Brown Swiss (BS) cows have greater urea concentrations in milk and blood compared with Holstein (HO) cows. We tested the hypothesis that BS and HO cows differ in kidney function and nitrogen excretion. Blood, saliva, urine, and feces were sampled in 31 multiparous BS and 46 HO cows kept under identical feeding and management conditions. Samples were collected at different lactational stages after the monthly DHIA control test-day. To test the glomerular filtration rate (GFR) and urea excretion, concentrations of creatinine and urea were measured in serum, urine, and saliva. As an additional marker to estimate GFR, we determined symmetric dimethylarginine (SDMA) in serum. Feces were analyzed for DM content and nitrogen concentration. Data on milk urea and protein concentrations and daily milk yield were obtained from the monthly DHIA test-day records. The effects of breed, time, and parity number on blood, saliva, urine, feces, and milk parameters were evaluated with the GLM procedure of SAS, with breed, time, and parity number as fixed effects. Differences between BS and HO were assessed by the Tukey-corrected t-test. Concentrations of urea, creatinine, and SDMA in serum, were greater in BS than in HO cows: 5.46 ± 0.19 versus 4.72 ± 0.13 mmol/L (urea), 105.96 ± 2.23 versus 93.07 ± 1.50 mmol/L (creatinine), and 16.78 ± 0.69 versus 13.39 ± 0.44 µg/dL (SDMA). We observed a greater urea concentration in BS cows (25.8 ± 0.7 vs. 21.8 ± 0.7 mg/dL) and protein content in milk (3.70% ± 0.08% vs. 3.45% ± 0.07%) than in HO cows. Urea and creatinine concentrations in urine and saliva did not differ between breeds. No differences between BS and HO were observed for milk yield, fecal DM, and fecal nitrogen content. Dry matter intake and BW were similar in BS and HO cows. Despite greater urea, creatinine, and SDMA concentrations in blood, as well as a higher milk urea content in BS compared with HO, respective concentrations in urine did not differ between breeds. In conclusion, our results demonstrate a lower renal GFR in BS compared with HO cows, thereby contributing to the greater plasma urea concentration in BS cows. The aim of the present study was to assess the effect of serum calcium (Ca) status in the early postpartum period on the quantity and quality of colostrum, milk production, and the health of Holstein dams and their calves. One hundred multiparous Holstein cows were enrolled in this study. To determine serum Ca status, blood samples were taken at 1 and 4 days in milk (DIM). Cows were categorized into 1 of 4 groups based on their serum Ca concentrations: normocalcemic (CON; Ca >1.87 at 1 DIM and >2.10 mmol/L at 4 DIM, n = 36), transient SCH (TSCH; Ca ≤1.87 at 1 DIM and >2.10 mmol/L at 4 DIM, n = 14), persistent SCH (PSCH; Ca ≤1.87 at 1 DIM and ≤2.10 mmol/L at 4 DIM, n = 15), or delayed SCH (DSCH; Ca >1.87 at 1 DIM and ≤2.10 mmol/L at 4 DIM, n = 35). The ROC curve was utilized to determine the optimal cut-off value for serum total Ca, maximizing both sensitivity and specificity in predicting metritis. Metabolic disorders such as dystocia, retained placenta, and culling within 60 DIM were not affected by serum Ca status in the early postpartum period (P > 0.10). However, the incidences of metritis and endometritis were different based on the serum Ca status of the cows. The PSCH cows had a higher incidence of metritis than the CON cows (P = 0.05). Also, the DSCH cows tended to have an increased risk of metritis than the CON cows (P = 0.09). However, metritis incidence in the TSCH and CON cows was similar (P = 0.83). Cows with PSCH (P = 0.07) and DSCH (P = 0.10) tended to be at a higher risk for endometritis than the CON cows. But, the incidence of endometritis in the TSCH and CON cows was not different (P = 0.50). The TSCH cows had higher milk yield than the PSCH and DSCH cows (P = 0.05). However, milk yield was not different between the TSCH and CON cows and the PSCH and DSCH and CON cows. The serum Ca status of cows had no effect on colostrum volume and its quality, age and weight of weaning, and average daily gain of calves (P > 0.10). The proportion of calves experiencing failure of passive transfer (FPT) was greater in the PSCH group than in the CON group (P = 0.03). Also, the FPT tended to be higher in the DSCH calves compared to the CON calves (P = 0.10). However, the FPT in the TSCH and CON calves was not different (P = 0.83). The incidence of diarrhea tended to be higher in the PSCH and DSCH calves compared to the CON calves (P = 0.10). However, diarrhea incidence between the TSCH and CON calves was similar (P = 0.97). Overall, the findings of this study suggest that cows with TSCH showed better adaptation to lactation, experienced a lower occurrence of diseases and produced higher milk compared to PSCH and DSCH cows. Genetic research for the assessment of mastitis and milk production traits simultaneously has a long history. The main issue that arises in this context is the known existence of a positive correlation between the risk of mastitis and lactation performance due to selection. The transcriptome-wide association study (TWAS) approach endeavors to combine the expression quantitative trait loci and genome-wide association study summary statistics to decode complex traits or diseases. Accordingly, we used the farmgtex project results as a complete bovine database for mastitis and milk production. The results of colocalization and TWAS approaches were used for the detection of functional associated candidate genes with milk production and mastitis traits on multiple tissue-based transcriptome records. Also, we used the david database for gene ontology to identify significant terms and associated genes. For the identification of interaction networks, the genemania and string databases were used. Also, the available z-scores in TWAS results were used for the calculation of the correlation between tissues. Therefore, the present results confirm that LYNX1, DGAT1, C14H8orf33, and LY6E were identified as significant genes associated with milk production in eight, six, five, and five tissues, respectively. Also, FBXL6 was detected as a significant gene associated with mastitis trait. CLN3 and ZNF34 genes emerged via both the colocalization and TWAS approaches as significant genes for milk production trait. The aims of this study were to conduct a single-step genome-wide association to identify genomic regions associated with milk urea (MU) and to estimate genetic correlations between MU and milk yield (MY), milk composition (calcium content [CC], fat percentage [FP], protein percentage [PP], and casein percentage [CNP]), and cheese-making properties (CMP; coagulation time [CT], curd firmness after 30 min from rennet addition [a30], and titratable acidity [TA]). The used data have been collected from 2015 to 2020 on 78,073 first-parity (485,218 test-day records) and 48,766 second-parity (284,942 test-day records) Holstein cows distributed in 671 herds in the Walloon Region of Belgium. Data of 565,533 SNP located on 29 BTA of 6,617 animals (1,712 males) were used. Random regression test-day models were used to estimate genetic parameters through the Bayesian Gibbs sampling method. The SNP solutions were estimated using a single-step genomic BLUP approach. The proportion of the total additive genetic variance explained by windows of 50 consecutive SNPs (with an average size of ∼216 kb) was calculated, and the top-3 genomic regions explaining the largest rate of the genetic variance were considered promising regions and used to identify potential candidate genes. Mean (SD) MU was 25.38 (8.02) mg/dL and 25.03 (8.06) mg/dL in the first and second lactation, respectively. Mean heritability estimates for daily MU were 0.21 and 0.23 for the first and second lactation, respectively. The genetic correlations estimated between MU and MY, milk composition, and CMP were quite low (ranged from -0.10 [CC] to 0.10 [TA] and -0.05 [CT] to 0.13 [TA] for the first and second lactations, respectively). The top-3 regions associated with MU were located from 80.61 to 80.74 Mb on BTA6, 103.26 to 103.41 Mb on BTA11, and 1.59 to 2.15 Mb on BTA14. Genes including KCNT1, MROH1, SHARPIN, TSSK5, CPSF1, HSF1, TONSL, DGAT1, SCX, and MAF1 were identified as positional candidate genes for MU. Milk production traits as the most important economic traits of dairy cows, they directly reflect the benefits of breeding and the economic benefits of pasture. In this study, A disintegrin and metalloproteinase-12 (ADAM12), Parkinson's disease gene 2 (PRKN) and dipeptidyl peptidase-like protein subtype 6 (DPP6) polymorphism in 384 Chinese Holstein cows were detected by time-of-flight mass spectrometry and through statistical analysis using software such as Popgene 32, SAS 9.4 and Origin 2022, the relationship between single nucleotide polymorphisms (SNPs) of three genes with four milk production traits such as daily milk yield (DMY), milk fat percentage (MFP), milk protein percentage (MPP) and somatic cell score (SCS) was verified at molecular level. The results showed that four polymorphic loci (116,467,133, 116,604,487, 116,618,268 and 116,835,111) of DPP6 gene, two polymorphic loci (97,665,052 and 97,159,837) of PRKN gene and two polymorphic loci (45,542,714 and 45,553,888) of ADAM12 gene were detected. PRKN-97665052, DPP6-116467133, ADAM12-45553888, DPP6-116604487 and DPP6-116835111 were all in Hardy-Weinberg equilibrium state (p > .05). ADAM12-45542714, PRKN-97159837 and PRKN-97665052 were moderately polymorphic (0.25 ≤ PIC <0.50) in Holstein. It is evident that the selection potential and genetic variation of these five loci are relatively large, and the genetic richness is relatively high. The correlation analysis of different genotypes between these eight loci and milk production traits of Holstein showed that ADAM12-45542714 and DPP6-116835111 (p < .01) had an extremely significant effects on the DMY of Chinese Holstein in Ningxia, while PRKN-97665052 had an extremely significant effect on MFP (p < .01). The effect of PRKN-97665052 and DPP6-116467133 on MPP of Holstein were extremely significant (p < .01). DPP6-116618268 had an extremely significant effect on the SCS of Holstein in Ningxia (p < .01), and AA genotype individuals showed a higher SCS than GG genotype individuals; the other two loci (ADAM12-45553888 and DPP6-116604487) had no significant effects on milk production traits of Holstein (p > .05). In addition, through the joint analysis of DPP6, PRKN and ADAM12 gene loci, it was found that the interaction effect between the three gene loci could significantly affect the DMY, SCS (p < .01) and MPP (p < .05). In conclusion, several different loci of DPP6, PRKN and ADAM12 genes can affect the milk production traits of Holstein to different degrees. This study investigated the age at first calving in Holstein heifers and its impact on various production parameters. A sample of 737 Holstein heifers born between 2015 and 2018 and finishing their first lactations between 2018 and 2020 was included. Cluster analysis revealed three groups based on age at first calving: high precocity, medium precocity and low precocity. Medium-precocity group exhibited the highest 305-day milk yield and peak milk production. Additionally, the same group demonstrated superior mean production per lactation, 305-day milk protein content, and 305-day milk yield. The lowest somatic cell count was found in the low-precocity group. In conclusion, medium-precocity cows showed better results such as higher 305-day milk yield and peak milk production.

This study aimed to find the parsimonious random regression model (RRM) to evaluate the genetic potential for milk yield (MY), fat content (FC), and protein content (PC) in Tunisian Holstein cows. For this purpose, 551,139; 331,654; and 302,396 test day records for MY, FC, and PC were analysed using various RRMs with different Legendre polynomials (LP) orders on additive genetic (AG) and permanent environmental (PE) effects, and different types of residual variances (RV). The statistical analysis was performed in a Bayesian framework with Gibbs sampling, and the model performances were assessed, mainly, on the predictive ability criteria. The study found that the optimal model for evaluating these traits was an RRM with a third LP order and nine classes of heterogeneous RV. In addition, the study found that heritability estimates for MY, FC, and PC ranged from 0.11 to 0.22, 0.11 to 0.17, and 0.12 to 0.18, respectively, indicating that genetic improvement should be accompanied by improvements in the production environment. We fit the Wood's lactation model to an extensive database of test-day milk production records of US Holstein cows to obtain lactation-specific parameter estimates and investigated the effects of temporal, spatial, and management factors on lactation curve parameters and 305-d milk yield. Our approach included 2 steps as follows: (1) individual animal-parity parameter estimation with nonlinear least-squares optimization of the Wood's lactation curve parameters, and (2) mixed-effects model analysis of 8,595,413 sets of parameter estimates from individual lactation curves. Further, we conducted an analysis that included all parities and a separate analysis for first lactation heifers. Results showed that parity had the most significant effect on the scale (parameter a), the rate of decay (parameter c), and the 305-d milk yield. The month of calving had the largest effect on the rate of increase (parameter b) for models fit with data from all lactations. The calving month had the most significant effect on all lactation curve parameters for first lactation models. However, age at first calving, year, and milking frequency accounted for a higher proportion of the variance than month for first lactation 305-d milk yield. All parameter estimates and 305-d milk yield increased as parity increased; parameter a and 305-d milk yield rose, and parameters b and c decreased as year and milking frequency increased. Calving month estimates parameters a, b, c, and 305-d milk yield were the lowest values for September, May, June, and July, respectively. The results also indicated the random effects of herd and cow improved model fit. This study was conducted to (1) estimate Wood's incomplete gamma function parameters (a, b and c) for individual registered Tunisian Holsteins in the 3 first lactations; (2) estimate persistency (Per), peak time (PT), peak yield (PY) and 305-day lactation milk yields (Y305); (3) estimate the genetic parameters of the predicted traits and (4) evaluate the genetic trend of the most heritable traits. Pedigree data and 380,709 test-day milk records of 20,758 Holstein cattle collected from 2008 to 2018 in 33 herds were analyzed. Initially, Wood's incomplete gamma function parameters were determined and subsequently along with some production characteristics (Per, PT, PY and Y305). Each estimated trait was subjected to a single trait animal model to estimate genetic parameters using MTDFREML program (Boldman et al. 1995). Multivariate animal model was used to estimate genetic and phenotypic correlations between the most heritable traits and their breeding values (EBVs). Genetic trends were defined using BREEDPLAN multi-trait EBVs by averaging the EBV of all animals born by year of birth. The resulting heritability estimates varied from 0.04 to 0.06 for Per, 0.01 to 0.02 for PT, 0.08 to 0.16 for PY and 0.09 to 0.16 for Y305. The greatest heritabilities were within the first lactation for PY and Y305 traits. Genetic and phenotypic correlations were very high between the most heritable two traits (0.91 and 0.83, respectively). Heritability estimates of PT and Per were close to zero indicating that these traits are more influenced by the management and the environment. The regression coefficients, relative to the genetic trend graph, mark an overall rate of increase of 13.18 kg/year and 0.056 kg/year in Y305 and PY, respectively, over 16-year period. Milk urea content is receiving growing interest from science and industry as a tool to infer the protein adequacy of dairy cows' diets, nitrogen excretion and its environmental impact, and efficiency of animals' protein metabolism. Fourier-transform infrared (FTIR) prediction is a high-throughput method for rapidly and cheaply evaluating milk urea content at the population level. Existing knowledge of the major sources of variation (e.g., year, season, farming system, individual herd, and the cow's breed, parity, stage of lactation, and productive potential) is fragmentary. The objective of this work was to study at the population level the simultaneous effects of all the major sources of variation and their most important interactions. Milk urea content in 1,759,706 test day milk samples collected from 291,129 lactations of 115,819 cows from 6,430 herds over 8 yr was predicted by FTIR. The milk urea content data (and also milk protein percentage, for comparison) were analyzed using a linear model that included the effects of parity, days in milk (DIM) class, year, month, herd intensiveness level, cow productivity level, breed, and herd intensiveness and cow productivity levels within breed. All sources of variation of milk urea content proved highly significant, the most important in terms of F-value being breed > year > herd intensiveness level > parity. The ranking for milk protein was very different (DIM class > herd intensiveness level > parity > breed). The patterns of the least squares means for urea and protein contents of milk were also very different and sometimes contrasting. The seasonal variation in urea was sinusoidal. Urea content increased during the first 4 mo of lactation and then remained almost stable before decreasing after 11 mo. Specialized dairy breeds had lower average milk urea content than dual-purpose breeds; in the former case it was lower in Holsteins than in Brown Swiss, and in the latter it was lower in Simmentals than in Alpine Greys. The effect of herd intensiveness level was much stronger than the effect of cow productivity level; the increase in milk urea with increasing herd average daily milk yield was almost linear in the case of dairy breeds but curvilinear in dual-purpose breeds. The large differences in breed and the modest relationships with the cow's productive potential require further analysis at the genetic level to obtain information of potential use in genetic improvement of the dairy cow populations., (© 2022, The Authors. Tropical and subtropical milk production herds in Mexico generally generate different types of milk-yield records as milk yield per month and mean production per month. Lactation curves generated by these types of records may contribute to understand milk production in the tropical regions of Mexico. The aim of this study was to compare five lactation-curve models fitted to two types of milk-yield records of Holstein, Brown Swiss, and F1 crossbred cows under subtropical conditions. The two types of records (n = 3756) used were: (1) milk yield per month (TR) and 2) mean production per months (MR). Goodness-of-fit statistics, including Akaike's information criterion (AIC) and root mean square error (RMSE), were applied to compare the models for each type of records. The Brody model provided the best goodness-of-fit when using monthly milk-yield records, while the Wilmink model provided the best goodness-of-fit for lactation milk-yield records. The RMSE and AIC values were similar between datasets. The final third of the lactation curve showed a little difference between model predictions in both datasets. The comparison of several models was useful to better describe the actual lactation curves of the herd. The specific fatty acid (FA) profile of colostrum may indicate a biological requirement for neonatal calves. The objective of this study was to characterize the FA profile and yields in colostrum, transition milk, and mature milk in primiparous (PP) and multiparous (MP) cows. Colostrum was milked from 10 PP and 10 MP Holstein cows fed the same pre- and postpartum rations. Milkings (M) 2 to 5 and 12 were respectively termed transition and mature milk. Overall, short-chain FA (C4:0 and C6:0) were 61 and 50% lower in colostrum than mature milk, respectively. A parity by milking interaction was also present, with higher C4:0 for PP cows at M2 and for MP cows at M12. Additionally, higher concentrations of C6:0 were present for PP cows at M2 through M4 and for MP cows at M12. Palmitic (C16:0) and myristic (C14:0) acids were 38% and 19% higher in colostrum than mature milk, respectively. However, total saturated FA remained relatively stable. Branched-chain FA were 13% lower in colostrum than mature milk and higher in PP than MP cows throughout the milking period. The proportion of trans-monounsaturated FA (MUFA) was 72% higher in PP cows throughout the milking period, as well as 13% lower in colostrum than mature milk. In contrast, cis-MUFA and total MUFA were not affected by milking nor parity. Linoleic acid (LA) was 25% higher in colostrum than transition and mature milks, but α-linolenic acid (ALA) did not differ. Consequently, the ratio of LA to ALA was 29% higher in colostrum than mature milk and 33% higher in MP cows. Linoleic acid was also 15% higher in MP cows, whereas ALA was 15% higher in PP cows. Conjugated linoleic acid (CLA, cis-9,trans-11) was 2.7-fold higher in PP cows, and no differences between colostrum and mature milk were detected. Overall, polyunsaturated FA (PUFA) from the n-6 and n-3 series were over 40% higher in colostrum compared with transition and mature milk. Milking by parity interactions were present for arachidonic acid (ARA), eicosapentaenoic acid (EPA), docosapentaenoic acid (DPA), docosahexaenoic acid (DHA), and total n-3 PUFA, translating to higher proportions in PP cows in M1 to M3, whereas proportions remained relatively stable throughout the milking period in MP cows. Despite increasing milk yields throughout the subsequent milkings, higher yields of EPA, ARA, DPA, and DHA were present in colostrum than in mature milk. Greater proportions and yields of n-3 and n-6 FA in colostrum may translate to specific requirements for newborn calves. The present study aimed to estimate covariance components of milk fatty acids (FA) and to compare the genomic estimated breeding values under general and heat-stress effects. Data consisted of 38,762 test-day records from 6,344 Holstein cows obtained from May 2012 through January 2018 on 4 dairy herds from Brazil. Single-trait repeatability test-day models with random regressions as a function of temperature-humidity index values were used for genetic analyses. The models included contemporary groups, parity order (1-6), and days in milk classes as fixed effects, and general and thermotolerance additive genetic and permanent environmental as random effects. Notably, differences in heritability estimates between environments (general and heat stress) increased (0.03 to 0.06) for unsaturated FA traits, such as unsaturated, monounsaturated, and polyunsaturated, at higher heat-stress levels. In contrast, heritability estimated between environments for saturated FA traits, including saturated FA, palmitic acid (C16:0), and stearic acid (C18:0) did not observe significant differences between environments. In addition, our study revealed negative genetic correlations between general and heat-stress additive genetic effects (antagonistic effect) for the saturated FA, C16:0, C18:0, and C18:1, which ranged from -0.007 to -0.32. Spearman's ranking correlation between genomic estimated breeding values ranged from -0.27 to 0.99. Results indicated a moderate to strong interaction of genotype by the environment for most FA traits comparing a heat-stress environment with thermoneutral conditions. We compared values of Wilmink's exponential term to describe the lactation curves of Holstein cows in Japan. Data were a total of 100,971,798 test-day records from the first through fifth parities during 1991 through 2018. The lactation curve model used fourth-order Legendre polynomials and Wilmink's exponential term. In total, 810 analyses were executed to compare six values (-0.02, -0.03, …, -0.07) for the exponential term to select the one that yielded the smallest root mean square error. For all parities, daily milk yield and lactation persistency increased consistently and peak lactation days occurred later from year to year. For the years evaluated, the optimal exponential term was -0.05 for first and second parities, -0.04 for third parity, and -0.03 for fourth and fifth parities. Accurately profiling and characterizing factors shaping raw milk microbiota would provide practical information for detecting microbial contamination and unusual changes in milk. The current work was an observational study aiming to profile the microbiota of raw milk collected across wide geographic regions in China in different seasons and to investigate the contribution of geographical, seasonal, and environmental factors in shaping the raw milk microbiota. A total of 355 raw cow milk samples from healthy Holsteins and 41 environmental samples (farm soil and surface of milking room floor) were collected from 5 dairy farms in 5 Chinese provinces (namely, Daqing in Heilongjiang province, Jiaozuo in Henan province, Qingyuan in Guangdong province, Suqian in Jiangsu province, and Yinchuan in Ningxia Hui Autonomous Region) in January, May, and September 2018. The microbial communities in raw milk and farm environmental samples were determined using the PacBio small-molecule real-time circular consensus sequencing, which generated high-fidelity microbiota profiles based on full-length 16S rRNA genes; such technology was advantageous in producing accurate species-level information. Our results showed that both seasonality and sampling region were significant factors influencing the milk microbiota; however, the raw milk microbiota was highly diverse according to seasonality, and sampling region was the less determining factor. The wide variation in raw milk microbial communities between samples made it difficult to define a representative species-level core milk microbiota. Nevertheless, 3 most universal milk-associated species were identified: Lactococcus lactis, Enhydrobacter aerosaccus, and Acinetobacter lwoffii, which were consistently detected in 99%, 95%, and 94% of all analyzed milk samples, respectively (n = 355). The top taxa accounting for the overall seasonal microbiota variation were Bacillus (Bacillus cereus, Bacillus flexus, Bacillus safensis), Lactococcus (Lactococcus lactis, Lactococcus piscium, Lactococcus raffinolactis), Lactobacillus (Lactobacillus helveticus, Lactobacillus delbrueckii), Lactiplantibacillus plantarum, Streptococcus agalactiae, Enhydrobacter aerosaccus, Pseudomonas fragi, and Psychrobacter cibarius. Unlike the milk microbiota, the environmental microbiota did not exhibit obvious pattern of seasonal or geographic variation. However, this study was limited by the relatively low number and types of environmental samples, making it statistically not meaningful to perform further correlation analysis between the milk and environmental microbiota. Nevertheless, this study generated novel information on raw milk microbiota across wide geographic regions of China and found that seasonality was more significant in shaping the raw milk microbiota compared with geographic origin., (Copyright © 2021 American Dairy Science Association. Milk and dairy products are considered important sources of iodine in several countries. Despite this, there is a paucity of studies that have investigated sources of variation of milk iodine, especially on a large scale. So far, it is not clear if milk iodine content could be increased through breeding in dairy cattle. Recently, a mid-infrared spectroscopy prediction model has been developed for an indirect quantification of iodine content in cow milk, as it is a faster and less expensive method that allows the prediction at population level. The model has coefficient of determination and ratio of performance to deviation in external validation of 0.57 and 1.44, respectively, and it was used in the present study to predict the iodine content from historical milk spectral data to investigate phenotypic and genetic aspects in the Italian Holstein cattle. Based on the accuracy of the model, the prediction was interpreted as proxy for the real milk iodine concentration (IOD P ). The data set comprised 33,776 test-day records with IOD P from 4,072 cows. Data of IOD P were transformed through natural logarithm to achieve a normal distribution. The effect of parity, lactation stage, and month of sampling were investigated, and genetic parameters were estimated using a test-day repeatability animal model. Milk IOD P decreased with parities and was the lowest in early lactation. Heritability of IOD P was low (0.025) and it was positively genetically correlated with milk yield and negatively with fat content. Results suggested that it would be challenging to directly improve this trait through breeding strategies in dairy cattle, because IOD P is mainly affected by temporary environmental factors and thus, cannot be easily improved through genetics. Although preliminary, findings of this study suggest that it would be more convenient to develop feeding and management strategies to drive milk iodine level than to put efforts and resources into breeding strategies. The identification of functional genetic variants and associated candidate genes linked to feed efficiency may help improve selection for feed efficiency in dairy cattle, providing economic and environmental benefits for the dairy industry. This study used RNA-sequencing data obtained from liver tissue from 9 Holstein cows [n = 5 low residual feed intake (RFI), n = 4 high RFI] and 10 Jersey cows (n = 5 low RFI, n = 5 high RFI), which were selected from a single population of 200 animals. Using RNA-sequencing, 3 analyses were performed to identify: (1) variants within low or high RFI Holstein cattle; (2) variants within low or high RFI Jersey cattle; and (3) variants within low or high RFI groups, which are common across both Holstein and Jersey cattle breeds. From each analysis, all variants were filtered for moderate, modifier, or high functional effect, and co-localized quantitative trait loci (QTL) classes, enriched biological processes, and co-localized genes related to these variants, were identified. The overlapping of the resulting genes co-localized with functional SNP from each analysis in both breeds for low or high RFI groups were compared. For the first two analyses, the total number of candidate genes associated with moderate, modifier, or high functional effect variants fixed within low or high RFI groups were 2,810 and 3,390 for Holstein and Jersey breeds, respectively. The major QTL classes co-localized with these variants included milk and reproduction QTL for the Holstein breed, and milk, production, and reproduction QTL for the Jersey breed. For the third analysis, the common variants across both Holstein and Jersey breeds, uniquely fixed within low or high RFI groups were identified, revealing a total of 86,209 and 111,126 functional variants in low and high RFI groups, respectively. Across all 3 analyses for low and high RFI cattle, 12 and 31 co-localized genes were overlapping, respectively. Among the overlapping genes across breeds, 9 were commonly detected in both the low and high RFI groups (INSRR, CSK, DYNC1H1, GAB1, KAT2B, RXRA, SHC1, TRRAP, PIK3CB), which are known to play a key role in the regulation of biological processes that have high metabolic demand and are related to cell growth and regeneration, metabolism, and immune function. The genes identified and their associated functional variants may serve as candidate genetic markers and can be implemented into breeding programs to help improve the selection for feed efficiency in dairy cattle., (The Authors. With the rapid development of dairy industry, the breeding process of dairy cows has been accelerated. In previous genome-wide association studies (GWAS), a large number of genetic markers have been reported which may contribute to the selection of Holstein populations with superior milk-producing traits, but they remain to be further verified before practical application. In this study, 90 single nucleotide polymorphisms (SNPs) were selected, which were reported to be significantly associated with five milk production traits, including 305-day milk yield (305MY), 305-day milk fat percent (305FC), 305-day milk protein percent (305PC), 305-day milk fat yield (305FY) and 305-day milk protein yield (305PY). Effective 305-day data and fresh DNA samples were obtained from 295 healthy cows with gestational age of 1-4. Matrix-assisted laser desorption/ionization-time of flight (MALDI-TOF) was used to perform precise genotyping of these loci, followed by site association and haplotype analysis. Results showed that 36 out of 90 loci were supported to be used as genetic markers. In particular, several novel and effective haplotypes were also presented. Overall, our results verified tens of useful markers and provided a basis for further development of breeding strategies.

The Azores archipelago is the most suitable region for dairy production in Portugal, representing 30% of the overall Portuguese dairy production. It has a production system characterized by an average milk yield of 6216 kg/cow/year, and the predominance of pasture-based feeding and cows that have longer productive lives and lower incidence of metabolic/production diseases, such as acidosis or mastitis. The biggest problem with the Azores Islands dairy sector is the cost of transport, as the main markets are located in continental Portugal, over 1500 km away, and local dairy products have to compete with dairy products produced in mainland Portugal and in the rest of the European Union. Herein, the evolution of the dairy sector in the Azores Islands from 2007 to 2017 is presented. A SWOT (Strength, Weakness, Opportunity, and Threat) analysis was performed to find potential solutions to increase the value of the Azorean dairy sector. The most relevant solution considered was the valorization of the dairy production through three major aspects: higher milk quality, namely, better organoleptic properties; lower carbon footprint (not considering transport costs); and higher levels of animal welfare. Three examples are shown of such valorization: protected denomination of origin (PDO) cheeses, the "happy cows" program, and the production of an organic milk, from the Terceira Island. Some of these programs are relatively recent, so, it will be interesting to see how their sales and acceptance by consumers evolve, particularly under the current economic framework.

Milk urea nitrogen (MUN) content is closely related to blood urea nitrogen (BUN) concentration and reflects the balance of dietary crude protein and energy supply for ruminal microbial metabolism. The present study investigated whether the higher MUN content in Brown Swiss (BSW) compared with Holstein (HOL) cows is related to milk production and feeding conditions. Therefore, we evaluated test-day records of three consecutive official milk recordings from 86 BSW and 200 HOL cows kept on the same farm under identical feeding and management conditions. In addition, the relationships between BUN and MUN assessed either by an enzymatic method or by mid-infrared spectroscopy were analysed. For the second objective, milk and blood samples (1,112 samples each) were collected in parallel from day 5 until day 150 of lactation from 141 dairy cows housed at different farms. Concentrations of MUN determined either enzymatically (MUN ENZ ) or by Fourier transform infrared spectroscopy (FTIR; MUN IR ) were regressed on the BUN concentrations of the concomitantly obtained blood samples. Both MUN IR and MUN ENZ were well correlated with BUN (r = .93 and .89 respectively). Concentration of MUN was higher in BSW compared with HOL independently of lactational stage or parity number (p < .0001). Protein and fat content were higher in BSW than in HOL (p < .05). Primiparous cows had lower milk yield and ECM (p < .001 for BSW, p < .0001 for HOL) than multiparous in both breeds. Multiparous BSW had a similar milk production than primiparous HOL (p = .13). In conclusion, BSW cows have higher MUN than HOL when kept under identical feeding and management conditions. The objectives of this study were (1) to evaluate the computational feasibility of the multitrait test-day single-step SNP-BLUP (ssSNP-BLUP) model using phenotypic records of genotyped and nongenotyped animals, and (2) to compare accuracies (coefficient of determination; R 2 ) and bias of genomic estimated breeding values (GEBV) and de-regressed proofs as response variables in 3 Australian dairy cattle breeds (i.e., Holstein, Jersey, and Red breeds). Additive genomic random regression coefficients for milk, fat, protein yield and somatic cell score were predicted in the first, second, and third lactation. The predicted coefficients were used to derive 305-d GEBV and were compared with the traditional parent averages obtained from a BLUP model without genomic information. Cow fertility traits were evaluated from the 5-trait repeatability model (i.e., calving interval, days from calving to first service, pregnancy diagnosis, first service nonreturn rate, and lactation length). The de-regressed proofs were only for calving interval. Our results showed that ssSNP-BLUP using multitrait test-day model increased reliability and reduced bias of breeding values of young animals when compared with parent average from traditional BLUP in Australian Holsten, Jersey, and Red breeds. The use of a custom selection of approximately 46,000 SNP (custom XT SNP list) increased the reliability of GEBV compared with the results obtained using the commercial Illumina 50K chip (Illumina, San Diego, CA). The use of the second preconditioner substantially improved the convergence rate of the preconditioned conjugate gradient method, but further work is needed to improve the efficiency of the computation of the Kronecker matrix product by vector. Records of methane emissions from 1,501 cows on 14 commercial farms in 4 regions of Spain were collected from May 2018 to June 2019. Methane concentrations (MeC) were measured using a nondispersive infrared methane detector installed within the feed bin of the automatic milking system during 14- to 21-d periods. Rumination time (RT; min/d) was collected using collars with a tag that registered time (minutes) spent eating and ruminating. The means of MeC and methane production (MeP) were 1,254.28 ppm and 182.49 g/d, respectively; mean RT was 473.38 min/d. Variance components for MeC, MeP, and RT were estimated with REML using pedigree and genomic information in a single-step model. Heritabilities for MeC and MeP were 0.11 and 0.12, respectively. Rumination time showed a slightly larger heritability estimate (0.17). The genetic correlation between MeP and MeC was high (>0.95), suggesting that selection on either trait would lead to a positive correlated response on the other. Negative correlations were estimated between RT and MeC (-0.24 ± 0.38) and MeP (-0.43 ± 0.35). Methane concentration and MeP had slightly positive correlations with milk yield (0.17 ± 0.39 and 0.21 ± 0.36), protein percentage (0.08 ± 0.32 and 0.30 ± 0.45), protein yield (0.22 ± 0.41 and 0.31 ± 0.35), fat percentage (0.02 ± 0.40 and 0.27 ± 0.36), and fat yield (0.27 ± 0.28 and 0.29 ± 0.28) from bivariate analyses. Rumination time had positive correlations with milk yield (0.41 ± 0.75) and protein yield (0.26 ± 0.57) and negative correlations with fat yield (-0.45 ± 0.32), protein percentage (-0.15 ± 0.38), and fat percentage (-0.40 ± 0.47). A positive approximated genetic correlation was estimated between fertility and MeC (0.10 ± 0.05) and MeP (0.18 ± 0.05), resulting in slightly higher CH 4 production when selecting for better fertility [days open estimated breeding values (EBV) are expressed with mean 100 and SD 10, inversely related to days from calving to conception; that is, greater days open EBV implies better fertility]. Positive correlations were also estimated for stature with MeC and MeP (0.30 ± 0.04 and 0.43 ± 0.04, respectively). Other type traits (chest width, udder depth, angularity, and capacity) were positively correlated with methane traits, possibly because of higher milk yield and higher feed intake from these animals. Rumination time showed positive EBV correlations with production traits and type traits, and negative correlations with somatic cell count and body condition score. Based on the genetic correlations and heritabilities estimated in this study, methane is measurable and heritable, and estimates of genetic correlations suggest no strong opposition to current breeding objectives in Spanish Holsteins., (Copyright © 2020 American Dairy Science Association. The objective of this study was to analyze the impact of incorporating enteric methane into the breeding objective of dairy cattle in Spain, and to evaluate both genetic and economic response of traits in the selection index under 4 scenarios: (1) the current ICO (Spanish total merit index), used as benchmark; (2) a hypothetical penalization of methane emissions through a carbon tax; (3) considering methane as a net energy loss for the animal; and (4) desired genetic response to reduce methane production by 20% in 10 yr. A bio-economic model was developed to derive the economic values for production and methane traits in each scenario. The estimated economic values for methane were estimated at -€1.21/kg and -€0.32/kg for scenarios 2 and 3, respectively. When merged with other traits in the selection index, methane had less economic importance (1-5%) than milk protein yield (39-42%) or milk fat yield (27-28%). Under these scenarios, selection resulted in an unfavorable response in methane emissions when it was included with an economic weight, with an increase in methane estimated from 0.52 to 0.60 kg/cow per year. Small differences in total profit per cow per year were observed between indices. The incorporation of methane production into the breeding objective had a negligible effect on production, with minor reductions in the expected genetic gain for fat and protein yields and in total economic benefits. However, total methane emissions in the dairy industry in Spain were estimated to decrease between 2 and 5% in the next 10 yr due to positive genetic trends for milk yield and an expected decrease in the total number of dairy cows. Additionally, methane intensity per 1 billion liters of milk would decrease in all scenarios. The uncertainty in the genetic parameters of methane and in carbon prices were tested in a sensitivity analysis, resulting in small deviations from the benchmark scenario. A major effect was observed only under the desired genetic response scenario. In this case, it was possible to achieve a 20% reduction of methane production in 10 yr via selective breeding but at the expense of a larger ad hoc weight (33%) of methane in the selection index and decelerating the genetic gain for production traits from 6 to 18%. The aim of this study was to identify genomic regions associated with 305-day milk yield and lactation curve parameters on primiparous (n = 9,910) and multiparous (n = 11,158) Holstein cows. The SNP solutions were estimated using a weighted single-step genomic BLUP approach and imputed high-density panel (777k) genotypes. The proportion of genetic variance explained by windows of 50 consecutive SNP (with an average of 165 Kb) was calculated, and regions that accounted for more than 0.50% of the variance were used to search for candidate genes. Estimated heritabilities were 0.37, 0.34, 0.17, 0.12, 0.30 and 0.19, respectively, for 305-day milk yield, peak yield, peak time, ramp, scale and decay for primiparous cows. Genetic correlations of 305-day milk yield with peak yield, peak time, ramp, scale and decay in primiparous cows were 0.99, 0.63, 0.20, 0.97 and -0.52, respectively. The results identified three windows on BTA14 associated with 305-day milk yield and the parameters of lactation curve in primi- and multiparous cows. Previously proposed candidate genes for milk yield supported by this work include GRINA, CYHR1, FOXH1, TONSL, PPP1R16A, ARHGAP39, MAF1, OPLAH and MROH1, whereas newly identified candidate genes are MIR2308, ZNF7, ZNF34, SLURP1, MAFA and KIFC2 (BTA14). Fatty acids (FA) have been related to effects on human health, sensory quality and shelf life of dairy products, cow's health and methane emission. However, despite their importance, they are not regularly measured in all dairy herds yet, which can affect the accuracy of estimated breeding values (EBV) for these traits. In this case, an alternative is to use genomic selection. Thus, the aim was to assess the use of genomic information in the genetic evaluation for milk traits in a tropical Holstein population. Monthly records (n = 36,457) of milk FA percentage, daily milk yield and quality traits from 4,203 cows as well as the genotypes of 755 of these cows for 57,368 single nucleotide polymorphisms (SNP) were used. Polygenic and genomic-polygenic models were applied for EBV prediction, and both models were compared through the EBV accuracy calculated from the prediction error and Spearman's correlation among EBV rankings. Prediction accuracy was assessed by using cross-validation. In this case, the accuracy was the correlation between the genomic breeding values (GEBV) obtained as the sum of SNP effects and the EBV obtained in the polygenic model in each validation group. For all traits, the use of the genomic-polygenic model did not alter the animals' ranking, with correlations higher than 0.87. Nevertheless, through this model, the accuracy increased from 1.5% to 6.8% compared to the polygenic model. The correlations between GEBV and EBV varied from 0.52 to 0.68. Improving feed efficiency (FE) is a major goal for the livestock industry. Previously, we have identified 48 SNP markers distributed over 32 genes significantly associated with residual feed intake (RFI) in Israeli Holstein male calves, the most significant of which are located in the bovine FABP4 gene. In the present study, we tested associations of eight of the FABP4 markers with RFI and feed conversion ratio (FCR), along with milk composition and feeding behavioral traits, in 114 lactating Israeli Holstein cows. Large allele effects were found, along with large contributions of FABP4 markers to the phenotypic variation [mean contribution of all significant markers ( P < 0.05), 15.4 and 12.0% for RFI and FCR, respectively] and genotypic variation [means of all significant markers ( P < 0.05), 75.7 and 32.4% in RFI and FCR, respectively]. However, the association of all significant FABP4 markers with FE and milk content traits was found in opposite directions, such that improved FE was accompanied by decreased milk content. Hence, before inclusion in breeding programs, the gain in FE must be economically balanced with the loss in milk contents. On the other hand, these findings imply that in any current improvement program concentrated on milk traits alone, without taking into account the effect on FE, the progress in milk composition is probably accompanied by deterioration of FE. These results, if confirmed in other populations and breeds, set FABP4 as a prime candidate in any marker-assisted selection program targeting FE as a whole and RFI in particular.

At a global level, dairy cow production systems (DCPS) are important sources of nourishment and profits, but they generate environmental impacts such as overexploitation of different resources including water, lands and fossil energy. Quantification of water and carbon footprint to define mitigation strategies and a more rational use of natural resources, is a reiterated claim. The aim of this study was to perform an economic evaluation of the environmental impact of the DCPS from the Comarca Lagunera, Mexico (24°N, 102°W, 220 mm, hot-semiarid climate) We contrasted the economic value (EV) generated by the DCPS with respect to the economic costs (EC) due to the greenhouse gas emissions (GHGE) and the water footprint (WFP) of this DCPS. While quantifications of GHGE considered those proposed by the Intergovernmental Panel on Climate Change, the WFP involved the use of blue, gray and green water by the DCPS and related activities. Quantification of the EC of WFP considered an international average price of water. In the year 2017, the Comarca Lagunera registered a dairy cow inventory of 493 144 heads, with 227 142 lactating cows, which produced 2386 million liters of milk per year with an annual average EV of €525.3 million. The EC (€, millions) generated by the GHGE and WFP were €311.8 and €11 980.7, respectively, with a total EC of € 12 292.5 million. When the EV of milk production and the total environmental EC are compared, the contrast demonstrates not only the noteworthy environmental impact but also the significant and senseless biological and EC. In addition, having a large dairy cow concentration creates pollution concerns and the DCPS transfers both nutrients and water resources from an ecologically vulnerable arid region. Therefore, some mitigation strategies such as, better cow genotype, feed and manure management combined with the production of forages and grains in a different geographical region are suggested to promote an optimum use of water in order to uphold the social, economic and biologic sustainability of the Comarca Lagunera, Mexico.

Non-perforating abomasal lesions are common in Danish Holstein cows, but the impact on production has not been studied. Our objective was to compare milk yield, carcass weight, days to first service and initiation of breeding among cows with different non- perforating abomasal lesion types and locations to cows with no such lesions. The occurrence, type and localisation of non- perforating abomasal lesions were characterised at slaughter in Danish Holstein cows, and first insemination dates (n = 592), milk yield (n = 999) and carcass information (n = 1,317) was also available. Production and reproduction data were retrieved from all other cows from the herds of origin. This information was used to evaluate the expected production performance in the herds. Of the 1,317 cows investigated at slaughter, 572, 428, 677, and 91 cows had at least one lesion Subtype 1a (mucosal erosion), 1b (acute deeper lesion), 1c (chronic deeper lesion) or 1d (scarring following a lesion), respectively. Cows with lesion Subtype 1c and lesions in the pyloric part of the abomasum were generally found to be culled later in lactation than their herd mates. Milk yield and days to first service were not associated with any lesion types. The occurrence of lesion Subtype 1c and lesions in the pylorus were associated with a higher weight at slaughter compared to cows without lesions of this subtype or in this location. Most Subtype 1c lesions was found in the pylorus. Initiation of breeding before 150 days in milk was positively associated with the number of lesions in the corpus of the abomasum. We discovered two sources of selection bias. First, cows with lesions had a higher carcass weight but also remained in the herds for longer after calving than cows without lesions. This would give them more time to overcome the period of negative energy balance in early lactation. Second, although the causes of culling were not known, cows with no lesions generally had a relatively low milk yield and could have been culled for this reason.". While non- perforating abomasal ulcers may have an effect on the production and reproduction data, the observational data presented show no evidence of such a relationship. It may be necessary to employ other study designs to identify any effects that might exist, but if the relationship cannot be demonstrated in a dataset of this magnitude, it is not deemed to be of practical relevance., (Copyright © 2019 Elsevier B.V. We used daily records from provincial Japanese weather stations and monthly test-day records of milk production to investigate the length of the lags in the responses of cows' milk yield and somatic cell score (SCS) to heat stress (HS). We also investigated the HS thresholds in milk yield and SCS. Data were a total of 17,245,709 test-day records for milk and SCS in Holstein cows that had calved for the first time between 2000 and 2015, along with weather records from 60 weather stations. Temperature-humidity index (THI) values were estimated by using average daily temperature and average daily relative humidity. Adjusted THI values were calculated by using temperature, relative humidity, wind speed, and solar radiation. The model contained herd, calving year, month of test day, age group, days in milk, and THI as a fixed effect. THIs for each day from 14 days before the test day until the test day were used to represent the HS effects. The HS occurring 3 days, and between 8 and 10 days, before the test day had the greatest effect on the milk yield and SCS, respectively. The procedure used for the genetic evaluation of dairy cattle in Japan has developed from a lactation sire-MGS model to a multiple-lactation random regression test-day animal model. Genetic evaluation of Holstein bulls in Japan began in 1989 with the use of field-style progeny testing; dairy herd improvement program data from all over Japan were used, along with a sire and maternal grandsire model. In 1993, an animal model was introduced to estimate breeding values for yield and type traits. A random regression test-day model was first applied in 2010. In the business of breeding dairy cattle, it is very important to users that estimated breeding values are reliable and stable among subsequent routine evaluations. With experience in the genetic evaluation of dairy cattle in Japan, Japanese researchers have found ways to improve the stability of estimated breeding values. These modifications involve changes in data editing, development of evaluation models, changes to the structures of unknown-parent groups, awareness of the problems of predicting lactation yield from partial test-day records, and adjustment for heterogeneity within herd variances. The aim of this study was to evaluate the effects of treating Holstein cows with pegbovigrastim on periparturient diseases, milk production, and reproductive performance while exploring the mode of action of an immunomodulatory protein. Cows were randomly allocated to 1 of 2 treatments, untreated control (CTR, n = 423) and pegbovigrastim (PEG, n = 417). At 7 d from the anticipated calving date (d -7), cows allocated to PEG received a subcutaneous injection of 15 mg of pegylated recombinant bovine granulocyte colony stimulating factor (pegbovigrastim injection, Imrestor, Elanco Animal Health, Greenfield, IN). A second injection was administered within 24 h after calving (d 0). Blood samples were obtained from a subset of cows (CTR, n = 103; PEG, n = 102) at -7 and 0, 3, 7, and 14 d relative to parturition. Samples were used for hemogram and quantification of haptoglobin, nonesterified fatty acids, β-hydroxybutyrate, and trace and macro minerals. Vaginal cytobrush was performed on the same subset cows at d 0, 7, and 14 to assess the relative neutrophil count. Additionally, colostrum samples were collected to measure IgG, IgM, IgA, and lactoferrin concentrations. Postpartum disease occurrence was recorded from calving until 30 d in milk (DIM). Weekly milk yield was recorded for the first 12 wk after calving. Cows treated with PEG had a 3- to 4-fold increase in circulating polymorphonuclear leukocyte, lymphocyte, and monocyte numbers, with a peak at 3 d after treatment followed by a gradual decline, but the counts remained significantly greater compared with CTR at 14 DIM. The administration of PEG did not affect the incidence of clinical and subclinical mastitis, retained fetal membranes, metritis, puerperal metritis, and endometritis. Primiparous cows treated with PEG tended to have lower odds of developing hyperketonemia than CTR [odds ratio (OR) = 0.57, 95% confidence interval (CI) = 0.23 to 1.42]. Cows treated with PEG had higher odds of being diagnosed with lameness within 30 DIM compared with CTR (OR = 1.79, 95% CI = 1.16 to 2.76); however, we found no significant differences by 60 DIM. Treatment with PEG increased the odds of displaced abomasum (OR = 8.27, 95% CI = 1.02 to 66.6). Cows treated with PEG had higher odds of being diagnosed with 1 or more clinical diseases compared with CTR cows (OR = 1.39, 95% CI = 1.02 to 1.90). We observed no differences in linear scores or milk composition between treatments. Furthermore, primiparous cows treated with PEG produced more milk than CTR primiparous cows during the first 12 wk postpartum (PEG = 37.51 ± 0.66; CTR = 35.91 ± 0.65 kg), but no differences were observed on energy-corrected milk. Treatment did not alter reproductive performance; additionally, cows diagnosed with metritis or puerperal metritis and treated with PEG tended to have higher proportion of neutrophils in the vaginal mucosa when compared with CTR metritic cows. Although PEG treatment increased circulating polymorphonuclear leukocyte, monocyte, and lymphocyte numbers, as expected, it was detrimental to cow health because it increased morbidity., (Copyright © 2018 American Dairy Science Association. As a result of the increase in herd size and the intensification of production, the complexity of reproductive management has been growing in dairy herds. The aim of our study was to examine the associations of management practices and reproductive performance in Holstein cows on large commercial dairy farms. Management practices applied to cows were surveyed between 22 May and 6 November 2015 in 34 large Holstein-Friesian dairy herds in Hungary. Individual data of 23 784 cows that calved between 1 January 2014 and 31 December 2014 in the surveyed herds were gathered. Associations between the management practices and the reproductive parameters were analyzed by mixed effects models. Regarding heat abatement we found that ventilation with sprinklers was associated with the shortest breeding interval (P0.05) results to lack of heat stress protection. It was also revealed, that lack of a well-established voluntary waiting period (VWP) or a VWP shorter than 50 days was associated with less days to first service (P<0.01), shorter breeding interval (P<0.01) and calving to conception interval (P<0.05), as well as higher odds of carrying a calf by 200 days in milk (P<0.01) compared with those using a VWP of at least 50 days. Those farms that applied transrectal ultrasonography were more likely to use ventilation with sprinklers (P<0.05), hormonal synchronization (P<0.01) and to perform early pregnancy diagnosis followed by pregnancy recheck (P<0.05). The application of transrectal ultrasonography with one of the aforementioned practices was associated with reduced days to first service (P<0.05), shorter breeding interval (P<0.05) and higher odds of pregnancy at 200 days in milk (P<0.05). Our study highlights the management practices most closely related to improved reproductive performance, which are, therefore, suggested to be applied on dairy farms, considering the local circumstances of the individual farms.

Milk yield (MY), composition, and fatty acid profiles of purebred Holstein (HO) cows, Brown Swiss (BS), and their F 1 crossbreds (HS) were compared under subtropical climate in Egypt. Pure HO had significantly greater 305-MY, total-MY, and daily-MY (p < 0.05) than pure BS and crossbred HS. Furthermore, HO and HS had significantly higher peak-MY (44.2 and 43.3 kg, respectively) than BS (36.1 kg). In comparison with HO, BS had significantly higher milk fat, protein, total solids, and solid-not-fat % (p < 0.05); however, no significant differences were observed between BS and HS for milk fat %. The milk fat of BS had higher concentrations of saturated (C17:0 and C18:0) fatty acids (FA) than that of the HO and HS (p = 0.001 and 0.008, respectively). Content of C4:0, C6:0, C8:0, C12:0, and C15.0 FA did not differ between genotypes. Milk from HO and BS had significantly higher concentrations of unsaturated (C20:1 and C20:5) FA than that from HS (p < 0.05). C14:1 (myristoleic), C16:1 (palmitoleic), and C18:2 (linoleic) FA contents were similar for all genotypes. In conclusion, BS surpassed HO for all milk composition traits and some FA components; however, HS had comparable milk fat percentage with BS. There appears to be an opportunity to modify the concentration of certain FA by breeding.

As ruminants are able to digest fibre efficiently and assuming that competition for feed v. food use would intensify in the future, cereals and other field crops should primarily be destined to cover the dietary needs of humans and monogastric animals such as poultry and pigs. Farming systems with a reduced or absent concentrate supplementation, as postulated by organic agriculture associations, require adapted dairy cows. The aim of this experiment was to examine the impact of concentrate supplementation on milk production, grazing and rumination behaviour, feed intake, physical activity and blood traits with two Holstein-Friesian cow strains and to conclude the consequences for sustainable and organic farming. The experiment was a cross-over study and took place on an organic farm in Switzerland. In all, 12 Swiss Holstein-Friesian (HCH) cows and 12 New Zealand Holstein-Friesian (HNZ) cows, which were paired according to lactation number, days in milk and age for primiparous cows, were used. All cows grazed full time and were supplemented either with 6 kg/day of a commercial, organic cereal-grain mix or received no supplement. After an adaptation period of 21 days, a measurement period of 7 days followed, where milk yield and composition, pasture dry matter intake estimated with the n-alkane double-indicator technique, physical activity based on pedometer measurements, grazing behaviour recorded by automatic jaw movement recorder and blood samples were investigated. Non-supplemented cows had a lower milk yield and supplemented HCH cows produced more milk than supplemented HNZ cows. Grazing time and physical activity were greater for non-supplemented cows. Supplementation had no effect on rumination behaviour, but HNZ cows spent longer ruminating compared with HCH cows. Pasture dry matter intake decreased with the concentrate supplementation. Results of blood analysis did not indicate a strong negative energy balance for either non-supplemented or supplemented cows. Minor differences between cow strains in this short-term study indicated that both cow strains are equally suited for an organic pasture-based production system with no concentrate supplementation. Many factors such as milk yield potential, animal welfare and health, efficiency, grazing behaviour and social aspects influence the decision to supplement grazing dairy cows with concentrates.

Mastitis, the most common and expensive disease in dairy cows, implies significant losses in the dairy industry worldwide. Many efforts have been made to improve genetic mastitis resistance in dairy populations, but low heritability of this trait made this process not as effective as desired. The purpose of this study was to identify genomic regions explaining genetic variation of somatic cell count using copy number variations (CNVs) as markers in the Holstein population, genotyped with the Illumina BovineHD BeadChip. We found 24 and 47 copy number variation regions significantly associated with estimated breeding values for somatic cell score (SCS&#95;EBVs) using SVS 8.3.1 and PennCNV-CNVRuler software, respectively. The association analysis performed with these two software allowed the identification of 18 candidate genes (TERT, NOTCH1, SLC6A3, CLPTM1L, PPARα, BCL-2, ABO, VAV2, CACNA1S, TRAF2, RELA, ELF3, DBH, CDK5, NF2, FASN, EWSR1 and MAP3K11) that result classified in the same functional cluster. These genes are also part of two gene networks, whose genes share the 'stress', 'cell death', 'inflammation' and 'immune response' GO terms. There is renewed interest in dairy cow crossbreeding in Ireland as a means to further augment productivity and profitability. The objective of the present study was to compare milk production and fertility performance for Holstein, Friesian, and Jersey purebred cows, and their respective crosses in 40 Irish spring-calving commercial dairy herds from the years 2008 to 2012. Data on 24,279 lactations from 11,808 cows were available. The relationship between breed proportion, as well as heterosis and recombination coefficients with performance, was quantified within a mixed model framework that also contained the fixed effects of parity; cow and contemporary group of herd-year-season of calving were both included as random effects in the mixed model. Breed proportion was associated with all milk production parameters investigated. Milk yield was greatest for Holstein (5,217kg), intermediate for Friesian (4,591kg), and least for Jersey (4,230kg), whereas milk constituents (i.e., fat and protein concentration) were greatest for Jersey (9.38%), intermediate for Friesian (7.91%), and least for Holstein (7.75%). Yield of milk solids in crossbred cows exceeded their respective parental average performance; greatest milk solids yield (i.e., fat kg + protein kg) was observed in the Holstein × Jersey first-cross, yielding 25kg more than the mid-parent mean. There was no consistent breed effect on the reproductive traits investigated. Relative to the mid-parent mean, Holstein × Jersey cows calved younger as heifers and had a shorter calving interval. Friesian × Jersey first-cross cows also had a shorter calving interval relative to their mid-parent mean. Results were consistent with findings from smaller-scale controlled experiments. Breed complementarity and heterosis attainable from crossbreeding resulted in superior animal performance and, consequently, greater expected profitability in crossbred cows compared with their respective purebreds., (Copyright © 2016 American Dairy Science Association. Reaction norm models have been widely used to study genotype by environment interaction (G × E) in animal breeding. The objective of this study was to describe environmental sensitivity across first lactation in Brazilian Holstein cows using a reaction norm approach. A total of 50,168 individual monthly test day (TD) milk yields (10 test days) from 7476 complete first lactations of Holstein cattle were analyzed. The statistical models for all traits (10 TDs and for 305-day milk yield) included the fixed effects of contemporary group, age of cow (linear and quadratic effects), and days in milk (linear effect), except for 305-day milk yield. A hierarchical reaction norm model (HRNM) based on the unknown covariate was used. The present study showed the presence of G × E in milk yield across first lactation of Holstein cows. The variation in the heritability estimates implies differences in the response to selection depending on the environment where the animals of this population are evaluated. In the average environment, the heritabilities for all traits were rather similar, in range from 0.02 to 0.63. The scaling effect of G × E predominated throughout most of lactation. Particularly during the first 2 months of lactation, G × E caused reranking of breeding values. It is therefore important to include the environmental sensitivity of animals according to the phase of lactation in the genetic evaluations of Holstein cattle in tropical environments.

Twenty lactating dairy cattle were intravenously infused with either lipopolysaccharide (LPS) (n = 10) or sterile saline (n = 10). Five cattle in each group received three doses of flunixin meglumine administered by either intravenous infusion or intramuscular injection at 24 h intervals. Milk, urine, and tissues were collected. Thirty-six hours after the last flunixin administration, milk from six cows contained 5-hydroxyflunixin (5OHF) levels greater than the milk tolerance of 2 ng/mL; by 48 h, milk from two cows, a saline and a LPS-treated animal, had violative milk concentrations of 5OHF. A single animal treated with LPS and intramuscular flunixin contained violative flunixin residues in liver. The ratio of urinary flunixin/5OHF was correlated (P < 0.01; R(2) = 0.946) with liver flunixin residues in LPS-treated animals, but not (P = 0.96; R(2) = 0.003) in cows treated with saline in lieu of LPS. Violative residues of flunixin in dairy cattle may be related to LPS inhibition of flunixin metabolism.

The objective of this paper was to compare efficiency measures, milk production, and feed intake for lactating cows in commercial herds using different breeds and production and milking systems. To accomplish this, we used all feed evaluations made by the Danish extension service during the period November 2012 to April 2013 for 779 herds, of which 508 were Holstein-Friesian (HOL); 100 were Jersey (JER); and 171 herds were a mixture of these 2 breeds, other dairy breeds, and crossbreeds (OTH). The annually recorded, herd-average energy-corrected milk (ECM) yield was 8,716kg (JER) and 9,606kg (HOL); and average herd size was 197 cows (HOL) and 224 cows (JER). All cows were fed a total mixed or partial mixed ration supplemented with concentrate from feeding stations, housed in loose housing systems with a slatted floor, and milked in either a parlor milking unit or an automatic milking system. Energy efficiency was calculated as net energy efficiency defined as total energy demand as a percentage of energy intake and as residual feed intake defined as energy intake (net energy for lactation; NEL) minus energy requirement. Production efficiency was expressed as kilograms of ECM per kilogram of dry matter intake (DMI), kilograms of ECM per 10 MJ of net energy intake (NEL), kilograms of ECM per 100kg of BW, and kilograms of DMI per 100kg of BW. Environmental efficiency was expressed by the nitrogen efficiency calculated as N in milk and meat as a percentage of N in intake, and as enteric emission of methane expressed as kilograms of ECM per megajoule of CH4. Mean milk yield for lactating cows was 30.4kg of ECM in HOL and 3kg less in JER, with OTH herds in between. Mean NEL intake was 122 MJ in JER, increasing to 147 MJ in HOL, whereas ration energy density between breeds did not differ (6.4-6.5 MJ of NEL per kg of DMI). The NEL intake and DMI explained 56 and 47%, respectively, of variation in production (ECM) for HOL herds but only 44 and 27% for JER. Jersey had a higher efficiency than HOL and OTH, except in nitrogen efficiency, where no significant difference between breeds existed. Most of the efficiency measures were internally significantly correlated and in general highly positively correlated with milk production, whereas the correlation to DMI was less positive and for JER negative for net energy efficiency, kilograms of ECM per kilogram of DMI, and nitrogen efficiency. Only little of the variation in efficiency between herds could be explained by differences in nutrient or roughage content of DMI. Unlabelled: Isolation of Staphylococcus aureus (Staph. aureus) from Holstein milk samples with mastitis and nonmastitis was conducted to estimate its prevalence, antimicrobial resistance and toxin genes. A total of 353 milk samples were collected from three Chinese Holstein herds. Fifty-three Staph. aureus isolates collected from 29 Staph. aureus-positive samples were characterized via antimicrobial susceptibility, toxin genes and Pulsed-field Gel Electrophoresis (PFGE) profiles. The prevalence of Staph. aureus was 4·0-9·5% in mastitic and 7·3-11·5% in nonmastitic samples in the analysed herds. Approximately 61·0% of Staph. aureus strains isolated from mastitis cows were resistant to ≥10 antimicrobials compared with 0% of isolates with nonmastitis. The most frequently observed super antigenic toxin gene was pvl (41·5%) followed by seh + pvl (13·2%). We did not find mecA-positive methicillin-resistant Staph. aureus (MRSA) strains, while mecA-negative MRSA strains were identified in the three herds. PFGE results suggested potential transmission of Staph. aureus strains in different farms. These results open new insights into Staph. aureus transmission and antimicrobial resistance of Holstein dairy cows and into developing strategies for udder health improvement of dairy cattle., Significance and Impact of the Study: Significantly higher levels of antibiotic resistance of Staphylococcus aureus strains were detected in the lactating Holstein milk with clinical mastitis compared with that without clinical mastitis. PFGE results suggest potential transmission of Staph. aureus strains in different dairy farms. Holstein is known to provide higher milk yields than most other cattle breeds, and the dominant position of Holstein today is the result of various selection pressures. Holstein cattle have undergone intensive selection for milk production in recent decades, which has left genome-wide footprints of domestication. To further characterize the bovine genome, we performed whole-genome resequencing analysis of 10 Holstein and 11 Hanwoo cattle to identify regions containing genes as outliers in Holstein, including CSN1S1, CSN2, CSN3, and KIT whose products are likely involved in the yield and proteins of milk and their distinctive black-and-white markings. The objective of this study was to compare calving traits, BCS, milk production, fertility, and survival of Holstein-Friesian (HF) and Norwegian Red (NR) dairy cattle in moderate-concentrate input systems. The experiment was conducted on 19 commercial Northern Ireland dairy farms, and involved 221 HF cows and 221 NR cows. Cows completed 5 lactations during the experiment, unless they died or were culled or sold. Norwegian Red cows had a lower calving difficulty score than HF cows when calving for the first and second time, but not for the third and fourth time. At first calving, the incidence of stillbirths for NR cows was 4%, compared with 13% for HF cows, whereas no difference existed between breeds in the proportion of calves born alive when calving for the second time. When calving for the first time, NR cows had a poorer milking temperament than HF cows, whereas milking temperament was unaffected by breed following the second calving. Holstein-Friesian cows had a higher full-lactation milk yield than NR cows, whereas NR cows produced milk with a higher milk fat and protein content. Full-lactation fat + protein yield was unaffected by genotype. Norwegian Red cows had a lower somatic cell score than HF cows during all lactations. Although NR cattle had a higher BCS than the HF cows during lactations 1 and 2, no evidence existed that the 2 genotypes either lost or gained body condition at different rates. Conception rates to first artificial insemination were higher with the NR cows during lactations 1 to 4 (57.8 vs. 40.9%, respectively), with 28.5% of HF cows and 11.8% of NR cows culled as infertile before lactation 6. A greater percentage of NR cows calved for a sixth time compared with HF cows (27.2 vs. 16.3%, respectively). Simple Summary: This study assessed the supplementation of two limiting milk production amino acids for dairy cows (methionine and lysine) on the lactational performance of fall-calving grazing dairy cows from Chile. Four groups were compared and 26 cows per group were randomly assigned to the following groups: (i) a control group without supplementation; (ii) a lysine group; (iii) a methionine group; and (iv) a lysine plus methionine group. Supplementation was conducted from 2 to 70 days postpartum. The group supplemented with both amino acids produced more milk protein and fat than the control group. In conclusion, the supplementation of methionine and lysine improved milk protein and fat yield in grazing dairy cows with fall parturitions; therefore, they can be used as a strategy to improve the performance of lactation in cows. The aim of this study was to evaluate the effect of the supplementation of rumen-protected (RP) methionine and lysine on milk yield, solids, and body weight over time on fall-calving grazing multiparous Holstein cows from Chile. Four treatment groups were studied and compared for the outcomes over time. The treatments were as follows: (i) CON: control (n = 26); (ii) RP lysine group (LYS; 20 g per cow per day; n = 26); (iii) RP methionine group (MET; 20 g per cow per day; n = 26); and (iv) LYS × MET (RP lysine and RP methionine 20 g of each amino acid per cow per day; n = 26). Data were analyzed with general linear mixed model ANOVAs for repeated measures to primarily test the main effects of each amino acid and their interactions. The supplementation of the amino acids was conducted from 2 to 70 days postpartum. Overall, milk production tended to be higher in the MET and in the LYS × MET group when compared to the control group. Furthermore, CON produced significantly less milk protein (kg) and milk fat (kg) than the LYS, MET, and LYS × MET groups. Milk urea tended to be lower during the entire study in the CON group than the rest of the groups. There was a trend for a reduction in the losses of postpartum body weight in the LYS × MET than the CON. In conclusion, RP methionine and lysine improved milk fat and protein yield in grazing multiparous cows with fall parturitions; consequently, both RP amino acids can be used as a strategy for improving grazing cows' production performance. In the present socio-economic context and environmental sustainability trend it is a high accent on milk production related with increased milk yield per cow. This study aims to explore the influence of milking technology for Holstein dairy cows on "milk" productivity and quality, at the farm level. Over three consecutive years, the dynamics of milk quantity and its quality parameters (% fat, % protein, somatic cells count, % dry matter) were followed. The values of these parameters changed as a result of the increase of milkings frequency per day, from two in the first year of study, to three milkings per day in the following years. The results were statistically analyzed, from the point of view of significance, using the Fisher and Student tests. The comparative analysis of the 3 years of production shows that milk production had a positive evolution, both in terms of quantity and quality. [ABSTRACT FROM AUTHOR]

Simple Summary: Animal breeding is one of the most profitable human activities. The ultimate goal is to join genetically high performance and good health. In the last decades, the study of genetic control of important traits on the molecular level has been launched. In the study of milk performance, major genes, namely those for milk proteins, are the focus of much research. Here, we studied the polymorphisms of CSN1S1 and LALBA genes and their associations with milk performance and quality. Also, other non-genetic effects were evaluated. The polymorphisms influenced milk, fat, and protein yield percentages in the second lactation but their effects in the first lactation were not significant. The technological properties of milk were not influenced. The effect of the farm was significant in most cases. Breed influenced milk performance but did not influence technological properties. Changes in lactose content reflect the health of the udder. We evaluated the influence of CSN1S1 and LALBA polymorphisms on cow's milk yield and quality. The analysis was done on Czech Simmental and Holstein cows. Non-genetic factors were included as well. CSN1S1 did not influence the milk performance in the first lactation. In the second lactation, cows with the BB genotype had significantly higher milk, protein, and fat yields than BC. The differences between LALBA genotypes were non-significant in the first lactation, while in the second lactation, the fat percentage was significantly higher in BB than in AB. The farm significantly influenced milk, protein, and fat yields in both the first and second lactations and fat percentage in the first lactation. The effect of CSN1S1 and LALBA genotypes on the milk technological quality was non-significant. Breed did not influence any of the evaluated technological traits and SCS. The ethanol test was not influenced by farm, season, lactation phase, protein percentage, breed, or non-fat solids percentage. Farm, season, and protein percentage significantly influenced milk fermentation ability, renneting, and SCS. The lactose content is a good indicator of udder health. [ABSTRACT FROM AUTHOR]

Simple Summary: Health treatments of Holsteins were recorded over a 7-year period and partitioned into five categories (mastitis, reproduction, lameness, metabolic, and miscellaneous). The fixed cost of health treatments was obtained from the veterinary clinics that serviced the eight herds, and they were assigned to each observation. The health treatment cost of cows was highest during the first 30 days in milk for all parities and ranged from USD 22.87 in first parity to USD 38.50 in fifth parity. The total health treatment cost in first parity ranged from USD 23.38 to USD 74.60 for the eight herds and usually increased with parity. Health treatments of Holstein cows (n = 2214) were recorded by the owners of eight high-performance dairy herds in Minnesota. Cows calved from March 2008 to October 2015, and 14 types of health treatments were uniformly defined across the herds. Specific types of health treatment were subsequently assigned a cost based on the mean veterinary cost obtained from the veterinary clinics that serviced the eight herds. A fixed labor cost for time (USD 18/h) associated with specific types of health treatment was determined based on interviews with the herd owners and was added to the veterinary cost. Health treatment cost was then partitioned into five health categories: mastitis (including mastitis diagnostic test), reproduction (cystic ovary, retained placenta, and metritis), lameness (hoof treatments), metabolic (milk fever, displaced abomasum, ketosis, and digestive), and miscellaneous (respiratory, injury, and other). Lactations of cows were divided into six intervals that corresponded with stage of lactation based on days in milk. The first interval of lactation was 30 days in length, followed by four intervals of 60 days each, and the final interval started on day 271 and had variable length because it continued to the end of lactation and included the dry period. Health treatment cost was summed within each interval of lactation and subsequently across lactations by parity. Statistical analysis by parity included the fixed effects of herd, interval, and the interaction of herd and interval, with interval regarded as a repeated measure of cows. Health treatment cost was highest during the first interval for all five parities of cows and ranged from USD 22.87 for first parity to USD 38.50 for fifth parity. Reproduction treatment cost was about one-half of the total health treatment cost during the first interval in all five parities. Metabolic treatment cost during the first interval ranged from USD 3.92 (in first parity) to USD 12.34 (in third parity). Compared to the other health categories, mastitis treatment cost was most evenly distributed across intervals of lactation in all parities. Lameness treatment cost was highest during mid- or late-lactation across parities and reflected the time when cows received routine hoof trimming. The aim of this study was to evaluate the effect of the supplementation of rumen-protected (RP) methionine and lysine on milk yield, solids, and body weight over time on fall-calving grazing multiparous Holstein cows from Chile. Four treatment groups were studied and compared for the outcomes over time. The treatments were as follows: (i) CON: control (n = 26); (ii) RP lysine group (LYS; 20 g per cow per day; n = 26); (iii) RP methionine group (MET; 20 g per cow per day; n = 26); and (iv) LYS × MET (RP lysine and RP methionine 20 g of each amino acid per cow per day; n = 26). Data were analyzed with general linear mixed model ANOVAs for repeated measures to primarily test the main effects of each amino acid and their interactions. The supplementation of the amino acids was conducted from 2 to 70 days postpartum. Overall, milk production tended to be higher in the MET and in the LYS × MET group when compared to the control group. Furthermore, CON produced significantly less milk protein (kg) and milk fat (kg) than the LYS, MET, and LYS × MET groups. Milk urea tended to be lower during the entire study in the CON group than the rest of the groups. There was a trend for a reduction in the losses of postpartum body weight in the LYS × MET than the CON. In conclusion, RP methionine and lysine improved milk fat and protein yield in grazing multiparous cows with fall parturitions; consequently, both RP amino acids can be used as a strategy for improving grazing cows’ production performance.

The aim of the study was to analyse beta-hydroxybutyric acid concentration in the blood (BBAC) and milk (MBAC) at postpartum week 2 (PPW2) and week 4 (PPW4) in Holstein (n=216, 8 farms), Montbeliard (n=23), Simmental (n=38) and Holstein/Montbeliard Crossbreed (n=23, BBAC only). Furthermore, the prevalence of subclinical ketosis (SCK) and its association with postpartum health disorders (PPHD), body condition score (BCS) and average daily milk yield in 90 days in milk (90DIM) were evaluated in Holstein. Holstein-Crossbreed and Montbeliard had significantly lower postpartum BBAC than others. Primiparous Montbeliard and Holstein had significantly higher MBAC at PPW2 than PPW4. Cows having BCS2 and 4 at calving had higher MBAC at PPW2 and 4 that was associated significantly with metritis and multiple diseases. Holstein with BCS4 at calving had higher BBAC at PPW2 and 4. SCK prevalence in the blood (BBAC≥1.2 mmol/L, BSCK) and milk (MBAC=100 µmol/L, MSCK1), (MBAC≥200 µmol/L, MSCK2) and (MBAC≥100 µmol/L, MSCK1/2) was 8.3, 11.8, 5.8 and 17.3% at PPW2 and 4.7, 4.9, 6.9 and 11.9% at PPW4 in Holstein respectively. Holstein with SCK was more likely to develop PPHD in 90DIM. MSCK1 and MSCK1/2 did not associate milk production loss in Holstein. Holstein with BSCK and MSCK2 at PPW2 had a 6.7 kg loss of average daily milk yield in 90DIM. Culling rate was 3.7% in Holstein and MSCK2 positive Holstein at PPW2 was significantly more likely to be culled (25%, Odds:11.2, P<0.05) in 90DIM. In conclusion, cows having BCS2 and 4 at calving had higher MBAC and BBAC at PPW2 and 4. Holstein-Crossbreed and Montbeliard had much lower postpartum BBAC than Holstein and Simmental. The aim of this study was to determine the difference between melatonin hormone in night milk and melatonin concentration in day milk of Holstein cows. In this study, daytime milk obtained from 40 heads Holstein cows in the first lactation raised in a private dairy cattle farm in Karapınar district of Konya state province in Turkey and night milk samples taken from the same cows that were kept in darkness for one week were used. The milk samples were collected from day and night milking in order to determine the melatonin concentration with the help of Bovine Melatonin (MLT) Elisa Kit. In this study, melatonin concentration in day time and night time milk were determined as respectively 103.70 ± 6.61 pg mL-1 and 163.13 ± 8.96 pg mL-1. The difference between melatonin levels of day and night milk was statistically significant (P<0.01). Since significant difference occurred between melatonin concentration between day and night milk at the end of the study, it can be stated that night milk can be used for medical purposes that is used cure of some illnesses and producers can be provided with a new production source.
