We describe a surface charge imaging method for heterogeneous biosilicas based on relationships between zeta ( ζ ) potential, feature size of nanoparticles, and PDMPO fluorescence and apply it to silicified structures from plants and diatoms. The methodology provides the first opportunity to map the surface charge of large heterogeneous biosilica materials and indicates that local surface charge is related to morphology below the diffraction limit ( ca. 20-130 nm) with sharper features showing less negative zeta potential equivalent surface charge suggesting that the zeta potential of silica structures can be adjusted by engineering surface morphology. We show that the approach can be used to study living silicified biological tissues without recourse to sectioning and fixation. Further, the approach could be used for the study of other metal oxides possessing hydroxylated moieties. The method has potential to open up opportunities for the engineering of materials with defined charge characteristics for the solution of biomedical engineering problems including materials for tissue replacement.

Molecular recognition and interactions at the interface between biomolecules and inorganic materials determine important phenomena such as protein adsorption, cell adhesion to biomaterials, or the selective response of biosensors. Events occurring at the biomolecule-inorganic interface, despite their importance, are still poorly understood, thus limiting control of interfacial properties and response. In this contribution, using well-characterized silica nanoparticles and a series of peptides having heterogeneous physicochemical properties (S1: KLPGWSG, S2: AFILPTG, and S3: LDHSLHS) identified from biopanning against the same particles, we identify the driving forces that govern peptide-silica binding. Binding isotherms obtained by fluorimetric assay under different pH conditions allowed us to demonstrate the impact of binding environment (pH) on adsorption behavior of a given peptide-surface silica nanoparticle. Our experimental data suggest a multistep adsorption mechanism leading to the formation of multilayers on silica, in which the prevailing interactions (i.e., electrostatic or hydrophobic/hydrogen bonding) and their relative contribution to the binding event are governed by the identity of the peptide itself, the substrate's surface functionality (hydrophilic or hydrophobic), and the peptide bulk concentration and solution bulk pH. Our studies show how it is possible to modulate peptide uptake on silica, or in fact on any particle, by changing either the surface properties or, more simply, the binding environment. In addition, the data reveal an intrinsic bias toward positively charged sequences in the elution conditions used in the biopanning protocol with much information about strong binder sequence diversity being lost during panning.

Control over selective recognition of biomolecules on inorganic nanoparticles is a major challenge for the synthesis of new catalysts, functional carriers for therapeutics, and assembly of renewable biobased materials. We found low sequence similarity among sequences of peptides strongly attracted to amorphous silica nanoparticles of various size (15-450 nm) using combinatorial phage display methods. Characterization of the surface by acid base titrations and zeta potential measurements revealed that the acidity of the silica particles increased with larger particle size, corresponding to between 5% and 20% ionization of silanol groups at pH 7. The wide range of surface ionization results in the attraction of increasingly basic peptides to increasingly acidic nanoparticles, along with major changes in the aqueous interfacial layer as seen in molecular dynamics simulation. We identified the mechanism of peptide adsorption using binding assays, zeta potential measurements, IR spectra, and molecular simulations of the purified peptides (without phage) in contact with uniformly sized silica particles. Positively charged peptides are strongly attracted to anionic silica surfaces by ion pairing of protonated N-termini, Lys side chains, and Arg side chains with negatively charged siloxide groups. Further, attraction of the peptides to the surface involves hydrogen bonds between polar groups in the peptide with silanol and siloxide groups on the silica surface, as well as ion-dipole, dipole-dipole, and van-der-Waals interactions. Electrostatic attraction between peptides and particle surfaces is supported by neutralization of zeta potentials, an inverse correlation between the required peptide concentration for measurable adsorption and the peptide pI, and proximity of cationic groups to the surface in the computation. The importance of hydrogen bonds and polar interactions is supported by adsorption of noncationic peptides containing Ser, His, and Asp residues, including the formation of multilayers. We also demonstrate tuning of interfacial interactions using mutant peptides with an excellent correlation between adsorption measurements, zeta potentials, computed adsorption energies, and the proposed binding mechanism. The role of polymer (poly(vinylamine)) size (238-11000 units) on silicic acid condensation to yield soluble nanoparticles or composite precipitates has been explored by a combination of light scattering (static and dynamic), laser ablation combined with aerosol spectrometry, IR spectroscopy, and electron microscopy. Soluble nanoparticles or composite precipitates are formed according to the degree of polymerization of the organic polymer and pH. Nanoparticles prepared in the presence of the highest molecular weight polymers have core-shell like structures with dense silica cores. Composite particles formed in the presence of polymers with extent of polymerization below 1000 consist of associates of several polymer-silica nanoparticles. The mechanism of stabilization of the "soluble" silica particles in the tens of nanometer size range involves cooperative interactions with the polymer chains which varies according to chain length and pH. An example of the use of such polymer-poly(silicic acid) nanoparticles in the generation of composite polymeric materials is presented. The results obtained have relevance to the biomimetic design of new composite materials based on silica and polymers and to increasing our understanding of how silica may be manipulated (stored) in the biological environment prior to the formation of stable mineralized structures. We suspect that a similar method of storing silicic acid in an active state is used in silicifying organisms, at least in diatom algae.

Interactions of aqueous solutions of aluminum polyoxocations (Al13-mers and Al30-mers) and aluminum hydroxide suspensions of varying particle sizes (26, 55, and 82 nm) with a model protein, bovine serum albumin (BSA), have been investigated using potentiometry, conductometry, viscometry, 27Al solution NMR, UV-vis spectroscopy, dynamic light scattering, zeta-potential measurements, thermogravimetry, X-ray diffraction, and scanning electron microscopy. Increasing amounts of BSA partially convert Al13-mers and, to a larger extent, Al30-mers into amorphous Al hydroxide without gel formation. At the same time, BSA molecules can form unstable aggregates in the Al polyoxocation solutions which redisperse easily upon standing. In the case of Al hydroxide sols, BSA addition causes substantial gelation, the extent of which is proportional to the amount of BSA added and inversely related to the Al hydroxide particle size. Upon freeze-drying or centrifugation of Al species-BSA solutions, an interesting sheetlike morphology with 150-200 nm wide nanoribbons is observed for pure Al hydroxide nanoparticles and for solutions of Al polyoxocations with the highest amount of BSA studied. On the basis of the combined solution, colloidal and solid-state characterization of model Al species-BSA systems, a qualitative model of possible interactions in the Al polyoxocation-BSA and Al hydroxide-BSA systems is proposed wherein core-shell hybrid nanoparticles are formed from protein "core" and Al polyoxocation "shell" or Al hydroxide "core" and protein "shell".

Calcium carbonate (E170) is a common food and pharmaceutical additive/ingredient. In addition to a source of calcium, the carbonate has uses including as a colour, acidity regulator and bulking agent. Globally, a range of regulatory agencies and pharmacopoeia control the analyses and specification of additives in food, supplements, pharmaceutical substances and excipients. Accordingly, a range of specifications and analyses exist for calcium carbonate depending on the application and market of the product. In this contribution, we analyse calcium carbonates from geological, synthetic and biogenic sources, focussing on acid insoluble impurities, a test required by current monographs. Analysis of calcium carbonate from different origins may require modification of existing tests to comply with regulatory bodies, due to the variation of impurities specific to the source of the material. We suggest an analytical approach involving centrifugation that improves analytical efficiency (up to 85% time reduction), especially for calcium carbonate of biological origin. Abstract: A novel synthesis method was described to prepare Au@SiO2 core-shell spheres, using a commercial gold colloid and tetraethyl orthosilicate as the precursors. The synthesis concept was based on surface modification of the gold nanoparticles with a binary system of surfactants, or using both of 2-Mercapto-ethanol and (3-Aminopropyl)-trimethoxysilane together, and the Stöber processing. The shape and size, and silica coatings of the resultant particles were investigated by transmitting electron microscopy (TEM). The Au@silica core-shell spheres, each containing a single gold nanoparticle or an aggregate of several gold nanoparticles at the center, could be prepared. [Copyright &y& Elsevier]

Silica skeletal architectures in diatoms are characterized by remarkable morphological and nanostructural details. Silk proteins from spiders and silkworms form strong and intricate self-assembling fibrous biomaterials in nature. We combined the features of silk with biosilica through the design, synthesis, and characterization of a novel family of chimeric proteins for subsequent use in model materials forming reactions. The domains from the major ampullate spidroin 1 (MaSp1) protein of Nephila clavipes spider dragline silk provide control over structural and morphological details because it can be self-assembled through diverse processing methods including film casting and fiber electrospinning. Biosilica nanostructures in diatoms are formed in aqueous ambient conditions at neutral pH and low temperatures. The R5 peptide derived from the silaff in protein of Cylindrotheca fusiformis induces and regulates silica precipitation in the chimeric protein designs under similar ambient conditions. Whereas mineralization reactions performed in the presence of R5 peptide alone form silica particles with a size distribution of 0.5-10 μm in diameter, reactions performed in the presence of the new fusion proteins generate nanocomposite materials containing silica particles with a narrower size distribution of 0.5-2 μm in diameter. Furthermore, we demonstrate that composite morphology and structure could be regulated by controlling processing conditions to produce films and fibers. These results suggest that the chimeric protein provides new options for processing and control over silica particle sizes, important benefits for biomedical and specialty materials, particularly in light of the all aqueous processing and the nanocomposite features of these new materials. Abstract: Osteoinductive and biodegradable composite biomaterials for bone regeneration were prepared by combining silk fibroin with silica particles. The influence of these composite systems on osteogenesis was evaluated with human mesenchymal stem cells (hMSCs) subjected to osteogenic differentiation. hMSCs adhered, proliferated, and differentiated towards osteogenic lineages on silk/silica films. The addition of the silica to the silk films influenced gene expression leading to upregulation of bone sialoprotein (BSP) and collagen type 1 (Col 1) osteogenic markers. Evidence for early bone formation in the form of collagen fibers and apatite nodules was obtained on the silk/silica films. Collagen fibers were closely associated with apatite deposits and overall collagen content was higher for the silica containing samples. Also, smaller sized silica particles (24 nm–2 μm) with large surface area facilitated silica biodegradation in vitro through particle dissolution, leading to ∼5-fold decrease in silica content over 10 weeks. These results indicate the suitability of silk/silica composite system towards bone regeneration, where degradation/remodeling rates of the organic and inorganic components can be controlled. Abstract: Aluminium doped barium and strontium hexaferrite nanoparticles BaAl x Fe(12−x)O19 and SrAl x Fe(12−x)O19 were synthesised via a sol–gel route using citric acid to complex the ions followed by an auto-combustion reaction. This method shows promise for the synthesis of complex ferrite powders with small particle size. It was found that around half of the iron could be substituted for aluminium in the barium ferrite with structure retention, whereas strontium aluminium ferrites could be produced with any aluminium content including total substitution of the iron. All synthesised materials consisted of particles smaller than 1μm, which is the size of a single magnetic domain, and various doping levels were achieved with the final elemental composition being within the bounds of experimental error. The materials show structural and morphological changes as they move from iron to aluminium ferrites. Such materials may be promising for imaging applications. [Copyright &y& Elsevier]
