The multitrophic interactions in the rhizosphere impose significant impacts on microbial community structure and function, affecting nutrient mineralisation and consequently plant performance. However, particularly for long-lived plants such as forest trees, the mechanisms by which trophic structure of the micro-food web governs rhizosphere microorganisms are still poorly understood. This study addresses the role of nematodes, as a major component of the soil micro-food web, in influencing the microbial abundance and community structure as well as tree growth. In a greenhouse experiment with Pedunculate Oak seedlings were grown in soil, where the nematode trophic structure was manipulated by altering the proportion of functional groups (i.e., bacterial, fungal, and plant feeders) in a full factorial design. The influence on the rhizosphere microbial community, the ectomycorrhizal symbiont Piloderma croceum, and oak growth, was assessed. Soil phospholipid fatty acids were employed to determine changes in the microbial communities. Increased density of singular nematode functional groups showed minor impact by increasing the biomass of single microbial groups (e.g., plant feeders that of Gram-negative bacteria), except fungal feeders, which resulted in a decline of all microorganisms in the soil. In contrast, inoculation of two or three nematode groups promoted microbial biomass and altered the community structure in favour of bacteria, thereby counteracting negative impact of single groups. These findings highlight that the collective action of trophic groups in the soil micro-food web can result in microbial community changes promoting the fitness of the tree, thereby alleviating the negative effects of individual functional groups. Plant residues provide a major source of nitrogen (N) for plant growth. Litter N mineralization varies with litter carbon-to-nitrogen (C-to-N) ratio and presence of bacterial-feeding fauna. We assessed the effect of amoebae, major bacterial feeders in soil, on mineralization of litter of low (high quality) and high C-to-N ratio (low quality) and evaluated consequences for plant growth. We used stable isotopes to determine plant N uptake from litter and plant C partitioning. Stable isotope probing of phospholipid fatty acids was used to follow incorporation of plant C into microorganisms. Amoebae increased plant N uptake independent of litter quality and thereby the biomass of shoots and roots by 33% and 66%, respectively. Plant allocation of total 13C to roots in low (42%) exceeded that of high-quality litter treatments (26%). Amoebae increased plant allocation of 13C to roots by 37%. Microbial community structure and incorporation of 13C into PLFAs varied significantly with litter quality and in the low-quality litter treatment also with the presence of amoebae. Overall, the results suggest that in particular at low nutrient conditions, root-derived C fosters the mobilization of bacterial N by protozoa, thereby increasing plant growth when microorganisms and plants compete for nutrients. Subsurface microorganisms are crucial for contaminant degradation and maintenance of groundwater quality. This study investigates the microbial biomass and community composition [by phospholipid fatty acids (PLFAs)], as well as physical and chemical soil characteristics at woodland flooding sites of an artificial groundwater recharge system used for drinking water production. Vertical soil profiles to c. 4 m at two watered and one nonwatered site were analyzed. The microbial biomass was equal in watered and nonwatered sites, and considerable fractions (25–42%) were located in 40–340 cm depth. The microbial community structure differed significantly between watered and nonwatered sites, predominantly below 100 cm depth. Proportions of the bacterial PLFAs 16:1ω5, 16:1ω7, cy17:0 and 18:1ω9t, and the long-chained PLFAs 22:1ω9 and 24:1ω9 were more prominent at the watered sites, whereas branched, saturated PLFAs (iso/anteiso) dominated at the nonwatered site. PLFA community indices indicated stress response ( trans/ cis ratio), higher nutrient availability (unsaturation index) and changes in membrane fluidity (iso/anteiso ratio) due to flooding. In conclusion, water recharge processes led to nutrient input and altered environmental conditions, which resulted in a highly active and adapted microbial community residing in the vadose zone that effectively degraded organic compounds. Increased carbon translocation to the rhizosphere via ‘leakage’ induced by low amounts of plant parasitic nematodes can foster microorganisms. The effects of the root-knot nematode Meloidogyne incognita on microbial biomass (Cmic) and community structure (phospholipid fatty acids) in the rhizosphere of barley were studied. Inoculation densities of 2000, 4000, and 8000 nematodes were well below the threshold level for plant damage. A 13CO2 pulse-labelling was performed to assess the distribution of assimilated 13C in the rhizosphere. Infection with M. incognita increased the carbon concentration in shoots, and enhanced root biomass slightly. The presence of nematodes did not affect microbial biomass, but significantly changed the allocation of the recent photosynthate. Less plant carbon was sequestered by microorganisms with increasing nematode abundance. Microbial community structure was distinctly altered in the early stages of the plant–nematode interactions. Both, bacteria and fungi, showed a positive response with 2000, and a negative one with 4000 and 8000 M. incognita added. The results suggest that low-level root herbivory still imposes a considerable carbon demand, and that proliferation of microorganisms due to increased rhizodeposition may be short-termed. Abstract: The lipid composition of Collembola and their potential food resources was assessed in three deciduous forest stands, in order to gain insight into food web linkages under field conditions. Fatty acids (FAs) previously assigned as trophic markers in laboratory experiments were used to investigate feeding strategies in situ. As potential food sources soil microbiota and plant debris were characterised by their phospholipid fatty acid (PLFA) composition. Both the amount and the pattern of PLFAs differed between sites and soil depth, in particular the bacterial and plant marker FAs in the upper soil layer. Thus, the availability of resources for micro-detritivores varied due to forest and soil layer. The lipid composition of vital and senescent beech leaves was predominantly influenced by metabolic status and represented a quite homogenous FA resource across forest stands. Comparing Folsomia quadrioculata, Lepidocyrtus lignorum, Neanura muscorum and Pogonognathellus longicornis between the different forests revealed FA profiles to be predominantly affected by site, suggesting a diet shift depending on resources at hand. However, species-specific differences in individual FAs occurred, likely related to feeding strategy and physiological activity. Lipids of Collembola comprised low amounts of bacterial marker FAs, and bacterial consumption may occur to some extent, particularly on Gram-positives. The marker FA for predatory feeding, , was found in several species, although in low amounts. This contradicts known feeding habits and caution is advisable in using as trophic marker. Overall, as indicated by high proportions of oleic or linoleic acid, most species were either plant litter or fungal feeders, with some transitions. This study assessed the carbon (C) budget and the C stocks in major compartments of the soil food web (bacteria, fungi, protists, nematodes, meso- and macrofauna) in an arable field with/without litter addition. The C stocks in the food web were more than three times higher in topsoil (0–10 cm) compared to subsoil (>40 cm). Microorganisms contained over 95% of food web C, with similar contributions of bacteria and fungi in topsoil. Litter addition did not alter C pools of soil biota after one growing season, except for the increase of fungi and fungal feeding nematodes in the topsoil. However, the C budget for functional groups changed with depth, particularly in the microfauna. This suggests food web resilience to litter amendment in terms of C pool sizes after one growing season. In contrast, the distinct depth dependent pattern indicates specific metacommunities, likely shaped by dominant abiotic and biotic habitat properties. [ABSTRACT FROM AUTHOR]

The agricultural application of organic fertilizer as well as irrigation with partially treated wastewater is proposed to increase crop yield, but also the incidence of food borne pathogens. Studies have concentrated on the impact of such management practice on soil health, and on spread of pathogens during harvest, transport, and marketing, whereas the role of soil organisms in these processes was largely neglected. Therefore, the objective of the present work was to study microorganisms and nematodes as potential agents in suppression or dissemination of human pathogens. Two experimental field sites were established in semi-arid soils in the West Bank in Palestine. Treatments comprised application of fresh water, wastewater, sewage sludge and chicken manure in three successive crop cycles, and the effects on the microbial and nematode communities were assessed. Microbial biomass was generally fostered by all three organic amendments but the increase was dependent on arable site as well as the type of urban waste. The introduced microorganisms were able to establish in the field environment, indicating that the autochthonous microbial community was not effective in suppressing allochthonous enteropathogens. Nematodes, as potential bacterial vectors, displayed a population increase with organic input across crops and sites. Effects were most distinct by amendment with chicken manure, which additionally resulted in a decrease in diversity, with a distinct increase in bacterial-feeding r -strategists and strongly enriched food web conditions. In contrast, application of treated wastewater and sewage sludge fostered K -strategist, predominantly omnivores. This enhanced biomass at higher trophic levels indicates ample energy flux along the trophic cascade as well as a stable and structured food web, despite possible contaminants introduced via urban residues. Abstract: Soil food webs are important determinants for the carbon flow through terrestrial systems, with the trophic networks between microbes and microfaunal grazers forming the basis for processing plant resources. At an agricultural field site cropped with maize or wheat, plant carbon input to soil was experimentally manipulated by amendment with maize litter. The community structure of dominant micro-food web components, the bacteria, fungi, protozoa and nematodes, was investigated across a depth gradient comprising plough layer, rooted soil below plough horizon, and deeper root free soil. The community composition and diversity within micro-food webs, and the response to resource supply, were assessed in summer, the vegetation period with highest root exudation. In the plough layer amendment with plant residues increased microbial biomass as well as density of fan shaped amoeba morphotypes and of bacterial- and fungal-feeding nematodes. Diversity of food web assemblages was assessed by operational taxonomic units (OTU) for bacteria and fungi, protozoa morphotypes and nematode families. Changes in diversity were either not apparent (fungi, protozoa), negatively related to litter (bacteria) or positively linked to the presence of a specific crop plant (bacteria, nematodes). Based on nematode functional guilds and the related enrichment and structure index, general food web conditions were assigned as nutrient enriched, with a high degree of disturbance, and a dominance of the bacterial energy channel. In sum, litter amendment fostered the abundance but not the diversity of organisms as food webs remained bottom heavy with only small amounts of carbon conserved at higher trophic levels. The microbiota in three distinct terrestrial habitats (biological soil crusts – BSC, crust-adhering soil beneath BSC – trans, and crust-free soil close to BSC – soil) was studied in 33 temperate forest stands differing in management intensity (ForMI). Specific lipid fractions and respective marker fatty acids were used to determine microbial biomass and community structure (phospholipid fatty acids – PLFA), energy reserves (neutral lipid fatty acids – NLFA), and community level lipid profiles (CLLP) of nematodes and enchytraeids. The density and diversity of nematode fauna were additionally determined morphologically. The highest total amounts of PLFA and NLFA occurred in the BSC, followed by soil and trans habitats, indicating stronger effects of habitats than forest management intensity. This corresponds to the distribution pattern of bacterial PLFA that dominated the soil microbiota. The highest nematode population density occurred in the BSC, followed by soil and trans. The BSC was dominated by bacterial feeders and omnivores, while plant feeding nematodes were most frequent in the crust-free soil habitat. The highest biomass of photoautotrophs also occurred in the BSC, followed by the crust-adhering and crust-free soil habitats. Redundancy analysis indicated strong effect of ForMI and total N on soil PFLAs and NFLAs across habitats. The C:N and C:S ratios were important for the separation of NLFAs while P:S accounted for the separation of PLFAs. Bacterial and fungal PLFAs largely separated the microbial community across habitats, and particularly between BSC and trans habitats. The separation of nematodes and enchytraeids by CLLP was less pronounced, and most distinct between the crust-free soil and BSC or crust-adhering soil habitats.
