Introduction: The assessment of body composition in female athletes of different sports is important for health monitoring. Objective: To compare body composition in university athletes of different team sports (indoor soccer, flag football and volleyball). Methods: A cross-sectional study carried out with 45 female athletes, aged 18 to 35 years (22.8 ± 3.55). The dependent variables were body fat mass (FM) and fat-free mass (FFM) measured by air displacement plethysmography. Bone mineral content, adjusted for height (BMC/height) and bone mineral density (BMD), were measured by dual energy X-ray absorptiometry. The independent variable was sport [flag football (n = 12); indoor soccer (n = 20); volleyball (n = 13)] and the covariates were age (complete years), training volume (minutes per week) and length of time playing the sport (complete years). Analysis of covariance was used. Results: Adjusting the model for covariates, volleyball athletes (19.27 kg ± 2.20) presented higher FM values compared to the flag football (16.00 kg ± 1.70) and indoor soccer players (12.20 kg ± 1.30). There was no significant difference in FFM, BMC/height and total BMD between sports, even after adjusting for covariates. Conclusion: Volleyball athletes presented higher FM compared to flag football and indoor soccer athletes. There were no differences in FFM, BMC/height and BMD among the players of the different team sports. This study can help coaches and other sports professionals to prevent injuries to athletes in sports such as higher FM (volleyball), or to prevent diseases such as menstrual irregularities, which are common in athletes who may have low levels of body fat (indoor soccer players), this being one of the risk factors for the female athlete triad (eating disorders, menstrual irregularities and low BMD). No hubo diferencia significativa en la MM, CMO/estatura y DMO total entre los deportes, incluso después del ajuste para las covariables. Conclusiones: Las atletas de vóleibol presentaron mayor MG en comparación con los atletas de flag football y fútbol de salón. No hubo diferencias en MM, CMO/estatura y DMO entre las jugadoras de diferentes modalidades colectivas. Conclusões: As atletas de voleibol apresentaram maior MG em comparação com as atletas de flag football e futebol de salão. Não houve diferenças em MM, CMO/estatura e DMO entre os jogadoras de diferentes modalidades coletivas. Este estudo pode ajudar treinadores e outros profissionais do esporte a prevenir lesões em atletas com maior MG (voleibol) ou prevenir doenças como irregularidades menstruais, comuns em atletas com baixos níveis de gordura corporal (futebol de salão), que é um dos fatores de risco para a síndrome da tríade da atleta feminina (distúrbios alimentares, irregularidades menstruais e baixa DMO). Resumen: Los inhibidores del cotransportador de sodio y glucosa tipo 2 (i-SGLT2) ejercen su efecto hipoglucemiante a través de la inhibición de la reabsorción de glucosa en el riñón. Sin embargo, los efectos cardiovasculares, renales y en falla cardiaca parecen ocurrir de manera independiente al efecto hipoglucemiante de estos fármacos. Los principales mecanismos de acción que explican sus beneficios cardiovasculares son el efecto hipotensor y la disminución en las presiones de llenado, efectos directos en el metabolismo de la célula miocárdica, reducción de la microalbuminuria, efectos metabólicos no glicémicos y disminución del tejido adiposo, aumento del hematocrito y uricosuria. En este artículo se describe cada uno de estos mecanismos novedosos. Abstract: Type 2 Sodium-Glucose Cotransporter (SGLT2) Inhibitors exercise their glucose-lowering effect through the inhibition of glucose reabsorption in the kidney. However, the cardiovascular and renal effects, as well as in those of cardiac failure, seem to occur independently from the glucose–lowering effects of these drugs. The principal mechanisms of action that explain their cardiovascular benefits are the hypotensive effect and the decrease in fill pressures, as well as direct effects on the metabolism of the myocardial cell. There is also a reduction in the urine albumin, as well as non-glycaemic effects, and a decrease in adipose tissue, with an increase in haematocrit and urine uric acid. In healthy individuals, white adipose tissue (WAT) has a relevant homeostatic role in glucose metabolism, energy storage, and endocrine signaling. Mast cells contribute to these functions promoting WAT angiogenesis and adipogenesis. In patients with T2D, inflammation dramatically impacts WAT functioning, which results in the recruitment of several leukocytes, including monocytes, that enhance this inflammation. Accordingly, the macrophages population rises as the WAT inflammation increases during the T2D status worsening. Since mast cell progenitors cannot arrive at WAT, the amount of WAT mast cells depends on how the new microenvironment affects progenitor and differentiated mast cells. Here, we employed a flow cytometry-based approach to analyze the number of mast cells from omental white adipose tissue (o-WAT) and subcutaneous white adipose tissue (s-WAT) in a cohort of 100 patients with obesity. Additionally, we measured the number of mast cell progenitors in a subcohort of 15 patients. The cohort was divided in three groups: non-T2D, pre-T2D, and T2D. Among them, metabolic alterations have been described. Adipose tissue is responsible for the secretion of several factors involved in controlling body weight and energy expenditure, the regulation of insulin sensitivity, and the development of inflammation, among others. Moreover, the liver regulates glucose homeostasis and lipid metabolism, playing an essential role in developing insulin resistance. In this work, we analyzed if prenatal stress leads to alterations in metabolism and the relationship between these alterations and gene expression in the adipose tissue and the liver. Prenatal stress-exposed animals developed disturbances in the glucose and insulin response curve, showing in both tests higher glycemia than the control group. However, they did not exhibit increased body weight. At the same time, in the adipose tissue, we observed an increase in mRNA expression of Leptin and Resistin and a decrease in Adiponectin. In the liver, we observed a lower mRNA expression of several genes involved in glucose metabolism and fatty acid oxidation, such as Sirt1, Pgc1α, Pparα, among others. In both tissues, we observed a lower expression of inflammatory genes. These results suggest that prenatal stress exposure produces insulin resistance at both physiological and molecular levels without pro-inflammatory signaling or obesity.

Just over a decade has passed since metabolically active brown adipose tissue (BAT) was unequivocally identified in healthy adult humans. Since then, researchers have provided evidence of expression of the thermogenic molecule uncoupling protein 1 (UCP1) in human BAT, as well as its energy dissipating capacity. Additionally, clinical cross-sectional analysis suggested that a decline in BAT activity with aging, as judged from [18F]FDG-PET/CT, coincides with the development of obesity and insulin resistance. These major observations provided a rationale to investigate BAT as a potential target for preventing obesity. Indeed, several studies have demonstrated that cold exposure and adrenomimetic agents strongly activate BAT thermogenesis, and thus energy expenditure in humans. However, the extent to which stimulating BAT thermogenesis decreases adiposity in humans remains unclear. Moreover, therapeutic strategies aimed at general stimulation of the sympathetic nervous system (SNS) and adrenergic receptors involved may not be applicable for obese and diabetic patients, because of potential negative side-effects on cardiovascular function. While implementation in humans remains challenging, recent studies in mice have yielded insights in molecular mechanisms underlying thermogenic regulation of brown and brown-like (brite/beige) adipocytes, highlighting their ability to reduce insulin resistance and atherosclerosis (1). These include identification of UCP1-independent thermogenesis as well as peripheral organ-derived endocrine factors and metabolites capable of triggering adipocyte thermogenesis. Activating these non-canonical mechanisms through genetic manipulation or pharmacological agents suppresses obesity and its associated complications in mice. The major challenge remains translating preclinical scientific advances to humans. Obesity is characterized by an excessive body fat percentage (BF%). Animal and cell studies have shown benefits of vitamin A (VA) on BF% and lipid metabolism, but it is still controversial in humans. Furthermore, although some genetic variants may explain heterogeneity in VA plasma levels, their role in VA metabolic response is still scarcely characterized. This study was designed as a combination of an observational study involving 158 male subjects followed by a study with a well-balanced genotype&ndash, phenotype protocol, including in the design an ex vivo intervention study performed on isolated peripheral blood mononuclear cells (PBMCs) of the 41 former males. This is a strategy to accurately identify the delivery of Precision Nutrition recommendations to targeted subjects. The study assesses the influence of rs5888 (SCARB1), rs659366 (UCP2), and rs1800629 (UCP1) variants on higher BF% associated with suboptimal VA consumption and underlines the cellular mechanisms involved by analyzing basal and retinoic acid (RA) response on PBMC gene expression. Data show that male carriers with the major allele combinations and following suboptimal-VA diet show higher BF% (adjusted ANOVA test p-value = 0.006). Genotype&ndash, BF% interaction is observed on oxidative/inflammatory gene expression and also influences lipid related gene expression in response to RA. Data indicate that under suboptimal consumption of VA, carriers of VA responsive variants and with high-BF% show a gene expression profile consistent with an impaired basal metabolic state. The results show the relevance of consuming VA within the required amounts, its impact on metabolism and energy balance, and consequently, on men&rsquo, s adiposity with a clear influence of genetic variants SCARB1, UCP2 and UCP1.

Low-grade chronic inflammation is associated with many chronic diseases and pathological conditions. The aim of the present study was to determine the effect of regular caffeine intake, physical activity levels, and sedentary behavior on the inflammatory status in healthy participants. In total, 112 men and 132 women aged 18 to 55 years and belonging to the staff and student population of the University of the Balearic Islands volunteered to participate in this descriptive cross-sectional study. Plasma concentrations of pro-inflammatory and anti-inflammatory markers were measured. Weight, height, and body composition (bioelectrical impedance) were determined. Caffeine intake, physical activity levels and sitting time, and diet quality were determined using questionnaires. Statistical regression analysis showed that caffeine intake was a negative predictor of C-reactive protein (CRP) (p = 0.001). Body fat percentage was positively associated with CRP (p &lt, 0.001) and inversely associated with adiponectin (p = 0.032) and interleukin (IL)-10 levels (p = 0.001). Visceral fat was the main predictor for IL-6 (p &lt, 0.001) and tumor necrosis factor (TNF)-&alpha, (p &lt, 0.001). Sitting time was found to be the main, inverse, predictor for IL-10 (p &lt, 0.001), and a positive predictor for TNF-&alpha, 0.001). In conclusion, regular caffeine consumption induced very limited anti-inflammatory effects. Sedentary behavior and body fat accumulation induced significant pro-inflammatory effects.

We performed a meta-analysis of the effects of exercise on epicardial adipose tissue (EAT). A systematic search was conducted in PubMed and Scopus (since inception to 1 February 2020) of randomized controlled trials assessing the effects of exercise interventions alone (with no concomitant weight loss intervention) on EAT. The standardized mean difference (Hedges' g) and 95% confidence interval between interventions were computed using a random effects model. Ten studies (including 521 participants who had, on average, overweight/obesity) met all inclusion criteria. Interventions were supervised and lasted 2 to 16 weeks (≥3 sessions·per week). Exercise significantly reduced EAT (g = 0.82 [0.57-1.07]) irrespective of the duration of the intervention or the EAT imaging assessment method. Exercise benefits were separately confirmed for endurance (six studies, n = 287; g = 0.83 [0.52-1.15]) but not for resistance exercise training (due to insufficient data for quantitative synthesis). It was not possible to compare the effect of high-intensity interval training (HIIT) versus moderate-intensity continuous training (two studies, one reporting higher benefits with HIIT and the other no differences). Physical exercise interventions-particularly endurance training, with further evidence needed for other exercise modalities-appear as an effective strategy for reducing EAT in individuals with overweight/obesity, which supports their implementation for cardiovascular risk reduction. [Background and Aims]: The synthetic atypical cannabinoid Abn-CBD, a cannabidiol (CBD) derivative, has been recently shown to modulate the immune system in different organs, but its impact in obesity-related meta-inflammation remains unstudied. We investigated the effects of Abn-CBD on metabolic and inflammatory parameters utilizing a diet-induced obese (DIO) mouse model of prediabetes and non-alcoholic fatty liver disease (NAFLD)., [Materials and Methods]: Ten-week-old C57Bl/6J mice were fed a high-fat diet for 15 weeks, following a 2-week treatment of daily intraperitoneal injections with Abn-CBD or vehicle. At week 15 mice were obese, prediabetic and developed NAFLD. Body weight and glucose homeostasis were monitored. Mice were euthanized and blood, liver, adipose tissue and pancreas were collected and processed for metabolic and inflammatory analysis., [Results]: Body weight and triglycerides profiles in blood and liver were comparable between vehicle- and Abn-CBD-treated DIO mice. However, treatment with Abn-CBD reduced hyperinsulinemia and markers of systemic low-grade inflammation in plasma and fat, also promoting white adipose tissue browning. Pancreatic islets from Abn-CBD-treated mice showed lower apoptosis, inflammation and oxidative stress than vehicle-treated DIO mice, and beta cell proliferation was induced. Furthermore, Abn-CBD lowered hepatic fibrosis, inflammation and macrophage infiltration in the liver when compared to vehicle-treated DIO mice. Importantly, the balance between hepatocyte proliferation and apoptosis was improved in Abn-CBD-treated compared to vehicle-treated DIO mice., [Conclusions]: These results suggest that Abn-CBD exerts beneficial immunomodulatory actions in the liver, pancreas and adipose tissue of DIO prediabetic mice with NAFLD, thus protecting tissues. Neonatal supplementation with resveratrol (RSV) or nicotinamide riboside (NR) programs in male mice brown adipocyte-like features in white adipose tissue (WAT browning) together with improved metabolism in adulthood. We tested the involvement in this programming of long-term epigenetic changes in two browning-related genes that are overexpressed inWAT of supplemented mice, Slc27a1 and Prdm16. Suckling mice received orally the vehicle, RSV or NR from postnatal days 2-to-20. After weaning (d21) onto a chow diet, male mice were habituated to a normal-fat diet (NFD) starting d75, and split on d90 into continuation on the NFD or switching to a high-fat diet (HFD) until euthanization on d164. CpG methylation by bisulfite-sequencing was analyzed on inguinal WAT. Both treatments modified methylation marks in Slc27a1 and Prdm16 and the HFD-dependent dynamics of these marks in the adultWAT, with distinct and common effects. The treatments also a ffected gene expression of de novo DNA methylases in WAT of young animals (euthanized at d35 in independent experiments). Studies in 3T3-L1 adipocytes indicated the direct effects of RSV and NR on the DNA methylation machinery and favoring browning features. Purpose: To compare the healing and clinical outcomes of anterior cruciate ligament (ACL) reconstruction between patients with or without intraoperative administration of adipose-derived regenerative stem cells (ADRC). Methods: Between 2013 and 2014, the outcomes of 20 soccer players undergoing ACL reconstruction using bone–patellar tendon–bone autograft infiltrated with ADRC at the end of the procedure were compared to a historical, matched cohort of 19 soccer players undergoing the same procedure without ADRC. Outcomes were obtained at baseline, and 6 and 12 months postop for IKDC (International Knee Documentation Committee), Lysholm, and Lequesne, and at 2, 4, 6, and 12 months postop for VAS (visual analogue scale) for pain and graft maturation to evaluate the ligamentization process (magnetic resonance imaging (MRI)-based). Results: Both groups significantly improved the IKDC (p < 0.001 in both groups), Lysholm (p < 0.001 in both groups), Lequesne index (p < 0.001 in both groups), VAS for pain (p = 0.002 for the ADRC and p < 0.001 for the control group), and MRI scores (p < 0.001 in both groups) in the 12 months postop compared to baseline scores. However, there were no significant differences in the improvement of the outcomes between groups across time (p > 0.05). All patients returned to sports after surgery, but 8 (40%) patients in the ADRC and 13 (68.4%) patients in the control group had lower Tegner activity score at 12 months postop. Conclusions: Patients receiving ADRC at the time of ACL reconstruction significantly improved knee function and healing/maturation of the graft at 12 months. Treatment with all-trans retinoic acid (ATRA), the carboxylic form of vitamin A, lowers body weight in rodents by promoting oxidative metabolism in multiple tissues including white and brown adipose tissues. We aimed to identify novel markers of the metabolic impact of ATRA through targeted blood metabolomics analyses, with a focus on acylcarnitines and amino acids. Blood was obtained from mice treated with a high ATRA dose (50 mg/kg body weight/day, subcutaneous injection) or placebo (controls) during the 4 days preceding collection. LC-MS/MS analyses with a focus on acylcarnitines and amino acids were conducted on plasma and PBMC. Main results showed that, relative to controls, ATRA-treated mice had in plasma: increased levels of carnitine, acetylcarnitine, and longer acylcarnitine species, decreased levels of citrulline, and increased global arginine bioavailability ratio for nitric oxide synthesis, increased levels of creatine, taurine and docosahexaenoic acid, and a decreased n-6/n-3 polyunsaturated fatty acids ratio. While some of these features likely reflect the stimulation of lipid mobilization and oxidation promoted by ATRA treatment systemically, other may also play a causal role underlying ATRA actions. The results connect ATRA to specific nutrition-modulated biochemical pathways, and suggest novel mechanisms of action of vitamin A-derived retinoic acid on metabolic health.

The endocannabinoid (eCB) system regulates energy homeostasis and is linked to obesity development. However, the exact dynamic and regulation of eCBs in the hypothalamus during obesity progression remain incompletely described and understood. Our study examined the time course of responses in two hypothalamic eCBs, 2-arachidonoylglycerol (2-AG) and arachidonoylethanolamine (AEA), in male and female mice during diet-induced obesity and explored the association of eCB levels with changes in brown adipose tissue (BAT) thermogenesis and body weight. We fed mice a high-fat diet (HFD), which induced a transient increase (substantial at 7 days) in hypothalamic eCBs followed by a progressive decrease to basal levels with a long-term HFD. This transient rise at early stages of obesity is considered a physiologic compensatory response to BAT thermogenesis, which is activated by diet surplus. The eCB dynamic was sexually dimorphic: hypothalamic eCBs levels were higher in female mice, who became obese at later time points than male. The hypothalamic eCBs time course positively correlated with thermogenesis activation but negatively matched body weight, leptinemia, and circulating eCB levels. Increased expression of eCB-synthetizing enzymes accompanied the transient hypothalamic eCB elevation. Intracerebroventricular injection of eCB did not promote BAT thermogenesis; however, administration of thermogenic molecules, such as central leptin or a peripheral β3-adrenoreceptor agonist, induced a significant increase in hypothalamic eCBs, suggesting a directional link from BAT thermogenesis to hypothalamic eCBs. Phaeodactylum tricornutum (P. tricornutum) comprise several lipophilic constituents with proposed anti-obesity and anti-diabetic properties. We investigated the effect of an ethanolic P. tricornutum extract (PTE) on energy metabolism in obesity-prone mice fed a high fat diet (HFD). Six- to eight-week-old male C57BL/6J mice were switched to HFD and, at the same time, received orally placebo or PTE (100 mg or 300 mg/kg body weight/day). Body weight, body composition, and food intake were monitored. After 26 days, blood and tissue samples were collected for biochemical, morphological, and gene expression analyses. PTE-supplemented mice accumulated fucoxanthin metabolites in adipose tissues and attained lower body weight gain, body fat content, weight of white adipose tissue (WAT) depots, and inguinal WAT adipocyte size than controls, independent of decreased food intake. PTE supplementation was associated with lower expression of Mest (a marker of fat tissue expandability) in WAT depots, lower gene expression related to lipid uptake and turnover in visceral WAT, increased expression of genes key to fatty acid oxidation and thermogenesis (Cpt1, Ucp1) in subcutaneous WAT, and signs of thermogenic activation including enhanced UCP1 protein in interscapular brown adipose tissue. The adipose tissue, which is currently viewed as an organ with neuroimmunoendocrine functions, participates in the homeostasis of the human organism. It has great plasticity and functional variability based on the intake of nutrients or to the increase or decrease of its tissue volume, which modifies both the function and the number of the cells that form it or reach it. The elements that are released abnormally by these cells, among other cytokines and adipokines, cause both local and systemic inflammation, mainly when they come from the visceral adipose tissue, and they can affect diverse organs like the liver and the cardio-vascular system. It has been pointed out that obesity entails a greater risk for developing inflammatory, metabolic, autoimmune, or allergic diseases, as well as alterations in scarring, and cancer.

Fat-to-muscle ratio has been proposed as an alternative approach for assessing body fat. The objective of this study was to explore fat-to-muscle ratio thresholds in metabolic syndrome (MetS) diagnosis, it was hypothesised that the fat-to-muscle ratio is a good predictive indicator of MetS in a large population of young Colombian adults. For this purpose, a cross-sectional study was conducted on 1416 subjects (66.6% female), aged from 18.1 to 25.1. As part of the study, measurements of the subjects&rsquo, anthropometric indicators, serum lipid indices, blood pressure, and fasting plasma glucose were taken. Body composition was measured through bioelectrical impedance analysis (BIA). A new variable (ratio of fat mass to muscle mass, in kg) was calculated. Following the International Diabetes Federation (IDF) definition, MetS includes three or more metabolic abnormalities. Receiver operating characteristic (ROC) curves and logistic regression determined the discriminatory ability of the fat-to-muscle ratio to predict MetS. According to the IDF, the best fat-to-muscle ratio cut-off point for detecting MetS in men was 0.225 kg, with an area under the curve (AUC) of 0.83, sensitivity of 80%, and specificity of 70%. For women, the fat-to-muscle ratio cut-off point was 0.495 kg, the AUC was 0.88, and the sensitivity and specificity were 82% and 80%, respectively. In conclusion, our results showed that the fat-to-muscle ratio cut-off points from ROC analyses demonstrate good discriminatory power for detecting MetS in young Colombian adults.

Polycystic ovary syndrome is a highly frequent reproductive-endocrine disorder affecting up to 8?10% of women worldwide atreproductive age. Although its etiology is not fully understood, evidence suggests that insulin resistance, with or withoutcompensatory hyperinsulinemia, and hyperandrogenism are very common features of the polycystic ovary syndrome phenotype.Dysfunctional white adipose tissue has been identified as a major contributing factor for insulin resistance in polycystic ovarysyndrome. Environmental (e.g., chronodisruption) and genetic/epigenetic factors may also play relevant roles in syndromedevelopment. Overweight and/or obesity are very common in women with polycystic ovary syndrome, thus suggesting thatsome polycystic ovary syndrome and metabolic syndrome female phenotypes share common characteristics. Sleep disturbanceshave been reported to double in women with PCOS and obstructive sleep apnea is a common feature in polycystic ovarysyndrome patients. Maturation of the luteinizing hormone-releasing hormone secretion pattern in girls in puberty is closelyrelated to changes in the sleep-wake cycle and could have relevance in the pathogenesis of polycystic ovary syndrome. Thisreview article focuses on two main issues in the polycystic ovary syndrome-metabolic syndrome phenotype development: (a) theimpact of androgen excess on white adipose tissue function and (b) the possible efficacy of adjuvant melatonin therapy toimprove the chronobiologic profile in polycystic ovary syndrome-metabolic syndrome individuals. Phenelzine has been suggested to have an antiobesity effect by inhibiting de novo lipogenesis, which led us to investigate the metabolic effects of oral chronic phenelzine treatment in high-sucrose-drinking mice. Sucrose-drinking mice presented higher body weight gain and adiposity versus controls. Phenelzine addition did not decrease such parameters, even though fat pad lipid content and weights were not different from controls. In visceral adipocytes, phenelzine did not impair insulin-stimulated de novo lipogenesis and had no effect on lipolysis. However, phenelzine reduced the mRNA levels of glucose transporters 1 and 4 and phosphoenolpyruvate carboxykinase in inguinal white adipose tissue (iWAT), and altered circulating levels of free fatty acids (FFA) and glycerol. Interestingly, glycemia was restored in phenelzine-treated mice, which also had higher insulinaemia. Phenelzine-treated mice presented higher rectal temperature, which was associated to reduced mRNA levels of uncoupling protein 1 in brown adipose tissue. Furthermore, unlike sucrose-drinking mice, hepatic malondialdehyde levels were not altered. In conclusion, although de novo lipogenesis was not inhibited by phenelzine, the data suggest that the ability to re-esterify FFA is impaired in iWAT. Summary Thyroid hormones (THs) act in the brain to modulate energy balance. We show that central triiodothyronine (T3) regulates de novo lipogenesis in liver and lipid oxidation in brown adipose tissue (BAT) through the parasympathetic (PSNS) and sympathetic nervous system (SNS), respectively. Central T3 promotes hepatic lipogenesis with parallel stimulation of the thermogenic program in BAT. The action of T3 depends on AMP-activated protein kinase (AMPK)-induced regulation of two signaling pathways in the ventromedial nucleus of the hypothalamus (VMH): decreased ceramide-induced endoplasmic reticulum (ER) stress, which promotes BAT thermogenesis, and increased c-Jun N-terminal kinase (JNK) activation, which controls hepatic lipid metabolism. Of note, ablation of AMPKα1 in steroidogenic factor 1 (SF1) neurons of the VMH fully recapitulated the effect of central T3, pointing to this population in mediating the effect of central THs on metabolism. En la actualidad, las células madre del tejido adiposo (ASC) juegan un papel crucial en las estrategias de reparación o regeneración tisular. Son células mesenquimales multipotenciales que se pueden obtener fácilmente de lipoaspirados. Tras un proceso de digestión enzimática y centrifugación, se recoge la fracción estromal vacular (SVF) en el sedimento obtenido y se purifican las ASC a través del cultivo en plástico y realización de varios subcultivos. Hay diversos aspectos que han convertido a las ASC en una importante opción en terapia celular en los últimos años: su seguridad demostrada en ensayos clínicos; la mayor cantidad de células mesenquimales que se obtienen del tejido adiposo con respecto a la médula ósea, con un proceso menos invasivo y con menor morbilidad; su carácter inmunorregulador, bien a través de la secreción de mediadores moleculares o interacción directa; y finalmente, su gran acción trófica con el objetivo de crear un microambiente adecuado para activar los procesos de reparación del propio tejido. En este trabajo también se indican las dosis celulares y las vías de administración que se están utilizando para la aplicación de las ASC y la SVF. Los próximos años serán cruciales para determinar, gracias a distintos ensayos clínicos en marcha, el potencial terapéutico de las ASC y sus mecanismos de acción.

Adipose tissue (AT) expansion is the result of two processes: hyperplasia and hypertrophy; and both, directly or indirectly, depend on the adipogenic potential of adipocyte precursor cells (APCs). Glucocorticoids (GCs) have a potent stimulatory effect on terminal adipogenesis; while their effects on early stages of adipogenesis are largely unknown. In the present work, we study, in a model of high GC levels, the adipogenic potential of APCs from retroperitoneal AT (RPAT) and its relationship with RPAT mass expansion. We employed a model of hyper-adiposity (30- and 60-day-old rats) due to high endogenous GC levels induced by neonatal treatment with L-monosodium glutamate (MSG).We found that the RPAT APCs from 30-day-old MSG rats showed an increased adipogenic capacity, depending on the APCs’ competency, but not in their number. Analyses of RPAT adipocyte diameter revealed an increase in cell size, regardless of the rat age, indicating the prevalence of a hypertrophic process. Moreover, functional RPAT alterations worsened in 60-day-old rats, suggesting that the hyperplastic AT expansion found in 30-day-old animals might have a protective role. We conclude that GCs chronic excess affects APCs’ adipogenic capacity, modifying their competency. The aim of this work was to determine the effect of a fructose rich diet (FRD) consumed by the pregnant mother on the endocrine-metabolic and in vivo and in vitro adipose tissue (AT) functions of the male offspring in adulthood. At 60 days of age, rats born to FRD-fed mothers (F) showed impaired glucose tolerance after glucose overload and high circulating levels of leptin (LEP). Despite the diminished mass of retroperitoneal AT, this tissue was characterized by enhanced LEP gene expression, and hypertrophic adipocytes secreting in vitro larger amounts of LEP. Analyses of stromal vascular fraction composition by flow cytometry revealed a reduced number of adipocyte precursor cells. Additionally, 60 day-old control (C) and F male rats were subjected to control diet (CC and FC animals) or FRD (CF and FF rats) for three weeks. FF animals were heavier and consumed more calories. Their metabolic-endocrine parameters were aggravated; they developed severe hyperglycemia, hypertriglyceridemia, hyperleptinemia and augmented AT mass with hypertrophic adipocytes. Our study highlights that manipulation of maternal diet induced an offspring phenotype mainly imprinted with a severely unhealthy adipogenic process with undesirable endocrine-metabolic consequences, putting them at high risk for developing a diabetic state., Facultad de Ciencias Médicas

The endocannabinoid (eCB) system can promote food intake by increasing odor detection in mice. The eCB system is over-active in human obesity. Our aim is to measure circulating eCB concentrations and olfactory capacity in a human sample that includes people with obesity and explore the possible interaction between olfaction, obesity and the eCB system. The study sample was made up of 161 females with five groups of body mass index sub-categories ranging from under-weight to morbidly obese. We assessed olfactory capacity with the “Sniffin´Sticks” test, which measures olfactory threshold-discrimination-identification (TDI) capacity. We measured plasma concentrations of the eCBs 2-arachidonoylglycerol (2-AG) and N-arachidonoylethanolamine or anandamide (AEA), and several eCB-related compounds, 2-acylglycerols and N-acylethanolamines. 2-AG and other 2-acylglycerols fasting plasma circulating plasma concentrations were higher in obese and morbidly obese subjects. AEA and other N-acylethanolamine circulating concentrations were lower in under-weight subjects. Olfactory TDI scores were lower in obese and morbidly obese subjects. Lower TDI scores were independently associated with higher 2-AG fasting plasma circulating concentrations, higher %body fat, and higher body mass index, after controlling for age, smoking, menstruation, and use of contraceptives. Our results show that obese subjects have a lower olfactory capacity than non-obese ones and that elevated fasting plasma circulating 2-AG concentrations in obesity are linked to a lower olfactory capacity. In agreement with previous studies we show that eCBs AEA and 2-AG, and their respective congeners have a distinct profile in relation to body mass index. The present report is the first study in humans in which olfactory capacity and circulating eCB concentrations have been measured in the same subjects. Si bien los injertos de tejido adiposo se han venido utilizando durante décadas a lo largo del siglo pasado, la técnica se venía considerando como poco útil dado que el injerto tendía a la reabsorción. Fue Coleman quien introdujo un sistema de obtención-procesamiento-infiltración atraumático que aumentó el porcentaje de supervivencia del injerto y la obtención de resultados positivos similares en su aplicación en todo el mundo. Aún así, la publicación de resultados dispares en la literatura respecto a la supervivencia del injerto, con diferentes porcentajes de reabsorción y el descubrimiento de las células madre en el tejido adiposo, han llevado a la realización de numerosos trabajos de investigación encaminados a comprender la biología del injerto de tejido adiposo, a estudiar el papel de las células madre en este proceso y la influencia que sobre la viabilidad celular tienen los distintos pasos a los que los cirujanos plásticos sometemos al tejido adiposo para su trasplante. Hemos hecho una revisión de la literatura científica al respecto para reunir la información disponible sobre estas controversias.

INTRODUÇÃO: O envelhecimento humano aliado ao estilo de vida sedentário é marcado por alterações metabólicas com modificação na composição corporal, que repercutem diretamente na proteína C-reativa (PCR), sendo este um importante marcador de risco de doenças cardiovasculares (DCV). O treinamento resistido (TR) é um método muito utilizado na prevenção de doenças associadas ao envelhecimento. Objetivo: Avaliar os efeitos de oito semanas de TR sobre a composição corporal, força muscular e PCR em um grupo de idosas.MÉTODO: Foi realizado um estudo experimental com 10 idosas (63 ± 2 anos de idade). Para o ensaio de PCR, análises bioquímicas de hemograma e lipidograma completos foram coletadas amostras de sangue venoso periférico antes do exercício e 24 horas depois. Para medidas antropométricas calcularam-se índice de massa corporal (IMC), relação cintura/quadril (RCQ) e composição corporal. O TR foi realizado por Série Combinada - Bi-Set. Para a análise estatística, primeiramente foi realizado o teste de normalidade de Shapiro-Wilk (p > 0,05) para testes paramétricos. As variáveis do grupo foram apresentadas como média e desvio padrão. Para variáveis de PCR, antropométricas, composição corporal e perfil lipídico foram realizados o teste t de Student,tanto anterior quanto posteriormente às oito semanas de treinamento. Para as amostras da evolução das cargas e médias do consumo alimentar foi realizado o teste One Way ANOVA e, quando necessário, o teste post hoc de Tukey. O nível de significância adotado foi de p ≤ 0,05.RESULTADOS: Houve redução estatisticamente significativa (p = 0,02) para as concentrações séricas de PCR, o que significa uma redução de 70,96%, além de diminuir a massa gorda e aumentar a massa magra e a carga de treino após oito semanas.CONCLUSÃO: Oito semanas de TR reduziram as concentrações séricas de proteína C-reativa, assim como diminuíram a massa gorda e aumentaram o volume muscular, demonstrando ser uma estratégia eficiente para a diminuição dos fatores de riscos de doenças cardiovasculares. Resistance Training (RT) is a widely used method in the prevention of diseases associated with aging. Objective: To evaluate the effects of eight weeks of TR on body composition, muscle strength and CRP in a group of elderly women.METHOD: An experimental study was conducted with 10 elderly women (63 ± 2 years). For the CRP analysis, biochemical analysis, complete blood count and lipid profile samples of peripheral venous blood were collected before exercise and 24 hours later. For anthropometric measurements were calculated the body mass index (BMI), waist/hip ratio (WHR) and body composition. The TR was conducted by Combined Series - Bi-Set. Statistical analysis was first conducted with the Shapiro-Wilk normality test (p>0.05) for parametric tests. Group variables were presented as mean and standard deviation. For CRP, anthropometric, body composition and lipid profile variables, the Student t test was performed, for both previous and later to eight weeks of training. For samples of the loads evolution and average food consumption was performed the One Way ANOVA test and, where necessary, the Tukey's post hoc test. The significance level was set at p≤0.05.RESULTS: There was a statistically significant reduction (p=0.02) for the serum concentrations of CRP, which means a decrease of 70.96%, and reduction of the fat mass and increased lean body mass and the training load after eight weeks.CONCLUSION: Eight weeks of RT reduced serum concentrations of C-reactive protein, as well as decreased fat mass and increased muscle volume, proving to be an effective strategy in reducing the risk factors for cardiovascular disease. El entrenamiento de resistencia (RT) es un método ampliamente utilizado en la prevención de las enfermedades asociadas con el envejecimiento. Objetivo: Evaluar los efectos de las ocho semanas de TR en la composición corporal, la fuerza muscular y la PCR en un grupo de personas de edad avanzada.MÉTODO: Un estudio experimental se llevó a cabo con 10 mujeres de edad avanzada (63 ± 2 años). Para prueba PCR, el análisis bioquímico, hemograma completo y perfil lipídico completo se recogieron muestras de sangre venosa periférica antes del ejercicio y 24 horas después. Para las medidas antropométricas se calcularon el índice de masa corporal (IMC), índice cintura/cadera (ICC) y composición corporal. El TR fue realizado por la Serie Combinada - Bi-Set. El análisis estadístico se realizó por primera vez con la prueba de normalidad de Shapiro-Wilk (p > 0,05) para las pruebas paramétricas. Las variables de grupo se presentan como media y desviación estándar. La PCR, las variables antropométricas, la composición corporal y el perfil lipídico se realizaron con la prueba de la t de Student, tanto anterior cuanto posteriormente a las ocho semanas de entrenamiento. Para las muestras de cambios de carga y el consumo medio de alimentos se realizó la prueba One Way ANOVA y, cuando necesario, la prueba post hoc de Tukey. El nivel de significación se fijó en p ≤ 0,05.RESULTADOS: Hubo una reducción estadísticamente significativa (p = 0,02) para las concentraciones séricas de PCR, lo que significa un descenso del 70,96%, y reducción la masa grasa y aumento de la masa corporal magra y la carga de entrenamiento después de ocho semanas.CONCLUSIÓN: Ocho semanas de RT reducen las concentraciones séricas de proteína C-reactiva, así como de la masa grasa, aumentaron el volumen muscular, demostrando ser una estrategia eficaz para la reducción de los factores de riesgo de enfermedad cardiovascular.

The restoration of body composition (BC) parameters is considered to be one of the most important goals in the treatment of patients with anorexia nervosa (AN). However, little is known about differences between AN diagnostic subtypes [restricting (AN-R) and binge/purging (AN-BP)] and weekly changes in BC during refeeding treatment. Therefore, the main objectives of our study were twofold: 1) to assess the changes in BC throughout nutritional treatment in an AN sample and 2) to analyze predictors of BC changes during treatment, as well as predictors of treatment outcome. The whole sample comprised 261 participants [118 adult females with AN (70 AN-R vs. 48 AN-BP), and 143 healthy controls]. BC was measured weekly during 15 weeks of day-hospital treatment using bioelectrical impedance analysis (BIA). Assessment measures also included the Eating Disorders Inventory-2, as well as a number of other clinical indices. Overall, the results showed that AN-R and AN-BP patients statistically differed in all BC measures at admission. However, no significant time×group interaction was found for almost all BC parameters. Significant time×group interactions were only found for basal metabolic rate (p = .041) and body mass index (BMI) (p = .035). Multiple regression models showed that the best predictors of pre-post changes in BC parameters (namely fat-free mass, muscular mass, total body water and BMI) were the baseline values of BC parameters. Stepwise predictive logistic regressions showed that only BMI and age were significantly associated with outcome, but not with the percentage of body fat. In conclusion, these data suggest that although AN patients tended to restore all BC parameters during nutritional treatment, only AN-BP patients obtained the same fat mass values as healthy controls. Put succinctly, the best predictors of changes in BC were baseline BC values, which did not, however, seem to influence treatment outcome. Fundamentos: La evaluación nutricional durante la premenopausia y la menopausia tiene un papel relevante para valorar los cambios que acontecen en la mujer. El objetivo de este estudio es comparar los parámetros grasa corporal e índice adiposo- muscular corporal (IAMC) estimados mediante impedanciometría respecto al índice de masa corporal (IMC) en la definición del estado nutricional y la composición corporal. Métodos: Estudio descriptivo transversal. Mediante muestreo aleatorio (base de tarjeta sanitaria) fueron seleccionadas 30 mujeres de 35 a 55 años, valorándose en ellas la asociación entre grasa corporal e IAMC con el IMC; diferentes estimadores de la distribución de la grasa corporal: circunferencias de la cintura y cresta ilíaca, cocientes cintura-cadera e ilíaca-cadera y diámetros ilíaco y sagital; presiones arteriales sistólica (PAS) y diastólica (PAD) y niveles plasmáticos de glucosa, colesterol, HDL-colesterol y triglicéridos. Resultados: El porcentaje de grasa corporal correlacionó intensamente (p < 0,001) con IMC (r=0,919), circunferencias de la cintura (r=0,866) e ilíaca (r=0,841) y diámetros sagital (r=0,783) e ilíaco (r=0,772), y, menos intensamente, con la glucemia (r=0,385; p=0,036) y la PAS (r=0,497; p=0,005) y PAD (r=0,582; p=0,001). El IAMC presentó similares resultados, sin asociarse significativamente con ningún parámetro bioquímico. El 46,7% de las mujeres obesas según su porcentaje de grasa corporal (> 33%) presentaba un IMC menor de 30. Conclusiones: La estimación de los parámetros grasa corporal e IAMC en mujeres de 35 a 55 años complementa la evaluación nutricional realizada mediante el IMC, aunque para definir su verdadero valor en esta evaluación es necesario establecer los parámetros de normalidad en la población.

Journal Article; Recent studies have linked gestational exposure to highly caloric diets with a disrupted endogenous cannabinoid system (ECS). In the present study, we have extended these studies by analyzing the impact of the exposure to a palatable diet during gestation and lactation on a) the adult expression of endocannabinoid-related behaviors, b) the metabolic profile of adult offspring and c) the mRNA expression of the signaling machinery of the ECS in the hypothalamus, the liver and the adipose tissue of adult offspring of both sexes. Exposure to a palatable diet resulted in a) sex-dimorphic and perinatal diet specific feeding behaviors, including the differential response to the inhibitory effects of the cannabinoid receptor inverse agonist AM251, b) features of metabolic syndrome including increased adiposity, hyperleptinemia, hypertriglyceridemia and hypercholesterolemia and c) tissue and sex-specific changes in the expression of both CB1 and CB2 receptors and in that of the endocannabinoid-degrading enzymes FAAH and MAGL, being the adipose tissue the most affected organ analyzed. Since the effects were observed in adult animals that were weaned while consuming a normal diet, the present results indicate that the ECS is one of the targets of maternal programming of the offspring energy expenditure. These results clearly indicate that the maternal diet has long-term effects on the development of pups through multiple alterations of signaling homeostatic pathways that include the ECS. The potential relevance of these alterations for the current obesity epidemic is discussed. Yes

Context: Glucose-dependent insulinotropic peptide (GIP) has a central role in glucose homeostasis through its amplification of insulin secretion; however, its physiological role in adipose tissue is unclear. Objective: Our objective was to define the function of GIP in human adipose tissue in relation to obesity and insulin resistance. Design: GIP receptor (GIPR) expression was analyzed in human sc adipose tissue (SAT) and visceral adipose (VAT) from lean and obese subjects in 3 independent cohorts. GIPR expression was associated with anthropometric and biochemical variables. GIP responsiveness on insulin sensitivity was analyzed in human adipocyte cell lines in normoxic and hypoxic environments as well as in adipose-derived stem cells obtained from lean and obese patients. Results: GIPR expression was downregulated in SAT from obese patients and correlated negatively with body mass index, waist circumference, systolic blood pressure, and glucose and triglyceride levels. Furthermore, homeostasis model assessment of insulin resistance, glucose, and G protein-coupled receptor kinase 2 (GRK2) emerged as variables strongly associated with GIPR expression in SAT. Glucose uptake studies and insulin signaling in human adipocytes revealed GIP as an insulin-sensitizer incretin. Immunoprecipitation experiments suggested that GIP promotes the interaction of GRK2 with GIPR and decreases the association of GRK2 to insulin receptor substrate 1. These effects of GIP observed under normoxia were lost in human fat cells cultured in hypoxia. In support of this, GIP increased insulin sensitivity in human adipose-derived stem cells from lean patients. GIP also induced GIPR expression, which was concomitant with a downregulation of the incretin-degrading enzyme dipeptidyl peptidase 4. None of the physiological effects of GIP were detected in human fat cells obtained from an obese environment with reduced levels of GIPR. Conclusions: GIP/GIPR signaling is disrupted in insulin-resistant states, such as obesity, and normalizing this function might represent a potential therapy in the treatment of obesity-associated metabolic disorders.

Context: Recently irisin (encoded by Fndc5 gene) has been reported to stimulate browning and uncoupling protein 1 expression in sc adipose tissue of mice. Objective: The objective of the study was to investigate FNDC5 gene expression in human muscle and adipose tissue and circulating irisin according to obesity, insulin sensitivity, and type 2 diabetes. Design, Patients, and Main Outcome Measure: Adipose tissue FNDC5 gene expression and circulating irisin (ELISA) were analyzed in 2 different cohorts (n = 125 and n = 76); muscle FNDC5 expression was also evaluated in a subcohort of 34 subjects. In vitro studies in human preadipocytes and adipocytes and in induced browning of 3T3-L1 cells (by means of retinoblastoma 1 silencing) were also performed. Results: In both sc and visceral adipose tissue, FNDC5 gene expression decreased significantly in association with obesity and was positively associated with brown adipose tissue markers, lipogenic, insulin pathway-related, mitochondrial, and alternative macrophage gene markers and negatively associated with LEP, TNFα, and FSP27 (a known repressor of brown genes). Circulating irisin and irisin levels in adipose tissue were significantly associated with FNDC5 gene expression in adipose tissue. In muscle, the FNDC5 gene was 200-fold more expressed than in adipose tissue, and its expression was associated with body mass index, PGC1α, and other mitochondrial genes. In obese participants, FNDC5 gene expression in muscle was significantly decreased in association with type 2 diabetes. Interestingly, muscle FNDC5 gene expression was significantly associated with FNDC5 and UCP1 gene expression in visceral adipose tissue. In men, circulating irisin levels were negatively associated with obesity and insulin resistance. Irisin was secreted from human adipocytes into the media, and the induction of browning in 3T3-L1 cells led to increased secreted irisin levels. Conclusions: Decreased circulating irisin concentration and FNDC5 gene expression in adipose tissue and muscle from obese and type 2 diabetic subjects suggests a loss of brown-like characteristics and a potential target for therapy.

El injerto de tejido adiposo se ha convertido en una de las principales herramientas con la que contamos en Cirugía Plástica y, como no podía ser de otra forma, la mama es el principal órgano diana de nuestra actuación. Desde que introdujimos la técnica en nuestra práctica privada en el año 2009 hemos ido incrementando las indicaciones de su uso. Las tres principales son el aumento mamario, la corrección de defectos y asimetrías y más recientemente el aumento con combinación de ambas técnicas. El aumento mamario con tejido adiposo está indicado en pacientes que quieran incrementar el volumen sin usar implantes. Hay que explicar muy bien a la paciente lo que podemos conseguir en cuanto a volumen y proyección de la mama, además de los problemas potenciales de aparición en las mamografías de quistes y microcalcificaciones. El tejido adiposo, en cambio, es un gran complemento de los implantes mamarios, ya que permite corregir deformidades y asimetrías que con los implantes son más difíciles de resolver. El aumento de mamas combinando ambas técnicas es la consecuencia final y evidente de esta evolución terapéutica, ya que aúna las ventajas de ambas: la firmeza y volumen estables que proporcionan los implantes y la plasticidad que aporta el tejido adiposo para lograr una mayor simetría.

El tejido adiposo obtenido mediante liposucción es una fuente idónea para aislar células con potencial terapéutico, las denominadas células de la fracción estromal vascular (FEV), que incluyen células madre mesenquimales. Estas células se han convertido en una de las principales herramientas de terapia celular autóloga para diversas aplicaciones médicas, y en los últimos años se han ido desarrollando diversas tecnologías para su aislamiento y uso clínico. En este trabajo presentamos un nuevo método rápido, sencillo y eficiente para el aislamiento de células de la FEV mediante un dispositivo médico cerrado que permite recoger y procesar lipoaspirados humanos en el mismo procedimiento quirúrgico de manera coste-efectiva. Además describimos los métodos llevados a cabo para cuantificar la calidad, seguridad y eficacia del inóculo celular obtenido.

Objective Munc18c is associated with glucose metabolism and could play a relevant role in obesity. However, little is known about the regulation of Munc18c expression. We analyzed Munc18c gene expression in human visceral (VAT) and subcutaneous (SAT) adipose tissue and its relationship with obesity and insulin. La terapia basada en células madre adultas constituye la forma de tratamiento más innovadora para la regeneración de los tejidos patológicos, ausentes o deficitarios; también de relleno estético apropiado para variedad de indicaciones cosméticas y reconstructivas, y una de las líneas prioritarias de investigación traslacional en todo el mundo, evitando los problemas de rechazo inmune de los homotrasplantes, las complicaciones inherentes a la implantación de otros materiales aloplásticos, las secuelas estéticas o funcionales de las zonas donantes de los autotrasplantes de mayor morbilidad, y los problemas éticos del uso de células madre embrionarias. El cirujano plástico tiene un papel protagonista tanto en la extracción del injerto de células madre del abdomen mediante una liposucción manual tumescente, como en la aplicación clínica rápida sobre distintos defectos, deformidades o pérdidas de sustancia. Presentamos el protocolo de extracción y procesamiento de células madre derivadas del tejido adiposo (ADSC) aprobado por el Comité Ético de Investigación Clínica, que resulta ser factible, seguro y reproducible, pudiendo servir de guía de actuación para el diseño de nuevos estudios clínicos controlados sobre la eficacia del autotrasplante de células madre aplicados a muchas otras patologías en el universo de la Cirugía Plástica en el que esté comprometida la cicatrización.

Breast Cancer 1 (BrCa1) interacts with acetyl-CoA carboxylase (ACC) reducing the rate of fatty acid biosynthesis. In this study, we aimed to evaluate BrCa1 in human adipose tissue according to obesity and insulin resistance, and in vitro cultured adipocytes. [Research Design and Methods]: BrCa1 gene expression, total and phosphorylated (P-) BrCa1, and ACC were analyzed in adipose tissue samples obtained from a total sample of 133 subjects. BrCa1 expression was also evaluated during in vitro differentiation of human adipocytes and 3T3-L1 cells. [Results]: BrCa1 gene expression was significantly up-regulated in both omental (OM; 1.36-fold, p = 0.002) and subcutaneous (SC; 1.49-fold, p = 0.001) adipose tissue from obese subjects. In parallel with increased BrCa1 mRNA, P-ACC was also up-regulated in SC (p = 0.007) as well as in OM (p = 0.010) fat from obese subjects. However, at present, there are no studies highlighting the relationship between human adipose tissue angiogenesis and obesity-associated insulin resistance (IR). [Results]: Our aim was to analyze and compare angiogenic factor expression levels in both subcutaneous (SC) and omentum (OM) adipose tissues from morbidly obese patients (n = 26) with low (OB/L-IR) (healthy obese) and high (OB/H-IR) degrees of IR, and lean controls (n = 17). Another objective was to examine angiogenic factor correlations with obesity and IR. Here we found that VEGF-A was the isoform with higher expression in both OM and SC adipose tissues, and was up-regulated 3-fold, together with MMP9 in OB/L-IR as compared to leans. This up-regulation decreased by 23% in OB/-H-IR compared to OB/L-IR. On the contrary, VEGF-B, VEGF-C and VEGF-D, together with MMP15 was down-regulated in both OB/H-IR and OB/L-IR compared to lean patients. Moreover, MMP9 correlated positively and VEGF-C, VEGF-D and MMP15 correlated negatively with HOMA-IR, in both SC and OM. However, the association between lipogenic and lipolytic genes and the evolution of morbidly obese subjects after bariatric surgery remains unknown. In this prospective study we analyze the association between the improvement in the morbidly obese patients as a result of bariatric surgery and the basal expression of lipogenic and lipolytic genes. METHODS We study 23 non diabetic morbidly obese patients who were studied before and 7 months after bariatric surgery. Also, we analyze the relative basal mRNA expression levels of lipogenic and lipolytic genes in epiploic visceral adipose tissue (VAT) and abdominal subcutaneous adipose tissue (SAT). RESULTS When the basal acetyl-CoA carboxylase 1 (ACC1), acetyl-CoA synthetase 2 (ACSS2) and ATP citrate lyase (ACL) expression in SAT was below percentile-50, there was a greater decrease in weight (P = 0.006, P = 0.034, P = 0.026), body mass index (P = 0.008, P = 0.033, P = 0.034) and hip circumference (P = 0.033, P = 0.021, P = 0.083) after bariatric surgery. In VAT, when the basal ACSS2 expression was below percentile-50, there was a greater decrease in hip circumference (P = 0.006). After adjusting for confounding variables in logistic regression models, only the morbidly obese patients with SAT or VAT ACSS2 expression ≥ P50 before bariatric surgery had a lower percentage hip circumference loss (

Context: Adipose tissue hypoxia and endoplasmic reticulum (ER) stress may link the presence of chronic inflammation and macrophage infiltration in severely obese subjects. We previously reported the up-regulation of TNF-like weak inducer of apoptosis (TWEAK)/fibroblast growth factor-inducible 14 (Fn14) axis in adipose tissue of severely obese type 2 diabetic subjects. Objectives: The objective of the study was to examine TWEAK and Fn14 adipose tissue expression in obesity, severe obesity, and type 2 diabetes in relation to hypoxia and ER stress. Design: In the obesity study, 19 lean, 28 overweight, and 15 obese nondiabetic subjects were studied. In the severe obesity study, 23 severely obese and 35 control subjects were studied. In the type 2 diabetes study, 11 type 2 diabetic and 36 control subjects were studied. The expression levels of the following genes were analyzed in paired samples of sc and visceral adipose tissue: Fn14, TWEAK, VISFATIN, HYOU1, FIAF, HIF-1a, VEGF, GLUT-1, GRP78, and XBP-1. The effect of hypoxia, inflammation, and ER stress on the expression of TWEAK and Fn14 was examined in human adipocyte and macrophage cell lines. Results: Up-regulation of TWEAK/Fn14 and hypoxia and ER stress surrogate gene expression was observed in sc and visceral adipose tissue only in our severely obese cohort. Hypoxia modulates TWEAK or Fn14 expression in neither adipocytes nor macrophages. On the contrary, inflammation up-regulated TWEAK in macrophages and Fn14 expression in adipocytes. Moreover, TWEAK had a proinflammatory effect in adipocytes mediated by the nuclear factor-κB and ERK but not JNK signaling pathways. Conclusions: Our data suggest that TWEAK acts as a pro-inflammatory cytokine in the adipose tissue and that inflammation, but not hypoxia, may be behind its up-regulation in severe obesity.

Durante la última década, la terapia celular ha emergido como una estrategia útil en el tratamiento de diversas enfermedades como la isquemia miocárdica y las fístulas en la enfermedad de Crohn; pero últimamente, hay también ya líneas de investigación centradas en su uso en reconstrucción mamaria, cuyos resultados van siendo publicados paulatinamente. Existen varios tipos de células madre adultas que han sido investigadas en estudios preclínicos y clínicos diseñados para este propósito: células de medula ósea, células del sistema circulatorio y mioblastos y, recientemente se está trabajando en una población de células madre en el tejido adiposo, que presentan una fácil extracción y manipulación. Estas células son capaces de diferenciarse en múltiples líneas celulares, como los adipocitos y las células endoteliales entre otras. En el presente artículo, trataremos de hacer una revisión de los principios básicos de las células madre derivadas del tejido adiposo (tipos, características, procesos de obtención y multiplicación), los primeros estudios experimentales y los ensayos clínicos que están siendo realizados en la actualidad.

[ES]: Se estudia la influencia de las 3 variedades más importantes de la grasa de la dieta (cantidad, calidad y composición) sobre la composición y distribución de ácidos grasos del tejido adiposo, utilizando ratas como animales de experimentación. Los resultados obtenidos a partir de 11 grupos de ratas permiten deducir las siguientes conclusiones: 1. - La composición de ácidos grasos de la grasa del tejido adiposo puede ser muy variable. Contenidos de grasa en la dieta superior al 12% originan un perfil de ácidos grasos en tejido adiposo muy similar al de la grasa ingerida. 2.- La alteración de la grasa de la dieta modifica la composición de ácidos grasos del tejido adiposo, debido a la menor cantidad de grasa absorbida. 3.- La distribución de los ácidos grasos en los triglicéridos del tejido adiposo es independiente de la de la grasa ingerida., [EN]: In this paper the effect of the 3 most important variables (quantity, quality and composition) of dietary fat on the distribution and composition of fatty acids in rat adipose tissue, is studied. From the results obtained after feeding 11 groups of rats, the following conclusions stand out: 1 . - Fatty acid composition of rat adipose tissue is very variable. Nevertheless, when the percentage of fat in the diet is higher than 12%, the fatty acid composition of the adipose tissue is similar to that of the ingested fat. 2.- The level of alteration of the fat in the diet clearly modifies the fat composition In adipose tissue. Journal Article; Research Support, Non-U.S. Gov't; It is well known that the adult human thymus degenerates into fat tissue; however, it has never been considered as a potential source of angiogenic factors. Recently, we have described that this fat (TAT) produces angiogenic factors and induces human endothelial cell proliferation and migration, indicating its potential angiogenic properties. DESIGN Adult thymus fat and subcutaneous adipose tissue specimens were obtained from 28 patients undergoing cardiac surgery, making this tissue readily available as a prime source of adipose tissue. We focused our investigation on determining VEGF gene expression and characterizing the different genes, mediators of inflammation and adipogenesis, and which are known to play a relevant role in angiogenesis regulation. RESULTS We found that VEGF-A was the isoform most expressed in TAT. This expression was accompanied by an upregulation of HIF-1alpha, COX-2 and HO-1 proteins, and by increased HIF-1 DNA binding activity, compared to SAT. Furthermore, we observed that TAT contains a high percentage of mature adipocytes, 0.25% of macrophage cells, 15% of endothelial cells and a very low percentage of thymocyte cells, suggesting the cellular variability of TAT, which could explain the differences in gene expression observed in TAT. Subsequently, we showed that the expression of genes known as adipogenic mediators, including PPARgamma1/gamma2, FABP-4 and adiponectin was similar in both TAT and SAT. Moreover the expression of these latter genes presented a significantly positive correlation with VEGF, suggesting the potential association between VEGF and the generation of adipose tissue in adult thymus. CONCLUSION Here we suggest that this fat has a potential angiogenic function related to ongoing adipogenesis, which substitutes immune functions within the adult thymus. The expression of VEGF seems to be associated with COX-2, HO-1 and adipogenesis related genes, suggesting the importance that this new fat has acquired in research in relation to adipogenesis and angiogenesis. Visfatin is an adipokine highly expressed in visceral AT (adipose tissue) of humans and rodents, the production of which seems to be dysregulated in excessive fat accumulation and conditions of insulin resistance. EPA (eicosapentaenoic acid), an n−3 PUFA (polyunsaturated fatty acid), has been demonstrated to exert beneficial effects in obesity and insulin resistance conditions, which have been further linked to its reported ability to modulate adipokine production by adipocytes. TNF-α (tumour necrosis factor-α) is a pro-inflammatory cytokine whose production is increased in obesity and is involved in the development of insulin resistance. Control of adipokine production by some insulin-sensitizing compounds has been associated with the stimulation of AMPK (AMP-activated protein kinase). The aim of the present study was to examine in vitro the effects of EPA on visfatin production and the potential involvement of AMPK both in the absence or presence of TNF-α. Treatment with the pro-inflammatory cytokine TNF-α (1 ng/ml) did not modify visfatin gene expression and protein secretion in primary cultured rat adipocytes. La obtención de tejido adiposo supone un nuevo y prometedor mercado de trabajo para los cirujanos plásticos, ya que los bancos de tejidos escogerán de forma acertada la grasa como el medio más fácil para obtener fuentes de células madre de alto rendimiento, en la medida en que este tejido es capaz de producir al menos cinco veces más unidades formadores de colonias (UFCs) que la médula ósea. El objetivo del presente trabajo es mostrar lo que se puede esperar del tejido adiposo como origen de células adultas de fracción vacular estromal (FVE), y señalar las mejores áreas del cuerpo humano para ser elegidas como donantes de tejido adiposo, extraído mediante liposucción. Describimos la rutina seguida para la obtención de células de FVE mediante la digestión de las muestras de tejido adiposo humano con colagenasa. En el momento de su recolección, esas células presentaban una viabilidad de 92+/- 1% basada en exclusión por Azul de Trypan. Las células de FVE recontadas después de permanecer 48 horas en medio de cultivo de Eagle modificado por Dulbecco (DMEM), dentro de una cámara de Neubauer, tras lo cual el rendimiento medio de las células de FVE fue de 7,2 +/- 1,3 x 103 células por mililitro de tejido lipoaspirado. En conclusión, pensamos que supone un desafío en la actualidad el mejorar las estrategias para la obtención de células de FVE. Este trabajo, por ahora preliminar, muestra que las células de FVE pueden ser fácilmente obtenidas por medio de lipoaspiración. La comparación entre las diferentes áreas donantes, mostró un rendimiento 22% más alto para las células de FVE cuando el tejido adiposo había sido obtenido del tronco, en comparación a cuando lo había sido de los miembros.

Biomarkers could provide a more accurate measure of long-term intake than questionnaires. Adipose tissue is considered the best indicator of long-term essential fatty acid intake, but other tissues may prove equally valid. The authors evaluated the ability of fasting whole blood, relative to fasting plasma and adipose tissue, to reflect fatty acid intake. Costa Rican men (n = 99) and women (n = 101) completed a 135-item food frequency questionnaire and provided adipose tissue and blood samples from 1999 to 2001. Fatty acids were identified by using capillary gas chromatography. Correlation coefficients adjusted for age, sex, and body mass index were calculated. Diet-tissue correlation coefficients for α-linolenic acid and linoleic acid, respectively, were 0.38 and 0.43 in whole blood, 0.51 and 0.52 in adipose tissue, and 0.39 and 0.41 in plasma. High correlations were observed between whole-blood α-linolenic and linoleic acid and adipose tissue (r = 0.59 and r = 0.67) and plasma (r = 0.96 and r = 0.88), respectively. Results show that fasting whole blood is a suitable biomarker of long-term essential fatty acid intake, and its performance is comparable to that of fasting plasma. Thus, fasting whole blood could be the sample of choice in epidemiologic studies because of its ability to predict intake, its accessibility, and minimum sample processing. This article analyzes food consumption in relation to levels of DDE (the principal metabolite of DDT) in the adipose tissue of 207 Mexican women residing in States with high and low exposure to DDT. Data on the women's dietary habits and childbearing history were obtained from a personal interview. Adipose tissue DDE levels were measured by gas-liquid chromatography and compared by analysis of variance (ANOVA) and multiple linear regression. Adipose tissue DDE levels increased significantly with age (p = 0.005) and residence in coastal areas (p = 0.002) and non-significantly with the consumption of onion, cauliflower, prickly pear, squash blossoms, sweet corn, broad beans, chili pepper sauce, ham, and fish. Even so, during breastfeeding there was a non-significant reduction in these levels. The findings suggest that certain foods serve as vehicles for DDE residues and confirm that breastfeeding is a mechanism for the elimination of this insecticide, which accumulates over the years in the human body.

Fueron evaluados los niveles de residuos de pesticidas organoclorados (p,p’-DDE, HCB, g-HCH, aldrín) presentes en 123 muestras de tejido adiposo mamario obtenidas de mujeres residentes en Córdoba (España). Además, se investigó la influencia de hábitos alimentarios y edad de las donantes sobre los niveles de organoclorados encontrados en el tejido. El p,p’-DDE fue el compuesto encontrado en concentraciones más altas (1,848mg/g), seguido de HCB (0,235mg/g), aldrín (0,005mg/g) y g-HCH (0,003mg/g). Las donantes mayores presentaron niveles de p,p’-DDE, aldrín y HCB superiores que las más jóvenes. Sin embargo, la edad no influyó en los niveles de g-HCH. La mayoría de las donantes (40,65%) indicaron las frutas como el alimento más consumido, seguido de la alimentación variada (34,15%), los legumbres (6,50%), los guisos y potajes (6,50%), las verduras (4,88%), la leche (3,25%), el pescado (2,44%) y la carne (1,63%). Una correlación no significativa fue observada entre los niveles de organoclorados en el tejido adiposo y los hábitos alimentarios de las donantes The levels of organochlorine pesticide residues (p,p’-DDE, HCB, g-HCH, aldrin) present in adipose tissue samples from mammary glands of 123 women living in Cordoba (Spain) were evaluated. Decreased M2 in adipose tissue is known to be associated with obesity and inflammation-related metabolic disturbances. Here we aimed to investigate MCR1 relative to CD68 (total macrophages) gene expression in association with adipogenic and mitochondrial genes, which were measured in human visceral [VWAT, n = 147] and subcutaneous adipose tissue [SWAT, n = 76] and in rectus abdominis muscle (n = 23). The effects of surgery-induced weight loss were also longitudinally evaluated (n = ).[Results]: MCR1 and CD68 gene expression levels were similar in VWAT and SWAT. A higher proportion of CD206 relative to total CD68 was present in subjects with less body fat and lower fasting glucose concentrations. The ratio MCR1/CD68was positively associated with IRS1gene expression and with the expression of lipogenic genes such as ACACA, FASN and THRSP, even after adjusting for BMI. The ratio MCR1/CD68 in SWAT increased significantly after the surgery-induced weight loss (+44.7%; p = 0.005) in parallel to the expression of adipogenic genes. In addition, SWAT MCR1/CD68ratio was significantly associated with muscle mitochondrial gene expression (PPARGC1A, TFAM and MT-CO3). AT CD206 was confirmed by immunohistochemistry to be specific of macrophages, especially abundant in crown-like structures. [Objective]: In patients with a wide BMI range, we analyze FABP4 expression in adipose and hepatic tissues in the settings of obesity and insulin resistance. Associations between FABP4 expression in adipose tissue and the FABP4 plasma level as well as the main adipogenic and lipolytic genes expressed in adipose tissue were also analyzed. [Methods]: The expression of several lipogenic, lipolytic, PPAR family and FABP family genes was analyzed by real time PCR. FABP4 protein expression in total adipose tissues and its fractions were determined by western blot. [Results]: In obesity FABP4 expression was down-regulated (at both mRNA and protein levels), with its levels mainly predicted by ATGL and inversely by the HOMA-IR index. The BMI appeared as the only determinant of the FABP4 variation in both adipose tissue depots. FABP4 plasma levels showed a significant progressive increase according to BMI but no association was detected between FABP4 circulating levels and SAT or VAT FABP4 gene expression. The gene expression of FABP1, FABP4 and FABP5 in hepatic tissue was significantly higher in tissue from the obese IR patients compared to the non-IR group. [Conclusion]: The inverse pattern in FABP4 expression between adipose and hepatic tissue observed in morbid obese patients, regarding the IR context, suggests that both tissues may act in a balanced manner. Objective:Carnitine palmitoyltransferase 1C (CPT1C) is implicated in central regulation of energy homeostasis. Our aim was to investigatewhether CPT1C in the ventromedial nucleus of the hypothalamus (VMH) is involved in the activation of brown adipose tissue (BAT) thermogenesisin the early stages of diet-induced obesity.Methods:CPT1C KO and wild type (WT) mice were exposed to short-term high-fat (HF) diet feeding or to intracerebroventricular leptinadministration and BAT thermogenesis activation was evaluated. Body weight, adiposity, food intake, and leptinemia were also assayed.Results:Under 7 days of HF diet, WT mice showed a maximum activation peak of BAT thermogenesis that counteracted obesity development,whereas this activation was impaired in CPT1C KO mice. KO animals evidenced higher body weight, adiposity, hyperleptinemia, ER stress, anddisrupted hypothalamic leptin signaling. Leptin-induced BAT thermogenesis was abolished in KO mice. These results indicate an earlier onsetleptin resistance in CPT1C KO mice. Since AMPK in the VMH is crucial in the regulation of BAT thermogenesis, we analyzed if CPT1C was adownstream factor of this pathway. Genetic inactivation of AMPK within the VMH was unable to induce BAT thermogenesis and body weight lossin KO mice, indicating that CPT1C is likely downstream AMPK in the central mechanism modulating thermogenesis within the VMH.
