Methane production was tested in membrane-less microbial electrolysis cells (MECs) under closed-circuit (R CC ) and open-circuit (R OC ) conditions, using glucose as a substrate, to understand the regulatory effects of bioelectrochemistry in anaerobic digestion systems. A dynamic model was built to simulate methane productions and microbial dynamics of functional populations, which were colonized in groups R CC and R OC during the start-up stage. The experiment results showed significantly greater methane production in R CC than R OC , the average methane production of R CC was 0.131 m 3 /m 3 /d, which was 1.4 times higher than that of R OC (0.055 m 3 /m 3 /d). The simulation results revealed that bioelectrochemistry had a significant influence on the abundance of microorganisms involved in acidogenesis and methanogenesis. The abundance of glucose-uptaking microorganisms was 87% of the total biomass in R OC without applied voltage, which was 20% higher than that in R CC (67%) when external voltages were applied between the anode and cathode. The abundance of hydrogenotrophic methanogens in R CC was 6% higher than that in R OC . The simulation results were verified through 16S rDNA high-throughput sequencing analysis. An electron balance analysis revealed that alteration of the acidogenesis type led to more acetate and hydrogen production from glucose fermentation, compared with the situation without bioelectrochemistry. An additional pathway from acetate to hydrogen was introduced by bioelectrolysis. These two factors resulted in significant enhancement of methane production in R CC . Bioelectrolysis process directly contributed to 26% of the total methane production after the start-up stage. When the applied voltages were cut down or decreased, R CC could maintain considerable methane productions, because the microbial communities and electron transfer pathways were already formed. The effects of three widely-used surfactants on waste activated sludge (WAS) fermentation and microbial community structures were investigated. Rhamnolipid bio-surfactants (RL) showed more positive effects on WAS hydrolysis and acidification compared to chemosynthetic surfactants, such as sodium dodecylsulphate (SDS) and sodium dodecyl benzene sulfonate (SDBS). The highest SCOD and VFAs concentrations obtained with RL were 1.15-fold and 1.16-fold that of SDS, and up to 1.73 and 3.63 times higher than those obtained with SDBS. Pyrosequencing analysis showed that an evident reduction in bacterial diversity in surfactant-treated WAS. Moreover, acid-producing bacteria (such as Megasphaera and Oscillibacter ), detected with RL, were (6.8% and 6.4% in proportion) more abundant than with SDS, and were rarely found in SDBS and the control. Abstract: GeoChip (II) and single strand conformation polymorphism (SSCP) were used to characterize anode microbial communities of a microbial electrolysis cell (MEC). Biofilm communities, enriched in a two-chamber MEC (R1, 0.6V applied) having a coulombic efficiency (CE) of 35±4% and a hydrogen yield of 31±3%, were used as the inoculum for a new reactor (R2). After three months R2 achieved stable performance with CE=38±4% and Few changes in the predominant populations were observed from R1 to R2. Unlike sludge inoculation process in R1 in the beginning, little further elimination was aroused by community competitions in anode biofilm reformation in R2. Functional genes detection of biofilm indicated that cytochrome genes enriched soon in new reactor R2, and four genera (Desulfovibrio, Rhodopseudomonas, Shewanella and Geobacter) were likely to contribute to exoelectrogenic activity. Microbial catalysed electrochemical systems are intensively used in basic and applied research as a sustainable platform for harnessing energy and generating value added bio-products. Recently more emphasis is being laid on enhancement of AD (Anaerobic Digestion) by integrating ME (Microbial Electrolysis) with it to convert CO 2 (Carbon dioxide) directly to CH 4 (Methane). This research attempts to shed light on the effects of electrode positioning and arrangement along with hydraulic retention time (HRT), on CH 4 generation, and organic removal in novel microbial electrolysis coupled anaerobic digestion (ME-AD) reactors. However, the positioning and placement of electrodes in a ME-AD reactor have not yet been evaluated. Four reactors (S1, S2, S3 and S4) are designed with different electrode arrangements and run in four different HRTs (12, 18, 24 and 36 h) with beer brewery wastewater as the substrate for treatment. The reactors with electrodes arranged at the bottom are better in performance than the reactors with electrodes placed at the top. They have maximum COD, TOC and Carbohydrate removal efficiencies of 92.1%, 64.2% and 98.9% respectively, high methane production rate (MPR) and methane yield (MY) with 304.5 mLCH 4 /Lreactor/day and 275.8 mL/gCOD respectively and a maximum current generation of 10 mA, all at 36 h HRT. Electrode placement also has crucial roles to play in microbial community prevalence on the electrode biofilm of the reactors. Methanogens and electrogens are well enriched on the electrode biofilms of the reactors with bottom positioned electrodes, revealing the basis behind their maximum organic removal, methane production efficiencies and current generation. Pyridine contamination poses a significant threat to human and environmental health. Due to the presence of nitrogen atom in the pyridine ring, the pi bond electrons are attracted toward it and make difficult for pyridine treatment with biological and chemical methods. In this study, coupling Fenton treatment with different biological process was designed to enhance pyridine biotransformation and further mineralization. After Fenton oxidation process optimized, pretreated pyridine was evaluated under three biological (anaerobic, aerobic and microaerobic) operating conditions. Under optimum Fenton oxidation, pyridine (30–75%) and TOC (5–25%) removal efficiencies were poor. Biological process alone also showed insignificant removal efficiency, particularly anaerobic (pyridine = 8.2%; TOC = 5.3%) culturing condition. However, combining Fenton pretreatment with biological process increased pyridine (93–99%) and TOC (87–93%) removals, suggesting that hydroxyl radical generated during Fenton oxidation enhanced pyridine hydroxylation and further mineralization in the biological (aerobic > microaerobic > anaerobic) process. Intermediates were analyzed with UPLC-MS and showed presence of maleic acid, pyruvic acid, glutaric dialdehyde, succinic semialdehyde and 4-formylamino-butyric acid. High-throughput sequencing analysis also indicated that Proteobacteria (35–43%) followed by Chloroflexi (10.6–24.3%) and Acidobacteria (8.0–29%) were the dominant phyla detected in the three biological treatment conditions. Co-existence of dominant genera under aerobic/microaerobic (Nitrospira > Dokdonella > Caldilinea) and anaerobic (Nitrospira > Caldilinea > Longilinea) systems most probably play significant role in biotransformation of pyridine and its intermediate products. Overall, integrating Fenton pretreatment with different biological process is a promising technology for pyridine treatment, especially the combined system enhanced anaerobic (>10 times) microbial pyridine biotransformation activity. Boosting acetate production from waste activated sludge (WAS) fermentation is often hindered by the inefficient solubilization in the hydrolysis step and the high hydrogen pressure ( p H 2) during the acidogenesis of C3–C5 short-chain fatty acid (SCFAs), i.e., propionate (HPr), butyrate (HBu) and valerate (HVa). Therefore, this study employed persulfate (PS) oxidation and C3–C5 incomplete-oxidative sulfate reducing bacteria (io-SRB) metabolizers to tailor SCFAs conversion from WAS fermentation. The decomposition efficiency, performance of SCFAs production was investigated. Results showed that the PS significantly promoted WAS decomposition, with a dissolution rate of 39.4%, which is 26.0% higher than the un-treated test. Furthermore, SCFAs yields were increased to 462.7 ± 42 mg COD/g VSS in PS-HBu-SRB, which was 7.4 and 2.2 times higher than that of un-treated and sole PS tests, respectively. In particular, the sum of acetate and HPr reached the peak value of 85%, indicating that HBu-SRB mediation promoted the biotransformation of HBu and macromolecular organics by reducing the p H 2 restriction. Meanwhile, sulfate radical (SO 4 ∙-)-based oxidation (SR-AOPs) was effective in the decomposition of WAS, the oxidative product, i.e., sulfate served the necessary electron acceptor for the metabolism of io-SRB. Further analysis of Mantel test revealed the cluster of the functional genus and their interaction with environmental variables. Additionally, molecular ecological network analysis explored the potential synergistic and competitive relationships between critical genera. Anaerobic digestion is widely employed for the treatment of waste activated sludge (WAS) due to its advantages like simultaneous energy recovery and sludge stabilization, promoting carbon-neutral operation of wastewater treatment plants. Natural zeolite, a low-cost and eco-friendly additive, has the potential to improve methane production from anaerobic digestion. This study investigated the effects of natural zeolite on anaerobic digestion when the substrate was WAS. It was found that methane production potential in response to natural zeolite was dosage-dependent. The optimal dosage was 0.1 g zeolite/g volatile suspended solids (VSS), with a methane yield of 181.89 ± 6.75 mL/g VSS, which increased by 20.1% compared to that of the control. Although the methane yields with other dosages of natural zeolite were higher than that of control, they were lesser than that with 0.1 g zeolite/g VSS. Natural zeolite affected transfer and conversion of proteins much more than polysaccharides in liquid phase and extracellular polymeric substances. In anaerobic digestion, natural zeolite had with little effects on WAS solubilization, while it improved hydrolysis, acidification, and methanogenesis. The dosages of natural zeolite did have significant effects on bacterial communities in biofilm rather than suspension, while the archaeal communities in biofilm and suspension were all greatly related to natural zeolite dosages. The developed biofilms promoted richness and functionality of microbial communities. The syntrophic metabolism relationships between methanogens and bacteria were improved, which was proved by selective enrichment of Methanosarcina , Syntrophomonas , and Petrimonas. The findings of this work provided some new solutions for promoting methane production from WAS, and the roles of natural zeolite in anaerobic digestion. This study extracted the extracellular polymeric substances (EPS) from sludge by low-temperature thermal-hydrolysis (LTH) and high-speed hydro-cyclone (HSHC) pretreatment and recovered it for high-quality VFAs bio-production in thermophilic fermentation. Microbial mechanism analysis disclosed that interspecific interaction networks composed of functional flora, which accumulate VFAs by bio-converting EPS primarily and supplemented by EPS synthesis, guaranteed the efficient bio-production of VFAs. Current studies have employed various chemicals for disintegrating and hydrolyzing microbial cells in waste activated sludge (WAS). However, a comprehensive process assessment over the whole anaerobic digestion process has seldom been proposed. Besides, the characterization of microbial community responses to these chemicals is not well understood. In this study, the effects of five typical chemicals: solubilizer (β-cyclodextrin, CD), alkaline (NaOH), peroxide (peracetic-acid, PA), biological (rhamnolipid, RL) and chemical (sodium dodecylsulphate, SDS) surfactants on WAS digestion were examined. Higher efficiencies of WAS solubilization, hydrolysis and acidification were achieved by CD treatment, followed by RL and SDS. Methanogenesis was also strongly chemicals-dependent. Shifts in microbial community structure were observed in all chemical-pretreated WAS. The community in RL, CD and PA was dominated by microorganisms that anaerobically hydrolyze organics to acids, while that in NaOH and SDS was mainly associated to biogas production. This study proved that the overall performance of WAS digestion was substantially depended on the initial chemical pretreatments, which in turn influenced and was related to the microbial community structures. Although the economic advantage might not be clear yet, the findings obtained in this work may provide a scientific basis for the potential implementation of chemicals for WAS treatment. Quorum sensing has been widely applied to enhance the energy recovery of bioelectrochemical system as a sustainable pathway to enhance communication between cells and electrodes. However, how signalling molecules (acyl-homoserine lactones, AHLs) regulate the microbial community to improve hydrogen generation in microbial electrolysis cells (MECs) is not well understood, especially the subsequent influence on interspecies relationships among not only electrode-respiring bacteria but also hydrogen scavengers. Understanding AHL regulation in a complicated and actual biofilm system will be valuable for future applications of microbial electrochemical technology. Herein, we added short-chain AHLs (3OC6) to regulate the biofilm community on bio-electrodes in MECs. As a result, hydrogen yields were enhanced with AHL addition, increasing by 5.57%, 38.68%, and 81.82% with varied external voltages (0.8 V, 0.6 V, and 0.4 V, respectively). Accordingly, overall reactor performance was enhanced, including coulombic efficiency, electron recovery efficiency, and energy efficiency. Based on an electrochemical impedance spectra analysis, the structured biofilm under simple nutrient conditions (acetate) showed a lower internal resistance with AHL addition, indicating that the microbial communities were altered to enhance electron transfer between the biofilm and electrode. The change in the cathodic microbial structure with more electrochemically active bacteria and fewer hydrogen scavengers could contribute to a higher electron recovery and hydrogen yield with AHL addition. • Nitrogen removal through MAB was promoted by applied voltage for the first time. • The optimal applied voltage was 1.5 V for MAB to treat N-rich saline wastewater. • Candidatus Scalindua was enriched and strengthened by proper applied voltage. • The EPS and heme c secretion were enhanced by low applied voltage (≤ 1.5 V). • MAB activity was inhibited at high voltage (2.5 V) but recovered after voltage-cut. To date, the application of marine anammox bacteria (MAB) is still a challenge in saline wastewater treatment due to the low growth rate and high sensitivity. Herein, bioelectrochemical system with applied voltage was exerted for the first time to promote the activity of MAB for removing nitrogen from saline wastewater. At the optimal voltage of 1.5 V, the mean total nitrogen removal rate (TNRR) reached the maximum of 0.65 kg/m3•d, which was 27.45% higher than that without applied voltage. Besides, applied voltage reduced the microbial diversity of MAB-based consortia, but the relative abundance of Candidatus Scalindua increased by 4.63% at 1.5 V compared with that without applied voltage. Also, proper applied voltage promoted the secretion of EPS and heme c, which resulted in the enhancement of MAB activity. Based on the remodified Logistic model analysis, the lag time of the nitrogen removal process was shortened by 0.72 h at the voltage of 1.5 V. Furthermore, it was found that higher voltage (> 2.0 V) had a negative effect on the MAB activity for low TNRR of 0.33 kg/m3•d (2.5 V). However, TNRR increased back to 0.61 kg/m3•d after removing the high applied voltage, which implied that the bioactivity was recoverable after being inhibited. These findings demonstrated that external electrical stimulation is an effective strategy to promote nitrogen removal and MAB activity for treating saline wastewater. The elemental sulfur (S 0 ) recovery was evaluated in the presence of nitrate in two development models of simultaneous desulfurization and denitrification (SDD) process. At the loading rates of 0.9 kg S/(m 3 ·day) for sulfide and 0.4 kg N/(m 3 ·day) for nitrate, S 0 conversion rate was 91.1% in denitrifying sulfide removal (DSR) model which was higher than in integrated simultaneous desulfurization and denitrification (ISDD) model (25.6%). A comprehensive analysis of functional diversity, structure and metabolic potential of microbial communities was examined in two models by using functional gene array (GeoChip 2.0). GeoChip data indicated that diversity indices, community structure, and abundance of functional genes were distinct between two models. Diversity indices (Simpson's diversity index (1/ D ) and Shannon–Weaver index ( H ′)) of all detected genes showed that with elevated influent loading rate, the functional diversity decreased in ISDD model but increased in DSR model. In contrast to ISDD model, the overall abundance of dsr genes was lower in DSR model, while some functional genes targeting from nitrate-reducing sulfide-oxidizing bacteria (NR-SOB), such as Thiobacillus denitrificans , Sulfurimonas denitrificans , and Paracoccus pantotrophus were more abundant in DSR model which were highly associated with the change of S 0 conversion rate obtained in two models. The results obtained in this study provide additional insights into the microbial metabolic mechanisms involved in ISDD and DSR models, which in turn will improve the overall performance of SDD process. Enhanced performance of anammox process has attracted public attention in treating saline wastewater with marine anammox bacteria. The addition of some key metal elements would enhance metabolic activity in wastewater treatment. Herein, responses of marine anammox bacteria (MAB) to Mo(VI) addition were explored in nitrogen-laden saline wastewater treatment. The experiment was conducted at a low temperature (15 ± 1 °C). With the optimal Mo(VI) concentration of 2.1 mg/(g VSS), the substrate removal rate reached the maximum of 1.41 kg N/(m3·d). Specific anammox activity and substrate conversion rate increased by 16.0% and 36.9%, respectively. The lower ΔNO 2 --N/ΔNH 4 +-N indicated the presence of the potential electron acceptor. Moreover, the relative abundance of Cadidatus Scalindua increased by 4.18% during this experiment. Dynamic analysis showed that the remodified Logistic model was proper to analyze nitrogen removal with Mo(VI) addition.
