Peach (Prunus persica (L.) Batsch) is a typical shallow-rooted fruit plant with a high respiratory intensity and oxygen demand, which makes it highly susceptible to oxygen-deficient soil conditions resulting from waterlogging. Rootstock waterlogging resistance is essential to the performance of cultivated peaches under waterlogging stress. In comparison to Prunus persica var. persica ('Maotao', M) and Prunus davidiana (Carr.) C. de Vos ('Shantao', S), Prunus persica f. Hossu ('Hossu', H) exhibited superior leaf photosynthetic electron transfer efficiency, a higher rate of mycorrhizal fungi infection in both fine roots and mesophyll palisade cells, as well as earlier air cavity formation in both leaf midvein and fine roots under waterlogging stress. Furthermore, under non-waterlogging conditions, Hossu had greater leaf superoxide dismutase (SOD) activity, higher proline content, and a greater content of starch granules in the pith and xylem ray cells of stems and roots than rootstocks M and S. As a result, Hossu's tolerance to waterlogging may be due to its higher photosynthetic efficiency, improved tissue oxygen permeability, higher energy metabolism, and increased intracellular mycorrhizal fungus infection rates in both root parenchyma cells and mesophyll palisade cells. The function of floral organ identity genes, APETALA1/2/3 , PISTILLATA, AGAMOUS , and SEPALLATA1/2/3 , in flower development is highly conserved across angiosperms. Emerging evidence shows that these genes also play important roles in the development of the fruit that originates from floral organs following pollination and fertilization. However, their roles in fruit development may vary significantly between species depending on the floral organ types contributing to the fruit tissues. Fruits of the Rosaceae family develop from different floral organ types depending on the species, for example, peach fruit flesh develops from carpellary tissues, whereas apple and strawberry fruit flesh develop from extra-carpellary tissues, the hypanthium and receptacle, respectively. In this review, we summarize recent advances in understanding floral organ gene function in Rosaceae fruit development and analyze the similarities and diversities within this family as well as between Rosaceae and the model plant species Arabidopsis and tomato. We conclude by suggesting future research opportunities using genomics resources to rapidly dissect gene function in this family of perennial plants. [ABSTRACT FROM AUTHOR]

Type 2C protein phosphatase (PP2C) plays an essential role in abscisic acid (ABA) signaling transduction processes. In the current study, we identify 719 putative PP2C genes in eight Rosaceae species, including 118 in Chinese white pear, 110 in European pear, 73 in Japanese apricot, 128 in apple, 74 in peach, 65 in strawberry, 78 in sweet cherry, and 73 in black raspberry. Further, the phylogenetic analysis categorized PbrPP2C genes of Chinese white pear into twelve subgroups based on the phylogenic analysis. We observed that whole-genome duplication (WGD) and dispersed gene duplication (DSD) have expanded the Rosaceae PP2C family despite simultaneous purifying selection. Expression analysis finds that PbrPP2C genes have organ-specific functions. QRT-PCR validation of nine PbrPP2C genes of subgroup A indicates a role in ABA-mediated response to abiotic stress. Finally, we find that five PbrPP2C genes of subgroup A function in the nucleus. Peach is a typical climacteric fruit that releases ethylene during fruit ripening. Several studies have been conducted on the transcriptional regulation of ethylene biosynthesis in peach fruit. Herein, an ethylene response factor, PpERF.A16, which was induced by exogenous ethylene, could enhance ethylene biosynthesis by directly inducing the expression of 1-aminocyclopropane-1-carboxylic acid synthase (PpACS1) and 1-aminocyclopropane-1-carboxylic acid oxidase (PpACO1) genes. Moreover, the NAM/ATAF1/2/CUC2 (NAC) transcription factor (TF) PpNAC.A59 was coexpressed with PpERF.A16 in all tested peach cultivars. Interestingly, PpNAC.A59 can directly interact with the promoter of PpERF.A16 to induce its expression but not enhance LUC activity driven by any promoter of PpACS1 or PpACO1. Thus, PpNAC.A59 can indirectly mediate ethylene biosynthesis via the NAC-ERF signaling cascade to induce the expression of both PpACS1 and PpACO1. These results enrich the genetic network of fruit ripening in peach and provide new insight into the ripening mechanism of other perennial fruits. [ABSTRACT FROM AUTHOR]

Abstract Homeobox transcription factors belong to a superfamily that has been widely studied in plant growth and development, but little is known regarding their role in fruit development and ripening. Using a genome-wide expression analysis of homeobox (HB) genes and quantitative real-time PCR, a HD-ZIP II member, PpHB.G7 , which presented higher levels of expression in ripening fruits than in developing fruits in all of the tested cultivars, was isolated from peach. Transient transformations showed that PpHB.G7 affects ethylene production and the expression of ethylene biosynthesis genes (PpACS1 and PpACO1). Both dual-luciferase and yeast one-hybrid assays confirmed that PpHB.G7 interacts with the promoters of PpACS1 and PpACO1. Thus, PpHB.G7 mediates ethylene biosynthesis by stimulating PpACS1 and PpACO1 activities. Furthermore, we also found that the other eight HB genes were differentially expressed in the developing fruits, with seven of these genes belonging to the HD-ZIP family. Anthocyanin pigmentation is an important consumer trait in peach ( Prunus persica). In this study, the genetic basis of the blood-flesh trait was investigated using the cultivar Dahongpao, which shows high levels of cyanidin-3-glucoside in the mesocarp. Elevation of anthocyanin levels in the flesh was correlated with the expression of an R2R3 MYB transcription factor, Pp MYB10.1. However, Pp MYB10.1 did not co-segregate with the blood-flesh trait. The blood-flesh trait was mapped to a 200-kb interval on peach linkage group ( LG) 5. Within this interval, a gene encoding a NAC domain transcription factor ( TF) was found to be highly up-regulated in blood-fleshed peaches when compared with non-red-fleshed peaches. This NAC TF, designated BLOOD ( BL), acts as a heterodimer with Pp NAC1 which shows high levels of expression in fruit at late developmental stages. We show that the heterodimer of BL and Pp NAC1 can activate the transcription of Pp MYB10.1, resulting in anthocyanin pigmentation in tobacco. Furthermore, silencing the BL gene reduces anthocyanin pigmentation in blood-fleshed peaches. The transactivation activity of the BL-Pp NAC1 heterodimer is repressed by a SQUAMOSA promoter-binding protein-like TF, Pp SPL1. Low levels of Pp MYB10.1 expression in fruit at early developmental stages is probably attributable to lower levels of expression of Pp NAC1 plus the presence of high levels of repressors such as Pp SPL1. We present a mechanism whereby BL is the key gene for the blood-flesh trait in peach via its activation of Pp MYB10.1 in maturing fruit.
