Pertusaria jurana var. confluens Erichsen (1935 [1936]: 678) Type:���[Switzerland] SCHWEIZ, Jura, Neuenburg, an Wegulmen bei Buttes, 800 m., 03.10.1928, leg. E. Frey (367) (holotype HBG!). Figure 3. The thallus of the type specimen of Pertusaria jurana var. confluens in HBG is quite thick, whitish-grey and sorediate, with a whitish to brownish prothallus. Soralia are grey-whitish, abundant, orbicular to irregular, discrete, and separated from each other (see Sipman 2006) (Figure 3). The presence of variolaric acid has been detected by TLC. The type specimen Pertusaria jurana var. confluens is morphologically and chemically concordant with Ochrolechia turneri (Smith 1801: 857) Hasselrot (1945: 130) (Kukwa 2011). Therefore it should be treated as a synonym of the latter name (Turland et al. 2018). According to Kukwa (2011) there are two other corticolous and sorediate Ochrolechia species with variolaric acid, O. alboflavescens (Wulfen, in Jacquin 1791 [1789]: 111) Zahlbruckner (1927 [1926]: 94) and O. microstictoides R��s��nen (1936: 226), which resemble O. turneri. Ochrolechia albofavescens has often well-separated soralia, like O. turneri, but it contains in addition to variolaric acid also fatty acids, lichesterinic and protolichesterinic acids. Ochrolechia microstictoides is very variable in thallus morphology, but in general its soralia are irregular and often confluent, but can be distinguished by the presence of lichesterinic acid, which is produced in addition to variolaric acid (Kukwa 2011). Another species which may produce variolaric acid is Varicellaria hemisphaerica (Fl��rke 1815: 6) I. Schmitt & Lumbsch in Schmitt et al. (2012: 29). The species is characterized by the pale bluish-grey tinged thallus with an often broad white margin and conspicuous, paler or concolorous, convex soralia. Pertusaria inopinata Erichsen (1936: 99) Type:���[GERMANY, Schleswig-Holstein] Bramsted, erratische Bl��cke bei Bim��hlen, auf einer Weide, 27.05.1915, leg. Erichsen (lectotype HBG ���designated here, Mycobank MB 10000452). Figure 1. The type specimen of Pertusaria inopinata is characterized by a hardly rimose, dark grey to olive-greenish, not areolated thallus, with numerous irregular, rounded, 0.2���0.3 mm wide, grey-whitish to olive-greenish soralia (Figure 1). According to Erichsen (1936) the thallus reacts P+ yellow, K+ yellow, C���and I���. Norstictic acid and traces of lecanoric and gyrophoric acids were detected by own chemical analyses. This name was reported from Denmark (S��chting & Alstrup 2002), Germany (Erichsen 1936) and Poland (Alstrup & Olech 1992). The type of Pertusaria inopinata is morphologically and chemically concordant with Lepra excludens (Nylander 1885: 296) Hafellner, in Hafellner & T��rk (2016: 172) (syn. Pertusaria excludens Nyl.). Therefore P. inopinata should be considered as a synonym of L. excludens. During herbarium studies three original specimen of P. inopinata were found in HBG, one collected by Erichsen and annotated as ���Typus��� consists of two pieces of stone with the same thallus (Figure 1). The other two were cited in the protologue, but collected by Lettau and Schade. All samples studied correspond in morphological and anatomical characters with the description provided by Erichsen (1936). The chemistry of the type specimen annotated as ���typus��� and other samples were checked by P. James and norstictic acid and traces of lecanoric and gyrophoric acid were detected (Hanko 1983). The presence of these secondary metabolites is confirmed by own research. Hanko (1983) cited as a ���typus��� the specimen collected by Schade (��� DDR. Sachsen: Leusitzer Niederung 7.1936, Schade, HBG-1213���), but the data presented by Hanko (1983: 201) do not match those given in the protologue by Erichsen (1936: 99), therefore, his typification cannot be considered effective, and consequently, a new lectotype of P. inopinata is designated (Turland et al. 2018). The lichen-forming genus Pertusaria under its current circumscription is polyphyletic and its phylogenetic ailiations are uncertain. Here we study the species of the genera Pertusaria and Varicellaria which contain lecanoric acid as major constituent, have disciform apothecia, strongly amyloid asci, non-amyloid hymenial gel, 1-2-spored asci, and 1- or 2-celled ascospores with thick, 1-layered walls. We infer phylogenetic relationships using maximum likelihood and Bayesian analyses based on four molecular loci (mtSSU, nuLSU rDNA, and the protein-coding, nuclear RPB1 and MCM7 genes). Our results show that the lecanoric acid-containing species form a well-supported, monophyletic group, which is only distantly related to Pertusaria s.str. he phylogenetic position of this clade is unclear, but placement in Pertusaria s.str. is rejected using alternative hypothesis testing. he circumscription of the genus Varicellaria is enlarged to also include species with non-septate ascospores. Seven species are accepted in the genus: Varicellaria culbersonii (Vezda) Schmitt & Lumbsch, comb. nov., V. hemisphaerica (Flörke) Schmitt & Lumbsch, comb. nov., V. kasandjeii (Szatala) Schmitt & Lumbsch, comb. nov., V. lactea (L.) The phylogeny and taxonomic placement of three species currently placed in the genus Pertusaria with gyalectoid ascomata were studied using maximum likelihood and Bayesian analysis of four molecular loci (mitochondrial SSU, nuclear LSU rDNA and the protein-coding, nuclear RPB1 and MCM7 genes). A total of 40 new sequences were generated for this study and aligned with 84 sequences retrieved from Genbank. Our results show that the gyalectoid Pertusaria species are only distantly related to Pertusaria s.str. They form a strongly supported sister-group relationship to Coccotrema. Consequently, the new genus Gyalectaria Schmitt, Kalb & Lumbsch is described in Coccotremataceae to accommodate these species and the new combinations G. diluta (C. Bjork, G. Thor & T. Wheeler) Schmitt, T. Sprib. & Lumbsch, G. gyalectoides (Vezda) Schmitt, Kalb & Lumbsch, and G. jamesii (Kantvilas) Schmitt, Kalb & Lumbsch are proposed. Abstract: The phylogeny of Agyriaceae was investigated using MP and Bayesian approaches based on a combined dataset of nuLSU rDNA, mtSSU rDNA, and RPB1 sequences of 78 ascomycetes. The type genus of the family is shown to be a strongly supported sister to Coccotremataceae + Pertusariaceae, whereas the remaining species currently classified in Agyriaceae have a well-supported sister-group relationship with Baeomycetales. Monophyly of Agyriaceae is significantly rejected using two independent alternative topology tests. The micromorphology in Agyriaceae s. lat., Coccotremataceae, and Pertusariaceae is restudied. It is confirmed that the ascus type of Agyrium agrees with that of other taxa currently placed in Agyriaceae, and hence the ascus type is interpreted as homoplasious and phylogenetically misleading in this group of fungi. Consequently, we suggest that Trapeliaceae be resurrected for the lichenized taxa currently classified in Agyriaceae, and that this family be placed in Baeomycetales. The ordinal classification in Ostropomycetidae, especially the circumscription of Pertusariales and its distinction from Agyriales, requires additional studies. [Copyright &y& Elsevier]

Pertusaria islandica sp. nov. is characterized by pertusariate to pseudolecanorate ascomata, ellipsoid ascospores, 8-spored asci, a strongly K+ violet reacting epithecium, and the presence of the 2′-O-methylperlatolic acid chemosyndrome, while P. occidentalis sp. nov. has pertusariate ascomata, globose to subglobose ascospores, 8-spored asci, and contains the arthothelin chemosyndrome. Abstract: Lichenized fungi synthesize a great variety of secondary metabolites. These are typically crystalline compounds, which are deposited extracellularly on the fungal hyphae. While we know a lot about the chemical properties and structures of these substances, we have very little information on the molecular background of their biosynthesis. In the current study we analyze the diversity of non-reducing polyketide synthase (PKS) genes in members of the lichenized Pertusariales. This order primarily contains fully oxidized secondary metabolites from different substance classes, and is chemically and phylogenetically well studied. Using a degenerate primer approach with subsequent cloning we detected up to five non-reducing PKS sequences in a single PCR product. Eighty-five new KS sequence fragments were obtained for this study. Analysis of the 157 currently available fungal KS sequence fragments in a Bayesian phylogenetic framework revealed 18 highly supported clades that included only lichenized taxa, only non-lichenized taxa, or both. Some Pertusarialean groupings of PKS sequences corresponded partly to phylogenetic groupings based on ribosomal DNA. This is reasonable, because a correlation between well-supported phylogenetic lineages and the occurrence of secondary metabolites in the Pertusariales has been observed before. However, no clear linkage was found between the PKS genes analyzed and the ability to produce a particular secondary substance. Several PKS clades did not reveal obvious patterns of secondary compound distribution or phylogenetic association. Compared with earlier phylogenetic analyses of KS sequences the increased sampling in the current study allowed us to detect many new groupings within the fungal non-reducing PKSs. Eight species of the lichen genus Pertusaria (Pertusariaceae) are reported from the Juan Fernandez Archipelago (Chile). Pertusaria fernandeziana is described as a new species and two species, Pertusaria phlyctaenula Nyl. and Pertusaria dehiscens Müll. Arg., are recorded for the first time from the archipelago. Range extensions of the known distribution within the archipelago are reported for Pertusaria hadrocarpa Zahlbr., Pertusaria melanospora Nyl., Pertusaria pachythallina (Räsänen) Messuti, Pertusaria papillulata Nyl., and Pertusaria velata (Turner) Nyl. Pertusaria polycarpa Kremp. vat. monospora Zahlbr. is reduced to synonymy with Pertusaria papillulata and Pertusaria chilena Zahlbr. is synonymized with Pertusaria velata. The Pertusariaceae is a diverse, cosmopolitan group of crustose lichen-forming fungi. It is particularly rich in secondary metabolites, containing a variety of compounds from different substance classes, such as xanthones, depsides, depsidones, and depsones. Morphology and chemistry-based studies have provided conflicting views of relationships among taxa, and phylogenetic relationships among genera in the Pertusariaceae are still unsettled. To evaluate these relationships, we generated a phylogeny based on nucleotide sequences of the nuclear large subunit (nu LSU) and the mitochondrial small subunit (mt SSU) rRNA genes. For a subset of taxa, we additionally sequenced the mitochondrial large subunit (mt LSU) rDNA. We studied a total of 49 taxa from the family Pertusariaceae and the enigmatic genus Loxosporopsis including four Agyrialean taxa used as outgroups. The alignments were analyzed using a Bayesian approach with Markov Chain Monte Carlo tree sampling (B/MCMC), and maximum parsimony methods. In the resulting phylogenetic estimates the genus Pertusaria in its current circumscription is polyphyletic comprising three major clades: the Pertusaria s.str.-group, the Variolaria-group and the Varicellaria-group. Loxosporopsis is closely related to Pertusaria s.str. We re-analyzed morphological, anatomical, and chemical features in the light of the molecular study and found novel character combinations to describe monophyletic entities. The taxonomic significance of particular secondary chemical constituents in the Pertusariaceae is corroborated. [Copyright &y& Elsevier]

The azooxanthellate scleractinian coral Lophelia pertusa has a near-cosmopolitan distribution, with a main depth distribution between 200 and 1000 m. In the northeast Atlantic it is the main framework-building species, forming deep-sea reefs in the bathyal zone on the continental margin, offshore banks and in Scandinavian fjords. Recent studies have shown that deep-sea reefs are associated with a highly diverse fauna. Such deep-sea communities are subject to increasing impact from deep-water fisheries, against a background of poor knowledge concerning these ecosystems, including the biology and population structure of L. pertusa. To resolve the population structure and to assess the dispersal potential of this deep-sea coral, specific microsatellites markers and ribosomal internal transcribed spacer (ITS) sequences ITS1 and ITS2 were used to investigate 10 different sampling sites, distributed along the European margin and in Scandinavian fjords. Both microsatellite and gene sequence data showed that L. pertusa should not be considered as one panmictic population in the northeast Atlantic but instead forms distinct, offshore and fjord populations. Results also suggest that, if some gene flow is occurring along the continental slope, the recruitment of sexually produced larvae is likely to be strongly local. The microsatellites showed significant levels of inbreeding and revealed that the level of genetic diversity and the contribution of asexual reproduction to the maintenance of the subpopulations were highly variable from site to site. These results are of major importance in the generation of a sustainable management strategy for these diversity-rich deep-sea ecosystems. [ABSTRACT FROM AUTHOR]

A new chlorinated phenolic compound, methyl-3-chloro-2-hydroxy-4-methoxy-6-pentylbenzoate (1) and 4 known compounds (2-5) were isolated from the Antarctic lichen, Pertusaria dactylina ( Pertusariaceae). The structure of the new compound was determined by means of One-dimensional and two dimensional nuclear magnetic resonance (1D and 2D NMR) and high-resolution fast atom bombardment mass spectrometry (HRFABMS) experiments. The antimicrobial activities of compounds 1 to 5 against Staphylococcus aureus and Candida albicans were evaluated. The results showed that compound 1 exhibited a weak inhibitory effect against C. albicans with an IC50 value of 67 ± 7 μg/mL.

Varicellaria carneonivea is lichenized with Asterochloris phycobiontica, a new member of the Chlorococcales. In the free-living state this species reproduces by zoo- and aplanospores; autospores are lacking. Sporangia develop an internal local thickening of the wall, which marks the later aperture. A group of dictyosomes apparently contributes to the formation of this thickening. Secondary carotenoids dissolved in droplets or irregular masses of oil are deposited around and within the pyrenoid. XVIII, f. 17 from Dillenius as lectotype. This represents Lecanora dispersa (Pers.)
