We previously showed that adult Paragonimus westermani, the causative agent of paragonimiasis and whose habitat is the host lung, possesses both aerobic and anaerobic respiratory chains, i.e., cyanide-sensitive succinate oxidase and NADH-fumarate reductase systems, in isolated mitochondria (Takamiya et al., 1994). This finding raises the intriguing question as to whether adult Paragonimus worms possess two different populations of mitochondria, one having an aerobic succinate oxidase system and the other an anaerobic fumarate reductase system, or whether the worms possess a single population of mitochondria possessing both respiratory chains (i.e., mixed-functional mitochondria). Staining of trematode tissues for cytochrome c oxidase activity showed three types of mitochondrial populations: small, strongly stained mitochondria with many cristae, localised in the tegument and tegumental cells; and two larger parenchymal cell mitochondria, one with developed cristae and the other with few cristae. The tegumental and parenchymal mitochondria could be separated by isopycnic density-gradient centrifugation and showed different morphological characteristics and respiratory activities, with low-density tegumental mitochondria having cytochrome c oxidase activity and high-density parenchymal mitochondria having fumarate reductase activity. These results indicate that Paragonimus worms possess three different populations of mitochondria, which are distributed throughout trematode tissues and function facultatively, rather than having mixed-functional mitochondria., (Copyright © 2010 Australian Society for Parasitology Inc. The mitochondrion and the apicoplast of the malaria parasite, Plasmodium spp. is microscopically observed in a close proximity to each other. In this study, we tested the suitability of two different separation techniques--Percoll density gradient centrifugation and fluorescence-activated organelle sorting--for improving the purity of mitochondria isolated from the crude organelle preparation of Plasmodium falciparum. To our surprise, the apicoplast was inseparable from the plasmodial mitochondrion by each method. This implies these two plasmodial organelles are bound each other. This is the first experimental evidence of a physical binding between the two organelles in Plasmodium.

Parasites have exploited unique energy metabolic pathways as adaptations to the natural host habitat. In fact, the respiratory systems of parasites typically show greater diversity in electron transfer pathways than do those of host animals. These unique aspects of parasite mitochondria and related enzymes may represent promising targets for chemotherapy. Natural products have been recognized as a source of the candidates of the specific inhibitors for such parasite respiratory chains. Chalcones was recently evaluated for its antimalarial activity in vitro and in vivo. However, its target is still unclear in malaria parasites. In this study, we investigated that licochalcone A inhibited the bc1 complex (ubiquinol-cytochrome c reductase) as well as complex II (succinate ubiquinone reductase, SQR) of Plasmodium falciparum mitochondria. In particular, licochalcone A inhibits bc1 complex activity at very low concentrations. Because the property of the P. falciparum bc1 complex is different from that of the mammalian host, chalcones would be a promising candidate for a new antimalarial drug.

We recently reported that Ascaris suum mitochondria express stage-specific isoforms of complex II: the flavoprotein subunit and the small subunit of cytochrome b (CybS) of the larval complex II differ from those of adult enzyme, while two complex IIs share a common iron-sulfur cluster subunit (Ip). In the present study, A. suum larval complex II was highly purified to characterize the larval cytochrome b subunits in more detail. Peptide mass fingerprinting and N-terminal amino acid sequencing showed that the larval and adult cytochrome b (CybL) proteins are identical. In contrast, cDNA sequences revealed that the small subunit of larval cytochrome b (CybS(L)) is distinct from the adult CybS (CybS(A)). Furthermore, Northern analysis and immunoblotting showed stage-specific expression of CybS(L) and CybS(A) in larval and adult mitochondria, respectively. Enzymatic assays revealed that the ratio of rhodoquinol-fumarate reductase (RQFR) to succinate-ubiquinone reductase (SQR) activities and the K(m) values for quinones are almost identical for the adult and larval complex IIs, but that the fumarate reductase (FRD) activity is higher for the adult form than for the larval form. These results indicate that the adult and larval A. suum complex IIs have different properties than the complex II of the mammalian host and that the larval complex II is able to function as a RQFR. Such RQFR activity of the larval complex II would be essential for rapid adaptation to the dramatic change of oxygen availability during infection of the host.

Parasites have developed a variety of physiological functions necessary for their survival within the specialized environment of the host. Using metabolic systems that are very different from those of the host, they can adapt to low oxygen tension present within the host animals. Most parasites do not use the oxygen available within the host to generate ATP, but rather employ anaerobic metabolic pathways. In addition, all parasites have a life cycle. In many cases, the parasite employs aerobic metabolism during its free-living stage outside the host. In such systems, parasite mitochondria play diverse roles. In particular, marked changes in the morphology and components of the mitochondria during the life cycle are very interesting elements of biological processes such as developmental control and environmental adaptation. Recent research on the respiratory chain of the parasitic helminth Ascaris suum has shown that the mitochondrial NADH-fumarate reductase system plays an important role in the anaerobic energy metabolism of adult parasites inhabiting hosts, as well as describing unique features of the developmental changes that occur during its life cycle.

Using N2 cavitation, we established a protocol to prepare the active mitochondria from Plasmodium falciparum showing a higher succinate dehydrogenase activity than previously reported and a dihydroorotate-dependent respiration. The fact that fumarate partially inhibited the dihydroorotate dependent respiration suggests that complex II (succinate-ubiquinone reductase/quinol-fumarate reductase) in the erythrocytic stage cells of P. falciparum functions as a quinol-fumarate reductase.

The respiratory chain of Caenorhabditis elegans was characterized in mitochondria isolated from aerobically grown nematodes. Nematode mitochondria contain ubiquinone-9 as a major component and rhodoquinone-9 as a minor component. The ratio of ubiquinone-9/rhodoquinone-9 is higher in C. elegans mitochondria than in mitochondria from second-stage larvae of Ascaris suum, the free-living stage of porcine gut-dwelling nematode. The individual oxidoreductase activities comprising succinate oxidase and the amount of substrate-reducible cytochromes are comparable to those of mitochondria from second-stage larvae of A. suum. The specific activity of fumarate reductase is lower in C. elegans mitochondria than in mitochondria from second-stage larvae of A. suum, but still higher than in mammalian mitochondria. These results indicate that the free-living nematode C. elegans is capable of synthesizing rhodoquinone, as distinguished from aerobic mammalian species, although its mitochondria appear more aerobic than A. suum larval mitochondria., (Copyright 1999 Academic Press.)

Parasitic helminths exhibit greater diversity in energy metabolism than do the host animals and many have exploited unique respiratory chains as adaptations to their natural habitats. Cytochromes are involved, not only in intracellular aerobic respiration found in free-living stages, but also in the reduction of relatively oxidized compounds such as fumarate during the adult stages of parasitic helminths. In addition, most helminths retain a significant capacity to produce energy via aerobic pathways and have a mammalian type respiratory chain in their mitochondria during their development in the host. In this review, we focus on recent advances in the study of cytochromes in the respiratory chain of parasitic helminths. These include the identification of unique features of anaerobic respiration in adult parasites, the elucidation of molecular structures of the components involved and an understanding of the developmental changes that occur during the life-cycle of these parasites.

Morphology and respiratory function were studied in situ and in the isolated mitochondria of Paragonimus ohirai. Two types of parenchymal cells (i.e., Pc1 and Pc2 cells), whose mitochondria differ in terms of morphology and staining for cytochrome c oxidase activity, were found in fluke tissues. Enzymatic and spectrophotometric analyses of the isolated mitochondria showed that fluke mitochondria possess both aerobic and anaerobic respiratory chains. These results suggest that there are two mitochondrial populations in fluke parenchymal cells, one possessing an aerobic respiratory chain and the other an anaerobic respiratory chain.

Cytochrome c oxidase in the mitochondria of the tegument and tegumental and parenchymal cells was examined cytochemically in Echinostoma trivolvis, Zygocotyle lunata, Schistosoma mansoni, Fasciola gigantica and Paragonimus ohirai, trematodes that inhabit different sites in their vertebrate hosts. Clear differences in enzyme activity occurred in the mitochondria of these species, probably reflecting the different energy metabolisms of these worms. Marked aerobic metabolism occurred in S. mansoni and P. ohirai adults that inhabit the host mesenteric veins and the lungs, respectively. The tegument and parenchymal cells of S. mansoni possess relatively few, small mitochondria with tabular cristae which are heavily reactive for cytochrome c oxidase. In P. ohirai, the activity for cytochrome c oxidase in tegumental mitochondria increased gradually from juveniles to adults, reflecting that the respiratory activity increased with growth and the aerobic metabolism is activated when the worms reach the lung. P. ohirai juveniles and adults had two types of mitochondria with different shapes and enzyme activities that were located in two different types of parenchymal cells. The intestinal species, E. trivolvis had mitochondria in the basal aspect of the tegument, and some variations in enzyme activity of their mitochondria in the tegumental and parenchymal cells were observed, suggesting that they possess both aerobic and anaerobic metabolic systems. Z. lunata that live in rodent caeca are devoid of mitochondria in the tegument and have many characteristic mitochondria with undeveloped cristae in the parenchymal cells. Mitochondria of F. gigantica showed weak or no activity for cytochrome c oxidase, suggesting that the worm is well-adapted to an anaerobic environment in the host bile duct.

Mitochondrial complex II functions as a fumarate reductase (FRD), the reverse reaction of succinate dehydrogenase (SDH), and plays an important role in the anaerobic respiratory chain of parasitic helminths. In this study, complex II from the dog heartworm, Dirofilaria immitis adult, which is thought to act as a homolactatic fermenter, was examined in terms of its enzymatic features and primary structure in order to investigate the possible role of mitochondria in this filaria. Mitochondria from D. immitis adult showed high FRD activity when the enzymatic assay was performed using methylviologen as an artificial electron donor. The ratio of SDH to FRD in D. immitis was comparable to that in Ascaris suum adult, which is known to have an anaerobic mitochondrial respiratory chain with a high FRD activity of complex II. The FRD activity of D. immitis mitochondria was inhibited by the sulfhydryl reagent N-ethylmaleimide (NEM), while that of A. suum complex II was resistant to this inhibitor. The presence of the flavoprotein (Fp) subunit, which contains the substrate binding active site, was confirmed in D. immitis mitochondria by immunoblotting using a monoclonal antibody against the A. suum Fp subunit. By homology probing with the polymerase chain reaction, the entire cDNA for the D. immitis adult Fp was cloned and sequenced. The deduced amino acid sequence showed significant homology to that of A. suum and other mitochondrial Fps, in contrast to much less similarity to bacterial FRD, even though the D. immitis complex II showed high FRD activity. These results are the first indication of the presence of a functional complex II in D. immitis mitochondria.

Complex II from mitochondria of the adult parasitic nematode, Ascaris suum, exhibits high fumarate reductase activity and plays a key role in the anaerobic electron transport observed in these organelles. In contrast, mitochondria isolated from free living second stage larvae (L2) of A. suum show much lower fumarate reductase activity than those from adults, whereas succinate dehydrogenase activities of mitochondria in both stages are comparable. In the present study, biochemical and antigenic properties of the partially purified enzymes from both larval and adult mitochondria were compared. Larval complex II eluted from the DEAE-Cellulofine column chromatography at a lower salt concentration than adult enzyme, whereas the apparent molecular size of both enzyme complexes estimated by gel permeation column chromatography was the same. The fumarate reductase activity of larval complex II was less than 3% of that of adult enzyme, and the Km values for substrates were significantly different between the two complexes. The flavoprotein subunit of larval complex II could be distinguished from that of adult complex II by two-dimensional gel electrophoresis and peptide mapping. The antibody against the smallest subunit (small subunit of cytochrome b558) of the adult enzyme did not cross-react with that of the larval enzyme. These results suggest that larval complex II differs from adult enzyme and is more similar to aerobic mammalian enzymes with low fumarate reductase activity. This is the first direct indication of the two different stage-specific forms of mitochondrial complex II.

By fractionation using polyacrylamide gel electrophoresis and/or a preparative hybrid selection method employing solid-phase DNA probes, we prepared and characterized mitochondrial tRNAs from the body wall muscle of Ascaris suum, all of which are thought to lack either the T stem or the D stem from their gene sequences (Okimoto, R., and Wolstenholme, D. R. (1990) EMBO J. 10, 3405-3411). Some of the partially purified tRNAs were appreciably aminoacylated with an extract of A. suum mitochondria. The three species sequenced had CCA sequence at their 3'-ends, and tRNA(Met) had 5-formylcytidine at the anticodon first position, a new modified nucleoside found at the same position of bovine mitochondrial tRNA(Met) (Moriya, J., Yokogawa, T., Wakita, K., Ueda, T., Nishikawa, K., Crain, P. F., Hashizume, T., Pomerantz, S. C., McCloskey, J. A., Kawai, G., Hayashi, N., Yokoyama, S., and Watanabe, K. (1994) Biochemistry 33, 2234-2239). Enzymatic probing of these tRNAs supported the secondary structural model proposed by Okimoto and Wolstenholme in the reference cited above. Chemical probing of tRNA(Phe) demonstrated the existence of tertiary interactions between the (T arm-variable loop)-replacement loop and the D arm. The results suggest that these tertiary interactions enable the bizarre tRNAs of nematode mitochondria to maintain an L-shape-like structure in order to function in the nematode mitochondrial translation system.

The respiratory chain of adult Paragonimus westermani, a lung fluke, was characterized in isolated mitochondria. The fluke mitochondria exhibited cyanide- and antimycin A-sensitive succinate oxidase activity at a rate of 16.8 nmol O2 min-1 mg-1 protein. The succinate oxidation was shown to be stimulated by ADP and linked to the formation of membrane potential. The specific activities of oxidoreductases composing the succinate oxidase system, i.e., succinate-ubiquinone and succinate--cytochrome c oxidoreductase (complex II and complex II-III, respectively) and cytochrome c oxidase (complex IV), were compared in mitochondria from adult Paragonimus, bovine heart (an aerobic tissue), and muscle of adult Ascaris suum which possesses an anaerobic respiratory chain. The activity values of complex II-III and complex IV were high, middle, and low for bovine heart, Paragonimus, and A. suum, respectively, whereas the activity of complex II was comparable among the three sources. The cytochrome contents of Paragonimus mitochondria as determined by difference absorption spectrophotometry ranged between those in Ascaris and bovine mitochondria for types c and aa3 cytochromes. Paragonimus mitochondria exhibited a high activity of NADH-fumarate reductase; the specific activity was about 18-fold higher in fluke submitochondria than in bovine heart submitochondria. Quinone analysis by HPLC and mass spectrometry showed that the fluke mitochondria contain both rhodoquinone-10 and ubiquinone-10 at concentrations of 0.572 and 0.321 nmol mg-1 mitochondrial protein, respectively. These data clearly show that mitochondria from adult P. westermani, unlike adult Ascaris mitochondria, possess both cyanide-sensitive succinate oxidase and NADH-fumarate reductase systems, indicating that the fluke mitochondria are facultatively anaerobic.

The flavoprotein (Fp) subunit of mitochondrial complex II contains covalently bound FAD as a prosthetic group. In this study, the primary structure of the flavin-bound tryptic peptide from the Fp subunit of Ascaris complex II was determined and found to be highly similar to those of the corresponding flavin-binding regions of bovine heart and bacterial Fp subunits. Furthermore, the Ascaris Fp subunit was shown to contain two regions exhibiting striking sequence similarity to the segments that have been predicted to interact noncovalently with the AMP moiety of FAD in bacterial Fp subunits. The conservation of these two regions also in the mitochondrial Fp subunit suggests their functional importance.

The effects of tetragalloylglucose (1,2,3,6-tetra-O-galloyl-beta-D-glucose) on purified complex II (succinate-ubiquinone oxidoreductase) of the mitochondrial electron transport system of Ascaris muscle were studied. Both succinate-ubiquinone-1 (Q1) oxidoreductase, and succinate dehydrogenase measured with 3-(4,5-dimethylthiazol-2-yl)- 2,5-diphenyltetrazolium bromide (MTT) in the presence of phenazine methosulfate (PMS) were inhibited by tetragalloylglucose. The inhibitions of both reductase activities of complex II were of competitive type, and the inhibitor constant (Ki) for Ascaris complex II (148 nM) was lower than that for rat liver complex II (1.5 microM). Thus, Ascaris complex II is much more sensitive to this inhibitor than the mammalian counterpart.

Complex II of the anaerobic respiratory chain in Ascaris muscle mitochondria showed a high fumarate reductase activity when reduced methyl viologen was used as the electron donor. The maximum activity was 49 mumol/min per mg protein, which is much higher than that of the mammalian counterpart. The mitochondria of Ascaris-fertilized eggs, which require oxygen for its development, also showed fumarate reductase activity with a specific activity intermediate between those of adult Ascaris and mammals. Antibody against the Ascaris flavoprotein subunit reacted with the mammalian counterparts, whereas those against the Ascaris iron-sulfur protein subunit did not crossreact, although the amino acid compositions of the subunits in Ascaris and bovine heart were quite similar. Cytochrome b-558 of Ascaris complex II was separated from flavoprotein and iron-sulphur protein subunits by high performance liquid chromatography with a gel permeation system in the presence of Sarkosyl. Isolated cytochrome b-558 is composed of two hydrophobic polypeptides with molecular masses of 17.2 and 12.5 kDa determined by gradient gel, which correspond to the two small subunits of complex II. Amino acid compositions of these small subunits showed little similarity with those of cytochrome b-560 of bovine heart complex II. NADH-fumarate reductase, which is the final enzyme complex in the anaerobic respiratory chain in Ascaris, was reconstituted with bovine heart complex I, Ascaris complex II and phospholipids. The maximum activity was 430 nmol/min per mg protein of complex II. Rhodoquinone was essential for this reconstitution, whereas ubiquinone showed no effect. The results clearly indicate the unique role of Ascaris complex II as fumarate reductase and the indispensability of rhodoquinone as the low-potential electron carrier in the NADH-fumarate reductase system.

Immunoblotting studies on mitochondria prepared from the striata of patients who died of Parkinson's disease were performed using specific antisera against Complexes I, III and IV. In 4 out of 5 patients with Parkinson's disease, the 30-, 25- and 24-kDa subunits of Complex I were moderately to markedly decreased. No clear difference was noted in immunoblotting studies on subunits of Complexes III and IV between the control and Parkinson's disease. Deficiencies in Complex I subunits seem to be one of the most important clues to elucidate pathogenesis of Parkinson's disease.
