorders and symptoms suggestive of asthma in Mashhad city, Northeast Iran. We performed a cross-sectional survey in a randomly selected representative sample of the Mashhad population. A questionnaire was used based on the standard questionnaire of the European Community Respiratory Health Survey (ECRHS). The questionnaires asked about allergy-related symptoms including nasal symptoms, eye symptoms, atopic dermatitis symptoms, confirmed asthma, asthma-related symptoms, and nonspecific allergic symptoms. Some of the environmental triggers of allergic reactions such as pollen, dried plant, dust, drugs, food, fruit, condiments, makeup, perfume, jewelry, latex gloves, and keeping a pet were considered in the questionnaire. The prevalence of allergic disorders in this study was found to be 27.5% in the city of Mashhad. According to our definitions, the rates detected of allergic rhinitis, atopic dermatitis, conjunctivitis, rhinoconjunctivitis, and asthma were 22.4, 6.6, 13.5, 9.5, and 2.3%, respectively. Half of the allergic group suffered from rhinorrhea and other symptoms were significantly higher than in the non-allergic group. Dust and pollen were found to be the most important triggers of allergic reactions in the allergic group. This report may be useful to understand the factors contributing to asthma and allergic disorders and may help in management and control of these problems.

Knowledge about the prevalence, risk factors and triggers of allergic disorders among populations of different geographical areas is important for the development of preventative strategies. The aim of this study was to investigate the prevalence, clinical pattern and triggers of allergic disorders among the inhabitants of the rural town, Boshrooye, in Iran. A written symptoms questionnaire derived from the standard questionnaire of the European Community Respiratory Health Survey (ECRHS) in the Farsi language was used. The prevalence of atopy, allergic rhinitis, atopic dermatitis, allergic conjunctivitis and physician-diagnosed asthma was 18.1%, 15.5%, 11.5%, 8.2% and 0.3%, respectively. Rhinorhea, sneezing and nose blockage were the most frequent symptoms among atopic cases (37.7%, 33% and 28.8%, respectively). Dust and pollen were the most important triggers of allergic symptoms. The findings of this study indicate a low to moderate prevalence of allergic disorders in a rural area in Iran and emphasize the importance of dust and pollens as the main triggers of allergic symptoms.

Nasal polyposis (NP) is a common inflammatory disorder with different proposed etiologies. There is some evidence that the local production of specific IgE against fungal allergens may contribute to the pathogenesis of this condition. The aim of this study was to compare local and systemic production of fungal-specific IgE among patients with NP. In a prospective cross-sectional study, 141 patients with sinonasal polyposis who were surgical candidates were enrolled. Total IgE and fungal-specific IgE antibodies in serum and tissue homogenates were measured by the ELISA and ImmunoCAP method, respectively. Twelve cases (9.45 %) met the allergic fungal rhinosinusitis (AFRS) criteria while the remaining 129 patients showed histopathological features of chronic hyperplastic rhinosinusitis. Considering a cutoff point of 0.4 KUI/L for specific IgE as a positive reaction against fungal elements, seven and 62 cases showed positive results in serum and homogenate tissue, respectively. All serum positive reactions were seen among patients with AFRS. In conclusion, IgE antibody could be locally produced in the nasal polyp tissue while not detectable in the serum.

Background Exposure to house dust mites (HDMs) is a major risk factor for the development of allergic symptoms. HDMs are worldwide in distribution. Assessing these allergens in each area is a critical step in evaluating the risk of sensitisation and controlling allergic symptoms. The aim of the present study was to evaluate the level of major HDMs, Der p 1 and Der f 1, in various parts of Iran. Methods In 2009, 257 dust samples were obtained from living rooms’ carpets in seven cities throughout Iran with different geoclimatic conditions. The level of Der p 1 and Der f 1 was measured by commercial ELISA. Results Detectable level of Der p 1 and Der f 1 levels were only found in Gorgan and Sari, two cities near the Caspian Sea with moderate temperatures and high relative humidity. In both of these cities, Der f 1 was more frequent than Der p 1 (100% vs. 85%) and was found to be at a higher level than Der p 1 (geometric mean 3128 vs. 439 ng/g dust, P Conclusion The results of this study confirm that house dust mites in Iran are restricted to the Caspian Sea coastal areas, and in other parts, due to seasonal variations of temperature and humidity mites are not able to grow well and therefore are not an important risk factor for sensitisation and respiratory allergies.

Allergic inflammation in upper airways can act as a predisposing factor for infectious ear diseases. There are some evidences about the role of allergic rhinitis in chronic otitis media with effusion, but its role in establishing chronic suppurative otitis media (CSOM) has not been clearly shown. 68 adult patients with established CSOM, who were candidates for ear surgery, and 184 age- and sex-matched controls were evaluated for the presence of allergic rhinitis. Standard questionnaire was filled out for all participants. All patients and controls underwent skin prick test for 28 com- mon regional aeroallergens, and serum total IgE was mea- sured by means of ELISA method. Allergic rhinitis were defined as a positive responses to the questionnaire, positive skin prick test to at least one allergen, and/or high level of serum total IgE. Allergic rhinitis was diagnosed in 20 (29.41%) and 41 (22.28%) of patients and controls, respectively (P = 0.241) (OR = 1.28, CI = 0.69-2.36). Outdoor allergens, especially grass pollen, were the most prevalent allergens among both groups, but indoor allergens like mites and molds have a low prevalence. The study did not show a significant difference in the prevalence of AR in the CSOM patients compared to the controls. The inter- mittent nature of allergy and other less known intervening factors in the etiopathogenesis of CSOM make such a con- clusion difficult.

Background: Allergic rhinitis is the most common allergic disease with negative impacts on patients’ quality of life. The prevalence and pattern of sensitisation vary between different countries and populations. Identifi cation of the most prevalent aeroallergens in each area has a very important role in diagnosis and treatment of allergic rhinitis. Iran is a fairly large country with different geo-climatic conditions and there are no data about the most prevalent aeroallergen in Mashhad City, which is the second largest city in Iran, with a semiarid climate. The aim of this study was to evaluate the prevalence of positive skin test to various common aeroallergens among allergic rhinitis patients in the city of Mashhad. Materials and Methods: Skin prick tests were performed with 27 common regional aeroallergens including grass, weed, tree, mite and mould in 311 patients with allergic rhinitis. Results: The overall rate of sensitisation to any allergen was 81 %. 76 % of patients were poly-sensitised and weed and grass were the most prevalent allergens (77 % and 62 % respectively). Salsola Kali and mould were the most and the least prevalent individual allergens (72.5 % and 6.5 % respectively). Mean total IgE in patients with positive skin prick test was signifi cantly higher than in patients without any positive skin prick test (308 vs 128 IU/mL, P = 0.016). 97 % of atopic patients were sensitised to Salsola kali and or Fraxinus americana. Conclusions: Our work showed the importance of weeds, especially the Amaranthaceae and Chenopodiaceae families. Diagnosis of pollen allergy can be simplifi ed by using a combination of a few common aeroallergens.

Background: Invariant natural killer T cells (iNKT) are a small subset of T lymphocytes which are involved in a wide variety of immune responses. Human studies have reported a wide variation in the number of circulating iNKT cell among healthy donors as well as in some immunologic disorders while there is no or little data about the factors which affect the pool of circulating iNKT cells in human. Given the importance of iNKT cells in immunologic responses and variation in the number and phenotype of iNKT cells, the aim of this study was to evaluate the frequency of circulating iNKT cells in a cohort of healthy Iranian adults. Methods: The number of circulating iNKT cells was measured among PBMC in a group of healthy Iranian adults by flow cytometry. iNKT cells were identified by using 6B11 monoclonal antibody or by coexpression of TCR Vα24 and Vβ11 chain. Results: The mean frequency of 6B11-iNKT cells and Vα24-Vβ11 iNKT cells among T lymphocytes, were 0.54% and 0.31%, respectively. Forty-five percent of 6B11-iNKT cells were CD4 positive, and there was a weak but significant inverse correlation between total 6B11-iNKT cells and their CD4-positive subset. There was no significant difference in the frequency of iNKT cells between males and females. Conclusions: Results of this study show that the frequency of iNKT cells is fairly high among the Iranian population and suggest matching of study population for age, sex, and ethnic background in iNKT-related research. More studies are necessary to understand the factors that can affect the variation in the number of iNKT cells in healthy humans. © 2009 Clinical Cytometry Society
