All biomaterials initiate a tissue response when implanted in living tissues. Ultimately this reaction causes fibrous encapsulation and hence isolation of the material, leading to failure of the intended therapeutic effect of the implant. There has been extensive bioengineering research aimed at overcoming or delaying the onset of encapsulation. Nanotechnology has the potential to address this problem by virtue of the ability of some nanomaterials to modulate interactions with cells, thereby inducing specific biological responses to implanted foreign materials. To this effect in the present study, we have characterised the growth of fibroblasts on nano-structured sheets constituted by BaTiO3, a material extensively used in biomedical applications. We found that sheets of vertically aligned BaTiO3 nanotubes inhibit cell cycle progression - without impairing cell viability - of NIH-3T3 fibroblast cells. We postulate that the 3D organization of the material surface acts by increasing the availability of adhesion sites, promoting cell attachment and inhibition of cell proliferation. This finding could be of relevance for biomedical applications designed to prevent or minimize fibrous encasement by uncontrolled proliferation of fibroblastic cells with loss of material-tissue interface underpinning long-term function of implants.

While in the last years applications of carbon nanotubes in the field of biotechnology have been largely proposed, biomedical applications of boron nitride nanotubes (BNNTs) are yet totally unexplored. BNNTs have very interesting physical properties that should be exploited in the biomedical field. At this date, studies on their biocompatibility are completely missing and the first issue behind this investigation is the dispersion of BNNTs in aqueous solutions. In this paper the authors propose, for the first time, a technique for obtaining BNNT stable dispersions suitable for biological applications, based on polyethyleneimine (PEI) water solutions. Based on authors' knowledge, in vitro testing performed on human neuroblastoma cell line (SH-SY5Y) is the first study of interaction between BNNTs and living material. Experimental results showed a satisfactory cell viability up to a concentration of 5.0 microg/ml PEI-BNNTs in the cell culture medium.

Boron nitride nanotubes (BNNTs) have unique physical properties, which can be exploited in the biomedical field. Hence, the surprising lack of reported studies on their biocompatibility and interactions with living cells, addressed by the present paper which deals the results of such an investigation based on 72 h culture of human neuroblastoma cell line (SH-SY5Y) in the presence of an aqueous suspension of polyethyleneimine (PEI)-coated BNNTs. BNNTs conjugated with fluorescent markers (quantum dots) are employed to enable tracking of their uptake by living cells. The results demonstrate good cytocompatibility together with unequivocal BNNT cellular uptake by an energy-dependent endocytic process.

Abstract The 3-D spatial and mechanical features of nano-topography can create alternative environments, which influence cellular response. In this paper, murine fibroblast cells were grown on surfaces characterized by protruding nanotubes. Cells cultured on such nano-structured surface exhibit stronger cellular adhesion compared to control groups, but despite the fact that stronger adhesion is generally believed to promote cell cycle progression, the time cells spend in G1 phase is doubled. This apparent contradiction is solved by confocal microscopy analysis, which shows that the nano-topography inhibits actin stress fiber formation. In turn, this impairs RhoA activation, which is required to suppress the inhibition of cell cycle progression imposed by p21/p27. This finding suggests that the generation of stress fibers, required to impose the homeostatic intracellular tension, rather than cell adhesion/spreading is the limiting factor for cell cycle progression. Indeed, nano-topography could represent a unique tool to inhibit proliferation in adherent well-spread cells. Boron neutron capture therapy (BNCT) is increasingly being used in the treatment of several aggressive cancers, including cerebral glioblastoma multiforme. The main requirement for this therapy is selective targeting of tumor cells by sufficient quantities of 10B atoms required for their capture/irradiation with low-energy thermal neutrons. The low content of boron targeting species in glioblastoma multiforme accounts for the difficulty in selective targeting of this very malignant cerebral tumor by this radiation modality. In the present study, we have used for the first time boron nitride nanotubes as carriers of boron atoms to overcome this problem and enhance the selective targeting and ablative efficacy of BNCT for these tumors. Following their dispersion in aqueous solution by noncovalent coating with biocompatible poly- l-lysine solutions, boron nitride nanotubes were functionalized with a fluorescent probe (quantum dots) to enable their tracking and with folic acid as selective tumor targeting ligand. Initial in vitro studies have confirmed substantive and selective uptake of these nanovectors by glioblastoma multiforme cells, an observation which confirms their potential clinical application for BNCT therapy for these malignant cerebral tumors. Because mass-produced carbon nanotubes (CNTs) are strongly aggregated and highly hydrophobic, processes to make them water soluble are required for biological applications. Suspensions in surfactant solutions are often employed. Among these, Pluronic F127 appear to be highly biocompatible if used at low concentrations. Starting from these results, this work involves a systematic study to clarify the dispersion behaviour of CNTs in Pluronic F127. The results suggest a two-step process: first, the bundles disaggregate, kinetically driven by the energy supplied to the system; second, they disperse (surfactant adsorption), thermodynamically driven by the surfactant concentration. The dispersion reaction data are well fitted by a first-order kinetics reaction. By performing a pretreatment step, consisting of stirring at 70°C, the achieved concentration of CNTs in solution is twice that of the traditional process. The proposed procedure provides an optimal compromise between a low Pluronic concentration and a high CNT concentration. [ABSTRACT FROM AUTHOR]

Abstract: Carbon nanotubes (CNTs) are widely used for biomedical applications as intracellular transporters of (bio)molecules, due to their high propensity to cross cell membranes. However, there is a clear discrepancy in the literature about their uptake mechanism, which should be related to the differences existing in the nanotube materials, as well as the experimental procedures. Despite the fact that there are some studies on the influence of the CNT surface chemistry, the role of the properties of non-functionalized CNTs in cellular uptake has not been much investigated to a great extent. In this work, different kinds of multi-wall CNTs (MWCNTs) are produced and fully characterized, in terms of diameter, length, metal impurity, carbon soot and surface chemistry. These MWCNT samples are tested in vitro, and the cellular uptake is indirectly evaluated by using standard fluorescent probes and confirmed by TEM images. Our assays demonstrate that nanotube length clearly influences their uptake and shorter (sub-1μm) MWCNTs are easier to be internalized through an energy-independent pathway. The results of this investigation may be useful for the design of promising CNT-based vectors for cell therapy. [Copyright &y& Elsevier]
