Determining surface topography of different tissues of the molar tooth with novel analytical methods has opened new horizons in dental surface measurements which characterize tooth surface quality in dentistry. Studying surface topological measurements and comparing surface morphology of hard tissue of the molar tooth are the ultimate goals of the present study. Ten molar teeth have been chosen for investigating their surface characteristics through image processing techniques. The power spectral density (PSD) and fast Fourier transform algorithms of every molar tooth containing enamel, dentin, and cementum have determined that the characterization of surface profiles is possible. As can be seen, PSD along with fractal dimensions leads to good results for teeth surface topography. The functional monomer 10-methacryloyloxydecyl dihydrogen phosphate (10-MDP), used in many dental adhesives, self-assembles in nano-layers at adhesive-tooth interfaces. Recently, several states of the POH groups of 10-MDP_Ca salts were suggested, while their actual status has not been elucidated yet. We mechanistically investigated 10-MDP_Ca-salt nano-layering at adhesive-dentin interfaces, correlatively using scanning transmission electron microscopy with energy-dispersive X-ray spectrometry (STEM-EDS), X-ray diffraction (XRD) and solid state nuclear magnetic resonance (NMR). STEM-EDS confirmed the presence of Ca and P in each nano-layer. Both XRD and NMR revealed that the two terminal POH groups of 10-MDP reacted with Ca. This twofold POH interaction of 10-MDP with Ca was stable in water and is therefore expected to contribute to durable nano-layering of 10-MDP_Ca salts in the hybrid and adhesive layer. STATEMENT OF SIGNIFICANCE: The functional monomer 10-methacryloyloxydecyl dihydrogen phosphate (10-MDP), commonly used in dental adhesives with favorable long-term clinical outcome, has been documented to self-assemble into nano-layers at adhesive-tooth interfaces. Characterizing ultra-morphologically (STEM) and chemically (STEM-EDS, XRD, NMR) the mechanisms of interaction of 10-MDP with bulk dentin in a similar manner as what occurs clinically, it was found that the water stable 10-MDP_Ca salts consist of CaRPO 4 , meaning that the two OH groups of the phosphate group of 10-MDP ionically reacted with Ca. Teeth are subjected to compressive loads during mastication. Under small loads the soft tissue periodontal ligament (PDL) deforms most. However when the loads increase and the PDL is highly compressed, the tooth and the alveolar bone supporting the tooth, begin to deform. Here we report on the structure of this alveolar bone in the upper furcation region of the first molars of mature minipigs. Using light microscopy and scanning electron microscopy (SEM) of bone cross-sections, we show that this bone is hypermineralized, containing abundant small pores around 1-5 μm in diameter, lacunae around 10-20 μm as well as larger spaces. This bone does not possess the typical lamellar motif or other repeating structures normally found in cortical or trabecular mammalian bone. We also use high resolution focused ion beam scanning electron microscopy (FIB-SEM) in the serial surface mode to image the 3D organization of the demineralized bone matrix. We show that the upper furcation bone matrix has a disordered isotropic structure composed mainly of individual collagen fibrils with no preferred orientation, as well as highly staining material that is probably proteoglycans. Much larger aligned arrays of collagen fibers - presumably Sharpey's fibers - are embedded in this material. This unusual furcation bone material is similar to the disordered material found in human lamellar bone. In the upper furcation region this disordered bone comprises almost all the volume excluding Sharpey's fibers. In this study, natural molar human tooth specimens were investigated for determining their micro- and nanoscale structural morphology, chemistry and crystallinity. The differences were tracked comparatively for both enamel and dentin layers and at their interfaces. Although dental material structures are hard and tough and the cross-sectioning of these materials using mechanical methods is challenging, FIB-SEM dual-beam instruments serve for preparing ultra-thin homogenous lamella sections. In this work, both FIB-SEM and TEM based advanced characterization methods were applied to reveal different morphological characteristics of dental tissue via complementary imaging and diffraction analysis. In addition, SEM-EDS and Raman spectroscopy techniques provided additional information about the elemental distribution and the chemical composition differences of the dental tissues. According to electron microscopy examinations at the intersection between the enamel and the dentin layers, it was shown that the enamel was denser and polycrystalline, while the dentin layer was porous, fibrillar and of negligible long-range order, due to its tubular structure and organic components. In particular, EDS mapping and linescan analyses showed almost no differences in the elemental distribution. Raman results confirmed that both tissues had similar chemical composition except dentin showed spectral background effects in the spectrum due to its tubular structure and organic components., (Copyright © 2018 Elsevier Ltd. This study aims to explore the feeding ecology of two terrestrial papionins, Papio and Theropithecus from the Shungura Formation in Ethiopia, the most complete stratigraphic and paleontological record of the African Plio-Pleistocene. Two aspects were evaluated using Dental Microwear Texture Analysis: differences in diet between the extinct genera and their extant relatives, and any potential dietary fluctuations over time. Amongst more than 2,500 cercopithecid dental remains, 154 Theropithecus molars and 60 Papio molars were considered. Thirty-nine extant wild baboons and 20 wild geladas were also considered. The results show that diets of extinct monkeys from Member G already differed between genera as it is the case for their extant representatives. The shearing facets on the Theropithecus molars display significant variations in microwear textures, suggesting several dietary shifts over time. Two events point to higher intakes of herbaceous monocots (tougher than dicots foliages), at about 2.91 Ma (between members B and C) and at 2.32 Ma (between members E and F). These two events are separated by an inverse trend at about 2.53 Ma (between members C and D). Some of these variations, such as between members E and F are supported by the enamel carbon isotopic composition of herbivorous mammals and with paleovegetation evidence.

The aim of the present study was to identify the stage during acid etching of dental enamel where scratches introduced by sectioning and grinding procedures were eliminated. Nitric acid was used as etchant. Four hundred 2-3 mm-thick longitudinal sections of human third molar crowns were assigned to 80 groups, each group differing with respect to grinding procedure (silicon carbide paper grit 600; 1,000; 1,200; and 1,200 + polishing), acid concentration (0.1%, 1%, 2.5%, and 5%) and etching time (15, 30, 45, 90, and 180 s). Observation of the specimens with scanning electron microscopy (SEM) showed that elimination of scratches was facilitated by increasing fineness of grinding paper and polishing, and by increasing acid concentration and increasing etching time. An acid concentration of 0.1% did not remove scratches totally irrespective of grinding procedure or etching time, 1% acid removed most scratches after 45-90 s, while 2.5% and 5% acids removed most scratches after 30-45 s. It is proposed that the scratches are eliminated/obscured through a combined effect of the acid and the inherent structure of the enamel. The results of the present study may serve as a guide in choosing the best procedures for obtaining scratch-free enamel surfaces through acid etching with the least possible loss of enamel substance. Aims: The structural organization of collagen from mineralized tissues, such as dentin and bone, has been a topic of debate in the recent literature. Recent reports have presented novel interpretations of the complexity of collagen type I at different hierarchical levels and in different tissues. Here, we investigate the nanostructural organization of demineralized dentin collagen following the digestion of non-collagenous components with a trypsin enzyme., Materials and Methods: Dentin specimens were obtained from healthy third-molars, cut into small cubes, and polished down to 1 µm roughness. Samples were then demineralized with 10% citric acid for 2 min. Selected specimens were further treated with a solution containing 1 mg/ml trypsin for 48 hours at 37 °C (pH 7.9-9.0). Both untreated and trypsin digested samples were analyzed using SDS-PAGE, Field Emission Scanning Electron Microscopy (FE-SEM), and nanoindentation, where surface hardness and creep properties were compared before and after treatments., Results: FE-SEM images of demineralized dentin showed the banded morphology of D-periodical collagen type I, which upon enzymatic digestion with trypsin appeared to dissociate longitudinally, consistently unraveling ~20 nm structures (microfibril bundles). Such nanoscale structures, to the best of our knowledge, have not been characterized in dentin previously. Mechanical characterization via nanoindentation showed that the unraveling of such microfibril bundles affected the creep displacement and creep rate of demineralized dentin., Conclusion: In summary, our results provide novel evidence of the organization of collagen type I from dentin, which may have important implications for the interaction of dental materials with the organic dentin matrix and the mechanical properties of mineralized tissues.

In the current study, we evaluate the equivalent stiffness of peritubular reinforcement effect (PRE) of porous dentine optimized by the thickness of peritubular dentine (PTD). Few studies to date have evaluated or quantitated the effect of PRE on composite dentine. The miscrostructure of porous dentine is captured by scanning electron microscope images, and then finite element modeling is used to quantitate the deformation and stiffness of the porous dentine structure. By optimizing the radius of PTD and dentine tubule (DT), the proposed FE model is able to demonstrate the effect of peritubular reinforcement on porous dentine stiffness. It is concluded that the dentinal equivalent stiffness is reduced and degraded with the increase of the radius of DT (i.e., porosity) in the certain ratio value of Ep/Ei and certain radius of PTD, where Ep is the PTD modulus and Ei is the intertubular dentine modulus. So in order to ensure the whole dentinal equivalent stiffness is not loss, the porosity should get some value while the Ep/Ei is certain. Thus, PTD prevents the stress concentration around DTs and reduces the risk of DTs failure. Mechanically, the overall role of PTD appears to enhance the stiffness of the dentine composite structure. These results provide some new and significant insights into the biological evolution of the optimal design for the porous dentine microstructure. These findings on the biological microstructure design of dentine materials are applicable to other engineering structural designs aimed at increasing the overall structural strength.

of this study was to analyze the effects of carbon dioxide (CO2) laser light on the structure and elemental composition of dentine., Methods: The evaluation was conducted on samples from extracted teeth. The surface of the dentine was exposed to the radiation from a CTL 1401 CO2 laser (Centre of Laser Technology, Poland). The radiation and frequency parameters were as follows: group I with 5 W and 1 Hz, group II with 10 W and 1 Hz, group III with 5 W and 5 Hz, and group IV with 10 W and 5 Hz. The altered dentine structure was macroscopically and microscopically evaluated using a Nova NanoSEM 200 Scanning Electron Microscope (FEI Europe) with integrated microanalysis X-ray system for elemental analysis in points., Results: There were significant differences between groups in the macro- and microstructure of laser defects., Conclusions: CO2 laser radiation causes irreversible, destructive changes in dentine. The structural dentine lesions developed under the influence of the CO2 laser radiation may hinder proper adhesion of bonding systems with the damaged tissue. Laser defects in the structure should be treated like defects of noncarious origin requiring preparation and filling with composite materials in accordance with the procedures.

Objective: To compare the effect of different endodontic irrigants on microleakage of adhesives used within pulp chamber of primary molars., Study Design: 72 Primary molars were divided into 6 groups, according to 3 irrigants and 2 adhesives used. After de-roofing the pulp chamber, pulp was extirpated. In 36 samples, pulp chambers were bonded with Xeno V + after irrigation with either QMix (Group1); 17%EDTA+5%NaOCl (Group2) or normal saline (Group3) and in other half samples, pulp chambers were bonded with SinglebondUniversal after irrigation with either QMix (Group 4); 17%EDTA+5% NaOCl (Group5) or normal saline (Group 6). All the samples were restored with Filtek Z350. Ten teeth from each group were assessed for dye penetration. Two samples in each group were viewed under scanning electron microscope. Data was statistically analyzed using Mann-Whitney and Kruskal Wallis tests at a significance level of P < 0.05., Results: Mean microleakage scores were: Group 1-1.5±0.70, Group 2-1.6±.51, Group 3-2.4±0.96, Group 4-1.2±0.42, Group 5-1.2±0.42, Group 6-1.1±0.32., Conclusions: Irrigation with QMix significantly reduced the microleakage of XenoV + but had no significant effect on microleakage of SinglebondUniversal. Irrigation with EDTA/NaOCl or QMix had no detrimental effect on the sealing ability of either of the adhesive tested.

Objective: The present study was designed to elucidate whether three soluble N-ethylmaleimide-sensitive fusion protein attachment protein receptor (SNARE) core proteins, syntaxin-1, synaptosomal-associated protein of 25kDa (SNAP-25), and vesicle-associated membrane protein-2 (VAMP-2), are present in the dental pulp of the rat molar at both the light and electron microscopic levels., Design: Immunohistochemistry for protein gene product 9.5 (PGP 9.5), a pan-neuronal marker, syntaxin-1, SNAP-25, and VAMP-2 was performed on decalcified rat molars for light and electron microscopic analyses. Double-immunolabeling of PGP 9.5 and the SNARE core proteins, as well as combinations of the SNARE core proteins, was also carried out., Results: PGP 9.5-immunoreactive nerve fibers ran toward the coronal region, ramified at the subodontoblast layer, and formed the subodontoblastic nerve plexus. Most nerve fibers penetrated the predentin and dentin along the dentinal tubules. Most, if not all, nerve fibers displayed immunoreactivity for syntaxin-1, SNAP-25, and VAMP-2. Immunoelectron microscopic analyses confirmed the presence of immunoreactivity for the SNARE core proteins within the intradental axonal elements., Conclusions: The present findings suggest that, since SNARE core proteins participate in the docking and exocytosis of synaptic vesicles in the central nervous system, they may contribute to vesicle exocytosis from the dental nerve fibers even though there are no apparent synapses., (Copyright © 2016 Elsevier Ltd. The "lead line" was described by Henry Burton in 1840. Rodents are used as sentinels to monitor environmental pollution, but their teeth have not been used to determine lead. To determine whether lead deposits can be observed in the teeth of lead-exposed animals, since the gingival deposits known as "lead line" would likely have a correlate in the calcified tissue to which the gums are opposed during life. Male Wistar rats were exposed to lead in the drinking water (30 mg/L) since birth until 60 days-old. Molars and the incisors of each hemimandible were analyzed by scanning electron microscopy (SEM) on regular and backscattered electrons (BSE) mode. Elements were determined using electron dispersive spectroscopy (EDS). Clean cervical margins were observed on control teeth, as opposed to the findings of extensive deposits on lead-exposed animals, even in hemimandibles that had been exhumed after being buried for 90 days. BSE/EDS indicated that those deposits were an exogenous material compatible with lead sulfite. Presence of calcium, phosphorus, magnesium, carbon, lead, and oxygen is presented. Lead-exposed animals presented marked root resorption. The lead deposits characterized here for the first time show that the "lead line" seen in gums has a calcified tissue counterpart, that is detectable post-mortem even in animals exposed to a low dose of lead. This is likely a good method to detect undue lead exposure and will likely have wide application for pollution surveillance using sentinels.

The aim of the study was to propose the artefact index on cone-beam computed tomography (CBCT) images of clinical prosthodontics materials, and to compare the effect of the artefacts on CBCT image clarity of normal oral tissues. Seven spheres of different materials were secured on the centre of a resin baseboard, respectively, and four human molars in vitro were placed at 10 mm front, back, left and right of the sphere. The board was scanned using CBCT with the same setting. 10 tomographic images from each of the seven data sets with clear artefacts was selected. The grayscale measuring tool of Photoshop software was used to measure the grayscale (G0) within the boundary of tomographic image and the grayscales of the streaky artefacts that were 1 mm and 20 mm outside the circular boundary (G1 and G2). The arc length, L1, of the circular boundary with artefacts was measured; the circumference, L2, was calculated. The artefact index, A, was determined as (G1/G0) × 0.5 + (G2/G1) × 0.4 + (L2/L1) × 0.1. The artefact index A can comprehensively represent the effect of artefacts on CBCT image clarity for oral tissue.

Matrix extracellular phosphoglycoprotein (MEPE) is an extracellular matrix protein found in dental and skeletal tissues. Although information regarding the role of MEPE in bone and disorders of phosphate metabolism is emerging, the role of MEPE in dental tissues remains unclear. We performed RNA in situ hybridization and immunohistochemistry analyses to delineate the expression pattern of MEPE during embryonic and postnatal development in craniofacial mineralizing tissues. Mepe RNA expression was seen within teeth from cap through root formation in association with odontoblasts and cellular cementoblasts. More intense expression was seen in the alveolar bone within the osteoblasts and osteocytes. MEPE immunohistochemistry showed biphasic dentin staining in incisors and more intense staining in alveolar bone matrix and in forming cartilage. Analysis of Mepe null mouse molars showed overall mineralized tooth volume and density of enamel and dentin comparable with that of wild-type samples. However, Mepe(-/-) molars exhibited increased thickness of predentin, dentin, and enamel over controls and decreased gene expression of Enam, Bsp, Dmp1, Dspp, and Opnby RT-PCR. In vitro Mepe overexpression in odontoblasts led to significant reductions in Dspp reporter activity. Objective: To evaluate the root canal surfaces in teeth previously treated using RF resin by SEM., Material and Methods: 20 distal roots of mandibular molars with typical discoloration of tooth crown were used in this study: 10 brownish - red and 10 non-colored roots. The roots were removed from the crowns, splinted longitudinally and subjected to SEM in a back-scattered electron mode., Results: In discolored root samples canal walls appeared without smear layer and varying amount of remaining debris, the observed morphology of dentine was ordinary. In the non-colored root samples the dentin profile revealed: in coronal third of the root just few open dentinal tubules were detected, while in the middle and apical thirds no open dentinal tubules were observed, dentin profile was similar to sclerotic dentin. The multiple dentinal defects were detected in all samples, however significantly more dentinal defects were found in the colored roots. The RF paste was well adhered to the root canal dentine and multiple tags of RF resin penetrating deeply into the dentinal tubules were detected., Conclusions: The surfaces of the root canal walls of the teeth treated using RF paste exhibits usual appearance and ordinary dentin morphology in colored roots while the dentin of non-colored samples has the profile of sclerotic dentin. Multiple dentinal defects could be determined by damaging effect of RF resin on dentin. The adaptation and adhesion of RF paste to the dentin, capability to penetrate into dentinal tubules is comparable with the modern resin based sealers.

Many types of behavioral and dietary information can be extracted from studies of tooth microwear. Some studies have even been successful at determining the overall directionality of microwear in order to establish gross masticatory movement (Williams et al., 2009, PNAS, 106, 11194-11199). However, microwear has never been successfully visualized in situ in 3 dimensions (3D), visualized virtually and quantified. The ability to accomplish this yields information on exact masticatory movement which can then be used to address any number of eco-biological and physiological questions in extant and extinct organisms. In order to create 3D virtual reality (VR) representation of microwear, fossil molars from the Javanese Sangiran 7 (S7) Homo erectus tooth collection and from historic hunter/gatherer meta-populations were imaged, the microwear in 3 dimensions was extracted, this information was then placed back on VR representations of the molars and quantified. The methodology contained herein demonstrates the efficacy and importance of such a technique in determining gross masticatory movement in fossil and recent hominin molars. This methodology could, in theory, be applied to any organism which produces microwear on its dentition. Objectives: To evaluate the effect of adhesion strategy on the enamel microshear bond strengths (μSBS), etching pattern, and in situ degree of conversion (DC) of seven universal adhesives., Methods: 84 extracted third molars were sectioned in four parts (buccal, lingual, proximal) and divided into 21 groups, according to the combination of the main factors adhesive (AdheSE Universal [ADU], All-Bond Universal [ABU], Clearfil Universal [CFU], Futurabond U [FBU], G-Bond Plus [GBP], Prime&Bond Elect (PBE), and Scotchbond Universal Adhesive [SBU]), and adhesion strategy (etch-and-rinse, active self-etch, and passive self-etch). Specimens were stored in water (37°C/24h) and tested at 1.0mm/min (μSBS). Enamel-resin interfaces were evaluated for DC using micro-Raman spectroscopy. The enamel-etching pattern was evaluated under a field-emission scanning electron microscope (direct and replica techniques). Data were analyzed with two-way ANOVA and Tukey's test (α=0.05)., Results: Active self-etch application increased μSBS and DC for five out of the seven universal adhesives when compared to passive application (p<0.001). A deeper enamel-etching pattern was observed for all universal adhesives in the etch-and-rinse strategy. A slight improvement in etching ability was observed in active self-etch application compared to that of passive self-etch application. Replicas of GBP and PBE applied in active self-etch mode displayed morphological features compatible with water droplets. The DC of GBP and PBE were not affected by the application/strategy mode., Conclusions: In light of the improved performance of universal adhesives when applied actively in SE mode, selective enamel etching with phosphoric acid may not be crucial for their adhesion to enamel., Clinical Significance: The active application of universal adhesives in self-etch mode may be a practical alternative to enamel etching in specific clinical situations., (Copyright © 2015 Elsevier Ltd. Objectives: To evaluate the fracture strength and the mode of failure of endodontically treated molars restored with CAD/CAM overlays with fiber reinforced composite build-up of the pulp chamber., Methods: 40 Devitalized molars were cut over the CEJ and divided into five groups (n=8). The pulp chamber area was filled with: group 0 (control), no resin build-up; group 1, hybrid composite build-up (G-aenial posterior, GC); group 2, as in group 1 but covered with 3 nets of bi-directional E-glass fibers (EverStickNET, Stick Tech Ltd.); group 3, a FRC resin (EverX posterior, GC); group 4, as in group 3 but covered by the bi-directional fibers. The crowns were restored with CAD-CAM composite restorations (Lava Ultimate, 3M ESPE). Maximum fracture loads were recorded in Newton and data were analyzed using Kruskal-Wallis test (p<0.05). Fragments were analyzed using SEM., Results: The mean static loads in Newton were: group 0: 2448 (546); group 1: 2817 (873); group 2: 2128 (952); group 3: 2429 (1091); group 4: 2577 (833). No significant differences were found between the groups (p>0.05). All specimens fractured in a catastrophic way, under the CEJ. The main crack evolved in the corono-apical direction. In groups 2 and 4 secondary fracture paths with apico-coronal direction were detected close to the bi-directional fibers' layer., Conclusions: For the restoration of endodontically treated molars, the incorporation of FRCs did not influence the load-bearing capacity of the tooth-restoration complex. The SEM analysis showed a low ability of the bi-directional fibers net in deviating the fracture but this effect was not sufficient to lead more favorable fracture patterns, over the CEJ., Clinical Significance: The use of FRCs to reinforce the "core" of devitalized molars against vertical fractures under static loads seems useless when the thickness of the CAD/CAM composite overlay restoration is high., (Copyright © 2015 Elsevier Ltd. Paleoanthropologists and vertebrate paleontologists have for decades debated the etiology of tooth wear and its implications for understanding the diets of human ancestors and other extinct mammals. The debate has recently taken a twist, calling into question the efficacy of dental microwear to reveal diet. Some argue that endogenous abrasives in plants (opal phytoliths) are too soft to abrade enamel, and that tooth wear is caused principally by exogenous quartz grit on food. If so, variation in microwear among fossil species may relate more to habitat than diet. This has important implications for paleobiologists because microwear is a common proxy for diets of fossil species. Here we reexamine the notion that particles softer than enamel (e.g., silica phytoliths) do not wear teeth. We scored human enamel using a microfabrication instrument fitted with soft particles (aluminum and brass spheres) and an atomic force microscope (AFM) fitted with silica particles under fixed normal loads, sliding speeds, and spans. Resulting damage was measured by AFM, and morphology and composition of debris were determined by scanning electron microscopy with energy-dispersive X-ray spectroscopy. Enamel chips removed from the surface demonstrate that softer particles produce wear under conditions mimicking chewing. Previous models posited that such particles rub enamel and create ridges alongside indentations without tissue removal. We propose that although these models hold for deformable metal surfaces, enamel works differently. Hydroxyapatite crystallites are "glued" together by proteins, and tissue removal requires only that contact pressure be sufficient to break the bonds holding enamel together.

Background: Mechanical compression of cells during mesenchymal condensation triggers cells to undergo odontogenic differentiation during tooth organ formation in the embryo. However, the mechanism by which cell compaction is stabilized over time to ensure correct organ-specific cell fate switching remains unknown., Results: Here, we show that mesenchymal cell compaction induces accumulation of collagen VI in the extracellular matrix (ECM), which physically stabilizes compressed mesenchymal cell shapes and ensures efficient organ-specific cell fate switching during tooth organ development. Mechanical induction of collagen VI deposition is mediated by signaling through the actin-p38MAPK-SP1 pathway, and the ECM scaffold is stabilized by lysyl oxidase in the condensing mesenchyme. Moreover, perturbation of synthesis or cross-linking of collagen VI alters the size of the condensation in vivo., Conclusions: These findings suggest that the odontogenic differentiation process that is induced by cell compaction during mesenchymal condensation is stabilized and sustained through mechanically regulated production of collagen VI within the mesenchymal ECM., (© 2015 Wiley Periodicals, Inc.)

Objective: Molar-incisor malformation (MIM) is a newly discovered type of dental anomaly that involves a characteristic root malformation of the permanent first molars. The aim of this study was to reveal the microstructure of MIM teeth in order to determine their origin., Study Design: Four MIM teeth were extracted from a 9-year-old girl due to severe mobility. The detailed microstructure of the teeth was determined by examinations with micro-computed tomography (micro-CT), hematoxylin and eosin (H&E) staining, immunohistochemical staining, and scanning electron microscopy to reveal the detailed microstructure., Results: Micro-CT and H&E staining revealed the pulpal floor comprising three layers: upper, middle, and lower. Amorphous hard tissues and hyperactive cells were observed in the middle layer of the pulpal floor, and the cells stained positively for dentin sialoprotein and osteocalcin, but not for collagen XII., Conclusion: The results of the present study imply that MIM-affected molars probably result from inappropriate differentiation of the apical pulp and dental follicle., (Copyright © 2015 Elsevier Inc. The purpose of this study was to investigate the effect of ultrasonic scaling on teeth restored with a light-cured resin. Ultrasonic scaling is a very popular periodontal therapy among dentists, and used for the removal of dental plaque and calculus in order to reduce and eliminate inflammation. Given the fact that most ultrasonic devices are used at high frequencies to perform scaling, undesirable consequences, such as loss of adhesion and increase in surface roughness, may occur in teeth that have been restored with light-cured resins. Electrochemical impedance spectroscopy (EIS) and scanning electron microscopy were used to investigate the effects of ultrasonic treatments at the dental material-hard dental tissue interface. After ultrasonic scaling, EIS measurements were performed on a human tooth that had been restored with a light-cured resin filling. The data were analyzed and the influence of ultrasound was shown after visualization of the hard dental tissues and the dental material as equivalent electrical circuits. The study revealed, through EIS measurements, that ultrasonic scaling affected the resistance of the light-cured resin filling and dentin, whereas the enamel was affected only slightly. To evaluate the preventive effect of Er:YAG laser (EYL) irradiation followed by silver diamine fluoride (SDF) application on dentine with cariogenic biofilm challenge. Twenty-four dentine slices were prepared from extracted sound human third molars. Each slice was cut into four parts for SDF application, followed by EYL irradiation (group SL), SDF application (group S), EYL irradiation (group L) and water (group W). The specimens were subjected to cariogenic biofilm challenge for 12 h, followed by immersion in a buffered remineralising solution containing calcium chloride and sodium hypophosphate for 12 h at 37 °C. Surface morphological changes in the specimens were examined using scanning electronic microscopy. Elemental analysis was performed using energy-dispersive X-ray spectrometry. Micro-mechanical properties were investigated by nano-indentation. The specimen surfaces of groups SL and L showed laser melting contours with narrowed dentinal orifices. Group S showed a partial tubular occlusion. A porous surface was observed in group W, indicating demineralisation. The mean (SD) fluoride weight percentages were 3.93 (0.91), 3.10 (0.61), 0.17 (0.09) and 0.32 (0.07) in groups SL, S, L and W, respectively, (p < 0.001; SL, S > L, W). The mean (SD) micro-hardness values in GPa were 1.84 (0.22), 0.49 (0.13), 0.41 (0.11) and 0.30 (0.06) in groups SL, S, L and W, respectively, (p < 0.001; SL > S > L, W). The mean (SD) elastic moduli in GPa were 75.1 (7.2), 20.0 (1.3), 24.3 (5.2) and 20.2 (2.8) in groups SL, S, L and W, respectively, (p < 0.001; SL > S, L, W). SDF application followed by EYL irradiation on a dentine surface increased its resistance to cariogenic biofilm challenge.

Aims: Presenting a new method for direct, quantitative analysis of enamel surface. Measurement of adhesive remnants and enamel loss resulting from debonding molar tubes., Material and Methods: Buccal surfaces of fifteen extracted human molars were directly scanned with an optic blue-light 3D scanner to the nearest 2 μm. After 20 s etching molar tubes were bonded and after 24 h storing in 0.9% saline - debonded. Then 3D scanning was repeated. Superimposition and comparison were proceeded and shape alterations of the entire objects were analyzed using specialized computer software. Residual adhesive heights as well as enamel loss depths have been obtained for the entire buccal surfaces. Residual adhesive volume and enamel loss volume have been calculated for every tooth., Results: The maximum height of adhesive remaining on enamel surface was 0.76 mm and the volume on particular teeth ranged from 0.047 mm3 to 4.16 mm3. The median adhesive remnant volume was 0.988 mm3. Mean depths of enamel loss for particular teeth ranged from 0.0076 mm to 0.0416 mm. Highest maximum depth of enamel loss was 0.207 mm. Median volume of enamel loss was 0.104 mm3 and maximum volume was 1.484 mm3., Conclusions: Blue-light 3D scanning is able to provide direct precise scans of the enamel surface, which can be superimposed in order to calculate shape alterations. Debonding molar tubes leaves a certain amount of adhesive remnants on the enamel, however the interface fracture pattern varies for particular teeth and areas of enamel loss are present as well.

An increasing body of data supports the concept that early humans ate invertebrate prey items, especially insects, and that insects may have been a substantial protein source. Insects are ubiquitous throughout the landscape and of high nutritional value. Given that all modern apes and many human groups eat insects, it is likely that early hominins did as well. However, it is unknown how much, which type(s), and what foraging strategy was utilized to obtain invertebrate prey. Given that gross masticatory anatomy of early hominins, like modern apes, fails to reflect their insectivorous proclivities, dental microwear analysis might be a useful tool for identifying this behavior in our fossil ancestors. Dental microwear data was collected on three primarily myrmecophagous mammals and compared to the literature on other faunivorous, frugivorous, and folivorous species. Results indicate that myrmecophagous species have comparable pit frequencies with other faunivores and pit frequency generally distinguishes them from folivores but not all frugivores. Additionally, myrmecophagous mammals have unusually high feature densities, like other faunivores, and density is the microwear signal most suggestive of some sort of faunivory., (Copyright © 2014 Elsevier Ltd. Vascularization is essential for organ and tissue development. Teeth develop through interactions between epithelium and mesenchyme. The developing capillaries in the enamel organ, the dental epithelial structure, occur simultaneously by mechanisms of vasculogenesis and angiogenesis at the onset of dentinogenesis. The vascular neoformation in the dental mesenchyme has been reported to start from the cap stage. However, the mechanisms of vascularization in the dental mesenchyme remain unknown. In the hope of understanding the mechanisms of the formation of dental mesenchymal vasculature, mouse lower molar germs from embryonic day (E) 13.5 to E16.5 were processed for immunostaining of CD31 and CD34, terminal deoxynucleotidyltransferase-mediated dUTP-biotin nick end labeling (TUNEL) and transmission electron microscopy (TEM). In addition, the role of apoptosis for the vascularization in dental mesenchyme was examined by in vitro culture of E14.0 lower molars in the presence of the apoptosis inhibitor (z-VAD-fmk) and a subsequent subrenal culture. Our results showed that CD31- and CD34-positive cells progressively entered the central part of the dental papilla from the peridental mesenchyme. For TEM, angioblasts, young capillaries with thick endothelium and endothelial cells containing vacuoles were observed in peripheral dental mesenchyme, suggesting vasculogenesis was taking place. The presence of lateral sprouting, cytoplasmic filopodia and transluminal bridges in the dental papilla suggested angiogenesis was also occurring. Inhibition of apoptosis delayed the angiogenic vascularization of the dental papilla. Therefore, these data demonstrated that molar mesenchyme is progressively vascularized by mechanisms of both vasculogenesis and angiogenesis and apoptosis partially contributes to the vascularization of the dental papilla.

Purpose: To compare the occlusal and gingival microleakage of Class-II composite restorations utilizing etch-and-rinse and self-etch adhesives and different liner materials in primary and permanent teeth., Study Design: Standardized class-II cavities were prepared in freshly-extracted sound primary and permanent molars (n = 80/each), with all cavosurface margins involving enamel. The main experimental groups were; A. Single Bond 2/primary teeth; B. Adper SE Plus/primary teeth; C. Single Bond 2/permanent teeth; and D. Adper SE Plus/permanent teeth. Each group comprised 4 subgroups (n = 10/each) with respect to the liner material employed (n = 10/subgroup): 1. Fuji VII; 2. Fuji Triage; 3. Filtek Supreme XT Flowable Composite, and 4. No liner All teeth were restored with Filtek Supreme XT Universal Nanofilled Composite. Following thermocycling and immersion in basic fuchsin, the extent of microleakage was measured on crown sections using image analysis. The data were analyzed with Wilcoxon Signed Ranks Test, Mann-Whitney U-Test and Kruskal-Wallis One-Way ANOVA at alpha = 0.05., Results: In both primary and permanent teeth the use of etch-and-rinse adhesive resulted in similar occlusal and gingival microleakage values (p > 0.05). As for the self-etch adhesive, similar results were observed (p > 0.05) with the exception of significantly less occlusal leakage in the Fuji Triage VII and Fuji Triage subgroups of primary teeth than those of permanent teeth (p < 0.05). When the effects of liner material and the type of adhesive were disregarded, significantly more gingival microleakage was observed in primary teeth than in permanent teeth (p < 0.01), while the occlusal microleakage values were similar (p > 0.05). Irrespective of the tooth type and adhesive material, comparison of subgroups containing a liner material with those without one revealed no significant differences for both occlusal and gingival microleakage values (p > 0.05)., Conclusions: Occlusal microleakage was similar in both primary and permanent teeth, while a lesser extent of gingival seal was observed in primary teeth. Overall, placement of a liner material did not improve resistance to microleakage.

Introduction: This study evaluated dentin morphology and microhardness at the furcation area of mandibular molars., Methods: Tooth segments of mandibular molars were embedded in resin blocks and bisected longitudinally (n = 20), and the furcation area was delimited by tracing 2 orthogonal lines from the most concave point of the outer surface of the furcation toward the mesial and distal canal entrances. In half of the specimens, Knoop microhardness was measured in 2 directions in the furcation area: vertically, parallel to the orthogonal lines and the bisector of the angle formed by them; and horizontally, in the outer (close to the pulp chamber floor), inner (close to the cementum), and middle dentin layers. Data were analyzed statistically by one-way analysis of variance and Tukey-Kramer test (α = 0.05). The other half of the specimens were examined by scanning electron microscopy to evaluate dentin morphology and trajectory of the tubules., Results: No statistically significant difference (P > .05) was found among the mesial (46.5 ± 6.4), central (47.3 ± 8.1), and distal (49.7 ± 6.5) orthogonal lines. The inner dentin layer (51.7 ± 2.5) was statistically similar to the middle (46.3 ± 2.9) (P > .05), which was similar to the outer layer (41.4 ± 2.4) (P > .05). The inner layer was significantly harder than the outer layer (P < .05). The morphologic analysis revealed a tendency of calcification of the tubules from the outer toward the inner layer., Conclusions: Dentin microhardness at the furcation area is uniform in its 3 vertical axes, but the inner dentin layer is harder than the outer layer. Objective: The purpose of the present study was to investigate the occurrence and periodicity of enamel incremental lines in mouse molars in an attempt to draw attention to some key questions about the rhythm in the activity of the secreting ameloblasts during formation of mouse molar enamel., Methods: The mouse molars were ground, etched, and studied using scanning electron microscopy., Results: Lines interpreted as incremental lines generally appeared as grooves of variable distinctness, and were only observed cervically, in the region about 50-250μm from the enamel-cementum junction. The lines were most readily observable in the outer enamel and in the superficial prism-free layer, and were difficult to identify in the deeper parts of enamel, i.e. in the inner enamel with prism decussation. However, in areas where the enamel tended to be hypomineralized the incremental lines were observed as clearly continuous from outer into inner enamel. The incremental lines in mouse molar enamel exhibited an average periodicity of about 4μm, and the distance between the lines decreased towards the enamel surface., Conclusions: We conclude that incremental lines are to some extent visible in mouse molar enamel. Together with data from the literature and theoretical considerations, we suggest that they probably represent a daily rhythm in enamel formation. We studied the structural characteristics and periodicities of regular incremental markings in sheep enamel using fluorochrome injections for vital labeling of forming enamel and backscattered electron imaging in the scanning electron microscope. Microscopic analysis of mandibular first molars revealed the presence of incremental markings with a daily periodicity (laminations) that indicated successive positions of the forming front of interprismatic enamel. In addition to the laminations, incremental markings with a sub-daily periodicity were discernible both in interprismatic enamel and in enamel prisms. Five sub-daily increments were present between two consecutive laminations. Backscattered electron imaging revealed that each sub-daily growth increment consisted of a broader and more highly mineralized band and a narrower and less mineralized band (line). The sub-daily markings in the prisms of sheep enamel morphologically resembled the (daily) prisms cross striations seen in primate enamel. Incremental markings with a supra-daily periodicity were not observed in sheep enamel. Based on the periodicity of the incremental markings, maximum mean daily apposition rates of 17.0 µm in buccal enamel and of 13.4 µm in lingual enamel were recorded. Enamel extension rates were also high, with maximum means of 180 µm/day and 217 µm/day in upper crown areas of buccal and lingual enamel, respectively. Values in more cervical crown portions were markedly lower. Our results are in accordance with previous findings in other ungulate species. Using the incremental markings present in primate enamel as a reference could result in a misinterpretation of the incremental markings in ungulate enamel. Thus, the sub-daily growth increments in the prisms of ungulate enamel might be mistaken as prism cross striations with a daily periodicity, and the laminations misidentified as striae of Retzius with a supra-daily periodicity. This would lead to a considerable overestimation of crown formation times in ungulate teeth.

Cleaning and shaping are important section for the root canal treatment. A number of different methodologies have been developed to overcome these problems, including the introduction of rotary instruments nickel-titanium (NiTi). In endodontics NiTi have been shown to significantly reduce procedural errors compared to manual techniques of instrumentation. The efficiency of files is related to many factor. Although previous investigations that have used µCT analysis were hampered by insufficient resolution or projection incorrect. The new generation of μCT performance best offer, as micron resolution and accurate measurement software for evaluating the accurate anatomy of the root canal. The aim the paper was to evaluate the efficiency of Reciproc files in root canal treatment, evaluated before and after instrumentation by using μ-CT analysis.

This study analysed the effects of cryopreservation on periodontal regeneration of autotransplanted rat molars. First and second maxillary molars (n=92) of 24 four-week-old Wistar rats were gently extracted and autotransplanted into the abdominal tissue immediately (control group n=44) or after cryopreservation in liquid nitrogen for 7 days (experimental group n=48). At 1, 2, 4 and 10 weeks after transplantation, the transplanted molars were excised and regeneration of the periodontal tissues was analysed on histological sections stained with routine H&E and Goldner method. Different tissue responses were scored on a tooth basis: inflammation, regeneration of the periodontal ligament, resorption/apposition of cementum, and alveolar bone formation. Sixty-two teeth were available for histological evaluation, including 30 experimental and 32 control samples. One week after transplantation, both control and test teeth were surrounded by granulation tissue and some areas of root resorption could be seen. After 2 weeks, signs of regeneration of the periodontal ligament, cementum apposition, and new bone formation roughly coincided in both groups, however markedly retarded in the experimental group. After 4 weeks, regeneration progressed equally in both groups, presenting fewer areas of cementum apposition in experimental samples. Finally, 10 weeks after baseline transplantation, no significant differences between both groups could be observed. Cryopreservation followed by autotransplantation of extracted teeth in rats appears to have minimal detrimental effects on regeneration of periodontal tissues after integration periods of 1-10 weeks. The periodontal ligament (PDL), a soft tissue connecting the tooth and the bone, is essential for tooth movement, bone remodeling and force dissipation. A collagenous network that connects the tooth root surface to the alveolar jaw bone is one of the major components of the PDL. The organization of the collagenous component and how it changes under load is still poorly understood. Here using a state-of-the-art custom-made loading apparatus and a humidified environment inside a microCT, we visualize the PDL collagenous network of a fresh rat molar in 3D at 1 μm voxel size without any fixation or contrasting agents. We demonstrate that the PDL collagen network is organized in sheets. The spaces between sheets vary thus creating dense and sparse networks. Upon vertical loading, the sheets in both networks are stretched into well aligned arrays. The sparse network is located mainly in areas which undergo compressive loading as the tooth moves towards the bone, whereas the dense network functions mostly in tension as the tooth moves further from the bone. This new visualization method can be used to study other non-mineralized or partially mineralized tissues, and in particular those that are subjected to mechanical loads. Odontogenesis consists of a series of consecutive tooth morphogenesis stages, in which apoptosis is involved to eliminate the unnecessary cells. Autophagy, a lysosome or endosome-mediated self-degradation process, is indicated to participate in embryogenesis and tissue morphogenesis associated with apoptosis. This study hypothesized that autophagy may be involved and associated with apoptosis in odontogenesis. The transcripts of autophagy-related genes (Atg5, Atg7, and Atg12) were positively detected in tooth germs at embryonic day (E) 14.5 and postnatal day (P) 5.5 by quantitative real-time PCR. The protein expression of Atg5-Atg12 conjugate and lipidation of LC3 (microtubule-associated protein 1 light chain 3, autophagic marker) were revealed in the developing tooth germs by western blot. Meanwhile, LC3 was immunolocalized in the enamel organ and dental papilla at embryonic stages (E13.5-E18.5), especially stage E14.5 cervical loop and the PEK that facing the mesenchyme. At postnatal stages (P1.5-P15.5), besides the dental epithelium cells, LC3 was detected in the differentiating and differentiated odontoblasts, dental follicle cells, and Hertwig's epithelium root sheath cells. Moreover, double-immunofluorescence analysis revealed the partial colocalization of LC3 and TUNEL signal in the E14.5 PEK that facing the mesenchyme, the E16.5 stratum intermedium and outer enamel epithelium, the P5.5 stratum intermedium and stellate reticulum. Nevertheless, LC3 was also found in non-apoptotic cells. Furthermore, the transmission electron microscopic images revealed the presence of autophagy, as well as the partial colocalization of autophagic vacuoles and apoptotic nuclei during tooth development. Our findings imply the developmental appearance of autophagy and its partial colocalization with apoptosis during odontogenesis.

Aim: The accessory canals in the furcation of primary molars can explain the frequent presence of radiolucency in the furcation area in case of pulpitis and necrosis of teeth. In fact these canals can represent a communication between periodontium and pulp tissue allowing bacteria to enter. The aim of this study was to determine the prevalence of foramina in the furcation area of primary molars., Materials and Methods: The presence of foramina was assessed using extracted deciduous molars examined with Scanning Electron Microscope (SEM) evaluation. Thirty extracted human primary molars (15 upper deciduous molars and 15 lower deciduous molars) were selected to evaluate the presence of extra foramina in their furcation. All teeth were prepared according to Koenigs' technique: the roots were cut in the furcation area perpendicular to the canals (1.5 mm apical to the external surface of the furcation and 1 mm apical to the cervical line) and then analysed with an electronic scanning microscope., Results and Conclusion: This study demonstrated that 23 teeth (77% of the samples) had one or more extra foramina near the furcation.

In the elastic fiber system of the periodontal ligaments only oxytalan fibers can be identified, whereas all three types of fibers, oxytalan, elaunin and elastic fibers, are present in the gingiva. However, little information is available concerning their organization in the developing periodontal ligament. In the present study, growth and distribution of the oxytalan fiber system were examined in the developing periodontal ligament of rat molars using the specific staining for oxytalan, elastic and collagen fibers, and electron-microscopic analyses. Oxytalan staining clearly confirmed the earliest oxytalan fibers in a bell-staged tooth germ at embryonic day 18, which were tiny violet-colored fibers in the dental follicle. Their cross images were made up of dot-like microfibrils of 10-15nm in diameter close to fibroblasts in the dental follicle of the rat molars aged 1 day. These microfibrils appeared to be linked to one another through delicate filaments in 3-nm-diameter. At the beginning of root formation, the cross figures of oxytalan fibers were found as dot-like structures around the root sheath as well as in areas very close to blood vessels. As development proceeded, longer oxytalan fibers were produced in the apico-occlusal direction along with blood vessels. In addition, the immunoreactive products to anti-amyloid β protein on the surface of blood vessels suggest that this molecule might be involved in the adhesion of oxytalan fibers to vascular basement membranes. Aim: To evaluate the accumulation of hard-tissue debris when using the Self-adjusting File (SAF) system in mesial roots of mandibular molars with isthmuses and to compare it with that occurring when the ProTaper file system was used., Methodology: Forty extracted human mandibular molars with joining mesial root canals and an isthmus between the two canals were randomly assigned to two experimental groups and scanned using micro-computed tomography. Root canals in the control group (N = 20) were instrumented using the ProTaper rotary system until F3. Irrigation with 1 mL of 3% NaOCl solution was applied after each instrument. Root canals in the experimental group (N = 20) were prepared using the SAF for 4 min, with continuous irrigation (3% NaOCl, 4 mL min(-1)). After rescanning, canals in both groups were further irrigated with 5 mL of 17% EDTA for 2 min. After final scanning, the per cent value of total canal system volume filled with hard-tissue debris was calculated. Statistical analysis was performed using Mann-Whitney U-test; the alpha-type error was set at 1%., Results: Instrumentation of the root canals with ProTaper left 10.1% (IQR 5.2) of the total canal system volume filled with hard-tissue debris while preparation with the SAF left 1.7% (IQR 1.6). This difference was highly significant (P < 0.0001). After subsequent EDTA irrigation, these values were significantly reduced (P < 0.01) to 7.9% (IQR 4.1) and 1.3% (IQR 0.8) in the ProTaper and SAF groups, respectively., Conclusion: Preparation with the SAF system resulted in less hard-tissue debris accumulation in isthmus-containing root canal systems compared with instrumentation with ProTaper rotary files., (© 2011 International Endodontic Journal.)

Femtosecond lasers have been widely used in laser surgery as an instrument for contact-free tissue removal of hard dental, restorative materials, and osseous tissues, complementing conventional drilling or cutting tools. In order to obtain a laser system that provides an ablation efficiency comparable to mechanical instruments, the laser pulse rate must be maximal without causing thermal damage. The aim of this study was to compare the different morphological characteristics of the hard tissue after exposure to lasers operating in the femtosecond pulse regime. Two different kinds of samples were irradiated: dentin from human extracted teeth and bovine femur samples. Different procedures were applied, while paying special care to preserving the structures. The incubation factor S was calculated to be 0.788±0.004 for the bovine femur bone. These results indicate that the incubation effect is still substantial during the femtosecond laser ablation of hard tissues. The plasma-induced ablation has reduced side effects, i.e., we observe less thermal and mechanical damage when using a superficial femtosecond laser irradiation close to the threshold conditions. In the femtosecond regime, the morphology characteristics of the cavity were strongly influenced by the change of the effective number of pulses.

Aim: To compare the cleaning effectiveness of manual, hybrid and rotary instrumentation techniques in primary molar teeth., Methodology: Fifteen primary molars were selected. After endodontic access, the teeth were immersed in a medium containing Enterococcus faecalis and divided into three groups, according to the root canal instrumentation technique: group 1 - manual, group 2 - hybrid and group 3 - nickel-titanium (NiTi) rotary files. For microbiological evaluation, comparisons before and after instrumentation were performed using the paired Student's t-test. One-way anova complemented with the Student's t-test was used to compare the percentage of microbial reduction. Instrumentation time was evaluated by Kruskal-Wallis and Student-Newman-Keuls tests. Images obtained under scanning electron microscopy were analysed by three blinded examiners, and kappa statistics was used to evaluate calibration among examiners. The most frequent results among examiners were analysed using Kruskal-Wallis and Student-Newman-Keuls tests., Results: The hybrid technique required a significantly longer instrumentation time than the manual and rotary techniques (P < 0.05). All techniques tested were able to significantly reduce the number of E. faecalis (P < 0.05). The hybrid technique was associated with the highest intracanal bacterial reduction, with a statistically significant difference compared with manual instrumentation (P = 0.01). Manual instrumentation resulted in the lowest amount of debris and the highest amount of smear layer when compared with the rotary and hybrid techniques (P < 0.05). There was no difference between rotary and hybrid instrumentation in the removal of debris and smear layer., Conclusion: The use of NiTi rotary files is an option for root canal instrumentation in primary teeth., (© 2011 International Endodontic Journal.)

In this study a 3D microtomography display of tooth surfaces after in vitro dental wear tests has been obtained. Natural teeth have been compared with prosthetic teeth, manufactured by three different polyceramic composite materials. The prosthetic dental element samples, similar to molars, have been placed in opposition to human teeth extracted by paradontology diseases. After microtomography analysis, samples have been subjected to in vitro fatigue test cycles by servo-hydraulic mechanical testing machine. After the fatigue test, each sample has been subjected again to microtomography analysis to obtain volumetric value changes and dental wear surface images. Wear surface images were obtained by 3D reconstruction software and volumetric value changes were measured by CT analyser software. The aim of this work has been to show the potential of microtomography technique to display very clear and reliable wear surface images. Microtomography analysis methods to evaluate volumetric value changes have been used to quantify dental tissue and composite material wear.

The goal of the present article is to illustrate and analyze the applications and the potential of microcomputed tomography (micro-CT) in the analysis of tooth anatomy and root canal morphology. The authors performed a micro-CT analysis of the following different teeth: maxillary first molars with a second canal in the mesiobuccal (MB) root, mandibular first molars with complex anatomy in the mesial root, premolars with single and double roots and with complicated apical anatomy. The hardware device used in this study was a desktop X-ray microfocus CT scanner (SkyScan 1072, SkyScan bvba, Aartselaar, Belgium). A specific software ResolveRT Amira (Visage Imaging) was used for the 3D analysis and imaging. The authors obtained three-dimensional images from 15 teeth. It was possible to precisely visualize and analyze external and internal anatomy of teeth, showing the finest details. Among the 5 upper molars analyzed, in three cases, the MB canals joined into one canal, while in the other two molars the two mesial canals were separate. Among the lower molars two of the five samples exhibited a single canal in the mesial root, which had a broad, flat appearance in a mesiodistal dimension. In the five premolar teeth, the canals were independent; however, the apical delta and ramifications of the root canals were quite complex. Micro-CT offers a simple and reproducible technique for 3D noninvasive assessment of the anatomy of root canal systems.

Using light and scanning electron microscopy thin epoxide processed sections of 12 impacted third molars, one molar developed in odontogenic follicular cyst and one tooth-like teratoma were studied. It was established that the 4 impacted teeth and the tooth developed in odontogenic follicular cyst had obvious signs of enamel damage in the form of erosion in the crown fissures. The features of enamel alteration make it possible to interpret the changes as carious process which apparently has endogenous origin. Tooth-like teratoma extracted from ovarian dermoid cyst looked like canine and incisor "hybrid" coronal portion of which is covered with thin enamel layer, marked by obvious signs of multiple erosive lesions of endogenous nature. But unlike alterations in impacted teeth it resembles either fluorosis or enamel hypoplasia.

Enamel mineralisation is a highly controlled process in which the deposition, growth, and maturation of inorganic crystallites are regulated by secreted matrix proteins at the molecular and cellular level. Maxillary and mandibular first molars from the col1-caPPR mutants as well as normal controls aged for 12 weeks were observed by SEM and nanoindentation, respectively. Several types of aberrations in enamel distribution and crystal organisation were encountered in the transgenic molars. Also, the gene alteration resulted in degradation by as much as 23.42% in hardness and 17.56% in the elastic modulus. These data suggested that gene mutation altered the ameloblastic differentiation and movement, resulting in variations of crystal arrangement patterns, aberrations of enamel distribution, and degradation of mechanical behaviour. Unlabelled: OVERALL AIM: The general objective of this thesis was to enhance the understanding of Molar Incisor Hypomineralization (MIH) in areas of the histological, chemical and mechanical properties of the hypomineralized enamel, objective and subjective clinical symptoms in relation to bacteria findings. Further, to estimate a time for onset of the disturbance and investigate possible etiological factors., Material & Methods: 22 teeth diagnosed with MIH were used in the histological and chemical studies. A number of analytical methods were used; Light microscopy, Polarized light microscopy, Scanning electron microscopy, X-ray microanalysis, Vickers hardness test and X-ray Micro Computed Tomography. Decalcified sections were stained with bacterial staining. An ozone device was tested for the ability to kill strains of oral bacteria. In collaboration with the prospective ABIS study, 17.000 individuals were examined and possible etiological causes of severe demarcated opacities were tested., Results & Conclusions: The hypomineralized enamel was mainly located in the buccal enamel of the teeth and had a high degree of porosity extending from enamel-dentin-junction with a distinct border to the normal cervical enamel. Teeth diagnosed MIH had lower hardness values in hypomineralized enamel and differences in the chemical composition. Bacteria were observed in the enamel and deep into the dentin. Ozone treatment for 20 seconds or more was effective to kill oral microorganisms. Significant relations were found between MIH in first molars and breast feeding more than 6 months, late introduction to gruel and infant formula (later than 6 months). The onset for the hypomineralized enamel was estimated to around 200 days from start of the enamel mineralization.

Unlabelled: Despite the advances in bonding materials, many clinicians today still prefer to place bands on molar teeth. Molar bonding procedures need improvement to be widely accepted clinically., Objective: The purpose of this study was to evaluate the shear bond strength when an additional adhesive layer was applied on the occlusal tooth/tube interface to provide reinforcement to molar tubes., Material and Methods: Sixty third molars were selected and allocated to the 3 groups: group 1 received a conventional direct bond followed by the application of an additional layer of adhesive on the occlusal tooth/tube interface, group 2 received a conventional direct bond, and group 3 received a conventional direct bond and an additional cure time of 10 s. The specimens were debonded in a universal testing machine. The results were analyzed statistically by ANOVA and Tukey's test (α=0.05)., Results: Group 1 had a significantly higher (p<0.05) shear bond strength compared to groups 2 and 3. No difference was detected between groups 2 and 3 (p>0.05)., Conclusions: The present in vitro findings indicate that the application of an additional layer of adhesive on the tooth/tube interface increased the shear bond strength of the bonded molar tubes.

Many researchers have suggested that Australopithecus anamensis and Australopithecus afarensis were among the earliest hominins to have diets that included hard, brittle items. Here we examine dental microwear textures of these hominins for evidence of this. The molars of three Au. anamensis and 19 Au. afarensis specimens examined preserve unobscured antemortem microwear. Microwear textures of these individuals closely resemble those of Paranthropus boisei, having lower complexity values than Australopithecus africanus and especially Paranthropus robustus. The microwear texture complexity values for Au. anamensis and Au. afarensis are similar to those of the grass-eating Theropithecus gelada and folivorous Alouatta palliata and Trachypithecus cristatus. This implies that these Au. anamensis and Au. afarensis individuals did not have diets dominated by hard, brittle foods shortly before their deaths. On the other hand, microwear texture anisotropy values for these taxa are lower on average than those of Theropithecus, Alouatta or Trachypithecus. This suggests that the fossil taxa did not have diets dominated by tough foods either, or if they did that directions of tooth-tooth movement were less constrained than in higher cusped and sharper crested extant primate grass eaters and folivores.

A chronology of dental development in Pan troglodytes is arguably the best available model with which to compare and contrast reconstructed dental chronologies of the earliest fossil hominins. Establishing a time scale for growth is a requirement for being able to make further comparative observations about timing and rate during both dento-skeletal growth and brain growth. The absolute timing of anterior tooth crown and root formation appears not to reflect the period of somatic growth. In contrast, the molar dentition best reflects changes to the total growth period. Earlier initiation of molar mineralization, shorter crown formation times, less root length formed at gingival emergence into functional occlusion are cumulatively expressed as earlier ages at molar eruption. Things that are similar in modern humans and Pan, such as the total length of time taken to form individual teeth, raise expectations that these would also have been the same in fossil hominins. The best evidence there is from the youngest fossil hominin specimens suggests a close resemblance to the model for Pan but also hints that Gorilla may be a better developmental model for some. A mosaic of great ape-like features currently best describes the timing of early hominin dental development.

The aim was to characterize the external structure, roughness, and absolute depth profile (ADP) of fluorotic enamel compared with healthy enamel. Eighty extracted human molars were classified into four groups [TFI: 0, control (C); 1-3, mild (MI); 4-5, moderate (MO); 6-9, severe fluorosis (S)] according to the Thylstrup-Fejerskov Index (TFI). All samples were analyzed by atomic force microscopy.The mean values of enamel surface roughness (ESR) in nm were: Group C, 92.6; Group MI, 188.8; Group MO, 246.9; and Group S, 532.2. The mean values of absolute depth profile in nm were: C, 1,065.7; MI, 2,360.7; MO, 2,536.7; and S, 6,146.2. The differences between mean ESR and mean ADP among groups were statistically significant (p < 0.05). This structural study confirms at the nanometer level that there is a positive association between fluorosis severity, ESR, and ADP, and there is an association with the clinical findings of fluorosis measured by TFI as well.

The tribosphenic molar is a dental apomorphy of mammals and the molar type from which all derived types originated. Its enamel coat is expected to be ancestral: a thin, evenly distributed layer of radial prismatic enamel. In the bat Myotis myotis, we reinvestigated the 3D architecture of the dental enamel using serial sectioning combined with scanning electron microscopy analyses, biometrics of enamel prisms and crystallites, and X-ray diffraction. We found distinct heterotopies in enamel thickness (thick enamel on the convex sides of the crests, thin on the concave ones), angularity of enamel prisms, and in distribution of particular enamel types (prismatic, interprismatic, aprismatic) and demonstrated structural relations of these heterotopies to the cusp and crest organization of the tribosphenic molar. X-ray diffraction demonstrated that the crystallites composing the enamel are actually the aggregates of much smaller primary crystallites. The differences among particular enamel types in degree of crystallite aggregation and the variation in structural microstrain of the primary crystallites (depending upon the duration and the mechanical context of mineralization) represent factors not fully understood as yet that may contribute to the complexity of enamel microarchitecture in a significant way., (© 2010 Wiley-Liss, Inc.)

Introduction: The purpose of this in vitro study was to compare the debris removal efficacy of the EndoActivator system, the F file, ultrasonic irrigation, or 6% NaOCl irrigation alone in human mandibular molars after hand-rotary instrumentation., Methods: A custom brass cube (K-Kube) was used to create a sealed canal system, allowing each tooth to serve as its own control. Forty extracted mandibular molars were randomly divided into 4 equal experimental groups. Each tooth was mounted, sectioned at 1, 3, and 5 mm from the working length, and then reassembled into the K-Kube, and the mesial roots were similarly prepared by using hand-rotary instrumentation. For final debridement, group 1 used F file for 30 seconds, group 2 used EndoActivator system for 30 seconds, group 3 used ultrasonic irrigation for 30 seconds, and group 4 used irrigation with 6% NaOCl within 1 mm of working length. All groups received a final irrigation with 6% NaOCl in each canal. Specimens were evaluated at 1, 3, and 5 mm from the working length for cleanliness by capturing a digital image with a stereomicroscope. All specimens had the percent cleanliness for each canal and isthmus calculated both before and after final debridement. Statistical analysis was completed by using a repeated-measures analysis of variance with Tukey post hoc tests., Results and Conclusions: The results showed no statistically significant difference in canal or isthmus cleanliness among the 4 groups, but there was a statistically significant difference (P < .001) in canal cleanliness between the 1-mm level versus the 3-mm and 5-mm levels for all of the groups., (Published by Elsevier Inc.)
