Um dos problemas associados a isto é a ocorrência de solos com problemas de drenagem, principalmente na região de Pelotas, que dependendo do período do ano, podem sofrer situações de alagamento, prejudicando potencialmente o desenvolvimento e a produtividade da cultura. Em situações de alagamento do solo, o oxigênio se torna insuficiente para raízes, prejudicando o processo de respiração e podendo levar as células a uma crise energética. Somado a isso, é sabido que as mudanças climáticas vêm alterando o clima global, sendo esperado que tais alterações aumentem a frequência e a gravidade dos eventos de inundação em muitas regiões agriculturáveis do planeta. Para que estudos possam auxiliar na seleção de genótipos tolerantes ao alagamento, é de fundamental importância entender as alterações fisiológicas e moleculares das plantas em situações de privação de oxigênio. Desta forma, no presente trabalho foram conduzidos três estudos, os quais tiveram como objetivo a caracterização de respostas dos parâmetros de trocas gasosas e moleculares em cinco porta-enxertos de Prunus spp. submetidos ao alagamento do solo. No primeiro estudo objetivou-se obter o perfil transcriptômico em folhas dos porta-enxertos de pessegueiro ‘Capdeboscq’ e da ameixeira ‘Julior’ submetidos ao alagamento por 48 horas, através do uso da técnica de RNA-Seq. Desta análise, foram identificados 3.011 genes diferencialmente expressos (DEGs) para ‘Capdeboscq’ (sensível), onde 1.566 foram up-regulated e 1.445 down-regulated, e 251 DEGs para ‘Julior’ (tolerante ao estresse), sendo 125 up-regulated e 126 downregulated. Embora tenha ativado vias de sinalização, ‘Capdeboscq’ teve a fotossíntese como o processo mais afetado em nível fisiológico e molecular apresentando maior número de termos GOs (Gene Ontology) enriquecidos down-regulated. ‘Julior’ foi mais eficiente ao conseguir acionar respostas de defesa, as quais incluíram a ativação de vias de biossíntese de flavonoides, segundo a análise funcional de vias metabólicas do KEGG (Kyoto Encyclopedia of Genes and Genomes) e termos GOs. A família de fatores de transcrição LSD parece ser um ponto-chave na elucidação dos mecanismos de defesa frente ao estresse, dentre as famílias identificadas neste estudo. A rede de interação proteína-proteína de ‘Capdeboscq’ apresentou altos graus de interação e indicam a grande probabilidade de alguns nós identificados serem hubs. No segundo estudo, foram testados oito genes candidatos à referência quanto a estabilidade de expressão para estudos de RT-qPCR em folhas e raízes de quatro porta-enxertos de Prunus spp. submetidos ao alagamento do solo por 0, 6, 12, 24, 48, 72, 120 e 192h. No geral, para as cultivares de pessegueiro estudadas (Capdeboscq e NR0170401) CYP2 e RPII foram os genes que apresentaram maior estabilidade, enquanto que para as ameixeiras ‘Marianna 2624’ e ‘Mirabolano 29-C), RPL13 e RPII foram os mais estáveis, indicando a sua utilização como genes normalizadores. Realizou-se a validação de expressão em LDH1 e ADH1 e estes confirmaram a importância estudos para seleção de genes de referência para atingir os resultados adequados. No terceiro estudo, foram avaliados parâmetros fisiológicos e expressão de nove genes envolvidos nos metabolismos glicolítico, fermentativo e de síntese de etileno em ‘NR0170401’, ‘Marianna 2624 e ‘Mirabolano 29-C’ nas mesmas condições experimentais do segundo estudo. Os três porta-enxertos avaliados ativaram o metabolismo fermentativo já nas seis horas iniciais de experimento, sendo que os tecidos radiculares são mais afetados em comparação com folhas. ‘Marianna 2426’ aparenta ter um sistema de resposta gênico mais eficiente em comparação com as demais cultivares estudadas em condições de privação de oxigênio. The State of Rio Grande do Sul is the largest producer of peaches from Brazil. However, it still has low values of productivity when compared to other States. One of the problems associated to it this is the occurrence of drainage soils problems, mainly in Pelotas and its surroundings, which depending on the time of year, it may suffer flooding situations, potentially hampering the development and productivity such culture. In soil flooding situations, oxygen becomes insufficient for roots, damaging the process of cellular respiration and may lead the cells to an energy crisis. In addition, global climate change is affecting the climate, and such changes are expected to increase the frequency and severity of flood events in many of the agricultural regions. For studies to assist in the selection of flood tolerant genotypes, it is essential to understand the physiological and molecular changes of the plants in situations of oxygen deprivation. Thus, in the present work three studies were conducted, which had as aim the characterization of physiological and molecular responses in five Prunus spp. rootstocks under soil flooding. The first study aimed to obtain the transcriptomic profile on leaves of 'Capdeboscq' peach and 'Julior' plum submitted to flooding for 48 hours using the RNA-Seq technique. From this analysis, 3,011 differentially expressed genes (DEGs) were identified for 'Capdeboscq' (sensitive), which 1,566 were up-regulated and 1,445 down-regulated, and 251 DEGs for 'Julior' (stress tolerant), 125 being up-regulated and 126 down-regulated. 'Capdeboscq' activated signaling pathways, but photosynthesis was the most affected process at physiological and molecular level presenting a higher number of down-regulated enriched Gene Ontology (GO) terms. 'Julior' was more efficient in triggering defense responses, which included the activation of flavonoid biosynthesis pathways, according to KEGG (Kyoto Encyclopedia of Genes and Genomes) metabolic pathways and GOs terms. The LSD transcription factors family seems to be a key point in elucidating the defense mechanisms against stress among the families identified in this study. The protein-protein interaction network of 'Capdeboscq' showed high degrees of interaction and indicate the high probability of some identified nodes being hubs. In the second study, eight candidate genes for expression stability were studied for RT-qPCR studies on leaves and roots of four Prunus spp. submitted to soil flooding by 0, 6, 12, 24, 48, 72, 120 and 192h. In general, for the studied peach cultivars (Capdeboscq and NR0170401), CYP2 and RPII were the most stable genes, whereas for plums 'Marianna 2624' and 'Mirabolano 29-C', RPL13 and RPII were the most stable, indicating their use as reference genes. Expression validation was performed on LDH1 and ADH1 genes and these have confirmed the importance of studies for selection of reference genes to achieve the appropriate results. In the third study, physiological parameters and expression of nine genes involved in glycolytic, fermentative, and ethylene synthesis pathways in 'NR0170401', 'Marianna 2624' and 'Mirabolano 29-C' were evaluated in the same experimental conditions of the second study. The three evaluated rootstocks activated the fermentative metabolism in the first six hours of the experiment, and the root tissues were more affected compared to leaves. 'Marianna 2426' appears to have a more efficient gene response system compared to the other cultivars studied under oxygen deprivation conditions.

This study aimed at determining and correlating the main morphometric characteristics of fruits and seeds with the germination potential and vigor of eight peach rootstocks. The experimental design was completely randomized, with four replications of 50 seeds per treatment. The analyzed variables were: length, width, thickness, fresh mass of fruit and seeds, moisture content of seeds, percentage of seeds attacked by fungi and intact seeds, germination percentage, germination speed index (GSI) and mean germination time (MGT). The Capdeboscq, Aldrighi and Tsukuba 1 cultivars showed higher values of length, width, thickness and fresh mass of seeds compared to other cultivars, presenting a relationship with the later period of fruit ripening. Regardless of the fruit ripening time, the germination percentage was high, ranging between 93% and 100% for all cultivars. In addition, the fresh mass of fruits has showed a high positive correlation with the fresh mass of seeds for the eight evaluated peach rootstocks. The GSI and MGT have a high relationship with the fresh mass of seeds. Resumo: Objetivou-se determinar e correlacionar as principais características morfométricas de frutos e sementes sobre o potencial de germinação e o vigor de oito porta-enxertos de pessegueiro. O delineamento experimental foi inteiramente casualizado com quatro repetições de 50 sementes por tratamento. As variáveis analisadas foram: comprimento, largura, espessura, massa fresca dos frutos e sementes, grau de umidade de sementes, porcentagem de sementes intactas e infectadas por fungos, porcentagem de germinação, índice de velocidade de germinação (IVG) e tempo médio de germinação (TMG). As cultivares Capdeboscq, Aldrighi e Tsukuba 1 apresentaram maiores valores médios de comprimento, largura, espessura e massa fresca de sementes comparados com as demais cultivares, apresentando relação com o período mais tardio de maturação dos frutos. Independente da época de maturação dos frutos, a porcentagem de germinação foi alta variando entre 93% e 100% para todas as cultivares. Somado a isso, a massa fresca do fruto apresenta alta correlação positiva com a massa fresca das sementes dos oito porta-enxertos de pessegueiro avaliados. O IVG e o TMG possuem alta relação com a massa fresca das sementes.

The present study evaluated the germination potential of seeds from eight peach tree rootstocks under different stratification methods, aiming at the production of high quality seedlings. In the first experiment, seeds with endocarp from eight genotypes (‘Aldrighi’, ‘Capdeboscq’, ‘Flordaguard’, ‘Okinawa’, ‘Okinawa roxo’, ‘Tsukuba 1’, ‘Tsukuba 2’, ‘Tsukuba 3’) were stratified on sand and moist vermiculite, at 7 ºC for 60 days. In the second experiment, endocarpless seeds of the same genotypes were stratified on Petri dishes at 7 ºC. In the third experiment, the time at which at least 75% of the seedlings reached transplanting height (15 cm high) was verified. The endocarp was a limiting factor for seed germination of the eight rootstocks, since the highest germination percentages were 21% and 48.3% for ‘Aldrighi’ and ‘Capdeboscq’, respectively. Germination of ‘Okinawa’ was only 19.2% and 4.2%, in sand and vermiculite, respectively. Seeds stratified without the endocarp, showed 100% germination, except for ‘Flordaguard’ and ‘Tsukuba 2’ and 3’, with 93% germination. Seed mass presented a positive correlation with the germination speed index and the initial growth of the seedlings. ‘Aldrighi’ and ‘Capdeboscq’ presented more vigorous seedlings with greater growth in height, stem diameter, and Dickson quality index. Resumo: Objetivou-se avaliar o potencial germinativo de sementes de oito porta-enxertos de pessegueiro, sob diferentes métodos de estratificação, visando à produção de seedlings com alta qualidade. No primeiro experimento, sementes com endocarpo de oito genótipos de pessegueiros (‘Aldrighi’, ‘Capdeboscq’, ‘Flordaguard’, ‘Okinawa’, ‘Okinawa roxo’, ‘Tsukuba 1’, ‘Tsukuba 2’, ‘Tsukuba 3’) foram estratificadas em areia e vermiculita úmida, à 7 ºC por 60 dias. No segundo experimento, sementes sem endocarpo foram estratificadas em placas de Petri, à 7 ºC. No terceiro experimento avaliou-se o tempo em que pelo menos 75% dos seedlings atingissem o ponto de transplantio (15 cm de altura). O endocarpo revelou ser uma barreira limitante na germinação das sementes dos oito porta-enxertos, pois as maiores porcentagens de germinação foram 21% e 48,3%, para ‘Aldrighi’ e ‘Capdeboscq’, respectivamente. A germinação de ‘Okinawa’ foi de apenas 19,2% e 4,2%, em areia e vermiculita, respectivamente. Sementes estratificadas sem o endocarpo, apresentaram 100% de germinação, exceto para ‘Flordaguard’, ‘Tsukuba 2’ e 3’, com 93% de germinação. Verificou-se correlação positiva da massa de sementes com o índice de velocidade de germinação e o crescimento inicial dos seedlings. ‘Aldrighi’ e ‘Capdeboscq’ apresentam seedlings mais vigorosos, com maior crescimento em altura, diâmetro do caule e índice de qualidade Dickson.

Há uma grande carência de pesquisas e literatura no Brasil sobre a reprodução do pessegueiro. A presente revisão de literatura aborda e confronta inúmeros resultados de pesquisas acerca dos principais fatores envolvidos na reprodução dessa frutífera, com o intuito de subsidiar futuras pesquisas que visam ao melhor entendimento e/ou à solução de problemas associados à sua reprodução. Através da presente revisão, foi possível verifi car que o processo de fi xação de frutos se mostra complexo para algumas cultivares, sendo dependente da passagem, com êxito, por cada estádio fenológico. Com isso, a habilidade dos técnicos e produtores em integrar os fatores porta-enxerto, cultivar, ambiente e manejo das plantas é crucial para o sucesso no cultivo dessa espécie frutífera. In Brazil there is a great lack of studies and literature about the peach reproduction. The present review discusses a lot of research results regarding the main factors involved in the reproduction of this species, and it has the intention to help future researches that aims to understand or to resolve the problems associated with the reproduction of the peach tree. By this review it was possible to verify that the process of fruit set is complex for some cultivars and highly dependent on each phenological stage. The ability of the professionals and producers to integrate the factors rootstocks, variety, environment and adequate cultural practices is very important for a successful peach tree production.

Dentre os efeitos prejudiciais causados por estes estresses, destacam-se a diminuição na taxa assimilatória líquida, fechamento de estômatos, redução das atividades celulares, produção de espécies reativas de oxigênio e desestabilização de membranas e proteínas. Tais efeitos possuem uma delicada regulação que transcorre ao nível molecular, onde genes e fatores de transcrição (TFs) modulam a expressão gênica diferencial sob condições estressantes. Desta forma, o objetivo deste trabalho foi investigar em que magnitude os parâmetros de trocas gasosas e o perfil transcriptômico de portaenxertos de Prunus spp. são influenciados diferencialmente sob estresse por seca e alagamento. O trabalho foi dividido em dois experimentos. No primeiro experimento foram avaliados parâmetros de trocas gasosas em três portaenxertos de Prunus spp. („Capdeboscq‟, „Julior‟ e „Marianna 2624‟), nas condições controle, de déficit hídrico e alagamento do solo durante sete dias. No geral, os três portaenxertos demonstraram ser mais suscetíveis ao alagamento do que ao déficit hídrico, apenas variando no tempo de resposta, o qual é intrínseco a cada genótipo. No segundo experimento objetivou-se obter o perfil transcriptômico e avaliar a expressão de TFs em folhas do portaenxerto de pessegueiro „Capdeboscq‟ submetido ao alagamento durante 48 horas, através da técnica de sequenciamento massivo de RNA (RNA-Seq). Desta análise foram identificados 2.971 genes diferencialmente expressos (DEGs), sendo 1.559 up-regulated e 1.412 down-regulated. Foi possível observar forte efeito sob a fotossíntese, a qual apresentou inúmeros genes down-regulated. TFs membros das famílias bHLH, ERF, NAC, WRKY, HD-Zip, NAC e MYB_related foram os mais afetados pelo estresse, apresentando regulação tanto up quanto down-regulated. Among the harmful effects caused by these stresses, the decrease in liquid assimilatory rate, closing of stomata, reduction of cellular activities, production of reactive oxygen species and destabilization of membranes and proteins are included. Such effects have a delicate adjustment that takes place at the molecular level, where genes and transcription factors (TFs) modulate the differential gene expression under stressful conditions. Thus, the aim of this study was to investigate in which magnitude gas exchange parameters and Prunus spp. rootstock transcriptomic are differentially influenced under stress by drought and flooding. The study was divided into two experiments. In the first one, gas exchange parameters were evaluated in three Prunus spp. rootstock (' Capdeboscq ', ' Julior ' and ' Marianna 2624 ') under control conditions of soil water deficit and flooding for seven days. Overall, the three rootstocks have shown to be more susceptible to flooding than water deficit, only varying in response time, which is intrinsic to each genotype. The second one, aimed to obtain the transcriptomic profile and evaluate the TFs expression in peach rootstock leaves ' Capdeboscq ' subjected to flooding during 48 hours, through RNA sequencing (RNA-Seq) technique. This analysis identified 2,971 (DEGs) differentially expressed genes, among them 1,559 were up-regulated and 1,412 were down-regulated. It was possible to observe a strong effect on photosynthesis, which presented numerous down-regulated genes. TFS bHLH families members, ERF, NAC, WRKY, HD-Zip, NAC and MYB_related were the most affected by stress, showing both setting up and down-regulated.

One of the problems related to low productivity are soils with poor drainage and low water storage capacity, mainly in the producing region of Pelotas, as well as the presence of root-knot nematodes (Meloidogyne spp.) on the orchards, whose damages have great importance because the novice action on the root system, affecting the nutrient and water absorption and translocation. These stresses can cause physiological changes in plants affecting fruit quality and productivity of orchards. The objective of this study was to evaluate the effects of these stresses on peach plants of rootstock cv. Capdeboscq, Flordaguard and plum rootstock cv. Mirabolano 29-C. The study was divided into two experiments. At first, we evaluated gas exchange and antioxidant enzymes activity of Prunus rootstocks submitted to the following stresses treatments: a) Waterlogging, for eight days; b) drought for eight days; and c) control, with regular plant watering. After the stresses period, plants returned to normal water regime. The Mirabolano 29-C rootstock shown to be more susceptible to drought than waterlogging, compared to the two peach rootstocks evaluated. On the other hand, Flordaguard and Capdeboscq shown more damages when the plants were subjected to waterlogging, verified by the higher rate of lipid peroxidation in relation to the control plants of these cultivars. The second experiment evaluated the effects of M. javanica plant-inoculation on the reproduction factor (RF) and gas exchange. 150 days after inoculation, the plants of cvs. Capdeboscq, Flordaguard, and Mirabolano C-29 were susceptible (FR = 1.77), resistant, and immune, respectively. In relation to gas exchange, only the inoculated plants of cv. Capdeboscq showed higher stomatal conductance, internal CO2 concentration and transpiration, and reduced efficiency in water use. For other genotypes there were no significant effects on the variables analyzed. O Rio Grande do Sul possui a maior área de produção de pêssego do Brasil, porém sua produtividade ainda é baixa quando comparado com outros estados. Um dos problemas relacionados à baixa produtividade são os solos com problemas de drenagem e baixa capacidade de armazenar água, principalmente na região produtora de Pelotas, assim como a presença de nematóides causadores de galhas nas raízes (Meloidogyne spp.), cujos danos têm grande importância em virtude da ação nociva sobre o sistema radicular, afetando a absorção e a translocação de água e nutrientes. Esses estresses podem causar alterações fisiológicas nas plantas, comprometendo a qualidade dos frutos e a produtividade dos pomares. O objetivo do presente trabalho foi avaliar os efeitos destes fatores de estresse nos portaenxertos de pessegueiro cv. Capdeboscq, Flordaguard e ameixeira cv. Mirabolano 29-C. O trabalho foi dividido em dois experimentos. No primeiro, avaliou-se as trocas gasosas e atividade antioxidante de portaenxertos do gênero Prunus submetidos aos seguintes tratamentos de estresse: a) alagamento do solo, por oito dias b) seca, por oito dias e c) controle, com irrigação conforme necessidade das plantas. Após o período de estresse, as plantas retornaram às condições normais de regime hídrico. O portaenxerto Mirabolano 29-C demonstrou ser mais suscetível ao déficit hídrico do que ao alagamento, quando comparado as duas cultivares de pessegueiro avaliadas. Por outro lado, nas cvs. Flordaguard e Capdeboscq ocorreram maiores danos quando as plantas foram submetidas ao alagamento, verificado por meio da maior taxa de peroxidação lipídica, em relação as plantas controle destas cultivares. No segundo experimento, avaliaram-se os efeitos da inoculação de Meloidogyne javanica sobre o fator de reprodução (FR) e das trocas gasosas. Aos 150 dias após a inoculação, as plantas das cvs. Capdeboscq, Flordaguard e Mirabolano 29-C mostraram-se suscetíveis (FR=1,77), resistentes e imunes, respectivamente. Em relação às trocas gasosas, apenas as plantas inoculadas da cv. Capdeboscq apresentaram maiores valores de condutância estomática, concentração intracelular de CO2 e de transpiração, e uma menor eficiência no uso da água. Para os demais genótipos não houve efeitos significativos nas variáveis analisadas.

In this context, this dissertation addressed the current situation and the prospects of using marker-assisted selection in peach breeding programs as well as to characterize the genetic variability and population structure of an association panel representative of peach germplasm available in Brazil through SSR analysis. Based on a literature review, it was seen that a high number of QTL (Quantitative Trait Loci) is identified in genetic maps of peach, but, despite the importance of the characters studied, the use of markerassisted selection has been incipient by breeding programs. By the other side, with the recent completion of the sequencing of the peach genome, the perspective is that the low use of molecular tools by peach breeding program will change in the coming years. In this trend, the genetic variability present in the association panel representative of peach germplasm in Brazil is quite relevant. On the other hand, this germplasm is structured into two subpopulations according to the fruit flesh type, melting or non-melting, with some genotypes being a mixture of both types of fruit flesh. Greater variability is available in the melting flesh group. These results show that the association panel is suitable for association studies given its relevant genetic variability. However, it should be taken into account in the association studies the genetic structure intrinsic to this germplasm to avoid the identification of false markertrait associations. Com base no avanço do conhecimento de genômica a seleção assistida por marcadores mostra-se como uma estratégia bastante promissora especialmente para acelerar o desenvolvimento de novas cultivares em espécies perenes, como o pessegueiro. Nesse contexto, esta dissertação aborda a situação atual e as perspectivas do uso de seleção assistida por marcadores moleculares no melhoramento genético de pessegueiro, assim como caracteriza a variabilidade genética e a estrutura de população do painel associativo de pessegueiro representativo do germoplasma disponível no Brasil por meio da análise de 10 loci SSR (Simple Sequence Repeats). Com base na literatura, um elevado número de QTLs (Quantitative Trait Loci) está identificado em mapas genéticos de pessegueiro, mas, apesar da importância dos caracteres envolvidos, até o momento o uso de seleção assistida por marcadores tem sido incipiente pelos programas de melhoramento. Por outro lado, com a conclusão recente do sequenciamento do genoma do pessegueiro, a perspectiva é de que essa realidade mude nos próximos anos. Corroborando com essa tendência, o primeiro passo para a seleção assistida por marcadores moleculares é conhecer a variabilidade genética presente no germoplasma de pessegueiro disponível no Brasil. Nesse sentido, os resultados desse trabalho mostram grande variabilidade genética no germoplasma avaliado. Porém, esse germoplasma está estruturado em duas subpopulações bem definidas e agrupadas de acordo com o tipo de polpa, fundente e não-fundente, com alguns acessos apresentando mistura de ambos, entre os quais a maior variabilidade está disponível dentro do subgrupo fundente. Com base nos resultados infere-se que o germoplasma avaliado é adequado para estudos de mapeamento associativo, dada a sua expressiva variabilidade genética, mas deve ser levada em consideração a estrutura genética desse germplasma a fim de evitar a identificação de associações falso-positivas entre marcadores e fenótipos de interesse.

O objetivo do trabalho foi avaliar agronomicamente o sistema de produção integrada (PI) x produção convencional (PC) em pomar de pessegueiro cv. Diamante, no município de Pelotas - RS, comparando-se dados de três anos de avaliação (1999 - 2001), em duas áreas distintas. Os sistemas de produção caracterizaram-se: PC no qual prevalece o manejo e práticas culturais utilizadas normalmente pelo produtor e PI no qual são utilizadas as práticas de manejo definidas nas "Normas para Produção Integrada de Pêssego". As avaliações compreenderam: produção total, classificação dos frutos, danos, análises pós-colheita e resíduos de agrotóxicos. Pela análise da produção por planta e número de frutas produzidas por planta, pode-se verificar que a produção foi maior na PI. Pela avaliação da qualidade das frutas, verificou-se que houve aumento no número de frutas na categoria I, no sistema PI. A ocorrência de danos na colheita por insetos, doenças, pássaros e, muitas vezes, pela interação destes foi maior no sistema PC. Pela análise de agrotóxicos na polpa das frutas, não foram encontrados resíduos nas mesmas, dentro dos limites máximos estabelecidos. As frutas do sistema PI apresentaram, na colheita, maiores firmeza da polpa e teor de acidez, e menor teor de sólidos solúveis totais.The objective of this work was to evaluate agronomically the system of integrated production (IP) x conventional production (CP) in orchard of peach tree cv. Diamante in the rural area of Pelotas, Rio Grande do Sul State, Brazil. The study was conducted during three years (1999- 2001), in two different areas. The production systems were characterized as: in the CP prevails the handling and cultural practices usually used by the producer and IP is where the defined handling practices are used in the "Norms for Integrated Peach Production". The evaluations were: total production, classification of the fruits, damages, analyses post-harvesting and pesticides residues. The analysis of the production kg/tree and number of fruits/tree demonstrated that the production was larger in IP. The evaluation of the quality of the fruits indicated that there was an increasing in the number of fruits in one category in the system IP. The occurrence of damages in the crop for attack of insects, diseases, birds and a lot of times by the interaction of these was larger in CP. The analysis of pesticides residues in the pulp of the fruits demonstrated that there were not found residues, inside of the maximum established limits. The fruits of the system IP presented, in the crop, a larger firmness of the pulp and acidity and smaller level of soluble solids.

No estabelecimento, testaram os meios MS, MS ¾, SH e Villegas, e na multiplicação: SH e MS ¾. O meio MS ¾ foi testado em diferentes concentrações de ágar (4,5; 5,5; 6,5 g.L-1). Avaliaram-se as percentagens de estabelecimento dos explantes, contaminação, oxidação e segmentos não brotados. O meio Villegas apresentou menor oxidação durante o período de estabelecimento in vitro. Com o meio MS ¾, verificou-se maior percentagem de estabelecimento dos explantes. Na fase de multiplicação, avaliaram-se a percentagem de crescimento, a taxa de multiplicação e o número de brotações. The establishment media used were as follow: MS, ¾ MS, SH and Villegas, and the multiplication media used were: SH and ¾ MS. O presente trabalho teve como objetivo estimar os coeficientes de repetibilidade de caracteres relacionados à produtividade do pessegueiro, além de identificar e selecionar clones promissores para o Litoral Sul de Santa Catarina. Foram selecionadas plantas híbridas que apresentavam boa adaptação ao clima, produção, tamanho, sabor e firmeza dos frutos. Essas seleções foram enxertadas sobre o porta -enxerto Okinawa e plantadas no ano de 2005, em três diferentes locais: 1- Estação Experimental de Urussanga, situada a 40 metros de altitude; 2- Propriedade de fruticultor, situada a 200 metros de altitude; 3 - Propriedade de fruticultor, situada a 350 metros de altitude. As mudas foram plantadas no espaçamento de 6 x 1 metros e conduzidas no sistema de V . As avaliações foram realizadas durante os anos de 2007 e 2008. Os coeficientes de repetibilidade foram estimados pelo método da máxima verossimilhança restrita (REML), e a predição dos valores fenotípicos e genotípicos, pela melhor predição linear não viciada (BLUP), por meio do software Selegen-REML/BLUP. Todos os caracteres apresentaram considerável variabilidade genética, com estimativas de coeficientes de repetibilidade (r) variando de média a alta magnitude (0,54 a 0,74) para o caráter massa média dos frutos (MMF) e de baixa a média magnitude (0,22 a 0,39) para o caráter produção de frutos/planta (PTF). Os clones 1770 e 1443 apresentaram bom desempenho na média de todos os locais, enquanto os clones 0470 e 1307 se destacaram no local 1, o clone 1444 no local 2, os clones 0740 e 0926 no local 3 e o clone 1770 nos locais 1 e 2 de avaliação. The aim of the present study was estimate the coefficients of repeatability for productive features in peach, in addition to identify and select promising clones for the south coastal region of the State of Santa Catarina, Brazil. Hybrid plants that showed good adaptation to the climate, production, size, flavor and firmness of fruit were selected. These selections were then grafted on Okinawa rootstock and planted in the year 2005 in three different locations: 1 - Experimental Station of Urussanga, at 40 meters of altitude; 2 - Grower's orchard, at 200 meters of altitude; 3 - Grower's orchard, at 350 meters of altitude. The seedling were planted in a spacing of 6 x 1 meters and conducted in V system. Evaluations were carried out during the years 2007 and 2008. Variance components were estimated by Restricted Maximum Likelihood (REML), and the prediction of phenotypic and genotypic values by Best Linear Unbiased Prediction (BLUP), using the software Selegen-REML/BLUP. All productive features showed considerable genetic variability, with the estimations of repeatability coefficient (r) ranging from medium to high magnitude (0.54 to 0.74) for mean fruit weight (MMF), and from low to medium magnitude (0.22 to 0.39) for the number of fruits per plant (PTF). Clones 1770 and 1443 showed a good performance on average across all sites, while clones 0470 and 1307 performed better in site 1, clone 1444 in site 2, clones 0740 and 0926 in site 3, clone 1770 in sites 1 and 2.

Dentre os principais patógenos que incidem em fruteiras temperadas, destacam-se o Prune dwarf virus (PDV), o Apple mosaic virus (ApMV) e o Grapevine leafroll-associated virus 1 (GLRaV-1). Neste trabalho foram realizadas a detecção e a caracterização molecular dos genes da proteína capsidial de isolados destas três espécies virais. RNAs totais foram extraídos de amostras de folhas de pessegueiros, macieiras e videiras e, nas reações de RT-PCR, foram utilizados oligonucleotídeos específicos para cada espécie viral. Os cDNAs amplificados foram clonados e sequenciados. Foram verificadas altas identidades entre as sequências de nucleotídeos dos genes da proteína capsidial dos isolados brasileiros de PDV, ApMV e GLRaV-1 e isolados de outros países, independente da origem geográfica e da hospedeira. O peso molecular da proteína capsidial destes vírus foi estimado por meio de Western blot em cerca de 24kDa (PDV), 26kDa (ApMV) e 39kDa (GLRaV-1). Among the main pathogens infecting temperate fruit trees are Prune dwarf virus, Apple mosaic virus and Grapevine leafroll-associated virus 1. In this work the detection and molecular characterization of the coat protein genes of isolates from these viral species were carried out. Total RNA was extracted from peach, apple and grapevine leaves and RT-PCR reactions were performed using specific primers to each virus. The amplified cDNA fragments were cloned and sequenced. High identities were observed between coat protein nucleotide sequences of Brazilian isolates of PDV, ApMV and GLRaV-1 and isolates from other countries, independently from geographic origin and host. Coat protein molecular weights of these viruses were estimated by Western blot to be ca. 24kDa (PDV), 26kDa (ApMV) and 39kDa (GLRaV-1).

Objetivou-se avaliar o efeito da fonte e concentração de citocinina e da composição dos sais presentes no meio de cultivo, na multiplicação in vitro do porta-enxerto 'Tsukuba 1' (Prunus persica L.). Explantes cultivados em meio QL/MS com 2,0 mg L-1 de BAP apresentaram maior formação de brotações e crescimento destas. Porém, é necessária a transferência dos explantes para o meio de cultura com redução na concentração de BAP (0,5 mg L-1) para as brotações adquirirem tamanho adequado para a fase de enraizamento. The objective was to assess the font and concentration's effect of cytokinin and salt composition, present in the medium of cultivation, on in vitro multiplication of the rootstock 'Tsukuba 1' (Prunus persica L.). Explants cultivated in QL/MS medium enriched with 2.0 mg/L of BAP showed more shoot formation and growth. However, it is necessary to transfer the explants to culture medium with reduced BAP concentration (0.5 mg/L), so that shoots may obtain the properly size for the rooting phase.

A cultura do pessegueiro na região da Beira Interior representa 23% da área total de pessegueiro de Portugal continental e 16% da área de culturas permanentes da região. Para a caracterização do potencial edáfico onde se desenvolve a cultura, foi efectuado um trabalho de sistematização dos dados referentes às análises de terra, através das Organizações de Agricultores reconhecidas para a prática da Protecção Integrada e Produção Integrada de Prunóideas (AAPIM, APPIZÊZERE e Cooperativa dos Fruticultores da Cova da Beira). O total das 117 análises disponíveis diz respeito ao conjunto de agricultores que exploram uma área de pessegueiros de 463 ha, o que corresponde a um terço da área total ocupada pela cultura na região (1500 ha). O conjunto dos dados analisados permitiu concluir que os solos são maioritariamente ácidos, apresentando textura grosseira, baixo teor de matéria orgânica e elevados teores de fósforo e potássio assimiláveis na camada superficial. Peach orchards in the region of Beira Interior, Portugal, account for 23% of the total Portuguese area of peach trees and 16% of the region fruit crops. The aim of this study is the characterization of the soil of peach orchards by utilizing the data of soils analysis supplied by the farmers’organization of integrated protection and integrated production (AAPIM, APPIZÊZERE and Cooperativa dos Fruticultores da Cova da Beira). The total of 117 soil analysis available corresponds to 463 ha of peach production which represent 1/3 of the total area of peach production in the region (1500 ha). The main results show that soil have a coarse texture, are acid, with low organic mater but high content of phosphorus and potassium.

Este trabalho foi conduzido no Departamento de Fitotecnia da Faculdade de Agronomia Eliseu Maciel, Universidade Federal de Pelotas, em estufa com nebulização intermitente, com o objetivo de verificar o efeito do ácido indolbutírico(AIB) e ethefon (ácido 2-cloroetil fosfônico) no enraizamento de estacas de ramos de pessegueiro. Utilizou-se estacas medianas retiradas de ramos de plantas com seis anos de idade, da cultivar Diamante, com comprimento de 15cm. Efetuou-se duas lesões laterais na base das estacas que foram tratadas com ácido indolbutírico nas concentrações 0; 1000; 2000 e 3000ppm, na forma líquida e, ethefon nas concentrações de 0; 50; 100 e 150ppm. Após a imersão de 5 segundos, acondicionou-se as estacas em sacos de polietileno contendo como substrato a mistura de areia+cinza (2:1 v/v). Pelos resultados obtidos concluiu-se que o ácido indolbutírico aumentou o percentual de estacas realizadas até a concentração de 1752ppm. e o número de raízes até a concentração de 1791ppm. O ethefon não apresentou resultados significativos na forma e concentração utilizadas. This work was conducted at the Plant Science Departament Eliseu Maciel Agronomy College of Federal University of Pelotas, in greenhouse with intermitent mist, aiming to evaluate the effect of indolebutyric acid and ethephon (2-chloroetyl phosphonic acid) on peach hranch cutting rooting. Median cuttings with 15cm length removed from Diamante cultivar branch were used. Two lateral lesions were made on cutting basis and they were treated with indolebutyric acid in liquid at 0; 1000; 2000; and 3000ppm concentrations and ethefon at 0; 50; 100 and 150ppm concentrations. The immersion time was five seconds and the cuttings were packed in polietylene sacks containing a mixture of sand and rice husk ash substrate (2:1 v/v). Results showed that the indolebutyric acid increased the rooted cuttings percentage up to 1752ppm concentration and the number of roots until 1791ppm concentration. The ethephon did not show significant variation for the form and concentrations used.

A seletividade de agrotóxicos indicados nas Normas Técnicas Específicas da Produção Integrada de Pêssego (NTE-PIP) foi avaliada sobre Trichogramma pretiosum Riley, utilizando-se da metodologia padronizada internacionalmente pela International Organization for Biological and Integrated Control of Noxious Animals and Plants (IOBC). Os agrotóxicos (% ingrediente ativo na calda) Orthocide 500 (0,120) e Persist SC (0,160) foram classificados como inócuos; Hokko Cihexatim 500 (0,025) e Roundup WG (1,387) foram levemente nocivos; Assist (1,512) foi moderadamente nocivo; Sevin 480 SC (0,173), Tiomet 400 CE (0,048), Dipterex 500 (0,150) e Kumulus DF (0,480) foram nocivos a adultos de T. pretiosum no teste de toxicidade inicial em laboratório. The side-effects of pesticides indicated in Technical Norms of the Integrated Production of Peache (NT-IPP) was assessment on Trichogramma pretiosum Riley, using the International Organization for Biological and Integrated Control of Noxious Animals and Plants (IOBC) standard methodology. The pesticides (% active ingredient) Orthocide 500 (0.120) and Persist SC (0.160) were considered harmless; Hokko Cihexatim 500 (0.025) and Roundup WG (1.387) were slightly harmful; Assist (1.512) was moderately harmful; Sevin 480 SC (0.173), Tiomet 400 CE (0.048), Dipterex 500 (0.150) and Kumulus DF (0.480) were harmful to parasitoids in inicial toxicity test on egg parasitoid T. pretiosum.

O objetivo deste trabalho foi avaliar o estabelecimento in vitro de cinco cultivares de porta-enxerto de Prunus sp. em variações de dois meios de cultura. O material vegetal foi obtido de plantas-matrizes cultivadas em telado, sendo a desinfestação realizada em soluções à base de álcool e hipoclorito de sódio. Utilizou-se um esquema fatorial 5 x 2 (cultivares Aldrighi, Barrier, Capdeboscq, Flordaguard e GF677 x formulações salinas ½MS e QL), distribuído em blocos casualizados, compostos por quatro repetições, com cinco tubos de ensaio cada uma, sendo inoculado um segmento nodal por tubo. O cultivo dos explantes foi realizado por quatro semanas, em ambiente com 20 µEm-2s-1, 24 ± 4ºC e fotoperíodo de 16 horas. Independentemente do meio de cultivo, os porta-enxertos 'Barrier', 'Capdebosq' e 'Flordaguard' apresentaram maiores porcentagem de explantes estabelecidos e número de brotações, sendo os mais responsivos in vitro. A formulação salina ½MS foi a mais indicada para o estabelecimento da cultivar GF677, enquanto a QL para a 'Aldrighi'. Não foram observadas plântulas com sintomas de clorose, vitrificação ou encarquilhamento, indicando que as condições de cultura utilizadas foram satisfatórias. As maiores porcentagens de contaminação foram verificadas nas cultivares Aldrighi (72%) e Capdeboscq (41%), as quais são as mais utilizadas na região Sul do Brasil. The aim of this work was to establish five Prunus sp. rootstocks on two media cultures. The explants were obtained from mother plants cultivated in greenhouse. Desinfestation was carried out in alcohol and sodium hypochlorid solutions. The experiment was a factorial with five cultivars ('Aldrighi', 'Barrier', 'Capdeboscq', 'Flordaguard' and 'GF677') and two culture media (½MS and QL). The experimental design was of completely randomized blocks, with four replications with five test tubes per plot. A nodal segment was introduced per test tub. The explant culture was maintained at 24 ± 4ºC, 20 µE m-2 s-1 and 16 hours photoperiod for four weeks. Independently of culture medium, the rootstocks 'Barrier', 'Capdeboscq' and 'Flordaguard' showed higher in vitro response rate and higher number of shoots. ½MS medium was indicated to 'GF677' establishment and QL medium to 'Aldrighi'. In vitro abnormal plantlets were not observed, suggesting satisfactory conditions to culture. The higher contamination rate was verified in 'Aldrighi' (72%) and 'Capdeboscq' (41%), that are the rootstocks most used in southern Brazil.

No Brasil, a falta de porta-enxertos para as Prunáceas, principalmente de origem clonal, tem incentivado a seleção de novas variedades e o uso de técnicas de cultura in vitro para a propagação. Neste sentido, o objetivo deste trabalho foi avaliar o potencial de multiplicação in vitro do porta-enxerto 'Carelli' sob efeito de diferentes concentrações da citocinina 6-benzilaminopurina (BAP). Segmentos nodais com 0,5 cm de comprimento foram inoculados em meio de cultura de Lepoivre, suplementado com 0,0; 0,5; 1,0; 2,0 e 4,0 mg.L-1 BAP. Estes segmentos nodais são oriundos de plântulas preestabelecidas in vitro, após duas subculturas em meio de cultura de Lepoivre, suplementado com 0,5 mg.L-1 de BAP. As avaliações para número de brotos por explante e altura média das brotações foram realizadas após 21 dias de cultura in vitro. Os resultados mostraram que os tratamentos com BAP não apresentaram diferenças significativas entre si. A taxa média de multiplicação foi de 3,3 a 3,4 brotos por explante. O tratamento sem adição de BAP não apresentou a formação de brotações axilares, mas resultou em brotos com maior altura média (16,2 mm). O uso de BAP afetou significativamente a altura das brotações, e o acréscimo nas suas concentrações reduziu o comprimento das mesmas. Concentrações de BAP superiores a 1,0 mg.L-1de BAP reduziram o comprimento das brotações e promoveram hiperidricidade. O uso de 0,5 mg.L-1 de BAP promoveu a formação de 3,3 brotos por explante com 11,0 mm de altura média, em condições adequadas para o enraizamento. In Brazil the lack of well adapted rootstocks in the Peach industry, mainly of clonal origin, forces the selection of new varieties and the use of tissue culture techniques for the mass clonal propagation. In the present work it was evaluated the in vitro multiplication potential of "Carelli" rootstock in response to different levels of BA. Nodal segments (0.5 cm length) were inoculated in test tubes containing 20 ml of Lepoivre culture medium supplemented with 0.0; 0.5; 1.0; 2.0 and 4.0 mg.L-1 BA. This nodal segments were originated from in vitro pre-established plants after two subcultures in Lepoivre culture medium supplemented with 0.5 mg.L-1 BA. Evaluations done after 21 days in culture showed that the different levels of BA resulted in values statistically similar for the multiplication rate and length of regenerated shoots. The values for multiplication rate ranged from 3.3 to 3.4 shoots/explant. The treatment control did not result in multiple shoots but showed shoots with the highest lengh (16.2 mm). BA significativelly affected the length of shoots and the increase in its levels reduced the shoot lengh. BA levels higher than 1.0 mg.L-1 reduced the shoot length and promoted vitrification. BA in the level of 0.5 mg.L-1 resulted in the induction of 3.3 shoots/explant with means values of 11.0 mm in length in adequate conditions for rooting.

As avaliações compreenderam: produção total, classificação dos frutos, danos, análises pós-colheita e resíduos de agrotóxicos. Pela análise da produção por planta e número de frutas produzidas por planta, pode-se verificar que a produção foi maior na PI. Pela avaliação da qualidade das frutas, verificou-se que houve aumento no número de frutas na categoria I, no sistema PI. A ocorrência de danos na colheita por insetos, doenças, pássaros e, muitas vezes, pela interação destes foi maior no sistema PC. Pela análise de agrotóxicos na polpa das frutas, não foram encontrados resíduos nas mesmas, dentro dos limites máximos estabelecidos. As frutas do sistema PI apresentaram, na colheita, maiores firmeza da polpa e teor de acidez, e menor teor de sólidos solúveis totais. The objective of this work was to evaluate agronomically the system of integrated production (IP) x conventional production (CP) in orchard of peach tree cv. Diamante in the rural area of Pelotas, Rio Grande do Sul State, Brazil. The study was conducted during three years (1999- 2001), in two different areas. The production systems were characterized as: in the CP prevails the handling and cultural practices usually used by the producer and IP is where the defined handling practices are used in the "Norms for Integrated Peach Production". The evaluations were: total production, classification of the fruits, damages, analyses post-harvesting and pesticides residues. The analysis of the production kg/tree and number of fruits/tree demonstrated that the production was larger in IP. The evaluation of the quality of the fruits indicated that there was an increasing in the number of fruits in one category in the system IP. The occurrence of damages in the crop for attack of insects, diseases, birds and a lot of times by the interaction of these was larger in CP. The analysis of pesticides residues in the pulp of the fruits demonstrated that there were not found residues, inside of the maximum established limits. The fruits of the system IP presented, in the crop, a larger firmness of the pulp and acidity and smaller level of soluble solids.

RESUMO Foi conduzido experimento para avaliar a eficiência agronômica de Triflumizole, Dicloran, Procymidone e Thiabendazole, em diferentes doses, no controle da podridão parda do pessegueiro, cultivar Diamante. Foi utilizado o delineamento experimental blocos ao acaso com quatro repetições. A unidade experimental foi composta por dez frutos. Os produtos foram aplicados através da imersão dos frutos na calda fungicida. Após, os frutos foram inoculados por meio de ferimento superficial e com inóculo obtido de frutos infectados a campo. Os parâmetros utilizados para avaliação foram o número de frutos infectados, relação entre o diâmetro total do fruto; presença de resíduo de fungicida visível no tegumento dos frutos. Os resultados obtidos permitiram concluir que Trifumizole (100g/100 litros), Dicloran, Procymidone e Thiabendazole foram eficientes no controle do patógeno. Apenas Dicloran apresentou deposição de resíduos visíveis no tegumento dos frutos. SUMMARY An experiment to evaluate the agronomic efficiency of Triflumizol, Dicloran, Procymidone and Thiabendazol and different doses for peach brown rot control of "Diamante" cultivar was conducted. The experimental design was complete random block, four replications and ten fruits/plot. The fungicides applications was done by fruits immersion. The fruits were inoculated with material directily obtained from diseased fruits after fungicide treatment The parameters utilized for evaluation were number of infected fruits, lesion diameter and fruit total diameter rate, residue presence over the fruits. The results obtained showed that Triflumizol (100g/100 liters), Dicloran, Procymidone and Thiabendazol were efficient on pathogen control and only Dicloran showed residue deposition on fruits.

Experimentos testando o tipo de auxina (AIB, AIA e ANA) e concentração (0; 0,01; 0,1 e 1,0 mM) foram realizados para cada um dos quatro porta-enxertos utilizados (G x N22, Mr.S 2/5, Marianna e Mirabolano). Cada porta-enxerto constituiu-se num experimento, uma vez que, apesar de ser o mesmo meio básico, MS ¾, com exceção de Marianna, onde o meio foi o MS, as concentrações da citocinina BAP foram diferentes. A cada meio, foram adicionados os tipos e as concentrações das auxinas a serem testadas. Os porta-enxertos apresentaram comportamentos distintos quanto ao tipo e concentração das auxinas. O AIB e o AIA foram superiores ao ANA nas variáveis analisadas. Na fase de multiplicação, o 'Mirabolano' não respondeu a concentrações de auxinas de até 1,0 mM. O 'G x N22' apresentou as melhores respostas na concentração de 0,01 miM de AIB ou AIA. O porta-enxerto Marianna respondeu positivamente a concentrações de auxinas entre 0,01 e 1,0 miM. O ANA não proporciona bons resultados na multiplicação in vitro de porta-enxertos do gênero Prunus, sendo muitas vezes superado pela testemunha. The experiments were carried out by testing the type of auxin (IBA, IAA and NAA) at four concentrations (0; 0.01; 0.1 and 1.0 muM) for each one of the four used rootstocks (G x N22, Mr.S 2/5, Marianna and Mirabolano). Each rootstock performed a trial with the same basic medium (MS ¾) except for Marianna, which was carried out with MS; the BAP concentrations were differents. To each medium, the different types and auxin concentrations were added to be tested. The rootstocks performed differently in relation to the type and auxin concentrations. The auxins IBA and IAA provided higher values than NAA for all the analyzed parameters. IBA or IAA provided the best responses at 0.01 mM for all the analyzed parameters of the rootstock G x N22. Marianna showed the best responses at 0.01 - 1.0 muM. Mirabolano rootstock, had no significantly response to the type and auxin concentration. NAA, did not provide good results for in vitro multiplication of Prunus rootstocks, and it was overcome as a whole by the control.

A cultura do pessegueiro na região da Beira Interior representa 23% da área total de pessegueiro de Portugal continental e 16% da área de culturas permanentes da região. Para a caracterização do potencial edáfico onde se desenvolve a cultura, foi efectuado um trabalho de sistematização dos dados referentes às análises de terra, através das Organizações de Agricultores reconhecidas para a prática da Protecção Integrada e Produção Integrada de Prunóideas (AAPIM, APPIZÊZERE e Cooperativa dos Fruticultores da Cova da Beira). O total das 117 análises disponíveis diz respeito ao conjunto de agricultores que exploram uma área de pessegueiros de 463 ha, o que corresponde a um terço da área total ocupada pela cultura na região (1500 ha). O conjunto dos dados analisados permitiu concluir que os solos são maioritariamente ácidos, apresentando textura grosseira, baixo teor de matéria orgânica e elevados teores de fósforo e potássio assimiláveis na camada superficial.Peach orchards in the region of Beira Interior, Portugal, account for 23% of the total Portuguese area of peach trees and 16% of the region fruit crops. The aim of this study is the characterization of the soil of peach orchards by utilizing the data of soils analysis supplied by the farmers’organization of integrated protection and integrated production (AAPIM, APPIZÊZERE and Cooperativa dos Fruticultores da Cova da Beira). The total of 117 soil analysis available corresponds to 463 ha of peach production which represent 1/3 of the total area of peach production in the region (1500 ha). The main results show that soil have a coarse texture, are acid, with low organic mater but high content of phosphorus and potassium.
