Background. Currently, pneumonia ranks 4-5 rate in the world in the structure of death causes after cardiovascular and cancer diseases, cerebrovascular pathology, injuries and poisonings. Objective. The aim of the study was to evaluate the indicators of the oxidative-antioxidant system and their relationship with the strength of the respiratory muscles in adult patients with community-acquired pneumonia. Methods. The study was carried out in the period 2017-2020 on the basis of the therapeutic department of the Municipal non-profit enterprise "City Clinical Multidisciplinary Hospital № 25" of Kharkiv City Council. The study involved 52 adult patients with CAP aged 18 to 80 years. The control group consisted of 20 apparently healthy humans. It was determinated the activity of malondialdehyde, catalase and superoxide dismutase, level of reduced glutathione, glutathione reductase and glutathione peroxidase. The assessment of the RM strength was investigated by recording the maximum static pressures at the level of the mouth and nose with “closed” airways using a MicroRPM apparatus on the 1st and 10th days of illness. Results. Dysfunction of expiratory respiratory muscles prevailed in patients with non-severe CAP, and inspiratory respiratory muscles in patients with severe CAP. Were established significant negative correlations of malondialdehyde with indicators of respiratory muscles strength and positive correlations with glutathione reductase, glutathione peroxidase, catalase, and superoxide dismutase. Conclusions. The presence of relationships between pro- and antioxidant indicators and respiratory muscles strength complements the concept of the body's systemic response on pulmonary inflammation – one of the markers of respiratory muscles dysfunction. Objective We aimed to evaluate apelin‐13 levels, total oxidant/antioxidant status in Alzheimer’s disease (AD) and to investigate the relationship between these parameters. Methods Patients newly diagnosed with AD were enrolled in the study. The control group consisted of age‐ and gender‐matched healthy individuals. Serum levels of apelin‐13, total antioxidant status (TAS), and total oxidant status (TOS) were measured. Oxidative stress index was calculated (TOS/TAS) for each participant. Results We reported that serum apelin‐13 and TAS values were significantly lower in the AD group compared with controls, and they found a fair but insignificant relationship between Apelin‐13 and TAS values. This study investigated the effects of curcumin and capsaicin on testicular and hepatic oxidant–antioxidant status in rats fed a high-fat diet (HFD). Male Sprague–Dawley rats were divided into 5 groups (8 rats per group). The control group was fed a normal control diet (standard laboratory chow), the HFD group was fed HFD (60% of total calories from fat), the HFD+CUR group received HFD supplemented with curcumin (1.5 g curcumin/kg HFD), the HFD+CAP group was given HFD supplemented with capsaicin (0.15 g capsaicin/kg HFD), and the HFD+CUR+CAP group received HFD supplemented with curcumin and capsaicin for 16 weeks. Hepatic and testicular thiobarbituric acid reactive substances (TBARS), reactive oxygen species (ROS), glutathione (GSH) levels, glutathione transferase activity, and Cu-Zn superoxide dismutase, glutathione peroxidase, and catalase protein expression and enzyme activities were measured. Protein expression was determined by Western blotting. GSH levels and antioxidant enzyme activities were measured with colorimetric methods. HFD slightly increased hepatic and testicular oxidative stress parameters. GSH levels did not change between groups. TBARS and ROS levels were significantly reduced in the HFD+CUR+CAP group compared with the HFD group. Liver and testis antioxidant enzyme activities and expression increased significantly with combined capsaicin and curcumin treatment. Curcumin and capsaicin treatment attenuated testicular and hepatic oxidative stress and enhanced the antioxidant defense system. The combination of capsaicin and curcumin with HFD seems to have some remarkable and beneficial effects on testicular oxidative damage in the fatty liver rat model. more...

Oxidative stress is a potential mechanism involved in the pathogenesis of iron deficiency anaemia (IDA). Although a tendency for hypercoagulability has been reported in IDA, its underlying mechanism is yet to be elucidated. This study investigated the probable relationship between oxidative stress and hypercoagulability in children with IDA. This study included 57 children diagnosed with IDA (IDA group) between October 2016 and October 2017 in addition to 48 healthy children (control group). The maximum clot firmness (MCF) index, and clot formation time (CFT) index, which are indicators of hypercoagulability in rotational thromboelastometry assays [intrinsic TEM (INTEM) and extrinsic TEM (EXTEM)] derived from our previous study, were recorded. In elderly, hormones and oxidant-antioxidant interplay are suggested to mediate biochemical balance between adipose tissue to other tissues. Thus the study attempts to explore metabolic traits, plasma resistin, and oxidant-antioxidant markers in metabolic syndrome (MetS) in comparison to non-metabolic syndrome (non-MetS) elderly individuals. A total of 541 healthy elderly Caucasians, with no acute and/or chronic disorders were invited. After taking into account inclusion/exclusion criteria’s the MetS was defined as the presence of three out of five abnormal findings and allowed to divided groups into: non-metabolic syndrome, non-MetS ( n = 25, median age 69.0 years), and newly diagnosed MetS ( n = 29; median age 70.5 years) individuals. Glucose, plasma lipids, resistin (Res), thiobarbituric acid-reacting substances (TBARS), total antioxidant status (TAS), and Cu,Zn-superoxide dismutase (SOD-1) were measured. The MetS had higher resistin than non-MetS ( p 2 = 0.23; p 2 = 0.53; p = 0.0004) moreover in that group TAS correlated negatively with HDL-C (β = −0.71; R = 0.72; R 2 = 0.37; p = 0.01). While age correlated negatively with systolic blood pressure (β = −0.60; R = 0.62; R 2 = 0.14; p = 0.03) independently from other factors in the non-MetS group. Various metabolic factors contribute to maintain serum resistin and oxidant-antioxidant balance in the elderly people in the presence or absence of MetS. Background Stroke is a major cause of mortality and morbidity. Also, free radicals and oxidative stress are deleterious factor in the stroke progression. We aimed to evaluate the association between oxidative stress markers and odds of having risk factor for stroke or developing stroke. Methods The present case control study conducted on 556 participants in Imam-Reza hospital, Tabriz, Iran. Subjects were divided into three group, including individuals with acute ischemic stroke, at risk of stroke, and healthy controls. All enrolled participants except for controls underwent neurological examinations and brain magnetic resonance imaging (MRI). Stroke-related disability and stroke severity were evaluated by modified Rankin Scale (mRS) and National Institutes of Health Stroke Scale (NIHSS), respectively. Serum malondialdehyde (MDA) level and total antioxidant capacity (TAC) were measured within 48 hours of stroke. One-way ANOVA and Chi-square tests for comparing characteristics between groups, multivariable logistic regression for odds of stroke based on MDA and TAC quartiles, and Spearman’s correlation were used. It is unclear how aerobic exercise program effects oxidative stress and antioxidant defense parameters in obese children; therefore, our objective is to examine the effects of aerobic exercise on parameters of oxidative stress and antioxidant in obese boys. Our study included 10 obese boys, aged from 13 to 15, and 10 healthy boys as control group. Before and after exercise, 10 ml blood specimens were taken from the obese boys, who exercised 3 days/week for 12 weeks. 10 ml blood specimens were also taken from the health control group. The oxidative stress and antioxidant defense parameter levels in blood samples were measured for both groups. In our study, it was found that control group and obese children after aerobic exercise had significantly higher superoxide dismutase, catalase, glutathione peroxidase and paraoxonase-1 values than those of obese boys before aerobic exercise. On the other hand, it was found that after aerobic exercise, control group and obese children had significantly lower malondialdehyde, nitric oxide and carbonyl values than those of obese boys before aerobic exercise. Aims Oxidative stress induced by diabetes mellitus (DM) is considered as one of the main causes of infertility in diabetic patients. The aim of the present study was to assess the effect of Tempol – as a synthetic antioxidant- on the testis oxidative stress and sperm parameters in type 2 diabetic (T2D) rats. Main methods Twenty male Wistar rats were divided into 4 groups. Control groups (C) and diabetic groups (D); the control and diabetic groups received Tempol (100 mg/kg) for one month. Sperm parameters and oxidative stress biomarkers were evaluated in testicular tissue. Key findings The results demonstrated that administration of Tempol in diabetic rats improved sperm motility and viability and decreased the count of abnormal sperms. Also Tempol decreased the fasting blood sugar (FBS) and lipid peroxidation (LPO). In addition, Tempol significantly increased total antioxidant capacity (TAC) levels in testis tissue of T2D rats. Histopathological changes were also improved in the diabetic treated group. The purpose of this study was to evaluate the prophylactic effects of quercetin (QUE) against oxidative damages and histological changes induced by 1, 2, 3, 4-tetrachlorodibenzo-p-dioxin (TCDD) in rat heart. For this purpose, 20 adult male Wistar rats were divided into four groups as fellow control group, TCDD group (10µg/kg BW/day by gavage daily for 60 consecutive days), QUE group (30 mg/kg BW/day) and TCDD + QUEgroup. The oxidative biomarkers of heart tissue and the levels of cTnI and lipid profile in serum were measured. Our results showed that the cTnI serum level and the heart lipid peroxidation significantly increased and the heart level of antioxidant profile significantly decreased in the TCDD group compared to control and QUEgroups. While pretreatment with QUEcould significantly improved these factors in serum and heart tissue in animals that consumed TCDD. It can be concluded that QUEat doses of 30 mg/kg/day could alleviate heart oxidative damage and histological changes induced by TCDD. more...

Introduction Rosacea is a chronic, inflammatory dermatosis which develops due to the effect of genetic and environmental factors. Aim To evaluate the oxidative stress in rosacea patients by measuring serum total antioxidant status (TAS), total oxidant status (TOS), oxidative stress index (OSI) and advanced oxidation protein products (AOPP) levels in our study. Material and methods Our study included rosacea patients and healthy volunteers aged between 18 and 65 years. Total antioxidant status, TOS and AOPP levels were measured and OSI was calculated. Results The study included 70 rosacea patients and 30 healthy volunteers as a control group. When TAS, TOS, OSI and AOPP levels were compared between rosacea and control groups, there was no difference for OSI levels; while TAS, TOS and AOPP levels were significantly higher in the rosacea group (p = 0.151, p = 0.013, p = 0.034, p = 0.017, respectively). In the rosacea group, there was no correlation between TAS, TOS, OSI and AOPP levels and disease duration. Besides there was no difference between family history, rosacea type, symptom frequency and ocular involvement and TAS, TOS, OSI and AOPP levels in the rosacea group. Conclusions We observed that serum TAS, TOS and AOPP levels were significantly higher in rosacea patients, but there was no significant difference among the disease activity parameters. Graphical abstract, Background Early detection of oxidant-antioxidant levels and special care in severe patients are important in combating the COVID-19 epidemic. However, this process is costly and time consuming. Therefore, there is a need for faster, reliable and economical methods. Methods In this study, antioxidant/oxidant levels of patients were estimated by Expert-models using biomarkers, which are effective in the diagnosis/prognosis of COVID-19 disease. For this purpose, Expert-models were trained and created between the white-blood-cell-count (WBC), lymphocyte-count (LYM), C-reactive-protein (CRP), D-dimer, ferritin values of 35 patients with COVID-19 and antioxidant/oxidant parameter values of the same patients. Error criteria and R2 ratio were taken into account for the performance of the models. The validity of the all models was checked by the Box-Jenkis-method. Results Antioxidant/Oxidant levels were estimated with 95% confidence-coefficient using the values of WBC, LYM, CRP, D-dimer, ferritin of different 500 patients diagnosed with COVID-19 with the trained models. The error rate of all models was low and the coefficients of determination were sufficient. In the first data set, there was no significant difference between measured antioxidant/oxidant levels and predicted antioxidant/oxidant levels. This result showed that the models are accurate and reliable. In determining antioxidant/oxidant levels, LYM and ferritin biomarkers had the most effect on models, while WBC and CRP biomarkers had the least effect. The antioxidant/oxidant parameter estimated with the highest accuracy was Native-Thiol divided by Total-Thiol. Conclusions The results showed that the antioxidant/oxidant levels of infected patients can be estimated accurately and reliably with LYM, ferritin, D-dimer, WBC, CRP biomarkers in the COVID-19 outbreak. more...

Conventional phototherapy converts unconjugated bilirubin to its oxidation products and consequently causes oxidative stress with lipid peroxidation products. New devices that deliver intensive phototherapy are efficacious in treating severe hyperbilirubinemia and minimizing the need for exchange transfusions. However, the oxidative stress status when using these devices has not been explored. Therefore, we aimed to study the impact of using intensive phototherapy on the oxidant-antioxidant status in severely jaundiced neonates.This prospective case-control study included term newborns admitted with severe hyperbilirubinemia managed with intensive phototherapy. Baseline oxidant-antioxidant concentrations were compared to healthy controls and re-measured after 8 h of intensive phototherapy exposure.The study included 40 cases with severe jaundice and 40 non-jaundiced apparently normal controls. Total serum bilirubin at enrollment was 23.4 ± 4.2 mg/dl that significantly decreased after 8 h of therapy to 15.4 ± 3.4 mg/dl (p 0.001). The decline of total serum bilirubin was 1 mg/dl/h. Bilirubin: albumin ratio decreased from 3.45 ± 0.28 to 2.7 ± 0.21 (p 0.001). Total antioxidant capacity (TAC), superoxide dismutase (SOD), malondialdehyde (MDA), and total oxidative stress (TOS) concentrations were lower in cases (p 0.001, p 0.001, p = 0.049, and p 0.001 respectively) compared to controls. Following 8 h of intensive phototherapy, further decline of TAC (p = 0.016) with increased concentrations of TOS (p = 0.005) were noted. SOD and MDA did not change.Although efficacious, intensive phototherapy was associated with increased oxidative stress. In the last two or three decades, physical activity (PA) has gained increasing recognition as being essential for maintaining good health and improving quality of life for all ages. To evaluate the nutritional status, oxidant/antioxidant, and inflammatory markers, in sports adolescents, a cross sectional study has done in west Algeria. A total of 110 athletes adolescents from special sport classes, aged 11-17 years, were compared to a non athletes group (n=60). Blood pressure, anthropometric and serum parameters were measured. Daily energy intake (DEI) was estimated using 24 hours recall, followed by a 3-days record. Energy expenditure (EE) was evaluated by the International Physical Activity Questionnaire-short form (IPAQ-S). Oxidant/antioxidant and inflammatory markers were determined. BACKGROUND Oxidative stress parameters such as total oxidant status (TOS), total antioxidant status (TAS), and oxidative stress index (OSI) have been studied in breast, thyroid, and simple esophageal cancers (EC). We evaluated these parameters in patients with EC and analyzed their correlations with treatment outcomes. MATERIAL AND METHODS Serum TOS, TAS, and OSI in 92 patients with EC at different clinical stages and in 64 healthy people (controls) were measured. RESULTS Serum TOS, TAS, and OSI were significantly different between patients with EC and healthy controls (all p0.001); however, there were no significant differences across different clinical stages (all p0.05). These factors are not correlated with smoking or drinking history (all p0.05). Patients with EC with higher TOS and OSI and lower TAS had better responses to chemotherapy and/or radiotherapy, but there was no significant correlation with different responses (all p0.05). In a receiver operating characteristic curve analysis comparing patients with EC with healthy controls, the Youden indices were 0.391, 0.886, and 1, respectively. CONCLUSIONS Serum TOS, TAS, and OSI were significantly different between patients with EC and healthy controls. In patients with EC, these factors were not correlated with smoking or drinking history or with clinical stage. Patients with EC with higher TOS and OSI and lower TAS had a trend towards better outcomes but it did not reach significance. Bulut O, Duruyen S. Impacts of phototherapy on DNA damage and total oxidant/antioxidant status in jaundiced newborns. Turk J Pediatr 2019; 61: 697-703. Jaundice is common in newborn babies, and pathologic hyperbilirubinemia is frequently treated with phototherapy. Although it is considered to be safe, the side effects of phototherapy in newborns are still a matter of debate. In this study, the impacts of two types of phototherapy, conventional fluorescence and intensive light-emitting diodes (LED), on DNA damage and total oxidant/ antioxidant status in jaundiced newborns were assessed. The study group included 40 newborns (gestation age ≥ 37 weeks) on days 2-8 after birth. Newborns were divided into two groups on the basis of need for phototherapy: 20 newborns were exposed to conventional phototherapy (Group I); and 20 infants were exposed to intensive phototherapy (Group II). Blood samples were taken from all infants at admission and after phototherapy to determine plasma 8-hydroxy-2-deoxy-guanosine (8-OH-dG; a marker of DNA damage), total oxidant status (TOS), and total antioxidant capacity (TAC). The oxidative stress index (OSI) was also calculated. Demographic information was recorded, and DNA damage, TOS, and TAC were compared. There were no differences in demographic information between the two groups. There were no significant differences in DNA damage, TOS, TAC, and OSI between Groups I and II before phototherapy (p > 0.05) and no significant differences in DNA damage, TOS, TAC, and OSI between the two groups after phototherapy (p > 0.05). However, the TAC decreased significantly in both groups after phototherapy (p < 0.01). These findings suggest that conventional and intensive phototherapies do not affect DNA damage and oxidative stress, supporting the safety of its use as the preferred treatment for jaundiced newborns. more...

Aim: This study has been conducted for the purpose of determining serum hepcidin, total antioxidant status (TAS), total oxidant status (TOS), and Fe levels in calves with suspected neonatal septicemia before and after treatment and the clinical significance of hepcidin in calves with suspected neonatal septicemia. Materials and Methods: The study material consisted of 15 calves of different ages and sexes brought to the Training, Research and Application Center at the Kafkas University Faculty of Veterinary Medicine with suspected neonatal septicemia. 8.5 mL of blood was drawn from the jugular vein of each animal into coagulant tubes before and after treatment for one-off biochemical analyses and centrifuged. Background/aim: Oxidative stress plays an important role in the pathogenesis of chronic obstructive pulmonary disease (COPD). Smoking is the leading source of oxidants in lungs. However, it is currently unknown why some individuals are more resistant to the detrimental effects of smoking and do not develop COPD. The aim in this study is to measure and compare the oxidant/antioxidant balance between in non-COPD individuals who smoke and COPD patients who smoke. Materials and methods: Included in the study were 137 patients with COPD and 102 healthy individuals. Participants were divided into groups as COPD patients (former and current smokers), non-COPD individuals who smoke and non-smokers healthy persons. In the following stage, the total antioxidant status (TAS), total oxidant status (TOS) and oxidative stress index (OSI) levels were measured in serum for all participants. Results: In the current-smoker COPD group, the level of oxidant status were significantly higher than the former-smoker COPD group (p < 0.001). Similarly, oxidant levels were significantly high in current-smoker healthy group than never smoker healthy group. According to these results TOS was associated with especially smoking status rather than COPD. Antioxidant status were similar between former-smoker COPD group and current-smoker COPD group. The antioxidant levels were found significantly low in current-smoker COPD patients, compared to the current-smoker non-COPD individuals (p = 0.007). Nevertheless, no significant difference was found in OSI levels between two groups. Briefly, high TOS and OSI values were correlated with only smoking, independently from COPD. Conclusion: It was concluded that there are complex pathogenetic mechanisms, including genetic and individual variations other than oxidant/antioxidant balance, involved in the development of smoking-related COPD. The AlterG antigravity treadmill allows running with a considerable weight reduction. Physical exercise practiced on this treadmill is an innovative method supporting the treatment of injuries in sports and rehabilitation of patients. The aim of the study was to investigate the effect of a 30 min run on the AlterG treadmill with 80% body weight reduction comparing the effect to the similar effort on the classic treadmill on the redox equilibrium and the activity of selected lysosomal enzymes and a serine protease inhibitor in the blood of amateur minor female volleyball players. Venous blood samples were taken before the exercise and 30 minutes and 24 hours after its completion. The obtained results were analysed using Tukey’s test and Pearson’s linear correlations were calculated. 24 h after the running test on classic treadmill, the erythrocytic superoxide dismutase activity was higher than before and 30 min after it, as well as compared to the run on AlterG treadmill (p<0.001). The erythrocytic-conjugated diene concentration 24 h after the exercise on the classic treadmill was meaningly higher compared to that after the exercise on the AlterG treadmill (p<0.001). The cathepsin D activity was significantly lower after the exercise in AlterG conditions compared to the baseline value and that measured after the exercise on classic treadmill (p<0.001). It seems that the exercise on the AlterG treadmill keeps the oxidant–antioxidant equilibrium and stabilizes lysosomal membranes in young, physically active women in contrast to the exercise on the classic treadmill. Our knowledge about a link between buprenorphine and hepatotoxicity is controversial. This study evaluated the effects of buprenorphine on the liver of young, adult, and aged rats. For this reason, young, adult, and aged rats received intraperitoneally 0.25, 0.5, and 1 mg/kg buprenorphine for 30 days. The present results revealed that the normal aging was associated with a significant decrease in the activities of antioxidant enzymes, and an increase in the liver lipid peroxidation, serum aspartate aminotransferase (AST), alanine aminotransferase (ALT), and lactate dehydrogenase (LDH) activities in the aged rats. This study also demonstrated that buprenorphine led to a significant increase in the serum activities of ALT, AST, and LDH as well as liver lipid peroxidation content with a decrease in the antioxidant enzymes in the liver of buprenorphine-treated aged rat versus the aged matched control animals. To analyze oxidative stress parameters in the aqueous humor with retinal vein occlusion.A total of 56 patients who underwent cataract surgery were enrolled in this study. The eyes with retinal vein occlusion were included in the study group. Aqueous humor was sampled from all patients and analyzed by an automated measurement method. Total oxidant status (TOS), total antioxidant status (TAS), and oxidative stres index (OSI) were calculated.OSI was significantly higher in the study group (p = 0.04). TAS was significantly lower (p = 0.01) and OSI was significantly higher (p = 0.01) in cases with both central retinal vein occlusion and macular edema. Previous laser photocoalugation to the retina and the duration of disease had no significant effect on the levels of oxidative stress parameters.OSI was found higher in patients with retinal vein occlusion. To establish the total oxidant status (TOS), total antioxidant capacity (TAC) and oxidative stress index (OSI) of the aqueous humour in patients presenting exfoliation syndrome (EXS) without glaucoma. The TOS, TAC and OSI of the aqueous humour of patients with EXS (group 1, n = 17) and patients without EXS (group 2, n = 25) who underwent cataract surgery were evaluated. Samples were measured spectrophotometrically using a colourimetric method. The Mann–Whitney U, independent-samples t tests, Pearson correlation and analysis of covariance tests were used in the statistical analyses. The mean TOS in group 1 and 2 patients was 57.6 ± 32.4 and 30.4 ± 22.6 mmol/L, respectively, which is a statistically significant difference (p = 0.001). The mean TAC level in group 1 and 2 patients was 2.3 ± 0.7 and 2.5 ± 0.7 mmol/L, respectively, and although TAC was decreased in group 1 relative to group 2, the difference was not statistically significant (p = 0.55). The mean OSI in group 1 and 2 patients was 27.4 ± 17.1 and 12.5 ± 8.3 mmol/L, respectively, with the mean OSI level statistically higher in group 1 (p = 0.03). Our findings provide evidence that the aqueous humour of EXS patients is characterised by increased oxidative stress and a disturbed oxidant/antioxidant balance. The increased oxidative stress and decreased levels of antioxidants in ocular fluids of EXS patients may play a significant role in the pathogenesis and complications of EXS. more...

Aim The aim of this study was to investigate the oxidant-antioxidant status in babies born to pre-eclamptic mothers (BBPM). Material and Methods The paraoxonase (PON)-1, total antioxidant status (TAS), and total oxidant status (TOS) levels were measured in the cord blood and venous blood (7th day) of BBPM (n = 31) and babies born to normotensive mothers (n = 25). Results The PON-1 and TOS levels in the cord blood and venous blood on the 7th day were not significantly different between the two groups; however, the cord blood TAS levels were higher in BBPM (P = 0.001), and the TAS levels in the venous blood were higher in the control group (P = 0.021). Furthermore, the cord blood PON-1 levels of babies born to severely pre-eclamptic mothers (n = 18) were higher than those of babies born to moderately pre-eclamptic mothers (n = 13) (P = 0.042). There were no differences in the cord blood TAS and TOS levels and venous blood PON-1, TAS, and TOS levels between babies born to severely and moderately pre-eclamptic mothers. Conclusion The increased TAS levels found in the cord blood of BBPM indicate that the fetus is protected against oxidative damage caused by increased oxidative stress in the mother. A high-fat diet (HFD) causes deficits in learning and memory by increasing oxidative stress. Antioxidants are known to improve learning and memory. Since Hypericum scabrum (H. scabrum) extract is rich in antioxidants, the aim of this study was to investigate the effects of the administration of H. scabrum extract on passive avoidance learning (PAL), novel object recognition (NOR), and locomotor activity in male rats on a HFD. Fifty-four male Wistar rats (weighing 220 ± 10 g) were divided into the following six groups: (1) Control (standard diet), (2) Ext100 (standard diet supplemented with 100 mg/kg extract once/day), (3) Ext300 (standard diet supplemented with 300 mg/kg extract once/day), (4) HFD (high-fat diet), (5) HFD + Ext100, and (6) HFD + Ext300. Rats in these groups were maintained on their respective diets for 3 months. In the PAL test, the step-through latencies in the retention test (STLr) were significantly higher in the HFD + extract group than in the HFD group. The time spent in the dark compartment (TDC) was significantly lesser and the time spent in exploring the novel object was significantly greater in the HFD + extract group than in the HFD group. In the HFD-fed rats, the activity of catalase had significantly decreased, and level of malondialdehyde had significantly increased; H. scabrum extract administration significantly reversed these changes. In conclusion, these results suggested that the administration of H. scabrum extract and its strong antioxidant properties enhanced learning and memory and reversed the memory impairment induced by chronic HFD consumption. more...

Aim: to evaluate the effect of levothyroxine replacement therapy on oxidative stress in patients with combined course of arterial hypertension (AH), type 2 diabetes mellitus (DM2T), and subclinical hypothyroidism (SHT). Object and methods. The study included 40 patients (9 males and 31 females) aged 44 to 75 years (mean age 59.3 ± 3.5 years) with stage II hypertension in combination with DM2T and SHT. The presence of SHT that developed as a result of autoimmune thyroiditis (AIT) and TSH levels in the range 6.0-10.0 µMU/ml in screening was an obigatory criterion. The control group consisted of 30 gender and age-representative patients who did not have cardiovascular disease and endocrinopathies. Thyroid metabolism, glutathione peroxidase (GPO) activity, levels of sulfhydryl groups (SH-groups), malonic dialdehyde (MDA) and 8-hydroxy-2-deoxyguanosine (8-OH-dG) in serum were determined. Ultrasound examination of the thyroid gland was performed on the “LOGIQ 5” according to the standard method. After determining the TSH level, patients were prescribed levothyroxine at doses of 12.5 to 50 μg/day with gradual dose titration (21 day titration step) until euthyroidism was achieved. Observation period was1 year. Statistical processing of data was performed using the computer program SPSS 21.0. Results. The presence of significantly higher levels of MDA and significantly elevated (almost 2.3-fold) levels of 8-OHdG in the patients of the study group indicates a pronounced stimulation of oxidative processes in such a polymorbid clinical situation. Method of study We studied 40 early and late onset pre-eclamptic patients and 40 healthy pregnant control and 39 healthy non-pregnant control subjects. Serum CA II antibodies, TAC and TOC, and MDA parameters were studied by ELISA. Results The mean values for TAC, TOC, OSI, MDA, and anti-CA II were significantly increased in patients with pre-eclampsia compared to the other groups. The anti-CA II antibody levels for the pregnant control subjects were 0.129 ± 0.04 and that for the pre-eclamptic patients were 0.282 ± 0.18. In this study, any absorbance value higher than 0.136, the mean absorbance + 2 S.D. of pregnant control subjects, was defined as positive. Positive results were obtained in 29 of 40 pre-eclamptic patients (72.5%). There were significant positive correlations between serum anti-CA II antibodies and TOC, MDA levels, and OSI levels. We investigated serum total oxidative and anti-oxidative status in manic patients. Group1 was formed as ECT+antipsychotic, group2 was antipsychotic and healthy volunteers as group3. The anti-oxidative status was significantly lower in group1 than group3. No significant change was found between pre and post-treatment oxidative and anti-oxidative status, whereas significantly increased oxidative stress index has been found in group2. The aim of the present study is to evaluate the relationship of total antioxidant capacity, total oxidant status, oxidative stres and total free sulfydryl groups in patients with diabetic and nondiabetic senil cataract. The study was carried out on venous blood samples from 70 subjects admitted to the ophtalmology clinic of a research hospital. Of these, 25 were senile Type 2 diabetic patients with cataract who were using oral hypoglycemic agents or insulin. Fourty-five patients with nondiabetic senile cataract comprised the control group. Total oxidant status (TOS), total antioxidant capacity (TAC) and total free -SH levels were measured in sera of patients with cataract. Serum TAS and TOS levels were determined using a novel automated measurement method. Oxidative status was determined using TAC and TOS measurement and with the calculation of oxidative stres index. TOS and OSI levels were significantly higher in diabetic cataract than senile cataract (p=0.008, p=0.013), respectively. TAC and total free -SH levels were similiar in the two groups (p = 0.17, p=0.54). Free sulfydryl groups levels are consistent with TAC. The results of present study suggest that TOS and intensity of the oxidative stress in diabetic patients are higher, compared to other age-related cataract patients without diabetes. Since oxidative stress is important in development of diabetic complications, increased TOS and oxidative stress might have a role in the pathophysiology of diabetic cataract. more...

In this study was investigated effects of borax (BX) on inflammation markers, haematolojical parameters and total oxidant (TOS)-antioxidant status (TAS) in rats applied 3-methylcholanthrene (3-MC). In this research a total of 24 Wistar Albino rats were used. They were divided into 4 groups each containing 6 rats. 1st group was separated as a control group. 3-MC was applied twice a week first 2 weeks 25 mg/kg dose to the 2nd group with i.p. way. BX was given to 3rd group 300mg/L/day dose with drinking water during 150 days. 3-MC was applied twice a week first 2 weeks 25 mg/kg dose with i.p. way and BX were given with drinking water during 150 days to 4th group. At the end of the study blood analysis, tumor necrosis factor alpha (TNF-alpha) and interleukin 1 beta (IL-1 beta) levels in 3-MC group; TOS and oxidative stress index (OSI), platelet (PLT) levels in 3-MC and 3-MC+BX groups showed significantly increases when compared to other groups. Possible effects of a high-cholesterol diet on the oxidant/antioxidant status in various tissues from rats and the effects of the combination of vitamins C and E on these parameters and on hypercholesterolemia were investigated. Twenty-four Sprague-Dawley type rats were randomly divided into three groups. The control group (group 1) was fed on a rat pellet diet. The study groups (groups 2 and 3) were fed on a high-cholesterol diet for 3 months. Rats in group 3 were supplemented with vitamins C (100 mg/kg intraperitoneally ) and E (60 mg/kg intramuscularly ) three times a week throughout the study. At the end of the study period, all animals were sacrificed and their heart, kidney, liver and testis surgically removed to measure oxidant and antioxidant parameters. Blood samples were also obtained from the animals. We observed that serum total cholesterol levels significantly increased in the study groups after the study period. The increase in the blood cholesterol level in group 3 was, however, significantly lower than that in group 2. As to the oxidant–antioxidant parameters, there were different results among the tissues. In conclusion, our results suggest that high serum cholesterol level does not lead to oxidation reactions in the early phase of hypercholesterolemia, and antioxidant vitamins supplementation increases antioxidant potential values of the tissues and decreases the blood total cholesterol level in the cholesterol-supplemented animals. The aim of this study was to evaluate the oxidant-antioxidant balance in patients with abdominal aortic aneurysms (AAA). Forty-two consecutive patients with AAA and 46 control subjects were included. Total oxidant status (TOS) and total antioxidant status (TAS) levels were measured and the oxidative stress index (OSI) value determined. Serum TOS and OSI values in patients with AAA were higher than those in the controls (p2O2 equivalent/L for TOS was associated with 86% sensitivity and 83% specificity and a cut-off value of 1.77 for OSI was associated with 86% sensitivity and 81% specificity for predicting AAA. We aimed to assess the plasma oxidant and antioxidant levels in systemic sclerosis (SS) patients with interstitial lung involvement. Twenty-seven female SS patients and 17 healthy female volunteers were included in the study. Plasma levels of oxidants and antioxidants levels were studied of two groups. The median oxidant and antioxidant levels in study and control groups were, MDA 5.2 +/- 0.4/3.7 +/- 0.5 nmol/ml (P0.001), NO 45.4 +/- 3.7/34.2 +/- 2.9 nmol/l (P0.001), SOD 25.6 +/- 2.3/24.6 +/- 2.0 U/ml (P0.05), catalase 99.9 +/- 9.9/140.0 +/- 10.0 U/ml (P0.001), vitamin E 20.5 +/- 1.3/22.6 +/- 2.0%mg (P0.001), vitamin C 70.6 +/- 8.7/83.5 +/- 7.3 mg/dl (P0.001), respectively. There was also no correlation between plasma levels of oxidants-antioxidants levels and disease duration, duration of pulmonary signs, pulmonary function test values, HRCT scores in SS patients (P0.05). The association between oxidative stress and cardiovascular diseases is a widely accepted fact today. Generally, men have a higher risk of cardiovascular incidents and mortality from acute myocardial infarction and strokes. We have examined sport-associated circannual rhythms of oxidant and antioxidant processes by measuring plasma LPO, erythrocyte SOD, CAT, Gpx activity and plasma hormonal status in both sedentary and long-term trained men and women. We have shown seasonal variations in both oxidant and antioxidant status in all examined groups. The largest difference was observed in the oxidant status between sedentary men and women during autumn and winter, which is considered a period of high coronary risk for men. Sport decreased LPO in trained men in autumn, while the same effect in trained women was shifted towards summer. These data state that regular, long-term physical exercise training induces adaptive responses that confer protection against oxidative stress, as well as the beneficial effect of exercise with regard to season, particularly in men during a period of high coronary risk (autumn and winter, respectively) and in women during summer. more...

This study investigated changing levels of serum oxidant/antioxidant with chemotherapy and their relation to treatment in 34 Hodgkin's lymphoma patients. The patient population consisted of 19 males and 15 females. Mean age was 30.41 ± 12.08 years. All patients received the adriamycin, bleomycin, vincristine and dexamethasone (ABVD) treatment protocol. Blood samples were taken before treatment, and on days 1 and 7 during treatment for measurement of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), catalase (CAT), malondialdehyde (MDA), nitric oxide (NO) and enzyme activities. After ABVD treatment, mean free radical levels were increased and antioxidant levels were significantly decreased in the serum. Erythrocyte antioxidant glutathione and malondialdehyde levels in erythrocytes and plasma glutathione S-transferase levels were estimated in patients with colorectal cancer and compared to controls. Further, the patients underwent four weeks of radiotherapy with adjuvant chemotherapy. The same parameters were estimated after four weeks of radiotherapy and compared with pretreatment levels. It was observed that there was a decrease in erythrocyte glutathione and malondialdehyde levels in patients with colorectal cancer compared to controls, but not in case of GST. However, after chemoradiotherapy, there were no statistically significant differences in all the parameters studied. more...
