Double-stranded RNA (dsRNA)-mediated gene silencing, also known as RNA interference (RNAi), has been a breakthrough technology for functional genomic studies and represents a potential tool for the management of insect pests. Since the inception of RNAi numerous studies documented successful introduction of exogenously synthesized dsRNA or siRNA into an organism triggering highly efficient gene silencing through the degradation of endogenous RNA homologous to the presented siRNA. Managing hemipteran insect pests, especially Halyomorpha halys (Stål) (Heteroptera: Pentatomidae), the brown marmorated stink bug (BMSB), is critical to food productivity. BMSB was recently introduced into North America where it is both an invasive agricultural pest of high value specialty, row, and staple crops, as well as an indoor nuisance pest. RNAi technology may serve as a viable tool to manage this voracious pest, but delivery of dsRNA to piercing-sucking insects has posed a tremendous challenge. Effective and practical use of RNAi as molecular biopesticides for biocontrol of insects like BMSB in the environment requires that dsRNAs be delivered in vivo through ingestion. Therefore, the key challenge for molecular biologists in developing insect-specific molecular biopesticides is to find effective and reliable methods for practical delivery of stable dsRNAs such as through oral ingestion. Here demonstrated is a reliable delivery system of effective insect-specific dsRNAs through oral feeding through a new delivery system to induce a significant decrease in expression of targeted genes such as JHAMT and Vg. This state-of-the-art delivery method overcomes environmental delivery challenges so that RNAi is induced through insect-specific dsRNAs orally delivered to hemipteran and other insect pests., Competing Interests: The authors have declared that no competing interests exist.

RNA interference is a post- transcriptional, gene regulation mechanism found in virtually all plants and animals including insects. The demonstration of RNAi in insects and its successful use as a tool in the study of functional genomics opened the door to the development of a variety of novel, environmentally sound approaches for insect pest management. Here the current understanding of the biogenesis of the two RNAi classes in insects is reviewed. These are microRNAs (miRNAs) and short interfering RNAs (siRNAs). Several other key approaches in RNAi -based for insect control, as well as for the prevention of diseases in insects are also reviewed. Invertebrate iridescent virus 6 (IIV6) was determined to cause infection in Phyllophaga vandinei Smyth (Coleoptera: Scarabaeidae) through a range of modes of transmissions. This is the first evidence of IIV6 infection in P. vandinei that caused both patent and sub-lethal infections in larvae and adults. Mortality rates were determined to be ~30% when virus inoculum was injected into larvae or adults. Adults injected with virus showed dramatically altered behavior; injected beetles were not observed feeding or mating compared with adults injected with buffer or adults that were not injected. Tissue collected from infected adults resulted in infection when injected into healthy adults, as confirmed with PCR. PCR also confirmed that frass of infected larvae and adults contained virus, and when reconstituted frass from infected individuals was injected into healthy adults or larvae they become infected. Healthy adults could be infected by coming into contact with soil or plant material that had been exposed to infected adults as much as two weeks prior to introduction of nonvirus exposed adults. Although relatively low mortality resulted when adults or larvae were injected with the virus, the demonstration of horizontal transmission, potentially through frass of infected individuals, identifies a mode of transmission that may be exploited as a potential management tool to reduce P. vandinei.

We report the toxicological effects of a new biorational, synthetic sucrose octanoate (AVA Chemical Ventures L.L.C., Portsmouth, NH), on brown citrus aphid, Toxoptera citricida (Kirkaldy), nymphs and adults and to its native parasitoid Lysiphlebus testaceipes (Cresson). Sucrose octanoate topically applied was equally toxic to brown citrus aphid adults and nymphs with LC50 and LC90 values ranging from 356 to 514 and 1029 to 1420 ppm, respectively. Mortalities of both stages did not differ significantly over time during the 3-24-h sampling period. Dry residues of sucrose octanoate exhibited similar levels of toxicity to both nymphs and adults. Mortality ranged from 60 to 70% at 6,000 ppm 4 h after exposure. L. testaceipes was not harmed by treatments as high as 4,000 ppm of sucrose octanoate as long as the parasitoid had mummified before treatment. Based on these results, sucrose octanoate would be a useful biorational in citrus integrated pest management programs.

Leaves of the tropical legume Tephrosia candida DC deterred feeding by adults of the Diaprepes root weevil, Diaprepes abbreviatus (L.), compared with leaves of Citrus macrophylla Wester, a common citrus rootstock, or T. vogelii Hook. f. When larvae were placed in pots containing plants of the three species for 28 d in a growth chamber, larval survival and weight gain were significantly reduced in pots containing plants of T. candida compared with larvae in pots with C. macrophylla or T. vogelii. Diet incorporation of lyophilized roots of T. candida into an artificial diet increasingly inhibited the growth of larvae and increased larval mortality with increased concentration of roots, whereas roots from C. macrophylla or T. vogelii had no effect compared with the diet-only control. T. candida, but not T. vogelii, contains at least one constituent that acts as an antifeedant toward adult D. abbreviatus and as a toxicant toward larvae. No antifeedant effect of roots of T. candida toward larvae was observed in no-choice pot tests or in a diet incorporation bioassay. In pots, larval feeding damage to roots of T. candida was evident. In the diet incorporation assay, 97% of larvae survived 29 d on a diet of cellulose powder (a nutritionally inert filler) despite losing weight. We conclude that decreased survival and weight gain of larvae-fed fresh or lyophilized roots of T. candida were the result of ingestion of a toxicant and not deterrence from feeding.
