Neste trabalho, as raízes de batata-baroa (Arracacia xanthorrhiza Bancroft) foram armazenas em câmaras frias, à temperatura de 5 e 10ºC, por 28 dias. A cada sete dias, as raízes eram retiradas da condição de frio, e realizada análise visual dos sintomas de injúria por frio. Em seguida, as raízes foram maceradas para determinação das atividades da peroxidase, polifenoloxidase e da concentração dos compostos fenólicos solúveis. As temperaturas de 5 e 10ºC estimularam o escurecimento externo e interno das raízes durante o armazenamento, com maior intensidade para a temperatura de 5oC. Em ambas as temperaturas, houve elevação da atividade da polifenoloxidase, peroxidase e da concentração de compostos fenólicos, a partir da exposição dos tecidos ao frio. A atividade da polifenoloxidase e a concentração de compostos fenólicos solúveis aumentaram após o 14º dia de exposição às duas temperaturas. A atividade da peroxidase aumentou até o 7º dia de armazenamento em ambas as temperaturas, mantendo-se praticamente constante, após este período, a 5 e a 10ºC, a maior atividade ocorreu no 14º dia de armazenamento. Estes resultados indicam atuação inicial da peroxidase como uma resposta inicial ao estresse causado pela colheita e o frio e posterior participação de polifenoloxidase no escurecimento enzimático e acúmulo de compostos fenólicos nos tecidos. In this experiment roots of arracacha (Arracacia xanthorrhiza Bancroft) were stored at temperature of 5 and 10oC for 28 days. At every seven days, the roots were removed from the cold storage and visually analyzed for the presence of chilling symptoms. Afterwards, the roots were grinded for polyphenoloxidase, peroxidase activities and soluble phenolic compounds. Both temperature of 5 and 10oC induced external and internal darkening of the roots, with higher intensity at 5oC. Activities of polyphenoloxidase, peroxidase and content of phenolic compounds were enhanced at cold storage. Activity of polyphenoloxidase and content of phenolic substances increased up to the 14th day of storage in both temperatures. The activity of peroxidase was elevated up to the seventh day of storage in both temperatures, which was kept constant at 5 and at 10oC. The highest activity was detected at the 14th day of storage. These results show that peroxidase is involved in the initial stress response to the harvest and chilling, followed by the involvement of polyphenoloxidase in the enzymatic darkening and accumulation of phenolic compounds in the tissues.

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior The Yellow Peruvian root is a vegetable of subtropical origin, whose tuber roots are highly perishable when stored at room temperature, requiring storage under refrigeration to extend the postharvest life. However, low temperatures of storage can induce the development of chilling symptoms, including the browning of the roots. In many species, it has been observed that the browning occurs due action of peroxidase and polyphenoloxidase. The objective of this work was to evaluate the role of peroxidase and polyphenoloxidase on the development of browning induced by chilling inducing temperatures and to study the behavior of these enzymes under different conditions of pH and temperature, determining optimal conditions for activity and possible ways for inactivation. The influence of the enzymes peroxidase and polyphenoloxidase in the enzymatic browning caused by the temperatures was determined by the visual analysis of the roots, determination of peroxidase and polyphenoloxidase activities, and by the total soluble phenolic concentration in roots stored at 5ºC and 10ºC. The influence of pH and temperature on the activity of enzymes were done in an ammonium sulphate saturated (0 80%) crude extract. In both, at 5ºC and 10ºC had an increase in the root browning, in the activity of peroxidase and polyphenoloxidase, and total phenolics content while being stored. The activity of the enzyme peroxidase increased fast up to the 7º day of storage in both the temperatures, remaining practically constant after this period at 5ºC until the 28º day of storage. The temperature of 10ºC showed higher activity at 14º day of storage, with posterior decline. The activity of polyphenoloxidase and the soluble phenolics concentration increased dramatically after the 14º day of exposition to both temperatures. These results indicate a probable involvement of the enzymes peroxidase and polyphenoloxidase in the browning, however, these two enzymes may not be the only responsible factors acting on the browning. Peroxidase had higher activity when the reaction was performed between the pH 5.5 and pH 6.0 and temperature of 30ºC; its inactivation was accomplished at pH 2.5 for 60 minutes and the temperature of 60ºC for 10 minutes or 70ºC, for 5 minutes. Polyphenoloxidase presented higher activity when the reaction was done at pH 7.5 and temperature of 30ºC. This enzyme was not inactivated by incubation in pH 2.5 or 9.0, but it was inactivated after 5 minutes at 80ºC. A batata-baroa é uma hortaliça de origem subtropical, cujas raízes tuberosas são altamente perecíveis em condições de temperatura ambiente, necessitando de armazenamento sob refrigeração para prolongar a durabilidade pós-colheita. Entretanto, baixas temperaturas de armazenamento podem acarretar o surgimento de sintomas de injúria por frio, dentre eles, o escurecimento das raízes. Em muitas espécies, têm-se observado que o escurecimento ocorre devido à atuação das enzimas peroxidase e polifenoloxidase. O objetivo deste trabalho foi avaliar a participação das enzimas peroxidase e polifenoloxidase no escurecimento provocado por baixas temperaturas causadoras da injúria por frio e verificar possíveis variações no comportamento dessas enzimas sob condições de pH e temperatura, determinando as condições ótimas da atividade e possíveis formas de inativação. A influência das enzimas peroxidase e polifenoloxidase no escurecimento enzimático ocasionado pelo armazenamento em temperaturas que causam injúria por frio, foi determinada por meio da análise visual das raízes, da determinação da atividade das enzimas peroxidase e polifenoloxidase, e da concentração de compostos fenólicos solúveis em raízes submetidas ao armazenamento a 5ºC e 10ºC. A influência do pH e da temperatura sobre atividade das enzimas foi feita via saturação do extrato bruto com sulfato de amônio (0 a 80%). Tanto a temperatura de 5ºC quanto de 10ºC houve progressivo aumento no grau de escurecimento, na atividade da peroxidase e da polifenoloxidase, e da concentração de compostos fenólicos durante o armazenamento. A atividade da enzima peroxidase aumentou expressivamente até o 7º dia de armazenamento em ambas as temperaturas, mantendo-se praticamente constante após esse período à temperatura de 5ºC até o 28º dia de armazenamento. A temperatura de 10ºC houve maior atividade, no 14º dia de armazenamento, com posterior declínio. A atividade da polifenoloxidase e a concentração de compostos fenólicos solúveis aumentaram acentuadamente após o 14º dia de exposição às duas temperaturas. Esses resultados indicam a atuação das enzimas peroxidase e polifenoloxidase no escurecimento, porém, essas duas enzimas não seriam os únicos fatores responsáveis pelo mesmo. A peroxidase teve maior atividade quando a reação ocorreu entre pH 5,5 e 6,0, e sob temperatura de 30ºC; houve inativação da enzima a pH 2,5 após 60 min de exposição, e às temperaturas de 60ºC, após 10 minutos, e 70ºC, após 5 minutos. A polifenoloxidase apresentou maior atividade quando a reação ocorreu em pH 7,5 e à temperatura de 30ºC. Essa enzima não foi inativada pela incubação em pH 2,5 ou 9,0, porém foi inativada após 5 minutos a 80 ºC.
