1. A Delayed Mass-Action Model for the Transcriptional Control of Hmp, an NO Detoxifying Enzyme, by the Iron-Sulfur Protein FNR

Abstract

In Escherichia coli, an enzyme called Hmp is a key contributor to the detoxification of nitric oxide (NO). In the absence of NO, the transcription of the hmp gene is repressed by an iron-sulfur protein called FNR. NO damages the iron-sulfur cluster of FNR, weakening the repression of hmp and allowing expression of Hmp to high levels. A delayed mass-action model for the Hmp-FNR network has been developed. This model has 33 parameters, all but three of which were estimated. One of the unknown parameters, the rate of NO inflow into the cell’s cytoplasm, was used as a control parameter in a study of the steady-state structure of this model. This study revealed bistability across a wide range of inflow rates, oxygen concentrations, and values of the unknown parameters. The bistability is caused by substrate inhibition of Hmp by NO, which allows for a high-NO steady state, which would likely be lethal, to coexist with a biologically desirable low-NO steady state.

Abstract

A mathematical model is devised to study the diffusion of mRNA in the nucleus from the site of synthesis to a nuclear pore where it is exported to the cytoplasm. This study examines the role that nuclear structure can play in determining the kinetics of export by considering models in which elements of the nuclear skeleton and confinement by chromatin direct the mRNA movement. As a rule, a dense chromatin layer favours rapid export by reducing the effective volume for diffusion. However, it may also result in a heavy tail in the export time distribution because of the low mobility of molecules that accidentally find their way deep into the dense layer. An anisotropic solid-state transport system can also assist export. There exist both an optimal ratio of the anisotropy and an optimal depth of the solid-state transport layer that favour rapid export. [Includes supplementary material]

Abstract

The Prehospital Fluid Conference was sponsored by the US Army Institute of Surgical Research and Combat Casualty Care Research, US Army Medical Research and Materiel Command. Some 65 conferees were invited in January 2010 to review the contemporary guidelines on the use of fluid resuscitation in treating combat casualties, discuss the state of the art of fluid resuscitation for combat casualties, and answer the following questions: - Are current Tactical Combat Casualty Care (TCCC) intravenous (IV) fluid resuscitation guidelines optimal for today? - Which IV fluid should be the top priority for future research? - What are the current indications for fluid resuscitation in the combat trauma patient? - What is the current practice in tactical fluid resuscitation? The objective of this conference was to identify the fluid to be used by the prehospital provider and not to address the needs once definitive hemorrhage control has been achieved. The fluids to be used are those that will be carried into the field on the back of the combat medic or in the vehicle used to transport the medic or the patient (echelon 1 care). There are several definitions of these echelons of care (North Atlantic Treaty Organization [NATO], European forces, etc.). The definitions used in this document are those of the Joint Theater Trauma System. The use of role and echelon can be interchangeable depending on the country of origin, but the terms can vary significantly. Echelon, as used in many/most of Joint Theater Trauma System presentations will be used throughout this document to indicate a level of care facility (see Table 1). Although discussions focused on military needs, it was understood and accepted by the consensus group that many, if not all, of the recommendations could and would be used for civilian prehospital providers with appropriate situational modifications.

Abstract

The inflammatory cytokine, TNF-alpha, induces IL-8 gene transcription via a mechanism involving proteasome-mediated IkappaBalpha degradation and NF-kappaB activation. Here, we investigated whether arsenic, which has been shown to inhibit the ubiquitin-proteasome pathway, could inhibit TNF-alpha-mediated increases in IL-8 expression. Using RT-PCR, we show that the addition of TNF-alpha to human bronchial epithelial (BEAS 2B) or embryonic kidney (HEK293) cells resulted in increased steady-state levels of IL-8 mRNA. This was preceded by a rapid decrease in cellular IkappaBalpha levels, as demonstrated by Western analysis, and an increase in nuclear levels of NF-kappaB, as demonstrated by gel shift analysis. Further demonstrating the activation of NF-kappaB, TNF-alpha induced the transcription of a NF-kappaB-dependent reporter gene. Exposing the cells to 500 microM arsenite, prior to adding TNF-alpha, completely inhibited IkappaBalpha degradation, NF-kappaB translocation, NF-kappaB-dependent gene transcription, and transcription of the endogenous gene for IL-8. In comparison with the proteasome inhibitor MG-132, which does not affect the phosphorylation and ubiquitination of IkappaBalpha, arsenite inhibited the phosphorylation of IkappaBalpha. Furthermore, arsenite directly blocked the activity of IKK, the kinase responsible for IkappaBalpha phosphorylation. These studies demonstrate that high levels of arsenic may inhibit NF-kappaB-mediated gene transcription by specifically blocking IKK activity, thereby limiting the phosphorylation and subsequent degradation of the NF-kappaB inhibitor, IkappaBalpha.

Abstract

In earlier studies, treatment with sodium arsenite was shown to decrease total hepatic CYP in rats. A concomitant increase in heme oxygenase, the rate-limiting step in heme degradation to biliverdin, was considered responsible for the decrease in CYP. Here we investigated the effect of sodium arsenite on induction of CYP2H, CYP1A, and heme oxygenase in primary cultures of chicken embryo hepatocytes. When added simultaneously with inducer, arsenite inhibited phenobarbital-mediated increases in CYP2H and 3-methylcholanthrene-mediated increases in CYP1A, as measured enzymatically and immunochemically. Near maximal decreases were observed in these forms of CYP at a concentration of 2.5 microM sodium arsenite. The concentration-dependent decreases in CYP2H and CYP1A by sodium arsenite were concomitant with increases in heme oxygenase. Sodium arsenite was not toxic at concentrations as high as 10 microM, as indicated by protein synthesis and the reduction of MTT by intact cells. Sodium arsenite had no effect on induction of CYP2H1 mRNA, suggesting that the decreases in this form of CYP occurred post-transcriptionally. Treatment of cells with tin mesoporphyrin (SnMeso), an inhibitor of heme oxygenase, resulted in inhibition of arsenite-induced heme oxygenase. However, SnMeso did not alter the effect of arsenite to prevent phenobarbital-mediated increases in CYP2H protein. SnMeso alone inhibited phenobarbital-mediated increases in CYP2H. Inclusion of 2 or 5 microM exogenous heme with arsenite did not prevent the arsenite-mediated decrease in CYP2H. Combined treatment with heme and phenobarbital induced heme oxygenase to the same extent as treatment with heme, arsenite, and phenobarbital. However, CYP2H activity was decreased only when the treatment included arsenite. These results suggest that elevated levels of heme oxygenase alone are not responsible for arsenite-mediated decreases in CYP2H.

Abstract

It has been noted that single-enzyme systems can undergo strongly damped transient oscillations. In this paper, we present a nonlinear dynamics analysis of oscillations in undriven chemical systems. This analysis allows us to classify transient oscillations into two groups. In the first group, oscillations arise from rapid oscillatory relaxation to a slower transient relaxation mode. These oscillations are always strongly damped. In the second group, it is the slowest relaxation mode which is implicated in the oscillations so these can be very lightly damped. This second class of oscillations has not previously been studied in enzymology. We show that a remarkably simple single-enzyme system, namely competitive inhibition with substrate flow, generates transient oscillations which belong to the second class. In an attempt to design an experimentally realizable version of this model, we then discovered a system which is capable of sustained oscillations. In this experimentally realizable model, two substrates compete to bind to a macromolecule. The flow of one substrate is controlled by a simple feedback device. Sustained oscillations are observed over a very wide range of parameters. In both models, oscillations are favored by a wide disparity in rates of binding and dissociation of the two substrates to the macromolecule.

Abstract

Glucocorticoid therapy is being used in a wide variety of systemic disorders. Reference papers, published more than 20 years ago, showed no correlation between adrenal insufficiency risk and dose or duration of glucocorticoid therapy.Our objective was to evaluate the extent to which long-term glucocorticoid therapy damages the pituitary-adrenal axis in patients with systemic inflammatory disorders.We conducted a retrospective observational study from January 2011 to August 2012.This was a monocentric study at the Department of Internal Medicine, Bichat Hospital, Paris-Diderot University, Paris, France.Sixty consecutive patients who were receiving long-term prednisone therapy for systemic inflammatory disorders and in whom discontinuation of glucocorticoid treatment was planned.A short Synacthen test was performed. A bolus of 0.25 mg 1-24-ACTH was injected in the morning, 24 hours after the most recent dose of prednisone. Cortisol was measured at baseline and 60 minutes after Synacthen injection.We assessed frequency and risk estimate of pituitary-adrenal dysfunction.Twenty-nine patients (48.3%) had adrenal insufficiency defined by a plasmatic cortisol100 nmol/L (n = 13) at baseline (time 0) or550 nmol/L (n = 16) 60 minutes after Synacthen injection. Cumulative dose (area under the receiver operating characteristic curve = 0.77 [95% confidence interval = 0.62-0.91], P = .007) and exposure (area under the receiver operating characteristic curve 0.80 [95% confidence interval = 0.67-0.93], P = .002) to prednisone were predictive for adrenal insufficiency based on a T0100 nmol/L. Prednisone was stopped in 29 of 31 patients (93.5%) showing a normal response to short Synacthen test; none of these patients required hydrocortisone replacement with a mean follow-up of 10 (± 6) months.Adrenal insufficiency is frequent in patients treated with long-term glucocorticoids for systemic inflammatory disorders and is related to duration and cumulative dose of steroids.

Abstract

Although the absence of intracellular (IC) free glucose is direct evidence of glucose transport being the rate-limiting step for muscle glucose disposal at euglycemia, the scarcity of data in humans during hyperglycemia precludes any definitive conclusion. In the present study, 13C and 31P in vivo nuclear magnetic resonance (NMR) data from two separate groups of subjects were combined to measure IC free glucose in the human skeletal muscle. When these noninvasive tools were used with an infusion of [1-13C]glucose, a steady-state concentration of 1.2 +/- 0.2 mmol IC glucose/l IC water was observed at the end of a 2-h hyperglycemic clamp with somatostatin infusion, during which glycemia was maintained at approximately 22 mmol/l and insulinemia at approximately 5 mU/l. Despite this moderate glucose accumulation, the persistence of a large transmembrane glucose gradient suggests that the posttransport steps do not play a significant role in the control of muscle glucose disposal in these specific conditions, relevant to insulinopenic diabetic patients.

Abstract

In early embryonic development, the cell cycle is paced by a biochemical oscillator involving cyclins and cyclin-dependent kinases (cdks). Essentially the same machinery operates in all eukaryotic cells, although after the first few divisions various braking mechanisms (the so-called checkpoints) become significant. Haase and Reed have recently shown that yeast cells have a second, independent oscillator which coordinates some of the events of the G1 phase of the cell cycle.(1) Although the biochemical nature of this oscillator is not known,it seems unlikely to be a redundant cyclin/cdk system. Abstract

Substance use disorders constitute a serious and persistent threat to military readiness and to the health and safety of military personnel and their families. Methamphetamine is among the most addictive and damaging of commonly abused drugs; this is of great concern for military health providers in Hawaii due to the unusually high prevalence in the local community. The effect of regional drug use on active duty subpopulations has not been previously studied. This study includes a 6-year retrospective sample of laboratory-confirmed methamphetamine-, cocaine-, and marijuana-positive drug tests among Army soldiers stationed in Hawaii and western and eastern continental U.S. installations. The findings suggest that active duty members are significantly affected by the local drug climate. However, current military drug policies also deter use as evidenced by low absolute drug-positive rates even in regions of high civilian prevalence.

Abstract

It is possible to produce anti-phosphopeptide antibodies (i.e., antibodies recognizing phosphorylated peptides) that recognize a protein only in its phosphorylated state, and that do not cross-react with either the cognate unphosphorylated protein or other phosphoproteins. Unlike conventional antibodies, anti-phosphopeptide antibodies provide information regarding not only the abundance of a protein but also its activity. Also, unlike general anti-phosphoamino acid (e.g., anti-phosphotyrosine) antibodies, which have broad reactivity, anti-phosphopeptide antibodies may have unique specificity toward the cognate proteins. Such reagents not only facilitate conventional in vitro analysis of phosphoproteins, but also allow heretofore impossible applications, e.g., differential isolation of species of a particular protein that have been phosphorylated at individual phosphorylation sites, as well as analysis of the functional state of a protein in situ by immunohistochemical techniques. This unit provides protocols for the production of both polyclonal and monocloncal anti-phosphopeptide antibodies. Support protocols are provided for the coupling of peptides and phosphotyrosine to the affinity matrix (Affi-Gel 10); BSA-agarose affinity matrix is commercially available.

Abstract

The aim of the present study was to investigate the effect of blood pressure lowering and intensive glucose control on the incidence and progression of retinopathy in type 2 diabetic patients. The Action in Diabetes and Vascular Disease: Preterax and Diamicron MR Controlled Evaluation (ADVANCE) Retinal Measurements Study, a sub-study of ADVANCE, is a randomised (using a central, computer-based procedure) controlled 2 × 2 factorial trial comprising a double blind comparison of blood pressure lowering with perindopril-indapamide vs placebo, and an open comparison of standard vs intensive glucose control targeting a HbA1c inferior or equal to 6.5% in 1,602 diabetic patients from ADVANCE centres with access to retinal cameras conducted from 2001 to 2008. At baseline and final visit, seven-field stereoscopic retinal photographs were taken and graded by blinded readers (gradable baseline and final photographs from 1,241 patients). Progression of more or less 2 steps in the Early Treatment of Diabetic Retinopathy Study Classification (using the eye with worst grading) was the primary outcome. Retinopathy progressed in 59 (4.8%) patients and developed in 128 (10.3%) patients over 4.1 years. Fewer patients on blood pressure-lowering treatment (N = 623) experienced incidence or progression of retinopathy compared with patients on placebo (N = 618), but the difference was not significant (OR: 0.78; 95% CI: [0.57–1.06]; P = 0.12). Blood pressure-lowering treatment reduced the occurrence of macular oedema (OR: 0.50; 95% CI: [0.29–0.88]; P = 0.016) and arteriovenous nicking compared with placebo (OR: 0.60; 95% CI: [0.38–0.94]; P = 0.025). Compared with standard glucose control (N = 611), intensive glucose control (N = 630) did not reduce (P = 0.27) the incidence and progression of retinopathy (OR: 0.84; 95% CI: [0.61–1.15]). Lower, borderline significant risks of microaneurysms, hard exudates and macular oedema were observed with intensive glucose control, adjusted for baseline retinal haemorrhages. These effects of the two treatments were independent and additive. Adverse events in the ADVANCE study are reported elsewhere. Blood pressure lowering or intensive glucose control did not significantly reduce the incidence and progression of retinopathy, although consistent trends towards a benefit were observed, with significant reductions in some lesions observed with both interventions. Abstract

Chronic human exposure to low levels of inorganic arsenic increases the incidence of vascular diseases and specific cancers. Exposure of endothelial cells to environmentally relevant concentrations of arsenic trioxide (arsenite) induces oxidant formation, activates the transcription factor NF-kappaB, and increases DNA synthesis (Barchowsky et al., Free Radic. Biol. Med. 21, 783-790, 1996). We show, in the current study, that arsenite induces concentration-dependent cell proliferation or death in primary porcine aortic endothelial cells. Low concentrations caused cell proliferation and were associated with increased superoxide and H(2)O(2) accumulation, cSrc activity, H(2)O(2)-dependent tyrosine phosphorylation, and NF-kappaB-dependent transcription. These concentrations were insufficient to activate MAP kinases. However, the MAP kinases, extracellular signal-regulated kinase and p38, were activated in response to levels of arsenite that caused cell death. These data suggest that arsenite-induced oxidant accumulation and subsequent activation of tyrosine phosphorylation represent a MAPK-independent pathway for phenotypic change and proliferation in vascular cells.

Abstract

An elongated endothelial cell phenotype, which demonstrated increased ICAM-1-dependent neutrophil adherence, was induced when these cells were exposed to noncytotoxic concentrations of asbestos (Treadwell et al., Toxicol. Appl. Pharmacol. 139, 62-70, 1996). The present study examined mechanisms underlying this phenotypic change by investigating the effects of asbestos on transcription factor activation and expression of urokinase-type plasminogen activator (uPA) and its receptor uPAR. In situ zymography was used to compare the effects of these fibers on the activity of uPA. Cultures incubated with chrysotile or crocidolite asbestos, but not refractory ceramic fiber 1 (RCF-1), demonstrate localized cleavage of plasminogen, which was inhibited by amiloride. Immunocytochemistry showed that chrysotile-stimulated uPA activity was associated with a time-dependent augmentation of uPAR protein levels. RT-PCR analysis was used to investigate molecular mechanisms for these increases. Chrysotile asbestos, but not RCF-1, increased endothelial cell uPA message, relative to changes in beta-actin mRNA. This response to asbestos was not limited to endothelial cells, since both uPA and uPAR mRNA levels increase in human bronchial epithelial BEAS-2B cells exposed to chrysotile fibers. Finally, both types of asbestos, but not RCF-1, increased nuclear levels of nuclear factor-kappaB (NF-kappa B), a transcription factor common to increased expression of ICAM-1 and uPA. These data demonstrate that asbestos caused fiber-specific activation of endothelial and pulmonary epithelial cells, resulting in phenotypes capable of facilitating tissue remodeling.

Abstract

Background In showing no correlation between adrenal insufficiency risk and dose or duration of steroid treatment, previous reference papers have blurred our comprehension and practice (1). Objectives To determine the extent to which long-term corticosteroid therapy damages the pituitary-adrenal axis in patients treated with prednisone medication for systemic disorders. Methods We retrospectively studied all consecutive patients followed in our department from January 2011 to August 2012 in whom short synacthen test (SST) was assessed when withdrawal of prednisone was planned. Age, sex, disease status, duration and cumulative dose of glucocorticoid therapy were systematically recorded. SST was considered blunted when the 60 minutes plasma cortisol concentration was below 550 nmol per liter. Results Sixty consecutive patients suffering various systemic diseases were studied (Table 1). The mean daily dose of prednisone at study time was 7 (±2) (range: 5-10) mg. Median duration and cumulative dose of prednisone were 3.8 years (range: 0.3-32) and 16.6 grams (1-132), respectively. The SST response was blunted in 29 patients (48.3%) and normal in 31. Blunted responses were associated with higher cumulative prednisone dose (p=0.04) and treatment duration (p=0.04) (Table1 and 2). Basal cortisol concentrations negatively correlated with both duration (r=-0.33, p=0.01) and cumulative dose (r=-0.28, p=0.03) of prednisone. Steroids were stopped in 29/31 (93.5 %) patients showing a normal response to SST. In this case, no patient required hydrocortisone replacement for a mean follow-up of 10 (± 6) months. Conclusions Adrenal insufficiency is frequent in patients treated with long-term glucocorticoid for systemic disorders. 26. The scale-free dynamics of eukaryotic cells.

Abstract

Temporal organization of biological processes requires massively parallel processing on a synchronized time-base. We analyzed time-series data obtained from the bioenergetic oscillatory outputs of Saccharomyces cerevisiae and isolated cardiomyocytes utilizing Relative Dispersional (RDA) and Power Spectral (PSA) analyses. These analyses revealed broad frequency distributions and evidence for long-term memory in the observed dynamics. Moreover RDA and PSA showed that the bioenergetic dynamics in both systems show fractal scaling over at least 3 orders of magnitude, and that this scaling obeys an inverse power law. Therefore we conclude that in S. cerevisiae and cardiomyocytes the dynamics are scale-free in vivo. Applying RDA and PSA to data generated from an in silico model of mitochondrial function indicated that in yeast and cardiomyocytes the underlying mechanisms regulating the scale-free behavior are similar. We validated this finding in vivo using single cells, and attenuating the activity of the mitochondrial inner membrane anion channel with 4-chlorodiazepam to show that the oscillation of NAD(P)H and reactive oxygen species (ROS) can be abated in these two evolutionarily distant species. Taken together these data strongly support our hypothesis that the generation of ROS, coupled to redox cycling, driven by cytoplasmic and mitochondrial processes, are at the core of the observed rhythmicity and scale-free dynamics. We argue that the operation of scale-free bioenergetic dynamics plays a fundamental role to integrate cellular function, while providing a framework for robust, yet flexible, responses to the environment.

Abstract

Publisher's version (útgefin grein)., Heavy alcohol consumption is an established risk factor for hypertension; the mechanism by which alcohol consumption impact blood pressure (BP) regulation remains unknown. We hypothesized that a genome-wide association study accounting for gene-alcohol consumption interaction for BP might identify additional BP loci and contribute to the understanding of alcohol-related BP regulation. We conducted a large two-stage investigation incorporating joint testing of main genetic effects and single nucleotide variant (SNV)-alcohol consumption interactions. In Stage 1, genome-wide discovery meta-analyses in ≈131K individuals across several ancestry groups yielded 3,514 SNVs (245 loci) with suggestive evidence of association (P < 1.0 x 10−5). In Stage 2, these SNVs were tested for independent external replication in ≈440K individuals across multiple ancestries. We identified and replicated (at Bonferroni correction threshold) five novel BP loci (380 SNVs in 21 genes) and 49 previously reported BP loci (2,159 SNVs in 109 genes) in European ancestry, and in multi-ancestry meta-analyses (P < 5.0 x 10−8). For African ancestry samples, we detected 18 potentially novel BP loci (P < 5.0 x 10−8) in Stage 1 that warrant further replication. Additionally, correlated meta-analysis identified eight novel BP loci (11 genes). Several genes in these loci (e.g., PINX1, GATA4, BLK, FTO and GABBR2) have been previously reported to be associated with alcohol consumption.
