The phytochrome superfamily comprises three groups of photoreceptors sharing a conserved GAF (cGMP-specific phosphodiesterases, cyanobacterial adenylate cyclases, and formate hydrogen lyase transcription activator FhlA) domain that uses a covalently attached linear tetrapyrrole (bilin) chromophore to sense light. Knotted red/far-red phytochromes are widespread in both bacteria and eukaryotes, but cyanobacteria also contain knotless red/far-red phytochromes and cyanobacteriochromes (CBCRs). Unlike typical phytochromes, CBCRs require only the GAF domain for bilin binding, chromophore ligation, and full, reversible photoconversion. CBCRs can sense a wide range of wavelengths (ca. 330-750 nm) and can regulate phototaxis, second messenger metabolism, and optimization of the cyanobacterial light-harvesting apparatus. However, the origins of CBCRs are not well understood: we do not know when or why CBCRs evolved, or what selective advantages led to retention of early CBCRs in cyanobacterial genomes. In the current work, we use the increasing availability of genomes and metagenome-assembled-genomes from early-branching cyanobacteria to explore the origins of CBCRs. We reaffirm the earliest branches in CBCR evolution. We also show that early-branching cyanobacteria contain late-branching CBCRs, implicating early appearance of CBCRs during cyanobacterial evolution. Moreover, we show that early-branching CBCRs behave as integrators of light and pH, providing a potential unique function for early CBCRs that led to their retention and subsequent diversification. Our results thus provide new insight into the origins of these diverse cyanobacterial photoreceptors.

The dissemination of antibiotic resistance in Escherichia coli poses a significant threat to public health worldwide. This review provides a comprehensive update on the diverse mechanisms employed by E. coli in developing resistance to antibiotics. We primarily focus on pathotypes of E. coli (e.g., uropathogenic E. coli) and investigate the genetic determinants and molecular pathways that confer resistance, shedding light on both well-characterized and recently discovered mechanisms. The most prevalent mechanism continues to be the acquisition of resistance genes through horizontal gene transfer, facilitated by mobile genetic elements such as plasmids and transposons. We discuss the role of extended-spectrum β-lactamases (ESBLs) and carbapenemases in conferring resistance to β-lactam antibiotics, which remain vital in clinical practice. The review covers the key resistant mechanisms, including: 1) Efflux pumps and porin mutations that mediate resistance to a broad spectrum of antibiotics, including fluoroquinolones and aminoglycosides; 2) adaptive strategies employed by E. coli, including biofilm formation, persister cell formation, and the activation of stress response systems, to withstand antibiotic pressure; and 3) the role of regulatory systems in coordinating resistance mechanisms, providing insights into potential targets for therapeutic interventions. Understanding the intricate network of antibiotic resistance mechanisms in E. coli is crucial for the development of effective strategies to combat this growing public health crisis. By clarifying these mechanisms, we aim to pave the way for the design of innovative therapeutic approaches and the implementation of prudent antibiotic stewardship practices to preserve the efficacy of current antibiotics and ensure a sustainable future for healthcare.

Antimicrobial resistant lower respiratory tract infections are an increasing public health threat and an important cause of global mortality. The lung microbiome can influence susceptibility of respiratory tract infections and represents an important reservoir for exchange of antimicrobial resistance genes. Studies of the gut microbiome have found an association between age and increasing antimicrobial resistance gene burden, however, corollary studies in the lung microbiome remain absent. We performed an observational study of children and adults with acute respiratory failure admitted to the intensive care unit. From tracheal aspirate RNA sequencing data, we evaluated age-related differences in detectable antimicrobial resistance gene expression in the lung microbiome. Using a multivariable logistic regression model, we find that detection of antimicrobial resistance gene expression was significantly higher in adults compared with children after adjusting for demographic and clinical characteristics. This association remained significant after additionally adjusting for lung bacterial microbiome characteristics, and when modeling age as a continuous variable. The proportion of adults expressing beta-lactam, aminoglycoside, and tetracycline antimicrobial resistance genes was higher compared to children. Together, these findings shape our understanding of the lung resistome in critically ill patients across the lifespan, which may have implications for clinical management and global public health.

Legacy effects after the removal of invasive plants present significant challenges to restoration. The pivotal role of soil microbial communities in shaping these legacy effects is increasingly recognized, yet there is a lack of effective methods to mitigate altered microbial communities. In Point Reyes National Seashore (California), although the invaded European beachgrass (Ammophila arenaria [L.] Link) was successfully controlled by herbicide treatment, beachgrass litter remained undecomposed for over 5 years, leaving pronounced legacy effects on soil organic matter and microbial community composition. We hypothesized that soil inoculation from uninvaded dune scrub can accelerate the decomposition of beachgrass litter in herbicide‐treated sites by restoring the soil microbial communities and the abundance of microbial decomposers. Three litterbags containing European beachgrass litter or litter from two common shrubs at dune scrub were deployed into each plot to assess the impact of soil inoculation on litter decomposition rates. Our results revealed that soil inoculation, regardless of the inoculation level, did not accelerate the decomposition of European beachgrass. Only the decomposition of bush lupine litter, which had the highest litter quality among three types of litter, was accelerated at the highest inoculation level (approximately 11,880 g/m2). Additionally, soil inoculation increased the richness and compositional homogeneity of soil microbial communities, along with the relative abundances of wood saprotrophic fungi, soil saprotrophic fungi, and lichenized fungi. Although these findings demonstrate the potential of soil inoculation, the cost‐effectiveness of soil inoculation limits its feasibility in accelerating the delayed decomposition of European beachgrass litter in Point Reyes. Textile industries utilize a variety of dyes and chemicals, resulting in wastewater that contains numerous hazardous components. The release of these dyes into aquatic systems poses a serious environmental and harms human health due to their persistence, recalcitrance, and nonbiodegradability. To address this, microbial bioremediation and nano‐photocatalysts are commonly employed for the effective removal of dyes and toxic compounds from textile effluents. This review explores the ecofriendly and efficient use of microbes, such as fungi, yeast, bacteria, and algae, in dye treatment, emphasizing their role in decolorization and degradation. Additionally, microbial remediation is highlighted as a clean, effective, and safe technology for detoxifying azo dyes in wastewater. The review also compares microbial and nanoparticle methods for dye removal from textile wastewater, examining the environmental impact of each approach. Background: Short courses of antibiotics (7-10 days) are effective for uncomplicated gram-negative bloodstream infections (GN-BSI). However, prior studies have been limited to small cohorts of critically ill patients. Objective: The objective of this study was to evaluate the safety and efficacy of short courses of therapy compared with longer courses in patients admitted to the intensive care unit (ICU) with GN-BSI. Methods: Propensity-matched, retrospective cohort study of critically ill patients with GN-BSI. The primary outcome was a composite of 30-day mortality or 60-day relapse. Secondary endpoints were components of the composite, 30-day relapse, cure with or without adverse drug events (ADE), and ADEs. Regression analysis was performed to identify factors predictive of the composite outcome. Results: 225 patients were included in the propensity analysis, 145 in the long cohort and 80 in the short cohort. The primary outcome occurred in 3.8% of patients in the short group and 9.0% of patients in the long group (P = 0.24). There was no difference in 30-day mortality (3.8% vs 5.5%, P = 0.79), 60-day relapse (0% vs 3.4%, P = 0.23), or 30-day readmission (20% vs 22.8%, P = 0.76). ADEs were more common in the long group (47.2% vs 34.1%, OR 1.7, 95% CI 1.04-2.9), primarily attributable to diarrhea. Conclusion and Relevance: In critically ill patients with GN-BSI, there were no efficacy outcome differences in patients treated with a short course of antibiotics compared with longer. However, patients in the short group were less likely to experience ADE. These findings suggest that short courses of antibiotics are effective for GN-BSI in critically ill patients. Fe-oxide deposits of the Lahn-Dill-type in the eastern Rhenish Massif comprise haematite and quartz with minor siderite, magnetite, and calcite. The deposits are located in the hanging wall of thick volcaniclastic rock sequences and mark the Middle to Late Devonian boundary. Varying ore types with accompanying footwall rocks were sampled from two formerly important ore deposits, the Fortuna mine (Lahn syncline) and the Briloner Eisenberg mine (East Sauerland anticline), in order to elucidate the interplay of processes leading to ore formation. Deposit geology, petrography, and whole-rock geochemistry suggest that the ores formed by iron mobilisation from deeply altered footwall volcaniclastic rocks, subsequent venting of a modified H2O-CO2-Fe-rich and H2S-poor fluid, and precipitation on the seafloor (sedimentary-type), or locally by metasomatic replacement of wall rocks (replacement-type). Petrographic analysis to the sub-micron scale revealed that the sedimentary-type ores most likely formed from a Fe-Si-rich gel and accompanying maturation. Early gel textures include the presence of spherules, aggregates, tubes, and filamentous stalks consisting of nanocrystalline haematite dispersed in a matrix of microcrystalline quartz. Local diagenetic Fe3+ reduction within the gel is indicated by siderite replacement of haematite. Replacement-type ores formed due to a two-step process including coprecipitation of (precursor) haematite and carbonates and subsequent metasomatic replacement by haematite. These ore-forming processes took place during a time when several restricted shallow marine basins in the north-eastern Rheic Ocean were influenced by extensive volcanism and associated hydrothermal fluid flux. Microplastics (MPs) are generated from diverse categories of plastic debris disposed in open. Its entry into the terrestrial ecosystem could not only impact soil physico-chemical attributes but also endanger the lives of biotas including the earthworms which play significant role in the decomposition of organics and sustaining the nutrient pool. There have been consistent efforts by various workers across the globe to study the impact of MPs on the terrestrial environment and exploring potential mitigation strategies to minimize contamination levels. This review presents a bibliometric analysis of scientific publications on impact of MPs on the earthworms from 2017 to 2023. The primary objective is to discern trends among authors, institutions, and countries contributing to research on MPs, particularly concerning their interaction with earthworms. The analysis reveals a steady escalation in the number of publications up to 2022. The VOS viewer software was utilized for data visualization and cluster analysis, unveiling three clusters highlighting keyword groups associated with "microplastics", "soil", and "earthworms". Eriocheir sinensis is the main aquaculture species in China. With the continuous expansion of the aquaculture scale, the demand for E. sinensis seedlings was also increased. The water used in breeding has well-nourished and its discharge into the sea posed significant risks. This study sampled the wastewater discharge points of the E. sinensis seedlings in Sheyang County, Jiangsu Province, and the areas far from the discharge points that were not affected in March and May 2023, respectively. A large number of antibiotic resistance genes (ARGs) were found in the sediment of the wastewater discharge area, and the highest ARG was sulfonamide ARG-sul1 using qPCR analysis, while ARGs were almost undetectable in the areas not affected by wastewater discharge. The 16S rRNA sequence analysis results showed that the main bacterial phyla at the wastewater discharge point were Bacteroidetes, Proteobacteria, and Thermodesulfobacteria. In the control point, the main bacterial phyla were Proteobacteria, Chlorobacterium, and Thermodesulfobacteria. There were significant differences in the composition of microbial communities between the two points, and the samples at the wastewater discharge point were more clustered and had higher similarity. The correlation network and redundancy analysis indicated that the phyla Bacteroidetes, Firmicutes, and Proteobacteria at the wastewater discharge points were positively correlated with most ARGs. The wastewater discharge had no effect on heavy metals from the two points. This study sets a foundation for future research by identifying key microbial taxa as potential ARG carriers and examining the interactions between microbial communities, ARGs, and heavy metals. The bacterial virulence and resistance to most antibiotics increased in the last decades and the public interest in traditional herbs has developed due to their confirmed medical properties and limited or no side effects. In the present study, the antibacterial activity of curcumin and its nanoparticles was investigated against Staph. aureus, E. coli and Pseudomonas aeroginosa. The prepared curcumin nanoparticles had the Z-average diameter of 103±32.12 nm and polydispersity index (PDI) of 0.152. In examining the freshly prepared nanoparticles by Transmission Electron Microscope (TEM) the spherical shape was confirmed, and the size was 47.83±5.45 nm and the flow of active functional groups was clarified by Fourier-transform infrared spectroscopy (FTIR). The minimum inhibitory concentration (MIC) of curcumin and its nanoparticles was 3.13% and 1.65%, on the examined bacteria, respectively. When curcumin and its nanoparticles were prepared in Tallaga cheese, a complete inhibition of the examined bacteria was recorded on the 4th day in samples supplemented with curcumin nanoparticles. However, the examined bacteria in the curcumin-free cheese samples reduced in count, but not completely inhibited. Background In the last decade, veterinary antimicrobial usage (AMU) and antimicrobial resistance (AMR) among indicator bacteria in livestock have decreased substantially in the Netherlands. The extent to which this decrease has affected AMR levels among human infections remains unclear. Objectives To assess the association between AMU in livestock and AMR in Escherichia coli isolates from human urinary tract infections (UTIs). Methods Data on AMR and AMU between 2009 and 2020 from Dutch national surveillance programmes for humans and livestock were used. Associations between AMU in four major livestock sectors and AMR in humans were assessed for 10 antimicrobial classes and the ESBL resistance profile, using logistic regression analysis. Associations between AMU and AMR in livestock, between AMR in livestock and in humans, and between AMU and AMR in humans were also assessed. Results Statistical significance was reached for 16/31 of the tested associations between AMU in livestock and AMR in human E. coli UTIs. Of the significant associations, 11 were positive (OR 1.01–1.24), whereas 5 were negative (OR 0.96–0.99). All associations between human AMU and AMR in E. coli isolates from UTIs were positive and statistically significant. Weak but significant positive correlations were also observed between livestock AMR and human AMR. Conclusions Although several significant associations between AMU in livestock and AMR in human UTIs caused by E. coli were observed, the associations between AMU and AMR were generally stronger within the human and animal populations. Objectives Klebsiella pneumoniae is a significant pathogen with increasing resistance and high mortality rates. Conventional antibiotic susceptibility testing methods are time-consuming. Next-generation sequencing has shown promise for predicting antimicrobial resistance (AMR). This study aims to develop prediction models using whole-genome sequencing data and assess their feasibility with metagenomic next-generation sequencing data from clinical samples. Methods On the basis of 4170 K. pneumoniae genomes, the main genetic characteristics associated with AMR were identified using a LASSO regression model. Consequently, the prediction model was established, validated and optimized using clinical isolate read simulation sequences. To evaluate the efficacy of the model, clinical specimens were collected. Results Four predictive models for amikacin, ciprofloxacin, levofloxacin, and piperacillin/tazobactam, initially had positive predictive values (PPVs) of 92%, 98%, 99%, 94%, respectively, when they were originally constructed. When applied to clinical specimens, their PPVs were 96%, 96%, 95%, and 100%, respectively. Meanwhile, there were negative predictive values (NPVs) of 100% for ciprofloxacin and levofloxacin, and 'not applicable' (NA) for amikacin and piperacillin/tazobactam. Our method achieved antibacterial phenotype classification accuracy rates of 95.92% for amikacin, 96.15% for ciprofloxacin, 95.31% for levofloxacin and 100% for piperacillin/tazobactam. The sequence-based prediction antibiotic susceptibility testing (AST) reported results in an average time of 19.5 h, compared with the 67.9 h needed for culture-based AST, resulting in a significant reduction of 48.4 h. Conclusions These preliminary results demonstrated that the performance of prediction model for a clinically significant antimicrobial–species pair was comparable to that of phenotypic methods, thereby encouraging the expansion of sequence-based susceptibility prediction and its clinical validation and application. Background Methicillin-resistant Staphylococcus haemolyticus (MRSH) is an important pathogenic agent of bovine mastitis. Among the prominent clone lineages in dairy cows are MRSH sequence types ST3 and ST42. Little information is available on the complete characterization of SCC mec elements in MRSH. Objective In this study, two clinical isolates of MRSH ST3 and ST42 from bovine mastitis milk were selected, and their nontypable SCC mec structures were compared. Methods Two MRSH strains, MRSH-ST3 strain M62.3 and MRSH-ST42 strain M81.1, were identified from bovine mastitis milk in Thailand in 2022. Minimum inhibitory concentration was used to screen for antimicrobial resistance susceptibility. Oxford Nanopore Technologies and Illumina sequencing were performed in combination to complete the genome. Their gene organization and structure of SCC mec types were analysed and compared with the whole sequences of other strains in the same sequence types. Results Both MRSH-ST3 strain M62.3 and MRSH-ST42 strain M81.1 possessed the class C1 mec complex but lacked the ccr gene complex. Notably, MRSH-ST42 strain M81.1 contained a novel variant of C1 mec complex, which consisted of IS 431 - mecA -IS Sha1 - paaZ - upgQ -IS 431 , with IS 431 organized in the same orientation. Apart from class C1 mec and the heavy metal-resistant cluster, the gene composition and order of the SCC mec element varied. In ST3, variations in the SCC mec type, gene content and organization were observed. Conclusions The distinct evolution of the MRSH lineage was indicated by the various SCC mec elements. The insertion of IS Sha1 resulted in a unique variant of class C1 mec complex that demonstrated the important role of the insertion sequence in SCC mec diversification. [ABSTRACT FROM AUTHOR]

Hospitals at home are increasingly offering outpatient parenteral antimicrobial therapy (OPAT) in an attempt to reduce costly inpatient care, but these settings favour broad-spectrum antibiotics that require less frequent dosing than penicillin. Benzyl penicillin could be delivered via continuous infusion pumps (eCIPs), but studies on their safety and efficacy in OPAT are scarce, and it remains unclear how much the availability of eCIPs increases penicillin use in real-life settings. We examined 462 electronic healthcare records of erysipelas patients treated between January 2018 and January 2022 in a large Finnish OPAT clinic. Average marginal effects from logistic models were estimated to assess how the introduction of eCIPs in December 2020 affected penicillin use and to compare clinical outcomes between patients with and without eCIPs. Introduction of eCIPs increased the predicted probability of penicillin treatment by 36.0 percentage points (95% confidence interval 25.5–46.5). During eCIP implementation, patients who received an eCIP had 73.1 (58.0–88.2) percentage points higher probability than patients without an eCIP to receive penicillin treatment. They also had about 20 percentage points higher probability to be cured at the time of discharge and 3 months after it. Patient and nurse satisfaction regarding eCIPs was very high. Benzyl penicillin eCIP treatment is effective and safe, and substantially increases the use of penicillin instead of broad-spectrum antibiotics. To reduce the risk of antimicrobial resistance, eCIPs could increasingly be promoted for use in OPAT clinics, and there should be adequate education and support in their implementation. Background National action plans (NAPs) are key instruments for governing antimicrobial resistance (AMR). In Europe, we can now observe many countries updating their NAPs which raise two key research questions; what substantial modifications are states opting for, and how do they wish to address challenges related to AMR governance in a comparative perspective? Methods Building on a previous analytical classification, we address these two questions by examining data of updated versions of NAPs in 13 European Union member states covering seventeen elements related to AMR governance. Results Our results substantiate the large variation with regard to both substantive issues and governance-related matters. Most tellingly, they highlight the growing importance of the One Health approach in updated versions of NAPs. Our analysis also shows that while substantive issues remain important, One Health and the coordination and collaboration issues it entails are becoming more salient in the second or third generation of NAPs. Conclusions Updated NAPs suggest that EU member states are becoming increasingly knowledgeable on the causes and consequences of AMR and how it needs to be addressed. The enhanced level of knowledge also leads these countries to address the next set of issues and challenges; to improve domestic and international coordination and collaboration. Background Antimicrobial resistance (AMR) is a significant global public health threat and key priority for the public health, especially health protection, workforce to lead by example. There is a paucity of learning resources on this for public health professionals (PHPs) in the UK. This project aimed to develop and disseminate a tailored interactive learning resource and evaluate impact on self-reported intention to change behaviour. Methods Learning objectives were agreed, content developed by the multi-disciplinary team and piloted by PHPs in 2022 alongside a matched pre- and post-implementation evaluation survey. Questions were mapped to the capability–opportunity–motivation–behaviour change model. Before and after responses were calculated to compare change in self-reported knowledge, understanding and behaviour. Significance of change in binary responses was estimated. Results The resource was delivered using an interactive, user-friendly and cost-free internal platform. Thirty-one PHPs completed the pilot e-learning and survey. Perceived and actual knowledge increased in parallel. Actual knowledge on AMR burden increased from 6.45 to 35.48% (P  = 0.004). Self-reported confidence to explain AMR to others improved by 0.71 (95% CI; 0.38–1.04: P  = 0.0001) Likert points on a five-point scale. Motivation to advocate for antimicrobial stewardship (AMS) in day-to-day work, improved by 0.71 (95% CI; 0.34–1.08: P  < 0.00001) Likert points. Case scenarios were well-received as an effective way to apply theory to practice. Conclusion Ensuring a well-informed and confident public health workforce is vital for reducing the AMR threat and advocating for AMS with the public and partner organizations. BACKGROUND RESULTS CONCLUSION Nannochloropsis algae contain approximately 20% polyunsaturated fatty acids (PUFA) and hold significant potential for high‐quality eicosapentaenoic acid (EPA) food industrialization. However, EPA in Nannochloropsis sp. is prone to oxidation, and microbial growth is a critical factor affecting the shelf life of fresh food. Storage composition and temperature are primary factors influencing microbial growth, yet these aspects are not fully understood. This study investigates the effects of temperature and encapsulation on EPA content in nano‐products over time. Nano‐powder and nanobeads derived from Nannochloropsis sp. served as raw materials. Additionally, changes in aerobic plate counts and coliform groups were monitored.The results indicated that nanobeads, due to their more complex processing and less mature packaging, were more susceptible to coliform contamination compared to nano‐powder. In terms of EPA stability, nanobeads exhibited a longer storage life than nano‐powder. The oxidation rate of both nano‐powder and nanobeads was faster at 37 °C than at 25 °C.These findings can inform general shelf life estimation, rapid detection of total lipid content in nano‐products and macro extraction of nano‐oil. The glycolipids cellobiose lipids (CL) and mannosylerythritol lipids (MEL) are biosurfactants mainly synthesized by microorganisms of the Ustilaginaceae family. They have a large structural diversity, varying in their sugar moieties and the attached fatty acids, resulting in a prospectively broad range of applications. This literature review provides a detailed overview of known microbial producers of CL and MEL, and their respective metabolic pathways that result in different molecular structures. Further, current advances in the aerobic fermentative synthesis of the glycolipids and their purification methods are illustrated. All influencing factors identified to date with regard to the fermentation are highlighted in detail: For CL synthesis usually hydrophilic carbon sources are used as substrate, whereas hydrophobic carbon sources are usually metabolized to MEL. Nitrogen limitation was described as a major trigger for glycolipid synthesis and an acidic pH range was favored for increased CL production. An overview of applied fermentation parameters in recent publications (e.g., substrate‐concentrations, feeding approaches) demonstrates the future potential of CL and MEL production optimization. Foaming during fermentation is either combated or exploited by foam fractionation as the first purification step. The current purification processes focus on solvent extractions and chromatography in the laboratory scale and a need for development was identified for future scale‐up. This study aimed to compare calcium oxide (CaO) and calcium chloride (CaCl2) as calcium sources for coconut fatty acid distillate (CFAD) calcium soap (Ca-soap) production and to evaluate the supplementation ratios of unprotected and protected CFAD in dairy rations to optimize rumen function. This research included two steps: (1) assessing the protection strength of Ca-soap made with CaO and CaCl2 at mole ratios of Ca to CFAD of 1, 1.5, 2, and 2.5; (2) evaluating CFAD supplementation in an in vitro dairy ration study using a 5 × 4 randomized factorial block design. Factor A compared unprotected and protected CFAD ratios of A1 = 100:0, A2 = 75:25, A3 = 50:50, A4 = 25:75, and A5 = 0:100, and factor B compared supplementation levels of B1 = 0%, B2 = 1%, B3 = 2%, and B4 = 3%. CaCl2 at a 2.5-mole ratio to CFAD produced the lowest acid value and the carboxylic acid (C=O) chemical bond. Complete protection (0:100) exhibited the highest densities of Bacteroides and nutrient digestibility (p < 0.05) without significantly affecting rumen fermentability (p > 0.05). Higher CFAD levels significantly reduced methanogens and protozoa (p < 0.05) without significantly affecting estimated methane production. Background Estimates of the prevalence of antimicrobial resistance (AMR) underpin effective antimicrobial stewardship, infection prevention and control, and optimal deployment of antimicrobial agents. Typically, the prevalence of AMR is determined from real-world antimicrobial susceptibility data that are time delimited, sparse, and often biased, potentially resulting in harmful and wasteful decision-making. Frequentist methods are resource intensive because they rely on large datasets. Objectives To determine whether a Bayesian approach could present a more reliable and more resource-efficient way to estimate population prevalence of AMR than traditional frequentist methods. Methods Retrospectively collected, open-source, real-world pseudonymized healthcare data were used to develop a Bayesian approach for estimating the prevalence of AMR by combination with prior AMR information from a contextualized review of literature. Iterative random sampling and cross-validation were used to assess the predictive accuracy and potential resource efficiency of the Bayesian approach compared with a standard frequentist approach. Results Bayesian estimation of AMR prevalence made fewer extreme estimation errors than a frequentist estimation approach [ n  = 74 (6.4%) versus n  = 136 (11.8%)] and required fewer observed antimicrobial susceptibility results per pathogen on average [mean = 28.8 (SD = 22.1) versus mean = 34.4 (SD = 30.1)] to avoid any extreme estimation errors in 50 iterations of the cross-validation. The Bayesian approach was maximally effective and efficient for drug–pathogen combinations where the actual prevalence of resistance was not close to 0% or 100%. Conclusions Bayesian estimation of the prevalence of AMR could provide a simple, resource-efficient approach to better inform population infection management where uncertainty about AMR prevalence is high. Objectives Carbapenem-resistant Klebsiella pneumoniae (CRKP) is a major nosocomial infectious pathogen with rapidly increasing prevalence. The genomic epidemiological characteristics of CRKP nationwide, especially the evolving trends within the predominant clones, should be evaluated clearly. Methods We collected 3415 K. pneumoniae strains from 28 hospitals across China. Antimicrobial susceptibility testing and WGS were performed. Subsequent genomic analyses, including sequence typing, K-locus (KL) identification, antimicrobial resistance gene screening, and virulence score assessment were performed. The phylogenetic relationship of clonal group 11 was determined based on core-genome analysis, and the presence of the pLVPK-like virulence plasmid in ST11 isolates was confirmed using plasmid core-gene analysis. Additionally, the trends of the ST11 lineage with different KL types on a global scale were investigated using Beast2. Results Of the K. pneumoniae strains, 708 were identified as CRKP isolates (20.7%), of which 97.7% were MDR. ST11 was the predominant clone, and KPC-2 was the prevalent carbapenemase in China, although the prevalence of specific clones and carbapenemases varied by geographic region. Among ST11 isolates, KL47 and KL64 were the predominant KL types, and KL64 gradually replaced KL47, with a higher percentage of KL64 isolates harbouring the pLVPK-like plasmid. Global genome data showed a significant increase in the effective population size of KL64 over the last 5 years. Conclusions The prevalence of CRKP was very high in certain regions in China. Objectives The novel dual-target triazaacenaphthylene, gepotidacin, recently showed promising results in its Phase III randomized controlled trial for the treatment of gonorrhoea. We investigated alterations in the gepotidacin GyrA and ParC targets in gonococci by in silico mining of publicly available global genomes (n  = 33 213) and determined gepotidacin MICs in isolates with GyrA A92 alterations combined with other GyrA and/or ParC alterations. Methods We examined gonococcal gyrA and parC alleles available at the European Nucleotide Archive. MICs were determined using the agar dilution method (gepotidacin) or Etest (four antimicrobials). Models of DNA gyrase and topoisomerase IV were obtained from AlphaFold and used to model gepotidacin in the binding site. Results GyrA A92 alterations were identified in 0.24% of genomes: GyrA A92P/S/V + S91F + D95Y/A/N (0.208%), A92P + S91F (0.024%) and A92P (0.003%), but no A92T (previously associated with gepotidacin resistance) was found. ParC D86 alterations were found in 10.6% of genomes: ParC D86N/G (10.5%), D86N + S87I (0.051%), D86N + S88P (0.012%) and D86G + E91G (0.003%). One isolate had GyrA A92P + ParC D86N alterations, but remained susceptible to gepotidacin (MIC = 0.125 mg/L). No GyrA plus ParC alterations resulted in a gepotidacin MIC > 4 mg/L. Modelling of gepotidacin binding to GyrA A92/A92T/A92P suggested that gepotidacin resistance due to GyrA A92T might be linked to the formation of a new polar contact with DNA. Conclusions In silico mining of 33 213 global gonococcal genomes (isolates from 1928 to 2023) showed that A92 is highly conserved in GyrA, while alterations in D86 of ParC are common. Objectives Antimicrobials can select for antimicrobial-resistant bacteria. After treatment the active compound is excreted through urine and faeces. As some antimicrobials are chemically stable, recirculation of subinhibitory concentrations of antimicrobials may occur due to coprophagic behaviour of animals such as chickens. Methods The persistence of three antimicrobials over time and their potential effects on antimicrobial resistance were determined in four groups of broilers. Groups were left untreated (control) or were treated with amoxicillin (unstable), doxycycline or enrofloxacin (stable). Antimicrobials were extracted from the faecal samples and were measured by LC-MS/MS. We determined the resistome genotypically using shotgun metagenomics and phenotypically by using Escherichia coli as indicator microorganism. Results Up to 37 days after treatment, doxycycline and enrofloxacin had concentrations in faeces equal to or higher than the minimal selective concentration (MSC), in contrast to the amoxicillin treatment. The amoxicillin treatment showed a significant difference (P  ≤ 0.01 and P  ≤ 0.0001) in the genotypic resistance only directly after treatment. On the other hand, the doxycycline treatment showed approximately 52% increase in phenotypic resistance and a significant difference (P  ≤ 0.05 and P  ≤ 0.0001) in genotypic resistance throughout the trial. Furthermore, enrofloxacin treatment resulted in a complete non-WT E. coli population but the quantity of resistance genes was similar to the control group, likely because resistance is mediated by point mutations. Conclusions Based on our findings, we suggest that persistence of antimicrobials should be taken into consideration in the assessment of priority classification of antimicrobials in livestock. Objectives Escherichia coli can cause infections in the urinary tract and in normally sterile body sites leading to invasive E. coli disease (IED), including bacteraemia and sepsis, with older populations at increased risk. We aimed to estimate the theoretical coverage rate by the ExPEC4V and 9V vaccine candidates. In addition, we aimed at better understanding the diversity of E. coli isolates, including their genetic and phenotypic antimicrobial resistance (AMR), sequence types (STs), O-serotypes and the bacterial population structure. Methods Blood and urine culture E. coli isolates (n  = 304) were collected from hospitalized patients ≥60 years (n = 238) with IED during a multicentric, observational study across three continents. All isolates were tested for antimicrobial susceptibility, O-serotyped, whole-genome sequenced and bioinformatically analysed. Results A large diversity of STs and of O-serotypes were identified across all centres, with O25b-ST131, O6-ST73 and O1-ST95 being the most prevalent types. A total of 45.4% and 64.7% of all isolates were found to have an O-serotype covered by the ExPEC4V and ExPEC9V vaccine candidates, respectively. The overall frequency of MDR was 37.4% and ST131 was predominant among MDR isolates. Low in-patient genetic variability was observed in cases where multiple isolates were collected from the same patient. Conclusions Our results highlight the predominance of MDR O25b-ST131 E. coli isolates across diverse geographic areas. These findings provide further baseline data on the theoretical coverage of novel vaccines targeting E. coli associated with IED in older adults and their associated AMR levels. The consumption of seafood is driven by flavour, yet achieving its high quality remains a challenge for many species reared in recirculating aquaculture systems (RAS). A comprehensive knowledge of off‐odour sources in aquatic foods is indispensable in ensuring flavour quality standards. At the beginning of the production chain, early post‐harvest lipid oxidation products develop into endogenous off‐odours and accumulate over time. These malodours add to those already absorbed exogenously, namely from the water and feeds, although the information on the interactions between these sources is currently scarce. Despite geosmin and 2‐methylisoborneol receiving significant attention in relation to fish off‐flavour, only limited knowledge on the molecular and ecological mechanisms driving their production in aquatic environments has been reported. Moreover, RAS‐hosted bacteria have been associated with a wide range of other odour‐active compounds, such as pyrazines, terpenoids, and other degradation byproducts that are frequently overlooked when studying flavour taint in fish. The influence of aquaculture feeds on the flavour of fish flesh has been underestimated, too, both as a source of off‐odours but also as a novel modulator strategy to achieve desirable aquatic food flavours. Finally, the influence of water treatment processes widely used in RAS operations, such as advance oxidation process, ozone, ultraviolet and hydrogen peroxide disinfections, is greatly underexplored with respect to odour quality. This article reviews the current scientific evidence with supporting data on the chemical diversity of off‐odours found in aquaculture fish worldwide and their potential sources and highlights knowledge gaps that should be addressed in future research. Background: Fishes being major sources of protein are susceptible to post-harvest losses. Appropriate processing and preservation makes food healthier, safer, tastier, and more shelf-stable. Therefore, an attempt was made to evaluate the effectiveness of different processing and preservation technologies on keeping quality of Labeo rohita (rohu fish), a widely found ray-finned species in South Asia that is important for aquaculture. Methods: Raw fish fillets were subjected to different processing and preservation technologies including freezing, sun drying, mechanical drying, salting, microwave processing, and marinating to determine their effects on keeping quality. Consequently, sensorial attributes, proximate composition, mineral (calcium (Ca), iron (Fe), and zinc (Zn)) contents and microbiological analyses were carried out. Obtained data were analyzed statistically and compared using one-way analysis of variance (ANOVA) followed by Fisher's LSD test at 5% level of significance. Results: The moisture, protein, fat, ash, and carbohydrate contents were 13.01-77.29, 8.62- 52.57, 0.86-18.69, 1.34-12.72, and 1.13-27.22%, respectively in different processing and preservation technologies. Drying manifested to be the efficient method in terms of preserving proteins and fats while retention of sensory attributes was obtained in freezing. The contents of Ca, Fe, and Zn were 120.05-227.97, 0.54-1.14, and 0.72-2.04 mg/100 g, respectively. In addition, maximum retention of Zn, Fe, and Ca contents were observed in freezing, sun drying, and marinating, respectively. Furthermore, total bacterial count was in the range of 0- 2.6×104 Colony Forming Unit (CFU)/g and no coliforms were detected in any of the raw and processed fish fillets. Results of microbiological study revealed that microwave processing is highly efficient to reduce microbial loads in processed fish. Conclusion: Combination of different processing and preservation technologies might be useful to the efficient management of fishery resources. On Mars, the lack of either plate tectonics or a prominent erosional hydrological cycle since the Noachian means geological changes caused by asteroid and comet impact events have been preserved. On Earth, surviving impact‐induced fractures are localized to the relatively few preserved craters on the planet. We estimate that the shell of impact‐fractured rock on Mars (the "impact‐sphere") could provide between 9,200 times the surface area of a Mars radius sphere and up to 100 times this value, depending on the assumptions made, as potential microbially accessible space. Although >93% of craters we consider are smaller than 10 km in diameter, they contribute only about 5% of the total fracture surface area generated by all craters, making complex craters the dominant process for potential habitat formation. Microbiological data from terrestrial impact craters suggest that these fractures could have significantly enhanced local habitability by providing pathways for fluid flow, and thus nutrients and energy. However, unlike on Earth, the geological history of Mars means that pervasive impact fractures may also have provided pathways for Hesperian and Amazonian brines to infiltrate the subsurface and locally reduce habitability. Combining the fracture data with previous microbiological observations provides testable hypotheses for Martian drilling missions. Plain Language Summary: The Martian surface and subsurface is pervasively fractured by asteroid and comet impacts, largely because, compared to Earth, they have not been subducted by plate tectonics or eroded by a vigorous water cycle. In this paper, we estimate the surface area of fractures generated by these impacts. We find that these impact fractures could be 9,200 times the equivalent surface area of a flat Mars‐sized sphere and up to a hundred times this, depending on the assumptions made. On the one hand, these fractures could have provided surfaces for life and improved the flow of nutrients, on the other hand they could have channeled deleterious salty waters into the subsurface, suggesting that habitability of the subsurface is profoundly influenced by impact, but its positive or negative effects depend on the history of water flow and brine formation. Abstract Microplastics (MPs) are generated from diverse categories of plastic debris disposed in open. Its entry into the terrestrial ecosystem could not only impact soil physico-chemical attributes but also endanger the lives of biotas including the earthworms which play significant role in the decomposition of organics and sustaining the nutrient pool. There have been consistent efforts by various workers across the globe to study the impact of MPs on the terrestrial environment and exploring potential mitigation strategies to minimize contamination levels. This review presents a bibliometric analysis of scientific publications on impact of MPs on the earthworms from 2017 to 2023. The primary objective is to discern trends among authors, institutions, and countries contributing to research on MPs, particularly concerning their interaction with earthworms. The analysis reveals a steady escalation in the number of publications up to 2022. The VOS viewer software was utilized for data visualization and cluster analysis, unveiling three clusters highlighting keyword groups associated with "microplastics", "soil", and "earthworms". Notably, "microplastics" and "earthworm" emerged as prominent research hotspots.

Drying is an age long practice which prevent fish from deterioration and spoilage, however, traditional drying is usually practiced in rural settings with poor hygienic conditions, thus, the nutritional and microbial quality of such products is uncertain. Methicillin-resistant Staphylococcus aureus (MRSA) is a major cause of morbidity and mortality. Colonization by MRSA increases the risk of infection and transmission, underscoring the importance of decolonization efforts. However, success of these decolonization protocols varies, raising the possibility that some MRSA strains may be more persistent than others. Here, we studied how the persistence of MRSA colonization correlates with genomic presence of antibiotic resistance genes. Our analysis using a Bayesian mixed effects survival model found that genetic determinants of high-level resistance to mupirocin was strongly associated with failure of the decolonization protocol. However, we did not see a similar effect with genetic resistance to chlorhexidine or other antibiotics. Including strain-specific random effects improved the predictive performance, indicating that some strain characteristics other than resistance also contributed to persistence. Study subject-specific random effects did not improve the model. Our results highlight the need to consider the properties of the colonizing MRSA strain when deciding which treatments to include in the decolonization protocol.

The ability of Candida albicans to switch its morphology from yeast to filaments, known as polymorphism, may bias the methods used in microbial quantification. Here, we compared the quantification methods [cell/mL, colony forming units (CFU)/mL, and the number of nuclei estimated by viability polymerase chain reaction (vPCR)] of three strains of C. albicans (one reference strain and two clinical isolates) grown as yeast, filaments, and biofilms. Metabolic activity (XTT assay) was also used for biofilms. Comparisons between the methods were evaluated by agreement analyses [Intraclass and Concordance Correlation Coefficients (ICC and CCC, respectively) and Bland-Altman Plot] and Pearson Correlation (α = 0.05). Principal Component Analysis (PCA) was employed to visualize the similarities and differences between the methods. Results demonstrated a lack of agreement between all methods irrespective of fungal morphology/growth, even when a strong correlation was observed. Bland-Altman plot also demonstrated proportional bias between all methods for all morphologies/growth, except between CFU/mL X vPCR for yeasts and biofilms. For all morphologies, the correlation between the methods were strong, but without linear relationship between them, except for yeast where vPCR showed weak correlation with cells/mL and CFU/mL. XTT moderately correlated with CFU/mL and vPCR and weakly correlated with cells/mL. For all morphologies/growth, PCA showed that CFU/mL was similar to cells/mL and vPCR was distinct from them, but for biofilms vPCR became more similar to CFU/mL and cells/mL while XTT was the most distinct method. As conclusions, our investigation demonstrated that CFU/mL underestimated cells/mL, while vPCR overestimated both cells/mL and CFU/mL, and that the methods had poor agreement and lack of linear relationship, irrespective of C. albicans morphology/growth.1

Objective: To determine the patterns of resistance among different macrolides and to detect macrolide resistance genes in Staphylococcus aureus collected from different clinical samples. Study Design: Cross-sectional observational study. Place and Duration of Study: The clinical samples sourced from the Pathology Labs of Railway Hospital, Tehsil Head Quarter (THQ) Hospital Taxila and Wah General Hospital Wah Cantt, Pakistan was collected from 01August 2017 to 01 February 2018. Materials and Methods: One hundred non-repetitive clinical isolates were obtained from different clinical samples (pus, urine, sputum and blood). Each Staphylococcus aureus isolate was obtained from only one sample, to avoid repetition of strain. They were selected on the criteria of their growth with bright yellow color colony formation on mannitol salt agar (MSA) media along with the color change of media from red to yellow and coagulase positive results. Kirbey-Baur disk diffusion test was followed for the detection of antimicrobial sensitivity, whereas genotypic resistance drift was determined using PCR. Results: Highest frequency of Staphylococcus aureus was observed in pus samples i.e. 45 (40.2%). Out of 100, 34 isolates of Staphylococcus aureus were resistant to macrolides group. Among 34 resistant isolates the frequency of ermC gene was 25 and msrAwas 14 respectively, whereas all isolates were negative for ermAgene. Maximum resistance was observed against erythromycin, n=33 (29.5%) and minimum against clarithromycin, n=26 (23.3%). The highest susceptibility trend was seen against Azithromycin, n=62 (55%). Conclusion: Resistance of Staphylococcus aureus may vary with different antibiotics within the same group. A high frequency of erythromycin resistance was seen in this study. The most predominant resistance was for ermC, among the resistance genes in isolates.

For investigating the microbial corrosion behavior of water supply pipelines in thermal power plants using regenerated water as the water source, a circular biofilm hanging plate reactor was used to dynamically simulate the operation status of the regenerated water pipeline. The influence process and mechanism of iron bacteria(IOB), sulfate reducing bacteria(SRB) and their mixture on the corrosion of Q235 steel were studied by measuring the pH value and conductivity of the solution, weight loss of the specimen, electrochemical parameters(such as redox potential, polarization curve, AC impedance), bacterial count, and combining SEM, EDS, XRD and other methods. Results showed that the number of two types of bacteria increased over time, and the pH value of the regenerated water first decreased and then increased, while the conductivity steadily increased. Furthermore, the number of bacteria in the environment was positively correlated with the corrosion rate of pipes at this time, and SRB were the main bacterial species that affected microbial corrosion. Through EDS and XRD analysis, it was found that the main components of corrosion products varied in different environments: the corrosion products during IOB process were mainly FeOOH, with a small amount of Fe2O3 and Fe3O4; the main corrosion products during SRB process were iron oxide and iron sulfide; When SRB+IOB coexisted, the main corrosion products were Fe2O3, Fe3O4 and FeS. Besides, the microbial film and corrosion products generated on the surface of the pipeline in the early stage would inhibit the corrosion of Q235. In the later stage, the deactivation of the microbial film and the detachment of the covered corrosion products would accelerate the corrosion.

Plant–microbe interactions are key to nutrient-use efficiency. Root microbes are influenced by rhizosphere soil and plant cultivars. The impact of cultivar-by-nitrogen (N) interactions on the maize-root microbiome remains unclear, yet it is crucial for understanding N use efficiency in maize. This study evaluated the effects of maize cultivars and N forms, along with their interactions, on the diversity and composition of root bacteria and fungi. Additionally, we examined correlations between soil microbes and root metabolites. The maize cultivar Zhengdan 958 (ZD958) showed a positive response to the mixture of nitrate and ammonium N, resulting in increased in biomass, grain yield, shoot N content, grain N content, and root area. In contrast, the cultivar Denghai605 (DH605) did not exhibit a similar response. The diversity and composition of root bacteria and fungi differed between ZD958 and DH605. The N form primarily affected the community structure of rhizospheric fungi in ZD958 and rhizospheric bacteria in DH605, rather than endophytic microbes. A mixed N supply increased the relative abundance of Basidiomycota, which was positively correlated with ZD958 yield. For DH605, a mixed N treatment enhanced nitrification functions involving Bacteroidetes and Proteobacteria, while it reduced the effects of ammonium N supply. The dominant rhizospheric microbes in DH605 showed a stronger response to changes in root metabolites compared to those in ZD958. A mixed N supply increased the content of palmitoleic acid in ZD958 root exudates, facilitating the recruitment of beneficial rhizospheric microbes, which promotes maize growth. In DH605, a mixed N supply decreased the concentration of sphinganine, which is significantly correlated with Acidobacteria (negatively), Proteobacteria (negatively), Bacteroidetes (positively), and TM7 (positively). Our findings suggest that different maize cultivars respond differently to N forms, causing distinct rhizospheric microbial effects, and that root metabolites send metabolic signals to regulate and recruit key bacterial and fungal communities. Background and Objectives: This study aimed to evaluate the frequency of multidrug-resistant (MDR) bacteria in biliary samples, MDR-bacteria risk factors, and the relationship between MDR-bacteria positivity and some clinical outcomes. Materials and Methods: The study was conducted between May 2018 and May 2023, including patients over the age of 18 who had positive culture results in biliary samples. The frequency of MDR-bacteria in biliary samples was evaluated. Risk factors for MDR bacteria were assessed using univariate and multivariate analyses. MDR and non-MDR groups were compared inappropriate empirical antibiotic treatment, total antibiotic treatment duration, length of stay, and in-hospital mortality. Results: 342 microorganisms were isolated from 202 patients. Escherichia coli was the most commonly (37.2%) isolated Gram-negative microorganism, and Enterococcus spp. was the most commonly (70.2%) isolated Gram-positive microorganism. The incidence of MDR microorganisms was 42.3%. Gastrointestinal malignancy (OR: 1.96; 95% CI, 1.03-3.71) and previous antibiotic use (OR: 2.26; 95% CI, 1.09-4.68) were independent risk factors for MDR-bacteria. In the MDR group, inappropriate empirical antibiotic treatment (56.6% vs. 41%, p = 0.091), total antibiotic treatment duration (13 vs. 8 days, p = 0.054), length of stay (24 vs. 15 days, p = 0.001), and in-hospital mortality (27.3% vs. 22.3%, p = 0.416) were higher compared to the non-MDR group. Conclusion: MDR-bacteria positivity is associated with inappropriate antibiotic treatment, prolonged hospitalization, and increased mortality. Screening, antibiotic prophylaxis, and empirical treatment approaches should be carefully performed in patients with malignancy and recent antibiotic use, which are significant risk factors for MDR-bacteria. Anaerobic wastewater treatment technology has been intensively and extensively investigated in the industry and scientific research. Inspired by the advantages of multi-stage and multi-phase anaerobic reactor technology (SMPA) in recent years, a three-stage anaerobic reactor (3S-AR) was designed and applied to treat poplar chemical–mechanical pulp wastewater, and various operation parameters, including the volume loading rate (VLR), hydraulic retention time (HRT), ascending velocity, reflux ratio, pH and temperature of the 3S-AR, were optimized to evaluate the reactor's removal efficiency for poplar wastewater. The properties of anaerobic granular sludge and the composition of wastewater were also characterized to assess microorganism growth and pollutant migration. Results show that the COD removal rate was over 75% with a volume loading rate range of 15–25 gCOD/(L·d) in the 3S-AR; the hydraulic retention time was also found to be an important factor affecting the performance of the 3S-AR reactor. The volume loading rate and degradation efficiency of the 3S-AR reactor are higher than those of the up-flow anaerobic sludge blanket (UASB) reactor. Microorganism separation can be achieved in the 3S-AR, which is conducive to the growth and methanogenesis activity of bacteria, thereby leading to enhanced removal and buffering efficiency. After treatment in the 3S-AR, the main pollutants of poplar wastewater were benzene aromatic acids and long-chain esters, which do no biodegrade easily; in contrast, most of the fatty acid substances with small molecules were completely degraded. Microbial infections caused by bacteria, viruses, and fungi pose significant global health threats in diverse environments. While conventional disinfection methods are effective, their reliance on frequent chemical applications raises concerns about resistance and environmental impact. Photodynamic self-disinfecting surfaces have emerged as a promising alternative. These surfaces incorporate photosensitizers that, when exposed to light, produce reactive oxygen species to target and eliminate microbial pathogens. This review explores the concept and mechanism of photodynamic self-disinfecting surfaces, highlighting the variety and characteristics of photosensitizers integrated into surfaces and the range of light sources used across different applications. It also highlights the effectiveness of these surfaces against a broad spectrum of pathogens, including bacteria, viruses, and fungi, while also discussing their potential for providing continuous antimicrobial protection without frequent reapplication. Additionally, the review addresses both the advantages and limitations associated with photodynamic self-disinfecting surfaces and concludes with future perspectives on advancing this technology to meet ongoing challenges in infection control. The Grand River watershed is the largest catchment in southern Ontario. The river's northern and southern sections are influenced by agriculture, whereas central regions receive wastewater effluent and urban runoff. To characterize in-river microbial communities, as they relate to spatial and environmental factors, we conducted two same-day sampling events along the entire 300 km length of the river, representing contrasting flow seasons (high flow spring melt and low flow end of summer). Through high-throughput sequencing of 16S rRNA genes, we assessed the relationship between river microbiota and spatial and physicochemical variables. Flow season had a greater impact on communities than spatial or diel effects and profiles diverged with distance between sites under both flow conditions, but low-flow profiles exhibited higher beta diversity. High-flow profiles showed greater species richness and increased presence of soil and sediment taxa, which may relate to increased input from terrestrial sources. Total suspended solids, dissolved inorganic carbon, and distance from headwaters significantly explained microbial community variation during the low-flow event, whereas conductivity, sulfate, and nitrite were significant explanatory factors for spring melt. This study establishes a baseline for the Grand River's microbial community, serving as a foundation for modeling the microbiology of anthropogenically impacted freshwater systems affected by lotic processes. Objective: Practitioners and scientists are re-examining marginalised wound care therapies to find strategies that combat the growing problem of antimicrobial resistance (AMR) without compromising patient outcomes. Maggot therapy (MT) makes up just an estimated 0.02% of UK's National Health Service spending on wound care. This study aims to uncover why MT is not used more often, despite its affordability and high level of efficacy for both debridement and disinfection, particularly in the context of AMR infections, and to determine what can be done to ensure MT is more effectively used in the future to improve patient outcomes and manage the growing problem of AMR. Method: For this investigation, a qualitative review of case studies using MT against AMR infections and a quantitative analysis of randomised control trials (RCTs) were performed using the Preferred Reporting Items for Systematic Reviews and Meta-Analyses framework. Results: Analysis showed that MT is highly effective against a range of infections and wound types, and compares well against conventional therapies. The low use of MT may be due in part to the documented 'yuck factor', often associated with maggots as well as misconceptions around the cost, efficacy and accessibility of MT. To overcome these factors, more RCTs on the spectrum and efficacy of MT across various clinical manifestations are needed, as well as professional and public engagement campaigns. Conclusion: MT is an underused therapy, particularly regarding AMR infections, and expanding its use in these circumstances appears warranted. MT could play a vital role in conserving the efficacy of the existing pool of antimicrobials available and should be considered in the development of antimicrobial stewardship programmes. This manuscript documents the diagenetic progression from deposition to early burial of freshwater low‐Mg calcite mud within a Holocene sequence of the Everglades and Florida Bay (Florida, USA). During initial, in situ burial, the texturally‐complex, organic‐rich mud, exhibits high susceptibility to alteration, within a primarily dissolution‐driven realm. Seaward, a brackish‐water transitional burial environment currently displays evidence of mixed precipitation and dissolution. This brackish stage burial alteration may be due to fluid exchange (a relatively high fluid‐to‐rock ratio) in a submerged estuary setting. Lastly, the burial environment in Florida Bay records a transgression from freshwater to near‐normal marine conditions. This island setting currently exhibits a diagenetic trend towards low‐Mg calcite precipitation in intervals of both the marine and freshwater mud. Burial of freshwater mud beneath nearly 3 m of marine mud during rising sea level appears to have preserved a considerable amount of original textures, as compared with younger freshwater deposits now within the transitional brackish‐water setting. Geochemical and textural data within the freshwater mud sections suggest that complex early diagenesis begins shortly after burial at the site of formation, and perhaps even during deposition. The findings herein document three major implications for coastal zone paludal carbonates: (i) that incipient textural diagenesis in low‐Mg calcite crystals can be extremely early and the stability and petrographic fidelity of these deposits are vulnerable due to the open, trellis‐like structure of the crystals; (ii) relatedly, the stable isotope signature of the original low‐Mg calcite crystals is also vulnerable to alteration by secondary precipitation within the open crystal structure; and (iii) during marine transgression, the paludal calcite crystals likely experience a shift from interstitial fluids that are dissolution‐dominated (freshwater) to more precipitation‐dominated as marine fluids overtake the pore fluid regime. Because palustrine deposits are used as palaeoenvironmental archives, it is important to understand their diagenetic and preservation potential. Biopesticides are made up of plant materials that kill insects and a variety of microorganisms that kill insects, such as entomopathogenic fungi, entomopathogenic bacteria, entomopathogenic viruses, and entomopathogenic nematodes. These are used to get rid of agricultural and household insect pests. Several new methods are being developed in the biopesticide sector, and production and marketing of biopesticides are increasing significantly worldwide because of the noteworthy results in integrated pest management. Lots of Bt-genic crops, different types of entomopathogenic bacteria, entomopathogenic viruses, entomopathogenic fungi, and nematode-associated entomopathogenic bacteria are available in the market and are being used in agriculture to protect crops from insect pests and reduce insect pest populations. In the present review, we describe the role of biopesticides, including microbial and plant-derived ones, in insect pest management and the importance of their uses. An updated illustration of the present information on biopesticides and the availability of their commercial products and active substances in plants that can be very helpful in Insect pest management is reported in this review. Investigators seeking to solve the mystery of rock varnish formation have yet to embrace hypothesis testing. Thus, this paper presents nine tests that assess the validity of the eight hypotheses proposed over the last four decades to explain varnish formation. An almost singular focus on manganese (Mn)-enrichment by many in varnish research may have led to six of the eight hypotheses failing to explain iron (Fe)-enrichment, as well as why clay minerals dominate the composition of rock varnish. Many varnish researchers displayed a sampling bias by collecting samples only from hot and dry deserts; thus, it should be of no surprise that four hypotheses failed a test of explaining varnishes in different climates; five hypotheses failed to explain varnishes in subsurface locations; and seven hypotheses failed to explain differences in varnish growth rates in hot deserts versus wetter locations. In the end, seven of eight proposed hypotheses to explain varnish formation failed more than five tests, any one of which would falsify the hypothesis. Only one hypothesis "passed" all nine tests. Background Staphylococcus pseudintermedius is a common opportunistic pathogen of companion dogs and an occasional human pathogen. Treatment is hampered by antimicrobial resistance including methicillin resistance encoded by mecA within the mobile genetic element SCC mec. Objectives SCC mec elements are diverse, especially in non- Staphyloccocus aureus staphylococci, and novel variants are likely to be present in S. pseudintermedius. The aim was to characterize the SCC mec elements found in four canine clinical isolates of S. pseudintermedius. Material and methods Isolates were whole-genome sequenced and SCC mec elements were assembled, annotated and compared to known SCC mec types. Results and discussion Two novel SSC mec are present in these isolates. SCC mec 7017-61515 is characterized by a novel combination of a Class A mec gene complex and a type 5 ccr previously only described in composite SCC mec elements. The other three isolates share a novel composite SCC mec with features of SCC mec types IV and VI. Conclusions S. pseudintermedius is a reservoir of novel SSC mec elements that has implications for understanding antimicrobial resistant in veterinary and human medicine. [ABSTRACT FROM AUTHOR]

Objectives To determine the frequencies and clonal distributions of putative genetic determinants of resistance to antimicrobials applied for treatment of Clostridioides difficile infection (CDI), as documented in the genomic record. Methods We scanned 26 557 C. difficile genome sequences publicly available from the EnteroBase platform for plasmids, point mutations and gene truncations previously reported to reduce susceptibility to vancomycin, fidaxomicin or metronidazole, respectively. We measured the antimicrobial susceptibility of 143 selected C. difficile isolates. Results The frequency of mutations causing reduced susceptibility to vancomycin and metronidazole, respectively, increased strongly after 2000, peaking at up to 52% of all sequenced C. difficile genomes. However, both mutations declined sharply more recently, reflecting major changes in CDI epidemiology. We detected mutations associated with fidaxomicin resistance in several major genotypes, but found no evidence of international spread of resistant clones. The pCD-METRO plasmid, conferring metronidazole resistance, was detected in a single previously unreported C. difficile isolate, recovered from a hospital patient in Germany in 2008. The pX18-498 plasmid, putatively associated with decreased vancomycin susceptibility, was confined to related, recent isolates from the USA. Phenotype measurements confirmed that most of those genetic features were useful predictors of antibiotic susceptibility, even though ranges of MICs typically overlapped among isolates with and without specific mutations. Conclusions Genomic data suggested that resistance to therapeutic antimicrobial drugs is rare in C. difficile. Public antimicrobial resistance marker databases were not equipped to detect most of the genetic determinants relevant to antibiotic therapy of CDI. [ABSTRACT FROM AUTHOR]
