Morchella steppicola (Morchellaceae) Zerova is included in the main list of the Red Book of the Belgorod Region of the Russian Federation. It has been assigned category and status 3 - a rare species. However, information about it is clearly incomplete, which is due to the peculiarities of its phenology and ecology, as well as the lack of systematic research aimed at finding its existing habitats. At the beginning of the 2020 season, reconnaissance surveys of characteristic ecotopes revealed 2 new localities of Morchella steppicola in unknown habitats (in the Belgorod and Shebekinsky administrative regions). The article describes these habitats in connection with the available data on the occurrence and distribution of this species, both within the region and beyond. The fact that new habitats have been identified should in no way call into question that Morchella steppicola is a species in relation to which the recommended measures should be followed: preserving existing habitats and searching for new ones, taking the identified populations under territorial protection, monitoring the abundance in known habitats. Morphological and molecular phylogenetic studies of true morels (Morchella) in North America, the Dominican Republic, Venezuela, Ecuador, and Peru led to the discovery of four undescribed species of Morchella. Two new species in the Elata clade, one from the Dominican Republic, initially distinguished by the informal designation Mel-18, and a newly discovered sister species from northern Arizona, are now recognized. Mel-18 is described as a novel phylogenetically distinct species, M. hispaniolensis. Its sister species from Arizona is described as M. kaibabensis, also recovered as an endophyte of Rocky Mountain juniper. Two additional species in the Esculenta clade, M. peruviana discovered in Peru and M. gracilis (previously reported as Mes-14) from the Dominican Republic, Venezuela, and Ecuador, are described as new. We also demonstrate that scanning electron microscopy (SEM) imaging of ascospores using rehydration/dehydration/critical point drying preparation techniques provides for enhanced resolution of spore wall surfaces, thereby increasing the number of morphological traits available to assess differences among otherwise closely related species. Morphological and molecular phylogenetic studies of true morels (Morchella) in the Canadian province of Newfoundland and Labrador resulted in the discovery of two undescribed species in the M. elata clade that we initially distinguished by the informal designations Mel-19 and Mel-36. The latter species, also collected in New Brunswick, Canada, is hitherto known only from the St Lawrence River Basin. Mel-36 is described here as a novel, phylogenetically distinct species, M. laurentiana. Before the discovery of Mel-19 in Newfoundland and Labrador, New Brunswick and Washington state it was only known from central China and central and northern Europe. Applying early names, with or without original material, to genealogical species is challenging. For morels this task is especially difficult because of high morphological stasis and high plasticity of apothecium color and shape. Here we propose a nomenclatural revision of true morels (Morchella, Pezizales) from Europe and North America, based on molecular phylogenetic analyses of portions of the genes for RNA polymerase II largest subunit (RPB1) and second largest subunit (RPB2), translation elongation factor-1a (TEF1), the nuc rDNA region encompassing the internal transcribed spacers 1 and 2, along with the 5.8S rDNA (ITS), and partial nuc 28S rDNA D1-D2 domains (28S). The 107 newly sequenced collections were from both continents, including 48 types, together with previously published sequences. Names are applied to 30 of the 65 currently recognized genealogical species. Results of the present study revealed that the number of Morchella species in Europe (n = 21) is nearly identical to that in North America (n = 22). Only seven species were found on both continents, consistent with previous reports of high continental endemism within the genus. Presently it is not possible to tell whether the transoceanic disjunctions were due to human activities, migration across a Bering land bridge or long-distance dispersal. In an effort to stabilize the taxonomy, due in part to the recent publication of synonyms for 11 of the species, accepted names are presented together with their corresponding later synonyms. A new subclade that includes holotypes of M. castanea and M. brunneorosea is identified in sect. Morchella (Esculenta Clade). Lectotypes for Morchella deliciosa, M. eximia and M. tridentina are designated here, as well as epitypes for M. dunalii, M. eximia, M. purpurascens and M. vulgaris. An abundant fruiting of a black morel was encountered in temperate northwestern New South Wales (NSW), Australia, during a mycological survey in Sep 2010. The site was west of the Great Dividing Range in a young, dry sclerophyll forest dominated by Eucalyptus and Callitris north of Coonabarabran in an area known as the Pilliga Scrub. Although the Pilliga Scrub is characterized by frequent and often large, intense wildfires, the site showed no sign of recent fire, which suggests this species is not a postfire morel. Caps of the Morchella data-like morel were brown with blackish ridges supported by a pubescent stipe that became brown at maturity. Because no morel has been described as native to Australia, the collections were subjected to multilocus molecular phylogenetic and morphological analyses to assess its identity. Results of these analyses indicated that our collection, together with collections from NSW and Victoria, represented a novel, genealogically exclusive lineage, which is described and illustrated here as Morchella australiana T. Recent molecular phylogenetic studies have revealed the existence of at least 50 species of Morchella worldwide and demonstrated a high degree of continental endemism within the genus. Here we describe 19 phylogenetic species of Morchella from North America, 14 of which are new (M. diminutiva, M. virginiana, M. esculentoides, M. prava, M. cryptica, M. frustrata, M. populiphila, M. sextelata, M. septimelata, M. capitata, M. importuna, M. snyderi, M. brunnea and M. septentrionalis). Existing species names (M. rufobrunnea, M. tomentosa, M. punctipes and M. angusticeps) are applied to four phylogenetic species, and formal description of one species (M. sp. "Mel-8") is deferred pending study of additional material. Methods for assessing morphological features in Morchella are delineated, and a key to the known phylogenetic species of Morchella in North America is provided. Phylogenetic analysis of LSU/ITS sequence data revealed two distinct lineages among 44 morphologically similar fruiting bodies of natural black morels {Morchella elata group) sampled at three non-burn locations in the St Joe and Kanisku National Forests in northern Idaho. Most of the sampled isolates (n = 34) represented a dominant LSU/ITS haplotype present at all three sites and identical to the Mel-12 phylogenetic lineage (GU551425) identified in a previous study. Variation at 1-3 nucleotide sites was detected among a small number of isolates (n = 6) within this well supported clade (94%). Four isolates sampled from a single location were in a well supported clade (97%) distinct from the dominant haplotypes and may represent a previously unsampled, cryptic phylogenetic species. Species-specific SNP and SCAR markers were developed for Mel-12 lineage isolates by cloning and sequencing AFLP amplicons, and segregation of AFLP markers were studied from single ascospore isolates from individual fruiting bodies. Based on the segregation of AFLP markers within single fruiting bodies, split decomposition analyses of two SCAR markers, and population genetic analyses of SNP, SCAR, and AFLP markers, it appears that members of the Morchella sp. Mel-12 phylogenetic lineage are heterothallic and outcross in nature similar to yellow morels. This is the first set of locus-specific molecular markers that has been developed for any Morchella species, to our knowledge. These markers will prove to be valuable tools to study mating system, gene flow and genetic structure of black morels at various spatial scales with field-collected fruiting bodies and eliminate the need to culture samples in vitro. Abstract: The in vitro growth of Morchella elata was characterized with respect to the effects of a variety of substrates, isolates, developmental status of the parental ascoma, temperature, and pH. Optimal substrates for growth included sucrose, mannose and lactose, but the growth of some isolates was substantially reduced in some composite media. Maltose and potato-dextrose media limited growth and caused changes in colony morphology; mycelial pigmentation was black in the case of maltose, and mycelial margins were plumose in potato-dextrose cultures. Rapid growth was most reliably achieved in a composite medium containing 1:1 sucrose:mannose. Isolates derived from single ascospores shortly after ejection from ascomata varied in ability to grow in the various substrates. This may be related to variable maturity or dormancy; increasing growth rates correlated with pileus length in the parental ascomata, and ascomata that initially produced slower-growing or abortive colonies produced faster-growing colonies after storage at 20°C for 96 wk. The growth of M. elata derived from recently ejected ascospores was optimal at 16–24°C or above for a faster-growing isolate, and 20–24°C or above for a slow-growing isolate. Although neither isolate grew at 8°C or below in an initial experiment, spawn cultured on puffed wheat at 28°C produced mycelia that proliferated when transferred to soil media and incubated at 8°C. Growth of M. elata in liquid cultures adjusted with potassium hydroxide was optimal at pH 7.0, and was relatively sensitive to more acidic or alkaline pH. When calcium carbonate was used to adjust pH, optimal growth shifted to pH 7.7 or above, suggesting that wood ash and other calcium compounds may not only stimulate growth in natural settings, but also alter the optimal pH for proliferation of M. elata. Further studies with other substrate combinations and incubation conditions will be necessary to fully understand the connections between in vitro growth and the ecological behaviour of the fungus. The external morphological changes occurring during initiation and early stages of fruit body development of a Morchella sp., before the development of asci, were examined by scanning electron microscopy. Four stages of primordial development were distinguished. First, disk-shaped knots of 0.51.5 mm were observed on the surface of the substrate. Next, the knot inflated and a primordial stem emerged from its centre. Afterward, the stem lengthened, oriented upward, and two types of hyphal elements developed: long, straight and smooth basal hairy hyphae and short stem hyphae, some of which were inflated and projected out of a cohesive layer of tightly packed hyphal elements. Finally, when the stem was 23 mm long, pre-apothecia emerged in the apical end, with ridges and pits having distinguished types of paraphyses. Morchella conica Pers. (M. conica) has been used both as a medical and edible mushroom and possesses antimicrobial properties and antioxidant activities. However, the antioxidant properties of polysaccharides purified from M. conica have not been studied. The aim of this study was to investigate the in vitro antioxidant properties of a polysaccharide NMCP-2 (neutral M. conica polysaccharides-2) purified from M. conica, as determined by radical scavenging assay and H2O2-induced oxidative stress in HEK 293T cells. Results showed that NMCP-2 with an average molecular weight of 48.3 kDa possessed a much stronger chelating ability on ferrous ions and a higher ability to scavenge radical scavenging 2,2-diphenyl-1-picrylhydrazyl (DPPH) than the other purified fraction of NMCP-1 from M. conica. Moreover, 3-(4, 5-Dimethylthiazol-2-yl)-2, 5-diphenyltetra-zolium bromide (MTT) assay showed that NMCP-2 dose-dependently preserved cell viability of H2O2-induced cells. The NMCP-2 pretreated group reduced the generation of reactive oxygen species (ROS) content and increased the mitochondria membrane potential (MMP) levels. In addition, Hoechst 33342 staining revealed cells treated with NMCP-2 declined nuclear condensation. Ultrastructural observation revealed that NMCP-2 pretreatment alleviated the ruptured mitochondria when exposed to H2O2. Furthermore, western blot analysis showed that NMCP-2 prevented significant downregulation of the protein expression of Bax, cleaved caspases 3, and upregulated Bcl-2 levels. These results suggest the protective effects of NMCP-2 against H2O2-induced injury in HEK 293T cells. NMCP-2 could be used as a natural antioxidant of functional foods and natural drugs. This article offers information regarding the rich history of wild mushroom usage in Asia. It discusses that there are several species of cultivated mushroom in Asia which includes morels which have been harvested and eaten for centuries in the various mountain ranges of the region. China, Afghanistan, turkey, and Pakistan exports morels to Europe. In addition, matsutake are highly valued in Japan and Korea as well as China are primary exporters of matsutake to Japan. It mentions that China's urban middle class enjoy getting out into the forest from time to time to harvest mushroom.
