Paper-based devices provide an alternative technology for simple, low-cost, portable, and disposable diagnostic tools for many applications, including clinical diagnosis, food quality control, and environmental monitoring. In this study we report a two-step fabrication process for creating two-dimensional microfluidic channels to move liquids on a hydrophobized paper surface. A highly hydrophobic surface was created on paper by TiO2 nanoparticle coating using a high-speed, roll-to-roll liquid flame spray technique. The hydrophilic pattern was then generated by UV irradiation through a photomask utilizing the photocatalytic property of TiO2. The flow dynamics of five model liquids with differing surface tensions 48-72 mN·m(-1) and viscosities 1-15 mN·m(-2) was studied. The results show that the liquid front (l) in a channel advances in time (t) according to the power law l=Zt0.5 (Z is an empirical constant which depend on the liquid properties and channel dimensions). The flow dynamics of the liquids with low viscosity show a dependence on the channel width and the droplet volume, while the flow of liquids with high viscosity is mainly controlled by the viscous forces.

An on-field colorimetric sensing strategy employing gold nanoparticles (AuNPs) and a paper-based analytical platform was investigated for mercury ion (Hg(2+)) detection at water sources. By utilizing thymine-Hg(2+)-thymine (T-Hg(2+)-T) coordination chemistry, label-free detection oligonucleotide sequences were attached to unmodified gold nanoparticles to provide rapid mercury ion sensing without complicated and time-consuming thiolated or other costly labeled probe preparation processes. Not only is this strategy's sensing mechanism specific toward Hg(2+), rather than other metal ions, but also the conformational change in the detection oligonucleotide sequences introduces different degrees of AuNP aggregation that causes the color of AuNPs to exhibit a mixture variance. To eliminate the use of sophisticated equipment and minimize the power requirement for data analysis and transmission, the color variance of multiple detection results were transferred and concentrated on cellulose-based paper analytical devices, and the data were subsequently transmitted for the readout and storage of results using cloud computing via a smartphone. As a result, a detection limit of 50 nM for Hg(2+) spiked pond and river water could be achieved. Furthermore, multiple tests could be performed simultaneously with a 40 min turnaround time. These results suggest that the proposed platform possesses the capability for sensitive and high-throughput on-site mercury pollution monitoring in resource-constrained settings.

We report a new type of paper analytical device that provides quantitative electrochemical output and detects concentrations as low as 767 fM. The model analyte is labeled with silver nanoparticles (AgNPs), which provide 250 000-fold amplification. AgNPs eliminate the need for enzymatic amplification, thereby improving device stability and response time. The use of magnetic beads to preconcentrate the AgNPs at the detection electrode further improves sensitivity. Response time is improved by incorporation of a hollow channel, which increases the flow rate in the device by a factor of 7 and facilitates the use of magnetic beads. A key reaction necessary for label detection is made possible by the presence of a slip layer, a fluidic switch that can be actuated by manually slipping a piece of paper. The design of the device is versatile and should be useful for detection of proteins, nucleic acids, and microbes.

There is a growing need for multiplexed specific IgE tests that can accurately evaluate patient sensitization profiles. However, currently available commercial tests are either single/low-plexed or require sophisticated instrumentation at considerable cost per assay. Here, we present a novel convenient lateral flow microarray-based device that employs a novel dual labelled gold nanoparticle-strategy for rapid and sensitive detection of a panel of 15 specific IgE responses in 35 clinical serum samples. Each gold nanoparticle was conjugated to an optimized ratio of HRP and anti-IgE, allowing significant enzymatic amplification to improve the sensitivity of the assay as compared to commercially available detection reagents. The mean inter-assay variability of the developed LFM assay was 12% CV, and analysis of a cohort of clinical samples (n = 35) revealed good general agreement with ImmunoCAP, yet with a varying performance among allergens (AUC = [0.54-0.88], threshold 1 kU). Due to the rapid and simple procedure, inexpensive materials and read-out by means of a consumer flatbed scanner, the presented assay may provide an interesting low-cost alternative to existing multiplexed methods when thresholds > 1 kU are acceptable.

Bioactive papers are usually challenged by four major limitations: sensitivity, selectivity, simplicity and strength (4S). Gold nanoparticles (AuNPs) treated paper has previously been demonstrated as a Surface Enhanced Raman Scattering (SERS) active substrate, capable of addressing the 4S issues. In this study, AuNPs on paper substrate were functionalized by a series of biomolecules to develop a generic SERS platform for antibody-antigen detection. The functionalization steps were performed by taking advantage of the high affinity association between Streptomyces avidinii-derived protein, streptavidin, and biotin. Streptavidin was firstly bound onto the AuNPs treated paper using biotinylated-thiol. Subsequently, desired biotinylated-antibody was bound onto the streptavidin. SERS spectra of each functionalization step were obtained to ensure specific adsorption of the bio-molecules. The binding interaction of the antibody with its specific antigen was detected using SERS. Shifts of Raman band associated with α-helix and β-sheet structures indicated structural modification of the antibody upon interaction with its antigen. Predominant tryptophan and tyrosine residue bands were also detected, confirming the presence of antigen. Reproducible spectral features were quantified as AuNP papers were subjected to different concentrations of antigen; the spectra intensity increased as a function of the antigen concentration. The retention of AuNPs on paper remained constant after all the consecutive washing and functionalization steps. The feasibility of AuNPs paper as a low-cost and generic SERS platform for bio-diagnostic applications was demonstrated.

Paper-based analytical devices (PADs) represent a growing class of elegant, yet inexpensive chemical sensor technologies designed for point-of-use applications. Most PADs, however, still utilize some form of instrumentation such as a camera for quantitative detection. We describe here a simple technique to render PAD measurements more quantitative and straightforward using the distance of colour development as a detection motif. The so-called distance-based detection enables PAD chemistries that are more portable and less resource intensive compared to classical approaches that rely on the use of peripheral equipment for quantitative measurement. We demonstrate the utility and broad applicability of this technique with measurements of glucose, nickel, and glutathione using three different detection chemistries: enzymatic reactions, metal complexation, and nanoparticle aggregation, respectively. The results show excellent quantitative agreement with certified standards in complex sample matrices. This work provides the first demonstration of distance-based PAD detection with broad application as a class of new, inexpensive sensor technologies designed for point-of-use applications.

Au–Ag bimetallic microfluidic, dumbbell-shaped, surface enhanced Raman scattering (SERS) sensors were fabricated on cellulose paper by screen printing. These printed sensors rely on a sample droplet injection zone, and a SERS detection zone at either end of the dumbbell motif, fabricated by printing silver nanoparticles (Ag NPs) and gold nanoparticles (Au NPs) successively with microscale precision. The microfluidic channel was patterned using an insulating ink to connect these two zones and form a hydrophobic circuit. Owing to capillary action of paper in the millimeter-sized channels, the sensor could enable self-filtering of fluids to remove suspended particles within wastewater without pumping. This sensor also allows sensitive SERS detection, due to advantageous combination of the strong surface enhancement of Ag NPs and excellent chemical stability of Au NPs. The SERS performance of the sensors was investigated by employing the probe rhodamine 6G, a limit of detection (LOD) of 1.1 × 10−13 M and an enhancement factor of 8.6 × 106 could be achieved. Moreover, the dumbbell-shaped bimetallic sensors exhibited good stability with SERS performance being maintained over 14 weeks in air, and high reproducibility with less than 15% variation in spot-to-spot SERS intensity. Using these dumbbell-shaped bimetallic sensors, substituted aromatic pollutants in wastewater samples could be quantitatively analyzed, which demonstrated their excellent capability for rapid trace pollutant detection in wastewater samples in the field without pre-separation.

In this study, a fast, low-cost, and facile spray method was proposed. This method deposits highly sensitive surface-enhanced Raman scattering (SERS) silver nanoparticles (AgNPs) on the paper-microfluidic scheme. The procedures for substrate preparation were studied including different strategies to synthesize AgNPs and the optimization of spray cycles. In addition, the morphologies of the different kinds of paper substrates were characterized by SEM and investigated by their SERS signals. The established method was found to be favorable for obtaining good sensitivity and reproducible results. The RSDs of Raman intensity of randomly analyzing 20 spots on the same paper or different filter papers depositing AgNPs are both below 15%. The SERS enhancement factor is approximately 2 × 10(7) . The whole fabrication is very rapid, robust, and does not require specific instruments. Furthermore, the total cost for 1000 pieces of chip is less than $20. These advantages demonstrated the potential for growing SERS applications in the area of environmental monitoring, food safety, and bioanalysis in the future.

This work reports a simple method to form and visualize individual polyelectrolyte-nanoparticle necklace-like structures on paper, which is applicable to other porous surfaces. In this work, one-dimensional necklaces of negatively charged gold nanoparticles (AuNPs) have been electrostatically assembled along the backbone of a cationic polyacrylamide (CPAM) chain adsorbed on paper. The process involves rapidly passing a dilute CPAM solution through filter paper, adsorbing the polyelectrolyte on the surface, followed by the immediate filtration of an AuNP suspension through the same filter paper. The nanoparticles used were negatively charged, citrate ion capped AuNPs with an average diameter of 20 nm. Scanning electron microscopy images of the dried paper sample showed that the AuNP necklaces were adsorbed in a perpendicular direction to that of the cellulose fibers and along the length of the CPAM molecules which were draped over the fibers. The effects of CPAM polymer concentration, charge density, and molecular weight on such assembly of AuNPs were studied. This technique enables the visualization of polyelectrolyte molecules and the formation of very well organized and reproducible polyelectrolyte-nanoparticle necklaces on a porous, three-dimensional substrate.

Paper-based devices have been broadly used for the point-of-care detection of dengue viral nucleic acids due to their simplicity, cost-effectiveness, and readily observable colorimetric readout. However, their moderate sensitivity and functionality have limited their applications. Despite the above-mentioned advantages, paper substrates are lacking in their ability to control fluid flow, in contrast to the flow control enabled by polymer substrates (e.g., agarose) with readily tunable pore size and porosity. Herein, taking the benefits from both materials, the authors propose a strategy to create a hybrid substrate by incorporating agarose into the test strip to achieve flow control for optimal biomolecule interactions. As compared to the unmodified test strip, this strategy allows sensitive detection of targets with an approximately tenfold signal improvement. Additionally, the authors showcase the potential of functionality improvement by creating multiple test zones for semi-quantification of targets, suggesting that the number of visible test zones is directly proportional to the target concentration. The authors further demonstrate the potential of their proposed strategy for clinical assessment by applying it to their prototype sample-to-result test strip to sensitively and semi-quantitatively detect dengue viral RNA from the clinical blood samples. This proposed strategy holds significant promise for detecting various targets for diverse future applications.

A colorimetric platform for influenza A virus detection was developed by using the high efficiency of enzymatic catalysis and the reduction of gold ions with hydrogen peroxide. Aptamer-functionalized magnetic microparticles were synthesized to capture the influenza A virus. This was followed by the binding of ConA-GOx-AuNPs to the H3N2 virus through the ConA-glycan interaction. The sandwich complex was subsequently dispersed in glucose solution to trigger an enzymatic reaction to produce hydrogen peroxide, which controlled the growth of gold nanoparticles and produced colored solutions. The determination of H3N2 concentration was realized by comparing the two differently colored gold nanoparticles. This method could detect the target virus as low as 11.16 μg ml−1. Furthermore, it opens new opportunities for sensitive and colorimetric detection of viruses and proteins.

A facile route that combines co-assembly and photothermal reduction was developed to synthesize free-standing, flexible FeF(3)-graphene papers. The papers contain well-dispersed FeF(3) nanoparticles and open diffusion channels in a porous, electrically conducting network of graphene sheets, and demonstrate promising applications as cathodes in high-energy density Li-ion batteries.

The silver-nanoparticle (AgNP) containing paper was successfully prepared. The AgNP is deposited by the in situ reduction of silver nitrate on the acrylamide grafted bagasse paper sheets in the presence of citrate molecules as stabilizing agent. In the present paper, grafting of acrylamide onto bagasse paper sheets using potassium persulfate was carried out under the influence of microwave radiations (MWR). The modified paper sheets were characterized by Fourier transform infrared spectroscopy (FTIR), UV-spectroscopy, and scanning electron microscopy (SEM). Antimicrobial activities of the prepared paper sheets were also investigated against G+ve bacterium Staphylococcus aureus, G-ve bacterium Pseudomomas aeruginosa, and yeast Candida albicans, which are model microorganisms for testing bactericidal properties. The AgNP containing paper sheets exhibited antibacterial activity.

This work investigates the effect of gold nanoparticle (AuNP) addition to paper substrate and examines the ability of these composite materials to amplify the surface enhanced Raman scattering (SERS) signal of a dye adsorbed. Paper has a three-dimensional (3D), porous, and heterogeneous morphology. The manner in which paper adsorbs the nanoparticles is crucial to its SERS properties, particularly with regards to aggregation. In this work, we sought to maintain the same degree of aggregation, while changing the concentration of nanoparticles deposited on paper. We achieved this by dipping paper into AuNP solutions of different, known concentration and found that the initial packing density of AuNPs in solutions was retained on paper with the same degree of aggregation. The surface coverage of AuNPs on paper was found to scale linearly to their concentration profile in solutions. The SERS performances of the AuNP-treated papers were evaluated with 4-aminothiophenol (4-ATP) as the Raman molecule, and their SERS intensities increased linearly with the AuNPs' concentration. Compared to AuNP-treated silicon, the Raman enhancement factor (EF) from paper was relatively higher due to a more uniform and greater degree of adsorption of AuNPs. The effect of the spatial distribution of AuNPs in their substrates on SERS activity was also investigated. In this experiment, the number of AuNPs was kept constant (a 1 μL droplet of AuNPs was deposited on all substrates), and the distribution profile of AuNPs was controlled by the nature of the substrate: paper, silicon, and hydrophobized paper. The AuNP droplet on paper showed the most reproducible and sensitive SERS signal. This highlighted the role of the z-distribution (through film) of AuNPs within the bulk of the paper, producing a 3D multilayer structure to allow inter- and intralayer plasmon coupling, and hence amplifying the SERS signal. The SERS performance of nanoparticle-functionalized paper can thus be optimized by controlling the 3D distribution of the metallic nanoparticles, and such control is critical if these systems are to be implemented as a low-cost and highly sensitive bioassay platform.

The development of a user-friendly sensing platform for the detection of Cu(2+) in water is necessary as there are wide concerns due to the substantial impact of Cu(2+) on human health, environmental monitoring, and so on. Motivated by this, we report a fluorescent silver nanoclusters (AgNCs)-based sensor for the detection of Cu(2+). These water-soluble AgNCs, as a new class of fluorescent probes, were synthesized by using azobenzene modified poly(acrylic acid) (MPAA) as templates. Their fluorescence can be quenched in the presence of Cu(2+), which enables the label-free detection of Cu(2+) in real water samples. Furthermore, such AgNCs can be integrated onto cellulose filter paper and used as fluorescent indicators for Cu(2+). The fluorescence quenching can be observed by the naked eye under UV light. It should be noted that this AgNCs-based paper assay performs successfully in barrelled drinking water and river water samples. Therefore, it opens up new avenues to the development of robust clusters-based sensing platforms.

A simple approach to the mass production of nanoporous gold electrode arrays on cellulose membranes for electrochemical sensing of oxygen using ionic liquid (IL) electrolytes was established. The approach, combining the inkjet printing of gold nanoparticle (GNP) patterns with the self-catalytic growth of these patterns into conducting layers, can fabricate hundreds of self-designed gold arrays on cellulose membranes within several hours using an inexpensive inkjet printer. The resulting paper-based gold electrode arrays (PGEAs) had several unique properties as thin-film sensor platforms, including good conductivity, excellent flexibility, high integration, and low cost. The porous nature of PGEAs also allowed the addition of electrolytes from the back cellulose membrane side and controllably produced large three-phase electrolyte/electrode/gas interfaces at the front electrode side. A novel paper-based solid-state electrochemical oxygen (O(2)) sensor was therefore developed using an IL electrolyte, 1-butyl-3-methylimidazolium hexafluorophosphate (BMIMPF(6)). The sensor looked like a piece of paper but possessed high sensitivity for O(2) in a linear range from 0.054 to 0.177 v/v %, along with a low detection limit of 0.0075% and a short response time of less than 10 s, foreseeing its promising applications in developing cost-effective and environment-friendly paper-based electrochemical gas sensors.

A silver nanoparticle colloid was prepared by a modified Tollens method using D-glucose as the reduction agent. The obtained nanoparticles were used for the modification of pine, linter and recycled cellulose fibers. Although the silver contents were relatively low (0.05-0.13 wt.%), the cellulose-sheets prepared from the modified fibers show improved mechanical and viscoelastic properties. The tensile index (strength) increased with up to 30% in comparison to the index of the sheets obtained from the untreated fibers. The influence of the nanoparticles on the viscoelastic properties of the cellulose sheets was investigated by dynamic mechanical analysis (DMA) in the temperature range from -120 to 20 degrees C and with a force frequency of 100Hz. A broad relaxation transition positioned at -80 degrees C was observed in the loss modulus spectrum of all the cellulose sheets, while the Ag-modified sheets exhibited higher storage moduli values in the whole temperature range. The antimicrobial activity tests show that the pine, silver and recycled cellulose fiber sheets with silver nanoparticles can be successfully employed to prevent the viability and growth of the common pathogens Staphylococcus aureus, Escherichia coli and Candida albicans. The successful in situ synthesis of platinum nanoparticles (PtNPs) on a microstructured paperlike matrix, comprising ceramic fibers as main framework and zinc oxide whiskers as selective support for the PtNPs, is reported. The as-prepared hybrid material (PtNPs@ZnO "paper") resembles ordinary paper products because it is flexible, lightweight, and easy to handle. In the catalytic reduction of nitrogen oxide (NO(x)) with propene for exhaust gas purification, the PtNPs@ZnO paper demonstrates a high catalytic performance at a low reaction temperature, with one-third the dosage of precious platinum compared to conventional platinum-loaded honeycomb catalysts. These results imply that the combination of easily synthesized PtNPs and a unique fiber-network microstructure can provide excellent performances, promoting the effective transport of heat and reactants to the active sites of the platinum nanocatalysts. Thus, PtNPs@ZnO materials with paperlike practical aspects are promising catalytic materials for efficient NO(x) gas purification.

The majority of bioassays utilize thermosensitive reagents (e.g., biomolecules) and laboratory conditions for analysis. The developing world, however, requires inexpensive, simple-to-perform tests that do not require refrigeration or access to highly trained technicians. To address this need, paper-based bioassays using gold nanoparticle (AuNP) colorimetric probes have been developed. In the two prototype DNase I and adenosine-sensing assays, blue (or black)-colored DNA-cross-linked AuNP aggregates were spotted on paper substrates. The addition of target DNase I (or adenosine) solution dissociated the gold aggregates into dispersed AuNPs, which generated an intense red color on paper within one minute. Both hydrophobic and (poly(vinyl alcohol)-coated) hydrophilic paper substrates were suitable for this biosensing platform; by contrast, uncoated hydrophilic paper caused "bleeding" and premature cessation of the assay due to surface drying. The assays are surprisingly thermally stable. During preparation, AuNP aggregate-coated papers can be dried at elevated temperatures (e.g., 90 degrees C) without significant loss of biosensing performance, which suggests the paper substrate protects AuNP aggregate probes from external nonspecific stimuli (e.g., heat). Moreover, the dried AuNP aggregate-coated papers can be stored for at least several weeks without loss of the biosensing function. The combination of paper substrates and AuNP colorimetric probes makes the final products inexpensive, low-volume, portable, disposable, and easy-to-use. We believe this simple, practical bioassay platform will be of interest for use in areas such as disease diagnostics, pathogen detection, and quality monitoring of food and water.

Disposable analytical devices for developing sensitive and label-free monitoring of cancerous cells would be attractive for cancer research. Here, paper-based electroanalytical devices based on impedance spectrometry were applied for the study of K562 cells and the toxic effect of anticancer drugs. The proposed device integrating gold nanorods modified ITO electrodes could provide a biocompatible surface for immobilization of living cells maintaining their bioactivity. The impedance results exhibited good correlation to the logarithmic value of cell numbers ranging from 7.5 × 10(2) to 3.9 × 10(6) cells mL(-1) with a detection limit of 500 cells mL(-1). Furthermore, this strategy was used to evaluate the cytotoxic effects of arsenic trioxide and cyclophosphamide. Results obtained by the impedimetric method correlate well with the conventional cell viability assay. Cells exposed to drugs exhibited a prominent reduction of impedance data, showing an inverse dose-dependent relationship. This simple, cost-effective and portable paper-based electrochemical analytical device could provide a new impedance platform for applications in monitoring cell behavior, pharmacological studies and toxicological analyses.

An efficient sequential, biological and photocatalytic treatment to reduce the pollutant levels in wastewater due to the bleaching process during paper production is reported. For a biological pre-treatment, 800 ml of non-sterilized effluent was inoculated with Trametes versicolor immobilized in polyurethane foam, with 25 g l(-1) glucose, 6.75 mM CuSO(4), and 0.22 mM MnSO(4) added, and cultured at 25 degrees C with an air flow of 800 ml min(-1) for 8d. The fungus did not inhibit growth of the heterotropic populations of the effluent. After 4d of culture, the chemical oxygen demand (COD) reduction and colour removal (CR) were 82% and 80%, respectively, with laccase (LAC) and manganese peroxidase (MnP) activities of 345 U l(-1) and 78 U l(-1), respectively. In this study, two different supramolecular recognition architectures for impedimetric detection of DNA hybridization have been formed on disposable paper-supported inkjet-printed gold electrodes. The gold electrodes were fabricated using a gold nanoparticle based ink. The first recognition architecture consists of subsequent layers of biotinylated self-assembly monolayer (SAM), streptavidin and biotinylated DNA probe. The other recognition architecture is constructed by immobilization of thiol-functionalized DNA probe (HS-DNA) and subsequent backfill with 11-mercapto-1-undecanol (MUOH) SAM. The binding capacity and selectivity of the recognition architectures were examined by surface plasmon resonance (SPR) measurements. SPR results showed that the HS-DNA/MUOH system had a higher binding capacity for the complementary DNA target. Electrochemical impedance spectroscopy (EIS) measurements showed that the hybridization can be detected with impedimetric spectroscopy in picomol range for both systems. EIS signal indicated a good selectivity for both recognition architectures, whereas SPR showed very high unspecific binding for the HS-DNA/MUOH system. The factors affecting the impedance signal were interpreted in terms of the complexity of the supramolecular architecture. The more complex architecture acts as a less ideal capacitive sensor and the impedance signal is dominated by the resistive elements.

In this work, microfluidic paper-based analytical device (μ-PAD) was applied in a photoelectrochemical (PEC) method and thus a truly low-cost, simple, portable, and disposable microfluidic PEC origami device (μ-PECOD) was demonstrated. The molecular imprinting technique was introduced into microfluidic paper-based analytical devices (μ-PADs) through electropolymerization of molecular imprinted polyaniline (MPANI) in a novel Au nanoparticle (AuNP)-modified paper working electrode (Au-PWE). This is fabricated through the growth of an AuNP layer on the surfaces of cellulose fibers in the PWE. Under visible light irradiation, MPANI can generate the photoelectric transition from the highest occupied molecular orbital (HOMO) to the lowest unoccupied molecular orbital (LUMO), delivering the excited electrons to the AuNPs, and then to the carbon working electrode. Simultaneously, it is believed that a positively charged hole of MPANI that took part in the oxidation process was consumed by ascorbic acid (AA) to promote the amplifying photocurrent response. On the basis of this novel MPANI-Au-PWE and the principle of origami, a microfluidic molecular imprinted polymer (MIP)-based photoelectrochemical analytical origami device (μ-MPECOD), comprised of an auxiliary tab and a sample tab, is developed for the detection of heptachlor in the linear range from 0.03 nmol L(-1) to 10.0 nmol L(-1) with a low detection limit of 8.0 pmol L(-1). The selectivity, reproducibility, and stability of this μ-MPECOD are investigated. This μ-MPECOD would provide a new platform for high-throughput, sensitive, specific, and multiplex assay in public health, environmental monitoring, and the developing world.

Silver nanoparticles (AgNPs) decorated filter papers combining solid-phase extraction (SPE) with surface enhanced Raman spectroscopy (SERS) achieved rapid collection of analytes and in situ detection. The AgNPs were fabricated by cellulose self-sacrificing reduction. Aqueous Ag(NH3)2OH was reduced by hydroxyl groups in cellulose under alkaline conditions. The AgNPs were highly uniform and firmly adhered to the microfibers. Reaction conditions were optimized by the probe molecule p-aminothiophenol (PATP) to attain the best Raman enhancement. Methylene blue trihydrate (MB) and 6-thioguanine (6-TG) were detected by flow-through method. The results exhibited outstanding SERS effect and an obvious improvement in detection limit was observed compared to common immersion methods. SERS detection was quantitative as the log-log plot of Raman intensity against MB and 6-TG concentrations showed a linear relationship. The SERS-active paper is degradable because it could be burned after analyte detection.

A novel 3D microfluidic paper-based immunodevice, integrated with blood plasma separation from whole blood samples, automation of rinse steps, and multiplexed CL detections, was developed for the first time based on the principle of origami (denoted as origami-based device). This 3D origami-based device, comprised of one test pad surrounded by four folding tabs, could be patterned and fabricated by wax-printing on paper in bulk. In this work, a sandwich-type chemiluminescence (CL) immunoassay was introduced into this 3D origami-based immunodevice, which could separate the operational procedures into several steps including (i) folding pads above/below and (ii) addition of reagent/buffer under a specific sequence. The CL behavior, blood plasma separation, washing protocol, and incubation time were investigated in this work. The developed 3D origami-based CL immunodevice, combined with a typical luminuol-H(2)O(2) CL system and catalyzed by Ag nanoparticles, showed excellent analytical performance for the simultaneous detection of four tumor markers. The whole blood samples were assayed and the results obtained were in agreement with the reference values from the parallel single-analyte test. This paper-based microfluidic origami CL detection system provides a new strategy for a low-cost, sensitive, simultaneous multiplex immunoassay and point-of-care diagnostics.

A method for patterning narrow lines of biomolecules onto nitrocellulose membranes using laboratory syringe pumps is described. One syringe pump is used to drive the biomolecule solution through a needle, while another modified syringe pump acts as a one-dimensional translation stage, moving the needle across the membrane much like a pen. This method consumes very small volumes of reagents, and is a viable option for laboratory-scale fabrication and prototyping of point-of-care rapid diagnostic test strips.

The quantum spin properties of nitrogen-vacancy defects in diamond enable diverse applications in quantum computing and communications1. However, fluorescent nanodiamonds also have attractive properties for in vitro biosensing, including brightness2, low cost3 and selective manipulation of their emission4. Nanoparticle-based biosensors are essential for the early detection of disease, but they often lack the required sensitivity. Here we investigate fluorescent nanodiamonds as an ultrasensitive label for in vitro diagnostics, using a microwave field to modulate emission intensity5 and frequency-domain analysis6 to separate the signal from background autofluorescence7, which typically limits sensitivity. Focusing on the widely used, low-cost lateral flow format as an exemplar, we achieve a detection limit of 8.2 × 10−19 molar for a biotin–avidin model, 105 times more sensitive than that obtained using gold nanoparticles. Single-copy detection of HIV-1 RNA can be achieved with the addition of a 10-minute isothermal amplification step, and is further demonstrated using a clinical plasma sample with an extraction step. This ultrasensitive quantum diagnostics platform is applicable to numerous diagnostic test formats and diseases, and has the potential to transform early diagnosis of disease for the benefit of patients and populations. Lateral-flow in vitro diagnostic assays based on fluorescent nanodiamonds, in which microwave-based spin manipulation is used to increase sensitivity, are demonstrated using the biotin–avidin model and by the single-copy detection of HIV-1 RNA.
